Summary: Pyemotes zhonghuajia Yu, Zhang & He is an ectoparasitic mite of various pests. Our studies found that P. zhonghuajia could kill Spodoptera frugiperda (Smith), Mythimna separata (Walker), and Spodoptera litura (Fabricius) at different developmental stages. Through an integrated approach of lipidomics, transcriptomics, and proteomics, we explored the immune response of S. frugiperda parasitized by P. zhonghuajia. In order to explore toxin proteins of P. zhonghuajia, we mined toxin genes by combining genome and transcriptome.

To date, no consistent oncogenic driver mutations have been identified in most adult soft tissue sarcomas; these tumors are thus generally insensitive to existing targeted therapies. Here we investigated alternate mechanisms underlying sarcomagenesis to identify potential therapeutic interventions. Undifferentiated pleomorphic sarcoma (UPS) is an aggressive tumor frequently found in skeletal muscle where deregulation of the Hippo pathway and aberrant stabilization of its transcriptional effector yes-associated protein 1 (YAP1) increases proliferation and tumorigenesis. However, the downstream mechanisms driving this deregulation are incompletely understood. Using autochthonous mouse models and whole genome analyses, we found that YAP1 was constitutively active in some sarcomas due to epigenetic silencing of its inhibitor angiomotin (AMOT). Epigenetic modulators vorinostat and JQ1 restored AMOT expression and wild-type Hippo pathway signaling, which induced a muscle differentiation program and inhibited sarcomagenesis. YAP1 promoted sarcomagenesis by inhibiting expression of ubiquitin-specific peptidase 31 (USP31), a newly identified upstream negative regulator of NFκB signaling. Combined treatment with epigenetic modulators effectively restored USP31 expression, resulting in decreased NFκB activity. Our findings highlight a key underlying molecular mechanism in UPS and demonstrate the potential impact of an epigenetic approach to sarcoma treatment.Significance: A new link between Hippo pathway signaling, NFκB, and epigenetic reprogramming is highlighted and has the potential for therapeutic intervention in soft tissue sarcomas. International audience; One-to-multiple medical image segmentation aims to directly test a segmentation model trained with the medical images of a one-domain site on those of a multiple-domain site, suffering from segmentation performance degradation on multiple domains. This process avoids additional annotations and helps improve the application value of the model. However, no successful one-to-multiple unsupervised domain adaptation (O2M-UDA) work has been reported in one-to-multiple medical image segmentation due to its inherent challenges: distribution differences among multiple target domains (among-target differences) caused by different scanning equipment and distribution differences between one source domain and multiple target domains (source–target differences). In this paper, we propose an O2M-UDA framework called dynamic domain adaptation (DyDA), for one-to-multiple medical image segmentation, which has two innovations: (1) dynamic credible sample strategy (DCSS) dynamically extracts credible samples from the target site and iteratively updates their number, thus iteratively expanding the generalization boundary of the model and minimizing the among-target differences; (2) hybrid uncertainty learning (HUL) reduces the voxel-level and domain-level uncertainty simultaneously, thus minimizing the source–target differences from the detail and entire perspective concurrently. Experiments on two one-to-multiple medical image segmentation tasks have been conducted to demonstrate the performance of the proposed DyDA. The proposed DyDA achieved competitive segmentation results and high adaptation with an average of 83.8% and 48.1% dice for the two tasks, respectively, which has improved by 21.7% and 9.2% compared with no adaptation, respectively. Chronic sclerosing sialadenitis of the submandibular gland (also known as Küttner tumor) is characterized by concomitant swelling of the submandibular glands secondary to strong lymphocytic infiltration and fibrosis. The pathogenesis of this disease has been unclear, but it is associated with immune disorders. ADAMTS18 is a member of the ADAMTS superfamily of extracellular proteinases. In this study, we showed that Adamts18 is highly expressed in submandibular salivary gland (SMG) during embryonic development and decreases but is retained in adult SMG tissue in mice. Adamts18 deficiency led to reduced cleft formation and epithelial branching in embryonic SMG before embryonic day 15.5 in mice. No significant histologic changes in the later stages of branching or the morphology of SMG were detected in Adamts18−/− mice. However, Adamts18 deficiency causes spontaneous SMG fibrogenesis and fibrosis in adult mice. At 8 wk of age, Adamts18−/− mice began to manifest the first signs of pathologic changes of mild fibrosis and CD11b+ cell infiltration in SMG tissues. At ≥8 mo, all male and female Adamts18−/− mice developed unilateral or bilateral SMG scleroma that is similar to patients with chronic sclerosing sialadenitis of the submandibular gland. Adamts18−/− mice also showed secretory dysfunction and severe dental caries. Histologically, SMG scleroma is characterized by progressive periductal fibrosis, acinar atrophy, irregular duct ectasis, and dense infiltration of IgG-positive plasma cells. A significant infiltration of CD4+ T lymphocytes and CD11b+ monocytes and macrophages was also detected in the SMG scleroma of Adamts18−/− mice. The levels of TGF-β1, IL-6, and IL-33 were significantly increased in Adamts18−/− SMGs, which induces chronic inflammation and myofibroblast activation, ultimately leading to fibrosis. This study indicates that Adamts18 regulates the early branching morphogenesis of embryonic SMG and plays a role in protecting from spontaneous SMG fibrogenesis via modulating local inflammation, autoimmune reaction, and myofibroblast activation in adult mice.

