Photoactivatable and photoconvertible fluorescent proteins capable of pronounced light-induced spectral changes are a powerful addition to the fluorescent protein toolbox of the cell biologist. They permit specific tracking of one subcellular structure (organelle or cell subdomain) within a differentially labelled population. They also enable pulse-chase analysis of protein traffic. The Kaede gene codes for a tetrameric protein found in the stony coral Trachyphyllia geoffroyi, which emits green fluorescence that irreversibly shifts to red following radiation with UV or violet light. We report here the use of Kaede to explore the plant secretory pathway. Kaede versions of the Golgi marker sialyl-transferase (ST-Kaede) and of the vacuolar pathway marker cardosin A (cardA-Kaede) were engineered. Several optical devices enabling photoconversion and observation of Kaede using these two constructs were assessed to optimize Kaede-based imaging protocols. Photoconverted ST-Kaede red-labelled organelles can be followed within neighbouring populations of non-converted green Golgi stacks, by their gradual development of orange/yellow coloration from de novo synthesis of Golgi proteins (green). Results highlight some aspects on the dynamics of the plant Golgi. For plant bio-imaging, the photoconvertible Kaede offers a powerful tool to track the dynamic behaviour of designated subpopulations of Golgi within living cells, while visualizing the de novo formation of proteins and structures, such as a Golgi stack.

Background information. The idea that GPCRs (G-protein-coupled receptors) may exist as homo- or hetero-oligomers, although still controversial, is now widely accepted. Nevertheless, the functional roles of oligomerization are still unclear and gaining greater insight into the mechanisms underlying the dynamics of GPCR assembly and, in particular, assessing the effect of ligands on this process seems important. We chose to focus our present study on the effect of MT7 (muscarinic toxin 7), a highly selective allosteric peptide ligand, on the oligomerization state of the hM1 (human M1 muscarinic acetylcholine receptor subtype). Results. We analysed the hM1 oligomerization state in membrane preparations or in live cells and observed the effect of MT7 via four complementary techniques: native-PAGE electrophoresis analysed by both Western blotting and autoradiography on solubilized membrane preparations of CHO-M1 cells (Chinese-hamster ovary cells expressing muscarinic M1 receptors); FRET (fluorescence resonance energy transfer) experiments on cells expressing differently tagged M1 receptors using either an acceptor photobleaching approach or a novel fluorescence emission anisotropy technique; and, finally, by BRET (bioluminescence resonance energy transfer) assays. Our results reveal that MT7 seems to protect the M1 receptor from the dissociating effect of the detergent and induces an increase in the FRET and BRET signals, highlighting its ability to affect the dimeric form of the receptor. Conclusions. Our results suggest that MT7 binds to a dimeric form of hM1 receptor, favouring the stability of this receptor state at the cellular level, probably by inducing some conformational rearrangements of the pre-existing muscarinic receptor homodimers.

The genus Ramonda includes three preglacial paleoendemic species surviving as the rare resurrection angiosperms of the Northern hemisphere in refugia habitats in the Balkan (Ramonda nathaliae and Ramonda serbica) and Iberian Peninsulas (Ramonda myconi). This study focuses on: assessing genome size and base composition, determining chromosome number and ploidy level in several populations, evaluating inter- and intra-specific variations in DNA content and chromosome number as well as looking for the possible hybridization in the sympatric zones of Balkan species. R. nathaliae and R. myconi are diploid species (2n = 2x = 48) while R. serbica is hexaploid (2n = 6x = 144). The mean 2C DNA values ranged from 2.30 pg for R. nathaliae to 2.59 pg for R. myconi compared to 7.91 pg for R. serbica. The base composition for R. nathaliae was 42.1% GC, for R. myconi 39.9% and for R. serbica 41.2%. In one population of R. serbica the DNA content ranged from 2C = 7.65 to 11.82 pg, revealing different ploidy levels among its individuals. In sympatric populations genome size was intermediary (∼5 pg) between the diploid and hexaploid classes which indicates the hybridization ability between R. serbica and R. nathaliae. It appears that polyploidization is the major evolutionary mechanism in the genus Ramonda.

Original and simple procedures for glassy carbon electrode modification with polyoxometalates (POMs), phosphotungstate [H7P8W48O184]33-, and Co(II)-containing silicotungstates [Co6(H2O)30{Co9Cl2(OH)3(H2O)9(β-SiW8O31)3}]5- and [{Co3(B-β-SiW9O33(OH))(B-β-SiW8O29OH)2}2]22- give stable and very active surfaces for the hydrogen-evolution reaction (HER). For this purpose, the selected POMs fixed on Vulcan XC-72 were adsorbed on the electrode surface or were directly entrapped in polyvinylpyridine films on the electrode. Cyclic voltammetry and confocal microscopy results converge to indicate that the activation is related to the proton and electron reservoir-like behaviors of these molecular oxides and not to any electrode surface area increase. However, the Tafel parameters of the HER process, which are different from one POM to the next, are in the range of those of the best metallic electrodes.

The phytohormone auxin has been known for >50 years to be required for entry into the cell cycle. Despite the critical effects exerted by auxin on the control of cell division, the molecular mechanism by which auxin controls this pathway is poorly understood, and how auxin is perceived upstream of any change in the cell cycle is unknown. Auxin Binding Protein 1 (ABP1) is considered to be a candidate auxin receptor, triggering early modification of ion fluxes across the plasma membrane in response to auxin. ABP1 has also been proposed to mediate auxin-dependent cell expansion, and is essential for early embryonic development. We investigated whether ABP1 has a role in the cell cycle. Functional inactivation of ABP1 in the model plant cell system BY2 was achieved through cellular immunization via the conditional expression of a single-chain fragment variable (scFv). This scFv was derived from a well characterized anti-ABP1 monoclonal antibody previously shown to block the activity of the protein. We demonstrate that functional inactivation of ABP1 results in cell-cycle arrest, and provide evidence that ABP1 plays a critical role in regulation of the cell cycle by acting at both the G1/S and G2/M checkpoints. We conclude that ABP1 is essential for the auxin control of cell division and is likely to constitute the first step of the auxin-signalling pathway mediating auxin effects on the cell cycle.

