Pyramiding of major resistance (R) genes through marker-assisted selection (MAS) is a useful way to attain durable and broad-spectrum resistance against Xanthomonas oryzae pv. oryzae pathogen, the causal agent of bacterial blight (BB) disease in rice (Oryza sativa L.). The present study was designed to pyramid four broad spectrum BB-R genes (Xa4, xa5, xa13 and Xa21) in the background of Basmati-385, an indica rice cultivar with much sought-after qualitative and quantitative grain traits. The cultivar, however, is susceptible to BB and was therefore, crossed with IRBB59 which possesses R genes xa5, xa13 and Xa21, to attain broad and durable resistance. A total of 19 F1 plants were obtained, some of which were backcrossed with Basmati-385 and large number of BC1F1 plants were obtained. In BC1F2 generation, 31 phenotypically superior genotypes having morphological features of Basmati-385, were selected and advanced up to BC1F6 population. Sequence-tagged site (STS)-based MAS was carried out and phenotypic selection was made in each successive generation. In BC1F6 population, potentially homozygous recombinant inbred lines (RILs) from each line were selected and evaluated on the bases of STS evaluation and resistance to local Xanthomonas oryzae pv. oryzae (Xoo) isolates. Line 23 was found pyramided with all four BB-R genes i.e., Xa4, xa5, xa13 and Xa21. Five genotypes including line 8, line 16, line 21, line 27 and line 28 were identified as pyramided with three R genes, Xa4, xa5 and xa13. Pathological study showed that rice lines pyramided with quadruplet or triplet R genes showed the highest level of resistance compared to doublet or singlet R genes. Thus, line 23 with quadruplet, and lines 8, 16, 21, 27, and 28 with triplet R genes, are recommended for replicated yield and resistance trials before release as new rice varieties. Strigolactones, a new group of phytohormones, are reported to improve plant tolerance to multiple abiotic stresses. A pot experiment was conducted to investigate the impact of synthetic strigolactone (GR24 at 0.001, 0.01 and 0.1 mg L−1) application on ornamental sunflowers (Helianthus annuus cv. Vincent's Choice) grown under salt stress (150 mM NaCl). Salt stress was applied after 14 days, and SL was applied 25 days seed sowing. The results showed that amongst various GR24 concentrations, 0.01 mg L−1 proved to be superior, as it enhanced the photosynthetic rate (9.29%), transpiration rate (0.76%), stomatal conductance (77.5%), total soluble protein (0.55%) and K+ (14.63% in roots; 14.87% in shoots) and Ca2+ (12.63% in roots; 11.48% in shoots) contents under control conditions. Similarly, the leaf turgor potential (Ψp), osmotic potential (Ψs) and free proline, glycinebetaine (GB), superoxide dismutase (SOD), catalase (CAT) and peroxide (POD) contents increased by 58.17, 89.95, 159.04, 101.54, 74.42, 175.68 and 53.62%, respectively, under salt stress conditions. The leaf water potential (Ψw) decreased (−0.14%) and the malondialdehyde (MDA) content increased (16.65%) when treated with the 0.001 mg L−1 GR24 level. Meanwhile, hydrogen peroxide (H2O2) and Na+ concentrations in roots and shoots increased by 62.53%, 74.66% and 38.55% under saline conditions with a GR24 level of 0 mg L−1. Regarding the plant biomass, a GR24 level of 0.01 mg L−1 with salt stress greatly decreased the root (−47.27% and −50.45%) and shoot (−44.79% and −59.42%) fresh and dry weights, respectively, compared to control conditions. These results reveal that exogenously applied GR24 might be an effective way to mitigate the perilous impacts of salt stress in ornamental sunflower production. It is suggested that the use of molecular techniques to study different processes in which GR24 could play a vital part in various commercial floricultural crops is extremely imperative and can open novel horizons for future investigations in this exhilarating field of plant hormones. Objectives: To compare outcomes of laparoscopic surgery versus open surgery for colorectal carcinoma in terms of hospital stay and post-operative complications. Study Design: Cross-sectional analytical study. Place and Duration of Study: Department of General Surgery, Combined Military Hospital, Rawalpindi, from Mar 2020 to Mar 2021. Methodology: We studied a total of 114 patients who were diagnosed with colorectal cancer and met the sample selection criteria. Patients with metastatic disease or previous history of abdominal surgery were excluded from the study. Patients were documented for hospital stay and followed for six months for the development of complications. Data was analyzed by SPSS version 26. Results: We studied a population where 45 (39.5%) were male while 69 (60.5%) were female. The mean age of the sample was 46.96 ± 14.47 years. The study showed that the mean hospital stay was shorter in Group A: 5.40 ± 1.88 days versus 6.59 ± 1.68 days in Group B (p=0.001). Mean blood loss was 65.99 ± 16.41 mL in Group A versus 366.93 ± 95.36 in Group B (p<0.001). The complication rates in Group A and B were 6 (10.5%) and 11 (19.2%), respectively (p=0.18), while the rate of incisional hernia formation was 0 (0%) and 4 (6.9%) (p=0.042). Conclusion: Laparoscopic surgery is superior to open surgery is the management in terms of shorter in-hospital stay and postoperative complications. [ABSTRACT FROM AUTHOR]

