Abstract Weak absorption contrast in biological tissues has hindered x‐ray computed tomography from accessing biological structures. Recently, grating‐based imaging has emerged as a promising solution to biological low‐contrast imaging, providing complementary and previously unavailable structural information of the specimen. Although it has been successfully applied to work with conventional x‐ray sources, grating‐based imaging is time‐consuming and requires a sophisticated experimental setup. In this work, we demonstrate that a deep convolutional neural network trained with a generative adversarial network can directly convert x‐ray absorption images into differential phase‐contrast and dark‐field images that are comparable to those obtained at both a synchrotron beamline and a laboratory facility. By smearing back all of the virtual projections, high‐quality tomographic images of biological test specimens deliver the differential phase‐contrast‐ and dark‐field‐like contrast and quantitative information, broadening the horizon of x‐ray image contrast generation.

IntroductionIssues related to fat, oil, and grease from kitchen waste (KFOG) in lipid-containing wastewater are intensifying globally. We reported a novel denitrifying bacterium Pseudomonas CYCN-C with lipid-utilizing activity and high nitrogen-removal efficiency. The aim of the present study was aim to explore the metabolic mechanism of the simultaneous lipid-utilizing and denitrifying bacterium CYCN-C at transcriptome level.MethodsWe comparatively investigated the cell-growth and nitrogen-removal performances of newly reported Pseudomonas glycinae CYCN-C under defined cultivation conditions. Transcriptome analysis was further used to investigate all pathway genes involved in nitrogen metabolism, lipid degradation and utilization, and cell growth at mRNA levels.ResultsCYCN-C could directly use fat, oil, and grease from kitchen waste (KFOG) as carbon source with TN removal efficiency of 73.5%, significantly higher than that (60.9%) with sodium acetate. The change levels of genes under defined KFOG and sodium acetate were analyzed by transcriptome sequencing. Results showed that genes cyo, CsrA, PHAs, and FumC involved in carbon metabolism under KFOG were significantly upregulated by 6.9, 0.7, 26.0, and 19.0-folds, respectively. The genes lipA, lipB, glpD, and glpK of lipid metabolic pathway were upregulated by 0.6, 0.4, 21.5, and 1.3-folds, respectively. KFOG also improved the denitrification efficiency by inducing the expression of the genes nar, nirB, nirD, and norR of denitrification pathways.ConclusionIn summary, this work firstly provides valuable insights into the genes expression of lipid-utilizing and denitrifying bacterium, and provides a new approach for sewage treatment with reuse of KFOG wastes.

Background Visual attention plays a crucial role in daily living and in sports, affecting an athlete’s performance and thus, potentially, the outcome of a match. However, studies assessing the association between the level of sports expertise and visual attention have yielded mixed results. This study was conducted to examine whether visual attention could be developed with increased sports expertise, and whether visual attention differed between male athletes and female athletes. Methods A total of 128 participants were included in this study: 64 first-level national soccer athletes recruited from college soccer teams (considered elite athletes; 32 men and 32 women with similar soccer performance requirements and training experience), and 64 physical education college students with limited soccer experience (considered novice athletes; 32 men and 32 women with matched soccer experience). To assess visual attention, we used a multiple object tracking (MOT) task with four targets among a total of 10 objects moving at a fixed speed of 10°/s in random directions across a computer monitor screen. Tracking accuracy on the MOT task was calculated for each participant as the proportion of correctly selected targets. A univariate analysis of variance was performed, with group (expert, novice) and sex (male, female) as independent variables, and tracking accuracy on the MOT task as the dependent variable to assess whether sports expertise or sex influenced visual attention. Simple effects tests followed by comparisons with Bonferroni corrections were used, and effect size calculations were performed using Cohen’s f statistic. Results Tracking accuracy on the MOT task was significantly affected by sports expertise (F(1,124) = 91.732, p < 0.001, ηP2 = 0.425), with accuracy among expert soccer athletes superior to that among novice soccer athletes. Abstract Low‐flow removal of refractory ascites is critical to treating cirrhosis and digestive system tumor, and thus, commercial ascites pump emerged lately. The rigid structure of clinically available pumps rises complication rate and lack of flow rate monitoring hinders early warning of abnormalities. Herein, a soft artificial system was proposed inspired by lymph for interactive ascites transfer with great biocompatibility. The implantable system is composed of pump cavity, valves and tubes, which are soft and flexible made by silica gel. Therefore, the system possesses similar modulus to tissues and can naturally fit surrounding tissues. The cavity with magnetic tablet embedded is driven by extracorporeal magnetic field. Subsequently, the system can drain ascites with a top speed of 23 mL min−1, much higher than that of natural lymphatic system and state‐of‐art devices. Moreover, integrated flexible sensors enable wireless, real‐time flow rate monitoring, serving as proof of treatment adjustment, detection and locating of malfunction at early stage. The liver function of experimental objects was improved, and no severe complications occurred for 4 weeks, which proved its safety and benefit to treatment. This artificial lymphatic system can serve as a bridge to recovery and pave the way for further clinical research.

59. Role of TET1-mediated epigenetic modulation in Alzheimer's disease

Alzheimer's disease (AD) is a neurodegenerative disorder influenced by a complex interplay of environmental, epigenetic, and genetic factors. DNA methylation (5mC) and hydroxymethylation (5hmC) are DNA modifications that serve as tissue-specific and temporal regulators of gene expression. TET family enzymes dynamically regulate these epigenetic modifications in response to environmental conditions, connecting environmental factors with gene expression. Previous epigenetic studies have identified 5mC and 5hmC changes associated with AD. In this study, we performed targeted resequencing of TET1 on a cohort of early-onset AD (EOAD) and control samples. Through gene-wise burden analysis, we observed significant enrichment of rare TET1 variants associated with AD (p = 0.04). We also profiled 5hmC in human postmortem brain tissues from AD and control groups. Our analysis identified differentially hydroxymethylated regions (DhMRs) in key genes responsible for regulating the methylome: TET3, DNMT3L, DNMT3A, and MECP2. To further investigate the role of Tet1 in AD pathogenesis, we used the 5xFAD mouse model with a Tet1 KO allele to examine how Tet1 loss influences AD pathogenesis. We observed significant changes in neuropathology, 5hmC, and RNA expression associated with Tet1 loss, while the behavioral alterations were not significant. The loss of Tet1 significantly increased amyloid plaque burden in the 5xFAD mouse (p = 0.044) and lead to a non-significant trend towards exacerbated AD-associated stress response in 5xFAD mice. At the molecular level, we found significant DhMRs enriched in genes involved in pathways responsible for neuronal projection organization, dendritic spine development and organization, and myelin assembly. RNA-Seq analysis revealed a significant increase in the expression of AD-associated genes such as Mpeg1, Ctsd, and Trem2. In conclusion, our results suggest that TET enzymes, particularly TET1, which regulate the methylome, may contribute to AD pathogenesis, as the loss of TET function increases AD-associated pathology.

