Abstract

AMPK regulates cellular energy balance using its γ subunit as an energy sensor of cellular AMP and ADP to ATP ratios. Here, the authors show that γ2 AMPK activation lowers heart rate by reducing the activity of pacemaker cells, whereas loss of γ2 AMPK increases heart rate and prevents the adaptive bradycardia of endurance training in mice.

Abstract

Per- and polyfluoroalkyl substance (PFAS) are increasingly becoming the contamination issue of our time, due to their environmental persistence, bioaccumulation and mounting evidence of their toxicity. Herein we examined the pyrolytic decomposition of a series of perfluorocarboxylic acids via the production of CO and CO2, utilizing FTIR and GC/MS. We observed the presence of a parallel surface-mediated reaction in conjunction with the gas-phase pyrolysis.

82. Thermal Decomposition of Chlorpyrifos - an Experimental Investigation

Abstract

Organophosphate compounds are a group of chemicals that are commonly used in pest control. One widely used organophosphate is chlorpyrifos (CPF), which has been associated with acetylcholinesterase inhibition (AChE), an acute neurotoxin. Further, under thermal decomposition conditions, such as those that occur in bushfires, CPF has been shown to produce a dioxin-like compound 2,3,7,8-tetrachloro-[1,4]-dioxinodipyridine (TCDDpy). To assist in understanding the reaction of CPF, an experimental methodology was developed which enabled the study of its thermal decomposition. CPF was pyrolysed in a three-zone furnace, leading to the formation of its major degradation product, 3,5,6-trichloro-2-pyridinol (TCP). The study revealed that ethylene gas and a solid sulfur/phosphorous compound were also generated with the TCP product. TCP undergoes further thermal decomposition to form several chlorinated products. Experiments undertaken under oxidative conditions revealed that TCP (at temperatures above 900 K and at a residence time of 9 s) also produced HCl and cis and trans forms of TCDDpy. The results of this study compare favourably with our previous theoretical work.

83. Thermal degradation of perfluorooctanoic acid (PFOA)

Abstract

Per - and polyfluoroalkyl substances (PFAS) are a group of environmental contaminants, that are dispersed throughout the world. A number of areas have been affected by their persistence and biomagnification. PFAS remediation of contaminated material requires extensive knowledge of the species involved. A method to simplify the study of the thermal degradation of PFAS is presented in this paper, providing a solution to experimental and analytical challenges. A three-zone furnace allowed manipulation of both reaction temperature and gas flowrate (and consequently residence time). The PFAS compound could be charged and vaporised in a separate zone. Adopting this experimental methodology, a kinetic model describing the decomposition of the PFAS can be devised. When reacting under an inert nitrogen atmosphere, perfluorooctanoic acid (PFOA) degraded at temperatures above 450 °C. The products observed were found to be hydrofluoric acid (HF) and carbon dioxide (CO2), along with a perfluoro-1-heptene species. Additional products of 1H-perfluorohexane or 1H-perfluoroheptane were also observed when residence times were increased. The effect of water vapour was also investigated, with similar behaviour to pyrolysis being observed, where a significantly higher concentration of HF was detected under otherwise similar reaction conditions. These preliminary results suggest water vapour accelerates the rate of PFAS decomposition.

Abstract

Catalytic cracking of high molecular weight hydrocarbons underpins the production of fossil fuels from petroleum vapour and the recycling of polyolefin waste plastic. However, thermal cracking over conventional microporous solid acids is hindered by poor mass‐transport. Here we explore the performance of hierarchical H−Y zeolites and Al‐SBA‐15 for the catalytic cracking of 1,3,5‐triisopropylbenzene (1,3,5‐TIPB) and low‐density polyethylene (LDPE) in a continuous fixed‐bed flow reactor. Dealumination by acid washing was used to create hierarchical mesoporosity in H−Y zeolite and modify the solid acidity. Physicochemical properties were studied by X‐ray diffraction (XRD), transmission electron microscopy (TEM), scanning electron microscopy (SEM), gas adsorption, in‐situ Fourier transform infrared (FTIR), ex‐situ pyridine DRIFT, 29Si and 27Al nuclear magnetic resonance (NMR), and 1H relaxation and pulsed field gradient (PFG) NMR diffusion studies. Despite weakening acidity, the introduction of hierarchical porosity promotes deep cracking of both feedstocks; HNO3 dealuminated H−Y produces five times more cumene and benzene from 1,3‐5‐TIPB, and 33 % more benzene and xylenes from LDPE, than the parent H−Y. Abstract

As sessile, photoautotrophic organisms, plants are subjected to fluctuating sunlight that includes potentially detrimental ultraviolet-B (UV-B) radiation. Experiments under controlled conditions have shown that the UV-B photoreceptor UV RESISTANCE LOCUS 8 (UVR8) controls acclimation and tolerance to UV-B in Arabidopsis thaliana ; however, its long-term impact on plant fitness under naturally fluctuating environments remain poorly understood. Here, we quantified the survival and reproduction of different Arabidopsis mutant genotypes under diverse field and laboratory conditions. We found that uvr8 mutants produced more fruits than wild type when grown in growth chambers under artificial low-UV-B conditions but not under natural field conditions, indicating a fitness cost in the absence of UV-B stress. Importantly, independent double mutants of UVR8 and the blue light photoreceptor gene CRYPTOCHROME 1 (CRY1) in two genetic backgrounds showed a drastic reduction in fitness in the field. Experiments with UV-B attenuation in the field and with supplemental UV-B in growth chambers demonstrated that UV-B caused the cry1 uvr8 conditional lethal phenotype. Using RNA-seq data of field-grown single and double mutants, we explicitly identified genes showing significant statistical interaction of UVR8 and CRY1 mutations in the presence of UV-B in the field. They were enriched in Gene Ontology categories related to oxidative stress, photoprotection and DNA damage repair in addition to UV-B response. Our study demonstrates the functional importance of the UVR8-mediated response across life stages in natura , which is partially redundant with that of cry1.
