Abstract Thermo‐electrochemical cells (TECs) provide a new potential for self‐powered devices by converting heat energy into electricity. However, challenges still remain in the fabrication of flexible and tough gel electrolytes and their compatibility with redox actives; otherwise, contact problems exist between electrolytes and electrodes during stretching or twisting. Here, a novel robust and neutral hydrogel with outstanding stretchability was developed via double‐network of crosslinked carboxymethyl chitosan and polyacrylamide, which accommodated both n‐type (Fe2+/Fe3+) and p‐type ([Fe(CN)6]3−/[Fe(CN)6]4−) redox couples and maintained stretchability (>300%) and recoverability (95% compression). Moreover, poly(3,4‐ethylenedioxythiophene):poly(styrene sulfonate) textile electrodes with porous structure are integrated into gel electrolytes that avoid contact issues and effectively boost the Pmax of n‐ and p‐type thermocell by 76% and 26%, respectively. The optimized thermocell exhibits a quick current density response and is continually fully operational under deformations, which satisfies the working conditions of wearable devices. Multiple thermocells (four pairs) are effectively connected in alternating single n‐ and p‐type cells in series and outputted nearly 74.3 mV at ΔT = 10°C. The wearable device is manufactured into a soft‐pack thermocells to successfully harvest human body heat and illuminate an LED, demonstrating the potential of the actual application of the thermocell devices.

The aim of this research was to investigate the impact of periplocin (PPLN) on oxaliplatin (OXA) resistance in hepatocellular carcinoma (HCC) cells and offer insights for improving clinical treatment of HCC. The IC50 value of HCC cell lines against OXA was detected by the CCK-8 assay, and an OXA-resistant HepG2 cell line (HepG2/OXA) was constructed. THP-1 cells were induced into M1 or M2 macrophages, and M2 macrophage-conditioned medium (M2-CM) was prepared. M1 and M2 macrophage polarization were detected using RT-qPCR and flow cytometry. CCK-8, EdU staining, clone formation assay, flow cytometry, and western blotting were used to assess the proliferation and apoptosis of HepG2/OXA cells treated with PPLN and M2-CM. Additionally, a nude mouse subcutaneous graft tumor model was constructed. PPLN enhanced the sensitivity of HepG2/OXA cells to OXA, reduced their clone-forming ability, and promoted their apoptosis. Notably, PPLN hindered M0 macrophage polarization to M2 macrophages, while M1 polarization remained unaffected. The proliferation-inhibiting and apoptosis-promoting effects of OXA+PPLN on HepG2/OXA cells were significantly attenuated by the addition of M2-CM, suggesting that PPLN improves the OXA sensitivity of HepG2/OXA cells by hindering M2 macrophage polarization. Furthermore, PPLN inhibited M2 macrophage polarization and improved the OXA sensitivity of HepG2/OXA cells in vivo. In conclusion, PPLN inhibited the proliferation of HepG2/OXA cells, promoted their apoptosis, and inhibited M2 macrophage polarization both in vivo and in vitro, which in turn enhanced the OXA sensitivity of HepG2/OXA cells.

Upon the large-scale integration of inverter-interfaced distributed generator (IIDG) into the grid, random faults in the distribution network can lead to momentary and sustained interruptions, significantly impacting system reliability. Although reliability methods have been widely used in distribution network adequacy assessment, using probabilistic method for reliability assessment including system dynamic security need to be investigated. To address this issue, a new method is designed to assess the reliability of distribution network using sequential Monte Carlo simulation. Firstly, the IIDG low-voltage ride-through (LVRT) control strategy is formulated, and the off-grid probability model for IIDG is developed based on the Gaussian distribution. Secondly, the component model is defined to consider the impact of random faults on power quality in security assessment. Following the fault tree analysis method, a protection action probability model was formulated to assess the failure probability of line current differential protection, IIDG anti-islanding protection, and reclosing protection. Finally, the method for momentary fault consequence analysis, based on depth-first search (DFS), and the method for sustained fault consequence analysis, based on the mixed-integer linear programming model, are developed. The study establishes a comprehensive probability reliability assessment framework. The validity of the method is demonstrated on the IEEE RBTS BUS6 F4 system, indicating good scalability.

Introduction: In prior reports, Jie-Du-Tong-Luo (JDTL) was reported to help control insulin secretion and blood glucose in patients with diabetes, while also protecting liver and pancreatic islet cells against injury caused by exposure to high glucose (HG) levels. This study was thus developed to assess the effects of JDTL on HG and palmitic acid (PA)-induced muscle injury and to explore the mechanistic basis for these effects. Methods: A model of muscle injury was established using mouse C2C12 myotubes treated with HG + PA. A proteomics approach was used to assess changes in protein levels following JDTL treatment, after which Western immunoblotting was employed to validate significantly affected pathways. Results: JDTL was able to protect against HG + PA-induced muscle cell injury in this experimental system, altering lipid metabolism and inflammatory activity in these injured C2C12 myotubes. Western blotting suggested that JDTL is capable of activating PI3K/Akt/PPARγ signaling to control lipid metabolism without any corresponding impact on the inflammatory NF-κB pathway. Conclusions: These data highlight the ability of JDTL to protect against HG + PA-induced injury to muscle cells, and suggest that the underlying basis for such efficacy is related to the PI3K/Akt/PPARγ pathway-mediated modulation of lipid metabolism.

BackgroundMalignant pleural effusion (MPE) is a common clinical problem that requires cytological and/or histological confirmation obtained by invasive examination to establish a definitive diagnosis. Radiomics is rapidly evolving and can provide a non-invasive tool to identify MPE.ObjectivesWe aimed to develop a model based on radiomic features extracted from unenhanced chest computed tomography (CT) images and investigate its value in predicting MPE.MethodThis retrospective study included patients with pleural effusions between January 2016 and June 2020. All patients underwent a chest CT scanning and medical thoracoscopy after artificial pneumothorax. Cases were divided into a training cohort and a test cohort for modelling and verifying respectively. The Mann-Whitney U test and the least absolute shrinkage and selection operator (LASSO) were applied to determine the optimal features. We built a radiomics model based on support vector machines (SVM) and evaluated its performance using ROC and calibration curve analysis.ResultsTwenty-nine patients with MPE and fifty-two patients with non-MPE were enrolled. A total of 944 radiomic features were quantitatively extracted from each sample and reduced to 14 features for modeling after selection. The AUC of the radiomics model was 0.96 (95% CI: 0.912-0.999) and 0.86 (95% CI: 0.657~1.000) in the training and test cohorts, respectively. The calibration curves for model were in good agreement between predicted and actual data.ConclusionsThe radiomics model based on unenhanced chest CT has good performance for predicting MPE and may provide a powerful tool for doctors in clinical decision-making.

