Abstract

Macrophages are known to depict two major phenotypes: classically activated macrophages (M1), associated with high production of pro-inflammatory cytokines, and alternatively activated macrophages (M2), which present an anti-inflammatory function. A precise control over M1-M2 polarization is a promising strategy in therapeutics to modulate both tissue regeneration and tumor progression processes. However, this is not a simple task as macrophages behave differently depending on the microenvironment. In agreement with this, non-consistent data have been reported regarding macrophages response to magnetic iron oxide nanoparticles (MNPs). To investigate the impact of both tissue microenvironment and MNPs properties on the obtained macrophage responses, single-core (SC) and multi core (MC) citrate coated MNPs, are synthesized and, afterwards, loaded with a macrophage polarization trigger, IL-4. The developed MNPs are then tested in macrophages subjected to different stimuli. We demonstrate that macrophages treated with low concentrations of MNPs behave differently depending on the polarization stage independently of the concentration of iron. Moreover, we find out that MNPs size and morphology determines the effect of the IL-4 loaded MNPs on M1 macrophages, since IL-4 loaded SC MNPs favor the polarization of M1 macrophages towards M2 phenotype, while IL-4 loaded MC MNPs further stimulate the secretion of pro-inflammatory cytokines

Abstract

Graphene oxide has been prepared from an industrial coke-like waste. The morphology of this material (GO-CW) is similar to that of standard graphene oxide (GO-G), although its surface has more defects. Both materials were used to prepare hybrid NHC-Ir(I)/graphene materials, consisting of molecular Ir(NHC) complexes covalently anchored to the graphene surface through the NHC moiety, following two different synthetic routes: (a) direct graphene electrografting of the previously synthesized aniline-functionalized imidazole-2-ylidene-Ir(I) complex, and (b) a two-step sequence comprising initial electrografting of aniline-imidazolium salts and subsequent chemical reaction with Ir(I) precursor anchoring imidazole-2-ylidene-Ir(I) molecular complexes. The synthesized NHC-Ir/graphene hybrid catalysts are active in the oxygen evolution reaction (OER) resulting in current densities in a similar range to those of other NHC-iridium(I) catalysts supported on GO. The highest activity corresponds to hybrid catalysts prepared by the two-step route, with even higher activity and stability when graphene oxide from industrial waste is used. EXAFS spectra of the materials prepared from both synthesis routes before oxidation catalysis reveal the local Ir coordination shell and a structural interaction between Ir and graphene. Both XANES and EXAFS spectra after electrocatalysis point to more oxidized species in which the molecular nature of the iridium catalysts is preserved.

Abstract

Durante 85 días (19 de septiembre – 13 de diciembre de 2021), la isla de La Palma sufrió el mayor evento eruptivo desde que se tienen registros históricos. El volcán Tajogaite emitió una gran cantidad de materiales volcánicos y gases, afectando a la población, a la economía local y a la biodiversidad. Tres ecosistemas se vieron afectados: matorral costero, matorral termófilo y pinar canario, siendo este último el que resultó más dañado, especialmente desde el cráter hasta 7 km hacia el sur de la isla. A pesar de ser un hábitat ampliamente estudiado, nunca se había evaluado el impacto de una erupción volcánica sobre su biodiversidad. Este estudio se ha centrado en este ecosistema, a similar altitud del cráter, a lo largo de la dorsal de Cumbre Vieja. Los resultados muestran que la velocidad de recuperación del pinar es más rápida de lo que se pensaba, permitiendo entender mejor los efectos de una erupción volcánica sobre la biodiversidad canaria.

Abstract

The filamentous cyanobacterium Limnospira platensis, formerly known as Arthrospira platensis or spirulina, is one of the most commercially important species of microalgae. Due to its high nutritional value, pharmacological and industrial applications it is extensively cultivated on a large commercial scale. Despite its widespread use, its precise manipulation is still under development due to the lack of effective genetic protocols. Genetic transformation of Limnospira has been attempted but the methods reported have not been generally reproducible in other laboratories. Knowledge of the transformation defense mechanisms is essential for understanding its physiology and for broadening their applications. With the aim to understand more about the genetic defenses of L. platensis, in this work we have identified the restriction-modification and CRISPR-Cas systems and we have cloned and characterized thirteen methylases. In parallel, we have also characterized the methylome and orphan methyltransferases using genome-wide analysis of DNA methylation patterns and RNA-seq. The identification and characterization of these enzymes will be a valuable resource to know how this strain avoids being genetically manipulated and for further genomics studies.