Background: Adipose-derived stem cells (ASCs) are a potential adult mesenchymal stem cell source for restoring endothelial function in critical limb ischemia patients. Fibroblast growth factors (FGF) play a major role in angiogenesis and wound healing. The purpose of this study was to evaluate the effects of FGF and VEGF on human ASCs proliferation, migration and potential endothelial differentiation regards to their use as endothelial cell (EC) substitutes. Methods: ASCs were isolated from clinical lipoaspirates and cultured in M199 medium with FBS (10%), FGF2 (10ng/ml), VEGF (50ng/ml) or combinations of FGF2 and VEGF. Cell proliferation rates, viability, and migration were measured by growth curves, MTT and scratch assays. For cell attachment determinations, ASCs were seeded onto scaffold (small intestinal submucosa) for 5d. Endothelial differentiation capabilities of ASC were confirmed by expression of EC specific markers using qPCR, immunofluorescence staining and cord formation on Matrigel. PD173074, a selective inhibitor of FGF receptor, was used to confirm the importance of FGF signaling. Results: Increased proliferation rates were observed in ASCs treated with FGF2 when compared to ASCs cultured with VEGF and control groups (3.2 fold, p Conclusions: Our results indicate that: 1) FGF is an ASC promoter for proliferation, migration, attachment and endothelial differentiation; 2) FGF and VEGF have a co-stimulatory effect on ASC endotheliogenesis; and 3) FGF signaling mediates ASC EC differentiation. These results suggest that ASCs with enhanced FGF2 signaling may potentially be used for tissue engineering and cell-based therapies in critical limb ischemia patients.

The structural diversity of natural sulfated glycosaminoglycans (GAGs) presents major promise for discovery of chemical biology tools or therapeutic agents. Yet, few GAGs have been identified so far to exhibit this promise. We reasoned that a simple approach to identify such GAGs is to explore sequences containing rare residues, for example, 2-O-sulfonated glucuronic acid (GlcAp2S). Genetic algorithm-based computational docking and filtering suggested that GlcAp2S containing heparan sulfate (HS) may exhibit highly selective recognition of antithrombin, a key plasma clot regulator. HS containing only GlcAp2S and 2-N-sulfonated glucosamine residues, labeled as HS2S2S, was chemoenzymatically synthesized in just two steps and was found to preferentially bind antithrombin over heparin cofactor II, a closely related serpin. Likewise, HS2S2S directly inhibited thrombin but not factor Xa, a closely related protease. The results show that a HS containing rare GlcAp2S residues exhibits the unusual property of selective antithrombin activation and direct thrombin inhibition. More importantly, HS2S2S is also the first molecule to activate antithrombin nearly as well as the heparin pentasaccharide although being completely devoid of the critical 3-O-sulfonate group. Thus, this work shows that novel functions and mechanisms may be uncovered by studying rare GAG residues/sequences.

Intraspecific variation in nuclear DNA content in 19 populations of Armeria maritima was investigated. Significant differences in DNA amount among ecogeographic groups and among populations were observed. These differences are related to geographic origin, with Californian populations having 7 % more DNA than their European counterparts. Within Europe, the four ecogeographic groups tested are not distinct, whatever their geographic origin (maritime versus continental) and their ecological characteristics (heavy-metal polluted versus non-toxic soils). However, significant differences in DNA amount appeared among population clusters identified from biochemical classification of European populations based on flavonoid compounds and isozyme markers. Variation in genome size among populations was found to be significantly correlated to variation in pollen size. Weak interpopulational variation has been detected for the nucleotidic base ratio, but was not correlated to variation in DNA amount.

Flow-cytometric analysis was carried out to determine the effect of low temperature on the changes in proportion of 2C:4C complement of DNA in root cells from three different zones (meristem apex, elongation and differentiation) in seedlings of Medicago sativa cv. Sitel. 2C:4C complement of DNA in cells were estimated after the treatments of the roots at control temperature of24°C, elevated temperatures of26°C, 30°C, 37°C and low temperatures—10°C and 5°C (in three different periods of duration 15, 38, 45 days). The observed results showed that at control temperature percentage of nuclei 2C:4C in meristem apex is 65:35, in elongation zone 50:50 and in differentiation zone is 40:60. The elevated temperatures (26°C, 30°C, 37°C) influenced this proportion very slightly while treatments of roots with low temperature especially in a period of 45 days changed 2C:4C complement of DNA drastically. Four wild Ribes species (Ribes alpinum L., Ribes petraeum Wulf., Ribes rubrum L., and Ribes uva- crispa L.; all 2n = 2x = 16) were surveyed for their chromosome and genome organisation. Their genome size was assessed using flow cytometry. Ribes alpinum had 5.3% more nuclear DNA than did the three other species, whose average was 2C = 1.91 pg with 40.4% GC. In addition, GC- and AT-rich heterochromatin and rDNA (18S–5.8S–26S and 5S) patterns were studied using fluorochrome banding and double-target fluorescence in situ hybridization (FISH), respectively. Only GC-rich heterochromatin was detected, co-localizing with 18S–26S rDNA. Fluorochrome banding and FISH patterns revealed marked differences between species. Ribes alpinum and R. uva-crispa differed from R. rubrum and R. petraeum by the number of 18S–26S sites and the localization of 5S rDNA. Ribes alpinum and R. uva-crispa were differentiated by the number of 5S sites. Ribes rubrum and R. petraeum also differed by the number of 5S sites and by the size of the GC-rich band on the satellite chromosome pair. This paper examines the molecular machinery involved in membrane exchange within the plant endomembrane system. A study has been undertaken on beta-COP-like proteins in plant cells using M3A5, an antibody raised against the conserved sequence of mammalian beta-COP proteins. In mammalian cells, beta-COP proteins are part of a complex named the coatomer, which probably recruits some specific areas of the endomembrane system. Immunofluorescence analyses by confocal laser scanning microscopy showed that beta-COP-like proteins marked predominantly the plant Golgi apparatus. Other proteins known to be part of a potential machinery for COPI vesicle formation (gamma-COP, beta'-COP and Arf1 proteins) were immunolocalized on the same membraneous structures as beta-COP. Moreover, beta-COP and other COPI antibodies stained the cell plate in dividing cells. It is further shown that, in maize root cells, and in contrast to observations upon mammalian cells, the drug Brefeldin A (BFA) does not induce the release of beta-COP and Arf1 proteins from the Golgi membrane into the cytosol. These data clearly demonstrate that the antibody M3A5 is a valuable marker for studies on trafficking events in plant cells. They also report for the first time the location of COP components in plant tissue at the light level, especially on a model well known for secretion, i.e. the maize root cells. They also suggest that the membrane recruitment machinery may function in a plant-specific way.

