Abstract Background Although knowledge of the sizes, contents, and forms of plant mitochondrial genomes (mitogenomes) is increasing, little is known about the mechanisms underlying their structural diversity. Evolutionary information on the mitogenomes of Primula, an important ornamental taxon, is more limited than the information on their nuclear and plastid counterparts, which has hindered the comprehensive understanding of Primula mitogenomic diversity and evolution. The present study reported and compared three Primula mitogenomes and discussed the size expansion of mitogenomes in Ericales. Results Mitogenome master circles were sequenced and successfully assembled for three Primula taxa and were compared with publicly available Ericales mitogenomes. The three mitogenomes contained similar gene contents and varied primarily in their structures. The Primula mitogenomes possessed relatively high nucleotide diversity among all examined plant lineages. In addition, high nucleotide diversity was found among Primula species between the Mediterranean and Himalaya-Hengduan Mountains. Most predicted RNA editing sites appeared in the second amino acid codon, increasing the hydrophobic character of the protein. An early stop in atp6 caused by RNA editing was conserved across all examined Ericales species. The interfamilial relationships within Ericales and interspecific relationships within Primula could be well resolved based on mitochondrial data. Transfer of the two longest mitochondrial plastid sequences (MTPTs) occurred before the divergence of Primula and its close relatives, and multiple independent transfers could also occur in a single MTPT sequence. Foreign sequence [MTPTs and mitochondrial nuclear DNA sequences (NUMTs)] uptake and repeats were to some extent associated with changes in Ericales mitogenome size, although none of these relationships were significant overall. Conclusions The present study revealed relatively conserved gene contents, gene clusters, RNA editing, and MTPTs but considerable structural variation in Primula mitogenomes. Relatively high nucleotide diversity was found in the Primula mitogenomes. In addition, mitogenomic genes, collinear gene clusters, and locally collinear blocks (LCBs) all showed phylogenetic signals. The evolutionary history of MTPTs in Primula was complicated, even in a single MTPT sequence. Various reasons for the size variation observed in Ericales mitogenomes were found.

The rare Primula brachystoma W.W.Sm. is an endemic species confined to Gaoligong mountain of Chinese-Burma frontier, which has been rediscovered from the same region after nearly 100 years. In total, 11 specimens from Gaoligong Mountain have been found in the herbaria worldwide, since its first collection in 1920 by Farrer, Reginald John. Previously, this species was described as homostylous but our finding shows the species also exhibited heterostyly. A complete description of the species, the distribution, morphological comparison and identification key from closely related species are provided here. Primula jiaozishanensis Z.K.Wu, W.H.Yang & Yuan Wu, a new species of Primulaceae is described and illustrated from Jiaozi Snow Mountain of Dongchuan District, Yunnan, China. Morphological evidence supports P. jiaozishanensis as a member of P. sect. Petiolares subsect. Davidii, which is characterized by firmly papery or leathery leaves, with veins impressed adaxially, often prominently raised and alveolate abaxially. The new species is characterized by having long and stout rhizomes, smaller leaves with short petioles, short or almost obsolete scape, and larger flowers. Hybridization is a key mechanism involved in lineage diversification and speciation, especially in ecosystems that experienced repeated environmental oscillations. Recently radiated plant groups, which have evolved in mountain ecosystems impacted by historical climate change provide an excellent model system for studying the impact of gene flow on speciation. We combined organellar (whole-plastome) and nuclear genomic data (RAD-seq) with a cytogenetic approach (rDNA FISH) to investigate the effects of hybridization and introgression on evolution and speciation in the genus Soldanella (snowbells, Primulaceae). Pervasive introgression has already occurred among ancestral lineages of snowbells and has persisted throughout the entire evolutionary history of the genus, regardless of the ecology, cytotype, or distribution range size of the affected species. The highest extent of introgression has been detected in the Carpathian species, which is also reflected in their extensive karyotype variation. Introgression occurred even between species with dysploid and euploid cytotypes, which were considered to be reproductively isolated. The magnitude of introgression detected in snowbells is unprecedented in other mountain genera of the European Alpine System investigated hitherto. Our study stresses the prominent evolutionary role of hybridization in facilitating