61. Research on visual search behaviors of basketball players at different levels of sports expertise

Abstract This study aims to examine visual search strategies of skilled basketball players in an anticipation task. This study selected 48 experienced and inexperienced basketball players. The participants were grouped into novice and expert groups based on their experience. The participants were asked to look at series of pictures of offensive patterns of play in a basketball game from a third person perspective and chose one of the three options: passing, shooting and break through. This study measured and recorded the response time, key pressing results, and eye movements. Eye movement data were recorded using the Tobii X-3 120 eye tracker. The results showed that, the expert group demonstrated superior anticipation performance, and were more accuracy. In addition, the results showed that participants in the expert group and faster predicted the offensive way faster than the novice group. The results also showed that experienced basketball players employed a simple and efficient visual search strategy including greater fixation counts and longer fixation duration on more informative areas than the novice group. Furthermore, the visualization metrics showed that the expert group had a more concise fixation trajectory and focused mainly on key information area. Generally, expert players exhibited a more efficient and effective visual search strategy demonstrating better performance on anticipation tasks.

62. A novel promising diagnosis model for colorectal advanced adenoma and carcinoma based on the progressive gut microbiota gene biomarkers

Abstract Background Colorectal cancer (CRC), a commonly diagnosed cancer often develops slowly from benign polyps called adenoma to carcinoma. Altered gut microbiota is implicated in colorectal carcinogenesis. It is warranted to find non-invasive progressive microbiota biomarkers that can reflect the dynamic changes of the disease. This study aimed to identify and evaluate potential progressive fecal microbiota gene markers for diagnosing advanced adenoma (AA) and CRC. Results Metagenome-wide association was performed on fecal samples from different cohorts of 871 subjects (247 CRC, 234 AA, and 390 controls). We characterized the gut microbiome, identified microbiota markers, and further constructed a colorectal neoplasms classifier in 99 CRC, 94 AA, and 62 controls, and validated the results in 185 CRC, 140 AA, and 291 controls from 3 independent cohorts. 21 species and 277 gene markers were identified whose abundance was significantly increased or decreased from normal to AA and CRC. The progressive gene markers were distributed in metabolic pathways including amino acid and sulfur metabolism. A diagnosis model consisting of four effect indexes was constructed based on the markers, the sensitivities of the Adenoma Effect Index 1 for AA, Adenoma Effect Index 2 for high-grade dysplasia (HGD) adenoma were 71.3% and 76.5%, the specificities were 90.5% and 90.3%, respectively. CRC Effect Index 1 for all stages of CRC and CRC Effect Index 2 for stage III–IV CRC to predict CRC yielded an area under the curve (AUC) of 0.839 (95% CI 0.804–0.873) and 0.857 (95% CI 0.793–0.921), respectively. Combining with fecal immunochemical test (FIT) significantly improved the sensitivity of CRC Effect Index 1 and CRC Effect Index 2 to 96.7% and 100%. Conclusions This study reports the successful diagnosis model establishment and cross-region validation for colorectal advanced adenoma and carcinoma based on the progressive gut microbiota gene markers. The results suggested that the novel diagnosis model can significantly improve the diagnostic performance for advanced adenoma.

Abstract This study aimed to develop a self-made C-arm camera shooting locator and verify its accuracy and advantages. A total of 60 physicians and nurses from the Surgical System of Sanmen People’s Hospital, Zhejiang Province, China, were randomly selected as filming operators. The C-arm machine with a self-made locator and a C-arm machine without a locator were used to measure the center of the circular plate. The iron nails were used to shoot. The distance between the iron nail and the center point of the circular display area on display was defined as the shooting deviation. When it was less than 3 cm, the shooting was stopped. The number of shots, total shooting time, and first-shot deviation in the C-arm camera shooting groups with and without the locator were statistically analyzed, and the advantages and disadvantages of the two were compared. The average number of shots, average total shooting time, and average first-shot deviation of the C-arm camera using the locator were significantly better than those in the group without the locator, and the differences were statistically significant. When the shooting distance (X) was equal to 30 cm and the shooting angle (Y) was equal to 0°, the average number of shots, average total shooting time, and average first-shot deviation were optimal. The C-arm camera shooting locator can improve the shooting accuracy of the C-arm camera and effectively reduce the number of shots and total shooting time. Hence, it can be applied in clinical and surgical practice.

Abstract Background Emerging evidence suggests that gut microbiota plays a predominant role in Crohn’s disease (CD). However, the microbiome alterations in the early stage of CD patients still remain unclear. The present study aimed to identify dysbacteriosis in patients with early CD and explore specific gut bacteria related to the progression of CD. Methods This study was nested within a longitudinal prospective Chinese CD cohort, and it included 18 early CD patients, 22 advanced CD patients and 30 healthy controls. The microbiota communities were investigated using high-throughput Illumina HiSeq sequencing targeting the V3–V4 region of 16S ribosomal DNA (rDNA) gene. The relationship between the gut microbiota and clinical characteristics of CD was analyzed. Results Differential microbiota compositions were observed in CD samples (including early and advanced CD samples) and healthy controls samples. Notably, Lachnospiracea_incertae_sedis and Parabacteroides were enriched in the early CD patients, Escherichia/Shigella, Enterococcus and Proteus were enriched in the advanced CD patients, and Roseburia, Gemmiger, Coprococcus, Ruminococcus 2, Butyricicoccus, Dorea, Fusicatenibacter, Anaerostipes, Clostridium IV were enriched in the healthy controls [LDA score (log10) > 2]. Furthermore, Kruskal–Wallis Rank sum test results showed that Blautia, Clostridium IV, Coprococcus, Dorea, Fusicatenibacter continued to significantly decrease in early and advanced CD patients, and Escherichia/Shigella and Proteus continued to significantly increase compared with healthy controls (P 2]. Causal inference prevails in the field of laparoscopic surgery. Once the causality between an intervention and outcome is established, the intervention can be applied to a target population to improve clinical outcomes. In many clinical scenarios, interventions are applied longitudinally in response to patients’ conditions. Such longitudinal data comprise static variables, such as age, gender, and comorbidities; and dynamic variables, such as the treatment regime, laboratory variables, and vital signs. Some dynamic variables can act as both the confounder and mediator for the effect of an intervention on the outcome; in such cases, simple adjustment with a conventional regression model will bias the effect sizes. To address this, numerous statistical methods are being developed for causal inference; these include, but are not limited to, the structural marginal Cox regression model, dynamic treatment regime, and Cox regression model with time-varying covariates. This technical note provides a gentle introduction to such models and illustrates their use with an example in the field of laparoscopic surgery.

