Infections caused by KPC-producing K. pneumoniae continue to pose a significant clinical challenge due to their emerging resistance to new antimicrobials. We investigated the association between two drugs whose roles have been repurposed against multidrug-resistant bacteria: fosfomycin and temocillin. Temocillin exhibits unusual stability against KPC enzymes, while fosfomycin acts as a potent “synergizer”. We conducted in vitro antimicrobial activity studies on 100 clinical isolates of KPC-producing K. pneumoniae using a combination of fosfomycin and temocillin. The results demonstrated synergistic activity in 91% of the isolates. Subsequently, we assessed the effect on Galleria mellonella larvae using five genetically different KPC-Kp isolates. The addition of fosfomycin to temocillin increased larvae survival from 73 to 97% (+Δ 32%; isolate 1), from 93 to 100% (+Δ 7%; isolate 2), from 63 to 86% (+Δ 36%; isolate 3), from 63 to 90% (+Δ 42%; isolate 4), and from 93 to 97% (+Δ 4%; isolate 10). Among the temocillin-resistant KPC-producing K. pneumoniae isolates (24 isolates), the addition of fosfomycin reduced temocillin MIC values below the resistance breakpoint in all isolates except one. Temocillin combined with fosfomycin emerges as a promising combination against KPC-producing K. pneumoniae, warranting further clinical evaluation.

Food safety is a topic of increasing relevance in the European Union (EU). Ensuring safe food from farm to fork is one of the main objectives of EU policies. In our study, we conducted an online survey to assess the food safety knowledge and eating habits in various categories. For young people, we chose school students aged 15–19, based on their ability to cook autonomously after school. For adults, we choose a representative sample consisting of students’ parents and teachers in Chieti and Pescara Provinces of Abruzzo Region (Italy). The survey was structured in eight sections with particular attention to questions regarding risks associated with microorganisms (e.g., Salmonella spp.) and bacterial cross-contamination. Water stress is an everyday issue in developing countries and especially in Foumban, a municipality located in the West Region of Cameroon between longitudes 10°40’-11°10’E and latitudes 05°30’-05°55’N. The failure of the water distribution system in the Municipality of Foumban (MoF) led the citizens as well as the municipal authorities to exploit groundwater. Unfortunately, an extensive hydrogeophysical field campaign, with incidental cost, is required because the groundwater potential is not priorly assessed. The main objective of this article is to demarcate groundwater potential zones of MoF. For this, six thematic layers contributing to groundwater occurrences are combined using analytical hierarchy process (AHP) on a GIS environment to produce a sound groundwater potential map. This map is classified into three zones: low potential for 39.06%, acceptable potential for 27.27% and high potential for 33.68% of the study area. This may help to increase the rate of productive wells (by focusing hydrogeophysical investigation on areas with high groundwater potential) using little financial resources.

The need to use renewable sources and matrices with energy potential is widely recognized. The development of innovative technologies aimed at the improvement of energy conversion processes and reducing environmental impacts is currently receiving increasing attention from the scientific community and policymakers. The presence of sugars in airborne particle materials is attributed to biomass combustion. For this reason, these compounds are considered markers of biomass burning. The purpose of this work was to evaluate the emissions produced by agroforestry biomass burning (citrus pruning) by simultaneously sampling both stack emissions and atmospheric particulates in the area around a biomass boiler to understand the real contribution of biomass burning to atmospheric pollution. The combustion tests were carried out by comparing the processes with and without particulate abatement system to see how biomass combustion’s contribution to particulate emission can be controlled and reduced. During the tests, the focus was on particulate matter (PM) speciation in terms of sugar marker identification and determination. This study aims to increase knowledge to better understand the contribution of biomass plants to air pollution and differentiate it from the contributions of other sources, such as vehicular traffic or domestic heating.

The injudicious usage of antibiotics during infections caused by Gram-negative bacteria leads to the emergence of β-lactamases. Among them, the NDM-1 enzyme poses a serious threat to human health. Developing new antibiotics or inhibiting β-lactamases might become essential to reduce and prevent bacterial infections. Nanobodies (Nbs), the smallest antigen-binding single-domain fragments derived from Camelidae heavy-chain-only antibodies, targeting enzymes, are innovative alternatives to develop effective inhibitors. The biopanning of an immune VHH library after phage display has helped to retrieve recombinant antibody fragments with high inhibitory activity against recombinant-NDM-1 enzyme. Nb02NDM-1, Nb12NDM-1, and Nb17NDM-1 behaved as uncompetitive inhibitors against NDM-1 with Ki values in the nM range. Remarkably, IC50 values of 25.0 nM and 8.5 nM were noted for Nb02NDM-1 and Nb17NDM-1, respectively. The promising inhibition of NDM-1 by Nbs highlights their potential application in combating particular Gram-negative infections.