Soil organic carbon (SOC) is crucial for soil health and quality, and its sequestration has been suggested as a natural solution to climate change. Accurate and cost-efficient determination of SOC and its functional fractions is essential for effective SOC management. Visible near-infrared spectroscopy (vis-NIR) has emerged as a cost-efficient approach. However, its ability to predict whole-profile SOC content and its fractions has rarely been assessed. Here, we measured SOC and its two functional fractions, particulate (POC) and mineral-associated organic carbon (MAOC), down to a depth of 200 ​cm in seven sequential layers across 183 dryland cropping fields in northwest, southwest, and south China. Then, vis-NIR spectra of the soil samples were collected to train a machine learning model (partial least squares regression) to predict SOC, POC, MAOC, and the ratio of MAOC to SOC (MAOC/SOC – an index of carbon vulnerability). We found that the accuracy of the model indicated by the determination coefficient of validation (Rval2) is 0.39, 0.30, 0.49, and 0.48 for SOC, POC, MAOC, and MAOC/SOC, respectively. Incorporating mean annual temperature improved model performance, and Rval2 was increased to 0.64, 0.31, 0.63, and 0.51 for the four carbon variables, respectively. Further incorporating SOC into the model increased Rval2 to 0.82, 0.64, and 0.59, respectively. These results suggest that combining vis-NIR spectroscopy with readily-available climate data and total SOC measurements enables fast and accurate estimation of whole-profile POC and MAOC across diverse environmental conditions, facilitating reliable prediction of whole-profile SOC dynamics over large spatial extents.

The ABCC1 gene belongs to the ATP-binding cassette membrane transporter superfamily, which plays a crucial role in the efflux of various endogenous and exogenous substances. Mutations in ABCC1 can result in autosomal dominant hearing loss. However, the specific roles of ABCC1 in auditory function are not fully understood. Through immunofluorescence, we found that ABCC1 was expressed in microvascular endothelial cells (ECs) of the stria vascularis (StV) in the murine cochlea. Then, an Abcc1 knockout mouse model was established by using CRISPR/Cas9 technology to elucidate the role of ABCC1 in the inner ear. The ABR threshold did not significantly differ between WT and Abcc1−/− mice at any age studied. After noise exposure, the ABR thresholds of the WT and Abcc1−/− mice were significantly elevated. Interestingly, after 14 days of noise exposure, ABR thresholds largely returned to pre-exposure levels in WT mice but not in Abcc1−/− mice. Our subsequent experiments showed that microvascular integrity in the StV was compromised and that the number of outer hair cells and the number of ribbons were significantly decreased in the cochleae of Abcc1−/− mice post-exposure. Besides, the production of ROS and the accumulation of 4-HNE significantly increased. Furthermore, StV microvascular ECs were cultured to elucidate the role of ABCC1 in these cells under glucose oxidase challenge. Notably, 30 U/L glucose oxidase (GO) induced severe oxidative stress damage in Abcc1−/− cells. Compared with WT cells, the ROS and 4-HNE levels and the apoptotic rate were significantly elevated in Abcc1−/− cells. In addition, the reduced GSH/GSSG ratio was significantly decreased in Abcc1−/− cells after GO treatment. Taken together, Abcc1−/− mice are more susceptible to noise-induced hearing loss, possibly because ABCC1 knockdown compromises the GSH antioxidant system of StV ECs. The exogenous antioxidant N-acetylcysteine (NAC) may protect against oxidative damage in Abcc1−/− murine cochleae and ECs.

Abstract Background Clinically, most patients with Kaposi's sarcoma (KS) are male, and several direct and indirect mechanisms may underlie this increased susceptibility in men, Kaposi's sarcoma-associated herpesvirus (KSHV), also known as human herpesvirus 8 (HHV-8), is considered to be the primary etiological agent responsible for KS. Thus, we propose the hypothesis that men are more susceptible to HHV-8 infection, leading to a higher incidence of Kaposi's sarcoma among males. A meta-analysis was conducted to evaluate the association between gender and HHV-8 seropositivity in the general population. Methods A comprehensive literature search was performed using 6 online databases: PubMed, EMBASE, Cochrane library, Web of Science, CNKI, and Wanfang. Studies published before March 15, 2023, were included. Results In all, 33 articles including 41 studies were included in the meta-analysis. In the included adult population. men had a higher risk of HHV-8 infection than did women in adult populations from all over the world (odds ratio [OR]: 1.08, 95% confidence interval [CI]: 1.01–1.15), but no differences were found in child population from all over the world (OR: 0.90, 95% CI: 0.79–1.01). There was a significant difference in HHV-8 seroprevalence between men and women in sub-Saharan Africa (SSA) adult population (OR: 1.15, 95% CI: 1.05–1.26). However, no significant differences were observed in sub-Saharan Africa (SSA) child population (OR: 0.90, 95%CI 0.78–1.03). As for other continents, the results showed no significant difference, such as the Asian population (OR: 1.03, 95%CI: 0.92–1.16). or the European and American populations (OR 1.01, 95%CI 0.87–1.17). Conclusion There was a slight gender disparity for HHV-8 infection in the adult population. Among the adult populations from SSA and globally, men were more likely to be infected with HHV-8 than were women. However, no statistical significance was observed in the child populations from SSA and globally. In the future, the inclusion of more standardized studies may strengthen the results of this study.

Abstract Background Patients with congenital myopathies may experience respiratory involvement, resulting in restrictive ventilatory dysfunction and respiratory failure. Pulmonary hypertension (PH) associated with this condition has never been reported in congenital ryanodine receptor type 1(RYR1)-related myopathy. Case presentation A 47-year-old woman was admitted with progressively exacerbated chest tightness and difficulty in neck flexion. She was born prematurely at week 28. Her bilateral lower extremities were edematous and muscle strength was grade IV−. Arterial blood gas analysis revealed hypoventilation syndrome and type II respiratory failure, while lung function test showed restrictive ventilation dysfunction, which were both worse in the supine position. PH was confirmed by right heart catheterization (RHC), without evidence of left heart disease, congenital heart disease, or pulmonary artery obstruction. Polysomnography indicated nocturnal hypoventilation. The ultrasound revealed reduced mobility of bilateral diaphragm. The level of creatine kinase was mildly elevated. Magnetic resonance imaging showed myositis of bilateral thigh muscle. Muscle biopsy of the left biceps brachii suggested muscle malnutrition and congenital muscle disease. Gene testing revealed a missense mutation in the RYR1 gene (exon33 c.C4816T). Finally, she was diagnosed with RYR1-related myopathy and received long-term non-invasive ventilation (NIV) treatment. Her symptoms and cardiopulmonary function have been greatly improved after 10 months. Conclusions We report a case of RYR1-related myopathy exhibiting hypoventilation syndrome, type II respiratory failure and PH associated with restrictive ventilator dysfunction. Pulmonologists should keep congenital myopathies in mind in the differential diagnosis of type II respiratory failure, especially in patients with short stature and muscle weakness.

Abstract Exciton–polaritons (polaritons) resulting from the strong exciton–photon interaction stimulates the development of novel low-threshold coherent light sources to circumvent the ever-increasing energy demands of optical communications1–3. Polaritons from bound states in the continuum (BICs) are promising for Bose–Einstein condensation owing to their theoretically infinite quality factors, which provide prolonged lifetimes and benefit the polariton accumulations4–7. However, BIC polariton condensation remains limited to cryogenic temperatures ascribed to the small exciton binding energies of conventional material platforms. Herein, we demonstrated room-temperature BIC polariton condensation in perovskite photonic crystal lattices. BIC polariton condensation was demonstrated at the vicinity of the saddle point of polariton dispersion that generates directional vortex beam emission with long-range coherence. We also explore the peculiar switching effect among the miniaturized BIC polariton modes through effective polariton−polariton scattering. Our work paves the way for the practical implementation of BIC polariton condensates for integrated photonic and topological circuits.

