52. Etude in vitro de l'impact de sediments artificiellement contamines par l'anthracene: effets sur les bacteries indigenes et les nematodes libres marins

Abstract

This study evaluates the in vitro impact of three bioremediation techniques: biostimulation, bioaugmentation, and a combination of biostimulation and bioaugmentation on microflora and nematofauna after an artificial contamination of lagoon sediments by anthracene. This sediment contamination tends to reduce the average load in aerobic heterotrophic bacteria in the water column, at the sediment interface and at the same time induces a highly significant decrease in the average densities of nematodes in all microcosms contaminated by this pollutant. After inoculation with the Staphylococcus sp. bacterial strain, the increase in bacterial load was significant (p < 0.05) in the three compartments of anthracene-contaminated microcosms with relation to the controls. The addition of nitrates and phosphates to the contaminated microcosms generated a highly significant increase in the average concentration of free-living nematodes with relation to the controls. La presente etude se propose d'evaluer, in vitro, l'impact de trois techniques de bioremediation a savoir la biostimulation, la bioaugmentation et la combinaison de la biostimulation et de la bioaugmentation sur la microflore et la nematofaune apres une contamination artificielle des sediments lagunaires par l' anthracene. Cette contamination sedimentaire, tend a provoquer une reduction de la charge moyenne des bacteries heterotrophes aerobies dans la colonne d' eau, a l' interface et dans le sediment et en parallele une diminution hautement significative des densites moyennes des nematodes dans tous les microcosmes contamines par ce polluant. Apres inoculation de la souche bacterienne Staphylococcus sp., l'augmentation de la charge bacterienne a ete significative (p < 0,05) au niveau des trois compartiments dans les microcosmes contamines par l'anthracene par rapport aux temoins. L'ajout des nitrates et des phosphates aux microcosmes contamines a entraine, par rapport aux temoins, une augmentation tres significative des effectifs moyens de nematodes libres. Abstract

An artificial benthic environment was used in the determination of the effects of oxygen, sulphide, pH and light on the growth of the CE 2401 strain of Prosthecochloris aestuarii, a green sulphur bacteria. Results indicate that bacterial growth is maximal beyond oxygen penetration depth, that sulphide flux limits microbial activity and that the bacteria exhibit adaptation to light regime changes.

Abstract

To assess the impact of two plastic derived chemicals: bisphenol A (BPA) and the di-2-ethylhexyl phthalate (DEHP), on phytoplankton biomass and community structure, microcosm incubations were performed during spring and summer, with offshore and lagoon waters of a south-western Mediterranean ecosystem. Phytoplankton were exposed to an artificial mixture of BPA and DEHP and to marine water previously enriched with plastic-derivative compounds, originated from in situ water incubations of plastic debris for 30 days. After 96 h of incubation, changes were observed in phytoplankton biomass in the contaminated microcosms, with a net decrease (up to 50% of the control) in the concentration of Chlorophyll a in offshore waters. Concomitantly, plastic-derivative contamination provoked structural changes, especially for offshore waters. This suggests a relative tolerance of the lagoon communities to BPA and DEHP contamination, related to the dominance of Chaetoceros spp., which could potentially be used as a bioindicator in bioassessment studies.

Abstract

International audience; Sediment resuspension can provoke strong water enrichment in nutrients, contaminants, and microorganisms. Microcosm incubations were performed in triplicate for 96 h, with lagoon and offshore waters incubated either with sediment elutriate or with an artificial mixt. of contaminants issued from sediment resuspension. Sediment elutriate provoked a strong increase in microbial biomass, with little effects on the phytoplankton and bacterioplankton community structures. Among the pool of contaminants released, few were clearly identified as structuring factors of phytoplankton and bacterioplankton communities, namely simazine, Cu, Sn, Ni, and Cr. Effects were more pronounced in the offshore waters, suggesting a relative tolerance of the lagoon microbial communities to contamination. The impacts of contamination on the microbial community structure were direct or indirect, depending on the nature and the strength of the interactions between phytoplankton and bacterioplankton.

