Intestinal ischemia and reperfusion injury is a model system of possible consequences of severe trauma and surgery, which might result into tissue dysfunction and organ failure. Neutrophils contribute to the injuries preceded by ischemia and reperfusion. However, the mechanisms by which intestinal ischemia and reperfusion stimulate and activate circulating neutrophils is still not clear. In this work, we used proteomics approach to explore the underlying regulated mechanisms in Wistar rat neutrophils after ischemia and reperfusion. We isolated neutrophils from three different biological groups; control, sham laparotomy, and intestinal ischemia/reperfusion. In the workflow, we included iTRAQ-labeling quantification and peptide fractionation using HILIC prior to LC-MS/MS analysis. From proteomic analysis, we identified 2,045 proteins in total that were grouped into five different clusters based on their regulation trend between the experimental groups. A total of 417 proteins were found as significantly regulated in at least one of the analyzed conditions. Interestingly, the enzyme prediction analysis revealed that ischemia/reperfusion significantly reduced the relative abundance of most of the antioxidant and pro-survival molecules to cause more tissue damage and ROS production whereas some of the significantly up regulated enzymes were involved in cytoskeletal rearrangement, adhesion and migration. Clusters based KEGG pathways analysis revealed high motility, phagocytosis, directional migration, and activation of the cytoskeletal machinery in neutrophils after ischemia and reperfusion. Increased ROS production and decreased phagocytosis were experimentally validated by microscopy assays. Taken together, our findings provide a characterization of the rat neutrophil response to intestinal ischemia and reperfusion and the possible mechanisms involved in the tissue injury by neutrophils after intestinal ischemia and reperfusion.

That objective reference is necessary for formation of reliable beliefs about the external world is almost axiomatic. However, Condorcet (1785) suggested that purely subjective information--if shared and combined via social interaction--is enough for accurate understanding of the external world. We asked if social interaction and objective reference contribute differently to the formation and build-up of collective perceptual beliefs. In three experiments, dyads made individual and collective perceptual decisions in a two-interval, forced-choice, visual search task. In Experiment 1, participants negotiated their collective decisions with each other verbally and received feedback about accuracy at the end of each trial. In Experiment 2, feedback was not given. In Experiment 3, communication was not allowed but feedback was provided. Social interaction (Experiments 1 and 2 vs. 3) resulted in a significant collective benefit in perceptual decisions. When feedback was not available a collective benefit was not initially obtained but emerged through practice to the extent that in the second half of the experiments, collective benefits obtained with (Experiment 1) and without (Experiment 2) feedback were robust and statistically indistinguishable. Taken together, this work demonstrates that social interaction was necessary for build-up of reliable collaborative benefit, whereas objective reference only accelerated the process but--given enough opportunity for practice--was not necessary for building up successful cooperation. Human communicational interaction can be mediated by a host of expressive means from words in a natural language to gestures and material symbols. Given the proper contextual setting even an everyday object can gain a mediating function in a communicational situation. In this study we used event-related fMRI to study the brain activity caused by everyday material objects when they are perceived as signals. We found that comprehension of material signals activates bilaterally areas of the ventral stream and pars triangularis of the inferior frontal cortex, that is, areas traditionally associated with verbal language and semantics. In addition, we found that right-hemisphere inferior frontal cortex is recruited as a function of the increasing unconventionality of communicative objects. Together these findings support an interpretation of the traditional language areas as playing a more general role across modalities in relation to communicational mediation of social semantic meaning. Binocular rivalry occurs when the eyes are presented with different stimuli and subjective perception alternates between them. Though recent years have seen a number of models of this phenomenon, the mechanisms behind binocular rivalry are still debated and we still lack a principled understanding of why a cognitive system such as the brain should exhibit this striking kind of behaviour. Furthermore, psychophysical and neurophysiological (single cell and imaging) studies of rivalry are not unequivocal and have proven difficult to reconcile within one framework. This review takes an epistemological approach to rivalry that considers the brain as engaged in probabilistic unconscious perceptual inference about the causes of its sensory input. We describe a simple empirical Bayesian framework, implemented with predictive coding, which seems capable of explaining binocular rivalry and reconciling many findings. The core of the explanation is that selection of one stimulus, and subsequent alternation between stimuli in rivalry occur when: (i) there is no single model or hypothesis about the causes in the environment that enjoys both high likelihood and high prior probability and (ii) when one stimulus dominates, the bottom-up, driving signal for that stimulus is explained away while, crucially, the bottom-up signal for the suppressed stimulus is not, and remains as an unexplained but explainable prediction error signal. This induces instability in perceptual dynamics that can give rise to perceptual transitions or alternations during rivalry. (Contains 5 figures.)

FAT/CD36 is a transmembrane protein that is thought to facilitate cellular long-chain fatty acid uptake. However, surprisingly little is known about the localization of FAT/CD36 in human skeletal muscle. By confocal immunofluorescence microscopy, we demonstrate high FAT/CD36 expression in endothelial cells and weaker but significant FAT/CD36 expression in sarcolemma in human skeletal muscle. No apparent intracellular staining was observed in the muscle cells. There are indications in the literature that caveolae may be involved in the uptake of fatty acids, possibly as regulators of FAT/CD36 or other fatty acid transporters.We show that in sarcolemma, FAT/CD36 colocalizes with the muscle-specific caveolae marker protein caveolin-3, suggesting that caveolae may regulate cellular fatty acid uptake by FAT/CD36. Furthermore, we provide evidence that FAT/CD36 expression is significantly higher in type 1 compared with type 2 fibers, whereas caveolin-3 expression is significantly higher in type 2 fibers than in type 1 fibers.

