Abstract

Rapid endothelialization of cardiovascular stents is critical to prevent major clinical complications such as restenosis. Reconstruction of the native endothelium on the stent surface can be achieved by the capture of endothelial progenitor cells (EPCs) or neighboring endothelial cells (ECs) in vivo. In this study, stainless steel cardiovascular stents were functionalized with recombinant scFv antibody fragments specific for vascular endothelial growth factor receptor‐2 (VEGFR2) that is expressed on EPCs and ECs. Anti‐VEGFR2 scFvs were expressed in glycosylated form in Escherichia coli and covalently attached to amine‐functionalized, titania‐coated steel disks and stents. ScFv‐coated surfaces exhibited no detectable cytotoxicity to human ECs or erythrocytes in vitro and bound 15 times more VEGFR2‐positive human umbilical vein ECs than controls after as little as 3 min. Porcine coronary arteries were successfully stented with scFv‐coated stents with no adverse clinical events after 30 days. Endovascular imaging and histology revealed coverage of the anti‐VEGFR2 scFv‐coated stent with a cell layer after 5 days and the presence of a neointima layer with a minimum thickness of 80 μm after 30 days. Biofunctionalization of cardiovascular stents with endothelial cell‐capturing antibody fragments in this manner offers promise in accelerating stent endothelialization in vivo. © 2019 Wiley Periodicals, Inc. Abstract

Phthalocyanines (Pc) are promising second-generation photosensitizers for the photodynamic therapy (PDT) of cancer. We report on the tetrasulfonated aluminum phthalocyanine (AlPcS4) localization in cultured Chinese hamster lung cells studied by means of confocal laser scanning microscopy (CLSM). In these cells AlPcS4 was found in granules surrounding Golgi apparatus and in the peripheral cytoplasmic region. Peripheral Pc-containing granules partially coincided with the acidic cellular compartments. The effect of irradiation with light on Pc intracellular distribution was also studied. In the Pc-free medium disruption of some Pc- containing granules was observed followed by appearance of Pc fluorescence in the cell plasma membrane, the nuclear envelope, and the near-nuclear region. When cells were irradiated in the presence of Pc in external medium a drastic increase of membrane permeability to Pc was observed, followed by Pc binding the cell plasma membrane, nuclear envelope, and some structures in the cytoplasm. Diffusive Pc fluorescence in the nucleus was also observed. Abstract

The surface passivation with titanium sol-gel coatings is a frequently used technique to control the adsorption of selected biological macromolecules and to reduce the exposure of the bulk material to biological matter. Due to the increasing number of new coating-preparation methods and new gel compositions with various types of additives, the quality and homogeneity determination of the surface covering is a critical factor affecting performance of any implanted material. While coating thickness is easy to determine, the homogeneity of the surface distribution of coating materials requires more elaborate methodologies. In the paper, the laser induced breakdown spectroscopy (LIBS) based method, capable to quantitate the homogeneity and uniformity of the europium in titanium dioxide sol-gel coatings on stainless steel surfaces prepared with two different procedures: spin-coating and dip-coating, is presented. The emission intensity of titanium has been used to determine the coating thickness whereas the relative values of europium and titanium emission intensities provide data on the coating homogeneity. The obtained results show that the spin-coating technique provides better surface coverage with titanium dioxide. However, when the surface coating compositions were compared the dip-coating technique was more reliable. [ABSTRACT FROM AUTHOR]

Abstract

T-cell contact with antigen-presenting B cells initiates an activation cascade which includes an increase in T-cell intracellular calcium and leads to T-cell proliferation and differentiation. We studied cell-cell contact requirements for T-cell activation using an optical trap to control the orientation of T-cell/B-cell pairs and fluorescence microscopy to measure subsequent T-cell [Ca2+]i response. B cells or beads coated with antibodies to the T-cell receptor (TCR) are trapped with a titanium-sapphire laser and placed at different locations along the T-cell, which has a polarized appearance defined by die shape and direction of crawling. T-cell [Ca2+]i is detected as an emission shift from the combination of fura-red and Oregon-green, two cytoplasmic [Ca2+] indicators. T cells which are presented antigen at the leading edge have a higher probability of responding and a shorter latency of response than those contacting B-cells or beads with their trailing end.

Abstract

Using a polarization sensitive Michelson interferometer, we measure two-dimensional images of optical birefringence in bovine tendon as a function of depth with a technique known as Optical Coherence Tomography (OCT). Detection of the polarization state of the signal, formed by interference of backscattered light from the sample and a mirror in the reference arm, gives the optical phase delay between light that has propagated along the fast and slow axes of the birefringent tendon. Images showing the change in birefringence in response to laser irradiation are presented. The technique allows rapid non-contact investigation of tissue structural properties through two-dimensional imaging of birefringence.

Abstract

We report the development of an optical technique for noninvasive in vivo imaging of tissue structure and blood flow dynamics with high spatial resolution (2 - 15 micrometer) in biological systems. The technique is based on coherence optical Doppler tomography (ODT), which combines Doppler velocimetry with optical coherence tomography to measure blood flow velocity at discrete spatial locations. The exceptionally high resolution of ODT allows noninvasive in vivo imaging of both blood microcirculation and tissue structures surrounding the vessel, which has significance for biomedical research and clinical applications. Abstract

Continuous-wave ultrasonic modulation of scattered laser light has been used to image objects in tissue-simulating turbid media for the first time. We hypothesized that the ultrasound wave focused into the turbid media modulates the laser light passing through the ultrasonic focal zone. The modulated laser light collected by a photomultiplier tube reflects the local mechanical and optical properties in the focal zone. Buried objects in 5-cm thick tissue phantoms (absorption coefficient µ_a = 0.1 cm^(-1), reduced scattering coefficient µ_s' = 10 cm^(-1)) were located with millimeter resolution by scanning and detecting alterations of the ultrasound-modulated optical signal.