With the advances of the Belt and Road Initiative (BRI), the geo-economic interactions between China and countries along the 21st-Century Maritime Silk Road counties (MSRCs) continue to increase, and analyzing the geo-economic relations between China and the MSRCs is extremely important for a scientific understanding of bilateral geo-economic cooperation. Differently from the traditional logic of geo-economic competition and cooperation, we constructed a new framework based on the dominant factors of geo-economic relations and used an extreme random forest regression model to classify the geo-economic relation types between China and MSRCs from 2006 to 2017. The results show that the unbalanced development of investment and trade between China and MSRCs hindered the enhancement of the intensity of bilateral geo-economic linkage from 2006 to 2017. The “Matthew effect” of China’s geo-economic flow linkage with MSRCs is significant. There are obvious differences in the dominant factors affecting the types of geo-economic relations between China and MSRCs, and the distribution of the importance of the indices of the types of geo-economic relations in each country is disordered. Geopolitics, markets, and resources have played important roles in the geo-economic linkages between China and MSRCs. There are five types of geo-economic relations between China and the MSRCs: market-oriented type, resource-oriented type, market-resource-oriented type, market-geopolitics-oriented type, and resource-geopolitics-oriented type, of which the market-oriented type is the most important type of geo-economic relations. In the future, China should focus on regional powers along the Maritime Silk Road for bilateral geo-economic cooperation, actively promote the balanced development of bilateral geo-economic elements flows, strengthen geopolitical cooperation with MSRCs, and formulate cooperation plans according to the types of geo-economic relations.

Pyemotes zhonghuajia (Acari: Pyemotidae) has paralyzing and lethal effects on a variety of important agricultural and forestry pests in Lepidoptera, Coleoptera, and Hymenoptera pests. A single P. zhonghuajia mite can paralyze and kill a host 680,000 times its own weight within 3 hours. However, although it is an efficient and broad-spectrum natural enemy that is unique to China, its mitochondrial genome is still unknown. In this study, the complete mitochondrial genome of P. zhonghuajia was sequenced and annotated. It consists of two circular genomes. The complete mitogenome of P. zhonghuajia is 14,772 bp in length and contains 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and two control regions. The phylogenetic tree of 29 species suggests that P. zhonghuajia belongs to Eleutherengona.

