52. Reliability-based topology optimization based on bidirectional evolutionary structural optimization using multi-objective sensitivity numbers

Abstract: Reliability-based topology optimization (RBTO) is used to obtain an optimal topology satisfying given constraints, as well as to consider uncertainties in design variables. In this study, RBTO was applied to obtain an optimal topology for the inner reinforcement of a vehicle’s hood based on bidirectional evolutionary structural optimization (BESO). A multi-objective topology optimization technique was implemented to obtain the optimal topology for two models with different curvatures while simultaneously considering the static stiffness of bending, torsion, and natural frequency. A performance measure approach (PMA) with probabilistic constraints formulated in terms of the reliability index was employed to evaluate the probabilistic constraints. The optimal topology obtained by RBTO was evaluated and compared to that obtained by deterministic topology optimization (DTO). A more suitable topology was obtained through RBTO than DTO even though the final volume obtained by RBTO was generally slightly greater than that obtained by DTO. The multiobjective optimization technique based on BESO can be applied very effectively with topology optimization for a vehicle’s hood reinforcement.

Abstract: The prevalence of two serotypes of Streptococcus parauberis isolated from the olive flounder, Paralichthys olivaceus, was evaluated in a total of 29 isolates between 2003 and 2010 in Korea. Streptococcus parauberis isolates were divided into two serologically distinct types (serotype 1 and serotype 2), except for one strain (S1091), using an agglutination assay with rabbit antiserum, and serotype 1 was identified as the dominant type (24 of 29 isolates) in this study. To identify the characteristics of the two serotypes of S. parauberis, we conducted a biochemical test using the API 20 Strep kit, a transmission electron microscopy (TEM) assay, sequence analysis of 16S-23S rRNA intergenic spacer region (ISR) and a pathogenicity test. In TEM, both serotypes possessed polysaccharide capsule layers around the cell surface when bacterial cells were treated with a homologous serotype of rabbit antiserum. However, we were unable to discriminate serotype-specific biochemical characteristics and genetic characteristics of 16S-23S rRNA ISR between the two serotypes. In the pathogenicity test, the serotype 1 strains induced significantly higher mortality than the serotype 2 strains in olive flounder when experimentally inoculated via the intraperitoneal route.

55. A Matched Asymptotic Expansion Analysis of Highly Unsteady Porous Media Flows

Abstract: The method of matched asymptotic expansions is used to analyse a mixture of wave and diffusive behaviours governing flow in a saturated porous medium inside an elastic pipe that is suddenly subjected to a large hydraulic gradient at its entrance. At early times and near the entrance, the head is a diffusing wave that can be reduced to the linear and non-linear telegrapher equations for the laminar and partially developed turbulent flows, respectively. At later times, laminar flows are diffusive and partially developed turbulent flows follow a ‘fast diffusion’ behaviour. In the case of fully developed turbulence, flows at later times follow a fast diffusion form which is complicated by advection at extremely high gradients. A matched asymptotic expansion approach is used to match flows at early times and near the entrance, with complementary forms that are away from the entrance and which occur at later times.

Abstract: Differential diagnoses of hepatic nodules include hepatocellular carcinoma, focal nodular hyperplasia, hepatic adenoma, regenerative nodule, focal fatty changes, and hemangioma. However, differentiation of these nodules can often be difficult. Hemangiomas are frequently encountered during ultrasonogram incidentally and can be diagnosed easily because they have an almost distinctive sonographic appearance: a homogeneous hyperechogenicity and discrete posterior acoustic enhancement. They also sometimes have atypical findings, for example an internal echogenicity including hypoechogenicity, heterogeneous echogenicity, hyperechoic rim, central hypoechogenicity due to various changes (e.g., internal hemorrhage, necrosis, thrombosis, myxomatous change, and fibrosis), and (rarely) calcification. We report herein the case of an atypical hemangioma presenting with a hypoechoic peripheral ring, mimicking a hepatic malignancy. To our knowledge, there have been no other reports demonstrating a cavernous hemangioma with a discrete hypoechoic ring and without a pseudocapsule.

58. Gong-ju WTP, the first full-scale membrane plant for drinking water treatment in Korea: the result of detailed design

Abstract: The construction of Gong-ju WTP (30,000 m3/day), the first practical full-scale integrated membrane system in Korea, has been launched after 6 months of pilot plant operation and 10 months of detailed design. Powdered activated carbon process was applied in order to remove taste and odor caused by algae and coagulation process was applied in consideration of reducing membrane fouling in this micro filtration system. Pilot testing concluded that the pressure type of hollow fiber membrane was more suitable for the raw water characteristic and economical aspects than submerged type membrane system. For the best flow distribution of each unit, CFD (computational fluid dynamics) analysis was performed on aspects of pipe arrangement and daily integrity test system was applied to ensure the safety for leakage under the criteria of 5-log removal. In view of the complicated and equipment-intensive nature of this membrane system, a 3D CAD system was utilized to minimize trial and errors of construction in accordance with combination of each equipments rendered by 3D CAD, and also optimised operation and maintenance were verified through the simulation of 3D rendering.

