Foram confeccionados 90 blocos cerâmicos: 60 blocos de silicato de lítio reforçado por zircônia – Celtra Duo (Dentisply-Sirona, Bensheim, Alemanha) e 30 em dissilicato de lítio - IPS e.max CAD (Ivoclar Vivadent Ltda, Schaan, Liechtenstein), nas dimensões 13 x 15 x 1,5 mm. As amostras foram divididas em 6 grupos, com e sem envelhecimento, de forma randomizada: Celtra S – Celtra sem queima; Celtra C – Celtra com queima; E – IPS E.max CAD; Celtra SE – Celtra sem queima e com envelhecimento; Celtra CE – Celtra com queima e com envelhecimento; EE – IPS E.max CAD com envelhecimento. A superfície das cerâmicas foram condicionadas com ácido fluorídrico 5% durante 30 segundos (Celtra Duo) e 20 segundos (IPS e.max), em seguida foi feita a aplicação do agente de união silano Monobond-N (Ivoclar Vivadent, Schaan, Liechtenstein). Para cada bloco, foram confeccionados 6 cilindros de 1 mm de diametro e 2 mm de altura de cimento resinoso para o teste de microcisalhamento. O teste de microcisalhamento foi realizado na máquina de ensaio universal DL 1000 (EMIC, São José dos Pinhais, Paraná, Brasil), com célula de carga de 50 kgf e velocidade de 0,5 mm/min. Os tipos de falhas foram analisados atraves do estereomicroscópico (Microscopia óptica ZEISS MC 80 DX) e microscopio eletrônico de varredura (Jeol-JSM-T330A- Scanning Microscope-Japan). Foi realizado o teste de rugosidade (Ra e Rz) e microscopia eletrônica de varredura (MEV) das superfícies cerâmicas. Os valores de resistência de união (n=10) e rugosidade (n=10) foram submetidos à análise estatística descritiva (média e desvio padrão) e inferencial, mediante o teste paramétrico ANOVA 2-fatores, seguido do teste de Tukey (α=0,05). O teste de microcisalhamento indicou que a condição sem cristalização apresentou resultados superiores aos demais grupos onde mostrou diferença estatisticamente significante (p=0,000). O envelhecimento hidrotérmico foi responsável pela diminuição dos valores de resistência de união para todos os grupos estudados. Conclui-se que o processo de queima de cristalização e degradação hidrotérmica geraram dimunição da resistência adesiva entre esses dois substratos. This study evaluated the influence of the crystallization firing process and hydrothermal degradation on the bond strength between lithium (di)silicatebased glass-ceramics and resin cement. Ninety ceramic blocks were made: 60 blocks of lithium silicate reinforced by zirconia – Celtra Duo (Dentisply-Sirona, Bensheim, Germany) and 30 blocks of lithium disilicate - IPS e.max CAD (Ivoclar Vivadent Ltd, Schaan, Liechtenstein), in dimensions 13 x 15 x 1.5 mm. The samples were randomly divided into 6 groups, with and without aging: Celtra S – Celtra without burning; Celtra C – Celtra with firing; E – IPS E.max CAD; Celtra SE – Celtra without burning and with aging; Celtra CE – Celtra with burning and aging; EE – IPS E.max CAD with aging. The ceramic surfaces were etched with 5% hydrofluoric acid for 30 seconds (Celtra Duo) and 20 seconds (IPS e.max), then the application of the Monobond-N silane bonding agent (Ivoclar Vivadent, Schaan, Liechtenstein) . For each block, 6 cylinders of 1 mm in diameter and 2 mm in height of resin cement were made for the microshear test. The microshear test was performed in a universal testing machine DL 1000 (EMIC, São José dos Pinhais, Paraná, Brazil), with a load cell of 50 kgf and a speed of 0.5 mm/min. The types of faults were analyzed through stereomicroscopic (optical microscope ZEISS MC 80 DX) and scanning electron microscope (Jeol-JSM-T330A-Scanning Microscope-Japan). The roughness test (Ra and Rz) and scanning electron microscopy (SEM) of the ceramic surfaces were performed. The bond strength (n=10) and roughness (n=10) values were subjected to descriptive (mean and standard deviation) and inferential statistical analysis using the 2-factor ANOVA parametric test, followed by Tukey's test (α= 0.05). The microshear test indicated that the condition without crystallization presented better results than the other groups, where it showed a statistically significant difference (p=0.000). Hydrothermal aging was responsible for the decrease in bond strength values for all groups studied. It is concluded that the crystallization firing process and hydrothermal degradation generated a decrease in the bond strength between these two substrates.