Maritime safety and the protection of the marine environment were the primary objectives of two European projects that the National Research Council of Italy had participated in, with numerical applications in two areas located in the northern part of Sardinia, Italy. Specifically, two operational Numerical Prediction Systems (NPS) for pollution risk management were developed; the first was applied to the area of the Bonifacio Strait and the Gulf of Asinara and the second to the port of Olbia. These systems are composed of many oceans and particle tracking numerical models. They are forced with meteorological and ocean data provided by the European Centre for Medium-Range Weather Forecasts and Copernicus Marine Service and their outputs have been compared with in situ measurements for preliminary calibration. A web graphical interface was ad hoc designed, specifically responding to projects’ needs, providing online access to a 3-day oceanographic forecast and advanced diagnostic variables like Oil Stranding Time, Risk Score and Water Age. These products, along with the interactive web platform, prove invaluable for marine spatial planning, prevention and emergency management at sea, for the use of competent governmental and local bodies.

In children, ureteropelvic junction obstruction (UPJO) is mostly caused by intrinsic factors (IUPJO) such as abnormal amounts of muscle and collagen deposition; extrinsic UPJO are rare and often due to crossing vessels (CVs). What is not clear is whether there is also intrinsic UPJ pathology in patients with CV. The aim of our study was to compare the histology of the two types of obstruction and to determine whether these histologic features are distinguishable enough to enable to identify the cause of obstruction based on histologic appearance alone. We retrospectively reviewed pathology reports of 38 children with UPJO that underwent surgery in our hospital from 2008 to 2022. The intrinsic and extrinsic groups consisted of 18 and 20 patients, respectively. After ematoxylin-eosin and Gomori's trichrome staining the specimens were scored for fibrosis and muscular hypertrophy in histhopatology, and CD117 antibody were used to detect interstitial Cajal-like cells. In our study, histological analyses revealed no differences between the CV and IUPJO specimens in terms of presence and degree of fibrosis and muscular hypertrophy; likewise, for presence of interstitial Cajal-like cells.

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) was identified as the pathogen responsible for the pandemic health emergency declared by the World Health Organization in March 2020. During the first part of the pandemic, adults showed mild to severe respiratory symptoms. Children seemed initially exempt, both from acute and subsequent complications. Hyposmia or anosmia were promptly identified as the main symptoms of acute infection, so neurotropism of SARS-CoV-2 was immediately suspected. (1, 2). As the emergency progressed, post infectious neurological complications were described also in pediatric population (3). Cases of cranial neuropathy in connection with acute SARS-CoV-2 infection have been reported in pediatric patients, as an isolate post infectious complication or in the context of the multisystem inflammatory syndrome in children (MIS-C) (4–6). Neuroinflammation is thought to be caused by several mechanisms, among which immune/autoimmune reactions (7), but so far, no specific autoantibody has been identified. SARS-CoV-2 can enter the central nervous system (CNS) directly and/or infect it retrogradely, through the peripheral nervous system (PNS), after replicating peripherally; several factors regulate invasion and subsequent neuroinflammation. Indeed, direct/secondary entry and replication can activate CNS-resident immune cells that, together with peripheral leukocytes, induce an immune response and promote neuroinflammation. In addition, as we will discuss in the following review, many cases of peripheral neuropathy (cranial and non-cranial) have been reported during or after SARS-CoV-2 infection. However, some authors have pointed out that the increase of cranial roots and ganglia in neurological imaging is not always observed in children with cranial neuropathy. (8). Even if a variety of case reports were published, opinions about an increased incidence of such neurologic diseases, linked to SARS-CoV-2 infection, are still controversial (9–11). Facial nerve palsy, ocular movements abnormalities and vestibular alterations are among the most reported issues in pediatric population (3–5). Moreover, an increased screen exposure imposed by social distancing led to acute oculomotion’s disturbance in children, not primarily caused by neuritis (12, 13). The aim of this review is to suggest food for thought on the role of SARS-CoV-2 in neurological conditions, affecting the peripheral nervous system to optimize the management and care of pediatric patients.

Plant roots are considered highly efficient soil explorers. As opposed to the push‐driven penetration strategy commonly used by many digging organisms, roots penetrate by growing, adding new cells at the tip, and elongating over a well‐defined growth zone. However, a comprehensive understanding of the mechanical aspects associated with root penetration is currently lacking. We perform penetration experiments following Arabidopsis thaliana roots growing into an agar gel environment, and a needle of similar dimensions pushed into the same agar. We measure and compare the environmental deformations in both cases by following the displacement of fluorescent beads embedded within the gel, combining confocal microscopy and Digital Volume Correlation (DVC) analysis. We find that deformations are generally smaller for growing roots. To better understand the mechanical differences between the two penetration strategies, we develop a computational model informed by experiments. Simulations show that, compared to push‐driven penetration, grow‐driven penetration reduces frictional forces and mechanical work, with lower propagation of displacements in the surrounding medium. These findings shed light on the complex interaction of plant roots with their environment, providing a quantitative understanding based on a comparative approach. Summary Statement: Inspired by the exceptional penetration abilities of plant roots, here we perform an experimental and computational analysis of root‐soil mechanical interactions. Aims Methods and Results Conclusion Giant cell interstitial pneumonia (GIP) is a fibrosing lung disease histologically characterized by centrilobular pulmonary fibrosis and cannibalistic intra‐alveolar multinucleated giant cells. It is considered a form of pneumoconiosis caused particularly by secondary exposure to hard metals (cemented carbide or tungsten carbide). Hard metals are commonly used in various industrial applications, such as cutting tools, drilling tools, machine inserts, and other wear‐resistant components. However, cases with unknown exposure that recurred in transplanted lungs have been described. This has led to the hypothesis of a complex etiopathogenesis, likely multifactorial, involving the coparticipation of immune mechanisms. We aimed to identify all the elements present in a series of GIP lung samples to better understand the pathogenic mechanisms of the disease.We describe five cases of histologically diagnosed GIP in patients with occupational exposure to metallic dust using ultrastructural characterization to identify metal dust and to quantify asbestos fibres. We found that tungsten was present in three cases, albeit in trace amounts in two of them. Numerous elements were identified in all samples, including asbestos fibres in patients with endstage pulmonary fibrosis. Furthermore, in one of the described cases the recurrence of the disease was also observed in transplanted lungs.These findings support the hypothesis that GIP may be due to elements other than hard metals, with asbestos possibly representing a contributory factor in the expression of a more severe fibrotic disease. The recurrence of GIP observed in transplanted organs strengthens the hypothesis of the existence of a not yet fully understood etiopathogenic immune mechanism. [ABSTRACT FROM AUTHOR]