59. Increasing the interval between neoadjuvant chemoradiotherapy and surgery in esophageal cancer: a meta-analysis of published studies

Abstract: The aim of this meta-analysis was to clarify whether a longer interval between the end of neoadjuvant chemoradiotherapy (nCRT) and surgery is associated with better outcomes in esophageal cancer. nCRT followed by surgery is the most common approach for patients with resectable esophageal cancer. Operations are performed within 2-8 weeks after nCRT; however, the optimal interval between nCRT and surgery for esophageal cancer is unknown. We performed a systematic literature search in MEDLINE, EMBASE, the Cochrane Central Register of Controlled Trials, and the Clinical Trials database for studies published between January 2000 and December 2014. Eligible studies were prospective or retrospective studies of esophageal cancer that assessed the effects of intervals longer or shorter than 7-8 weeks between the end of nCRT and surgery. The primary end-points were the overall survival (OS) and pathologic complete response (pCR). Secondary end-points were anastomotic leak, R0 resection, and postoperative mortality rate. A meta-analysis was performed to estimate odds ratios (ORs) using fixed-effect and random-effect models, with Review Manager 5.2. The five studies that met the eligibility requirements included 1,016 patients: 520 in the shorter interval group (≤7-8 weeks) and 496 in the longer interval group (7-8 weeks). The results of our meta-analysis indicate that a longer interval between nCRT and surgery may be disadvantageous for 2-year OS (OR = 1.40, 95% confidence interval [CI]: 1.09-1.80, P = 0.010) and R0 resection rate (OR = 1.71, 95% CI: 1.14-2.22, P = 0.009). The pCR, anastomotic leak rate, and postoperative morbidity were similar in the two groups. A longer interval (more than the standard 7-8 weeks) from the end of preoperative nCRT to surgery did not increase the rate of pCR in esophageal cancer, and the different intervals had similar effects on anastomotic leak rate and postoperative mortality rates. However, the longer interval between nCRT and surgery may be disadvantageous for long-term OS. These results should be validated prospectively in a randomized trial.

Abstract: Jet production rates are measured in p+p and d+Au collisions at sqrt[s_{NN}]=200 GeV recorded in 2008 with the PHENIX detector at the Relativistic Heavy Ion Collider. Jets are reconstructed using the R=0.3 anti-k_{t} algorithm from energy deposits in the electromagnetic calorimeter and charged tracks in multiwire proportional chambers, and the jet transverse momentum (p_{T}) spectra are corrected for the detector response. Spectra are reported for jets with 12p_{T}50 GeV/c, within a pseudorapidity acceptance of |η|0.3. The nuclear-modification factor (R_{dAu}) values for 0%-100% d+Au events are found to be consistent with unity, constraining the role of initial state effects on jet production. However, the centrality-selected R_{dAu} values and central-to-peripheral ratios (R_{CP}) show large, p_{T}-dependent deviations from unity, challenging the conventional models that relate hard-process rates and soft-particle production in collisions involving nuclei.

Abstract: We present the first measurement of elliptic ($v_2$) and triangular ($v_3$) flow in high-multiplicity $^{3}$He$+$Au collisions at $\sqrt{s_{_{NN}}}=200$ GeV. Two-particle correlations, where the particles have a large separation in pseudorapidity, are compared in $^{3}$He$+$Au and in $p$$+$$p$ collisions and indicate that collective effects dominate the second and third Fourier components for the correlations observed in the $^{3}$He$+$Au system. The collective behavior is quantified in terms of elliptic $v_2$ and triangular $v_3$ anisotropy coefficients measured with respect to their corresponding event planes. The $v_2$ values are comparable to those previously measured in $d$$+$Au collisions at the same nucleon-nucleon center-of-mass energy. Comparison with various theoretical predictions are made, including to models where the hot spots created by the impact of the three $^{3}$He nucleons on the Au nucleus expand hydrodynamically to generate the triangular flow. The agreement of these models with data may indicate the formation of low-viscosity quark-gluon plasma even in these small collision systems.

Abstract: We present a systematic study of charged-pion and kaon interferometry in Au + Au collisions at root s(NN) = 200 GeV. The kaon mean source radii are found to be larger than pion radii in the outward and longitudinal directions for the same transverse mass; this difference increases for more central collisions. The azimuthal-angle dependence of the radii was measured with respect to the second-order event plane and similar oscillations of the source radii were found for pions and kaons. Hydrodynamic models qualitatively describe the similar oscillations of the mean source radii for pions and kaons, but they do not fully describe the transverse-mass dependence of the oscillations.

Abstract: The standard model (SM) of particle physics is spectacularly successful, yet the measured value of the muon anomalous magnetic moment (g−2)μ deviates from SM calculations by 3.6σ. Several theoretical models attribute this to the existence of a “dark photon,” an additional U(1) gauge boson, which is weakly coupled to ordinary photons. The PHENIX experiment at the Relativistic Heavy Ion Collider has searched for a dark photon, U, in π0,η→γe+e− decays and obtained upper limits of O(2×10−6) on U−γ mixing at 90% C.L. for the mass range 30

Abstract: The aim of this paper is to establish that there exist suitable objective functions for mathematical programming formulations of dynamic user equilibrium assignment in the sense of Wardrop (1952) . Recently, Lin and Lo (2000) showed that we cannot necessarily calculate a dynamic user equilibrium assignment by minimising the integral over time of Beckmann’s objective function that is widely used to solve for static user equilibrium assignment. In the present paper, two distinct objective functions are presented that result from formulations of assignment either as a variational inequality or as an infinitesimal optimisation: each of these formulations can be solved as a forward dynamic programme. With these formulations and solution approaches, we solve an example based upon Lin and Lo’s one correctly.