No. of bitstreams: 0 Previous issue date: 2021-01-01 Systemic scleroderma is a relatively rare, autoimmune, chronic, and debilitating disease, and its etiology is unknown.The disease is characterized by vasomotor disturbances causing skin and mucosa hardening, hand stiffness, excess production of collagen fibers, and impairment of the internal organs associated with the immune system. Extreme mouth-opening limitation is one of the symptoms in the oral cavity, and compromises dental intervention. One of the fundamental steps when performing a dental procedure is dental arch impression. In the presented clinical case, limited mouth opening hindered the impression accuracy. The objective was to present an alterna-tive impression technique for a patient affected by scleroderma, with limited mouth opening and the need for oral rehabilitation with a removable partial prosthesis in the maxillary arch. An individualized tray was developed with design characteristics that allowed access to the oral cavity and removal, after which it was possible to perform the oral impression, reproducing the areas of interest without causing injury to the oral tissues.The orientation plan was elaborated after obtaining the cast. The teeth were assembled, tested in the mouth, and sent to the laboratory for acrylization. The prosthesis was placed once ready. The alternative impression technique enabled oral rehabilitation of the patient despite the mouth-opening limitation, and provided an accurate impression and a good prosthetic result. 1244389). Purpose Disappointing results from clinical studies assessing the efficacy of therapies targeting vascular endothelial growth factor (VEGF) for the treatment of pterygia suggest that other angiogenic mediators may also play a role in its development. We therefore explore the relative contribution of VEGF, hypoxia-inducible factor (HIF)-1α (the transcription factor that regulates VEGF expression in ocular neovascular disease), and a second HIF-regulated mediator, angiopoietin-like 4 (ANGPTL4), to the angiogenic phenotype of pterygia. Methods Expression of HIF-1α, VEGF, and ANGPTL4 were examined in surgically excised pterygia, and in immortalized human (ih) and primary rabbit (pr) conjunctival epithelial cells (CjECs). Endothelial cell (EC) tubule formation assays using media conditioned by ihCjECs in the presence or absence of inducers/inhibitors of HIF-1 or RNA interference (RNAi) targeting VEGF, ANGPTL4, or both were used to assess their relative contribution to the angiogenic potential of these cells. Results HIF-1α and VEGF expression were detected in 6/6 surgically excised pterygia and localized to CjECs. Accumulation of HIF-1α in was confirmed in ihCjECs and prCjECs, including stratified prCjECs grown on collagen vitrigel, and resulted in expression of VEGF and the promotion of EC tubule formation; the latter effect was partially blocked using RNAi targeting VEGF mRNA expression. We demonstrate expression of a second HIF-regulated angiogenic mediator, ANGPTL4, in CjECs in culture and in surgically excised pterygia. RNAi targeting ANGPTL4 inhibited EC tubule formation and was additive to RNAi targeting VEGF. Conclusions Our results support the development of therapies targeting both ANGPTL4 and VEGF for the treatment of patients with pterygia.

Engenharia de Energia O presente trabalho apresenta uma análise de confiabilidade de sistemas de energia elétrica (SEEs) por intermédio da utilização de métodos probabilísticos. As principais metodologias utilizadas foram as de Reserva Estática, Monte Carlo Sequencial e Markoviana. Foi pesquisado o impacto ocasionado pela inserção de energias renováveis – usinas eólicas e hídricas – e de termelétricas à carvão no desempenho do SEE, assim como analisou-se a repercussão do nível de imprevisibilidade do perfil de carga na confiabilidade geral do sistema. O estudo só foi possível devido à confecção, pelo autor, de um código de simulação na plataforma MATALB. This paper presents a reliability analysis of electrical energy systems (SEEs) through the use of probabilistic methods. The main methodologies used were Static Reserve, Monte Carlo Sequencial and Markoviana. It was investigated the impact caused by the insertion of renewable energies - wind and hydroelectric plants - and of coal-fired power plants in the performance of the ESS, as well as the repercussion of the level of unpredictability of the load profile in the general reliability of the system. The study was only possible due to the creation, by the author, of a simulation code in the MATALB platform.

We present a case of a patient with resolved central serous chorioretinopathy (CSC) in the left eye with persistent mild visual alterations 4 years after the resolution of the disease. Left eye exam revealed a best corrected visual acuity of 20/25 and a slight change of macular pigmentation. Optical coherence tomography revealed only minor irregularities in the topography of retinal pigment epithelium and Bruch's membrane. Adaptive optics (AO) optics demonstrated lower density, spacing, and changes in the photoreceptor mosaic pattern in the left eye than in the right eye, suggesting that CSC may cause damage to cones after clinical recovery. We conclude that AO can provide additional information to assist in the treatment and follow-up of patients with CSC or other macular pathologies. RESUMO Apresentamos o relato de caso de paciente com coriorretinopatia serosa central (CSC) cicatrizada em olho esquerdo e queixa de discreta alteração visual, mesmo após quatro anos da resolução do quadro. O exame do olho esquerdo apresenta melhor acuidade visual corrigida de 20/25 e discreta alteração de pigmentação macular. Tomografia de coerência óptica (OCT) apresentou apenas pequenas irregularidades em topografia de EPR e Bruch. Foi realizado exame com Adaptive Optics (AO), evidenciando valores inferiores de densidade, espaçamento e alterações no padrão de mosaico dos fotorreceptores em olho esquerdo quando comparado com olho direito, sugerindo que a CSC pode causar danos em cones, mesmo após uma recuperação considerada satisfatória. Concluímos que o AO é uma tecnologia que traz novas informações para auxiliar o tratamento e seguimento dos pacientes com CSC ou outras patologias maculares.