Abstract

Mer intercontinentale presque entièrement fermée, la Méditerranée s'étend sur environ 2,5 millions de km2. Elle s'ouvre sur l'océan Atlantique via le détroit de Gibraltar et est reliée à la mer Noire par le Bosphore et, depuis 1869, à la mer Rouge par le canal de Suez. Située à la limite des plaques africaine et eurasienne, elle est le siège d'une intense activité sismique et volcanique. D'un point de vue physique, elle se divise principalement en deux bassins bien individualisés (Méditerranée occidentale et orientale), eux-mêmes subdivisés en nombreuses entités contrastées. Sous l'influence des grands fleuves qui s'y jettent (Pô, Rhône, Nil, Ebre etc) et des différents courants dont elle est le siège, elle se caractérise par des transferts horizontaux et verticaux rapides, fortement dépendants des conditions météo-climatiques et soumis à une grande variabilité à toutes les échelles temporelles. D'un point de vue biologique, la mer Méditerranée est identifiée comme l'un des ' hot spots ' de la biodiversité marine, abritant de très nombreuses espèces, dont un fort pourcentage d'endémiques. Lagunes littorales, marais maritimes, estuaires, deltas, côtes rocheuses et sableuses, herbiers, coralligènes, fonds meubles et rocheux, canyons, plateaux, montagnes sous marines, sont autant d'habitats remarquables qui favorisent la diversité des organismes en Méditerranée. D'un point de vue géopolitique, la mer Méditerranée représentait dans l'Antiquité un carrefour d'échanges commerciaux et culturels entre les peuples de la région. Elle constitue aujourd'hui un espace partagé par 23 pays riverains, traversé et exploité par ces pays et des tierces parties. C'est un espace disputé, avec des tensions entre les usagers et des compétitions sur les ressources. Néanmoins, sur l'ensemble des problématiques liées à la mer, les pays riverains sont liés par une ' communauté de destin '. D'où la nécessité de mieux coopérer, à l'échelle de l'ensemble du bassin, en termes d'analyse et d'évaluation des ressources ; d'observation, de suivi et de contrôle ; et de gestion des ressources et des usages qui en découlent. D'un point de vue humain, les pays riverains rassemblaient en 2011 une population de 475 millions d'habitants, dont un tiers sur le littoral et une forte proportion dans des ' mégacités ' côtières ou proches de la mer (Le Caire, Istanbul, Athènes, Rome, Barcelone, Marseille, Alger etc). Depuis le littoral jusqu'au large, la mer Méditerranée est le siège de nombreuses activités : espace de navigation, exploitation de ses ressources vivantes ou minérales, mais aussi de ses particularités paysagères, culturelles et climatiques qui attirent chaque année sur le littoral méditerranéen plus de touristes que celui-ci ne compte d'habitants ! Elle procure également une grande diversité de services écosystémiques non marchands (nurseries pour les espèces exploitées, recyclage des éléments nutritifs, absorption et séquestration de CO2 atmosphérique, filtration des substances toxiques, protection contre l'érosion des côtes etc), dont dépendent la qualité de vie des populations riveraines et certaines de leurs activités économiques, comme la pêche, l'aquaculture, le tourisme etc.

70. Environmental controlling factors of copepod communities in the Gulf of Tunis (south western Mediterranean Sea)

Abstract

The copepod community structure and the distribution of the main groups of zooplankton were studied along an inshore-offshore gradient in the Gulf of Tunis during the rainy and dry seasons of 2007-2008. Hydrological parameters were also measured to assess the potential role of abiotic and biotic factors in the distribution of copepod species. The copepod community in the Gulf of Tunis comprises 86 species dominated by Paracalanus parvus, Clausocalanus lividus, Centropages kroyeri and Acartia clausi. Time had a greater influence than space (horizontal and vertical gradients) in shaping the copepod community structure with a significant influence of the seasons; winter (cold and rainy) resulted in hydrological conditions that were strongly different from those observed in summer (warm and dry). These hydrological differences were concomitant with changes in the community structure, with a high copepod diversity observed in winter while the summer period was characterized by a low specific richness and the dominance of a few species, Centropages kroyeri and Paracalanus parvus along the inshore-offshore gradient and Paracalanus aculeatus along the vertical. Canonical correspondence analysis showed that temperature, salinity and to a lesser extent chlorophyll a were the most important environmental factors structuring the copepod community. Interestingly, temperature and salinity showed a negative significant correlation with copepod specific richness. Competition with grazers (cladoceran) as well as top down control by predators (chaetognaths and siphonophors) were also identified as key factors for the copepod community structure.