Abstract Ferroptosis is a newly identified iron-dependent form of death that is becoming increasingly recognized as a promising avenue for cancer therapy. N6-methyladenosine (m6A) is the most abundant reversible methylation modification in mRNA contributing to tumorigenesis. However, the crucial role of m6A modification in regulating ferroptosis during colorectal cancer (CRC) tumorigenesis remains elusive. Herein, we find that m6A modification is increased during ferroptotic cell death and correlates with the decreased m6A demethylase fat mass and obesity-associated protein (FTO) expression. Functionally, we demonstrate that suppressing FTO significantly induces CRC ferroptotic cell death, as well as enhancing CRC cell sensitivity to ferroptosis inducer (Erastin and RSL3) treatment. Mechanistically, high FTO expression increased solute carrier family 7 member 11 (SLC7A11) or glutathione peroxidase 4 (GPX4) expressions in an m6A-YTHDF2 dependent manner, thereby counteracting ferroptotic cell death stress. In addition, we identify Mupirocin as a novel inhibitor of FTO, and Mupirocin induces CRC ferroptosis and inhibits tumor growth. Clinically, the levels of FTO, SLC7A11, and GPX4, are highly correlated expression in CRC tissues. Our findings reveal that FTO protects CRC from ferroptotic cell death in promoting CRC tumorigenesis through triggering SLC7A11/GPX4 expression.

Abstract Nonlinear transport is a unique functionality of noncentrosymmetric systems, which reflects profound physics, such as spin-orbit interaction, superconductivity and band geometry. However, it remains highly challenging to enhance the nonreciprocal transport for promising rectification devices. Here, we observe a light-induced giant enhancement of nonreciprocal transport at the superconducting and epitaxial CaZrO3/KTaO3 (111) interfaces. The nonreciprocal transport coefficient undergoes a giant increase with three orders of magnitude up to 105 A−1 T−1. Furthermore, a strong Rashba spin-orbit coupling effective field of 14.7 T is achieved with abundant high-mobility photocarriers under ultraviolet illumination, which accounts for the giant enhancement of nonreciprocal transport coefficient. Our first-principles calculations further disclose the stronger Rashba spin-orbit coupling strength and the longer relaxation time in the photocarrier excitation process, bridging the light-property quantitative relationship. Our work provides an alternative pathway to boost nonreciprocal transport in noncentrosymmetric systems and facilitates the promising applications in opto-rectification devices and spin-orbitronic devices.

Highlights The boosted heterogeneous interfaces in titanium nitride (TiN) nanotube/polydimethylsiloxane (PDMS) composite contributed to strong polarization loss relaxation ability. The TiN nanotubes/PDMS composite possessed both good impedance matching behavior and strong dielectric loss ability in wide temperature spectrum. The TiN nanotubes/PDMS composite exhibited excellent EMWA performances (effective absorption bandwidth value of 3.23 GHz and minimum reflection loss value of − 44.15 dB) at the varied temperature from 298 to 573 K.

Cereal foods are the primary source of daily energy, protein, and dietary fiber intake for humans. With the continuous improvement of people's living standards, cereal foods' nutritional value, safety, and food quality have attracted much attention. Cereal metabonomics, as a new and rapidly developing branch of "metabonomics", has made rapid progress in quantifying and analyzing various metabolites. This article reviews the analysis process of metabolomics technology, and its application in the detection of component identification, the identification of flavor substances, the traceability of origin, and the product of new developments in cereal foods, and the development trend of metabonomics in cereal food field has prospected, to provide a reference for the in-depth development and application of cereal foods.

In response to the difficult problem of gas control in soft and low permeability outburst coal seam, gas control technology is proposed to use physical modification hydraulic punching to improve permeability. Theoretical calculations and FLAC3D numerical simulation methods were used to analyze the plastic failure characteristics of coal bodies around holes under different drilling spacing conditions, and the corresponding stress evolution characteristics were quantitatively characterized. After hydraulic punching, the radial stress of coal bodies between holes significantly decreased, and the closer they were to the holes, the greater the reduction; the influence range of tangential stress is equivalent to the range of plastic failure zone; the peak vertical stress showed a significant increase, and as the spacing between boreholes decreased, the vertical stress curve evolved from a bimodal curve to a unimodal curve; it was found that the pore space provides a free surface for coal unloading, which can remove the confining pressure and transfer concentrated stress to the coal between the pores. Subsequently, based on the relationship between gas coal permeability and three-dimensional stress and gas pressure, the evolution characteristics of permeability under different drilling spacing conditions were quantitatively characterized. Compared with the original coal, the permeability of the coal around the holes was greatly increased, which promoted the gas desorption and flow, and determined the reasonable spacing of the holes, and a reasonable drilling spacing was determined. Field experiments have shown that hydraulic punching greatly increases the exposed area of soft coal bodies, provides pressure relief and transparency space for coal bodies, gas flow and concentration is significantly increased in high negative pressure extraction branch pipes, and the pure gas extraction volume fluctuates within the range of 6.20-9.8 m3/min. The gas concentration is above 34%.

Background Myocardial ischemia is a prevalent cardiovascular disorder associated with significant morbidity and mortality. While prompt restoration of blood flow is essential for improving patient outcomes, the subsequent reperfusion process can result in myocardial ischemia–reperfusion injury (MIRI). Mitophagy, a specialized autophagic mechanism, has consistently been implicated in various cardiovascular disorders. However, the specific connection between ischemia–reperfusion and mitophagy remains elusive. This study aims to elucidate and validate central mitophagy-related genes associated with MIRI through comprehensive bioinformatics analysis. Methods We acquired the microarray expression profile dataset (GSE108940) from the Gene Expression Omnibus (GEO) and identified differentially expressed genes (DEGs) using GEO2R. Subsequently, these DEGs were cross-referenced with the mitophagy database, and differential nucleotide sequence analysis was performed through enrichment analysis. Protein–protein interaction (PPI) network analysis was employed to identify hub genes, followed by clustering of these hub genes using cytoHubba and MCODE within Cytoscape software. Gene set enrichment analysis (GSEA) was conducted on central genes. Additionally, Western blotting, immunofluorescence, and quantitative polymerase chain reaction (qPCR) analyses were conducted to validate the expression patterns of pivotal genes in MIRI rat model and H9C2 cardiomyocytes. Results A total of 2719 DEGs and 61 mitophagy-DEGs were identified, followed by enrichment analyses and the construction of a PPI network. HSP90AA1, RPS27A, EEF2, EIF4A1, EIF2S1, HIF-1α, and BNIP3 emerged as the seven hub genes identified by cytoHubba and MCODE of Cytoscape software. Functional clustering analysis of HIF-1α and BNIP3 yielded a score of 9.647, as determined by Cytoscape (MCODE). In our MIRI rat model, Western blot and immunofluorescence analyses confirmed a significant elevation in the expression of HIF-1α and BNIP3, accompanied by a notable increase in the ratio of LC3II to LC3I. Subsequently, qPCR confirmed a significant upregulation of HIF-1α, BNIP3, and LC3 mRNA in the MIRI group. Activation of the HIF-1α/BNIP3 pathway mediates the regulation of the degree of Mitophagy, thereby effectively reducing apoptosis in rat H9C2 cardiomyocytes. Conclusions This study has identified seven central genes among mitophagy-related DEGs that may play a pivotal role in MIRI, suggesting a correlation between the HIF-1α/BNIP3 pathway of mitophagy and the pathogenesis of MIRI. The findings highlight the potential importance of mitophagy in MIRI and provide valuable insights into underlying mechanisms and potential therapeutic targets for further exploration in future studies.