The genome sizes of eight species from different wild population of diploid Medicago with origin from Algeria have been assessed by flow cytometry. This parameter vary from 2C=0.94 pg in Medicago orbicularis (the smallest genome) to 2C=1.80 pg in Medicago laciniata (the largest one). The relationship between genome size and competence for direct somatic embryo formation in liquid medium for four of the tested species (M. orbicularis Orbi DZ19 C 2C=0.94; M. truncatula Tru DZ 2B 2C=1.08; M.scutelata Scu 274 2C=1.11; M. arabica Ara 46 Ina 2C=1.22) was studied. It was found that Medicago orbicularis with the smallest genome size formed somatic embryos for a shortest period of time, with a high percent of reacting explants and number of somatic embryos per explant. It was followed by M. truncatula, M. scutelata and M. arabica.

The spatial organization of chromatin can be subject to extensive remodeling in plant somatic cells in response to developmental and environmental signals. However, the mechanisms controlling these dynamic changes and their functional impact on nuclear activity are poorly understood. Here, we determined that light perception triggers a switch between two different nuclear architectural schemes during Arabidopsis postembryonic development. Whereas progressive nucleus expansion and heterochromatin rearrangements in cotyledon cells are achieved similarly under light and dark conditions during germination, the later steps that lead to mature nuclear phenotypes are intimately associated with the photomorphogenic transition in an organ-specific manner. The light signaling integrators DE-ETIOLATED 1 and CONSTITUTIVE PHOTOMORPHOGENIC 1 maintain heterochromatin in a decondensed state in etiolated cotyledons. In contrast, under light conditions cryptochrome-mediated photoperception releases nuclear expansion and heterochromatin compaction within conspicuous chromocenters. For all tested loci, chromatin condensation during photomorphogenesis does not detectably rely on DNA methylation-based processes. Notwithstanding, the efficiency of transcriptional gene silencing may be impacted during the transition, as based on the reactivation of transposable element-driven reporter genes. Finally, we report that global engagement of RNA polymerase II in transcription is highly increased under light conditions, suggesting that cotyledon photomorphogenesis involves a transition from globally quiescent to more active transcriptional states. Given these findings, we propose that light-triggered changes in nuclear architecture underlie interplays between heterochromatin reorganization and transcriptional reprogramming associated with the establishment of photosynthesis.

Summary Imaging or quantifying protein synthesis in cellulo through a well-resolved analysis of the cell cycle (also defining G1 subcompartments) is a methodological challenge. Click chemistry is the method of choice to reveal the thymidine analogue 5-ethynyl-2′-deoxyuridine (EdU) and track proliferating nuclei undergoing DNA synthesis. However, the click reaction quenches fluorescent proteins. Our challenge was to reconcile these two tools. A robust protocol based on a high-resolution cytometric cell cycle analysis in tobacco (Nicotiana tabacum) BY2 cells expressing fluorescent Golgi markers has been established. This was broadly applicable to tissues, cell clusters, and other eukaryotic material, and compatible with Scale clearing. EdU was then used with the photoconvertible protein sialyl transferase (ST)-Kaede as a Golgi marker in a photoconversion pulse-chase cytometric configuration resolving, in addition, subcompartments of G1. Quantitative restoration of protein fluorescence was achieved by introducing acidic EDTA washes to strip the copper from these proteins which were then imaged at neutral pH. The rate of synthesis of this Golgi membrane marker was low during early G1, but in the second half of G1 (30% of cycle duration) much of the synthesis occurred. Marker synthesis then persisted during S and G2. These insights into Golgi biology are discussed in terms of the cell's ability to adapt exocytosis to cell growth needs.

Background and Aims Allopolyploidy and intraspecific heteroploid crosses are associated, in certain groups, with changes in the mating system. The genus Sorbus represents an appropriate model to study the relationships between ploidy and reproductive mode variations. Diploid S. aria and tetraploid apomictic S. austriaca were screened for ploidy and mating system variations within pure and sympatric populations in order to gain insights into their putative causalities. Methods Flow cytometry was used to assess genome size and ploidy level among 380 S. aria s.l. and S. austriaca individuals from Bosnia and Herzegovina, with 303 single-seed flow cytometric seed screenings being performed to identify their mating system. Pollen viability and seed set were also determined. Key Results Flow cytometry confirmed the presence of di-, tri- and tetraploid cytotype mixtures in mixed-ploidy populations of S. aria and S. austriaca. No ploidy variation was detected in single-species populations. Diploid S. aria mother plants always produced sexually originated seeds, whereas tetraploid S. austriaca as well as triploid S. aria were obligate apomicts. Tetraploid S. aria preserved sexuality in a low portion of plants. A tendency towards a balanced 2m : 1p parental genome contribution to the endosperm was shared by diploids and tetraploids, regardless of their sexual or asexual origin. In contrast, most triploids apparently tolerated endosperm imbalance. Conclusions Coexistence of apomictic tetraploids and sexual diploids drives the production of novel polyploid cytotypes with predominantly apomictic reproductive modes. The data suggest that processes governing cytotype diversity and mating system variation in Sorbus from Bosnia and Herzegovina are probably parallel to those in other diversity hotspots of this genus. The results represent a solid contribution to knowledge of the reproduction of Sorbus and will inform future investigations of the molecular and genetic mechanisms involved in triggering and regulating cytotype diversity and alteration of reproductive modes.