Abstract

Disrupted sleep has a profound adverse impact on lives of Parkinson's disease (PD) patients and their caregivers. Sleep disturbances are exceedingly common in PD, with substantial heterogeneity in type, timing, and severity. Among the most common sleep-related symptoms reported by PD patients are insomnia, excessive daytime sleepiness, and sleep fragmentation, characterized by interruptions and decreased continuity of sleep. Alterations in brain wave activity, as measured on the electroencephalogram (EEG), also occur in PD, with changes in the pattern and relative contributions of different frequency bands of the EEG spectrum to overall EEG activity in different vigilance states consistently observed. The mechanisms underlying these PD-associated sleep-wake abnormalities are poorly understood, and they are ineffectively treated by conventional PD therapies. To help fill this gap in knowledge, a new progressive model of PD - the MCI-Park mouse - was studied. Near the transition to the parkinsonian state, these mice exhibited significantly altered sleep-wake regulation, including increased wakefulness, decreased non-rapid eye movement (NREM) sleep, increased sleep fragmentation, reduced rapid eye movement (REM) sleep, and altered EEG activity patterns. These sleep-wake abnormalities resemble those identified in PD patients. Thus, this model may help elucidate the circuit mechanisms underlying sleep disruption in PD and identify targets for novel therapeutic approaches.

Abstract

Loss of functional mitochondrial complex I (MCI) in the dopaminergic neurons of the substantia nigra is a hallmark of Parkinson's disease.sup.1. Yet, whether this change contributes to Parkinson's disease pathogenesis is unclear.sup.2. Here we used intersectional genetics to disrupt the function of MCI in mouse dopaminergic neurons. Disruption of MCI induced a Warburg-like shift in metabolism that enabled neuronal survival, but triggered a progressive loss of the dopaminergic phenotype that was first evident in nigrostriatal axons. This axonal deficit was accompanied by motor learning and fine motor deficits, but not by clear levodopa-responsive parkinsonism--which emerged only after the later loss of dopamine release in the substantia nigra. Thus, MCI dysfunction alone is sufficient to cause progressive, human-like parkinsonism in which the loss of nigral dopamine release makes a critical contribution to motor dysfunction, contrary to the current Parkinson's disease paradigm.sup.3,4. Abstract

Sea turtles are key imperilled species that interact with numerous threats in their critical in‐water and inland habitats, which they occupy along their complex life cycle. Therefore, analysing these interactions is essential for adapting ongoing conservation and management actions.The long‐term recording of stranded sea turtle individuals and the potential causes of death in their breeding habitats is crucial for defining and adapting conservation actions.Twenty‐nine years of sea turtle stranding and mortality data (1992–2019) at nesting beaches in Campeche, Mexico, were analysed to describe their spatiotemporal patterns per species and possible causes of death.The most impacted species was the hawksbill turtle, followed by green and Kemp's ridley individuals. The most frequent probable causes of death in this region were bycatch, seismic surveys, vehicle strikes and slaughter at nesting beaches. Both immature and breeding individuals were stranded, mostly the former, and the most frequent months were in the summer (June–August).This study establishes a historic reference baseline for the spatiotemporal mortality patterns of imperilled sea turtle species in the southeastern Gulf of Mexico. It shows their interactions with anthropogenic activities and the circumstances jeopardizing their population recovery. It also points out management and conservation actions to implement to improve their long‐term viability in the context of the operationalization of their key attributes and interactions with their seascapes. Abstract

The energy storage mechanism operating in carbon-based supercapacitors using ionic liquids as electrolytes is not yet fully understood. In this paper, the interactions of ions of two widely used ionic liquids, i.e. EMImTFSI and EMImBF4, with a high specific surface area microporous carbon are investigated. Galvanostatic cycling experiments performed on each electrode and in the full cell demonstrate that a specific energy close to 46 Wh kg-1 can be achieved with this carbon. More interestingly, impedance spectroscopy studies reveal the presence of some unusual behavior, such as the presence of inductive elements in some of the electrodes, either in the positive or in the negative electrode. These inductive elements are identified, for the first time, as another type of possible experimental evidences of some phenomena previously proposed, such as ion-swapping or co-ions desorption, both phenomena leading to the formation of the superionic state.