Abstract: We report on J/psi production from asymmetric Cu + Au heavy-ion collisions at root S-NN = 200 GeV at the Relativistic Heavy Ion Collider at both forward (Cu-going direction) and backward (Au-going direction) rapidities. The nuclear modification of J/psi yields in Cu + Au collisions in the Au-going direction is found to be comparable to that inAu + Au collisions when plotted as a function of the number of participating nucleons. In the Cu-going direction, J/psi production shows a stronger suppression. This difference is comparable in magnitude and has the same sign as the difference expected from shadowing effects due to stronger low-x gluon suppression in the larger Au nucleus.

66. Hydraulic performance and stability of coarse rockfill deposits

Abstract: Open-pit coal mining operations often generate enormous quantities of waste rock. This is dumped into nearby valleys and the coarsest fraction ends up at the bottom of the deposit; the final result is often referred to as a ‘rock drain’. These ‘flow-through’ rockfill drains can be 100 m high, thousands of metres long, and usually have a stream flowing through their base. When the stream that supplies the rock drain floods, the ability of the drain to convey water is exceeded by its supply, and a large but temporary pool develops at the upstream end of the dump. More importantly, the free surface within the rock drain becomes elevated, as does the point of exit at the downstream face. Because of historic instances of failures at the downstream face, the management of these buried streams and the hydraulic behaviour of the rock drains through which they flow are of interest. The behaviour of homogeneous embankments comprised of coarse rockfill has many similarities to the behaviour of long rockfill drains. Methods for assessing the hydraulic performance of either are presented herein. Definition of the complete water surface ‘envelope’ and the tendency toward unravelling of the downstream face receive primary focus. It is demonstrated that the hydraulic gradient directly beneath the seepage face and small variations in its direction play a far greater role than seepage-face overflow in promoting unravelling failure, and argues that all such deposits are, in effect, embankment dams that should fall under the applicable national dam safety regulations.

Abstract: In order to improve efficiency of a system with three-dimensional flow characteristics, this paper presents a new method that overcomes the computational difficulties associated with three-dimensional effects by using two-dimensional CFD and a neural network. The method was applied to the shape optimization of cut-off in a multi-blade fan/scroll system. As for the entrance conditions of two-dimensional CFD analysis, the experimental values at the positions apart from the inactive zone were used. The distributions of velocity and pressure obtained by two-dimensional CFD analysis were compared with those of three-dimensional CFD analysis and experimental results. It was found that the distributions of velocity and pressure have qualitative similarity. The results of two-dimensional CFD analysis were used for learning as target values of a neural network. The optimal angle and radius of cut-off were determined as 71° and 0.092 times the outer diameter of impeller, respectively. It was concretized in a previous report that the optimal angle and radius of cut-off are approximately 72° and 0.08 times the outer diameter of impeller, respectively.

68. Piribedil and bromocriptine in Parkinson's disease: a single-blind crossover study

Abstract: Introduction – Clinicians switch from one dopamine agonist to another for various reasons. However, each change may inadvertently result in certain potential risks such as decreased medication efficacy or new side-effects. Objective – We evaluated the tolerability of a switch of bromocriptine to piribedil using two conversion ratios as a primary outcome measure, with motor function as a secondary outcome measure, in patients with mild to moderate Parkinson's disease (PD). Methods – Twenty consecutive patients with mild to moderate PD (Hoehn and Yahr, stage II–III) on treatment with stable doses of bromocriptine and levodopa were randomized to two groups of 10 patients each, to receive piribedil based on 1:5 or 1:10 conversion ratios. Blinded evaluations were performed: 1) United Parkinson's Diseased Rating Scale (UPDRS) scores both in ‘on’ and ‘off’, 2) Open-ended interviews for adverse events, 3) Epworth Sleepiness Scale, 4) Purdue Pegboard assessment during ‘on’ and ‘off’, 5) Hand-arm movement test during ‘on’ and ‘off’, and 6) Walking test during ‘on’ and ‘off’. Results – Major adverse events included ‘sleep attacks’ in one patient and minor side-effects included giddiness, nausea, hallucinations, sleepiness and lethargy. However, these were mild and 19 (95%) of the 20 patients completed the study. There was a significant improvement in both the UPDRS ‘off’ total and motor scores at 1 month compared with baseline for the group on 1:10 ratio. The walking times during the ‘off’ state at 1 and 2 months were significantly better compared with baseline in the 1:5 group. There were otherwise no significant differences in the rating tests during both ‘off’ and ‘on’ states before and after the bromocriptine switch. Conclusions – We demonstrated that patients with mild to moderate PD who were on relatively low doses of bromocriptine can be safely switched to piribedil based on a conversion ratio of either 1:5 or 1:10. However, the higher conversion ratio has to be carried out with caution in patients with daytime somnolence.