We report retinal functional and structural changes of a 40-year-old man diagnosed with occult macular dystrophy. Comprehensive ophthalmological evaluation was performed, followed by spectral-domain optical coherence tomography (SD-OC - Heidelberg) and image acquisition using an adaptive optics (AO) camera (RTX1, Imagine Eyes) for photoreceptor density analysis. Functional tests included full-field ERG (ERG) and multifocal electroretinography (mfERG) (Diagnosys, LLC) and microperimetry with scanning laser ophthalmoscope (SLO) fixation controlled (MAIA, CenterVUE). OCT revealed a line of discontinuity corresponding to cone outer-segment photoreceptors associated with a loss of cone density, highlighted by a dark blue spot on the AO co ne-density map on the fovea in both eyes. Loss of central sensitivity was revealed using microperimetry; ERG was within the normal range, although the mfERG showed a reduced central response amplitude. RESUMO Relatamos exames de função e estrutura retiniana de paciente masculino, de 40 anos, com diagnóstico clínico de Distrofia Macular Oculta (DMO). Avaliação oftalmológica completa foi seguida por tomografia de Coerência Óptica (SD-OCT - Heidelberg) e exame com câmara de fundo de olho com tecnologia "Adaptive Optics" (AO - RTX1, Imagine Eyes) para análise da densidade de fotorreceptores. Os exames funcionais incluíram: Eletroretinografia de campo total (ERG) e multifocal (mfERG) (Diagnosys - LLC) e microperimetria com controle de fixação (MAIA - CenterVUE). Os exames revelam descontinuidade da camada de fotorreceptores na região central da fóvea em ambos os olhos pelo SD-OCT em associação com perda de densidade no mosaico cones, representado por mancha azulada no mapa do AO. Os exames de função apresentam diminuição da acuidade visual (20/80; 20/50), redução de sensibilidade central na microperimetria. Como esperado, o ERG está dentro da normalidade, mas há redução da amplitude das respostas centrais do mfERG em ambos os olhos.

Vinte e um novilhos (n=11) e novilhas (n=10) foram alocados em baias individuais no d -15 e receberam água, mistura de sais minerais e vitaminas, e feno de alfafa ad libitum até d 21. No d -1, os animais foram classificados por sexo e peso corporal (PC) e designado para um dos três tratamentos a seguir: 1) Administração via oral de Meloxicam (1mg/Kg PC, diariamente; Carlsbad Technologies Inc., Carlsbad, CA, EUA) do d -1 até d 6 (MEL8); 2) Administração via oral de Meloxicam (1mg/Kg PC; Carlsbad Technologies Inc., Carlsbad, CA, EUA) no d 0 e de monoidrato de lactose (1mg/Kg PC; Avantor Performance Materials, Center Valley, PA, EUA) no d -1 e do d 1 até d 6 (MEL1); ou 3) Administração via oral de monoidrato de lactose (1mg/Kg PC, diariamente; Avantor Performance Materials, Center Valley, PA, EUA) do d -1 até d 6 (CON). Para o Experimento 1, no d 0, os animais receberam infusão intravenosa de LPS (0,5 μg/Kg PC; Escherichia coli 0111:B4, Sigma-Aldrich, St. Louis, MO, EUA) simultaneamente à administração do tratamento. Temperatura retal (RTEMP) foi medida, e amostras de sangue coletadas nas horas −2, 0, 2, 4, 6, 8, 12, 16, 24, 36, 48, 60, 72, 96, 120, e 144 h relativa à administração de LPS. Não foram detectados efeitos de tratamentos (P ≥ 0,36) para RTEMP, concentração sérica de Fator de Necrose Tumoral Alfa (TNFα), concentrações plasmáticas de haptoglobina, cortisol, insulina e leptina, assim como para expressão de RNAm sanguíneo para TNFα e ciclooxigenase-2 (COX2), contudo todas as variáveis se elevaram (P < 0,01) dentre os tratamentos, após administração de LPS. No Experimento 2, os animais receberam os mesmos tratamentos para os quais foram designados no Exp. 1, dos dias d 7 até d 13 e foram vacinados contra patógenos respiratórios juntamente com a administração dos tratamentos no d 8. Amostras de sangue foram coletadas e RTEMP registradas nos mesmos momentos realizados no Exp. 1 com adição das horas 168, 240 e 336 h relativas ao momento da vacinação. Nenhum efeito de 27 tratamento foi detectado (P ≥ 0,26) para RTEMP, nem para nenhumas das mesmas concentrações plasmáticas e séricas avaliadas no Exp. 1, e concentrações séricas de anticorpos contra Mannheimia haemolytica ou títulos contra Vírus Sincicial Bovino, Herpesvirus-1, Diarreia Viral Bovina vírus-1, e Vírus