speciation and diversification on the one hand, but also enriching previously isolated genetic pools. Primula vialii Delavay ex Franch. (1905) is an alpine species with an ornamental value. In this study, we sequenced, assembled, and annotated the chloroplast genome of P. vialii. The results showed that it was a double-stranded, closed circular DNA with 154,897 bp in length, comprising a small single-copy (SSC) region of 17,766 bp, a large single-copy (LSC) region of 85,379 bp and a pair of inverted repeat (IR) regions of 25,876 bp. A total of 113 unique genes were annotated, including 79 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. The phylogenetic analysis revealed that P. vialii is closely related to Primula flaccida. The cp genomic data will be useful for systematics and evolutionary studies of Primula. Primula sugongii J.D.Ya, Bin Yang & Y.H. Tan and P. zhengyii Bin Yang & Y.H. Tan, two new species of Primulaceae from Yunlong County and Luchun County, Yunnan Province, China, are described based on morphological characters and phylogenetic evidence, and colour plates of these two species are also provided. The molecular phylogenetic analysis using the nuclear ribosomal internal transcribed spacer (nrITS) and chloroplast genome, Primula sugongii was nested within the clade with most members of sect. Obconicolisteri, for Primula zhengyii, the phylogenetic position and its affiliation is somewhat controversial. Based on the comprehensive morphological evidence, they are affiliated to Primula sect. Obconicolisteri Balf. f. Primula sugongii is morphologically similar to P. barbicalyx C.H. Wright in having short and pilose scapes and calyx, but it can be easily distinguished by its longer corolla tube and corolla exannulate at throat, and series of other characters of the leaf blade. Primula zhengyii is morphologically similar to P. zhui Y.H. Tan & Bin Yang and P. intanoensis T. Yamaz., but clearly differs from P. zhui by its shorter corolla tube and smaller flowers size and differs from P. intanoensis by its distylous flowers and several other characters of leaf blade. The chemical composition and anti-inflammatory activity of the endemic Lysimachia baviensis were investigated for the first time in this study. A phytochemical fractionation of the methanol extract of L. baviensis resulted in the isolation of a new stilbene (bavienside A, 1) and two new chalcone glycosides (baviensides B and C, 2 and 3). Their structures were elucidated via the interpretation of NMR and HRESIMS spectroscopic data. Compounds 1–3 strongly inhibited the production of nitric oxide in LPS-induced RAW264.7 cells with the IC50 values of 6.23, 2.86 and 3.51 μM, respectively. The C-acetylstilbene and carbomethyl chalcone structures in compound 1 and 3 were found for the first time from natural source and could be important markers for chemotaxonomy of Lysimachia baviensis. Timing of seed germination is directly related to the survival probability of seedlings. For alpine plants, autumn-dispersal seeds should not germinate immediately because the cold temperature is not conducive to the survival of seedlings. Seed dormancy is a characteristic of the seed that prevents it from germinating after dispersal. Primula florindae is an alpine perennial forb endemic to eastern Tibet, SW China. We hypothesized that primary dormancy and environmental factors prevent seeds of P. florindae to germinate in autumn and allow them to germinate at the first opportunity in spring. We determined how GA3, light, temperature, dry after-ripening (DAR) and cold-wet stratification (CS) treatments affect seed germination by conducting a series of laboratory experiments. Firstly, the effects of gibberellic acid (GA3; 0, 20, and 200 mg L-1) on germination of freshly shed seeds at alternating temperatures (15/5 and 25/15 ℃) were immediately investigated to characterize seed with a physiological dormancy component. Then, the fresh seeds treated with 0, 3, and 6 months of after-ripening (DAR) and cold-wet stratification (CS) were incubated at seven constant (1, 5, 10, 15, 20, 25, and 30 ℃) and two alternating temperatures (5/1, 15/5, and 25/15 ℃) at light and dark conditions. Fresh seeds were dormant, which only germinated well (>60%) at 20, 25, and 25/15 ℃ in light but not at -15 °C and to higher percentages in light than in dark. GA3 increased germination percentage of fresh seeds, and DAR or CS treatments increased final germination percentage, germination rate (speed), and widened the temperature range for germination from high to low. Moreover, CS treatments reduced the light requirement for germination. Thus, after dormancy release, seeds germinated over a wide range of constant and alternating temperatures, regardless of light conditions. Our results demonstrated that P. florindae seeds have type 2 non-deep physiological dormancy. Timing of germination should be restricted to early spring, ensuring a sufficient length of the growing season for seedling recruitment. Primula