Exposure of cells to the diarrhetic shellfish poison, okadaic acid, leads to a dramatic reorganization of cytoskeletal architecture and loss of cell-cell contact. When cells are exposed to high concentrations of okadaic acid (100–500 nM), the morphological rearrangement is followed by apoptotic cell death. Okadaic acid inhibits the broad acting Ser/Thr protein phosphatases 1 and 2A, which results in hyperphosphorylation of a large number of proteins. Some of these hyperphosphorylated proteins are most likely key players in the reorganization of the cell morphology induced by okadaic acid. We wanted to identify these phosphoproteins and searched for them in the cellular lipid rafts, which have been found to contain proteins that regulate cytoskeletal dynamics and cell adhesion. By using stable isotope labeling by amino acids in cell culture cells treated with okadaic acid (400 nM) could be combined with control cells before the isolation of lipid rafts. Protein phosphorylation events and translocations induced by okadaic acid were identified by mass spectrometry. Okadaic acid was shown to regulate the phosphorylation status and location of proteins associated with the actin cytoskeleton, microtubules and cell adhesion structures. A large number of these okadaic acid-regulated proteins have previously also been shown to be similarly regulated prior to cell proliferation and migration. Our results suggest that okadaic acid activates general cell signaling pathways that induce breakdown of the cortical actin cytoskeleton and cell detachment.

The aim of this study was to, from a holistic perspective, describe the effects of a forage-only feeding system and a conventional training program on young Standardbred horses and compare data with similar observations from the literature. Sixteen Standardbred colts fed a forage-only diet for 4 months from breaking (August to December) and with the goal to vigorously trot 5 to 7 km at a speed of 5.6 m/s (3 min/km) were studied. The horses were fed grass haylage (56 to 61% dry matter (DM), 2.80 to 3.02 Mcal DE/kg DM and 130 to 152 g CP/kg DM) ad libitum, 1 kg of a lucerne product and minerals. The amount of training and number of training sessions were documented daily, and feed intake and body development were measured once every month. Heart rate (HR) was measured during and after a standardized exercise test in October and December. In December, a postexercise venous blood sample was collected and analyzed for plasma lactate concentration. Muscle biopsies (m. gluteus medius) were taken and analyzed for glycogen and fiber composition. Health was assessed in October and November by an independent veterinarian using a standardized health scoring protocol. BW and height at withers increased from 402 to 453 kg (root mean square error (RMSE) 6) and from 148.7 to 154.1 cm (RMSE 0.7), respectively, and the body condition score was 4.9 (RMSE 0.2) at the end of the study. Muscle glycogen content was 532 mmol/kg dry weight (s.d. 56). There was a significant decrease in postexercise HR (81 v. 73 bpm, RMSE 8), and the individual amount of training was negatively correlated with HR during and after exercise. The restrictions on the use of antibiotic and anthelmintic treatments in organic pig farming necessitate alternative non-medical control strategies. Therefore, the antibiotic and parasite-reducing effect of a fructan-rich (prebiotic) diet of dried chicory was investigated in free-ranging piglets. Approximately half of 67 piglets from nine litters were experimentally infected with Ascaris suum and Trichuris suis in the suckling period (1 to 7 weeks of age) and 58 of the piglets were challenged daily with Eschericia coli O138:F8 for 9 days after weaning to induce weaning diarrhoea. The litters were fed either chicory (30% dry matter) or a control diet. The effect of chicory on intestinal helminths, intestinal microbiota, especially Bifidobacteria and Campylobacter spp. and E. coli post-weaning diarrhoea was assessed. The weight gain of the piglets was not impaired significantly by chicory. The intestinal A. suum worm burden was reduced by 64% (P = 0.034) in the chicory-fed piglets, whereas these same piglets had 63% more T. suis worms (P = 0.016). Feeding with chicory elicited no changes among the main bacterial groups in ileum according to terminal restriction fragment length polymorphism analysis. However, the terminal-restriction fragment (T-RF) 208 bp, which may belong to Lachnospiraceae, was stimulated by the chicory feed (P = 0.03), and T-RF 370 bp that matches Enterobacter belonging to the Enterobacteria was reduced (P = 0.004). In addition, chicory increased the level of Bifidobacteria (P = 0.001) and the faecal Campylobacter excretion level was transitorily reduced in chicory-fed piglets at 7 weeks of age (P = 0.029). Unfortunately, it was not possible to assess the effect of chicory on post-weaning diarrhoea as it did not develop. In conclusion, feeding piglets chicory around the time of weaning caused complex changes of the microbiota and parasite communities within the intestinal tract, and feeding piglets chicory may therefore serve as an animal-friendly strategy to control pathogens.

The mathematical framework of superbundles as pioneered by D. Quillen suggests that one consider the Higgs field as a natural constituent of a superconnection. I propose to take as superbundle the exterior algebra obtained from a Hermitian vector bundle of rank n where n=2 for the electroweak theory and n=5 for the full Standard Model. The present setup is similar to but avoids the use of non-commutative geometry.