Neotoxoptera formosana (Takahashi), the onion aphid, is an oligophagous pest that mainly feeds on plants from the Allium genus. It sucks nutrients from the plants and indirectly acts as a vector for plant viruses. This aphid causes severe economic losses to Allium tuberosum agriculture in China. To better understand the host plant specificity of N. formosana on Allium plants and provide essential information for the control of this pest, we generated the entire genome using Pacific Biosciences long-read sequencing and Hi-C data. Six chromosomes were assembled to give a final size of 372.470 Mb, with an N50 scaffold of 66.911 Mb. The final draft genome assembly, from 192 Gb of raw data, was approximately 371.791 Mb in size, with an N50 contig of 24.99 Kb and an N50 scaffold of 2.637 Mb. The average GC content was 30.96%. We identified 73 Mb (31.22%) of repetitive sequences, 14,175 protein-coding genes, and 719 noncoding RNAs. The phylogenetic analysis showed that N. formosana and Pentalonia nigronervosa are sister groups. We found significantly expanded gene families that were involved in the THAP domain, the DDE superfamily endonuclease, zinc finger, immunity (ankyrin repeats), digestive enzyme (serine carboxypeptidase) and chemosensory receptor. This genome assembly could provide a solid foundation for future studies on the host specificity of N. formosana and pesticide-resistant aphid management.

Resting state electromagnetic recordings have been analyzed in epilepsy patients aiding presurgical evaluation. However, it has been rarely explored how pathological networks can be separated and thus used for epileptogenic focus localization purpose. We proposed here a resting state EEG/MEG analysis framework, to disentangle brain functional networks represented by electrophysiological oscillations. Firstly, by using an Embedded Hidden Markov Model (EHMM), we constructed a state space for resting state recordings consisting of brain states with different spatiotemporal patterns. After that, functional connectivity analysis along with graph theory were applied on the extracted brain states to quantify the network features of the extracted brain states, and we determine the source location of pathological states based on these features. The EHMM model was rigorously evaluated using computer simulations. Our simulation results revealed the proposed framework can extract brain states with high accuracy regarding both spatial and temporal profiles. We than validated the entire framework as compared with clinical ground truth in 10 patients with drug-resistant focal epilepsy who underwent MEG recordings. We segmented the resting state MEG recordings into a few brain states with diverse connectivity patterns and extracted pathological brain states by applying graph theory on the constructed functional networks. We showed reasonable localization results using the extracted pathological brain states in 6/10 patients, as compared to the invasive clinical findings. The framework can serve as an objective tool in extracting brain functional networks from noninvasive resting state electromagnetic recordings. It promises to aid presurgical evaluation guiding intracranial EEG electrodes implantation.

The luminescence of traditional phosphorescence-based hypoxia probes is limited to the visible and first near-infrared wavelength regions (1000 nm), which has defects of higher light scattering and lower penetration depth in contrast with the second near-infrared wavelength window (NIR-II, 1000-1700 nm) for optical bioimaging. Herein, 5,15-bis(2,6-bis(dodecyloxy)phenyl)-porphyrin platinum(II) (PpyPt) with J-aggregation induced NIR-II phosphorescence is reported. J-aggregates of PpyPt are confirmed by the X-ray diffraction data in the crystalline state. Moreover, the emission and excitation spectra of PpyPt in the solid states reveal NIR-II luminescence feature of PpyPt in J-aggregates. The prescribed-time containment control problem for second-order multi-agent systems with multiple leaders is studied in this paper. The control objective is that all the followers are able to enter the convex hull formed by leaders at any preassigned time. Suppose that only a few followers can obtain the information of leaders, we design the distributed observer to estimate the information of the convex hull spanned by leaders firstly. Furthermore, the prescribed-time containment controller is designed with the estimation of the distributed observer. Based on a nonlinear scaling function, the tracking process is divided into four stages. The results imply that the tracking error between the followers and the convex hull could converge to zero within prescribed time. Finally, two numerical examples illustrate the effectiveness of the proposed algorithm.

We report a facile one-pot solvothermal way to prepare two-dimensional Ni-based metal–organic framework microsheets (Ni-MOFms) using only Ni precursor and ligand without any surfactant. The Ni-MOFms exhibit good specific capacities (91.4 and 60.0 C g−1 at 2 and 10 A g−1, respectively) and long-term stability in 5000 cycles when used for a supercapacitor electrode.