pingbaensis Na Zhang, X.Q.Jiang & Z.K.Wu, a new species of Primulaceae from Gaofeng Mountain of Pingba county, Guizhou, China, is described and illustrated. Morphological evidence supports P pingbaensis as a member of P. sect. Petiolares on account of scape elongating, pedicels conspicuously thickening in fruit, and its capsule cracking irregularly round the top and crumbling away. Amongst the members of subsect. Davidii, the new species is characterized by having a uniquely smooth leaf blade due to inconspicuously raised veinlets and homostylous flowers with the style usually extending beyond the anthers. Ardisia anamalaiana is a new species described from Anamalai Hills, the southern Western Ghats of India. The new species is morphologically similar to A. gardneri C.B.Clarke, but differs in its leaves, petiole, inflorescence, bract, flower and pedicel. Ardisia anamalaiana is described and illustrated in detail. Morphological comparison between the new species and related taxa, a key to the genus Ardisia from the Western Ghats, colour photographs and conservation status is also provided. Labisia pumila is a traditional medicinal herb that is widely used by women to firm the uterus after childbirth. Recent scientific studies have also proven its pharmacological importance. However, the comparison of phytochemicals in the different varieties of the herb is very limited in literature. Therefore, 3 common varieties of L. pumila leaves were extracted in a reflux system, and consequently the crude extract was fractionated using reversed phase solid phase extraction (SPE) into individual fractions according to solvent polarity. L. pumila var. alata (LPA) showed the highest yield of crude extract and total phenolics content which was also in line with high DPPH radical scavenging activity. While, L. pumila var. pumila with red leaves (LPPR) exhibited higher total flavonoids and triterpenoids than its green leaves counterpart (LPPG). Higher antioxidant capacity was possibly due to the presence of plant co-pigments in LPPR. Glycosylated flavones including orientin were putatively detected in the intermediate polar fraction of LPA based on mass fragments. However, 0 % methanol (polar) and 100 % methanol (less polar) fractions notably contained compounds competing oxygen with nitric oxide radicals from the formation of nitrite ions. Proton NMR spectral analysis proposed N-[1-(2,3-dihydro-1,4-benzodioxin-6-yl)-1-hydroxy-3- (morpholin-4-yl)propan-2-yl]decanamide and 4-[({1,3-bis[(2-ethylpenta-3,4-dien-1-yl)oxy]propan-2- yl}oxy)methyl]hexa-1,2-diene in the 0 % methanol and 100 % methanol fractions, respectively. This study concluded that the plant variety of alata was more antioxidative than pumila. The detection of orientin and its derivatives are first time reported in LPA. A new species, Primula luquanensis Z.K.Wu and Wei Zhou sp. nov. (Primulaceae) is described and illustrated from Yunnan Province, China. It is morphologically assigned to P. sect Aleuritia based on its dwarf and hairless habit and coverage by farina on both sides of the leaf blade and scape. This new species is similar to P. nutantiflora and P. yunnanensis, but it is easily distinguished by its stolons, solitary bract, bell-shaped corolla and monomorphic floral form. A new species, Lysimachia fenghwaiana G.Hao & H.F.Yan (Primulaceae), from Hunan Province, China, is described and illustrated. This new species belongs to Lysimachia subgen. Lysimachia sect. Nummularia and is morphologically similar to L. crista-galli and L. carinata, but is distinctive in its leaf shape and arrangement of flowers. It can be further distinguished from L. crista-galli by the absence of calyx lobule spur, and from L. carinata by the black glandular striates in the corolla lobes, rather than punctate. This paper describes and illustrates a new species of Primulaceae, Primula wolongensis sp. nov. from Wolong National Nature Reserve in Sichuan Province, China. It is very rare and currently only known from its type locality. The new species belongs to subsection Chartacea of the section Petiolares on account of lacking bud scales at flowering, being efarinose and having distinct petiolate leaves with more or less rounded lamina. The new species can be differentiated from other members of the subsection by leaf blade margin dentate, and leaf veins which are not raised, scape shorter than or equal to pedicels, yellow flowers and location of stamens of the corolla tube at thrum flower. Molecular phylogenetic analysis based on nuclear ribosome internal transcribed spacer (nrITS) demonstrated that P. wolongensis was sister to subgen. Auriculastrum. Primula wolongensis is currently known from a single location in Wolong Town, and its conservation status is assessed as Data Deficient (DD). [ABSTRACT FROM AUTHOR]