Abstract This study aimed to evaluate the effect of previous surgical termination of pregnancy (STP) on pregnancy outcomes in women undergoing FET cycles of IVF/ICSI. Retrospective cohort study. Reproductive Center of Maternal and Child Health Care Hospital in Lianyungang city. Data were selected from all IVF/ICSI FET cycles performed between January 2014 and December 2020. A total of 761 cycles met the criteria were included in this study. The primary outcome measures were clinical pregnancy and live birth rates. Secondary outcome measures were biochemical pregnancy rate, spontaneous abortion rate, and preterm birth rate. After adjustments for a series of potential confounding factors, the previous STP was an influential factor in reducing FET cycle clinical pregnancy rate compared with women who had not previously undergone STP (OR = 0.614, 95% CI 0.413–0.911, P = 0.016). The effect of the previous STP on the live birth rate was not statistically significant. (OR = 0.745, 95% CI 0.495–1.122, P = 0.159). Also, an increase in the number of previous STPs relative to only 1-time abortion was an independent risk factor in reducing clinical pregnancy rate and live birth rate (OR = 0.399,95% CI 0.162–0.982, p = 0.046; OR = 0.32,95% CI 0.119–0.857, p = 0.023). Previous STP was an independent factor contributing to the decline in FET cycle clinical pregnancy rates.

Abstract Mechanical antennas have garnered considerable attention due to their ability to address the challenges posed by the significant size and high energy consumption associated with traditional electric antennas when operating at low frequencies. Here, a compact and structurally stable mechanical antenna design is presented. The proposed antenna is constructed using cylindrical piezoelectric ceramics, which have dimensions smaller than 1/1000 of the wavelength. The scrutiny of the influence exerted by the antenna feed area and material thickness on the radiation performance was undertaken, followed by an exhaustive discourse on these effects. Experimental measurements demonstrate the practical functionalities of signal coding, transmission, and reception in the low‐frequency communication system. Notably, at a frequency of 288 kHz, a single proposed antenna achieves an effective information transmission distance of 60 m using binary information coding. This remarkable outcome underscores the potential of this design in facilitating the development of portable and cost‐effective wireless communication equipment for low‐frequency applications.

The two-phase titanium alloy Ti–10 V–5Al-2.5Fe-0.1 B was taken as the experimental material, and thermal compression experiments were carried out at a deformation temperature of 770–920 °C and a strain rate of 0.0005–0.5 s−1. An Arrhenius model, a modified Johnson-Cook model, and an improved BP neural network model based on the sparrow search algorithm (SSA-BP) model were established to predict the high temperature rheological stress of the alloy. A comparison of the prediction accuracy of the three models was made. When the partial random data in the rheological curves was used for model building and relatively independent data were used for predicting the rheological stress, the SSA-BP model had higher prediction accuracy, which exhibits the highest mean square correlation coefficient (R2) value of 0.9992 and the lowest root mean square error (RMSE) and average absolute relative error (AARE) values of 1.3031, and 2.0947 %, respectively. The ability of three models to predict the rheological stress for the new process parameters was verified. Results show that the SSA-BP model still has better prediction ability, which exhibits the highest mean square correlation coefficient (R2) value of 0.9720 and the lowest root mean square error (RMSE) and average absolute relative error (AARE) values of 5.0099, and 6.0382 %, respectively. The predicted values of SSA-BP for the rheological stress were used to construct the hot processing map. Results show that the trend of the power dissipation factor (η) value from the hot processing map predicted by SSA-BP can well agree with the microstructure evolution of the alloy.

Due to software and hardware differentiation, the problem of false positives mistakenly identified as network attack behavior caused by inconsistent mimic decision results frequently occurs.Therefore, a mimic decision method based on deep learning was proposed.By constructing an unsupervised autoencoder-decoder deep learning model, the deep semantic features of diverse normal response data were explored from different executions and its statistical rules were analyzed and summarized.Additionally, the offline learning-online decision-making mechanism and the feedback optimization mechanism were designed to solve false positive problem, thereby accurately detecting network attacks and improving target system security resilience.Since statistical rules of normal response data was understood and mastered by deep learning model, the mimic decision results among different executions could remain consistent, indicating that the target system was in a secure state.However, once the target system was subjected to a network attacks, the response data outputted by the different executions was deviated from statistical distribution of deep learning model.Therefore, inconsistent mimic decision results were presented, indicating that the affected execution was under attack and the target system was exposed to potential security threats.The experiments show that the performance of the proposed method is significantly superior to the popular mimic decision methods, and the average prediction accuracy is improved by 14.89%, which is conducive to integrating the method into the mimic transformation of real application to enhance the system’s defensive capability.

Objective(s): The primary gene mutations associated with nasopharyngeal carcinoma (NPC) are located within the phosphoinositide 3-kinase-mammalian target of rapamycin signaling pathways, which have inhibitory effects on autophagy. Compounds that target autophagy could potentially be used to treat NPC. However, autophagy-related molecular targets in NPC remain to be elucidated. We aimed to examine levels of autophagy-related genes, including autophagy-related 4B cysteine peptidase (ATG4B) and gamma-aminobutyric acid (GABA) type A receptor-associated protein-like 1 (GABARAPL1), in NPC cells and explored their potential role as novel targets for the treatment of NPC.Materials and Methods: The mRNA and protein expression of autophagy-related genes were detected in several NPC cells. Levels of GABARAPL1 were modified by either overexpression or knockdown, followed by examining downstream targets using RT-qPCR and western blotting. The role of GABARAPL1 in NPC proliferation and apoptosis was examined by flow cytometry. Furthermore, the role of GABARAPL1 was assessed in vivo using a nude mouse xenograft tumor model. The underlying mechanism by which GABARAPL1 regulated nasopharyngeal tumor growth was investigated.Results: Autophagy-related 4B cysteine peptidase (ATG4B), GABARAPL1, and Unc-51-like kinase 1 (ULK1) were significantly down-regulated in multiple NPC cell lines. Overexpression of GABARAPL1 up-regulated the expression of autophagy-related proteins, decreased the level of hypoxia-inducible factor (HIF)-2α, and induced apoptosis in NPC cells. Importantly, overexpression of GABARAPL1 slowed tumor growth. Western blotting showed that autophagy was activated, and HIF-2α was down-regulated in tumor tissues.Conclusion: HIF-2α, as a substrate for autophagic degradation, may play an interesting role during NPC progression.