Abstract: The PHENIX experiment at the Relativistic Heavy Ion Collider has performed a systematic study of K-S(0) and K*(0) meson production at midrapidity in p + p, d + Au, and Cu + Cu collisions at root s(NN) = 200 GeV. The K-S(0) and K*(0) mesons are reconstructed via their K-S(0) -> pi(0)(-> gamma gamma) pi(0)(-> gamma gamma) and K*(0) -> K-+/-pi(-/+) decay modes, respectively. The measured transverse-momentum spectra are used to determine the nuclear modification factor of K-S(0) and K*(0) mesons in d + Au and Cu + Cu collisions at different centralities. In the d + Au collisions, the nuclear modification factor of K-S(0) and K*(0) mesons is almost constant as a function of transverse momentum and is consistent with unity, showing that cold-nuclear-matter effects do not play a significant role in the measured kinematic range. In Cu + Cu collisions, within the uncertainties no nuclear modification is registered in peripheral collisions. In central collisions, both mesons show suppression relative to the expectations from the p + p yield scaled by the number of binary nucleon-nucleon collisions in the Cu + Cu system. In the p(T) range 2-5 GeV/c, the strange mesons (K-S(0), K*(0)) similarly to the phi meson with hidden strangeness, showan intermediate suppression between the more suppressed light quark mesons (pi(0)) and the nonsuppressed baryons (p, (p) over bar). At higher transverse momentum, p(T) > 5 GeV/c, production of all particles is similarly suppressed by a factor of approximate to 2.

Abstract: We present a measurement of the cross section and transverse single-spin asymmetry (AN) for. mesons at large pseudorapidity from root s = 200 GeV p up arrow + p collisions. The measured cross section for 0.5 = 0.061 +/- 0.014. The results are consistent with prior transverse single-spin measurements of forward eta and pi(0) mesons at various energies in overlapping x(F) ranges. Comparison of different particle species can help to determine the origin of the large observed asymmetries in p up arrow + p collisions.

Abstract: The PHENIX experiment at the Relativistic Heavy Ion Collider has measured low-mass vector-meson ,omega, rho, and phi, production through the dimuon decay channel at forward rapidity (1.2 mu mu) = 80 +/- 6(stat) +/- 12(syst)nb and d sigma/dy(phi -> mu mu) = 27 +/- 3(stat) +/- 4(syst)nb. These results are compared with midrapidity measurements and calculations.

73. Early rehabilitation after total knee replacement surgery: a multicenter, noninferiority, randomized clinical trial comparing a home exercise program with usual outpatient care

Abstract: To determine, at 6 weeks postsurgery, if a monitored home exercise program (HEP) is not inferior to usual care rehabilitation for patients undergoing primary unilateral total knee replacement (TKR) surgery for osteoarthritis.We conducted a multicenter, randomized clinical trial. Patients ages 45-75 years were allocated at the time of hospital discharge to usual care rehabilitation (n = 196) or the HEP (n = 194). Outcomes assessed 6 weeks after surgery included the Western Ontario and McMaster Universities Osteoarthritis Index pain and physical function subscales, knee range of motion, and the 50-foot walk time. The upper bound of the 95% confidence interval (95% CI) mean difference favoring usual care was used to determine noninferiority.At 6 weeks after surgery there were no significant differences between usual care and HEP, respectively, for pain (7.4 and 7.2; 95% CI mean difference [MD] -0.7, 0.9), physical function (22.5 and 22.4; 95% CI MD -2.5, 2.6), knee flexion (96° and 97°; 95% CI MD -4°, 2°), knee extension (-7° and -6°; 95% CI MD -2°, 1°), or the 50-foot walk time (12.9 and 12.9 seconds; 95% CI MD -0.8, 0.7 seconds). At 6 weeks, 18 patients (9%) allocated to usual care and 11 (6%) to the HEP did not achieve 80° knee flexion. There was no difference between the treatment allocations in the number of hospital readmissions.The HEP was not inferior to usual care as an early rehabilitation protocol after primary TKR.

74. DNA damage in peripheral blood lymphocytes of ovarian cancer patients after radiotherapy

Abstract: Radiotherapy is a common mode of treatment for many types of cancer, particularly cancers that are not detected until late stages, as is common with ovarian cancer. Although radiotherapy is effective in preferentially killing tumor cells, DNA damage induced by ionizing radiation can also have toxic effects on non-tumor cells. The aim of this study was to investigate the extent of toxicity on non-tumor cells following radiotherapy for ovarian cancer.The authors used the comet assay to assess DNA damage in peripheral blood lymphocytes of 60 ovarian cancer patients undergoing radiotherapy. Venous blood samples were collected from patients before radiotherapy and after accumulated doses of 10, 20, 30, 40, and 50 Gy of radiotherapy.Comet frequencies, reflecting the proportion of damaged cells, were significantly higher after radiotherapy than before radiotherapy (f = 69.66, p0.05) and demonstrated a linear relationship with accumulated dose (y = 9.87 + 0.2987x, r = 0.9497, p0.05). Additionally, the comet tail length, reflecting the relationship between undamaged and damaged DNA, was significantly longer after radiotherapy (f = 175.13, p0.05).These results demonstrate that radiotherapy induces DNA damage in lymphocytes of ovarian cancer patients and suggest that radiotherapy doses should be limited during clinical treatment to reduce toxic side-effects.

