Abstract

Renal fibrosis is the final manifestation of chronic kidney disease (CKD); its prevention is vital for controlling CKD progression. Indoxyl sulfate (IS), a typical sulfate-conjugated uremic solute, is produced in the liver via the enzyme sulfotransferase (SULT) 1A1 and accumulates significantly during CKD. We investigated the toxicopathological role of IS in renal fibrosis using Sult1a1-KO mice and the underlying mechanisms. The unilateral ureteral obstruction (UUO) model was created; kidney IS concentrations, inflammation, and renal fibrosis were assessed on day 14. After UUO treatment, inflammation and renal fibrosis were exacerbated in WT mice, with an accumulation of IS in the kidney. However, they were significantly suppressed in Sult1a1-KO mice. CD206+ expression was upregulated, and β-catenin expression was downregulated in Sult1a1-KO mice. To confirm the impact of erythropoietin (EPO) on renal fibrosis, we evaluated the time-dependent expression of EPO. In Sult1a1-KO mice, EPO mRNA expression was improved considerably; UUO-induced renal fibrosis was further attenuated by recombinant human erythropoietin (rhEPO). Thus, UUO-induced renal fibrosis was alleviated in Sult1a1-KO mice with a decreased accumulation of IS. Our findings confirmed the pathological role of IS in renal fibrosis and identified SULT1A1 as a new therapeutic target enzyme for preventing and attenuating renal fibrosis. [ABSTRACT FROM AUTHOR]

Abstract

Species diversity encompasses both richness and evenness, but the interrelationship between these two aspects remains poorly understood. The long‐term dynamics of species diversity were examined in three natural secondary forests in Hokkaido, northern Japan, using permanent plot data gathered over 65–66 years after severe windthrow damage. The relationships among species diversity, species richness, and evenness were analyzed in the context of stand dynamics. Temporal trends in species diversity were unimodal‐shaped in three permanent plots, consistent with the pattern predicted by the intermediate disturbance hypothesis. Species richness and evenness increased following the windthrow, but decreased 27–37 years later in all three plots. In two plots, species diversity was closely associated with species richness and evenness at the early and late successional stages, respectively; in the other plot, species diversity was significantly related to both species richness and evenness in the later stage. The former two plots and the latter one plots differed markedly in terms of the extents of fatal tree damage caused by windthrow. Thus, windthrow severity affected the relationships among diversity, richness, and evenness as succession proceeded in these secondary forests. The tree density, species richness, evenness, and species diversity in all three plots decreased when the basal areas exceeded 18–24 m2/ha, indicating that increased competition among trees affected the temporal trends in these metrics. Abstract

The stimulation of photosynthesis is a strategy for achieving sustainable plant production. Red light is useful for plant growth because it is absorbed by chlorophyll pigments, which initiate natural photosynthetic processes. Ultraviolet (UV)-to-red wavelength-converting materials are promising candidates for eco-friendly plant cultures that do not require electric power. In this study, transparent films equipped with a UV-to-red wavelength-converting luminophore, the Eu3+ complex, were prepared on commercially available plastic films for plant growth experiments. The present Eu3+-based films absorb UV light and exhibit strong red luminescence under sunlight. Eu3+-painted films provide significant growth acceleration with size increment and biomass production for vegetal crops and trees in a northern region. The plants cultured with Eu3+-painted films had a 1.2-fold height and 1.4-fold total body biomass than those cultures without the Eu3+ luminophores. The present film can promote the plant production in fields of agriculture and forestry.

Abstract

Background. The objective of this study was to assess the relation between serum levels of retinol and other antioxidants and hearing impairment in Japanese older adults. Methods. This is a community-based cross-sectional study comprising 762 residents aged 65 years or older in Kurabuchi, Gumma, Japan. We measured serum retinol and other antioxidants (alpha- and gamma-tocopherols, and carotenoids including beta-cryptoxanthin, alpha- and beta-carotenes, lycopene, and lutein plus zeaxanthin) by high-performance liquid chromatography and divided each measurement into quartiles. Hearing impairment was defined as a failure to hear a 30-dB hearing level (HL) signal at 1 kHz and a 40-dB HL signal at 4 kHz in the better ear in pure-tone audiometric tests. The odds ratios (OR) for hearing impairment were calculated for each of the upper three quartiles of retinol and other antioxidant levels relative to the lowest quartile. Results. Crude analysis showed that serum levels of retinol and provitamin A carotenoids (beta-cryptoxanthin, and alpha- and beta-carotenes) were inversely related to the prevalence of hearing impairment. The multiadjusted ORs (95% confidence intervals) for the highest quartile of retinol and the provitamin A family (combinations of provitamin A carotenoids) compared with the lowest were 0.51 (0.26-1.00) and 0.53 (0.27-1.02), respectively. A dose-response relationship was observed for retinol (p = .03) and provitamin A (p = .09). Conclusion. Increased serum levels of retinol and provitamin A carotenoids were clearly associated with a decreased prevalence of hearing impairment. Key Words: Retinol--Provitamin A--Hearing--Aged--Japan.

Abstract

The neuropeptide hormone arginine-vasopressin (AVP) is well known to exert its antidiuretic effect via the vasopressin V2 receptor (V2R), whereas the role of the vasopressin Vla receptor (V1aR) in the kidney remains to be clarified. Previously, we reported decreased plasma volume and blood pressure in V1a receptor-deficient ([V1aR.sup.-/-]) mice (Koshimizu T, Nasa Y, Tanoue A, Oikawa R, Kawahara Y, Kiyono Y, Adachi T, Tanaka T, Kuwaki T, Mod T. Proc Natl Acad Sci USA 103: 7807-7812, 2006). In this study, we investigated the role of V 1 aR in urine concentration, renal function, and the renin-angiotensin system (RAS) using [V1aR.sup.-/-] mice. Urine volume of [V1aR.sup.-/-] mice was greater than that of wild-type mice, particularly when water was loaded, while the glomerular filtration rate (GFR), urinary NaCl excretion, AVP-dependent cAMP generation, V2R, and aquaporin 2 (AQP2) expression in the kidney were lower, indicating that the diminished GFR and V2R-AQP2 system led to impaired urinary concentration in [V1aR.sup.-/-] mice. Since the GFR and V2R-AQP2 system are regulated by RAS, we analyzed renin and angiotensin II in [V1aR.sup.-/-] mice and found that the plasma renin and angiotensin II were decreased. The expression of renin in granule cells was decreased in [V1aR.sup.-/-] mice, which led to a decreased level of plasma renin. In addition, the expression of renin stimulators such as neuronal nitric oxide synthase and cyclooxygenase-2 in macula densa (MD) cells, where V1aR was specifically expressed, was decreased in [V1aR.sup.-/-] mice. These data indicate that AVP regulates body fluid homeostasis and GFR via the V1aR in MD cells by activating RAS and subsequently the V2R-AQP2 system. renin-angiotensin system