Abstract Background Pichia pastoris is a widely utilized host for heterologous protein expression and biotransformation. Despite the numerous strategies developed to optimize the chassis host GS115, the potential impact of changes in cell wall polysaccharides on the fitness and performance of P. pastoris remains largely unexplored. This study aims to investigate how alterations in cell wall polysaccharides affect the fitness and function of P. pastoris, contributing to a better understanding of its overall capabilities. Results Two novel mutants of GS115 chassis, H001 and H002, were established by inactivating the PAS_chr1-3_0225 and PAS_chr1-3_0661 genes involved in β-glucan biosynthesis. In comparison to GS115, both modified hosts exhibited a looser cell surface and larger cell size, accompanied by faster growth rates and higher carbon-to-biomass conversion ratios. When utilizing glucose, glycerol, and methanol as exclusive carbon sources, the carbon-to-biomass conversion rates of H001 surpassed GS115 by 10.00%, 9.23%, and 33.33%, respectively. Similarly, H002 exhibited even higher increases of 32.50%, 12.31%, and 53.33% in carbon-to-biomass conversion compared to GS115 under the same carbon sources. Both chassis displayed elevated expression levels of green fluorescent protein (GFP) and human epidermal growth factor (hegf). Compared to GS115/pGAPZ A-gfp, H002/pGAPZ A-gfp showed a 57.64% higher GFP expression, while H002/pPICZα A-hegf produced 66.76% more hegf. Additionally, both mutant hosts exhibited enhanced biosynthesis efficiencies of S-adenosyl-L-methionine and ergothioneine. H001/pGAPZ A-sam2 synthesized 21.28% more SAM at 1.14 g/L compared to GS115/pGAPZ A-sam2, and H001/pGAPZ A-egt1E obtained 45.41% more ERG at 75.85 mg/L. The improved performance of H001 and H002 was likely attributed to increased supplies of NADPH and ATP. Specifically, H001 and H002 exhibited 5.00-fold and 1.55-fold higher ATP levels under glycerol, and 6.64- and 1.47-times higher ATP levels under methanol, respectively, compared to GS115. Comparative lipidomic analysis also indicated that the mutations generated richer unsaturated lipids on cell wall, leading to resilience to oxidative damage. Conclusions Two novel P. pastoris chassis hosts with impaired β-1,3-D-glucan biosynthesis were developed, showcasing enhanced performances in terms of growth rate, protein expression, and catalytic capabilities. These hosts exhibit the potential to serve as attractive alternatives to P. pastoris GS115 for various bioproduction applications.

Abstract Reservoirs play a crucial role in regulating water availability and enhancing water security. Here, we develop NASA’s Visible Infrared Imaging Radiometer Suite (VIIRS) based Global Water Reservoir (GWR) product, consisting of measurements of reservoir area, elevation, storage, evaporation rate, and evaporation loss for 164 large global reservoirs. The dataset is available at 8-day and monthly temporal resolutions. Since the Moderate Resolution Imaging Spectroradiometer (MODIS) is close to the end of its life, we further evaluated the consistency between MODIS and VIIRS-based GWR to ensure continuity to the 20+ year MODIS GWR product. Independent assessment of VIIRS reservoir storage (8-day) retrievals against in-situ measurements shows an average of R2 = 0.84, RMSE = 0.47 km3, and NRMSE = 16.45%. The evaporation rate has an average of R2 = 0.56, RMSE = 1.32 mm/day, and NRMSE = 28.14%. Furthermore, results show good consistency (R2 ≥ 0.90) between the VIIRS and MODIS-based product components, confirming that long-term data continuity can be achieved. This dataset can provide valuable insights for long-term trend analysis, hydrological modeling, and understanding hydroclimatic extremes in the context of reservoirs.

Abstract Elevated serum ferritin (SF) levels have been associated with poor prognosis in various cancer types, but its impact on nasopharyngeal carcinoma (NPC) remains unclear. This retrospective study analyzed clinical data from 252 non-metastatic NPC patients admitted to Hainan General Hospital between January 2014 and May 2016. SF levels were measured using the chemiluminescence method. Patients were categorized into low, medium, and high-level SF groups based on tertile median SF levels. Survival outcomes were assessed using Kaplan–Meier analysis and Cox regression models. The overall survival rates of the entire patient cohort at 1, 3, 5, and 8 years were 95.2%, 85.7%, 76.2%, and 68.9% respectively. The high-level SF group (SF > 164.00 ng/mL) had significantly worse overall survival (83.1 vs 96.3 months, P = 0.023) and progression-free survival (77.8 vs 93.3 months, P = 0.019) compared to the low-level SF group. Univariate and multivariate analyses confirmed that high SF levels, along with T3/T4 staging and N3 staging, were independent risk factors for poor prognosis. In conclusion, high SF levels are associated with shorter overall survival and progression-free survival in NPC patients.

Abstract Exciton transport in two-dimensional Ruddlesden−Popper perovskite plays a pivotal role for their optoelectronic performance. However, a clear photophysical picture of exciton transport is still lacking due to strong confinement effects and intricate exciton-phonon interactions in an organic-inorganic hybrid lattice. Herein, we present a systematical study on exciton transport in (BA)2(MA) n−1Pb n I3n+1 Ruddlesden−Popper perovskites using time-resolved photoluminescence microscopy. We reveal that the free exciton mobilities in exfoliated thin flakes can be improved from around 8 cm2 V−1 s−1 to 280 cm2V−1s−1 by anchoring the soft butyl ammonium cation with a polymethyl methacrylate network at the surface. The mobility of the latter is close to the theoretical limit of Mott-Ioffe-Regel criterion. Combining optical measurements and theoretical studies, it is unveiled that the polymethyl methacrylate network significantly improve the lattice rigidity resulting in the decrease of deformation potential scattering and lattice fluctuation at the surface few layers. Our work elucidates the origin of high exciton mobility in Ruddlesden−Popper perovskites and opens up avenues to regulate exciton transport in two-dimensional materials.

Abstract Paraneoplastic syndromes occur in cancer patients and originate from dysfunction of organs at a distance from the tumor or its metastasis. A wide range of organs can be affected in paraneoplastic syndromes; however, the pathological mechanisms by which tumors influence host organs are poorly understood. Recent studies in the fly uncovered that tumor secreted factors target host organs, leading to pathological effects. In this study, using a Drosophila gut tumor model, we characterize a mechanism of tumor-induced kidney dysfunction. Specifically, we find that Pvf1, a PDGF/VEGF signaling ligand, secreted by gut tumors activates the PvR/JNK/Jra signaling pathway in the principal cells of the kidney, leading to mis-expression of renal genes and paraneoplastic renal syndrome-like phenotypes. Our study describes an important mechanism by which gut tumors perturb the function of the kidney, which might be of clinical relevance for the treatment of paraneoplastic syndromes.

Abstract The search for new physics beyond the Standard Model is one of the central problems of current high energy physics interest. As the luminosities of current and near-future colliders continue to increase, the search for new physics has increased the requirements for processing large amounts of data. Meanwhile, quantum computing which is rapidly evolving, has great potential to become a powerful tool to help search for new physics signals. Since the k-means algorithm is known to be able to be accelerated with the help of quantum computing, we investigate and propose an event selection strategy based on k-means algorithm to search for new physics signals. Taking the case of tri-photon processes at the muon colliders as an example, the event selection strategy is shown to be effective in helping to search for the signals of dimension-8 operators contributing to anomalous quartic gauge couplings. Compared with traditional event selection strategy, the expected constraints are generally tighter.

