Genetic analyses of speciation have focused nearly exclusively on retrospective analyses of reproductive isolation between highly divergent species. Yet, a full understanding of the speciation process must encompass analysis of the consequences of genomic divergence in young lineages still capable of exchanging genes under natural conditions. The accumulation of conditionally neutral genetic variation may lead to the evolution of divergent gene networks. In a hybrid background, such mutations may no longer compensate one another, resulting in the appearance of selectively disadvantageous traits, including disruption of gene expression regulation. Here, we documented genome-wide patterns of gene expression divergence between young lineages of normal and dwarf lake whitefish and their backcross hybrids for which strong, yet incomplete post-zygotic isolation barriers exist. A significant proportion (33%) of backcross hybrids showed developmental abnormalities not seen in parental forms and eventually leading to death. Although the transcriptome of parental forms was nearly identical during embryonic development, suggesting a role for stabilizing selection, all hybrids displayed strongly divergent patterns of gene expression. By comparing healthy, surviving hybrids against moribund ones showing abnormal development, we observed that over 2000 genes were misregulated in these abnormal embryos. In particular, misregulation was significantly biased toward essential developmental genes, which were severely underexpressed. Furthermore, genes previously documented to be highly transgressive (exaggerated inter-individual variance) were almost invariably underexpressed in hybrids. Our results thus clearly showed a transcriptome-wide signature of hybrid breakdown in young, incipient species and demonstrated a persuasive link between misexpression of essential developmental genes and post-zygotic isolation.

We present here phenotypic and genetic parameters for the major quality and production traits of farmed European whitefish. A total of 70 families were produced by mating each of 45 sires to an average of 1.6 dams and each of the 52 dams to an average of 1.3 sires. A total of 2,100 individuals were recorded for survival, and 507 individuals for growth and quality-related traits. The 4 major results were as follows: first, all traits exhibited nonzero heritabilities except for fillet gaping and fillet protein%. The heritabilities for the production traits were harvest weight (0.42 ± 0.10), gutted weight (0.40 ± 0.10), fillet weight (0.36 ± 0.09), maturity score (0.27 ± 0.11, on liability scale), survival (0.19 ± 0.05, on liability scale), carcass% (0.14 ± 0.07), and fillet% (0.11 ± 0.06). The heritabilities for the quality traits were condition factor (0.49 ± 0.10), fillet lipid% (0.37 ± 0.10), muscle texture (0.30 ± 0.09), Distell lipid reading (0.26 ± 0.09), fillet lightness (0.16 ± 0.07), fillet gaping (0.04 ± 0.06), and fillet protein% (0.04 ± 0.06). Second, the quality traits that were significantly genetically correlated with each other were all related to lipid deposition. Increasing fillet lipid% (an undesired change in whitefish) was genetically related to desired lighter fillet color [genetic correlation (r(G)) = 0.70 ± 0.22] and to undesired greater condition factor (0.39 ± 0.17). None of the other genetic correlations between condition factor, fillet lipid%, muscle texture, fillet lightness, fillet gaping, and fillet protein% were significant. Third, BW and gutted weight were genetically related to the quality traits that were genetically related to lipid deposition. Increasing harvest weight was genetically related to high fillet lipid% (r(G) = 0.59 ± 0.14), lighter fillet color (0.61 ± 0.25), and to greater condition factor (0.60 ± 0.12). All other genetic correlations of harvest weights with the quality traits were nonsignificant, indicating that rapid growth was not genetically related to gaping and softer flesh. Fourth, none of the genetic correlations of carcass%, fillet%, maturity, and survival with the quality traits were significant, implying weak genetic integration between the traits. Yet, marginally significant genetic correlations were found for fillet lipid% with maturity score (r(G) = -0.46 ± 0.24) and survival (0.36 ± 0.19). These results provide the genetic basis for assessing the potential to improve product quality via selective breeding.

As populations adapt to novel environments, divergent selection will promote heterogeneous genomic differentiation via reductions in gene flow for loci underlying adaptive traits. Using a data set of over 100 SNP markers, genome scans were performed to investigate the effect of natural selection maintaining differentiation in five lakes harbouring sympatric pairs of normal and dwarf lake whitefish (Coregonus clupeaformis). A variable proportion of SNPs (between 0% and 12%) was identified as outliers, which corroborated the predicted intensity of competitive interactions unique to each lake. Moreover, strong reduction in heterozygosity was typically observed for outlier loci in dwarf but not in normal whitefish, indicating that directional selection has been acting on standing genetic variation more intensively in dwarf whitefish. SNP associations in backcross hybrid progeny identified 16 genes exhibiting genotype-phenotype associations for four adaptive traits (growth, swimming activity, gill rakers and condition factor). However, neither simple relationship between elevated levels of genetic differentiation with adaptive phenotype nor conspicuous genetic signatures for parallelism at outlier loci were detected, which underscores the importance of independent evolution among lakes. The integration of phenotypic, transcriptomic and functional genomic information identified two candidate genes (sodium potassium ATPase and triosephosphate isomerase) involved in the recent ecological divergence of lake whitefish. Finally, the identification of several markers under divergent selection suggests that many genes, in an environment-specific manner, are recruited by selection and ultimately contributed to the repeated ecological speciation of a dwarf phenotype., (© 2010 Blackwell Publishing Ltd.)

There is some concern that incidental consumption of eggs cured with commercially available cures for the purpose of sport fishing causes mortality in juvenile salmon. We evaluated this by feeding juvenile spring Chinook (Oncorhynchus tshawytscha) and steelhead (O. mykiss) with eggs cured with one of five commercially available cures. We observed significant levels of mortality in both pre-smolts and smolts. Depending on the experiment, 2, 3, or 4 of the cures were associated with mortality. Mortality tended to be higher in the smolts than in the parr, but there was no clear species effect. The majority of mortality occurred within the first 10 d of feeding. Removal of sodium sulfite from the cure significantly reduced the level of mortality. Soaking the eggs prior to feeding did not reduce mortality. We observed a clear relationship between the amount of cured egg consumed each day and the survival time. We conclude that consumption of eggs cured with sodium sulfite has the potential to cause mortality in juvenile steelhead and Chinook salmon in the wild.

Arctic cisco Coregonus autumnalis young-of-year (YOY) growth was used as a proxy to examine the long-term response of a high-latitude fish population to changing climate from 1978 to 2004. YOY growth increased over time (r² = 0·29) and was correlated with monthly averages of the Arctic oscillation index, air temperature, east wind speed, sea-ice concentration and river discharge with and without time lags. Overall, the most prevalent correlates to YOY growth were sea-ice concentration lagged 1 year (significant correlations in 7 months; r² = 0·14-0·31) and Mackenzie River discharge lagged 2 years (significant correlations in 8 months; r² = 0·13-0·50). The results suggest that decreased sea-ice concentrations and increased river discharge fuel primary production and that life cycles of prey species linking increased primary production to fish growth are responsible for the time lag. Oceanographic studies also suggest that sea ice concentration and fluvial inputs from the Mackenzie River are key factors influencing productivity in the Beaufort Sea. Future research should assess the possible mechanism relating sea ice concentration and river discharge to productivity at upper trophic levels., (Journal of Fish Biology © 2010 The Fisheries Society of the British Isles. River-dwelling fish, such as European graylings (Thymallus thymallus), are susceptible to changes in climate because they can often not avoid suboptimal temperatures, especially during early developmental stages. We analyzed data collected in a 62-year-long (1948-2009) population monitoring program. Male and female graylings were sampled about three times/week during the yearly spawning season in order to follow the development of the population. The occurrence of females bearing ripe eggs was used to approximate the timing of each spawning season. In the last years of the study, spawning season was more than 3 weeks earlier than in the first years. This shift was linked to increasing water temperatures as recorded over the last 39 years with a temperature logger at the spawning site. In early spring water temperatures rose more slowly than in later spring. Thus, embryos and larvae were exposed to increasingly colder water at a stage that is critical for sex determination and pathogen resistance in other salmonids. In summer, however, fry were exposed to increasingly warmer temperatures. The changes in water temperatures that we found embryos, larvae, and fry were exposed to could be contributing to the decline in abundance that has occurred over the last 30-40 years., (© 2010 Society for Conservation Biology.)