Abstract

An important part of the ecological evolution of many lineages consists of adaptations to new diets as climate and available resources changed. In this study, we inferred when squirrels adapted morphologically to new diets by analyzing data from both extant and extinct species. We relied on the relationship between tooth morphology in extant squirrels and their dietary preferences to infer the diets of extinct species. To achieve this, we employed two approaches, namely geometric morphometrics and Elliptic Fourier analyses, to measure the outline of the fourth lower premolar (p4) in occlusal view. We then used both datasets to infer the diets of extinct species. Additionally, we used phylogenetic comparative methods on extant data to explore the evolution of diet and tooth size, identifying potential drivers of squirrel morphological evolution in the context of past climatic changes. Our findings indicate that the morphology of squirrel teeth has been maintained through the evolutionary history of most lineages, but that it also has changed in some lineages as a result of adaptations to particularly demanding diets, sometimes mediated by climatic factors, which have played a crucial role in shaping tooth morphology. Furthermore, we demonstrated that both geometric morphometrics and Elliptic Fourier analyses can yield a similar percentage of correct classifications and congruent dietary inferences. This study provides valuable insights into the macroevolutionary changes in squirrels and provides further evidence of the usefulness of tooth morphology for inferring the diets of extinct mammalian species.

Abstract

Two new species of ctenodactylid rodents, Prodistylomys taatsinius sp. nov. and Prodistylomys mongoliensis sp. nov., are described from the Valley of Lakes, Central Mongolia. They represent the first record of this genus in Mongolia. Prodistylomys is characterized by a simple dental pattern and high hypsodonty, reflecting an adaptation for tough vegetation in a dry environment, which is congruent with the aridification event that took place during the Late Oligocene and Early Miocene. We performed the most complete phylogenetic analysis combining the largest sampling in the family Ctenodactylidae and the best-represented stratigraphical distribution for the sample. This allowed us to propose that the origin of ctenodactylids lies in East Asia during the Eocene, and after that, the group dispersed through western Asia, Europe, and Africa. The presence of several similar rodent taxa in Mongolia, Kazakhstan, and China confirms that these areas were part of a large bioprovince covering most of central Asia during the Oligocene and Early Miocene.

Abstract

Hydrogen as an energy carrier plays a significant role in tackling energy transition challenges. Its production via water electrolysis can be powered by renewable sources of energy and it has been identified as the key to a secure and sustainable energy system. Therefore, the development of efficient and abundant electrocatalysts is very important to realize the required large-scale production. In this context, transition metals have been postulated as promising alternatives to noble metal oxides for water splitting in alkaline media. The efficient incorporation of these metals into supports can improve their distribution and particle size control, making carbon supports an ideal choice due to their high conductivity and electrochemical stability. In this work, reduced graphene oxide 3D aerogels doped with very low concentrations of nickel were prepared by freeze-casting. A fully water-based approach that enables an outstanding integration of nickel hydroxide precursor in a colloidal graphene oxide (GO) aqueous suspension was developed. The as-prepared Ni/graphene 3D networks were extensively characterized showing a low loading of nickel (<10 wt%), and ultradispersed, and nanosized Ni-based particles (15-40 nm). Electrochemical experiments show that Ni/graphene 3D networks exhibit very good catalytic properties towards the oxygen evolution reaction (OER) and outstanding Fe-ion activation from the impurities present in the alkaline (KOH 1 M) electrolyte media. Previous studies have shown that Fe incorporation can increase two-fold the activity of Ni-based electrocatalysts. In contrast, the ultradispersed Ni/graphene hybrid prepared here exhibits an impressive over ten-fold performance increase, highlighting the remarkable influence of Fe in these materials. The high-dispersion and surface availability of Ni species promotes the in situ formation of highly active Ni-Fe oxyhydroxide on the surface of the catalysts.