Genome size has been studied for the first time in the Colchicum genus. Values obtained by flow cytometry were quite stable and specific to each taxon: C. autumnale L., 2C=5.89±0.22 pg, equivalent to 5.7×10 9 bp (2 n=4 x=36); C. alpinum DC., 2C=8.06±0.24 pg, equivalent to 7.8×10 9 bp (2 n=6 x=56); C. lusitanum Brot., 2C=10.7±0.67 pg, equivalent to 10.3×10 9 bp (2 n= approx. 10 x=90–92 and 94–96); C. multiflorum Brot., 2C=16.5±0.69 pg, equivalent to 15.9×10 9 bp, (2 n= approx. 16 x=140–148); C. corsicum Baker, 2C=21.3±0.99 pg, equivalent to 20.6×10 9 bp (2 n=22 x= approx. 198±2). Study of HS structure-function relations is, however, hampered due to the lack of availability of HS in sufficient quantities as well as the molecular heterogeneity of naturally occurring HS. Enzymatic synthesis of HS is an attractive alternative to the use of naturally occurring HS, as it reduces molecular heterogeneity, or a long and daunting chemical synthesis of HS. Heparosan, produced by E. coli K5 bacteria, has a structure similar to the unmodified HS backbone structure and can be used as a precursor in the enzymatic synthesis of HS-like polysaccharides. Here, we describe an enzymatic approach to synthesize several specifically sulfated HS polysaccharides for biological studies using the heparosan backbone and a combination of recombinant biosynthetic enzymes such as C5-epimerase and sulfotransferases.

One example, the extensively studied heparin pentasaccharide sequence-which binds antithrombin-III, inducing a conformational change that increases its serpin protease activity by 1,000-fold-is unique in that no other specific GAG-protein structure-function relations have been described to the same degree. Thus, production of heparan sulfate (HS) oligosaccharides is critical for obtaining specific structural information regarding the binding interactions of GAG and their ligands (typically proteins). Purely synthetic methods of oligosaccharide synthesis are possible, but the cost, time requirement, and difficulty of their preparation prohibit library synthesis in significant amounts. Herein, the use of bacterial heparin lyases for the production of HS oligosaccharides via enzymatic depolymerization of HS polymers is discussed. The separation and purification of these oligosaccharides by liquid chromatography are also described.

Genome size and base composition in 16 species and subspecies of the Hydrangea, a woody ornamental genus of Hydrangeaceae, were evaluated by flow cytometry in relation to their chromosome number. This is the first such study concerning the genome size of these species together with a karyotype study of the most important species, Hydrangea macrophylla subsp. macrophylla (Hortensia), from an economical point of view. The 2C DNA content ranged from 1.95 pg in Hydrangea quercifolia to 5.00 pg in Hydrangea involucrata. The base composition ranged from 39.9% GC in Hydrangea aspera subsp. sargentiana to 41.1% in Hydrangea scandens subsp. scandens (significant difference at p < 0.05). The smallest genome sizes were those of the three species originating from North or South America. Most of the species studied presented a chromosome number of 2n = 2x = 36, except for those of the section Aspereae which showed 2n = 30, 34 and 36. A primary karyotype has been made for the first time for H. macrophylla subsp. macrophylla. Phylogenetic relationships between species, the origin of chromosome number and an exploration of the genetic diversity within the genus are discussed.

To avoid polyploidy in regenerants the source of explant material should be monosomatic. Therefore, the leaf and petiole tissue of five diploid Medicago species (Medicago ciliaris, Medicago murex, Medicago orbicularis, Medicago polymorpha and Medicago truncatula cv. Jemalong, and the ecotype R108-1) was assessed for polysomaty by flow cytometry. For the species studied the frequency of 2C nuclei was about 90% in leaves compared with that in petioles. Embryos were readily formed from tissue of leaves in liquid media containing 1 mg l(-1) or 4 mg l(-1) dichlorophenoxyacetic acid (2,4-D). For embryo development two procedures were tested - prolonged use of induction medium and treatment with polyethylene glycol Mw 6000 (PEG). The highly regenerable genotypes M. truncatula cv. Jemalong and R108-1 showed efficient conversion of embryos after maturation in liquid medium. The regenerated plants were diploid and with normal phenotype.

Nod factors are important elicitors in legume-bacterium symbiosis. Any candidate plant receptor(s) for these lipo-oligosaccharides can be expected to show some lectin-like properties. A novel protein (P60), a native tetramer with 60 kDa monomers, has been isolated from a membrane fraction of Medicago sativa (lucerne, alfalfa) roots by using affinity chromatography with either GlcNAc or N,Nʹ,Nʹʹ-triacetyl-(1 → 4)-β-D-chitotriose [(GlcNAc)3] grafted to agarose beads as the matrix and, in a second step, Sephadex G-200 gel filtration. With (GlcNAc)3-agarose an additional protein of 78 kDa was isolated. P60 showed haemagglutination activity with specificity for GalNAc, GalN, GlcNAc and GlcN. Binding experiments with radioactive GlcNAc gave a Kd of 95 nM and one binding site per monomer of P60; Nod factor competed strongly for this binding. In native PAGE, protein incubated with O-sulphated Nod factors had a higher electrophoretic mobility as a consequence of binding. However, the largest modification was observed with a natural mixture of Nod factors, containing the O-acetylated and O-sulphated tetrasaccharidic NodRm-IV(Ac,S) (in which Ac stands for an O-acetylated group at the non-reducing end and S for O-sulphation at the reducing end) in addition to the non-O-acetylated NodRm-IV(S) (which alone had little effect) and NodRm-V(S). The native PAGE study was also performed with known lectins from other sources, but only the 34 kDa lectin of Phytolacca americana (pokeweed) showed any such interaction, although without discrimination between Nod factors. Finally, one peptide of each isolated protein was sequenced; the peptide from P60 showed some similarity with dihydrolipoamide dehydrogenase and ferric leghaemoglobin reductase, whereas the peptide from P78 was identical with an analogous region of 70 kDa heat shock protein.