To validate age determination from scales in European grayling Thymallus thymallus, the scale-read age of fish was compared with the true age obtained by tag-recapture analysis. A total of 3997 individuals were tagged with visible implant tags and passive integrated transponder (PIT) tags in the River Wylye, south-west England during 1999-2007. Annual repeat surveys were undertaken and collected scales read without prior knowledge of tag-recapture age. Accuracy of fish ageing by scales was highest in 1 and 2 year-old fish but decreased in older fish. In later life stages (>4 years old), underestimation of age occurred and the error in reading scales rose to 51.9% in 5 year-old fish. Age assigned from scales underestimated the tag-recapture assigned age by as much as 3 years. This study suggests that use of scales is an appropriate method to age a short-lived population of T. thymallus inhabiting productive lotic systems. The underestimation of age in older fish, however, needs to be considered in the management of fish stocks because it may lead to undesirable exploitation of population.

Increasing research is highlighting the fact that streams provide crucial ecosystem services through the biogeochemical and ecological processes they sustain. Freshwater land-based salmonid farms commonly discharge their effluents into low order, headwater streams, partly due to the fact that adequate freshwater resources for production are commonly found in undisturbed areas. We review the effects of salmonid farm effluents on different biological components of stream ecosystems. Relevant considerations related to the temporal and spatial scales of effluent discharge and ecological effects are discussed. These highlight the need to characterize the patterns of stressor discharge when assessing environmental impacts and designing ecological effects studies. The potential role of multiple stressors in disrupting ecosystem structure and function is discussed with an emphasis on aquaculture veterinary medicines. Further research on the effects of veterinary medicines using relevant exposure scenarios would significantly contribute to our understanding of their impact in relation to other effluent stressors., (Copyright 2009 Elsevier Ltd. Decabromodiphenyl ether (BDE 209) is the main congener in the commonly used commercial flame retardant mixture, "deca-BDE". There is evidence showing that fish can debrominate BDE 209 into potentially more toxic congeners. The objective of this study was to evaluate BDE 209 uptake and its potential effects on juvenile lake whitefish (Coregonus clupeaformis). Lake whitefish were fed BDE 209 at four nominal concentrations (control, 0.1, 1, and 2 microg/g-diet) for 30 days. Livers and carcasses were analyzed for 11 polybrominated diphenyl ether (PBDE) congeners (BDE 47, 99, 100, 153, 154, 196, 197, 206, 207, 208, and 209) and daily otolith increment width was measured as an estimate of growth before and after exposure. Four congeners (BDE 206, 207, 208, and 209) were detected in livers and carcasses. Hepatic BDE 209 concentrations in the 1 and 2 microg/g treatments were significantly higher than in the control group (1.25 and 5.80 nmol/g-lipid compared to 0.183 nmol/g-lipid). The concentration of BDE 209 detected in the tissues of the control group resulted from BDE 209 in the base diets. Concentrations of all congeners from the 1 and 2 microg/g groups were higher in livers than carcasses, indicating the liver was the primary organ of BDE 209 accumulation. Compared to the fraction in diets, the molar fraction of BDE 209 was lower in livers and carcasses, whereas the fractions of BDE 206, 207, and 208 were higher. These different distributions of PBDE congeners resulted from differential adsorption and metabolism. One congener, BDE 206, could be a major metabolite from BDE 209 debromination. Otolith increment widths were narrower in fish from the highest diet concentration administered, suggesting BDE 209 may have affected growth rates. In conclusion, this in vivo study with lake whitefish showed that BDE 209 was debrominated into lower PBDE congeners and that exposure to 2 microg/g may have affected fish growth.

The growth, and dispersal of stocked European grayling Thymallus thymallus, reared in a hatchery (fed dry food pellets) or in a pond (fed natural food), compared with their wild conspecifics was assessed from the recapture of individually tagged fish 168 days after their release into the Blanice River, Czech Republic. Recapture rates and site fidelity were higher for wild T. thymallus than for artificially reared fish. Specific growth rate and upstream or downstream dispersal did not significantly differ between any of the groups of fish. An influence of rearing conditions (pond v. hatchery) on the overall performance of stocked fish was not demonstrated. Initially, lower condition factors of reared T. thymallus were equal to wild fish after recapture, suggesting adaptation of artificially reared fish that remained in the sections studied.

Fish reproduction is likely to be affected by increasing water temperatures arising from climate change. Normal changes in environmental temperature have the capacity to affect endocrine function and either advance or retard gametogenesis and maturation, but above-normal temperatures have deleterious effects on reproductive processes. In Atlantic salmon Salmo salar, exposure to elevated temperature during gametogenesis impairs both gonadal steroid synthesis and hepatic vitellogenin production, alters hepatic oestrogen receptor dynamics and ultimately results in reduced maternal investment and gamete viability. Exposure to high temperature during the maturational phase impairs gonadal steroidogenesis, delaying or inhibiting the preovulatory shift from androgen to maturation-inducing steroid production. There are also deleterious effects on reproductive development of female broodstock of rainbow trout Oncorhynchus mykiss and Arctic charr Salvelinus alpinus when they are exposed to elevated temperature. Less is known about temperature effects on male fishes but inhibition of spermiation has been observed in S. salar and O. mykiss. Among wild stocks, the response to elevated temperature will involve behavioural thermoregulation with consequent change in geographical ranges and the possibility of local extinctions in some regions. For domesticated stocks, containment in the culture environment precludes behavioural thermoregulation and aquaculturists will be required to develop adaptive strategies in order to maintain productivity. The most direct strategy is to manage the thermal environment using one or more of a range of developing aquaculture technologies. Alternatively, there is potential to mitigate the effects of elevated temperature on reproductive processes through endocrine therapies designed to augment or restore natural endocrine function. Studies largely on S. salar have demonstrated the capacity for synthetic luteinizing hormone-releasing hormone to offset the inhibitory effects of elevated temperature on maturational events in both sexes, but the potential for hormone therapy to provide protection during gametogenesis is still largely unexplored.

Compensatory growth (CG) is a key issue in work aiming at a full understanding of the adaptive significance of growth plasticity and its carryover effects on life-history. The number of studies addressing evolutionary explanations for CG has increased rapidly during the last few years, but there has not been a parallel gain in our understanding of the methodological difficulties associated with the analysis of CG. We point out two features of growth that can have serious consequences for detecting CG: (1) size dependence of growth rates, which causes nonlinearity of growth trajectories, and; (2) temporal overlapping of structural growth and replenishment of energy reserves after a period of famine. We show that the currently used methods can be prone to spurious detection of CG (Type I error) under conditions of nonlinear growth, and therefore lead to the accumulation of a significant amount of false "empirical support." True and simulated growth data provided consistent results suggesting that a substantial fraction of the existing evidence for CG may be spurious. A small curvature in the growth trajectory can lead to spurious "detection" of CG when control and manipulated trajectories are compared over the same time interval (the "simultaneous" approach). We present a novel, robust method (the "asynchronous" approach) based on the accurate selection of control trajectories and comparison of control and treatment growth rates at different times. This method enables a reliable test to be performed for compensation under asymptotic growth. While the general results of our simulations do not support the application of conventional methods to the general case of nonlinear growth trajectories under the simultaneous approach, simple methods may prove valid if the experimental design allows for asynchronous comparisons. We advocate an alternative approach to deal with "safe" detection of CG that overcomes the problems associated with the occurrence of nonlinear and asymptotic growth, and provide recommendations for improving CG study designs.