Abstract Background Immune checkpoint inhibitors, which have attracted much attention in recent years, have achieved good efficacy, but their use is limited by the high incidence of acquired drug resistance. Therefore, there is an urgent need to develop new immunotherapy drugs. Compound taxus chinensis capsule (CTC) is an oral paclitaxel compound drug, clinical results showed it can change the number of regulatory T cells and T helper cell 17 in peripheral blood. Regulating the balance between regulatory T cells and T helper cell 17 is considered to be an effective anticancer strategy. Paclitaxel and ginsenoside metabolite compound K are the main immunomodulatory components, it is not clear that paclitaxel combined with compound K can inhibit tumor development by regulating the balance between regulatory T cell and T helper cell 17. Methods MTT, EdU proliferation and plate colony formation assay were used to determine the concentration of paclitaxel and compound K. AnnexinV-FITC/PI staining, ELISA, Western Blot assay, Flow Cytometry and Immunofluorescence were used to investigate the effect of paclitaxel combined with compound K on Lewis cell cultured alone or co-cultured with splenic lymphocyte. Finally, transplanted tumor C57BL/6 mice model was constructed to investigate the anti-cancer effect in vivo. Results According to the results of MTT, EdU proliferation and plate colony formation assay, paclitaxel (10 nM) and compound K (60 μM) was used to explore the mechanism. The results of Flow Cytometry demonstrated that paclitaxel combined with compound K increased the number of T helper cell 17 and decreased the number of regulatory T cells, which induced pyroptosis of cancer cells. The balance was mediated by the JAK–STAT pathway according to the results of Western Blot and Immunofluorescence. Finally, the in vivo results showed that paclitaxel combined with compound K significantly inhibit the progression of lung cancer. Conclusions In this study, we found that paclitaxel combined with compound K can activate CD8+ T cells and induce pyroptosis of tumor cells by regulating the balance between regulatory T cells and T helper cell 17. These results demonstrated that this is a feasible treatment strategy for lung cancer.

Abstract The Weibull distribution is used to describe the heterogeneity of rock hydraulics and embedded into the Fish program which is based on the discrete element method. The developed program overcomes the limitation of the Universal Distinct Element Code (UDEC) software regarding the number of parameter groups, which cannot exceed 50. A method for parameter assignment of heterogeneous rocks is proposed together with a method for estimating the initial flow rate value of heterogeneous models. Based on the established heterogeneity calculation model, the influence of block homogeneity, hydraulic aperture homogeneity, and stress on the seepage characteristics is studied. The results indicate that under zero stress conditions, the flow rate is positively correlated with N0.5 showing a strong linear relationship. The linear relationship is gradually enhanced with the increase in the shape parameters. The relationship between the flow rate and shape parameters is logarithmic with a correlation coefficient greater than 0.9654. The relationship between the flow rate and the axial pressure and confining pressure can be described by quadratic and cubic polynomials, respectively, based on which we further discuss the variation characteristics of equivalent hydraulic apertures under the various axial pressures, confining pressures, and shape parameters.

The weld crack leakage due to stress concentration and external load is a significant safety risk in pressure pipelines. Microstructural variations and dynamic propagation lead to unpredictable changes in leakage rate over time and conditions. To address the above problems, a novel framework called OILS-TCN for weld crack pattern recognition and leakage rate prediction is proposed. Firstly, the adaptive threshold optimization algorithm is introduced into the self-organizing incremental neural network to update and increase the crack leakage pattern. Secondly, the depth first search algorithm is combined with the radial basis function neural network to perform online increment labelling of the leakage state. Then, according to the attenuation characteristics of acoustic emission signals, a portable input-attention module is designed to add different weights to the input sequence. Finally, the accurate prediction of leakage rate under different conditions is realized based on the temporal convolutional network. Compared with other advanced methods, the proposed method has obvious advantages in the adaptability and accuracy of pipeline weld crack leakage rate prediction. In addition, the validity and necessity of each part of the framework proposed are discussed based on ablation experiments. The proposed method can predict the leakage rate in real time without modifying the hyperparameters of the model, and can provide a powerful guide for the online monitoring of the leakage AE technology of pressure pipeline in complex systems.

Water plugging by grouting in rock microfracture is a technical challenge in deep underground engineering. In this paper, a novel grouting test system was developed, which can simulate microfracture grouting tests under triaxial stress conditions. A specimen preparation method that can monitor the seepage and diffusion of microfine cement slurry in microfracture was also proposed. Microfracture grouting model tests were conducted on sandstone-like specimens to investigate the effects of confining pressure, roughness, and microfracture aperture on the seepage and diffusion characteristics of sandstone-like specimens. The test results show that microfine cement slurry can effectively seal the microfracture. However, when the microfracture aperture is small or the roughness is large, the slurry is easy to gather and deposit at the inlet, resulting in the phenomenon of ''seepage filtration'', which makes it difficult for the slurry to be injected into the microfracture. The pressure at the measuring point gradually decreases along the seepage and diffusion direction of the slurry, and the decreasing trend increases with the increase of microfracture roughness. The nonlinear relationship between the slurry volume flow rate and the hydraulic gradient can be described by Forchheimer's law. Moreover, both coefficients a and b in Forchheimer's equation show a gradual increase with increasing microfracture roughness. This study can provide a meaningful reference for the development of rock microfracture grouting technology in deep engineering.

A cost-effective approach is urgently needed to utilize molybdenum tailings (MoT), a mining byproduct with rising output. This study explored the preparation of molybdenum tailings collaborative fly ash (FA) geopolymers (MFG) from mechanically activated MoT and FA raw materials. Orthogonal experiments investigated the impacts of various parameters on the compressive strengths of the MFG. The macroscopic properties of MFG with different mixture ratios were evaluated. The microstructures and components of the MFG were characterized using various methods. Fractal dimensions (Ds) of pore structures in the MFG samples were determined. Carbon emissions resulting from the production of MFG geopolymers were calculated and evaluated using Low carbon Assessment. The findings showed that increasing the content of mechanically activated MoT in the MFG system decreased flowabilities and prolonged setting times. The optimal MoT-to-FA ratio for achieving the highest compressive strength of 31.26 MPa was 1:4, resulting in a Si/Al ratio of 2.03 in the MFG system. XRD, TG, SEM-EDS, and FTIR analyses confirmed the primary product of the MFG was a glassy hydrated sodium-aluminum silicate (N-A-S-H) gel. MIP analysis revealed that addition of mechanically activated MoT improved the pore structure of the MFG, leading to smaller pores. Additionally, for MFG composed solely of MoT or FA, the Ds was smaller, and smaller particles of the raw materials also led to smaller Ds values. Conversely, when the raw materials were mixed, larger Ds values were observed, and a closer volume fraction for the two raw materials resulted in larger Ds values. Furthermore, the carbon emission analysis revealed that the carbon emissions of MFG geopolymers were reduced by 45.4 %-64.3 % compared to traditional OPC slurries.

Debris flows have occurred in two adjacent gullies, namely Sedongpu Gully (SDP) and Zelongnong Gully (ZLN), since the 1950s. The most recent event occurred in ZLN in 2020, causing damage to human communities and infrastructure. Despite their close geographical proximity to the Great Bend of the Yarlung Zangbo River, SDP has experienced more frequent and severe events compared to ZLN. This paper aims to compare the evolutionary characteristics of mass movement between these two gullies. A comprehensive review of 12 debris flow events in both gullies from 1950 to 2020 was conducted. A novel method was proposed and verified for quantifying the debris flow mass movement by integrating InSAR, multi-DSMs, optical images, and field investigations. Comparative analysis revealed distinct evolutionary stages between the gullies, with SDP exhibiting mixed hillslope and channelized debris flows, whereas ZLN primarily experienced channelized debris flows. SDP witnessed a decrease in rainfall and temperature triggering debris flows, contrasting with ZLN where both factors increased. These explained the smaller frequency and magnitude of debris flows in ZLN. Overall, this study emphasizes the need for tailored risk prevention and control measures that reflect the specific dynamics of each gully.