Biotoxins present in foods represent a major source of contamination. Although different analytical methods have been developed, a new detection method for identification and quantification of these toxins is necessary for extending their application sphere. Therefore, in this study, an enzyme-controlled citrate-iron(III) chelating system was used to build a novel colorimetric immunoassay sensing platform for detection of ochratoxin A (OTA). The experimental signal was derived from the reaction of iron(III) and 3,3′,5,5′-tetramethylbenzidine (TMB). However, the addition of citrate could combine with iron(III) rapidly, thereby hindering the reaction of iron(III) and TMB for production of the signal (i.e., colorless to blue). Additionally, hydrogen peroxide (H2O2), which could be produced from glucose in the presence of glucose oxidase (GOx), could affect the binding capacity of citrate and iron(III), thereby releasing iron(III) to react with TMB. That is to say, different amounts of GOx could regulate the intensity of the signal. Based on these findings, magnetic bead (MB)-based OTA competed with dissociative OTA (added) for glucose oxidase (GOx)-labelled anti-OTA. As the dissociative OTA concentration increased, the amount of GOx-labelled anti-OTA on MB conveniently reduced and the absorbance decreased. The method exhibited a good linear relationship as the concentration of the target (OTA) increased from 0.005 to 5 ng mL−1; the detection limit was 4.2 pg mL−1, as estimated based on the 3Sblank level under optimal conditions. The specificity and feasibility for a real sample of the colorimetric immunoassay was acceptable, respectively. The current results provided important insights into the development of citrate-iron(III) chelating system-based biological detection methods and colorimetric immunoassays.

Catalyst wettability regulation has emerged as an attractive approach for high catalytic performance for the past few years. By introducing appropriate wettability, the molecule diffusion of reactants and products can be enhanced, leading to high activity. Besides this, undesired molecules are isolated for high selectivity of target products and long-term stability of catalyst. Herein, we summarize wettability-induced high-performance heterogeneous thermocatalysis in recent years, including hydrophilicity, hydrophobicity, hybrid hydrophilicity-hydrophobicity, amphiphilicity, and superaerophilicity. Relevant reactions are further classified and described according to the reason for the performance improvement. It should be pointed out that studies of utilizing superaerophilicity to improve heterogeneous thermocatalytic performance have been included for the first time, so this is a comparatively comprehensive review in this field as yet.

Stereotactic-electroencephalography (SEEG) is a common neurosurgical method to localize epileptogenic zone in drug resistant epilepsy patients and inform treatment recommendations. In the current clinical practice, localization of epileptogenic zone typically requires prolonged recordings to capture seizure, which may take days to weeks. Although epilepsy surgery has been proven to be effective in general, the percentage of unsatisfactory seizure outcomes is still concerning. We developed a method to identify the seizure onset zone (SOZ) and predict seizure outcome using short-time resting-state SEEG data. In a cohort of 43 drug resistant epilepsy patients, we estimated the information flow via directional connectivity and inferred the excitation-inhibition ratio from the 1/f power slope. We hypothesized that the antagonism of information flow at multiple frequencies between SOZ and non-SOZ underlying the relatively stable epilepsy resting state could be related to the disrupted excitation-inhibition balance. We found higher excitability in non-SOZ regions compared to the SOZ, with dominant information flow from non-SOZ to SOZ regions, probably reflecting inhibitory input from non-SOZ to prevent seizure initiation. Greater differences in information flow between SOZ and non-SOZ regions were associated with favorable seizure outcome. By integrating a balanced random forest model with resting-state connectivity, our method localized the SOZ with an accuracy of 85% and predicted the seizure outcome with an accuracy of 77% using clinically determined SOZ. Overall, our study suggests that brief resting-state SEEG data can significantly facilitate the identification of SOZ and may eventually predict seizure outcomes without requiring long-term ictal recordings.