Abstract

Little information exists on the ecological adaptive responses of riverine microorganisms to the salinity changes that typically occur in transitional waters. This study examined the precise effects of a gradual increase in salinity (+3 units per day for 12 days) on freshwater virus and prokaryote communities collected in the Red River Delta (northern Vietnam). The abundance, activity, morphology and diversity of both communities were examined along this simulated salinity gradient (0–36). Three main successive ecological stages were observed: (1) a continuous decline in prokaryotic and viral abundance from the start of the salinization process up to salinity 12–15 together with a strong decrease in the proportion of active cells, (2) a shift in both community compositions (salinity 9–15) and (3) a marked prevalence of lysogenic over lytic cycles up to salinity 21 followed by a collapse of both types of viral infection. Finally, after salinity 21, and up to seawater salinities (i.e. 36) the prokaryotic community showed multiple signs of recovery with their abundance and function even reaching initial levels. These results suggest that most of the physiological and phylogenetic changes that occurred within the salinity range 10–20 seemed to favor the installation of osmotically adapted prokaryotes accompanied by a specific cortege of viral parasites which might both be able to survive and even proliferate in saltwater conditions.

Abstract

We describe a new method to produce marine aggregates from natural organic material based on the sticking properties of transparent exopolymeric particles. Seawater samples were prescreened and ultrafiltered to concentrate the 30 kDa to 10 mu m size fraction. First, we produced small magnetizable aggregates by combining glass microfibers and paramagnetic 1-mu m beads with the organic matter present in the concentrated solution. The second step involved clustering the small aggregates into a single macro-aggregate, using a small ring-shaped magnet as an aggregation nucleus. Viral and bacterial densities, determined after dissolution of the newly formed aggregates with methanol, averaged 13.8 x 10(6) +/- 3.6 x 10(6) vir. agg.(-1) and 4.1 x 10(6) +/- 1.1 x10(6) bact. agg.(-1). Bacterial respiration and production measurements of single aggregates averaged 8.47 +/- 1.72 nmol O-2 agg.(-1) h(-1) and 1.54 +/- 0.45 ng C agg.(-1) h(-1), respectively. Particulate organic carbon and nitrogen content of the newly formed macro-aggregates averaged 31.92 +/- 2.67 mu g C agg.(-1) and 3.44 +/- 0.43 mu g N agg.(-1), respectively. This approach allows the concentration and isolation of the organic matter precursors that compose natural aggregates and provides a simple protocol for recombining those precursors into single newly formed macro-aggregates, which can then be easily manipulated for further investigation. This method is a new tool for investigations into the interactions between microorganisms and marine aggregates and their implications at the ecosystem level, but also into the interactions between aggregates and dissolved organic or inorganic substances.

Abstract

Microbial mats are characterized by high primary production but low growth rates, pointing to a limitation of growth by the lack of nutrients or substrates. We identified compounds that instantaneously stimulated photosynthesis rates and oxygen consumption rates in a hypersaline microbial mat by following the short-term response (c. 6 h) of these processes to addition of nutrients, organic and inorganic carbon compounds, using microsensors. Net photosynthesis rates were not stimulated by compound additions. However, both gross photosynthesis and oxygen consumption were substantially stimulated (by a minimum of 25%) by alanine (1 mM) and glutamate (3.5 mM) as well as by phosphate (0.1 mM). A low concentration of ammonium (0.1 mM) did not affect photosynthesis and oxygen consumption, whereas a higher concentration (3.5 mM) decreased both process rates. High concentrations of glycolate (5 mM) and phosphate (1 mM) inhibited gross photosynthesis but not oxygen consumption, leading to a decrease of net photosynthesis. Photosynthesis was not stimulated by addition of inorganic carbon, nor was oxygen consumption stimulated by organic compounds like glycolate (5 mM) or glucose (5 mM), indicating that carbon was efficiently cycled within the mat. Photosynthesis and oxygen consumption were apparently tightly coupled, because stimulations always affected both processes to the same extent, which resulted in unchanged net photosynthesis rates. These findings illustrate that microsensor techniques, due to their ability to quantify all three processes, can clarify community responses to nutrient enrichment studies much better than techniques that solely monitor net fluxes.

Abstract

The objectives of this study were (1) to assess the responses of benthic nematodes to a polycyclic aromatic hydrocarbon (PAH) contamination and (2) to test bioremediation techniques for their efficiency in PAH degradation and their effects on nematodes. Sediments with their natural nematofauna communities from Bizerte lagoon (Tunisia) were subjected to a PAH mixture (100 ppm) of phenanthrene, fluoranthene, and pyrene during 30 days. Nematode abundance and diversity significantly decreased, and the taxonomic structure was altered. Results from multivariate analyses of the species abundance data revealed that PAH treatments were significantly different from the control. Spirinia parasitifera became the dominant species (70 % relative abundance) and appeared to be an 'opportunistic' species to PAH contamination while Oncholaimus campylocercoides and Neochromadora peocilosoma were strongly inhibited. Biostimulation (addition of mineral salt medium) and bioaugmentation (inoculation of a hydrocarbonoclastic bacterium) were used as bioremediation techniques. Bioremediation treatments enhanced degradation of all three PAHs, with up to 96 % degradation for phenanthrene resulting in a significant stimulation of nematode abundance relative to control microcosms. Nevertheless, these treatments, especially the biostimulation provoked a weak impact on the community structure and diversity index relative to the control microcosms suggesting their feasibility in biorestoration of contaminated sediments. Abstract