Small GTP-binding Rho proteins are involved in signalling, cell polarity, membrane outgrowths and actin stabilization in eukaryotes. Known plant homologues represent essentially the Rac subfamily and an original Rop (Rho in pollen). Mammalian Rho proteins are preferential targets of clostridial toxins. In alfalfa ( Medicago sativa L.) cells, Clostridium botulinum C3-exoenzyme (C3) provoked disassembly of the actin cytoskeleton, similar to its effect in mammalian cells. In alfalfa proteins, several epitopes appear to be recognized by commercial antibodies raised against peptides characteristic for human Rho. One ≈ 40-kDa band was detected immunologically by anti-RhoB: a protein of this size was ADP-ribosylated by C3 and glucosylated in vitro by Clostridium difficile toxin B, without interference between the two nor from phosphatidyl inositide. C3 was also active upon a 34-kDa band which contained protein(s) immunoreactive with anti-Rac2 and which bound [γ 35 S]-GTP, but was glucosylated by neither toxin B nor Clostridium sordellii Lethal Toxin. An 18-kDa band detected by [γ 35 S]-GTP overlay was immunologically recognized by anti-Rac1. Anti-Cdc42Hs recognized a 54-kDa band. Substrates to toxin B and C3 were purified from alfalfa cell culture and partially sequenced: they included two proteins, P40 and P41, of ≈ 40 kDa (by SDS-electrophoresis). P40 appears to constitute a tetrameric aldolase (160 kDa by gel filtration; EC 4.1.2.13) whose activity is partially inhibited by toxin B and the anti-RhoB.

Leaf trichomes in Arabidopsis are unicellular epidermal hairs with a branched morphology. They undergo successive endoreduplication rounds early during cell morphogenesis. Mutations affecting trichome nuclear DNA content, such as triptychon or glabra3, alter trichome branching. We isolated new mutants with supernumerary trichome branches, which fall into three unlinked complementation groups: KAKTUS and the novel loci, POLYCHOME and RASTAFARI. They map to chromosomes IV, II, and V, respectively. The trichomes of these mutants presented an increased DNA content, although to a variable extent. The spindly-5 mutant, which displays a constitutive gibberellin response, also produces overbranched trichomes containing more nuclear DNA. We analyzed genetic interactions using double mutants and propose that two independent pathways, defined by SPINDLY and TRIPTYCHON, act to limit trichome growth. KAKTUS and POLYCHOME might have redundant actions mediating gibberellin control via SPINDLY. The overall leaf polysomaty was not notably affected by these mutations, suggesting that they affect the control of DNA synthesis in a tissue- or cell type-specific manner. Wild-type tetraploids also produce overbranched trichomes; they displayed a shifted polysomaty in trichomes and in the whole leaf, suggesting a developmental program controlling DNA increases via the counting of endoreduplication rounds.

The anion PW 12 O 3− 40 is very sparingly soluble in concentrated mineral acid solutions. This shortcoming has impeded the kind of study that demonstrated the merging of the first two waves of SiW 12 O 4− 40 and P 2 W 18 O 6− 62 by an EEC mechanism in aqueous solutions. Entrapment of PW 12 O 3− 40 in protonated slightly quaternized poly(4-vinylpyridine) films (QPVP) or in polyaniline films (PANI) has now permitted such a study. As also observed previously, a negative potential shift of the first redox wave of the heteropolyanion is obtained when the acid concentration in the bathing solution increases. All the results are compared with those obtained for QPVP–SiW 12 O 4− 40 , QPVP–P 2 W 18 O 6− 62 , PANI–SiW 12 O 4− 40 and PANI–P 2 W 18 O 6− 62 composites. In each case, the same EEC mechanism is operative. EQCM experiments were also very useful in this issue. Except for the case of PW 12 O3− 40 which could not be studied in concentrated acid solutions, all other systems show that the merging of waves necessitates higher acid concentrations in solution than when the heteropolyanions are entrapped in polymer matrices. This observation was termed the ‘microenvironment effect’. In the present work, confocal microscopy measurements of fluorescence intensity variation between bathing solutions and appropriate composites show directly the existence of differences of pH between the two phases. The results concord with predictions from Donnan effects, and reinforce such an explanation for the ‘microenvironment effect’.

The genome size and the base composition (GC%) of eight Hypochaeris species were determined by flow cytometry in order to establish the pattern of nuclear DNA variation within the genus. The species analysed showed an almost fivefold range of variation from 1.68 pg in H. cretensis to 8.10 pg in H. uniflora. This variation in DNA content is greater between taxonomic sections of Hypochaeris species than within a section. There was no correlation between 2C DNA content and GC% indicating that neither the GC fraction nor the AT fraction were preferentially associated with variation in genome size. Because there is little heterochromatin, these results show that it is interspersed repeated sequences that are most probably implicated in this variation. From phylogenetic analysis, it is likely that genome size has evolved by loss of DNA content in some lineages and by gain in one lineage from an ancestral genome which was probably similar to genomes of intermediate size in Hypochaeris.

We have used mevinolin, an inhibitor of 3-hydroxy-3-methylglutaryl- CoA reductase, to study the importance of mevalonic acid (MVA) for cell cycle progression of tobacco (Nicotiana tabacum L.) BY-2 (TBY-2) cells. Previously we have shown that in principal mevinolin blocks cells mainly at the end of G1 phase, but that cells are sensitive towards the inhibitor at the end of mitosis/entry G1 phase (Hemmerlin & Bach, 1998). The mevinolin—induced cell-cycle arrest was reversed by addition of exogenous MVA. It is shown that blockage of MVA synthesis might lead to variations in intracellular pH, which could be involved in the regulation of cell proliferation. SDS—PAGE analyses of total proteins and GTP—blot analyses indicate mevinolin—induced changes in the pattern, which can be reversed by addition of MVA. The genome size and base composition of diploid plant species from three genera of the Casuarinaceae family were determined by flow cytometry. Casuarina glauca Sieb. ex Spring. and Gymnostoma deplancheana (Miq.) L. Johnson showed a small genome with 2C = 0.70 pg, 58.6% AT, 40.5% GC for the first species and 2C = 0.75 pg, 58.7% AT, 40.5% GC for the second. Allocasuarina verticillata (Lam.) L. Johnson had a larger genome: 2C = 1.90 pg, 59.3% AT, 41.1% GC. One haploid genome of C. glauca is therefore about 340×106 base pairs. In leaves, roots or bark of these three species, polysomaty was virtually absent: a maximum frequency of 4C nuclei of only 0.08 was found in bark of C. glauca. The genome sizes of C. glauca and G. deplancheana are among the smallest described for higher plants. Small genome size, diploidy and the absence of polysomaty are advantageous traits for facilitating molecular approaches to improvement of these actinorhizal plants and developing the study of their symbiotic interactions with Frankia.