Publisher Copyright: IEEE In this letter, indium phosphide (InP) nanowires (NWs) are fabricated by Au-nanoparticle assisted vapor-liquid-solid method and applied as a saturable absorber (SA) for continuous-wave (CW) mode-locked femtosecond Yb:KYW bulk laser. I-scan method was used to characterize the saturable absorption properties of the prepared InP NWs SA. Pulses as short as 394 fs with the repetition rate of 41.5 MHz and maximum average output power of 315 mW are achieved. To the best of our knowledge, this is the first demonstration of InP NWs working as SA for the fs-pulse generation in the solid-state bulk laser. The results indicate that InP NWs are a promising SA candidate for applications in ultrafast photonic devices.

We report herein a versatile new approach for improving the photodegradation performance of UiO-66 by introducing 2-methylimidazole through a straightforward in situ one-pot solvothermal method. The resultant complex UiO-66-M-0.7 has demonstrated a reaction rate constant of 0.0183 min−1 in the photodegradation of rhodamine B under visible light, which is 69.4% higher than that with unmodulated UiO-66. Further investigation has confirmed that the improved activity can be attributed to enhanced adsorption of the dye by the photocatalyst during the adsorption–desorption equilibrium. The main active species involved in the photodegradation process have been identified as h+ and O2−·. In addition, the complex has shown ideal stability in recycling tests and excellent activity in the degradation of methylene blue. This study provides a novel perspective for the future design of various photocatalysts with superior performance. 2-Methylimidazole-modulated UiO-66 exhibits increased catalytic performance in the photodegradation of rhodamine B (RhB) under visible light because of enhanced adsorption of RhB on the catalyst during the adsorption-desorption equilibrium.

Background and aim Circular RNAs (circRNAs) have been highlighted to exert essential biological functions in papillary thyroid cancer (PTC). The purpose of this study was explore diagnostic utility of circRNAs in PTC patients. Patients and methods The distinctive expression profile of serum circRNAs was determined by individual quantitative real-time PCR (qRT-PCR) in two independent cohorts of 113 PTC patients, 80 thyroid nodules, and 111 healthy controls (HCs). A combination of circRNAs (circRNA-based combination index) was constructed by logistic regression. Results Individual qRT-PCR identification showed that two circRNAs (circRAPGEF5 and hsa_circ_0058124) were significantly up-regulated in PTC patients compared with HCs and thyroid nodules. Receiver-operating characteristic (ROC) curve analysis suggested that a combination of circRNAs was superior to individual circRNA in distinguishing PTC patients from HCs and thyroid nodules with area under ROC curve of more than 0.80. Furthermore, the combination of circRNAs increased significantly after systematic treatment, suggesting that it could monitor PTC dynamics. Additionally, the combination of circRNAs was independently correlated with PTC presence. Conclusion The combination of these altered circRNAs was correlated with PTC and may serve as a novel diagnostic approach.

High-grade sarcomas are metastatic and pose a serious threat to patient survival. Undifferentiated pleomorphic sarcoma (UPS) is a particularly dangerous and relatively common sarcoma subtype diagnosed in adults. UPS contains large quantities of extracellular matrix (ECM) including hyaluronic acid (HA), which is linked to metastatic potential. Consistent with these observations, expression of the HA receptor, hyaluronan-mediated motility receptor (HMMR/RHAMM), is tightly controlled in normal tissues and upregulated in UPS. Moreover, HMMR expression correlates with poor clinical outcome in these patients. Deregulation of the tumor-suppressive Hippo pathway is also linked to poor outcome in these patients. YAP1, the transcriptional regulator and central effector of Hippo pathway, is aberrantly stabilized in UPS and was recently shown to control RHAMM expression in breast cancer cells. Interestingly, both YAP1 and RHAMM are linked to TGFβ signaling. Therefore, we investigated crosstalk between YAP1 and TGFβ resulting in enhanced RHAMM-mediated cell migration and invasion. We observed that HMMR expression is under the control of both YAP1 and TGFβ and can be effectively targeted with small-molecule approaches that inhibit these pathways. Furthermore, we found that RHAMM expression promotes tumor cell proliferation and migration/invasion. To test these observations in a robust and quantifiable in vivo system, we developed a zebrafish xenograft assay of metastasis, which is complimentary to our murine studies. Importantly, pharmacologic inhibition of the TGFβ–YAP1–RHAMM axis prevents vascular migration of tumor cells to distant sites. Implications: These studies reveal key metastatic signaling mechanisms and highlight potential approaches to prevent metastatic dissemination in UPS.YAP1 and TGFβ cooperatively enhance proliferation and migration/invasion of UPS and fibrosarcomas.