To investigate the light intensity suitable for the growth of Ardisia gigantifolia Stapf, morphology, photosynthetic parameters, and indicators of oxidative stress were analyzed under different light intensities. Compared to highirradiance treatment, medium and low-irradiance treatments promoted plant growth and restricted transpiration. Compared to medium irradiance, plants under high and low irradiance exhibited significantly lower maximal photochemical efficiency, potential photochemical efficiency, and electron transport rate, but significantly higher malondialdehyde content. This indicated that both excessive light and severe shading inhibited photosynthetic activity and induced oxidative stress, which resulted in a significant decrease in net photosynthetic rate. A. gigantifolia can adapt to different light intensities, improving light harvesting and utilizing capacity under low irradiance by increasing Chl (a+b) content and reducing Chl a/b ratio, and adapting to high irradiance by enhancing heat dissipation and activity of peroxidase. Primula veris (cowslip) is an important medicinal–aromatic plant used traditionally for its expectorant and anti-inflammatory properties, as well as a valuable horticultural plant with ornamental value and agroalimentary interest (edible flowers and leaves). With extensive illegal harvesting across northern Greece to date, the long-term survival of Greek cowslip wild-growing populations seems compromised. With the aim to characterize the phytochemical content of locally adapted native genotypes of P. veris subsp. veris, we examined samples from 13 wild-growing populations sourced from 8 mountain areas along a longitudinal gradient of northern Greece. Flowers, leaves, and roots were separately evaluated with HPLC-PDA-MS for their chemical content to select the genotypes associated with the best phytochemical traits. The flowers were found to contain mainly flavonoids, isorhamnetin, quercetin, and kaempferol triglycosides, with varied population contents, generally higher in northwestern population samples with a colder and more humid climate; however, all in line with the European Medicines Agency’s (EMA) reports. The leaves were characterized by the prevalence of kaempferol and quercetin triglycosides, being generally higher than that of the flowers. In the roots, saponins were detected in relatively low percentages, and the phenolic glycosides were found up to nearly 2%. The results of this study suggest a potential specificity of the marginal Greek genotypes being at the edge of the species’ native distribution range in Europe and Asia. The phytochemical characterization herein supports the domestication process of Greek native cowslip genotypes. In turn, this may alleviate the overharvesting pressure in wild populations, thus contributing to species conservation and offering an incentive for farmers to sustainably cultivate at local scales well-adapted indigenous genotypes of high value.

A new species, Lysimachia coriacea, from Chongqing, China, is described and illustrated. It is assigned to subgen. Lysimachia sect. Nummularia ser. Paridiformes and resembles L. paridiformis var. stenophylla, but is characterised by smaller leathery leaves with black glandular striations near the margin. It is also similar to L. nanpingensis in its two to three pairs of leaves sub-whorled at the stem apices, but differs by smaller leathery leaves and densely glandular stem, petiole and pedicel, and calyx lobes with sparse black glandular stripes. The syntypes (H.R.E. Handel-Mazzetti 8896 and H.R.E. Handel-Mazzetti 9280) of Androsace mollis Hand.-Mazz. are identified as two separate taxa based on critical examinations of herbarium specimens and field investigation. While H.R.E. Handel-Mazzetti 8896 has been designated as the lectotype of A. mollis, we describe the other taxon, represented by H.R.E. Handel-Mazzetti 9280, as A. chimingiana Y.Xu & G.Hao sp. nov. A new species, Primula surculosa, is described and illustrated. In gross morphology, it is clearly allied to section Petiolares and is most similar to P. taliensis from the group Taliensis, but is distinctive in its indumentum in the throat of the corolla tube, and the markedly stoloniferous habit. Hottonia palustris L. is from the genus Hottonia (Primulaceae), and the understanding of its phytochemical and pharmacological properties is limited. In this study, the use of chromatographic techniques led to the isolation of a further eleven compounds, including three new flavonoids: 2′,5-dihydroxyflavone 2′-O-β-glucopyranoside, 5,6-dihydroxyflavone 6-O-(6"-O-glucopyranosyl)-β-glucopyranoside (hottonioside A), and 4′,5,7-trihydroxyflavone 7-O-(2"-O-β-glucuronide)-β-glucopyranoside. Their structures were determined using extensive 1D and 2D NMR data and mass spectrometry (HRMS). The qualitative assessment of the chemical composition of the investigated extracts and fractions was performed using the LC-HRMS technique. Furthermore, the antioxidant potential of extracts, fractions, and compounds and their ability to inhibit acetylcholinesterase were also evaluated. Thus, we may conclude that the observed biological effects are the result of the presence of