Abstract: A rapid and sensitive HPLC method with fluorescence detection has been developed for the micro-sample analysis of bisphenol A in rat serum. Compounds were separated on a Spherisorb C18 analytical column with acetonitrile-0.1% triethylamine in deionized water, 54:46 (v/v), as mobile phase. Bisphenol A was isolated by one-step extraction with dichloromethane. The calibration plot was linear (r=0.999) over the concentration range 1–1000 ng mL−1. The limits of detection and quantitation were 0.2 ng mL−1 and 1 ng mL−1, respectively, for 100-μL serum samples. The assay was successfully used for a pharmacokinetic study after intravenous administration of a low dose (0.1 mg kg−1) of bisphenol A to rats.

Abstract: The magnetoconductivity of two kinds of iodine doped helical polyacetylene, R- and S-polyacetylene, were investigated to understand the effect of the intrafibril and the interfibrillar interaction in the polyacetylene system. The zero-field resistivity ratio, ρr=ρ(1.2 K)/ρ(300 K), is comparable to that of stretch-oriented high-density polyacetylene film, which indicates the partial alignment of chains inside a polymer fiber. At low magnetic fields, the small negative component of magnetoconductivity (positive magnetoresistance) was observed and its magnitude increases as the ρr value increases. In the high field region, the magnetoconductivity is positive and it clearly shows the linear dependence on the magnetic field up to H=30 T. The linear field dependence of magnetoconductivity is different from what is expected in the three-dimensional localization-interaction picture. For the same ρr value samples, the magnitude of negative magnetoconductivity of S-polyacetylene is much bigger than that of R-polyacetylene, which could be attributed to the difference in the degree of helicity determining the strength of interfibrillar interaction.

Abstract: The growth-strain method was used for general two-dimensional shape optimization. It was verified in previous papers that the growth-strain method is very effective for shape optimization of structures with only one free surface to be deformed. But it could not provide reasonable optimized shapes for structures with two or more free surfaces such as structures with holes inside. Problems occurred, as the growth-strain method was applied to structures with two or more free surfaces. Then, an improved method was suggested. Finally, an automatic shape optimization system was built by the improved growth-strain method with commercial software using the finite element method. The effectiveness and practicability of the developed shape optimization system was verified by some examples.

Abstract: We present a reliable method to account for the magnetoresistance of resistance sensors which are used as thermometers in many low temperature (T⩽20 K) experiments carried out in high magnetic fields (to 31 T). To apply the method, a set of isothermal magnetoresistance data, and a zero magnetic field temperature calibration are first necessary. A simple algorithm, which uses this data set, can then be applied to compute the temperature from the measured resistance at any field. The method is particularly useful for temperature dependent measurements at fixed field, or where, in cases where the temperature may change unpredictably during a change in magnetic field. We apply this method to the treatment of data in two separate experiments with the two different thermometers, RuO2 (below 1 K) and Cernox (above 1 K) sensors, respectively.

Abstract: We consider the magnetoresistance oscillation phenomena in the Bechgaard salts (TMTSF)(2)X, where X = ClO4, PF6, and AsF6 in pulsed magnetic fields to 51 T. Of particular importance is the observation of a new magnetoresistance oscillation for X = ClO4 in its quenched state. In the absence of any Fermi-surface reconstruction due to anion order at low temperatures, all three materials exhibit nonmonotonic temperature dependence of the oscillation amplitude in the spin-density-wave (SDW) state. We discuss a model where, below a characteristic temperature T* within the SDW state, a magnetic breakdown gap opens. [S0163-1829(99)00904-2].

Abstract: Customer requirements used in the decision-making at early stage of design process is critically important but easily outdated in ever changing market places due to unstable global economic factors. These uncertain factors that have not been considered in a product design often negatively affect the product market performance at the time point of product introduction. To solve this critical problem, we propose a framework for strategic product design, in which designers may predict product market performance of their own product concepts at a targeted market and time with given levels of product attributes and predicted levels of economic factors. Conceptual design of mid-sized passenger aircraft is employed as a validation example in this work. Trends of customer preferences among three main aircraft attributes (number of seats, range, and speed) affected by oil price and interest rates are modeled based on historical sales data of existing aircraft models for 12 years.