Extended range electric vehicles (EREVs) are an effective solution to solve the lack of driving range of pure electric vehicles. Reducing the fuel consumption of EREVs and prolonging the service lifetime of battery play a positive role in solving environmental pollution and energy crisis. In this paper, hybrid energy storage system (HESS) is introduced into EREVs, and two energy management strategies based on HESS are proposed to reduce fuel consumption and prolonging the service life of battery. This paper proposes two energy management strategies for EREVs based on a HESS to reduce fuel consumption and the amount of energy flowing through the battery. HESS adopts the rule-based energy management strategy, whereas Auxiliary power unit (APU) adopts the equivalent fuel consumption minimization strategy (ECMS) or the rule-based energy management strategy as its first and second hierarchical energy management strategies respectively, both of which are embedded into the charge depleting/charge sustaining (CD/CS) control strategy. A simulation model for an EREV is built in AVL Cruise to verify the effectiveness of these strategies. The battery charge and discharge C-rate and the energy consumption of EREVs without HESS under the ECMS and rule-based energy management strategy are then compared. Results show that under the first hierarchical energy management strategy, the average discharge C-rate and cumulative ampere hours of the battery are the lowest, thereby reducing its operating load and extending its service lifetime. Meanwhile, for those EREVs without HESS, using the rule-based energy management strategy can reduce fuel consumption by 11.33%, whereas using ECMS increases fuel consumption by no more than 1.61%. Therefore, the first hierarchical energy management strategy proposed in this study can effectively extend the service lifetime of the battery and reduce the fuel economy of EREVs.

Abstract Background Few studies focused on the risk factors for hand rehabilitation of intracerebral hemorrhage (ICH) using of soft robotic hand therapy (SRHT). The aim of this study was to establish a predictive nomogram for soft robotic hand rehabilitation in patients with ICH. Methods According to the Brunnstrom motor recovery (BMR) stage, the patients were grouped into poor and good motor function groups. The data of patient demographic information and serum level of C-terminal Agrin Fragment (CAF), S100B and neurofilament light (NfL) were collected. The logistic regression was used to analyze the risk factors for poor hand function. Results Finally, we enrolled 102 and 103 patients in the control and SRHT groups. For the SRHT group, there were 17 and 86 cases with poor and good motor function at 6-months follow-up respectively. Objective: The postoperative survival rates of patients with gastric cancer at different stages treated at the National Cancer Center (NCC) of China have not been reported. Therefore, we evaluated the survival of patients with gastric cancer who underwent surgery at the NCC from 2011 to 2018 to provide baseline information for further studies. Methods: We identified 7,301 patients with primary gastric carcinoma who underwent surgery at the NCC between January 2011 and May 2018; among these, 5,008 patients were enrolled in this study. Follow up was completed on April 30, 2020. We analyzed the 5-year survival according to the stage, sex, age, and other clinicopathological features. Results: Five-year survival rates of patients with pTNM stages IA, IB, IIA, IIB, IIIA, IIIB, IIIC, and IV treated at the NCC were 94.9%, 91.8%, 86.5%, 76.1%, 61.1%, 44.2%, 29.7%, and 8.1%, respectively, which were similar to those reported in Japan during the same period. Five-year survival rates of patients with ypTNM stages I, II, III, and IV were 93.1%, 63.2%, 27.2%, and 0.0%, respectively. In multivariable analysis, age, signet ring cell carcinoma, adjuvant chemotherapy, and degree of differentiation were revealed as important prognostic factors. Conclusion: The survival rates of patients with gastric cancer treated at the NCC have been significantly improved, reaching the levels of Japan in the same period.

Abstract Background Pyroptosis is a programmed cell death mediated by inflammasomes. Previous studies have reported that inhibition of neurokinin receptor 1 (NK1R) exerted neuroprotection in several neurological diseases. Herein, we have investigated the role of NK1R receptor inhibition using Aprepitant to attenuate NLRC4-dependent neuronal pyroptosis after intracerebral hemorrhage (ICH), as well as the underlying mechanism. Methods A total of 182 CD-1 mice were used. ICH was induced by injection of autologous blood into the right basal ganglia. Aprepitant, a selective antagonist of NK1R, was injected intraperitoneally at 1 h after ICH. To explore the underlying mechanism, NK1R agonist, GR73632, and protein kinase C delta (PKCδ) agonist, phorbol 12-myristate 13-acetate (PMA), were injected intracerebroventricularly at 1 h after ICH induction, and small interfering ribonucleic acid (siRNA) for NLRC4 was administered via intracerebroventricular injection at 48 h before ICH induction, respectively. Neurobehavioral tests, western blot, and immunofluorescence staining were performed. Results The expression of endogenous NK1R and NLRC 4 were gradually increased after ICH. NK1R was expressed on neurons. Aprepitant significantly improved the short- and long-term neurobehavioral deficits after ICH, which was accompanied with decreased neuronal pyroptosis, as well as decreased expression of NLRC4, Cleaved-caspase-1, GSDMD (gasdermin D), IL-1β, and IL-18. Activation of NK1R or PKCδ abolished these neuroprotective effects of Aprepitant after ICH. Similarly, knocking down NLRC4 using siRNA produced similar neuroprotective effects. Conclusion Aprepitant suppressed NLRC4-dependent neuronal pyroptosis and improved neurological function, possibly mediated by inhibition of NK1R/PKCδ signaling pathways after ICH. The NK1R may be a promising therapeutic target for the treatment of ICH.