The development of isolation by distance (IBD) and isolation by time (IBT) was contrasted among demes of European grayling (Thymallus thymallus) that have diverged within the last 25 generations following colonization of a lake (Lesjaskogsvatnet). We find low but significant levels of genetic differentiation among spawning tributaries and a pattern of IBD among them. We do not, however, find evidence for IBT despite an up to four-week difference in spawning date between "warm/early" and "cold/late" spawning demes and differences in the incubation temperatures experienced by offspring. It appears that IBD has developed more rapidly than IBT in this system and that adaptive divergence has been initiated in the absence of IBT. Although analysis of selected loci could reveal reduced recombination in parts of the genome associated with temporal divergence, our analysis of neutral genetic data suggests that IBD is a more important isolating mechanism in the early stages of adaptive divergence in European grayling.

The hypothesis that diatom carbon (C) produced during the spring peak supported spring zooplankton production and, ultimately, the growth of Coregonus lavaretus early larval stages from March to May 2006 in Lake Annecy, France, was tested using gut content analyses and fatty acid biomarkers. Gut content results showed that C. lavaretus larvae from stages 1 to 4 preferentially fed on copepods with Daphnia sp. only a minor proportion of larval diet. The levels of diatom-marker fatty acids (C16:1n-7 and C20:5n-3) were high in Daphnia sp., but lower in both copepods and C. lavaretus larvae from stages 0 to 4. These results indicated that the spring diatom biomass was actually grazed by Daphnia sp., but, contrary to what was expected, the spring bloom was not the only C source supporting copepods secondary production and, consequently, the growth of C. lavaretus early larval stages. In contrast, levels of terrestrial fatty acid marker (C24:0) were low in Daphnia sp. but high in copepods and C. lavaretus larvae, indicating a significant contribution of terrestrial carbon to copepods and, ultimately, to the growth of C. lavaretus early larval stages.

Since 2000, a surprisingly high number of macroscopical gonad alterations has been reported in whitefish (Coregonus spp.) from Lake Thun, Switzerland. This unique phenomenon is still unexplained and has received much public attention. As one possible trigger for these effects, the presence of persistent, bioaccumulative and toxic compounds acting as endocrine disruptors in the lake has been discussed. In this study, concentrations of selected persistent organic pollutants were examined in two morphs of whitefish from Lake Thun and their link to the observed abnormalities was investigated. Analyzed compound classes included polychlorinated biphenyls, polychlorinated dibenzo-p-dioxins and dibenzofurans, polychlorinated naphthalenes, polybrominated diphenyl ethers and hexabromocyclododecanes. The target substances were identified in all samples and concentrations of the analyzed compounds were highly correlated among each other. These correlations show that the analyzed substances have the same distribution pattern throughout the lake and that uptake, accumulation and elimination processes are similar. Significant differences in contaminant levels within the samples existed between the two analyzed morphs of whitefish, most likely due to different age, food patterns and growth rate. No difference in contaminant levels was observed between fish with abnormal gonads and fish with normal gonads, suggesting no causal link between the investigated lipophilic organohalogen compounds present in fish and the observed gonad abnormalities in whitefish from Lake Thun. A comparison to existing data shows that concentrations in Lake Thun whitefish are at the lower bound of contaminant levels in whitefish from Swiss lakes or from European waters.

In this study, we demonstrate the power of applying complementary DNA (cDNA) microarray technology to identifying candidate loci that exhibit subtle differences in expression levels associated with a complex trait in natural populations of a nonmodel organism. Using a highly replicated experimental design involving 180 cDNA microarray experiments, we measured gene-expression levels from 1098 transcript probes in 90 individuals originating from six brown trout (Salmo trutta) and one Atlantic salmon (Salmo salar) population, which follow either a migratory or a sedentary life history. We identified several candidate genes associated with preparatory adaptations to different life histories in salmonids, including genes encoding for transaldolase 1, constitutive heat-shock protein HSC70-1 and endozepine. Some of these genes clustered into functional groups, providing insight into the physiological pathways potentially involved in the expression of life-history related phenotypic differences. Such differences included the down-regulation of genes involved in the respiratory system of future migratory individuals. In addition, we used linear discriminant analysis to identify a set of 12 genes that correctly classified immature individuals as migratory or sedentary with high accuracy. Using the expression levels of these 12 genes, 17 out of 18 individuals used for cross-validation were correctly assigned to their respective life-history phenotype. Finally, we found various candidate genes associated with physiological changes that are likely to be involved in preadaptations to seawater in anadromous populations of the genus Salmo, one of which was identified to encode for nucleophosmin 1. Our findings thus provide new molecular insights into salmonid life-history variation, opening new perspectives in the study of this complex trait.

Despite the progress achieved in elucidating the ecological mechanisms of adaptive radiation, there has been little focus on documenting the extent of adaptive differentiation in physiological functions during this process. Moreover, a thorough understanding of the genomic basis underlying phenotypic adaptive divergence is still in its infancy. One important evolutionary process for which causal genetic mechanisms are largely unknown pertains to life-history trade-offs. We analysed patterns of gene transcription in liver tissue of sympatric dwarf and normal whitefish from two natural lakes, as well as from populations reared in controlled environments, using a 16,006-gene cDNA microarray in order to: (i) document the extent of physiological adaptive divergence between sympatric dwarf and normal species pairs, and (ii) explore the molecular mechanisms of differential life history trade-offs between growth and survival potentially involved in their adaptive divergence. In the two natural lakes, 6.45% of significantly transcribed genes showed regulation either in parallel fashion (2.39%) or in different directions (4.06%). Among genes showing parallelism in regulation patterns, we observed a higher proportion of over-expressed genes in dwarf relative to normal whitefish (70.6%). Patterns observed in controlled conditions were also generally congruent with those observed in natural populations. Dwarf whitefish consistently showed significant over-expression of genes potentially associated with survival through enhanced activity (energy metabolism, iron homeostasis, lipid metabolism, detoxification), whereas more genes associated with growth (protein synthesis, cell cycle, cell growth) were generally down-regulated in dwarf relative to normal whitefish. Overall, parallelism in patterns of gene transcription, as well as patterns of interindividual variation across controlled and natural environments, provide strong indirect evidence for the role of selection in the evolution of differential regulation of genes involving a vast array of potentially adaptive physiological processes between dwarf and normal whitefish. Our results also provide a first mechanistic, genomic basis for the observed trade-off in life-history traits distinguishing dwarf and normal whitefish species pairs, wherein enhanced survival via more active swimming, necessary for increased foraging and predator avoidance, engages energetic costs that translate into slower growth rate and reduced fecundity in dwarf relative to normal whitefish.

Understanding how abundance regulates the effects of organisms on their ecosystems remains a critical goal of ecology, especially for understanding inter-ecosystem transfers of energy and nutrients. Here we examined how territoriality and nest-digging by anadromous salmon mediate trophic subsidies to stream fishes. Salmon eggs become available for consumption primarily by the digging of salmon that superimpose their nests on previous nests. An individual-based model of spawning salmon predicted that territoriality and habitat saturation produce a nonlinear effect of salmon density on numbers of available eggs to resident predators. Field studies in Alaskan streams found that higher densities of salmon produce disproportionately more eggs in stream drift and in diets of resident fishes (Arctic grayling and rainbow trout). Bioenergetics model simulations indicated that these subsidies produce substantially enhanced growth rates of trout. These results demonstrate that small changes in salmon abundance can drive large changes in subsidies to stream food webs. Thus, the ecological consequences of population declines of keystone species, such as salmon, will be exacerbated when behavior generates nonlinear impacts.