Abstract: The endocannabinoid system is important in the pathogenesis of obesity-related metabolic disorders. However, the effect of inhibiting the endocannabinoid system in type 2 diabetic nephropathy is unclear. Therefore, we examined the effect of the cannabinoid (CB)1 receptor antagonist, SR141716, on insulin resistance and diabetic nephropathy in db/db mice. Six-week-old db/db mice were treated with the CB1-specific antagonist SR141716 (10 mg/kg · d) for 3 months. Treatment with SR141716 significantly improved insulin resistance and lipid abnormalities. Concomitantly, CB1 antagonism improved cardiac functional and morphological abnormality, hepatic steatosis, and phenotypic changes of adipocytes into small differentiated forms, associated with increased adiponectin expression and decreased lipid hydroperoxide levels. CB1 receptor was overexpressed in diabetic kidneys, especially in podocytes. Treatment with the SR141716 markedly decreased urinary albumin excretion and mesangial expansion and suppressed profibrotic and proinflammatory cytokine synthesis. Furthermore, SR141716 improved renal lipid metabolism and decreased urinary 8-isoprostane levels, renal lipid hydroperoxide content, and renal lipid content. In cultured podocytes, high-glucose stimulation increased CB1 receptor expression, and SR141716 treatment abolished high-glucose-induced up-regulation of collagen and plasminogen activator inhibitor-1 synthesis. Additionally, knockdown of CB1 receptor expression by stealth small interfering RNA abolished high-glucose-induced sterol-regulatory element-binding protein-1 expression in podocytes. These findings suggest that CB1 blockade improves insulin resistance and protect against renal injury through both metabolic and antifibrotic effects in type 2 diabetic nephropathy. Targeting CB1 blockade could therefore provide a new therapeutic target to prevent type 2 diabetic nephropathy.

Abstract: The prevalence of two serotypes of Streptococcus parauberis isolated from the olive flounder, Paralichthys olivaceus, was evaluated in a total of 29 isolates between 2003 and 2010 in Korea. Streptococcus parauberis isolates were divided into two serologically distinct types (serotype 1 and serotype 2), except for one strain (S1091), using an agglutination assay with rabbit antiserum, and serotype 1 was identified as the dominant type (24 of 29 isolates) in this study. To identify the characteristics of the two serotypes of S. parauberis, we conducted a biochemical test using the API 20 Strep kit, a transmission electron microscopy (TEM) assay, sequence analysis of 16S-23S rRNA intergenic spacer region (ISR) and a pathogenicity test. In TEM, both serotypes possessed polysaccharide capsule layers around the cell surface when bacterial cells were treated with a homologous serotype of rabbit antiserum. However, we were unable to discriminate serotype-specific biochemical characteristics and genetic characteristics of 16S-23S rRNA ISR between the two serotypes. In the pathogenicity test, the serotype 1 strains induced significantly higher mortality than the serotype 2 strains in olive flounder when experimentally inoculated via the intraperitoneal route.

Abstract: In the past 4 years, incidences of endemic or epidemic respiratory diseases associated with canine influenza H3N2 virus in Asian dogs have been reported in countries such as South Korea and China. Canine species were considered to be the new natural hosts for this virus. However, at the beginning of 2010, influenza-like respiratory signs, such as dyspnoea, were also observed among cats as well as in dogs in an animal shelter located in Seoul, South Korea. The affected cats showed 100 % morbidity and 40 % mortality. We were able to isolate a virus from a lung specimen of a dead cat, which had suffered from the respiratory disease, in embryonated-chicken eggs. The eight viral genes isolated were almost identical to those of the canine influenza H3N2 virus, suggesting interspecies transmission of canine influenza H3N2 virus to the cat. Moreover, three domestic cats infected with intranasal canine/Korea/GCVP01/07 (H3N2) all showed elevated rectal temperatures, nasal virus shedding and severe pulmonary lesions, such as suppurative bronchopneumonia. Our study shows, for the first time, that cats are susceptible to canine influenza H3N2 infection, suggesting that cats may play an intermediate host role in transmitting the H3N2 virus among feline and canine species, which could lead to the endemic establishment of the virus in companion animals. Such a scenario raises a public health concern, as the possibility of the emergence of new recombinant feline or canine influenza viruses in companion animals with the potential to act as a zoonotic infection cannot be excluded.

Abstract: Background/Aims: Endoscopic submucosal dissection (ESD) is commonly used for radical resection of gastric adenoma and mucosal cancer, but there is about 30% of discrepancy rate between the histology of the endoscopic biopsy and that of the resected specimen obtained from the same lesion by ESD. The aim of this study was to clarify the clinical significance of IL-6, VEGF, CRP before ESD. Methods: We investigated the correlation between serum IL-6, VEGF, CRP level and discrepancy rate of gastric neoplastic lesions (10 low-grade dysplasias, 18 high-grade dysplasias, and 25 early gastic cancers). Results: Serum levels of IL-6 in gastric adenoma and mucosal cancer patients were significantly higher than in healthy controls (p＜0.05). Especially, serum IL-6 level of high-grade dysplasia patient was significantly higher than low-grade dysplasia and mucosal cancer patients, and the positive rate, sensitivity, and negative predictive value of serum IL-6 levels were higher in high-grade dysplasia patient compared to low-grade dysplasia patient and mucosal cancer patient. Serum levels of VEGF in patients with gastric adenoma and mucosal cancer were significantly higher than healthy controls (p＜0.01). Serum levels of CRP in patients with mucosal cancer were significantly higher than in the controls (p＜0.05), and the positive rate, sensitivity, and positive predictive value of serum CRP levels were higher in high-grade dysplasia and mucosal cancer patients compared to low-grade dysplasia patient. Conclusions: Serum levels of IL-6, VEGF, and CRP in patients with gastric neoplastic lesions were significantly higher than healthy controls, especially, serum IL-6 level of high grade dysplasia patient was significantly higher than low-grade dysplasia and mucosal cancer patients.