We have investigated the influence of age on B-cell responsiveness. The present study showed that the B-cell mitogen, lipopolysaccharide (LPS), similarly stimulated the proliferation of purified B lymphocytes obtained from either young mice (3 months) or old mice (24 months). In contrast, expression of the differentiation marker, alkaline phosphatase (ALP), was about fourfold higher in young mice than in older mice upon stimulation with LPS or with dextran sulfate (DXS) and interleukin-5 (IL-5). The occurrence of apoptosis during aging was then studied: unexpectedly, spontaneous cell death was double in B lymphocytes from young mice compared to older animals. Stimulation with DXS with or without IL-5 rescued B lymphocytes from cell death in young mice but protection decreased with aging, and no longer occurred in 24-month-old mice B cells. Meanwhile, the protective activity conferred by IL-4 was maintained at similar levels throughout aging. However, B cells from old mice were more responsive to apoptosis induction with cycloheximide, dibutyryl cAMP and dexamethasone. Together, the present results indicate an age-associated alteration in apoptosis and activation of B lymphocytes which could contribute to the age-related decline of the immune response.

Seven Quercus species, four deciduous (Q. cerris, Q. petraea, Q. pubescens, andQ. robur) and three evergreen (Q. coccifera, Q. ilex, andQ. suber), were assessed for DNA content. Their genome sizes ranged from 1.88 to 2.00 pg/2C, namely an interspecific DNA content variation of 6%. In addition, the DNA content of several populations of Q. petraea andQ. robur with different geographical origins, French and Croatian, was estimated. DNA content did not vary among four populations of Q. robur, but did differ between two populations of Q. petraea, the French population having a higher 2C DNA value than the Croatian. The genome size of this French population showed important dispersion among 10 measured individuals. This was in accordance with subsequent cytogenetic data that revealed extra chromosomes in some individuals from this population. The percent GC varied little between the species; a GC content of 39.9% is established as typical for oak.Key words: Quercus, flow cytometry, genome size, GC percent, DNA content variations, extra chromosomes.

The present report examines whether the presence of an additional chromosome can be detected as modifying the nuclear DNA amount and base composition of the cell, determined here by flow cytometry of interphasic nuclei, using four monosomic additions (chromosomes 3C, 4C, 7C and 8C transmitted from Allium cepa to Allium fistulosum L.). A. cepa differs significantly from A. fistulosum in genome size (2C DNA = 33.2 pg in A. cepa and 23.3 pg in A. fistulosum) as well as in GC content (38.7% and 39.8%, respectively). The presence of an extra chromosome of A. cepa obviously increases the nuclear DNA amount above the A. fistulosum value but also alters the apparent mean GC content. By comparing the monosomic additions and the parental background, the DNA amount and base composition of each of the four single chromosomes were calculated to quantify the GC content per chromosome and therefore to deduce their initial contribution to the A. cepa genome. Taken individually, some chromosomes are atypical in terms of GC content: the single chromosome 3C is AT-rich, having only about only 25% GC. However, the three other chromosomes examined are typical of the A. cepa genome in base composition. Indeed, this biological panel gives access to chromosomal features via a cytometric assay of nuclei. It should facilitate quantification of GC-rich repetitive sequences forming heterochromatic domains located mainly at the telomeres in the monocotyledonous A. cepa genome.

This paper describes black/white differences in risk factors for atherosclerosis in the large multicenter Atherosclerosis Risk in Communities (ARIC) Project sponsored by the National Heart, Lung, and Blood Institute. It is based on data collected at baseline in ARIC's four geographically distinct clinical centers. Participants were randomly selected (4264 black and 11,479 white men and women, ages forty-five to sixty-four years at entry). There were striking differences in obesity between black and white women, higher fasting glucose and greater prevalence of diabetes in blacks, and lower high-density lipoprotein values in white men. Not unexpectedly, blood pressure in black participants exceeded that in whites. Clustering of multiple risk factors was more common in the black population. Conversely, prevalence of no risk factors was greatest among whites. In conclusion, while African-Americans and Caucasians share much the same group of risk factors for atherosclerosis, there are clinically important racial differences in emphasis.

A systematic analysis of Pennisetum species has shown their cytological variability for basic chromosome number (x = 5, 7, 8, and 9), ploidy level, and chromosome size. We have identified the genome size and base composition of 15 species using flow cytometric procedure in order to test the relationship of these characters with the variation in basic chromosome number and to evaluate the ascendant dysploidy hypothesis from an ancestral x = 5 group. Ethidium bromide, bisbenzimlde Hoechst 33342, and chromomycln dyes have been used for genome size, percentage of AT, and percentage of GC nucleotides determination, respectively. Results show that genome sizes are variable among the Pennisetum species. This variation appears to be negatively related to the basic chromosome number. On the contrary, genome size Is not correlated with percentage of GC. This result shows that loss or gain of DNA that has occurred during evolution within this genus seems not to be especially associated with GCor AT-rich sequences.

The eukaryotic cell division cycle is coordinated by cyclin-dependent kinases (CDKs), represented by a single major serine/threonine kinase in yeasts (Cdc2/CDC28) and a family of kinases (CDK1 to CDK8) in human cells. Previously, two cdc2 homologs, cdc2MsA and cdc2MsB, have been identified in alfalfa (Medicago sativa). By isolating cDNAs using a cdc2MsA probe, we demonstrate here that at least four additional cdc2 homologous genes are expressed in the tetraploid alfalfa. Proteins encoded by the new cdc2MsC to cdc2MsF cDNAs share the characteristic functional domains of CDKs with the conserved and plant-specific sequence elements. Transcripts from cdc2MsA, cdc2MsB, cdc2MsC, and cdc2MsE genes are synthesized throughout the cell cycle, whereas the amounts of cdc2MsD and cdc2MsF mRNAs peak during G2-to-M phases. The translation of Cdc2MsA/B, Cdc2MsD, and Cdc2MsF proteins follows the pattern of transcript accumulation. The multiplicity of kinase complexes with cell cycle phase-dependent activities was revealed by in vitro phosphorylation experiments. Proteins bound to p13suc1-Sepharose or immunoprecipitated with Cdc2MsA/B antibodies from cells at G1-to-S and G2-to-M phase boundaries showed elevated kinase activities. the Cdc2MsF antibodies separated a G2-to-M phase-related kinase complex. Detection of histone H1 phosphorylation activities in fractions immunoprecipitated with antimitotic cyclin (CyclinMs2) antibodies from G2-to-M phase cells indicates the complex formation between this cyclin and a kinase partner in alfalfa. The observed fluctuation of transcript levels, amounts, and activities of kinases in different cell cycle phases reflects a multilevel regulatory system during cell cycle progression in plants.