Body composition traits have potential use in fish breeding programs as indicator traits for selective improvement of feed efficiency. Moreover, feed companies are increasingly replacing traditional fish meal (FM) based ingredients in feeds for carnivorous farmed fish with plant protein ingredients. Therefore, genetic relationships of composition and feed utilization traits need to be quantified for both current FM-based and future plant-based aquaculture feeds. Individual whole-body lipid% and protein%, daily gain (DG), ADFI, and G:F (daily gain/daily feed intake) were measured on 1,505 European whitefish (Coregonus lavaretus) from 70 half/full-sib families reared in a split-family design with either a typical FM or a novel soybean meal (SBM) based diet. Diet-specific genetic parameters were estimated with multiple-trait animal models. Lipid% was significantly greater in the FM diet group than in the SBM group, even independent of final BW or total feed intake. In both diets, lipid% showed moderate heritability (0.12 to 0.22) and had positive phenotypic and genetic correlations with DG (0.37 to 0.82) and ADFI (0.36 to 0.88). Therefore, selection against lipid% can be used to indirectly select for lower feed intake. Protein% showed low heritability (0.05 to 0.07), and generally very weak or zero correlations with DG and ADFI. In contrast to many previous studies on terrestrial livestock, lipid% showed zero or very weak phenotypic and genetic correlations with G:F. However, selection index calculations demonstrated that simultaneous selection for high DG and reduced lipid% could be used to indirectly increase G:F; this strategy increased absolute genetic response in G:F by a factor of 1.5 to 1.6 compared with selection on DG alone. Lipid% and protein% were not greatly affected by genotype-diet environment interactions, and therefore, selection strategies for improving body composition within current FM diets should also improve populations for future SBM diets.

In a 3 months experiment, the effects of three different kinds of food rations including culture rotifers, commercial food and the equal amount mixture of these two foods were studied on some growth specifications and survival of the whitefish (Coregonus lavaretus) fry. The initial average weight and length of the fry were 0.009 g and 4 mm, respectively. The period of experiment was among April to July 2003. The assessed characteristics variance analysis showed that the different food treatments caused very significant differences among the all averages (p < 0.01). Comparing the characteristics averages showed that during the first month of experiment, the best results of weight (0.029 g), total length (8.78 mm), specific growth rate (5.36%) and food efficiency (63.48%) were obtained in live food treatment, whereas the weakest averages of referred characteristics were observed in commercial food treatment. During the last month of experiment, some of the above results reversed so that the best averages of specific growth rate (3.73%) and food efficiency (44.3%) were observed in commercial food treatment. The results of this study showed that the use of rotifer as live food for fry has provided better survival rate than the other two treatments. However, this food could not provide the best results for the all measured characteristics in all of the experiment period, for the reason that during the third month of experiment period, using commercial food treatment which consist of more miscellaneous nutrient components, created better results for some fry's characteristics than the live food.

The aquaculture industry is increasingly replacing fishmeal in feeds for carnivorous fish with soybean meal (SBM). This diet change presents a potential for genotype-environment (G x E) interactions. We tested whether current salmonid breeding programmes that evaluate and select within fishmeal diets also improve growth and efficiency on potential future SBM diets. A total of 1680 European whitefish from 70 families were reared with either fishmeal- or SBM-based diets in a split-family design. Individual daily gain (DG), daily feed intake (DFI) and feed efficiency (FE) were recorded. Traits displayed only weak G x E interactions as variances and heritabilities did not differ substantially between the diets, and cross-diet genetic correlations were near unity. In both diets, DFI exhibited moderate heritability and had very high genetic correlation with DG whereas FE had low heritability. Predicted genetic responses demonstrated that selection to increase DG and FE on the fishmeal diet lead to favourable responses on the SBM diet. Selection for FE based on an index including DG and DFI achieved at least double FE gain versus selection on DG alone. Therefore, current breeding programmes are improving the biological ability of salmonids to use novel plant-based diets, and aiding the aquaculture industry to reduce fishmeal use.

The purpose of this review is to collate physiological knowledge on salmonid eggs and larvae in relation to water quality criteria. Salmonid genera reviewed include Coregonus, Thymallus, Salvelinus, Salmo, and Oncorhynchus spp. When physiological data for salmonids are lacking, the zebrafish and medaka models are included. The primary focus is on the underlying mechanisms involved in the hydro-mineral, thermal, and respiratory biology with an extended section on the xenobiotic toxicology of the early stages. Past and present data reveal that the eggs of salmonids are among the largest shed by any broadcast spawning teleost. Once ovulated, the physicochemical properties of the ovarian fluid provide temporary protection from external perturbations and maintain the eggs in good physiological condition until spawning. Following fertilisation and during early development the major structures protecting the embryo from poor water quality are the vitelline membrane, the enveloping layer and the chorion. The vitelline membrane is one of the least permeable membranes known, while the semi-permeable chorion provides both physical and chemical defense against metals, pathogens, and xenobiotic chemicals. In part these structures explain the lower sensitivity of the eggs to chemical imbalance compared to the larvae, however the lower metabolic rate and the chronology of gene expression and translational control suggest that developmental competence also plays a decisive role. In addition, maternal effect genes provide a defense potential until the mid-blastula transition. The transition between maternal effect genes and zygotic genes is a critical period for the embryo. The perivitelline fluids are an important trap for cations, but are also the major barrier to diffusion of gases and solutes. Acidic environmental pH interferes with acid-base and hydromineral balance but also increases the risk of aluminium and heavy metal intoxication. These risks are ameliorated somewhat by the presence of ambient humic acid. High temperatures during development may be teratogenic, cause sexual bias, or long-term effects on muscle cellularity. Xenobiotics cause inhibition of neural acetylcholine esterase and carboxylases and disrupt the normal signalling pathways of hormones by binding to relevant receptors and mimicking their actions. A complex suite of genes is activated in response to environmental or parentally transmitted xenobiotics. The primary defense mechanism in embryos involves resistance to uptake but later biotransformation via the aryl hydrocarbon receptor (AHR)-mediated activation of members of the cytochrome mixed-function mono-oxygenase superfamily (CYP1A, CYP2B, and CYP3A) and subsequent glucuronidation or glutathionation. Due to the number of duplicate or triplicate genes coding for intermediates in the signalling pathways, and cross-talk between nuclear orphan receptors and steroid hormone receptors, a large number of complications arise in response to xenobiotic intoxicaton. One such syndrome, known as blue-sac disease causes an anaphylactoid response in hatched larvae due to increased permeability in the vascular endothelium that coincides with AHR-mediated CYP induction. Early embryos also respond to such xenobiotic insults, but apparently have an immature translational control for expression of CYP proteins, which coincides with a lack of excretory organs necessary for the end-point of biotransformation. Other syndromes (M74 and Cayuga) are now associated with thiamine deficiency. Where possible guidelines for water quality criteria are suggested.

1. Empirical studies show that average growth of stream-dwelling salmon and trout often declines with increasing density in a characteristic concave relationship. However, the mechanisms that generate negative density-growth relationships in populations in natural streams are not certain. 2. In a recent study, Imre, Grant & Cunjak (2005; Journal of Animal Ecology, 74, 508-516) argue that density-dependent growth due to exploitative competition for prey causes the negative density-growth relationships for stream salmonids. They argue that the concave shape of empirical density-growth relationships is consistent with a simple model of exploitative competition and not consistent with interference competition for space. 3. We use a simple model to show that competition for space can yield concave density-growth relationships consistent with the empirical pattern when individuals compete for foraging sites that vary spatially in quality and lower-quality sites predominate. Thus, the predictions of the exploitative competition and spatial competition models overlap. 4. The shape of the density-growth relationship does not differentiate between candidate mechanisms underlying density-dependent growth for stream salmonids. Our results highlight the general problem with determining the mechanism driving an ecological process from patterns in observational data within the context of linking population demographics to habitat structure and animal behaviour.