Macroscopic deformation and microscopic pores are the main defects affecting the comprehensive performance of laser powder bed fusion (LPBF) parts. The LPBF process involves highly coupled multiple physical mechanisms over a wide time scale, making it difficult to obtain strongly correlated defect characteristics and achieve online monitoring. Thus, this study proposes an acoustic-based macro–micro defect synchronous monitoring technology. It enables monitoring by analysing process information progressively from intra-layer to inter-layer. The research identifies that high energy density leads to deformation defects, with helical scan strategy energy fluctuations exacerbating deformation. A correlation between keyhole pores and high-frequency signal components allows quantitative pore analysis directly from acoustic signals. Furthermore, an intelligent diagnosis model (gn-Res_SC) is proposed, incorporating high-order information interaction and residual structure for in-depth process analysis. The model demonstrates superior performance with accuracy (90.70%), recall (90.70%) and F1-score (90.40%) in LPBF acoustic monitoring signal evaluation.

ABSTRACTIn modern manufacturing industry, in order to adapt to changes in the general environment, the manufacturing industry must improve production efficiency. To this end, this article introduces an improved genetic algorithm based on rule selection to tackle the nondeterministic polynomial hard problem stemming from inventory fibre resources and fibre selection principles in optical cable production. The algorithm aims to maximize inventory score and minimize fibre segmentation rate. It employs a permutation encoding approach to link the genetic algorithm with fibre allocation solutions and applies a self-attention mechanism to determine subset solution weight within each solution. To boost the recombination of favourable gene segments from different chromosomes, a rule optimization strategy is integrated into the crossover operation based on the weights. This operations enhance the algorithm's global search capability and convergence speed. A feasibility repair strategy is then used to inspect and rectify chromosomes, preventing the generation of illegal solutions. The legitimate mutation operation, founded on weight optimization rules, effectively reduces the algorithm's running time by avoiding illegal solutions. By leveraging actual production data from an optical cable manufacturer for simulation, the experimental results confirm the effectiveness of the improved genetic algorithm in addressing the fibre allocation problem. Comparative simulations with the unimproved genetic algorithm and a stepwise greedy algorithm underscore the superiority of the improved genetic algorithm in resolving the fibre allocation problem.

To explore the influence of dynamic impacts on the creep properties of underground pillars, an underground coal sample is selected as the research material, and the mechanical response characteristics under dynamic impact are explored by the split Hopkinson pressure bar and high-speed camera. The results show that (1) when the creep stress level is higher, the initial deformation of coal is greater, and the damage to the sample through long-term creep is more severe. (2) With increasing levels of static stress, the dynamic strength and elastic modulus of the coal sample first increase and then decrease, and the dynamic elastic modulus is positively correlated with the impact velocity. (3) The static stress level determines the process of crack propagation and the degree of fracture, while the external dynamic impact determines the propagation speed of the crack. The results of this research enrich the theory of control systems for the prevention of coal pillar failure and instability and provide a theoretical basis for safe and efficient mining of working faces.

Blackcurrant (Ribes nigrum L.), a member of the Saxifragaceae family, is a perennial deciduous shrub. Seeds, leaves, and fruits contain a large number of bioactive substances, which have high nutritional value and medicinal health functions. They can be widely used as natural resources in the food and pharmaceutical industries (Huo et al. 2011). Blackcurrant is mainly distributed in Heilongjiang, Jilin, Inner Mongolia, Xinjiang, Qinghai, and other places in China. In the alpine region of Heilongjiang Province, the climate is cold, with a large temperature difference between day and night, sufficient sunlight, fertile soil, and high organic matter content, which is conducive to the accumulation of nutrients in the fruit (Zhu et al. 2022). At the same time, blackcurrant has become one of the characteristic and advantageous shrub species in Heilongjiang Province. The breeding and cultivation research on blackcurrant in this province is in its infancy, therefore the germplasm resources are relatively scarce. Currently, there are less than 10 cultivars available in production. Blackcurrant generally has shortcomings such as weak resistance to powdery mildew and poor cold resistance. Mudanjiang Branch of Heilongjiang Academy of Agricultural Sciences has bred four blackcurrant cultivars, ‘Zaofeng’, ‘Heifeng’, ‘Hanfeng’, and ‘Wanfeng’, through hybridization and selection that are used in production. ‘Danjianghei’ (Heilongjiang Provincial Crop Variety Approval Committee 2012046) is a new cultivar approved (registered) by Heilongjiang Provincial Crop Variety Approval Committee in 2012. ‘Danjianghei’ has a large fruit size, high yield, good quality, and resistance to blackcurrant disease characters.

Due to the uncertainty in soil landslide failure mechanisms, lack of early warning systems for soil landslides and adoption of improper excavation configurations, soil landslides accidents triggered by highway excavation in Chinese mountainous areas generally require expensive remedial measures. This paper describes a soil landslide associated with excavation through integrating field reconnaissance and finite element method simulation. According to the obtained results, the adoption of toe excavation and the presence of a silty clay layer are the two main factors contributing to the failure of the soil landslide, and a strong negative correction was observed between the toe excavation and surface displacement and the safety factor of the investigated cut slope; therefore, a four-level early warning system for this excavation-induced soil landslide was established by employing toe excavation and surface displacement thresholds as the warning indicators. Lastly, a preferable excavation configuration was proposed to facilitate excavation designs in similar landslide-prone areas.

Background Rostral ventrolateral medulla (RVLM) neuron hyperactivity raises sympathetic outflow, causing hypertension. MicroRNAs (miRNAs) contribute to diverse biological processes, but their influence on RVLM neuronal excitability and blood pressure (BP) remains widely unexplored. Methods and Results The RVLM miRNA profiles in spontaneously hypertensive rats were unveiled using RNA sequencing. Potential effects of these miRNAs in reducing neuronal excitability and BP and underlying mechanisms were investigated through various experiments. Six hundred thirty‐seven miRNAs were identified, and reduced levels of miR‐193b‐3p and miR‐346 were observed in the RVLM of spontaneously hypertensive rats. Increased miR‐193b‐3p and miR‐346 expression in RVLM lowered neuronal excitability, sympathetic outflow, and BP in spontaneously hypertensive rats. In contrast, suppressing miR‐193b‐3p and miR‐346 expression in RVLM increased neuronal excitability, sympathetic outflow, and BP in Wistar Kyoto and Sprague‐Dawley rats. Cdc42 guanine nucleotide exchange factor (Arhgef9) was recognized as a target of miR‐193b‐3p. Overexpressing miR‐193b‐3p caused an evident decrease in Arhgef9 expression, resulting in the inhibition of neuronal apoptosis. By contrast, its downregulation produced the opposite effects. Importantly, the decrease in neuronal excitability, sympathetic outflow, and BP observed in spontaneously hypertensive rats due to miR‐193b‐3p overexpression was greatly counteracted by Arhgef9 upregulation. Conclusions miR‐193b‐3p and miR‐346 are newly identified factors in RVLM that hinder hypertension progression, and the miR‐193b‐3p/Arhgef9/apoptosis pathway presents a potential mechanism, highlighting the potential of targeting miRNAs for hypertension prevention.