Rhizobium meliloti can interact symbiotically with Medicago plants thereby inducing the formation of root nodules. Screening of a young nodule cDNA library led to the isolation of a cDNA from Medicago sativa, Msgbl, that comprises a new member of the RACK1 (Receptor of Activated C Kinase) subfamily of WD-repeat proteins. This subfamily shows homology to the β-subunit of heterotrimeric G proteins. Besides RACK1, this subfamily contains several plant genes including the well characterized auxin-inducible ArcA of tobacco. The Msgbl gene is strongly expressed in young embryos and in leaves, and is induced upon cytokinin treatment of roots. Whereas northern analysis failed to reveal differences in expression between total RNA from roots and nodules, in situ hybridization demonstrated that the transcript was most abundant in dividing cells of nodule primordia and in the nodule meristem. Msgbl may be related to the signal transduction acting in response to hormone-mediated cell division.

International audience; The semi-aquatic legumes belonging to the genus Aeschynomene constitute a premium system for investigating the origin and evolution of unusual symbiotic features such as stem nodulation and the presence of a Nod-independent infection process. This latter apparently arose in a single Aeschynomene lineage. But how this unique Nod-independent group then radiated is not yet known. We have investigated the role of polyploidy in Aeschynomene speciation via a case study of the pantropical A. indica and then extended the analysis to the other Nod-independent species. For this, we combined SSR genotyping, genome characterization through flow cytometry, chromosome counting, FISH and GISH experiments, molecular phylogenies using ITS and single nuclear gene sequences, and artificial hybridizations. These analyses demonstrate the existence of an A. indica polyploid species complex comprising A. evenia (C. Wright) (2n = 2x = 20), A. indica L. s. s. (2n = 4x = 40) and a new hexaploid form (2n = 6x = 60). This latter contains the two genomes present in the tetraploid (A. evenia and A. scabra) and another unidentified genome. Two other species, A. pratensis and A. virginica, are also shown to be of allopolyploid origin. This work reveals multiple hybridization/polyploidization events, thus highlighting a prominent role of allopolyploidy in the radiation of the Nod-independent Aeschynomene.

N-terminal fatty acylations (N-myristoylation [MYR] and S-palmitoylation [PAL]) are crucial modifications affecting 2 to 4% of eukaryotic proteins. The role of these modifications is to target proteins to membranes. Predictive tools have revealed unexpected targets of these acylations in Arabidopsis thaliana and other plants. However, little is known about how N-terminal lipidation governs membrane compartmentalization of proteins in plants. We show here that h-type thioredoxins (h-TRXs) cluster in four evolutionary subgroups displaying strictly conserved N-terminal modifications. It was predicted that one subgroup undergoes only MYR and another undergoes both MYR and PAL. We used plant TRXs as a model protein family to explore the effect of MYR alone or MYR and PAL in the same family of proteins. We used a high-throughput biochemical strategy to assess MYR of specific TRXs. Moreover, various TRX–green fluorescent protein fusions revealed that MYR localized protein to the endomembrane system and that partitioning between this membrane compartment and the cytosol correlated with the catalytic efficiency of the N-myristoyltransferase acting at the N terminus of the TRXs. Generalization of these results was obtained using several randomly selected Arabidopsis proteins displaying a MYR site only. Finally, we demonstrated that a palmitoylatable Cys residue flanking the MYR site is crucial to localize proteins to micropatching zones of the plasma membrane.

In the date palm, a dioecious mode (separate male and female individuals) and the late initial reproductive age (5–10 years) are major practical constraints for genetic improvement. Early selection on young seedlings could enhance breeding programmes and generate experimental male and female genetic stocks, but no cytogenetic protocol exists for sex determination in an immature date palm. Here we describe a cytological method based on chromomycin staining which demonstrates the occurrence of sexual chromosomes carrying distinctive nucleolar heterochromatin and thus offers, for the first time, the possibility of identifying male and female individuals by simple analysis of root meristems. This observation has been extended by in situ rDNA hybridization, confocal microscopy and dual-label flow cytometry of nuclei.

Phosphoenolpyruvate carboxylase (PEPC) was characterized in extracts from C4 mesophyll protoplasts isolated from Digitaria sanguinalis leaves and shown to display the structural, functional, and regulatory properties typical of a C4 PEPC. In situ increases in the apparent phosphorylation state of the enzyme and the activity of its Ca2+-independent protein-serine kinase were induced by light plus NH4Cl or methylamine. The photosynthesis-related metabolite 3-phosphoglycerate (3-PGA) was used as a substitute for the weak base in these experiments. The early effects of light plus the weak base or 3-PGA treatment were alkalinization of protoplast cytosolic pH, shown by fluorescence cytometry, and calcium mobilization from vacuoles, as suggested by the use of the calcium channel blockers TMB-8 and verapamil. The increases in PEPC kinase activity and the apparent phosphorylation state of PEPC also were blocked in situ by the electron transport and ATP synthesis inhibitors DCMU and gramicidin, respectively, the calcium/calmodulin antagonists W7, W5, and compound 48/80, and the cytosolic protein synthesis inhibitor cycloheximide. These results suggest that the production of ATP and/or NADPH by the illuminated mesophyll chloroplast is required for the activation of the transduction pathway, which presumably includes an upstream Ca2+-dependent protein kinase and a cytosolic protein synthesis event. The collective data support the view that the C4 PEPC light transduction pathway is contained entirely within the mesophyll cell and imply cross-talk between the mesophyll and bundle sheath cells in the form of the photosynthetic metabolite 3-PGA.