Abstract Background Mycoplasma pneumoniae pneumonia (MPP) is a prevalent disease in community-acquired pneumonia among children. However, in addition to respiratory manifestations, it may also develop extra-pulmonary complications. Embolism is one of the uncommon extra-respiratory manifestations prone to severe sequelae and even death. This study aims to analyze the clinical features of MPP with embolism in children, and explore the associated risk factors of embolism in MPP patients. Methods A retrospective case–control analysis was performed on 48 children with MPP admitted to our hospital wards between January 2010 and December 2021. Embolism group comprised children with embolism by CTA or MRA results, whereas the non-embolism group comprised children with clinical suspicion of embolism but negative diagnostic imaging support. The clinical features, laboratory findings and imaging were analyzed to explore the risk factors for embolism in children with MPP. Results A total of 48 children with MPP were enrolled in the study (16 cases and 32 controls). In the embolism group, 10 patients (62.5%) had pulmonary embolism, 3 patients (18.75%) presented ventricle embolism, 2 patients (12.5%) presented cerebral and carotid artery embolism, one patient (6.25%) had a cerebral embolism, limb, and spleen, respectively. The univariate analysis revealed the maximum body temperature (Tmax), CRP, D-dimer (closest to CTA/MRA), the percentage of neutrophils (N%), pulmonary consolidation (⩾ 2/3 lobe), pleural effusion and atelectasis have significant differences between the embolism group and non-embolism group (P 3.55 mg/L [OR = 1.255 (95% CI: 1.025—1.537), P

Fushun west open-pit mine, which has a history of 100 years of mining, entered the pit-closing period. Due to the exploitation of resources, the west open-pit mine was threatened by geological disasters for a long time. In order to summarize the occurrence regularity of geological disasters in the west open-pit mine, ensure the safety of production and life in the mining area and near-mining city, and provide reference cases for disaster analysis and safety protection of other similar open-pit mines, the distribution law and influencing factors of landslides and ground fissures in west open-pit mine are studied and analyzed from time and space scale, based on the DPSR model, the corresponding measures of chain break disaster reduction and safety protection are put forward from three aspects: driving force response, pressure response and state response. Spatial-temporal distribution characteristics of landslide: in space, it first appeared in the western part of the southern slope in 1927. In recent hundred years, due to the driving influence of open-pit mining and underground mining, the main location of landslide developed from the western part of the southern slope to the western part of the northern slope, the western part of the northern slope, the central part of the northern slope and the eastern part of the northern slope. In recent years, large-scale rock slope sliding deformation occurred in the southern slope. Actually, from 1970 to 2000, landslide disasters occurred most frequently. After 2000, the frequency of landslide disasters decreased, and most of them occurred from May to September. Spatial-temporal distribution of ground fissures: the ground fissures near the north slope appeared after 1960, and accelerated in 1970s and 1980s, with the strike basically consistent with that of F1 and F1A faults; the ground fissures in the southern slope are distributed in the rear edge of the huge slip deformation body in the southern slope, which are arc-fan shaped and appeared in 2009 and developed rapidly after 2012. Through the analysis of disaster influencing factors, it is found that there is a chain relationship of homology and mutual causality between landslides and ground fissures, and the main factors affecting the occurrence of geological disasters in mining areas are geological structure and control of unfavorable engineering geological environment, driving of mining activities and inducing of rainfall.

Acute myeloid leukemia (AML) is a highly aggressive cancer with great heterogeneity and variability in prognosis. Though European Leukemia Net (ELN) 2017 risk classification has been widely used, nearly half of patients were stratified to “intermediate” risk and requires more accurate classification via excavating biological features. As new evidence showed that CD8+ T cell can kill cancer cells through ferroptosis pathway. We firstly use CIBERSORT algorithm to divide AMLs into CD8+ high and CD8+ low T cell groups, then 2789 differentially expressed genes (DEGs) between groups were identified, of which 46 ferroptosis-related genes associated with CD8+ T cell were sorted out. GO, KEGG analysis and PPI network were conducted based on these 46 DEGs. By jointly using LASSO algorithm and Cox univariate regression, we generated a 6-gene prognostic signature comprising VEGFA, KLHL24, ATG3, EIF2AK4, IDH1 and HSPB1. Low-risk group shows a longer overall survival. We then validated the prognostic value of this 6-gene signature using two independent external datasets and patient sample collection dataset. We also proved that incorporation of the 6-gene signature obviously enhanced the accuracy of ELN risk classification. Finally, gene mutation analysis, drug sensitive prediction, GSEA and GSVA analysis were conducted between high-risk and low-risk AML patients. Collectively, our findings suggested that the prognostic signature based on CD8+ T cell-related ferroptosis genes can optimize the risk stratification and prognostic prediction of AML patients.

This work focuses on the epitaxial growth process and kinetics of silicon-based Ir/yttria-stabilized zirconia (YSZ) bilayer films prepared by PLD and magnetron sputtering, including the sequential growth of a YSZ film, an Ir seed layer, and an Ir film on a silicon substrate. The results show that for growing YSZ films by PLD at the target-to-substrate distance of 60 mm, the higher the laser energy density and the oxygen pressure, the lower the laser frequency, the higher the epitaxy quality. In addition, for the preparation of the Ir-seed layer by PLD, background gas decelerates the plasma and suppresses self-sputtering and implantation, which is beneficial to obtaining a high-quality seed layer. In particular, introducing a small amount of oxygen is able to improve the epitaxial quality of the films during the growth of Ir-seed and Ir films, and the mechanism of oxygen is proposed. For the growth of Ir films by magnetron sputtering, the growth rate and the energy of argon ions increased with the increase of RF power, while the epitaxial quality is the opposite. The kinetic theory and simulation results demonstrate that this trend is mainly caused by the increase in the number of particles reaching the substrate without significantly increasing the distribution of high-energy particles. Finally, high-quality Ir (100) films on the YSZ (100) with a fresh surface are prepared. It is shown that increasing the epitaxy quality of the YSZ (100) film and the growth temperature of the Ir film can improve the quality of Ir film. Based on our results, the growth of Ir films on YSZ films with fresh surfaces resulting from in-situ argon plasma cleaning provides a new idea for preparing Ir (100)/YSZ (100) bilayer epitaxial film.

Background: Although traumatic brain injury (TBI) occurs in a very short time, the biological consequence of a TBI, such as Alzheimer’s disease, may last a lifetime. To date, effective interventions are not available to improve recovery from a TBI. Herein we aimed to ascertain whether recovery of neurosurgical high-frequency irreversible electroporation (HFIRE) injury in brain tissues can be accelerated by 7,8-dihydroxyflavone (7,8-DHF). Methods: The HFIRE injury was induced in the right parietal cortex of 8 adult healthy and neurologically intact male dogs. Two weeks before HFIRE injury, each dog was administered orally with or without 7,8-DHF (30 mg/kg) once daily for consecutive 2 weeks (n = 4 for each group). The values of blood-brain barrier (BBB) disruption, brain edema, and cerebral infarction volumes were measured. The concentrations of beta-amyloid, interleukin-1β, interleukin-6 and tumor necrosis factor-α in the cerebrospinal fluid were measured biochemically. Cortical neural dynamics mediate information processing for the cerebral cortex, which is implicated in fundamental biological processes such as vision and olfaction, in addition to neurological and psychiatric diseases. Spontaneous pain is a key feature of human neuropathic pain. Whether spontaneous pain pushes the cortical network into an aberrant state and, if so, whether it can be brought back to a “normal” operating range to ameliorate pain are unknown. Using a clinically relevant mouse model of neuropathic pain with spontaneous pain–like behavior, we report that orofacial spontaneous pain activated a specific area within the primary somatosensory cortex (S1), displaying synchronized neural dynamics revealed by intravital two-photon calcium imaging. This synchronization was underpinned by local GABAergic interneuron hypoactivity. Pain-induced cortical synchronization could be attenuated by manipulating local S1 networks or clinically effective pain therapies. Specifically, both chemogenetic inhibition of pain-related c-Fos–expressing neurons and selective activation of GABAergic interneurons significantly attenuated S1 synchronization. Clinically effective pain therapies including carbamazepine and nerve root decompression could also dampen S1 synchronization. More important, restoring a “normal” range of neural dynamics through attenuation of pain-induced S1 synchronization alleviated pain-like behavior. These results suggest that spontaneous pain pushed the S1 regional network into a synchronized state, whereas reversal of this synchronization alleviated pain.