Abstract: Background: Recurrence after percutaneous ethanol ablation (PEA) of benign hepatic and renal cysts has been common, resulting in re-treatment or additional surgery. However, in recent years, a few cases of spontaneous regression of recurrent cysts following PEA have been experienced, which led to the design of this study to evaluate cyst recurrence after PEA and the necessity of additional treatment. Purpose: To evaluate whether the initial recurrence after PEA of benign hepatic, renal, and splenic cysts is true recurrence, and to decide whether additional treatment is needed. Material and Methods: Thirty-nine benign cysts (21 hepatic, 17 renal, and one splenic) were treated with PEA. PEA was performed with injection of 13–900 ml (40–50% of the volume of aspirated fluid) of absolute ethanol into the cysts. For cysts larger than 100 ml, two or more PEAs were given in one session. Ultrasonography was then performed during a period of 12 months with 1–2-month intervals. Results: Two months after PEA, eight cysts (20.5%) regressed completely; another 31 cysts recurred with decreased size. After 6 months, 10 of the recurrent cysts had regressed spontaneously. Another four recurrent cysts regressed after 8 months, and three regressed after 12 months. Hence, 25 out of 39 (64.1%) cysts regressed within 12 months after PEA. The mean regression time of the 25 recurrent cysts was 6.3 months. All recurrent cysts, including the 14 that were lost to complete follow-up, showed gradual decrease overtime. There were no major complications associated with PEA. Conclusion: Initial relapse of a cyst following PEA does not signify true recurrence, but transient, reactive, or inflammatory fluid collections which eventually disappear within several months, and thus does not necessitate additional treatment.

Abstract: Transgenic chickens, ubiquitously expressing a human protein, could be a very useful model system for studying the role of human proteins in embryonic development as well as for efficiently producing pharmaceutical drugs as bioreactors. Human parathormone (hPTH) secreted from parathyroid glands plays a significant role in calcium homeostasis and is an important therapeutic agent for the treatment of osteoporosis in humans. Here, by using a robust replication-defective Moloney murine leukemia virus-based retrovirus vector encapsidated with vesicular stomatitis virus G glycoprotein, we generated transgenic chickens expressing hPTH under the control of a ubiquitous Rous sarcoma virus promoter. The recombinant retrovirus was injected into the subgerminal cavity of freshly laid eggs at the blastodermal stage. After 21 d of incubation, 42 chicks hatched from 473 retrovirus-injected eggs. All 42 living chicks were found to express the vector-encoded hPTH gene in diverse organs, as revealed by PCR and reverse transcription-PCR analysis by using primer pairs specific for hPTH. Four days after hatching, 6 chicks died and 14 chicks showed phenotypic deformities. At 18 wk of age, only 3 G0 chickens survived. They also released the hPTH hormone in their blood and transmitted the hPTH gene to G1 embryos. However, although the embryos were alive at d 18 of incubation, none hatched. An electrochemiluminescence immunoassay further showed that the hPTH expression level was markedly elevated in mammalian cells infected by the retrovirus vector. Thus, we demonstrated that transgenic chickens, expressing a human protein under the control of a ubiquitous promoter, not only could be an efficient bioreactor for the production of pharmaceutical drugs, but also could be useful for studies on the role of human proteins in embryonic development. To our knowledge, this is the first report on the production of a human protein (hPTH) in transgenic chickens under the control of a ubiquitous promoter by using a replication-defective Moloney murine leukemia virus-based retrovirus vector system.

Abstract: Combination therapy with inteferon-alpha and ribavirin is an approved therapy for patients with chronic hepatitis C. However, even with the use of pegylated interferon, response rates are still poor in many difficult-to-treat groups, especially with genotype 1 and high viral loads. Retreatment of these patients remains challenging. Newer combinations are being investigated to optimize chances of attaining a sustained response in these groups. Thymosin alpha 1 is a polypeptide with immunomodulatory properties that has been suggested to increase response rates in patients with chronic hepatitis C. Herein, we describe two cases of retreatment patients with chronic hepatitis C who have failed prior pegylated interferon and ribavirin therapy. They received triple combination therapies of thymosin alpha 1, pegylated interferon and ribavirin and achieved sustained virological responses. These cases support that thymosin-alpha 1 may increase the efficacy of pegylated interferon plus ribavirin in the treatment of non-responders to previous combination therapy.