Heavy metal pollution has become a serious environmental concern to global biological diversity. The continuous accumulation of toxic metals in plants is affecting the growth, development and reproduction of herbivore insects. In the present study, heavy metal-induced toxicity through tomato, Solanum lycopersicum L., leaves in Hadda beetle, Epilachna vigintioctopunctata (Coccinellidae; Coleoptera), was assessed. Second instar grubs of E. vigintioctopunctata were exposed to two concentrations i.e. 300 gg ml-1 and 100 gg ml-1 of zinc (Zn) and lead (Pb) metals. Experiment was performed using CRD design under controlled conditions (28 °C, 65% R.H. and 16L: 8D h). Feeding indices: relative growth rate (RGR), relative consumption rate (RCR), efficiency of conversion of ingested food (ECI), growth parameters: grub weight gain, grub length, leaf weight loss, and percent survival rate were recorded. Zn showed to have higher feeding indices (RGR, RCR, and ECI) at both concentrations. Pb, on the other hand, showed a significant reduction in RGR due to low consumption rate and poor efficiency of conversion of ingested food at both concentrations. Due to decreased feeding indices, grub growth also reduced in Pb treated insects compared to Zn. Survival rate was significantly decreased in Zn and Pb treated grubs compared to control. Observed effects were more detrimental in insects exposed to Pb. Future studies should focus other herbivore insects as bioindicators of heavy metal accumulation in agricultural ecosystems which will help improving environmental safety guidelines. Purpose: This study delineated the effect of calcium (Ca) and ethylenediaminetetraacetic acid (EDTA) addition on arsenic (As) accumulation and physiological attributes of Vicia faba L.Materials and methods: Two separate experiments were performed. In the first experiment, V. faba seedlings, grown under hydroponic conditions, were exposed to three levels of As (25, 125 and 250 μM) in the presence and absence of three levels of EDTA (25, 125, 250 μM) and calcium (CaCl2: 1, 5 and 10 mM). The effect of EDTA and Ca on As accumulation and physiological attributes of V. faba was assessed by determining As contents in roots and shoot, chlorophyll contents, H2O2 contents, and lipid peroxidation in young and old leaves. In the second experiment, V. faba seeds were grown in As-contaminated sand culture using the same treatment plan.Results and discussion: The accumulation and toxicity of As to V. faba plants increased with increasing As levels in nutrient solution. Arsenic exposure enhanced the production of reactive oxygen species (ROS) in both roots and leaves, which resulted in lipid peroxidation and decreased chlorophyll contents. The presence of both EDTA and Ca, in general, significantly decreased As accumulation by V. faba seedlings, Ca being more effective than EDTA. Both the amendments decreased As-induced reactive oxygen species (ROS) production and lipid peroxidation. In the case of chlorophyll contents, EDTA significantly decreased chlorophyll contents, while Ca significantly increased chlorophyll contents compared to As. The effect of all the treatments was more pronounced in roots than leaves and in young leaves compared to old leaves.Conclusions: It is proposed that EDTA and Ca greatly affect As accumulation and toxicity to V. faba, and the effect varies greatly with their applied levels as well as type and age of plant organs. The germinating seedlings of V. faba may be preferred for risk assessment studies, while transplanting 1-week-old V. faba seedlings to As-contaminated soils can decrease its toxicity. Insecticide exposure can affect honey bees in agro-ecosystems, posing behavioral stresses that can lead to population decline. In this study, insecticide incidence, DNA damage, and antioxidant enzyme activity were studied in Apis florea and A. dorsata honey bee samples collected from insecticide-treated and insecticide-free areas of Punjab, Pakistan. Seven insecticides: chlorpyrifos, dimethoate, imidacloprid, phorate, emamectin, chlorfenapyr, and acetamiprid were detected in seven samples of A. florea and five samples of A. dorsata . In total, 12 samples (22.2%) of honey bees were found positive to insecticide presence out of 54 samples. The most frequently detected insecticide was chlorpyrifos, which was found in four samples (7.4%), with a concentration ranging from 0.01 to 0.05 μg/g and an average concentration 0.03 μg/g. The comet assay or single cell gel electrophoresis assay, a simple way to measure DNA strand breaks in eukaryotic cells, was used to microscopically find damage of DNA at the level of a single cell. Comet tail lengths of DNA in A. florea and A. dorsata samples from insecticide-treated areas were significantly higher ( P  < 0.05) than samples from insecticide-free areas. The highest comet tail length (19.28 ± 2.67 μm) was observed in DNA of A. dorsata from insecticide-treated areas, while the minimum one (3.18 ± 1.46 μm) was noted in A. dorsata from insecticide-free areas. Catalase (CAT) activity did not vary significantly between honey bee samples from insecticide-treated and insecticide-free areas, while glutathione S -transferase (GST) activity showed a significant reduction in response to insecticide exposure. Significant positive correlations were detected between enzyme activity and insecticide concentration in honey bee species from insecticide-treated areas compared with control groups. Toxicity from pesticide exposure at sub-lethal levels after application or from exposure to pesticide residues should not be underestimated in honey bees, as it may induce physiological impairment that can decline honey bees' health.
