Abstract

The present study endeavored to design and develop a self-nanoemulsifying drug delivery system to improve the solubility and dermatological absorption of curcumin and naringin. Curcumin and naringin-loaded self-nanoemulsifying drug delivery system formulations were developed using aqueous phase titration. Phase diagrams were used to pinpoint the self-nanoemulsifying drug delivery system zones. Tween 80 and Labrasol (surfactants), Transcutol (cosurfactant), and cinnamon oil were chosen from a large pool of surfactants, cosurfactants, and oils based on their solubility and greatest nano-emulsion region. Fourier transform infrared spectroscopy, zeta sizer, and atomic force microscopy were used to characterize the optimized formulations and test for dilution and thermodynamic stability. The optimized curcumin-naringin-self-nanoemulsifying drug delivery system demonstrated the following characteristics: polydispersity index (0.412 ± 0.03), % transmittance (97%), particle size (212.5 ± 05 nm), zeta potential (− 25.7 ± 1.80 mV) and having a smooth and spherical droplet shape, as shown by atomic force microscopy. The ability of their combined formulation to cure wounds was tested in comparison to pure curcumin suspension, empty self-nanoemulsifying drug delivery system, and standard fusidic acid. Upon topical administration, the optimized curcumin-naringin-self-nanoemulsifying drug delivery system demonstrated significant wound healing activity in comparison with a pure curcumin suspension, empty self-nanoemulsifying drug delivery system, and standard fusidic acid. Based upon this result, we assume that skin penetration was increased by using the optimized curcumin-naringin-self-nanoemulsifying drug delivery system with enhanced solubility. Abstract

Mercury (Hg2+) as environmental pollutant is a widespread concern due to its cytotoxic effect in humans and animals and needs to be monitored through cost effective methods based on naked-eye detection. Herein, in this study, we extracted and isolated the limonin (LMN) via a facile procedure and then employed it as capping agent for the synthesis of silver nanoparticles (LMN-AgNPs) as a first report. LMN-AgNPs were characterized through Field Emission Scanning Electron Microscopy (FESEM), Atomic Force Microscopy (AFM), Dynamic Light Scattering (DLS), Zeta Potential Analyzer (ZPA) and Fourier Transform Infrared (FTIR) spectroscopy. As-formed LMN-AgNPs were recognized as extremely selective and highly sensitive colorimetric sensor for Hg2+ with potential analytical application. The developed sensor showed an outstanding linear correlation with the concentration of Hg2+ in the range of 0.002 − 55 µM via a color change from deep yellow to transparent showing hypsochromic-hypochromic shift with the limit of detection (LOD) and the limit of quantification (LOQ) as low as 0.21 nM and 0.7 nM respectively. The sensor was further allied with smartphone for immediate and on-site quantification of Hg2+. The LOD and LOQ of 0.42 µM and 1.4 µM was true for smartphone based sensing in the range of 7.5–55 µM Hg2+. The detection of Hg2+ was not disrupted by the presence of other metals in either of mentioned cases. The practical applicability of the proposed Hg2+ sensor was tested using spectrophotometric and smartphone based approaches in human serum and urine as well as in tap water samples with acceptable ranges of recovery. Abstract

The present study explores the potential of Valeriana jatamansi shoot extract for Ag-metal bio-reduction and its antimicrobial activity. Among the different ratios of AgNO3 and extract tested, 1:5 (1 mL AgNO3 and 5 mL extract) gave maximum SPR peak at 411.0 nm during UV-Vis spectrophotometric analysis, indicating the synthesis of maximum amount of AgNPs in solution. XRD analysis reported the crystalline nature of AgNPs with 13.32 nm nanocrystallite size. FTIR studies suggested the involvement of carboxylic acid (–[C–O–O–H]) and methane (–CH–) functional groups of different compounds in AgNPs reduction and fabrication. Average size of synthesized uniform shaped nanospheres was 32 nm by SEM image analysis. The produced AgNPs (1.5 mg/disc) showed growth inhibition of 71.46, 65.97, 61.5, 55.32, and 54.83% against Pseudomonas aeruginosa, Escherichia coli, Candida albicans, Xanthomonas campestris, and Staphylococcus aureus. While the least growth inhibition of 48.55% was recorded for Klebsiella pneumonia, suggesting it as the least-susceptible microbe among all the tested microbial species. P. aeruginosa was found to be most sensitive of all tested microbes, while E. coli, C. albicans, and X. campestris reported moderate susceptibility to AgNPs.