A Maxwell coil is a gradient magnetic field source with high uniformity, and it is widely used in the experiment of magnetic effect in space gravitational wave detection, magnetic resonance molecular imaging, and other precision magnetic measurement experiments. In this paper, taking the square Maxwell coil as a research object, the influences of four kinds of assembly errors on the uniformity of magnetic field gradient (UOMFG) are systematically analyzed and the results are verified by simulation using the finite element method. We obtained variation rules of UOMFG changing with the four assembly errors and summarized three useful conclusions. These rules and conclusions are instructive in the design, processing, and assembly of the square Maxwell coil and are beneficial for the optimal utilization of the working area in the precision magnetic measurement experiments.

BackgroundPinus massoniana is an important timber species with high ecological and economic value in southern China and a pioneer species for the reforestation of barren mountains. The structure and function of the existing Masson pine ecosystem have been seriously affected by worsening habitats under current climate change. Its suitable habitat is likely to change greatly in the near future.MethodsTo estimate the potential geographic distribution of P. massoniana and its response to climate change, the Maxent model was selected to simulate the potentially suitable habitat and corresponding changes in the distribution pattern of P. massoniana under current and future climate scenarios (SSP1-2.6, SSP2-4.5, and SSP5-8.5) with two periods (2050s and 2090s).ResultsUnder current climate conditions, the total suitable habitat area of P. massoniana was 2.08 × 106 km2, including 0.76 × 106 km2 of highly suitable habitat, concentrated mainly in Jiangxi and Zhejiang, central and southeastern Hunan, northern Fujian, central and western Chongqing, southern Anhui, central and surrounding areas of Guangdong, central and eastern Guangxi, and northern Taiwan. The areas of moderately suitable habitat and poorly suitable habitat were 0.87 × 106 km2 and 0.45 × 106 km2, respectively. Temperature and precipitation appear to be the most important predictors: precipitation of the driest month (14.7–215.6 mm), minimum temperature of the coldest month (−3.5–13.8°C), annual temperature range (8.1–32.9°C), and mean temperature of the warmest quarter (23.6–34.7°C). P. massoniana are predicted to expand their potential distribution under future climate change: by the end of this century, their total suitable habitat area increased 0.26 × 106 km2 (10.61%) and 0.45 × 106 km2 (17.05%) under the most moderate (SSP1-2.6) and severe (SSP5-8.5) warming scenarios, respectively, by mainly extending northward.ConclusionUnder the different future climate scenarios, the total suitable habitat area of P. massoniana increased by mainly extending northward. Overall, our study clarifies the potential habitat distribution of P. massoniana and provides a critical empirical reference for future P. massoniana conservation and planting practices.

Abstract Background Achalasia is a rare primary esophageal motility disorder disease. It is reported that the long-term effect of fully coated anti-reflux metal stent (FCARMS) implantation is satisfactory. Operated by a skilled and experienced endoscopist, the effect of per-oral endoscopic myotomy (POEM) treatment is equivalent to that of surgical myotomy. So far, there is still few evidence to prove FCARMS implantation or POEM which is better for achalasia. The choice of treatment for achalasia is still controversial. The aim of this study is to find a more suitable therapy for achalasia by comparing the efficacy of FCARMS implantation and POEM. Methods A propensity score (PS) matching (1:2) was used in this retrospective cohort study. Data collected from consecutive patients of Achalasia, receiving FCARMS implantation or POEM therapy at the department of gastroenterology, the Seventh Medical Center of the Chinese People’s Liberation Army General Hospital from May 2007 to May 2018. According to their previous treatment, they are divided into two groups, FCARMS group and POEM group. Clinical efficacy and complications were compared between the two groups. Results A total of 166 cases were collected, including 113 cases of FCARMS and 53 cases of POEM. By PS matching, 150 patients were enrolled (100 cases of FCARMS and 50 cases of POEM). The adequate assessment of the spontaneous combustion and oxidation characteristics of coking coal can help to reduce its spontaneous combustion potential. In this study, the natural ignition period of the Hex coking coal seam at Ping Coal was calculated based on the programmed-temperature-rise test and the thermal property parameters measured during the spontaneous combustion of the coal. Typical gas concentrations were measured at different oxygen concentrations and coal particle sizes to investigate the changes in the oxygen, carbon–oxygen, and hydrocarbon gas concentrations during the low-temperature oxidation of the coking coal and to determine the indicator gases at different oxidation stages. The following results were obtained: The minimum critical oxygen concentration required for spontaneous ignition was 8%, and fire prevention measures should be implemented below 200 °C. When the temperature of the coal sample reaches 50–60 °C, the rates of CO and CO2 production increase, and when the temperature of the coal sample reaches 100–120 °C, the spontaneous combustion and oxidation of the coal generates alkane gas, in which the coal particle size has a negligible effect on the concentration of each generated gas. CO and C2H4 were selected as the indicator gases for different coking coal oxidation stages, and C2H6 and the C2H4/C2H6 ratio were used as secondary indicators to assist in the analysis. Utilizing the enhanced mathematical model for the shortest spontaneous combustion period of coal seams, in conjunction with a programmed-temperature test device, experimental calculations were conducted to determine the adiabatic spontaneous combustion period. The results indicate that the natural ignition period for the Hex coking coal seam at Ping Coal is approximately 60 days, representing a brief timeframe, and the coal seam is characterized by a high risk of spontaneous combustion.