Elucidating the genetic architecture of population divergence may reveal the evolution of reproductive barriers and the genomic regions implicated in the process. We assembled genetic linkage maps for the dwarf and Normal lake whitefish species complex and their hybrids. A total of 877 AFLP loci and 30 microsatellites were positioned. The homology of mapped loci between families supported the existence of 34 linkage groups (of 40n expected) exhibiting 83% colinearity among linked loci between these two families. Classes of AFLP markers were not randomly distributed among linkage groups. Both AFLP and microsatellites exhibited deviations from Mendelian expectations, with 30.4% exhibiting significant segregation distortion across 28 linkage groups of the four linkage maps in both families (P < 0.00001). Eight loci distributed over seven homologous linkage groups were significantly distorted in both families and the level of distortion, when comparing homologous loci of the same phase between families, was correlated (Spearman R = 0.378, P = 0.0021). These results suggest that substantial divergence incurred during allopatric glacial separation and subsequent sympatric ecological specialization has resulted in several genomic regions that are no longer complementary between dwarf and Normal populations issued from different evolutionary glacial lineages.

Understanding the genetic architecture of reproductive barriers and the evolutionary forces that drove their divergence represents a considerable challenge towards understanding speciation. The objective of this study was to determine the genetic basis of intrinsic and extrinsic post-zygotic isolation in diverging populations of dwarf and normal lake whitefish with allopatric glacial origins. We found that the rate of embryonic mortality was 5.3-6.5 times higher in dwarf-normal hybrid backcrosses during development than in F1 dwarf and normal crosses. When comparing embryos that died during development against larvae that successfully hatched, patterns of Mendelian segregation at 101 loci whose linkage is known identified 13 loci distributed over seven linkage groups that exhibited significant shifts in segregation ratios leading to significant segregation distortion at these loci in the surviving progeny. Controlled crosses and quantitative trait loci analysis revealed a significant genetic basis for developmental time until emergence, a trait critical to fish larval survival in nature. Hatching backcross progeny exhibited asynchronous emergence and transgressive segregation, suggesting that extrinsic post-zygotic isolation may select against hybridization in specific environmental contexts. Evidence of a genetic basis for increased embryonic mortality followed by asynchronous emergence indicated that intrinsic and extrinsic mechanisms are not mutually exclusive in the formation and maintenance of reproductive isolation, but may be jointly promoting population divergence and ultimately speciation.

Brown trout (Salmo trutta) collected from a number of Danish streams impacted by sewage effluent were examined for alterations to gonadal development and induction of vitellogenin synthesis. Among fish collected in June/July 2000/2001 and November 2002 higher levels of plasma vitellogenin were found in males from six streams impacted by sewage effluent compared to males from their respective reference sites. A direct non-competitive ELISA was developed for brown trout vitellogenin in order to perform the vitellogenin measurements. Intersex in females with no apparent relation to sewage effluent exposure was observed at all sites. In one stream, male brown trout with a very high level of vitellogenin were concomitantly found to have a high degree of vacuolation of the testes and a presence of only the early spermatogenic stage, spermatogonia. The cause of these alterations to the testis structure is unclear. However, as a high level of plasma vitellogenin in these males indicated estrogenic exposure, the vacuolation might also be a result of endocrine disruption causing delayed or disrupted spermatogenesis.

The activities of alanine and aspartate aminotransferases were assessed in the embryos and juveniles of the rainbow trout Parasalmo mukiss L. and chum salmon Oncorchyncus keta W. Changes in subcellular localization of these enzymes and their activities were found at different pH optima in each subcellular fraction of the rainbow trout during ontogenesis.

As natural selection must act on underlying genetic variation, discovering the number and location of loci under the influence of selection is imperative towards understanding adaptive divergence in evolving populations. Studies employing genome scans have hypothesized that the action of divergent selection should reduce gene flow at the genomic locations implicated in adaptation and speciation among natural populations, yet once 'outlier' patterns of variation have been identified the function and role of such loci needs to be confirmed. We integrated adaptive QTL mapping and genomic scans among diverging sympatric pairs of the lake whitefish (Coregonus clupeaformis) species complex in order to test the hypothesis that differentiation between dwarf and normal ecotypes at growth-associated QTL was maintained by directional selection. We found evidence of significantly high levels of molecular divergence among eight growth QTL where two of the strongest candidate loci under the influence of directional selection exhibited parallel reductions of gene flow over multiple populations.

Organisms often shown enhanced growth during recovery from starvation, and can even overtake continuously fed conspecifics (overcompensation). In an earlier paper (Ecology 84, 2777-2787), we studied the relative role played by hyperphagia and resource allocation in producing overcompensation in juvenile (non-reproductive) animals. We found that, although hyperphagia always produces growth compensation, overcompensation additionally requires protein allocation control which routes assimilate preferentially to structure during recovery. In this paper we extend our model to cover reproductively active individuals and demonstrate that growth rate overcompensation requires a similar combination of hyperphagia and allocation control which routes the part of enhanced assimilation not used for reproduction preferentially towards structural growth. We compare the properties of our dynamic energy budget model with an earlier proposal, due to Kooijman, which we extend to include hyperphagia. This formulation assumes that the rate of allocation to reserves is controlled by instantaneous feeding rate, and one would thus expect that an extension to include hyperphagia would not predict growth overcompensation. However, we show that a self-consistent representation of the hyperphagic response in Kooijman's model overrides its fundamental dynamics, leading to preferential allocation to structural growth during recovery and hence to growth overcompensation.

We studied the activities of digestive enzymes responsible for the digestion of food carbohydrate and protein components in plant-eating fish at various stages of larval development. The activities of all digestive enzymes tend to rise during larval development. Species specific features of the alimentary canal functioning have been described.

A comparative cytogenetic analysis of coregonid fish was carried out in the river and lake ecosystems of the northern Tyumen' District. The occurrence of chromosomal aberrations in embryonic cells of fish steadily increased during the period from 1989 until 2001. Possible causes of this dynamic are discussed.

Background: Although fish allergy is common, no studies have been published on allergy to fish roe., Objective: To describe 2 cases of IgE-mediated allergy to 2 roe species., Methods: Two patients, one with local symptoms and the other with anaphylaxis following ingestion of roe, underwent skin prick testing (SPT) with 2 roe species, whitefish roe (WFR) and rainbow trout roe (RTR). Serum samples were taken for IgE measurement and immunoblotting to identify roe allergens. Inhibition studies were performed to scrutinize the cross-reactivity between the roes and to fish., Results: The results of the SPTs with the roes were clearly positive in both patients but negative in control persons. The results of SPTs to all other foods were negative. Roe-specific IgE levels were elevated in the serum samples of both patients. Immunoblotting revealed different IgE-binding patterns of the extracts and different inhibition profiles with the serum samples. In WFR blotting, both serum samples detected a heavy IgE-binding band at approximately 20 kDa, which was not inhibited with fish. Cross-reactivity between the roes was demonstrated in the patient with local symptoms from RTR but not in the patient with anaphylaxis from WFR. The first serum sample also detected several IgE-binding bands in the RTR blot, the most intensive at 21 to 23 kDa and 30 kDa, which were partially inhibited by WFR and more completely with fish. The anaphylaxis patient did not detect allergens in the RTR blot. After the investigation, the patients have remained symptom free and able to consume all kinds of fish without problems., Conclusions: IgE-mediated allergy to roe is possible without concomitant fish allergy. Roe allergy should be explored in patients who test negative to fish but are suspected of having seafood-related allergy.

