Abstract

This chapter deals with the advancement of nanomaterial-based sensors in the last 10 years. The use of different types of nanomaterials, including graphene, carbon nanotubes, and metallic nanoparticles, was described, highlighting that graphene represents a rising star in the plethora of nanomaterials. Among the different transducers, the chapter describes the electrochemical and optical (bio)sensors, being the most promising devices. The use of materials at the nanodimension scale provides several improvements in terms of analytical features including sensitivity, rapidity of response, selectivity, and robustness, demonstrating the huge advantage of using the nanomaterials over the micromaterials in the development of smart and high-performant analytical tools.

Abstract

Biosensors for Sustainable Food - New Opportunities and Technical Challenges addresses the challenges associated with sustaining the globally increasing demand for food that has been forecast for the next centuries and the immediate need for the food production system to adopt sustainable practices to protect the environment and human health. It provides a comprehensive overview of established, cutting-edge, and future trends in biosensor technology and its application in the agrifood sector. In particular, different biosensing advances are covered, outlining the newest research efforts in the cross-disciplines of chemistry, biology, and materials science with biosensing research, in order to develop novel detection principles, sensing mechanisms, and device engineering methods. Food production and consumption have a strong impact on the environment in terms of greenhouse gas emissions, water, and soil contamination, the reduction of arable land, water consumption, and many other factors, which in turn, negatively affect human health. These issues have consequences for economic development, too. To address these challenges, it is necessary for scientists with different expertise, policymakers, and economists work together to develop new smart technologies and introduce them to the market, along with adequate regulations. In this regard, a sustainable food production system can be thought of as a chain of procedures with a low impact on the environment that guarantees a secured supply of healthier and fortified food while supporting economic growth. Abstract

The growing spread of pathogens, caused by anthropogenic activities, pushes the interest of the scientific community towards developing biosensors with improved performance for rapid, simple, and on-site pathogen detection. In this study, we present and discuss a label-free gold nanorod (Au NR) array for the rapid detection of Escherichia colicells in water, resulting in an effective optical transducer, based on the phenomenon of localized surface plasmon resonance (LSPR). Au NRs with different aspect ratios are functionalized with a suitable antibody by an electrostatic-linking method, resulting in two different Au NR-based bioconjugates. We investigate the ability of the two bioconjugates to detect and spectroscopically recognize E. colicells dispersed in water by specific antigen–antibody interaction. The results allow selecting the Au NR morphology more suited for preparing the Au NR bioactive array on a glass substrate with excellent optical and morphological properties. The antibody-functionalized Au NR array can detect E. colicells with high sensitivity and a limit of detection of 8.4 CFU mL−1, resulting in an excellent label-free spectroscopic biosensor. In addition, the multicolor thermoplasmonic properties of the Au NR array, triggered by appropriate light sources, are suited to enable on-demand photothermal disinfection, thus providing an extraordinary capacity for the biosensor to be both disinfected and, more importantly, reutilized.

Abstract

The conformational changes of the Escherichia coli glutamine-binding protein (GlnBP) induced by GdnBP and the effect of the binding of glutamine (Gln) on these processes is studied. The use of the parametric presentation of fluorescence data showed that both GlnBP unfolding and GlnBP-Gn unfolding are three-step processes (N->I(sub 1)->I9sub 2)->U), though in the case of the GlnBP-Gln complex these stages essentially overlap.