Human movement recognition is the use of perceptual technology to collect some of the limb or body movements presented. This practice involves the use of wireless signals, processing, and classification to identify some of the regular movements of the human body. It has a wide range of application prospects, including in intelligent pensions, remote health monitoring, and child supervision. Among the traditional human movement recognition methods, the widely used ones are video image-based recognition technology and Wi-Fi-based recognition technology. However, in some dim and imperfect weather environments, it is not easy to maintain a high performance and recognition rate for human movement recognition using video images. There is the problem of a low recognition degree for Wi-Fi recognition of human movement in the case of a complex environment. Most of the previous research on human movement recognition is based on LiDAR perception technology. LiDAR scanning using a three-dimensional static point cloud can only present the point cloud characteristics of static objects; it struggles to reflect all the characteristics of moving objects. In addition, due to its consideration of privacy and security issues, the dynamic millimeter-wave radar point cloud used in the previous study on the existing problems of human body movement recognition performance is better, with the recognition of human movement characteristics in non-line-of-sight situations as well as better protection of people’s privacy. In this paper, we propose a human motion feature recognition system (PNHM) based on spatiotemporal information of the 3D point cloud of millimeter-wave radar, design a neural network based on the network PointNet++ in order to effectively recognize human motion features, and study four human motions based on the threshold method. The data set of the four movements of the human body at two angles in two experimental environments was constructed. This paper compares four standard mainstream 3D point cloud human action recognition models for the system. The experimental results show that the recognition accuracy of the human body’s when walking upright can reach 94%, the recognition accuracy when moving from squatting to standing can reach 84%, that when moving from standing to sitting can reach 87%, and the recognition accuracy of falling can reach 93%.

The premutation of the fragile X messenger ribonucleoprotein 1 (FMR1) gene is characterized by an expansion of the CGG trinucleotide repeats (55 to 200 CGGs) in the 5’ untranslated region and increased levels of FMR1 mRNA. Molecular mechanisms leading to fragile X-premutation-associated conditions (FXPAC) include cotranscriptional R-loop formations, FMR1 mRNA toxicity through both RNA gelation into nuclear foci and sequestration of various CGG-repeat-binding proteins, and the repeat-associated non-AUG (RAN)-initiated translation of potentially toxic proteins. Such molecular mechanisms contribute to subsequent consequences, including mitochondrial dysfunction and neuronal death. Clinically, premutation carriers may exhibit a wide range of symptoms and phenotypes. Any of the problems associated with the premutation can appropriately be called FXPAC. Fragile X-associated tremor/ataxia syndrome (FXTAS), fragile X-associated primary ovarian insufficiency (FXPOI), and fragile X-associated neuropsychiatric disorders (FXAND) can fall under FXPAC. Understanding the molecular and clinical aspects of the premutation of the FMR1 gene is crucial for the accurate diagnosis, genetic counseling, and appropriate management of affected individuals and families. This paper summarizes all the known problems associated with the premutation and documents the presentations and discussions that occurred at the International Premutation Conference, which took place in New Zealand in 2023.

In this study, the research aim is to enhance the activity index of activated coal gangue and study its activation mechanism. The activation process of coal gangue was optimized through orthogonal tests, and the Back-Propagation (BP) neural network model was improved using a genetic algorithm. With the effects of grinding duration, calcination temperature, and calcination duration, the morphological changes and phase transformation processes of coal gangue were studied at the micro and meso levels to clarify the activation mechanism. The results indicated that the effect of calcination temperature on the strength activity index of coal gangue was most significant, followed by grinding duration and calcination duration. The potential activity of coal gangue can be effectively stimulated through mechanical and thermal activation, and the content of potential active minerals in coal gangue powders was also increased. The activation process of coal gangue for the optimal scheme was obtained as grinding at 76 min first and thermal treatment at 54 min at 749 °C. As the thermal activation under 950 °C, some unstable external hydroxyls, and internal hydroxyls in kaolinite from coal gangue were removed, the AlⅥ-O octahedron was destroyed, and kaolinite was transformed into spatially disordered metakaolinite with very high activity.

Abstract In advancing age, some individuals maintain a stable cognitive performance over time, while others experience a rapid decline. Such variation in cognitive trajectory is only partially explained by common neurodegenerative pathologies. Hence, we aimed to identify new molecular processes underlying variation in cognitive trajectory using brain microRNA profile followed by an integrative analysis with brain transcriptome and proteome. Individual cognitive trajectories were derived from longitudinally assessed cognitive-test scores of older-adult brain donors from four longitudinal cohorts. Postmortem brain microRNA profiles, transcriptomes, and proteomes were derived from the dorsolateral prefrontal cortex. The global microRNA association study of cognitive trajectory was performed in a discovery (n = 454) and replication cohort (n = 134), followed by a meta-analysis that identified 6 microRNAs. Among these, miR-132-3p and miR-29a-3p were most significantly associated with cognitive trajectory. They explain 18.2% and 2.0% of the variance of cognitive trajectory, respectively, and act independently of the eight measured neurodegenerative pathologies. Furthermore, integrative transcriptomic and proteomic analyses revealed that miR-132-3p was significantly associated with 24 of the 47 modules of co-expressed genes of the transcriptome, miR-29a-3p with 3 modules, and identified 84 and 214 downstream targets of miR-132-3p and miR-29a-3p, respectively, in cognitive trajectory. This is the first global microRNA study of cognitive trajectory to our knowledge. We identified miR-29a-3p and miR-132-3p as novel and robust contributors to cognitive trajectory independently of the eight known cerebral pathologies. Our findings lay a foundation for future studies investigating mechanisms and developing interventions to enhance cognitive stability in advanced age.

Hydrogen sulfide (H2S) has emerged as a signaling molecule that plays a crucial role in the postharvest preservation of fruits and vegetables. This review summarizes the various functions of H2S such as delaying ripening and senescence, enhancing the resistance to cold and disease, and emphasizes the underlying mechanisms. Appropriate concentrations of H2S primarily operate through stimulating the antioxidant system, while showing positive effects on physiological metabolism relevant to storage quality and shelf life including energy, sugar, proline, phenolic, membrane lipid and cell wall metabolism. Moreover, H2S may reduce storage loss by modulating the expression patterns of senescence-related genes, like those linked to ethylene. The coordination of H2S and nitric oxide (NO) combats ethylene-derived negative effects during ripening and senescence. High concentrations of H2S not only act as a regulator to induce disease resistance, but also as a fungicide to inhibit the growth and pathogenicity of fungi. The intricate crosstalk between H2S and other molecules exists via synergistic and antagonistic roles based on protein persulfidation, which is the major signaling process of H2S and appears to compete with other post-translational modifications (PTMs) for the same cysteine residues. This review summarizes H2S synthesis pathways and also discusses the correlation between the signals of H2S, Ca2+ and ABA, while highlighting the intrinsic mechanisms of H2S performing its functions in postharvest preservation.

