68. Gene targeting as a tool to investigate granulocyte function

Abstract

Neutrophils provide the body's first line of defence against infection by bacteria and fungi. The aim of this project was to use gene targeting to determine the effect of disrupting three genes thought to be important in neutrophil function. Mice lacking p47phox. a component of the NADPH oxidase, were found to suffer from a condition identical to the human disorder chronic granulomatous disease (CGD). Like sufferers of CGD the mutant mice lacked the activity of the NADPH oxidase, developed granulomas and were extremely susceptible to infection by C.albicans. Mice deficient in grancalcin, an EF hand calcium binding protein expressed in neutrophils, were healthy and fertile. Neutrophils from the mutant mice could kill S.aureus and migrate to the peritoneum normally. The mutant mice themselves were able to resolve infection by S.aureus in an identical manner to wild type mice. Further research will be required using the mutant mice to define the role of grancalcin. Mice lacking Csk specifically in their neutrophils were generated using conditional gene targeting and found to suffer from pneumonia. Csk is thought to down regulate the Src family of tyrosine kinases and the tenfold elevation in tyrosine phosphorylation in the neutrophils of the mutant mice implied that Csk actively plays this role in neutrophils. The results obtained not only show how essential p47phox and Csk are for normal neutrophil function but how essential neutrophils are to the immune system.

Abstract

In this essay we take up two related questions facing us in everyday, contemporary analytic practice: How can we grasp what we don’t know? How can something new come into being through the analytic encounter? We approach both questions from within the perspective offered us by Bion and those colleagues who have further developed his concepts and his insights. In his late writings Bion introduced the concept of O, subsequently viewing atone- ment with O as the goal of analytic work, and transformations from O (Being) to K (Knowing) the fundamental process of analysis and of the patient’s growth. In this theorizing he stressed an analytic stance centered on Negative Capability (the absence of memory, desire and coherence) fostering a coincidence of intuition and O. Despite the elegance of his theorizing Bion did not offer clinical examples of his own to ground the concepts of O and at-one-ment. In this paper we discuss the challenges for the analyst in attempting to practice without memory, desire or coherence, noting that this idealized model is more aspirational than fully realizable. We also discuss the dread shared by analyst and patient alike of intersecting with O due to the threats of de-differentiation, the shared sense of being lost, and the narcissistic deflation (loss of personal integrity) that can accompany it. To ground our discussion, we offer a detailed clinical example of at-one-ment with O, suggesting that at-one-ment with O can be thought of as a form of communion or shared holding and transforming of experience. Understood this way, at-one-ment is a deeply somato/psychic experience arising from the vertex of O and is distinguishable from the vertex of projective identification/container-contained processes. Abstract

Pavement surface condition rating is an essential part of road infrastructure maintenance and asset management, and it is performed manually by the data analyst. The manual rating requires cognitive skills built through training and experience, which is quantitatively challenging and timeconsuming. This paper first analyses the complexity of the current manual visual rating system. This paper then investigates the suitability and robustness of a state-of-the-art convolutional neural network (CNN) classifier to automate the pavement surface condition index (PSCI) system used to rate pavement surfaces in Ireland. The dataset contains 3735 images of flexible asphalt pavements from Irish urban and rural environments taken from a video camera mounted in front of a van. The PSCI ratings were applied by experts using a scale of 1-10 to indicate surface conditions. The classification models are evaluated for different input pre-processing variations, image size, learning techniques, and the number of classes. Using 10 PSCI classes, the best classifier achieved a precision of 57% and a recall of 58%. Adjacent combination of classes (e.g., ratings 1 and 2 combined into a single class) to form a 5-class problem produced a classifier with a precision of 70% and recall of 77%. Given the complexity of the problem, classification using CNN holds promise as a first step towards an automated ranking system.
