Abstract

We study the compression of bundles of aligned macroscopic fibers with intrinsic shape disorder, as found in human hair and in many other natural and man-made systems. We show by a combination of experiments, numerical simulations and theory how the statistical properties of the shapes of the fibers control the collective mechanical behaviour of the bundles. This work paves the way for designing aligned fibrous matter with pre-required properties from large numbers of individual strands of selected geometry and rigidity., Comment: 14 pages, 15 figures

Abstract

Belantamab mafodotin (BM) is an anti-BCMA antibody-drug conjugate (GSK2857916) that represents an alternative option in multiple myeloma. We sought to assess the efficacy and safety of BM in a real-world setting in patients who benefited from an early access program. We conducted an observational, retrospective, multicenter study. Eligibility criteria were treatment of relapsed or refractory multiple myeloma (RRMM) in monotherapy in adult patients who have received at least three lines of therapy previously, including at least one immunomodulatory agent (IMiD), a proteasome inhibitor (PI) and an anti-CD38 monoclonal antibody, and whose disease progressed during the last treatment period. The primary endpoint of the study is to assess the overall survival (OS). Between November 2019 and December 2020, 106 patients were treated with BM; 97 were eligible for the efficacy evaluation and 104 for safety. The median age was 66 (range, 37–82) years. High-risk cytogenetics were identified in 40.9% of patients. Fifty-five (56.7%) patients were triple-class refractory and 11 (11.3%) were penta-class refractory. The median number of prior lines of treatment was five (range, 3–12). The median number of BM cycles administered was three (range, 1–22). The overall response rate at best response was 38.1% (37/97). The median OS was 9.3 months (95% confidence interval [CI]: 5.9-15.3), and median progression-free survival was 3.5 months (95% CI: 1.9-4.7). The median duration of response was 9 months (range, 4.65-10.4). Treatment was delayed for 55 (52.9%) patients including 36.5% for treatment-related toxicity. Ophthalmic adverse events, mainly grade ≤2, were the most common toxicity (48%). The occurrence of keratopathy was 37.5%. Overall, our data are concordant with the results from DREAMM-2 in terms of efficacy and safety on a non-biased population.

Abstract

Network reconnaissance and measurements play a central role in improving Internet security and are important for understanding the current deployments and trends. Such measurements often require coordination with the measured target. This limits the scalability and the coverage of the existing proposals. IP Identification (IPID) provides a side channel for remote measurements without requiring the targets to install agents or visit the measurement infrastructure. However, current IPID-based techniques have technical limitations due to their reliance on the idealistic assumption of stable IPID changes or prior knowledge, making them challenging to adopt for practical measurements. In this work, we aim to tackle the limitations of existing techniques by introducing a novel approach: predictive analysis of IPID counter behavior. This involves utilizing a machine learning (ML) model to understand the historical patterns of IPID counter changes and predict future IPID values. To validate our approach, we implement six ML models and evaluate them on realistic IPID data collected from 4,698 Internet sources. Our evaluations demonstrate that among the six models, the Gaussian Process (GP) model has superior accuracy in tracking and predicting IPID values. Using the GP-based predictive analysis, we implement a tool, called ZPredict, to infer various favorable information about target networks or servers. Our evaluation on a large dataset of public servers demonstrates its effectiveness in idle port scanning, measuring Russian censorship, and inferring Source Address Validation. 73. Carboniferous Barrovian to Permian Buchan‐type metamorphic cycles in the Mongolian Altai Zone: Implication for pressure cycles in accretionary orogens.

Abstract

In the Altai Accretionary Wedge, several periods of Barrovian‐ and Buchan‐type metamorphic cycles were dated from Ordovician to Permian. However, the timing and link between these cycles are not clear, and their causes are debated. In order to contribute to the understanding of Barrovian‐ to Buchan‐type evolution of the accretionary wedges, we studied an area composed of three parallel belts in the easternmost extremity of the Hovd domain located in Mongolian Altai Zone: garnet gneiss in the north, garnet–staurolite–kyanite schist overprinted by ±sillimanite±cordierite±andalusite‐bearing assemblages in the centre and garnet–sillimanite gneiss in the south. Petrography, garnet zoning and thermodynamic modelling indicate that the garnet gneiss from the northern belt records burial from ~510°C and ~3–4 kbar to ~600°C and ~5 kbar, followed by heating to ~660°C and decompression to ~4.5 kbar. The garnet–staurolite–kyanite schist from the central belt records burial from ~550°C and ~3–4.5 kbar to ~640–680°C and ~7 kbar, followed by decompression to the sillimanite stability field at ~650°C and ~6 kbar. Crystallization of cordierite, andalusite, late muscovite and chlorite in some samples indicates cooling on decompression to ~540°C and ~3.5 kbar. In the southern gneiss belt, the garnet–sillimanite gneiss with almost unzoned garnet suggests re‐equilibration at ~6 kbar and ~710°C. In situ U–Pb monazite and xenotime dating carried out inclusions in porphyroblasts and matrix grains revealed Carboniferous and Permian ages. The monazite and xenotime from gneisses of the northern and southern belts record Carboniferous and Permian ages, which are interpreted as Carboniferous crystallization at c. 347 Ma associated with metamorphic peak, followed by Permian (re)crystallization at c. 300 and 283 Ma. In the central belt, rare Carboniferous xenotime grains in a garnet–staurolite–kyanite–andalusite–muscovite schist indicate a possible Carboniferous age of the prograde metamorphism. Predominant ages between c. 280 and 260 Ma recorded by monazite are interpreted as a result of complete recrystallization during an LP metamorphic overprint. The Carboniferous ages from the gneisses can be interpreted as constraining the timing of the exhumation of deep crustal rocks to shallow crustal levels. This event corresponds to the formation of crustal‐scale migmatite‐magmatite domes in the Mongolian Altai Zone. The prograde Barrovian assemblages in the central schist belt are interpreted as having formed contemporaneously during burial in a synform between the migmatite‐magmatite domes. The Permian ages reflect LP–HT metamorphism, best recorded by the Buchan‐type assemblages in the central schist belt, and are related to massive heat flux from tectonically mobile deep partially molten crust. Correlation with similar Barrovian‐ and Buchan‐type episodes from the Chinese Altai Zone indicates multiple compressional and extensional events in the upper plate accretionary wedge, probably related to retreating and advancing modes of the subduction zone. Abstract

Background: IMAGE is a retrospective cohort study of patients enrolled in early access programs (EAPs) in France with relapsed/refractory multiple myeloma (RRMM) receiving isatuximab with pomalidomide and dexamethasone (Isa‐Pd). Methods: Patients aged ≥18 years with RRMM who received ≥1 dose of Isa under the EAPs between July 29, 2019 and August 30, 2020 were included. Effectiveness endpoints included progression‐free survival (PFS) and response rates. Verbatim terms for adverse events (AEs) were coded using the Medical Dictionary for Regulatory Activities and not graded for severity. Results: A total of 294 and 299 patients were included in the effectiveness and safety populations, respectively. IMAGE included patients who received one prior line of treatment (10.2%) and were daratumumab‐refractory (19.1%). At median follow‐up of 14.2 months, median PFS in the effectiveness population was 12.4 months (95% CI 9.0–15.0). Overall response and very good partial response rates were 46.3% and 27.9%, respectively. Subgroup analyses reflected similar results. In the safety population, 26.4% of patients reported at least one AE; the most common any‐grade AE was neutropenia (9.4%). Conclusion: IMAGE demonstrated Isa‐Pd had meaningful effectiveness in median PFS and depth of response and no new safety signals in a real‐world context, consistent with clinical trial results.
