Abstract

Structural characterization and determination of molecular properties for a protein fragment isolated from the cortex of nonpathological calf lenses were done. The polypeptide was found to be a stable fragment of the N-terminal region of alpha-crystallin B chain produced by in vivo hydrolysis. Its folding both in vivo and in vitro is independent of the native C-terminal region. It is proposed that proteolytic enzymes of the lenses catalyze the fragmentation of the polypeptide.

Abstract

Abstract Hepatocellular carcinoma (HCC) is a complex and heterogeneous tumor with multiple genetic aberrations. Several molecular pathways involved in the regulation of proliferation and cell death are implicated in the hepatocarcinogenesis. The major etiological factors for HCC are both hepatitis B virus (HBV) and hepatitis C virus infection (HCV). Continuous oxidative stress, which results from the generation of reactive oxygen species (ROS) by environmental factors or cellular mitochondrial dysfunction, has recently been associated with hepatocarcinogenesis. On the other hand, a distinctive pathological hallmark of HCC is a dramatic down-regulation of oxido-reductive enzymes that constitute the most important free radical scavenger systems represented by catalase, superoxide dismutase and glutathione peroxidase. The multikinase inhibitor sorafenib represents the most promising target agent that has undergone extensive investigation up to phase III clinical trials in patients with advanced HCC. The combination with other target-based agents could potentiate the clinical benefits obtained by sorafenib alone. In fact, a phase II multicenter study has demonstrated that the combination between sorafenib and octreotide LAR (So.LAR protocol) was active and well tolerated in advanced HCC patients. The detection of molecular factors predictive of response to anti-cancer agents such as sorafenib and the identification of mechanisms of resistance to anti-cancer agents may probably represent the direction to improve the treatment of HCC.

Abstract

Pentose phosphate pathway (PPP) is a major glucose catabolism pathway that supplies the cell with a reduced form of nicotinamide adenine dinucleotide phosphate (NADPH) and ribose-5-phosphate. NADPH is necessary for the detoxification of reactive oxygen species (ROS) and reductive biosynthesis. A key player in this pathway is the enzyme glucose-6-phosphate dehydrogenase (G6PD) that reduces NADP+ to NADPH, oxidizes glucose-6-phosphate and prevents ROS accumulation. Here, we show that the natural molecule 3,4’,5-trihydroxystilbene-3-β-d-glucoside (Polydatin) inhibits glucose-6-phosphate dehydrogenase (G6PD). As expected, G6PD inhibition causes an imbalance in NADP+/NADPH ratio, leading to a redox imbalance, followed by Endoplasmic Reticulum (ER) stress, autophagy, cell cycle block and apoptosis. we have demonstrated a link between G6PD inhibition and ER stress, showing that Unfolded Protein Response mediator such as PERK and IRE-1 have a key role in inducing autophagy and apoptosis after PPP block. Moreover, combination of PPP inhibition with autophagy inhibitors, such as chloroquine, strongly potentiate cytotoxicity on cancer cells, evidencing the role of autophagy as an escaping mechanism. Abstract

Polydatin (3,4’,5-trihydroxystilbene-3-β-d-glucoside) is a natural precursor of resveratrol already approved for commercialization as food supplement. It is present in many types of plants, including grape, peanut, and cassia seed where it acts as antimycotic agent. Polydatin has been proposed to have effect on cancer, including HNSCC and breast cancer, with promising results, but its mechanism of action seems to be different from resveratrol and is poorly understood. Glucose-6-phosphate dehydrogenase (G6PD) is the limiting enzyme of the pentose phosphate pathway which have been widely shown to be fundamental for tumor growth and metastasis formation. In this work our results show that polydatin inhibit G6PD in a dose dependent manner affecting cancer cell viability, causing a dose-dependent apoptosis and cell cycle arrest. Moreover, treated cells showed a strong increase of the Unfolded Protein Response (UPR), activated by a stress in the endoplasmic reticulum (ER), autophagy, reduced migration and invasion ability. Moreover, we developed a metastatic orthotopic HNSCC model in immunocompromised mice and showed that treated group had reduced tumor growth and reduce lymph nodes metastases. In conclusion here we show that Polydatin exerts a significant inhibitory effect on pentose phosphate pathway inhibiting HNSCC growth and metastases, pointing out that polydatin may be a reliable anticancer drug., Italian Journal of Anatomy and Embryology, Vol. 121, No. 1 (Supplement) 2016