Seasonal occurrence in terms of prevalence, intensity of infection, abundance and density of the tapeworm Proteocephalus longicollis (Zeder, 1800) and its transmission between its intermediate host (Cyclops abyssorum prealpinus) and definitive host (common whitefish, Coregonus lavaretus) in Lake Annecy, an oligotrophic lake in the western part of the Alps, France, were studied in the period of 1998-2000. A copepod Cyclops abyssorum prealpinus (Kiefer, 1939), the dominant species among planktonic crustaceans in the lake, served as the only intermediate host for this parasite. Infection with plerocercoids was higher in adult copepods (predominantly females) than in copepodite stages IV and V. The prevalence rate of 25% found in C. abyssorum prealpinus females in June 1998 represents a unique infection rate of intermediate hosts with fish tapeworms in natural conditions. The final host, the common whitefish Coregonus lavaretus (L.), was heavily infected with P. longicollis throughout the year (prevalence 90%; mean abundance 40.3; maximum intensity of infection more than 500 tapeworms per fish); immature tapeworms predominated in all samples (P<0.01). Transmission of tapeworm larvae from copepods to the common whitefish took place most intensively in summer and autumn, and depended on seasonal changes in the density of the C. abyssorum prealpinus population, infection of this copepod with plerocercoids and their density in the lake. In addition, transmission efficiency also seems to be determined by the longevity of tapeworm larvae in the intermediate host, timing of predation of the fish host and rapid development of the parasite within this host during the summer period. Overall transmission potential of P. longicollis was low, with only about 9% of juvenile specimens reaching maturity in common whitefish.

Low phosphorous (P) feeds in aquaculture are recommended to reduce eutrophication of water systems. However, the feed should be adequate for normal growth and intact immune defence. Influence of low dietary P supply on non-specific and specific immune defence of European whitefish (Coregonus lavaretus) was studied in two trials. For Trial 1, a semi-purified, low-P diet was formulated and supplied with 0, 2.5, 5.0, 7.5, or 10.0 g P kg(-1)to obtain a P gradient of 4.4-14.9 g kg(-1)diet. Diets were fed to four replicate groups of fingerling whitefish for 42 days in a flow-through, freshwater system maintained at 15 degrees C. Fish fed with the P-unsupplemented diet had significantly lower plasma immunoglobulin M (IgM) levels than fish fed with the P-fortified diets (means 2.53 vs. 3.19 mg ml(-1); P=0.047). Plasma lysozyme activity did not differ between fish fed with the P-unsupplemented diet and fish fed with the P-fortified diets (means 21.8 vs. 25.5 U ml(-1); P=0.107). For Trial 2, fish were acclimated for 49 days by feeding the lowest or highest dietary P contents, and thereafter immunised with a single intraperitoneal injection of a novel antigen, bovine gamma globulin (BGG). A clear antibody response against BGG was elicited but the antibody levels did not differ between fish fed low-P or high-P contents. Similar to Trial 1, plasma IgM was lower in fish fed low-P diets, and low-P diet did not influence plasma lysozyme activity. In both trials the growth of fish, having received low-P diet, was significantly lowered. It is concluded that P deficiency has only minor effects on the immune parameters in whitefish, and the practical aquafeed with P contents sufficient for normal growth does not compromise immune functions of this species.

Positively phototactic fish larvae may be exposed to increased ultraviolet-B (UV-B) radiation alone or, potentially and in addition, to polycyclic aromatic hydrocarbons (PAHs) such as retene (7-isopropyl-1-methylphenanthrene) at the egg or larval stages. Suspended and sedimental particulate material near pulp and paper mills may act as sources of retene in chemically contaminated lake areas. In laboratory conditions whitefish larvae were pre-exposed to retene (10, 32 and 100 microg/l), with relevant controls, and irradiated in semi-static tests for 3 h once a day (2 consecutive days) with two UV-B doses (CIE-weighted 2.8 or 5.4 kJ per m(2) per day) or with visible light only. These UV-B doses correspond with slightly subambient and 80% increases relative to the natural maximum daily doses of the solar UV-B in Finland in early May. The UV-B radiation alone increased mortality only slightly (2.7 and 4.0%, respectively). Similarly, no mortality was observed due to retene alone. On the contrary, simultaneous UV-B and retene exposure caused very high mortality to whitefish and all larvae died in the highest retene concentration. The photoinduced acute LC(50) for retene was 13.3 microg/l. Retene treated fish exhibited signs of behavioral irritation and hypoxia during and after the exposure to UV light. Severe skin damages were detected in larvae exposed simultaneously to retene and UV-B. The structural signs of sunburn could also be seen in UV-B and solvent controls (DMSO) with UV-B. Even at the lowest retene concentration, the number of mucous cells increased significantly in simultaneous chemical and UV-B treatment. We consider the tissue reaction as protective response against UV induced retene toxicity. Further, regarding liver parenchyma, fish exposed to retene with UV-B had lesions, revealing hepatotoxicity. We suggest that synergism of the UV-B radiation and the photoactivating pollutants such as retene is a potential risk factor to be taken into consideration in lake areas chemically contaminated by the pulp and paper industry.

Selection against large eggs has been proposed for aquatic environments, putatively because large eggs should have more difficulty obtaining the required oxygen. Here, we use brown trout (Salmo trutta) eggs to provide an experimental test of this hypothesis. At high levels of dissolved oxygen (14 mg l(-1)), egg survival was high and independent of egg size. At low oxygen levels (2.3 mg l(-1)), survival decreased overall, and was higher for large-egged than small-egged siblings. Thus, contrary to conventional expectation, low oxygen levels selected for large rather than small eggs. A second experiment using Atlantic salmon (S. salar) eggs indicated that oxygen consumption increases relatively slowly with increasing egg mass (allometric constant = 0.44). The failure of the conventional 'bigger is worse during incubation' hypothesis may thus be due to the erroneous assumption that oxygen consumption increases at a greater rate with increasing egg mass than does the egg surface area that is available for oxygen diffusion. We also demonstrate, using data from Atlantic salmon, that nest-specific oxygen consumption decreases with increasing egg size, but that this effect is more pronounced for large than for small females. This may help to explain the positive correlation between adult body size and egg size observed in fishes that cluster their eggs.

The sea louse (Lepeophtheirus salmonis Krøyer) is a major health problem for both farmed and wild salmonids. This paper investigates louse epidemiology and management in the salmon-farming zone of western Scotland. Based on a review of the marine ecology of wild salmon (Salmo salar L) and sea trout (Salmo trutta L), and catch and farm production statistics, best estimates were made for numbers of wild and farm hosts present in coastal waters in March-June 2000. Applying data for ovigerous female louse infections and fecundity, the sources and risks of larval transmission to wild salmon and sea trout were modelled. Farm salmon in the second spring of production were the primary host group (98% of fish), while numbers of wild salmonids (< 1%) and escaped farm salmon (2%) were relatively insignificant. Farm salmon produced 97% of louse eggs at high levels (eight ovigerous lice per fish), and 78% at low levels (one per fish). Wild salmonids produced < 1% of eggs under both scenarios, but escaped farm salmon produced 3% and 21%, respectively. All hosts potentially cross-infect one another, but farm salmon are more likely to infect wild and farm smolts, and also other farm salmon. Monitoring of lice on sea trout in June 1998-2000 by the Association of West Coast Fisheries Trusts corroborated the model's conclusions. Localised epizootics occurred every year and coincided with the presence of ovigerous lice on local farms. In areas of mixed-year class production on farms, epizootics were evident every spring, but occurred every second spring in areas of single-year class production. In 1998-2000 at least 14-40% of sea trout were infected with potentially lethal infestations of lice. Ovigerous louse levels of < 0.005 per fish were required on farm salmon in the spring of 2000 to produce less eggs than those emitted by wild salmonids. With the industry's continued expansion, and thus increased numbers of farm salmon, a target of zero ovigerous lice will be required on farms to minimise impacts on wild salmonids. Due to the limited long-term efficacy and availability of louse medicines, management strategies are discussed which will improve control, including single-year class production over large areas, alternate S1-S1/2 smolt inputs, and 11-month production cycles.