Abstract: AIMS: The expression of TGF beta-inducible gene h3(beta ig-h3) has been used to assess the biological activity of TGF beta in the kidney. In this study, we investigated whether the urinary concentration of beta ig-h3 is associated with diabetic nephropathy in patients with Type 2 diabetes mellitus. We also evaluated the relationship between the urinary concentration of beta ig-3 and proteinuria and microalbuminuria (AER) in a normal healthy population and in Type 2 diabetes patients. METHODS: Four hundred and seventy-nine Type 2 diabetic patients without non-diabetic kidney diseases and 528 healthy control subjects were enrolled. The study subjects were divided into five groups: a non-diabetic healthy control group with normal ACR (n = 443), a non-diabetic healthy control group with microalbuminuria (n = 85), a normoalbuminuric diabetic group (n = 198), a microalbuminuric diabetic group (n = 155) and an overt proteinuria group (n = 126). Urinary levels of beta ig-h3 were measured by enzyme-linked immunosorbent assay. RESULTS: (i) Urinary excretion of beta ig-h3 was significantly higher in the diabetic groups than in the controls, even in the normoalbuminuric stage (25.02 +/- 8.84 vs. 18.67 +/- 6.56, P = 0.03). In diabetic patients, urinary beta ig-h3 levels increased significantly as diabetic nephropathy advanced (25.02 +/- 8.84 vs. 34.06 +/- 24.55 vs. 169.63 +/- 57.33, P < 0.001). (ii) Proteinuria was found to be significantly correlated with urinary beta ig-h3 (healthy control; r = 0.137, P = 0.019, diabetic patients; r = 0.604, P < 0.001). ACR was also found to be significantly related with urinary beta ig-h3 in diabetic patients (r = 0.383, P = 0.006). (iii) In diabetic patients, urinary beta ig-h3 was significantly related with systolic and diastolic blood pressure (systolic blood pressure: r = 0.436, P = 0.024; diastolic blood pressure, r = 0.365, P = 0.042), total cholesterol and HbA(1c) (cholesterol: r = 0.169, P = 0.03, HbA(1c); r = 0.387, P = 0.044). Logistic regression analyses showed that urinary beta ig-h3 was associated with a significant increase in the risk of microalbuminuria and proteinuria in diabetic patients. CONCLUSIONS: Longitudinal monitoring of urinary beta ig-h3 may improve the likelihood of detecting diabetic nephropathy at an earlier stage and beta ig-h3 could be a sensitive marker of diabetic kidney disease progression.

Abstract: Dendritic cells (DCs) infected with recombinant avipox vectors express the introduced genes and activate antigen-specific T cells. DCs exhibit distinct differentiation-dependent immune functions. Moreover, immature DCs are readily infected by canarypox vectors, but undergo tumor necrosis factor (TNF)-alpha-dependent death, while fewer mature DCs get infected and resist dying. A pilot study was performed using the rhesus macaque system to explore whether immature and mature DCs infected with SIV-recombinant canarypox (vCP180) ex vivo could induce primary virus-specific immune responses in vivo. After subcutaneous (sc) reinjection, functional monocyte-derived DCs migrated to lymph nodes (LNs) within 1-2 days and primed T cells in vivo. This was observed by monitoring dye-labeled DCs in the draining LNs and tetanus toxoid (TT)-specific T cell responses after injection of TT-loaded DCs. DCs from simian immunodeficiency virus (SIV)-naive rhesus macaques were infected with vCP180 (SIVmac142 gag, pol, and env genes), and sc reinjected into donor animals. Low-level SIV-specific T cell proliferation, but little if any interferon (IFN)-gamma production was detected. DCs pulsed with vCP180 in combination with TT and keyhole limpet hemocyanin (KLH) (to activate additional T cells and provide "helper" cytokines) induced SIV-, TT-, and KLH-specific T cell responses, including IFN-gamma responses not seen when vCP180-carrying DCs were used alone. Interleukin (IL)-10 and low-level antibody responses were also observed. This pilot study provides the proof of principle that sc injected ex vivo SIV-recombinant canarypox-infected DCs safely induce low-level SIV-specific immune responses in vivo.

Abstract: Aims Vascular endothelial growth factor (VEGF) has been implicated in the pathogenesis of diabetes mellitus. We determined whether alterations of plasma and urinary VEGF levels are related to diabetic nephropathy in Type 2 diabetic patients. Methods One hundred and seven patients and 47 healthy controls were studied. Study subjects were divided into four groups using urinary albumin-to-creatinine ratio (ACR): a non-diabetic healthy control group (n = 47), a normoalbuminuric diabetic group (n = 37), a microalbuminuric diabetic group (n = 37) and an overt proteinuric diabetic group (n = 33). VEGF levels were measured by enzyme-linked immunosorbent assay. Results (i) Urinary VEGF concentrations were significantly higher in the diabetic groups, even at the normoalbuminuric stage (log VEGF/Cr, normoalbuminuria; 4.33 ± 1.06 vs. control; 3.53 ± 0.79, P = 0.009). Urinary VEGF excretions increased as diabetic nephropathy advanced. (ii) Plasma and urinary VEGF levels were higher in hypertensive diabetic patients than in the normotensive individuals with diabetes. (iii) In those with diabetes, plasma VEGF levels were found to be positively correlated with plasma urea (r = 0.398, P = 0.039) and urinary ACR (r = 0.251, P = 0.044), and urinary VEGF to be positively correlated with urinary ACR (r = 0.645, P