Parainfluenza-3. Todas as variáveis aumentaram (P < 0,01) dentre os tratamentos após a vacinação, exceto para TNFα sérico e títulos contra Diarreia Viral Bovina vírus-1 (P ≥ 0,40). Coletivamente, este estudo não encontrou evidencias de que administração oral de meloxicam, nas doses e intervalos adotados, mitigaram reações metabólicas, inflamatórias e de fase-aguda desencadeadas pela infusão de LPS ou vacinação contra doenças respiratórias. This study evaluated the effects of oral meloxicam administration on metabolic, inflammatory, and acute-phase responses of beef cattle receiving a lipopolysaccharide (LPS) challenge (Exp. 1; d −1 to 6) or vaccinated against respiratory pathogens (Exp. 2; d 7 to 21). Twenty-one Angus steers (n = 11) and heifers (n = 10) were housed in individual pens on d −15 and were offered free-choice water, mineral-vitamin mix, and hay until d 21. In Exp. 1, cattle were ranked on d −1 by sex and BW and assigned to 1) oral meloxicam administration (1 mg/kg BW daily) from day −1 to 6 (MEL8), 2) oral meloxicam administration (1 mg/kg BW) on d 0 and oral lactose monohydrate administration (1 mg/kg BW) on d −1 and from d 1 to 6 (MEL1), or 3) oral lactose monohydrate administration (1 mg/ kg BW daily) from d −1 to 6 (CON). On d 0, cattle received an intravenous LPS bolus (0.5 μg/kg BW) concurrently with treatment administration. Rectal temperature (RTEMP) was assessed, and blood samples were collected at −2, 0, 2, 4, 6, 8, 12, 16, 24, 36, 48, 60, 72, 96, 120, and 144 h relative to LPS administration. No treatment effects were detected (P ≥ 0.36) for RTEMP, concentrations of serum tumor necrosis factor α (TNFα), plasma haptoglobin, cortisol, insulin, and leptin, as well as blood mRNA expression of TNFα and cyclooxygenase-2, although all variables increased (P < 0.01) across treatments after LPS administration. In Exp. 2, cattle received the same treatments that they were assigned to in Exp. 1 from d 7 to d 13 and were vaccinated against respiratory pathogens concurrently with treatment administration on d 8. Blood samples were collected, and RTEMP was assessed as in Exp. 1 in addition to 168, 240, and 336 h relative to vaccination. No treatment effects were detected (P ≥ 0.26) for RTEMP, the same plasma and serum variables evaluated in Exp. 1, and serum concentrations of antibodies against Mannheimia haemolytica or serum titers against bovine respiratory syncytial virus, bovine herpesvirus-1, bovine viral diarrhea virus-1, and parainfluenza-3 virus. All variables increased (P < 0.01) across treatments after vaccination, except for serum TNFα and titers against bovine viral diarrhea virus-1 (P ≥ 0.40). Collectively, this study found no evidence that oral meloxicam administration, at the doses and intervals utilized herein, mitigated the metabolic, inflammatory, and acute-phase reactions elicited by LPS administration or vaccination against respiratory pathogens.

Introduction. Blue rubber bleb nevus syndrome is a rare disorder that is characterized by multiple recurrent vascular malformations that involve the skin and the gastrointestinal tract. The disease can present chronic anemia and severe episodes of gastrointestinal bleeding. Case Report. A 41-year-old man was admitted with recurrent episodes of lower gastrointestinal bleeding and anemia that had worsened over the last 3 months. The physical examination showed soft, diffuse, compressible, bluish nodules on all of the skin surfaces of the body. A biopsy from one of these skin lesions allowed a histological diagnosis of cavernous hemangioma. He submitted to a colonoscopy, which showed hemorrhoids and a plane vascular lesion mainly located on the right colon with recent signs of bleeding; this lesion was treated by local excision and sclerosis. The pathological study of the colon specimens also reflected the presence of cavernous hemangioma. The cutaneous hemangiomas and the presence of colonic venous malformations were compatible with blue rubber bleb nevus syndrome. The patient presented a favorable follow-up with clinical control of the anemia and without relapse of the gastrointestinal bleeding two years after the procedure. Conclusion. Although rarely diagnosed, blue rubber bleb nevus syndrome may be responsible for lower digestive bleeding.