Spatial increases and temporal shifts in outbreaks of gelatinous plankton have been observed over the past several decades in many estuarine and coastal ecosystems. The effects of these blooms on marine ecosystem functioning and particularly on the dynamics of the heterotrophic bacteria are still unclear. The response of the bacterial community from a Mediterranean coastal lagoon to the addition of dissolved organic matter (DOM) from the jellyfish Aurelia aurita, corresponding to an enrichment of dissolved organic carbon (DOC) by 1.4, was assessed for 22 days in microcosms (8 l). The high bioavailability of this material led to (i) a rapid mineralization of the DOC and dissolved organic nitrogen from the jellyfish and (ii) the accumulation of high concentrations of ammonium and orthophosphate in the water column. DOM from jellyfish greatly stimulated heterotrophic prokaryotic production and respiration rates during the first 2 days; then, these activities showed a continuous decay until reaching those measured in the control microcosms (lagoon water only) at the end of the experiment. Bacterial growth efficiency remained below 20 %, indicating that most of the DOM was respired and a minor part was channeled to biomass production. Changes in bacterial diversity were assessed by tag pyrosequencing of partial bacterial 16S rRNA genes, DNA fingerprints, and a cultivation approach. While bacterial diversity in control microcosms showed little changes during the experiment, the addition of DOM from the jellyfish induced a rapid growth of Pseudoalteromonas and Vibrio species that were isolated. After 9 days, the bacterial community was dominated by Bacteroidetes, which appeared more adapted to metabolize high-molecular-weight DOM. At the end of the experiment, the bacterial community shifted toward a higher proportion of Alphaproteobacteria. Resilience of the bacterial community after the addition of DOM from the jellyfish was higher for metabolic functions than diversity, suggesting that jellyfish blooms can induce durable changes in the bacterial community structure in coastal lagoons. Udgivelsesdato: August 1999, The dynamics of sulphide oxidation in an experimental biofilm of the green sulphur bacterium, Prosthecochloris aestuarii, were studied using a newly developed light–dark cycling procedure. The biofilm was grown for 6 weeks in a benthic gradient chamber, in which gradients of light, sulphide and oxygen were imposed experimentally. The H2S concentrations and pH were measured with microsensors as a function of depth in the biofilm and of time after a change in illumination status. The sulphide oxidation rates were calculated as a function of time and depth in the biofilm using a numerical procedure to solve the non-stationary general diffusion equation. A close agreement was found between the areal rates of anoxygenic photosynthesis during the cycling procedure and the steady state before the cycling experiment. For the different layers of the biofilm, the maximum activity was observed after 10–12 min of light exposure. After this maximum, sulphide oxidation decreased concomitantly with sulphide concentration, indicating sulphide limitation of anoxygenic photosynthesis. This lag time limits the application of the standard dark–light shift method with a brief light exposure of a few seconds and, therefore, the numerical procedure described in this study enables the depth distribution of anoxygenic photosynthesis rates in microbial mats to be determined more accurately.

Abstract

To assess the impact of two plastic derived chemicals: bisphenol A (BPA) and the di-2-ethylhexyl phthalate (DEHP), on phytoplankton biomass and community structure, microcosm incubations were performed during spring and summer, with offshore and lagoon waters of a south-western Mediterranean ecosystem. Phytoplankton were exposed to an artificial mixture of BPA and DEHP and to marine water previously enriched with plastic-derivative compounds, originated from in situ water incubations of plastic debris for 30 days. After 96 h of incubation, changes were observed in phytoplankton biomass in the contaminated microcosms, with a net decrease (up to 50% of the control) in the concentration of Chlorophyll a in offshore waters. Concomitantly, plastic-derivative contamination provoked structural changes, especially for offshore waters. This suggests a relative tolerance of the lagoon communities to BPA and DEHP contamination, related to the dominance of Chaetoceros spp., which could potentially be used as a bioindicator in bioassessment studies., (Copyright © 2018.