The development of brown trout (Salmo trutta f. fario L.) in water of two differently polluted streams and in a control situation was monitored in order to get insights into the impact of anthropogenic chemical stressors on the reproductive success of this fish species indigenous to both streams. The test streams, situated in the south of Stuttgart, Germany, were the complexly polluted Körsch stream and the less polluted Krähenbach stream. Bypass systems connected to the streams and a laboratory control system were used for continuous exposure of early brown trout stages shortly after fertilisation up to the end of the embryonic development. Temperature and oxygen conditions were standardised in all test series in order to minimise unspecific effects. The examined endpoints were: (1) mortality, (2) developmental rate, (3) time course of hatching, (4) malformations, and (5) growth. A retarded development, reduced growth rates and higher mortality rates of Körsch stream water exposed embryos indicated an embryotoxic potential for the more polluted stream. High infection-related mortality rates of embryos suggested the presence of confounding factors also in the less polluted Krähenbach stream. In parallel to the exposure experiment, physicochemical and limnochemical parameters as well as concentrations of organic contaminants and heavy metals were monitored. Analytical data confirm the different degrees of pollution of both streams.

Water quality was measured in 42 streams in the Colebrooke and Upper Bann catchments in Northern Ireland over the period 1990-1998. Despite ongoing pollution control measures, biological water quality, as determined by the invertebrate average score per taxon (ASPT) index, did not improve and there was no appreciable decline in recorded farm pollution incidents. However, the lack of decline in pollution incidents could reflect changes in detection policy, as a greater proportion of incidents were recorded from less polluting discharges such as farm-yard runoff. In contrast, there was an improvement during 1997 and 1998 in annual chemical water quality classification based on exceedence values (90th percentiles) for dissolved oxygen, ammonium and BOD concentrations. In 1998, 11.9% of streams were severely polluted compared to 26.2% in 1990, while the proportion classed as of salmonid water quality, increased from 40.5% in 1990 to 59.6% in 1998. Although water quality in 1996 did not improve relative to 1990 values, there was a notable increasing trend from 1990 in the numbers of samples taken during the summer which had good water quality with low ammonium (<0.6mgN l(-1)) and high dissolved oxygen (> 70% sat). The trend for samples with low BOD (<4 mgl(-1)) was more erratic, but an improvement was apparent from 1994. These improvements in chemical water quality suggest that point-source farm pollution declined after 1990. The fact that this was not reflected in stream biology may reflect the limited time scale for biological recovery. An important factor preventing biological recovery may be the high pollution capacity of manures and silage effluent, so that even reduced numbers of farm pollution incidents can severely perturb stream ecosystems. The intractable nature of farm pollution suggests that there is a need to consider an interactive approach to problem resolution involving both farmers and regulators.

Synchronic and allochronic data sets consisting of phenotypic values of various life-history traits from five grayling Thymallus thymallus populations with common ancestors were analysed for the purpose of estimating evolution and divergence rates. The synchronic data contained both juvenile and adult traits from populations that have been segregated for 44-88 years (9-22 generations). The allochronic time series contained growth- and maturation data spanning 95 years (16 generations). Estimated evolution and divergence rates were high compared with other life-history studies on the same temporal scale (0.002-1.008 haldanes, 10-30, 500 darwins). The divergence of adult traits were most probably caused by differential mortalities induced by variation in fishing intensity. For the population with allochronic data, 48 years (eight generations) of intense and consistent size-selective gill-net fishing resulted in a constant reduction in age (-0.33 years pr 10 year) and length (-18mm pr 10 year) at maturity. Length-at-age for ages one to five also decreased during the same period. When gill-net fishing was relaxed, age and length at maturity and length-at-age increased. Divergence rates for juvenile traits derived from a common-garden experiment were high, and standardized selection differentials (s') were high, especially for yolk-sac volume (s' = 2.6). We also document that low divergence rates for juvenile traits were lower between populations having similar spawning/nursery habitats (running water) than populations having relatively different habitats (running water v.s. still water). We suggest that the major part of the observed phenotypic divergence is mostly due to adaptive evolution, although microsatellite data indicate that genetic drift also has occurred.

There is limited knowledge about the effects of oil-based vaccines on the growth of Arctic charr Salvelinus alpinus, in particular at different rearing temperatures. One-year-old Arctic charr were immunized intraperitoneally at 2.9 degrees C with a metabolizable oil-adjuvanted, bivalent vaccine containing killed typical and atypical Aeromonas salmonicida bacteria. After vaccination the non-vaccinated (controls) and vaccinated individually marked fish were held for 20 d at 10.0 degrees C and then for 7 wk at 10.3, 14.1 or 18.1 degrees C. During the first 20 d at 10.0 degrees C the growth rate (G) was higher for non-vaccinated than vaccinated fish. Thereafter vaccinated charr had higher G than control fish at 10.3 and 14.1 degrees C. In contrast, at 18.1 degrees C there was no difference in G and therefore no compensation of earlier growth suppression in vaccinated fish was observed at that temperature. The study indicates that vaccination has no ultimate negative effects on the growth of Arctic charr at temperatures ranging from 10.3 to 14.1 degrees C.

Fish ecotypes found in north temperate lakes are increasingly used as model organisms to explore patterns and processes of population divergence that may ultimately cause speciation. Processes involved in their reproductive isolation are, however, still poorly understood. Recent experimental studies on whitefish ecotypes from different glacial races revealed that embryonic mortality of hybrids was 2.4-4.7 times higher than for parental forms. In this study, we compared fluctuating asymmetry (FA) in morphological traits of these same hybrid and pure crosses to test the hypothesis that genetic stress observed in hybrids at embryonic stages is also manifested at later developmental stages. Twelve morphological traits were used to measure asymmetry. Variable degrees of asymmetry were observed depending on traits and crosses, however there was no significant difference in FA among crosses. These results thus provided no evidence in support of the working hypothesis and indicated that genetic stress may differ among life stages. It is more likely that high hybrid embryonic mortality acts together with ecological factors at later stages to increase further the extent of reproductive isolation between sympatric whitefish ecotypes.

To examine whether the utilization of protein and lipids is altered in the genetically modified, rapidly growing charr, we compared CMVOnGH1 transgenic and sibling fish. Muscle composition and rates of gas exchange were analyzed. Plasma metabolites were determined in the recently fed and post-absorptive state. No difference was found in muscle composition. At equal rates of protein accretion, the rate of NH4 excretion was 43% greater in sibling charr. The lower molar ratio of NH4 to O2 exchange implied the reduced expenditure of metabolized protein in transgenic charr. Plasma NH4 concentration in transgenic fish did not differ from that in sibling charr whereas the greater level of total CO2 indicated enhanced oxidation of non-protein nutrients. Decreased plasma triglycerides concentration and lower triglyceride to cholesterol ratio showed faster utilization of ingested lipids in transgenic charr, especially of energy-containing fraction. However, this was not accompanied with a reduced lipid content or altered fatty acid composition of muscle triglycerides or phospholipids. Comparative studies suggested that the transgenic charr had acquired features of domesticated salmonid fish. Their increased metabolic rate and enhanced utilization of dietary lipids, especially triglycerides, resembled the characteristics of domestic rainbow trout rather than wild counterparts.