The introduction of a graphene (Gr) buffer layer between a ferromagnetic substrate and a metallorganic molecule is known to mediate the magnetic coupling between them, an effect attributed to a weak hybridization between graphene and molecule. In this paper, we present experimental evidence of this effect through a detailed investigation of the frontier electronic properties of iron phthalocyanine deposited on cobalt‐supported graphene. Despite being physisorbed, the molecular adsorption on Gr/Co induces a sizeable charge transfer from graphene to the molecular macrocycle leading to the partial occupation of the LUMO and the appearance of an energetically localized hybrid state, which can be attributed to the overlap between the graphene pz state and the molecular macrocycle. Graphene is not inert either; the adsorption of the molecule induces doping and alters the Fermi velocity of both the hybrid minicone state and the Dirac cone. Similar effects are observed when the molecular periphery is decorated with fluorine atoms, known for their electron‐withdrawing properties, with minimal changes in the energy alignment. This paper presents a passive Envelope Detector (ED) to be used for reception of OOK-modulated signals, such as in Wake-Up Receivers employed within Wireless Sensor Networks, widely used in the IoT. The main goal is implementing a temperature compensation mechanism in order to keep the passive ED input resistance roughly constant over temperature, making it a constant load for the preceding matching network and ultimately keeping the overall receiving chain sensitivity constant over temperature. The proposed ED was designed using STMicroelectronics 90 nm CMOS technology to receive 1 kbps OOK-modulated packets with a 433 MHz carrier frequency and a 0.6 V supply. The use of a block featuring a Proportional-to-Absolute Temperature (PTAT) current yields a 5 dB reduction in sensitivity temperature variation across the −40 °C to 120 °C range. Moreover, two different implementations were compared, one targeting minimal mismatch and the other one targeting minimal area. The minimal area version appears to be better in terms of estimated overall chain sensitivity at all temperatures despite a higher sensitivity spread. [ABSTRACT FROM AUTHOR]

In the present study, we analyzed the genome of two S. enterica strains TS1 and TS2 from stool and blood cultures, respectively, and one strain of C. freundii TS3, isolated from a single hospitalized patient with acute myeloid leukemia. The S. enterica Goldcoast ST358 (O:8 (C2-C3) serogroup), sequenced by the MiSeq Illumina system, showed the presence of β-lactamase genes (blaVIM-1, blaSHV-12 and blaOXA-10), aadA1, ant(2″)-Ia, aac(6′)-Iaa, aac(6′)-Ib3, aac(6′)-Ib-cr, qnrVC6, parC(T57S), and several incompatibility plasmids. A wide variety of insertion sequences (ISs) and transposon elements were identified. In C. freundii TS3, these were the blaVIM-1, blaCMY-150, and blaSHV-12, aadA1, aac(6′)-Ib3, aac(6′)-Ib-cr, mph(A), sul1, dfrA14, ARR-2, qnrVC6, and qnrB38. IncA plasmid isolated from E.coli/K12 transconjugant and C. freundii exhibited a sequence identity >99.9%. The transfer of IncA plasmid was evaluated by conjugation experiments.

Objectives: This study assessed a new technology-aided program to support independent leisure and communication engagement with people with intellectual and motor or sensory-motor disabilities. Methods: Six adults participated. The program relied on the use of a Samsung Galaxy Tab S2 LTE tablet with Android 6.0 Operating System, a NANO SIM card, and the WhatsApp Messenger and MacroDroid applications. The participants were presented with leisure and communication (i.e. text messaging) alternatives, and, based on their choices, with series of steps allowing them to access leisure events or send messages. Results: During baseline, the participants were unable to access leisure events or send messages. During intervention and post-intervention phases, they used the program successfully and spent large percentages of the session time independently engaged in leisure and messaging. Conclusions: A program based on readily available technology may be functional to support leisure and communication in people like the participants of this study.