2,4,6-Trinitrophenol (TNP) is widely used in our daily life; however, excessive use of TNP can lead to a large number of diseases. Therefore, it is necessary to find an effective method to detect TNP. Herein, the rapid fluorescence quenching by TNP was developed for the fluorometric determination of TNP in aqueous medium based on the internal filter effect. Nitrogen-sulfur-codoped carbon nanoparticles (N,S-CNPs), synthesized by a one-pot solvothermal method with the precursors of L-cysteine and citric acid, were applied for the determination of TNP as a fluorescent probe. The excitation peak center of N,S-CNPs and the emission peak center are 340 nm and 423 nm, respectively. The probe can be used in a variety of conditions to detect TNP due to its relatively stable properties. Meanwhile, it has a fast response time ( 1 min), wide linear response range (0.1-40 μM), and low detection limit (43.0 nM). This probe still has excellent selectivity and high sensitivity. The method was also used to detect standard water samples with a satisfactory recovery rate, and it will be used in the application of pollutants and clinical diseases. Graphical abstract.

Two-dimensional Bi2O2Se has drawn a lot of attention recently, due to its ultrahigh mobility and excellent performance in electronics and optoelectronics. However, the facile synthesis of two-dimensional Bi2O2Se nanocrystals is urgently required for their fundamental study and practical applications. In this work, we develop a controllable synthetic route to ultrathin Bi2O2Se nanosheets through the assistance of lithium nitrate (LiNO3) in a hydrothermal process. TEM, AFM, XRD, XPS and Raman demonstrate the successful synthesis of ultrathin Bi2O2Se nanosheets with a tetragonal phase. With Li+ adsorbing on {001} facets to inhibit crystal growth along the [001] direction, the lateral size of the Bi2O2Se nanosheets can be controlled through the amount of LiNO3 in the system. Photoelectrochemical tests reveal the good performance of the ultrathin Bi2O2Se nanosheets as a photoanode material in neutral solution. Detailed analysis of electrochemical impedance spectra and specific surface areas reveals that a high surface area and low recombination rate are responsible for the good photoelectrochemical activity of the larger Bi2O2Se nanosheets. Besides, the stability test shows negligible reduction of current density after 10 hours, and the TEM test demonstrates that the structure of the Bi2O2Se nanosheets is well preserved after long-term electrolysis, revealing great stability of the Bi2O2Se nanosheets as a photoanode material. This work indicates that two-dimensional Bi2O2Se could be a competitive candidate for the future energy systems.

Many kinds of nanoparticles and organic dyes as fluorescent probes have been used in the stimulated emission depletion (STED) nanoscopy. Due to high toxicity, photobleaching and non-water solubility, these fluorescent probes are hard to apply in living cell imaging. Here, we report a new fluorescence carbon dots (FNCDs) with high photoluminescence quantum yield (56%), low toxicity, anti-photobleaching and good water-solubility that suitable for live-cell imaging can be obtained by doping fluorine element. Moreover, the FNCDs can stain the nucleolus and tunneling nanotubes (TNTs) in the living cell. More importantly, for STED nanoscopy imaging, the FNCDs effectively depleted background signals and improved imaging resolution. Furthermore, the lateral resolution of single FNCDs size under the STED nanoscopy is up to 22.1 nm for FNCDs deposited on a glass slide was obtained. And because of their good water dispersibility, the higher resolution of single FNCDs size in the nucleolus of a living cell can be up to 19.7 nm. After the image optimization steps, the fine fluorescence images of TNTs diameter with ca. 75 nm resolution is obtained living cell, yielding a threefold enhancement compared with that in confocal imaging. Additionally, the FNCDs show excellent photobleaching resistance after 1,000 scan cycles in the STED model. All results show that FNCDs have significant potential for application in STED nanoscopy.
