Abstract

Several types of diseases are caused by the production of antibodies against the body's own red blood cells, white blood cells or platelets. Immune hemolytic anemia occurs when the antibodies directed against red blood cells cause their destruction. Warm antibodies react at body temperature and cold antibodies react at lower temperatures. Hemolytic disease of the newborn occurs when antibodies produced by the mother cross the placenta and attack fetal red blood cells. This usually occurs when there is an Rh blood group incompatibility. Immune thrombocytopenia is a reduction in platelets caused by the production of antibodies against platelet antigens. Patients with idiopathic thrombocytopenic purpura also have circulating immune complexes. Immune neutropenia is a reduction in neutrophils caused by the production of antibodies against neutrophil antigens.

Abstract

The mechanistic basis of biological activity loss of human interleukin-1beta (IL-1beta) upon mutation of Arg127 to Gly was investigated by multiple amino acid substitutions. T-helper cell proliferation assay was used to determine the biological activity of the mutants. The results suggest that the packing of amino acid residues, not their specific properties, is critical to IL-1beta function. Moreover, molecular modeling analyses were conducted to examine the structural features of IL-1beta required for biological activity.

Abstract

Exposure to microorganisms has repeatedly been found to influence development of atopic diseases, such as asthma. Innovative techniques have been developed that can comprehensively characterize microbial communities. The objective of this study was to characterize the home microbiota of asthmatic children utilizing 16S rRNA-based phylogenetic analysis by microarray.In this cross-sectional study, DNA was extracted from home dust and bacterial 16S rRNA genes amplified. Bacterial products were hybridized to the PhyloChip Array and scanned using a GeneArray scanner (Affymetrix, Santa Clara, CA). The Adonis test was used to determine significant differences in the whole microbiome. Welch's t-test was used to determine significant abundance differences and genus-level richness differences.Nineteen homes were included in the analysis (14 asthma and five no asthma). About 1741 operational taxonomic units (OTUs) were found in at least one sample. Bacterial genus richness did not differ in the homes of asthmatics and non-asthmatics (p = 0.09). The microbial profile was significantly different between the two groups (p = 0.025). All the top 12 OTUs with significant abundance differences were increased in homes of asthmatics and belonged to one of the five phyla (p = 0.001 to p = 7.2 × 10(-6)). Nearly half of significant abundance differences belonged to the phylum Cyanobacteria or Proteobacteria.These results suggest that home dust has a characteristic microbiota which is disturbed in the homes of asthmatics, resulting in a particular abundance of Cyanobacteria and Proteobacteria. Further investigations are needed which utilize high-throughput technology to further clarify how home microbial exposures influence human health and disease.

Abstract

In the past, asthma was considered mainly as a childhood disease. However, asthma is an important cause of morbidity and mortality in the elderly nowadays. In addition, the burden of asthma is more significant in the elderly than in their younger counterparts, particularly with regard to mortality, hospitalization, medical costs or health-related quality of life. Nevertheless, asthma in the elderly is still been underdiagnosed and undertreated. Therefore, it is an imperative task to recognize our current challenges and to set future directions. This project aims to review the current literature and identify unmet needs in the fields of research and practice for asthma in the elderly. This will enable us to find new research directions, propose new therapeutic strategies, and ultimately improve outcomes for elderly people with asthma. There are data to suggest that asthma in older adults is phenotypically different from young patients, with potential impact on the diagnosis, assessment and management in this population. The diagnosis of AIE in older populations relies on the same clinical findings and diagnostic tests used in younger populations, but the interpretation of the clinical data is more difficult. The challenge today is to encourage new research in AIE but to use the existing knowledge we have to make the diagnosis of AIE, educate the patient, develop a therapeutic approach to control the disease, and ultimately provide a better quality of life to our elderly patients.

Abstract

Eosinophilia is commonly seen in medical practice and can underlie a variety of medical conditions. Eosinophilia, defined as peripheral blood eosinophil counts greater than 500 per microliter, may vary from mild-severe. Idiopathic hypereosinophilic syndrome has been identified as an unusual cause of moderate to severe eosinophilia. Progress in treatment of this condition has accompanied greater understanding about the basic biology of eosinophils. This review will highlight the approach to patients with eosinophilia, along with associated conditions and syndromes.