Phosphoenolpyruvate carboxylase (PEPC, EC 4.1.1.31) is a key enzyme of photosynthesis in C4 plants; it is specifically localized in the cytosol of mesophyll cells and is regulated by a phosphorylation/dephosphorylation process. The light-dependent phosphorylation of PEPC is triggered by an increase in the cytosolic pH (pHc) of mesophyll cell protoplasts. An epifluorescence and confocal microscopy analysis showed that the specific pH probe 2′,7′-bis-(2-carboxyethyl)-5-(and-6) carboxyfluorescein, acetoxymethyl ester (BCECF-AM), when used at low concentration, was essentially localized in the protoplast cytosol. By the nigericin null-point method and flow cytometry, the pHc of freshly isolated protoplasts was estimated to be 6.4. To observe the full activity of PEPC kinase and maximal phosphorylation of PEPC in vivo, such protoplast suspensions must first be treated with a permeant weak base. The present report shows that 20 mM NH4Cl raised the final pHc to 7.4. This method can be useful for estimating rapid changes of pHc in plant cells. © 1996 Wiley-Liss, Inc.

International audience; A 15-day survey of autofluorescence has been conducted upon infection by downy mildew [Plasmopara viticola (Berk. & M.A. Curtis) Berl. & de Toni] of leaves of a susceptible grapevine genotype. Different autofluorescence signals were followed from the cellular to the whole-leaf level by using four types of devices for fluorosensing: a macroscope, a spectrofluorimeter, a portable field optical sensor (the Multiplex 3), and a field fluorescence sensor prototype with 335 nm excitation. It was shown for the first time, by the three different techniques and at three different scales, that the stilbene-dependent violet-blue autofluorescence (VBF) had a transitory behaviour, increasing to a maximum 6 days post-inoculation (DPI) and then decreasing to a constant lower level, nevertheless significantly higher than in the control leaf. This behaviour could be sensed from both sides of the leaf. On the abaxial side, VBF could discriminate the presence of infection from 1 DPI, and on the adaxial side from 3 DPI. There was a constant increase in blue-excited green fluorescence starting from 8 DPI, concomitant with a decrease in leaf chlorophyll content sensed by one reflectance and two fluorescence indices available on the Multiplex 3 sensor. These results show that a pre-symptomatic and symptomatic sensing of downy mildew is possible by autofluorescence-based sensors, and this is potentially applicable in the field.

International audience; Research on the nitrogen-fixing symbiosis has been so far focused on two model legumes, Medicago truncatula and Lotus japonicus, which use a sophisticated infection process involving infection thread formation. However, in 25% of the legumes, the bacterial entry occurs more simply in an intercellular fashion. Among them, some semi-aquatic Aeschynomene species present the distinctive feature to form nitrogen-fixing nodules on both roots and stems following elicitation by photosynthetic bradyrhizobia that do not produce Nod factors. This interaction is believed to represent a living testimony of the ancestral state of the rhizobium-legume symbiosis. To decipher the molecular mechanisms of this unique Nod-independent nitrogen-fixing symbiosis, we previously identified A. evenia C. Wright as an appropriate model legume, because it displays all the requisites for molecular and genetic approaches. To advance the use of this new model legume species, here we characterized the intraspecific diversity found in A. evenia. For this, the accessions available in germplasm banks were collected and subjected to morphological investigations, genotyping with RAPD and SSR markers, molecular phylogenies using ITS and single nuclear gene sequences, and cross-compatibility tests. These combined analyses revealed an important intraspecific differentiation that led us to propose a new taxonomic classification for A. evenia comprising two subspecies and four varieties. The A. evenia ssp. evenia contains var. evenia and var. pauciciliata whereas A. evenia ssp. serrulata comprises var. serrulata and var. major. This study provides information to exploit efficiently the diversity encountered in A. evenia and proposes subsp. evenia as the most appropriate subspecies for future projects aimed at identifying plant determinants of the Nod-independent symbiotic process.

International audience; Endopolyploidy, i.e. the amplification of genomic DNA without mitosis, is a widespread process in plants. Cells from the tomato fruit pericarp are characterized by a wide range of ploidy levels (from 2C to 256C). Although various functional hypotheses have been attributed to endoreduplication according to the literature, evidence for a specific role of endoreduplication in transcription and metabolism control is still lacking. We have developed a new method based on bacterial artificial chromosome fluorescent in situ hybridization (BAC-FISH) that allows the in situ determination of DNA ploidy levels of individual nuclei. The advantage of this method is illustrated by the analysis of ploidy levels and cell sizes within the pericarp tissue from mature green tomato fruits. Using this cellular approach we established the ploidy map of the pericarp tissue. Based on this map, we performed a structural analysis of endoreduplicated nuclei at the level of chromatin organization, nuclear shape and relationship with mitochondria. We demonstrated a link between the ploidy level of nuclei, the complexity of their shape and the number of mitochondria at the vicinity of polyploid nuclei. The use of the DNA FISH method demonstrated that endopolyploidy leads to the formation of polytene chromosomes, whereas the use of a RNA FISH method demonstrated that the rDNA transcription was increased during polyploidization. Performing quantitative PCR (qPCR) and RT-qPCR on sorted nuclei respectively, we confirmed that endoreduplication did amplified exponentially loci for a set of specific genes allowing us to demonstrade that endoreduplication results in an increasing transcriptional activity.

Farnesylacetone is a natural terpene extracted from androgenic glands of the crustacean Carcinus maenas and is capable of inhibiting proliferation, notably in transformed mammalian cells. Flow cytometry with three lipophilic probes, diphenylhexatriene, trimethylammonium-diphenylhexatriene, and Nile red, has revealed modifications of the lipidic metabolism in transformed FR3T3-mTT4 rat fibroblasts treated by farnesylacetone, including changes in membrane fluidity. Farnesylacetone strongly increased the number of neutral lipidic droplets in the cytoplasm. Moreover, after prolonged terpene treatment, the membrane fraction of cells contained a substantial level of triglycerides. Farnesylacetone provoked an immediate but transitory increase in membrane fluidity of the cell membrane. The change in measured lipid fluidity appears to be due to these triglycerides rather than to the phospholipids.