The co-chaperone DnaJ plays an important role in protein folding and regulation of various physiological activities, and participates in several pathological processes. DnaJ has been extensively studied in many species including humans, drosophila, mushrooms, tomatoes, and Arabidopsis. However, few studies have examined the role of DnaJ in wheat (Triticum aestivum), and the interaction mechanism between TaDnaJs and plant viruses. Here, we identified 236 TaDnaJs and performed a comprehensive genome-wide analysis of conserved domains, gene structure and protein motifs, chromosomal positions and duplication relationships, and cis-acting elements. We grouped these TaDnaJs according to their domains, and randomly selected six genes from the groups for tissue-specific analysis, and expression profiles analysis under hormone stress, and 17 genes for plant virus infection stress. In qRT-PCR, we found that among the 17 TaDnaJ genes tested, 16 genes were up-regulated after wheat yellow mosaic virus (WYMV) infection, indicating that the TaDnaJ family is involved in plant defense response. Subsequent yeast two-hybrid assays verified the WYMV NIa, NIb and 7KD proteins interacted with TaDJC (TraesCS7A02G506000), which had the most significant changes in gene expression levels after WYMV infection. Insights into the molecular mechanisms of TaDnaJ-mediated stress tolerance and sensitivity could inform different strategies designed to improve crop resistance to abiotic and biotic stress. This study provides a basis for future investigation of the TaDnaJ family and plant defense mechanisms.

Abstract. Variants in the solute carrier family 40 member 1 (SLC40A1) gene are the molecular basis of ferroportin disease, which is an autosomal dominant hereditary hemochromatosis. Here, we present a patient with pure red cell aplasia (PRCA) and large granular lymphocytic leukemia (LGLL) associated with an extremely high levels of serum ferritin and iron overload syndrome. Whole exon sequencing revealed a novel heterozygous variant in SLC40A1 (p.T419I), which was found in his daughter as well. A series of functional studies in vitro of the T419I variant in ferroportin were conducted and the results revealed a reduced capacity of iron export from cells without changes in protein localization and its sensitivity to hepcidin. Intracellular iron storage in mutated cells was significantly higher than that of wild-type. These findings suggest that the novel variant p.T419I can cause the classical form of ferroportin disease and an elevated intracellular iron level indicates a potential novel pathogenic mechanism underlying PRCA and LGLL.

The expeditious development of the digital economy has posed critical challenges for tax compliance. Recent reforms and technological changes, such as the emergency of platform data sharing, perform as potential instruments to potentially solve the tax compliance issue in the digital economy. This article innovatively combines tax compliance and digital economy taxation and proposes an expected utility theory model and a multiagent simulation model of tax compliance in the context of the digital economy, incorporating an important impact factor—platform data quality. In addition, the discrepancy and applicable conditions of three audit modes have been first compared and analyzed in this research: unconstrained signal audit, constrained random audit, and constrained signal audit. Applying computational experiments, this paper further investigates the effect of platform data quality on tax compliance in the digital economy. The key findings include: (1) the signal role of platform data is affected by three factors—the quality of platform data, the audit intensity and audit rate; (2) the signal role of platform data can be negative in given situations; and (3) the tax compliance rate varies in three different audit modes, which depend on the platform data quality, audit rate, and audit intensity. Based on the findings, this article provides policy recommendations on tax collection and management in digital economy.

Simulated reality encompasses virtual, augmented, and mixed realities—each characterized by different degrees of truthfulness in the visual perception: “all false,” “coexistence of true and false,” and “difficult distinction between true and false,” respectively. In all these technologies, however, the temperature rendering of virtual objects is still an unsolved problem. Undoubtedly, the lack of thermal tactile functions substantially reduces the quality of the user’s real-experience perception. To address this challenge, we propose theoretically and realize experimentally a technological platform for the in situ simulation of thermal reality. To this purpose, we design a thermal metadevice consisting of a reconfigurable array of radiating units, capable of generating the thermal image of any virtual object, and thus rendering it in situ together with its thermal signature. This is a substantial technological advance, which opens up new possibilities for simulated reality and its applications to human activities.

94. Mechanisms of chilling tolerance in melatonin treated postharvest fruits and vegetables: a review

Cold storage of fruits is the best strategy to extend shelf life. However, storage temperature below 8°C mostly causes chilling injury (CI). Thus, exogenous melatonin treatment is an alternative safe preservative to alleviate CI and induce cold tolerance in postharvest of fruits and vegetables. This paper reviewed the recent progresses of melatonin-mediated physiological, biochemical and molecular mechanisms on chilling tolerance of fruits and vegetables. Among cold tolerance mechanisms, melatonin substantially enhances enzymatic and non-enzymatic antioxidants removing reactive oxygen species (ROS). Melatonin triggered phenol accumulation which associated with 2,2-diphenyl-1picryl hydrazyl (DPPH) scavenging capacity and reduction of oxidative damage. In addition, melatonin extensively maintained intracellular energy and enhanced energy regulation associated enzymes, thereby maintaining cellular stability. Melatonin induces unsaturated/saturated fatty acid (unSFA/SFA) ratio, improving cellular fluidity. Besides, melatonin regulates arginine metabolisms via increased polyamines, proline and γ-aminobutyric acid (GABA) contents. Furthermore, melatonin possibly encodes calcium-dependent protein kinases (CDPK) and mitogen-activated protein kinases (MAPK) cascades. Moreover, it substantially induces Ca2+ signaling transduction pathways. As part of the signaling pathway, melatonin activates C-repeat binding factors (CBFs) as transcriptional factors for cold acclimation. Therefore, this review concluded that exogenous melatonin improves chilling stress tolerance by coordinating biochemical and molecular mechanisms at post-harvest life of fruits and vegetables and extending shelf life.

Objectives: We evaluated the contribution of lung lesion quantification on chest CT using a clinical Artificial Intelligence (AI) software in predicting death and intensive care units (ICU) admission for COVID-19 patients. Methods: For 349 patients with positive COVID-19-PCR test that underwent a chest CT scan at admittance or during hospitalization, we applied the AI for lung and lung lesion segmentation to obtain lesion volume (LV), and LV/Total Lung Volume (TLV) ratio. ROC analysis was used to extract the best CT criterion in predicting death and ICU admission. Two prognostic models using multivariate logistic regressions were constructed to predict each outcome and were compared using AUC values. The first model (“Clinical”) was based on patients’ characteristics and clinical symptoms only. The second model (“Clinical+LV/TLV”) included also the best CT criterion. Results: LV/TLV ratio demonstrated best performance for both outcomes; AUC of 67.8% (95% CI: 59.5 - 76.1) and 81.1% (95% CI: 75.7 - 86.5) respectively. Regarding death prediction, AUC values were 76.2% (95% CI: 69.9 - 82.6) and 79.9% (95%IC: 74.4 - 85.5) for the “Clinical” and the “Clinical+LV/TLV” models respectively, showing significant performance increase (+ 3.7%; p-value

In this study, we demonstrate the direct growth of high-quality h-BN films with a controllable thickness on sapphire substrates by using the pulsed laser deposition (PLD) technique. The effects of the deposition conditions and laser parameters on the growth of the h-BN films are systematically investigated by evaluating their characteristic Raman peaks. Among the various growth parameters studied, the substrate temperature has the greatest influence on the crystalline quality of the h-BN films, and the optimal pressure varies depending on the target-substrate distance. The h-BN film grown under optimal conditions exhibits a narrow Raman line width of ∼30 cm−1, indicating a high crystalline quality. The photodetectors fabricated from the PLD-grown h-BN films exhibit superior deep-ultraviolet detection performance with a large on/off ratio of >104, high photoresponsivity, and a sharp cut-off wavelength of 220 nm. This study presents the possibility of producing high-quality h-BN films by applying PLD on dielectric substrates for optoelectronic applications.