Abstract

Here, we report the design of new analogues of spirooxoindolepyrrolidine nucleus as modulators of p53 activity. Compounds (3R,7aR)-6-(4-chlorobenzyl)-1H-spiro[imidazo[1,5-c]thiazole-3,3′-indoline]-2′,5,7(6H,7aH)-trione (9c) and (3R,7aR)-5′-methyl-6-(3,4,5-trimethoxybenzyl)-1H-spiro[imidazo[1,5-c]thiazole-3,3′-indoline]-2′,5,7(6H,7aH)-trione (10d) are the most potent compounds of this series, inhibiting cell growth of different human tumor cells at submicromolar and micromolar concentrations, respectively. Compound 9cinduces apoptotic cell death in human melanoma cell line M14 at 24 h, while in the same condition, treatment with 10dshowes a clear arrest at G2/M phase inducing delay of cell cycle progression. Possibly, these activities may be due to inhibition of p53−MDM2 interaction and subsequent p53 release and activation.

Abstract

Several works have reported on the pharmaceutical usefulness of grape phytochemicals. Nevertheless, the scientific literature needs further studies to consider grape extracts as useful dietary supplements. The aim of the present work was to hypothesize for the first time the use of whole commercial red grape juices as food supplements potentially useful against both physiological and induced cardiac oxidative stress. First of all, the results indicated a good antioxidant stability of the juice sample to lyophilization that may be reasonably regarded as a suitable process for the formulation of food supplements. Then, the processed sample (lioRGJ) was tested on cardiac-derived H9C2 myocytes to ascertain its effects on reactive oxygen species (ROS) generation and caspase-3 activity incubating cardiomyocytes with lioRGJ at increasing doses (0.01-1 μg). Experiments showed an appreciable direct radical-scavenging activity at a maximum sample dose of 0.01 μg that made the caspase-3 activity decrease by about 47% (P < 0.001). Cardiac cells were exposed to 1 μM doxorubicin and its combination with different doses of lioRGJ. A maximum sample aliquot of 0.01 μg seemed to effectively contrast the induced oxidant injury, decreasing the ROS levels by about 31% and depressing the caspase-3 activity by about 60% (P < 0.001). In both assays, pro-oxidant effects at higher sample concentrations were detected as indicated by the increase in both ROS generation and apoptotic activity. The data suggested the possible employment of the juice sample as a food supplement with prospective cardioprotective benefits, although further studies are needed to optimize its dosages to avoid harmful pro-oxidant effects.

Abstract

Bacterial porins enhance the thrombin activity upon chromogen substrate chromozym. Should porin-dependent enhancement of thrombin activity take place also upon fibrinogen in vivo, this might greatly increase the fibrin production which, in turn, might lead to blood vessel obstruction. In this study, we demonstrate fibrin hyperproduction in a simplified coagulative system, consisting of fibrinogen and thrombin-pure molecules, in the presence of bacterial porins. In particular, bacterial porins, in the presence of thrombin, significantly increased the fibrin production compared with thrombin alone. Also, fibrin hyperproduction took place even in the presence of the thrombin inhibitors antithrombin III (AT III) or alpha2 macroglobulin (alpha2M). However, the thrombin-fibrinogen reaction in the presence of AT III or alpha2M did not generate fibrin, unless porins were present. In conclusion, porins not only enhance thrombin activity but also inhibit the antithrombin activity exerted by AT III or alpha2M. We hypothesize that, because of porins activity, fibrin is largely generated due to thrombin hyperactivation. Moreover, further fibrin is produced by thrombin, which is not blocked by two serpins for the presence of porins. These results might be relevant as to the occurrence of disseminated intravascular coagulation in sepsis by gram-negative bacteria, which are known to produce porins.

Abstract

Chromatographic separation of alpha-crystallin incubated with [3H]-labelled galactose showed the radioactivity to be concentrated in the low molecular mass subunits (20 and 40 kDa). The effect of glycation on the structural organization of alpha-crystallin was evaluated by FPLC analysis of native (pH 6.8 and 8.2) and glycated protein in dissociating conditions. Results suggest that the glycation acts on the protein surface by altering its charge distribution.