Small cell lung cancer (SCLC) is a fatal tumor type that is prone to drug resistance. In our previous study, we showed that human rhomboid-5 homolog-1 (RHBDF1) was differentially expressed in 5 intrinsic cisplatin-resistant SCLC tissues compared with 5 intrinsic cisplatin-sensitive SCLC tissues by RNA sequencing, which intrigued us. We performed gain- and loss-of-function experiments to investigate RHBDF1 function, bioinformatics analysis, qRT-PCR, western blotting, and immunoprecipitation to elucidate the molecular mechanisms as well as detect RHBDF1 expression in SCLC by immunohistochemistry. We found that RHBDF1 knockdown promoted cell proliferation and cisplatin chemoresistance and inhibited apoptosis in vitro and in vivo. These effects could be reversed by overexpressing RHBDF1 in vitro. Mechanistically, RHBDF1 interacted with YAP1, which increased the phosphorylation of Smad2 and transported Smad2 to the nucleus. Among clinical specimens, the RHBDF1 was a low expression in SCLC and was associated with clinicopathological features and prognosis. We are the first to reveal that RHBDF1 inhibited cell proliferation and promoted cisplatin sensitivity in SCLC and elucidate a novel mechanism through RHBDF1/YAP1/Smad2 signaling pathway which played a crucial role in cisplatin chemosensitivity. Targeting this pathway can be a promising therapeutic strategy for chemotherapy resistance in SCLC.

Semantic segmentation of large-scale point clouds is crucial for advancing smart city infrastructure and supporting autonomous driving technology. However, existing semantic segmentation techniques designed for indoor environments often struggle to adapt to vast outdoor scenes. Moreover, networks for large-scale scenes face challenges such as limited receptive fields and computational complexity, hindering their ability to accurately perceive small target features. To address these challenges, we propose PVCFormer, a novel cross-attention architecture that leverages both point and voxel representations. By feeding concurrently sampled data at varying voxel resolutions into the network, PVCFormer enhances the segmentation of small-scale features while expanding the receptive field. Additionally, the cross-transformer block facilitates better fusion of point and voxel features, and the introduction of CosFormer improves the computational efficiency of the network.Simultaneously, we introduce SYSU9, a new dataset labeled with 9 categories covering an area of over 7 square kilometers, to serve as a benchmark for evaluating point cloud semantic segmentation algorithms. We proposed two model versions, PVCFormer-CA and PVCFormer-SA. PVCFormer-CA achieves an overall accuracy of 92.4 % on SensatUrban, 94.6 % on DALES, and 91.1 % on SYSU9. For semantic segmentation, PVCFormer-CA achieves 61.5 % mIoU on SensatUrban, 73.6 % mIoU on DALES, and 62.4 % mIoU on SYSU9.Our experiments demonstrated promising results in large-scale outdoor point cloud semantic segmentation and introduce novel methodologies leveraging attention mechanisms for handling large-scale point clouds.

Temozolomide (TMZ) is a widely utilized chemotherapy treatment for patients with glioblastoma (GBM), although drug resistance constitutes a major therapeutic hurdle. Emerging evidence suggests that ferroptosis-mediated therapy could offer an appropriate alternative treatment option against cancer cells that are resistant to certain drugs. However, recurrent gliomas display robust ferroptosis resistance, although the precise mechanism of resistance remains elusive. In the present work, we report that proline rich protein 11 (PRR11) depletion significantly sensitizes GBM cells to TMZ by inducing ferroptosis. Mechanistically, PRR11 directly binds to and stabilizes dihydroorotate dehydrogenase (DHODH), which leads to glioma ferroptosis-resistant in a DHODH-dependent manner in vivo and in vitro. Furthermore, PRR11 inhibits HERC4 and DHODH binding, by suppressing the recruitment of E3 ubiquitin ligase HERC4 and polyubiquitination degradation of DHODH at the K306 site, which maintains DHODH protein stability. Importantly, downregulated PRR11 increases lipid peroxidation and alters DHODH-mediated mitochondrial morphology, thereby promoting ferroptosis and increasing TMZ chemotherapy sensitivity. In conclusion, our results reveal a mechanism via which PRR11 drives ferroptosis resistance and identifies ferroptosis induction and TMZ as an attractive combined therapeutic strategy for GBM.

Cold mixed epoxy asphalt mixture (CMEAM) is widely applied on steel deck bridge pavements due to its excellent high temperature and fatigue resistance. However, previous studies investigated above two properties through destructive test or without considering water-damage factor, respectively, which is far away from real servicing condition. To distinguish above two properties of EA-05 and EA-10 (mixture with nominal maximum particle size of 4.75 mm and 9.5 mm), uniaxial static creep tests, dynamic modulus test and four-point bending beam fatigue test were applied in this paper. At the same time, to comprehensively acquire elastic, viscous-elastic and plastic performance of CMEAM, four kinds of constitutive models were also put forward. Results showed that seven-element viscous-elastic-plastic creep model has the highest correlation coefficients which proves there exists plastic deformation in CMEAM. Meanwhile, EA-10 is likely to present viscous-elastic deformation while EA-05 is more likely to show plastic deformation. Dynamic modulus among different gradation type mixture at 40 °C is not so apparent. But with the increase of frequency and testing temperature, this difference would be clear. The middle gradation of EA-10 has the most excellent high temperature stability under dynamic load. In terms of the resistance to fatigue damage of control or freeze-thaw treated samples, EA-05-U(the upper limit of EA-05 gradation type) always takes the first place and has the lowest sensitivity to the change of stress level and water damage.

Pollution of aquatic environment by difenoconazole (DFZ) significantly affects survival and safety of freshwater aquaculture. Due to its anti-inflammatory and antioxidant properties, plant-derived quercetin (QUE) is often used as a dietary supplement. The main purpose of this study was to explore the protective effect of QUE on heart damage caused by exposure to pesticide DFZ and the potential mechanisms of these effects. This study found that QUE could effectively reduce the myocardial enzyme spectrum index of carp after exposure to DFZ, increase the levels of total antioxidant capacity (T-AOC), glutathione (GSH), and catalase (CAT), inhibit the content of malondialdehyde (MDA), and increase the transcription levels of superoxide dismutase (sod), glutathione peroxidase (gsh-px), and cat. By reducing the expression of keap1, QUE increased the transcription levels of nrf2 and nqo-1, reduced the production of reactive oxygen species (ROS) in the heart after exposure to DFZ, and restored the redox homeostasis of the heart. QUE upregulated the expression of anti-inflammatory cytokines il-10 and tgf-β1 levels, downregulated the transcription levels of pro-inflammatory factor inos, il-1β, il-6 and tnf-α, and alleviated the in vivo redox imbalance caused by exposure to DFZ, leading to myocardial interstitial inflammatory cell infiltration. The results showed that QUE inhibited the apoptosis after exposure to DFZ by affecting the expression of apoptotic factors in the mitochondrial apoptotic pathway. In summary, QUE can effectively decrease myocardial injury caused by exposure to DFZ in aquatic environment.