97. Melatonin treatment induces chilling tolerance by regulating the contents of polyamine, γ-aminobutyric acid, and proline in cucumber fruit

The mechanism of melatonin (MT) induced chilling tolerance in harvested cucumber fruit was investigated at commercial maturity. In this study, cucumber fruits were treated with 100 µmol L–1 MT at 4°C and 90% relative humidity for 15 d of storage. In comparison with the control, cucumber treatment with MT resulted in reduced chilling injury (CI), decreased electrolyte leakage and enhanced firmness. The fruits treated with MT showed higher chlorophyll contents in storage conditions with suppressed chlorophyllase enzyme activity. MT treatment increased arginine decarboxylase (ADC) and ornithine decarboxylase (ODC) enzyme activities. Moreover, enhanced expression of the Cucumis sativus ADC (CsADC) and C. sativus ODC (CsODC) genes resulted in the accumulation of polyamine contents. Similarly, proline levels exhibited higher levels among treated fruits. Meanwhile, the proline synthesizing enzymes △1-pyrroline-5-carboxylate syntheses (P5CS) and ornithine aminotransferase (OAT) were significantly increased, while a catabolic enzyme of proline dehydrogenase (PDH) activity was inhibited by treatment. In addition, MT induced expression of C. sativus OAT (CsOAT) and C. sativus P5CS (CsP5CS) genes. Cucumber fruits treated with MT also exhibited higher γ-aminobutyric acid (GABA) content by enhanced GABA transaminase (GABA-T) and glutamate decarboxylase (GAD) enzyme activities and a higher C. sativus GAD (CsGAD) gene expression. To sum up, the results show that MT treatment enhanced chilling tolerance, which was associated with the regulation of polyamines, as well as proline and γ-aminobutyric acid.

Abstract Background Hypoxic–ischemic encephalopathy (HIE) is a severe anoxic brain injury that leads to premature mortality or long-term disabilities in infants. Neuroinflammation is a vital contributor to the pathogenic cascade post-HIE and a mediator to secondary neuronal death. As a plasma membrane G-protein-coupled receptor, GPR39, exhibits anti-inflammatory activity in several diseases. This study aimed to explore the neuroprotective function of GPR39 through inhibition of inflammation post-hypoxic–ischemic (HI) injury and to elaborate the contribution of sirtuin 1(SIRT1)/peroxisome proliferator-activated receptor-γ coactivator 1α (PGC-1α)/nuclear factor, erythroid 2 like 2(Nrf2) in G-protein-coupled receptor 39 (GPR39)-mediated protection. Methods A total of 206 10-day-old Sprague Dawley rat pups were subjected to HIE or sham surgery. TC-G 1008 was administered intranasally at 1 h, 25 h, 49 h, and 73 h post-HIE induction. SIRT1 inhibitor EX527, GPR39 CRISPR, and PGC-1α CRISPR were administered to elucidate the underlying mechanisms. Brain infarct area, short-term and long-term neurobehavioral tests, Nissl staining, western blot, and immunofluorescence staining were performed post-HIE. Results The expression of GPR39 and pathway-related proteins, SIRT1, PGC-1α and Nrf2 were increased in a time-dependent manner, peaking at 24 h or 48-h post-HIE. Intranasal administration of TC-G 1008 reduced the percent infarcted area and improved short-term and long-term neurological deficits. Moreover, TC-G 1008 treatment significantly increased the expression of SIRT1, PGC-1α and Nrf2, but downregulated the expressions of IL-6, IL-1β, and TNF-α. GPR39 CRISPR EX527 and PGC-1α CRISPR abolished GPR39’s neuroprotective effects post-HIE. Conclusions TC-G 1008 attenuated neuroinflammation in part via the SIRT1/PGC-1α/Nrf2 pathway in a neonatal rat model of HIE. TC-G 1008 may be a novel therapeutic target for treatment post-neonatal HIE injury.

Optical deep learning based on diffractive optical elements offers unique advantages for parallel processing, computational speed, and power efficiency. One landmark method is the diffractive deep neural network (D2NN) based on three-dimensional printing technology operated in the terahertz spectral range. Since the terahertz bandwidth involves limited interparticle coupling and material losses, this paper extends D2NN to visible wavelengths. A general theory including a revised formula is proposed to solve any contradictions between wavelength, neuron size, and fabrication limitations. A novel visible light D2NN classifier is used to recognize unchanged targets (handwritten digits ranging from 0 to 9) and targets that have been changed (i.e., targets that have been covered or altered) at a visible wavelength of 632.8 nm. The obtained experimental classification accuracy (84%) and numerical classification accuracy (91.57%) quantify the match between the theoretical design and fabricated system performance. The presented framework can be used to apply a D2NN to various practical applications and design other new applications.

Abstract In recent years, addressing ill-posed problems by leveraging prior knowledge contained in databases on learning techniques has gained much attention. In this paper, we focus on complete three-dimensional (3D) point cloud reconstruction based on a single red-green-blue (RGB) image, a task that cannot be approached using classical reconstruction techniques. For this purpose, we used an encoder-decoder framework to encode the RGB information in latent space, and to predict the 3D structure of the considered object from different viewpoints. The individual predictions are combined to yield a common representation that is used in a module combining camera pose estimation and rendering, thereby achieving differentiability with respect to imaging process and the camera pose, and optimization of the two-dimensional prediction error of novel viewpoints. Thus, our method allows end-to-end training and does not require supervision based on additional ground-truth (GT) mask annotations or ground-truth camera pose annotations. Our evaluation of synthetic and real-world data demonstrates the robustness of our approach to appearance changes and self-occlusions, through outperformance of current state-of-the-art methods in terms of accuracy, density, and model completeness.