Four constructs containing salmonid growth hormone (GH) genes were transferred to Arctic charr (Salvelinus alpinus L.). Cytomegalovirus (CMV) and piscine metallothionein B (OnMT) and histone 3 (OnH3) promoters connected to sockeye salmon growth hormone 1 gene (OnGH1) were used for ectopic expression, and Atlantic salmon growth hormone 2 gene with 5'flanking region (SsGH2) was tested for pituitary-specific expression. Charr carrying the OnGH1 constructs showed a dramatic increase in growth rate. The 10-month old transformed fish were 14-fold heavier than control siblings. The ability of the CMVGH1 construct to promote growth was greater than that obtained in fish with piscine promoters. Analysis of individual growth curves of charr carrying the OnH3GH1 transgene indicated a stable ratio of specific growth rates in transformed and control fish regardless of fish size. No alteration in growth performance was found in fish carrying the SsGH2 transgene. There was evidence that the transformed rainbow trout (Oncorhynchus mykiss) were unable to produce SsGH2 mRNA in their pituitary glands. The presence of the transgene in various tissues was examined in trout to evaluate the reliability of one-tissue sampling.

Morphology, histology, and immunohistochemistry of the adrenocortical and adrenomedullary homologs (adrenal glands) of the following developing and adult teleosts were examined: Salmoniformes-Oncorhynchus mykiss (rainbow trout), Salmo trutta fario (brown trout), Coregonus lavaretus (white fish); Cyprinodontiformes-Gambusia affinis (mosquito fish). Perciformes-Dicentrarchus labrax (sea bass), Sparus aurata (sea bream), Diplodus sargus (white bream), Oblada melanura (saddled bream). The anatomical relationships of the gland with the renal system and venous vessels were also noted. In adults of all species steroidogenic and catecholaminergic chromaffin cells were found in the head kidney, which is pronephric in origin and subsequently transformed into a hematopoietic lymphatic organ. In Perciformes, chromaffin cells are distributed around the anterior and posterior cardinal veins and ducts of Cuvier; in Salmoniformes, around the posterior cardinal veins and in the hematopoietic tissue; and in G. affinis, around the ducts of Cuvier and posterior cardinal veins, while a few are visible also around the sinus venosus. In Perciformes and Salmoniformes, numerous chromaffin cells are also present in the posterior kidney, derived from the opisthonephros, in contact with the caudal vein. Steroidogenic cells are always confined to the head kidney. During development chromaffin and steroidogenic cells appear early after hatching in the pronephric kidney, at the level of the ducts of Cuvier and of the cephalic part of the posterior cardinal veins. Later, chromaffin cells in Perciformes reach the anterior cardinal veins, and subsequently, in both Perciformes and Salmoniformes, they reach the developing posterior kidney. Their localization along the posterior kidney is still in progress about 4 months after hatching and is completed about a year after hatching. These findings support the concept that the structure of the adrenal gland in teleosts is intermediate between that of the other actinopterygians and that of tetrapods. The development differs from that of tetrapods in that it occurs mainly in the pronephros and only later do chromaffin cells reach the opisthonephric kidney., (Copyright 1997 Academic Press.)

1. During early ontogeny, the serotonergic neurons in the brain stem of the three-spined stickleback shows a temporal and spatial developmental pattern that closely resembles that of amniotes. 2. However, in the adult fish, only the midline nuclei of the rostral group (dorsal and median raphe nuclei) and the dorsal lateral tegmental nucleus are consistently serotonin-immunoreactive (5-HTir), whereas the groups of the upper and lower rhombencephalon (raphe pontis, raphe magnus, and raphe pallidus/obscurus nuclei) are variable and, when present, contain relatively small numbers of 5-HTir neurons. 3. Using specific antisera against tryptophan 5-hydroxylase and aromatic L-amino acid decarboxylase, we have shown that the lateral B9 group and the groups of the upper and lower rhombencephalon are consistently present in adult sticklebacks. The results are discussed in relation to other known instances of neurotransmitter plasticity or transient neurotransmitter expression in teleost fish. 4. While there are several instances of transient expression of neurotransmitter markers by discrete neuronal populations, there is so far no evidence of changes from one neurotransmitter phenotype to another in the brain of teleost fish. However, there are indications of plasticity of expression of catecholamines and indoleamines, and their respective synthesizing enzymes, as reflected in age-dependent changes and variation between individuals of different physiological status. 5. As the brain grows continuously in teleost fish, and new neurons are added from proliferative regions, synaptic connections may be expected to undergo remodeling in all brain regions throughout life. Thus, the teleostean brain may be considered a suitable model for experimental studies of different aspects of neural plasticity.

Brook charr (Salvelinus fontinalis) were maintained at one of two stocking densities (SD) (30 or 120 kg/m3) and fed either a control or a T3-supplemented (20 mg/kg) diet for 30 days in order to investigate possible interactive effects of SD and T3-administration on growth, feeding rate, food conversion efficiency, and hepatic and dark muscle enzyme activity. In addition, liver slices were incubated in vitro for 6 h with epinephrine, norepinephrine, isoproterenol, propranolol, insulin, glucagon, or somatostatin to evaluate possible SD-T3 interactive effects on hepatic responses to hormonal stimulation. Maintaining the fish at high SD appeared to increase the clearance rate of T3 from the T3-supplemented group. There was no clear evidence of SD-T3 interactive effects on growth rate, feeding rate, or food conversion efficiency, although T3-administration decreased food conversion efficiency, and high SD decreased growth and feeding rates. Of the hepatic enzymes studied, HOAD, malic enzyme, G6PDH, CS, PFK, HK, and GDH activities all showed changes suggestive of interactive SD-T3 effects. Although hepatic FBPase was stimulated by both high SD and T3-administration, there was no evidence of interactive SD-T3 effects. Dark muscle HOAD, CS, and PFK also showed SD-T3-related responses; dark muscle malic enzyme, G6PDH, HK, and GDH were unaffected by either altered SD or T3-administration. Prior treatment of the fish with T3 and high SD had significant effects on free fatty acid (ffa) release to the medium and on hepatic lipid content, but had no effect on the responses to the various endocrine agents used. Glucose release from liver slices of fish stocked at high density (both T3-supplemented and controls) was higher than that of the fish stocked at low density; with the exception of insulin and glucagon, glucose release was similar in all pre-treatment groups. The insulin- and glucagon-stimulated changes in glucose release seen in the fish fed non-supplemented diets were not found in the two groups of fish fed the T3-supplemented diets. High SD and/or T3-administration induced significant lowering of hepatic glycogen content, but there was no effect of pre-treatment on the response to any of the endocrine agents used. The data show a marked effect of SD on energy partitioning processes in brook charr and the animal's ability to respond to T3-stimulation, but provided no evidence of such effects on the liver response to the various agents used.

Dietary requirements for amino acids and fatty acids have been reported for channel catfish (Ictalurus punctatus), salmonids (Oncorhynchus spp.), common carp (Cyprinus carpio), tilapias (Oreochromis spp.), and eel (Anguilla japonicus). Most of the vitamin and mineral requirements are available for channel catfish and salmonids, and some are available for common carp, tilapia, eel, and other finfish and crustaceans. From this available information, cost-effective feeds can be formulated for the major commercial aquaculture species. Major differences in nutrient requirements between fish and mammals or birds are as follows: fish have a lower digestible energy:protein ratio (8 to 10 kcal of DE/g of CP for fish vs 15 to 20 kcal of DE/g of CP for livestock); fish require n-3 fatty acids and land animals require n-6; fish can absorb minerals from the water, which negates the need for some minerals in the diet; and fish have limited ability to synthesize vitamin C and must depend on a dietary source. Areas for further research include 1) refinement of nutrient requirements of the major culture species considering effects of fish size, temperature, and management; 2) nutrient requirements of crustaceans; 3) effects of nutrition on fish health and product quality; and 4) feeding technology.