many biologically active compounds, of which dibenzoylmethane is the most active. Lysimachia arvensis var. caerulea (L.) Turland & Bergmeier, a variety belonging to the family Primulaceae is reported as new to the state flora of Assam, northeastern India. The authors encountered the specimen during field survey at Kamrup Rural District of Assam in 2020. Primula odontocalyx (Franch.) Pax (1905) is a perennial herb of the genus Primula in Primulaceae with heterostyly and ornamental value. Here, the chloroplast genome of P. odontocalyx was sequenced, assembled, and annotated. The complete chloroplast genome was a closed-circular molecule of 151,738 bp in length, containing a large single-copy region (LSC) of 83,817 bp, a small single-copy region (SSC) of 17,529 bp, and two inverted repeat (IR) regions of 25,196 bp. A total of 115 unique genes were annotated in the whole cp genome, including 81 protein-coding genes, 4 rRNA genes, and 30 tRNA genes. Phylogenetic analysis confirmed the close relationship between P. odontocalyx and Primula moupinensis, and both species belong to Sect. Petiolares Pax. [ABSTRACT FROM AUTHOR]

The rare plant Primula esquirolii , a karst cave species in the family Primulaceae, endemic to Guizhou, China, and with a presumed extremely small population, had not previously been observed since 1910. It is categorized as Endangered on the China Species Red List. In surveys during 2018–2022, we were unable to locate the species in its type location in Pingba county, but we discovered a previously unrecorded population of 44 mature individuals, 37 of which were long homostylous and seven of which were heterostylous. These individuals were discovered in Xiuwen county in February 2022, in an area of c. 50 m2, c. 40 km from the type location. All seven heterostylous individuals showed the short-styled morph. The almost homostylous P. esquirolii potentially faces the same high genetic load as other homostylous plants, and this may have led to its small population size and potential sensitivity to habitat destruction. Field surveys and informal interviews with local people indicated that the main threats to this species are its small population size and the loss of suitable habitat as a result of human activities. The most urgent requirement for the conservation of this species is to protect the habitat of this single known extant population and to carry out ex situ conservation. Primula chishuiensis C.M. Hu, G. Hao & Y. Xu sp. nov. (Primulaceae) is described and illustrated from Guizhou, China. It is morphologically belonging to P. sect. Monocarpicae and allied to P. centellifolia and P. scopulicola, but can be distinguished by its homostylous flowers and broadly campanulate calyx which split to 3/4 of its length, and the broadly lanceolate lobes. Active and passive restoration treatments were monitored for wetland vegetation recruitment on a large river floodplain in southwestern Montana. Species richness gains at the end of the monitoring periods were greatest in passively restored rich fen, extremely rich fen stream terraces, and alkaline meadows adjoining fens. Restored hydrology resulted in removal of exotics and natives intolerant of longer hydroperiods and recruitment of natives adapted to fully restored hydrology. Both active and passive treatments resulted in establishment of 5 rare species from borrowed sod, redistributed soils, and undisturbed in situ seedbanks. Seed characteristics and dormancy types can influence seed longevity in native wetland seedbanks, which can contain cryptic diversity in refugium populations of some rare species appearing after restoration treatments. A phytochemical study of the methanol extract of the fruit of Maesa lanceolata resulted in the isolation of a new alkenylbenzoquinone (1), alongside the known compounds (Z)-2,5-dihydroxy-6-methyl-3-(pentadec-10'-enyl)-1,4-benzoquinone (2), 2,5-dihydroxy-6-methyl-3-(nonadec-14'-enyl)-1,4-benzoquinone (3), 2,5-dihydroxy-6-methyl-3-(tridecyl)-1,4-benzoquinone (4), (2S,3S,4R,2'R,9E)-[2'-hydroxytetraeicosanoyl]-2-aminooctadec-9-ene-1,3,4-triol (5), monopalmitin (glyceryl palmitate) (6), lupeol (7), and 3-O-(β-D-glucopyranoside)-β-sitosterol (8). The structures of the compounds were established by the means of spectroscopic (1 D- and 2 D-NMR) and spectrometric techniques (MS). The isolated compounds were assessed for their antibacterial, cytotoxic, and antiradical activities. Compound 2 showed moderate activity against Staphylococcus warneri (DSMZ 20036), while the other compounds were inactive. The bulb flora of prefecture (nomos) Ilias in NW Peloponnese, Greece is documented with an emphasis on its distribution within the administrative unit. Four new compounds, 5-hydroxy-2′,6′-dimethoxyflavone (4), 5-hydroxy-2′,3′,6′-trimethoxyflavone (5), 5-dihydroxy-6-methoxyflavone (6), and 5,6′-dihydroxy-2′,3′-dimethoxyflavone (7), and three known compounds, 1,3-diphenylpropane-1,3-dione (1), 5-hydroxyflavone (2), and 5-hydroxy-2′-methoxyflavone (3), were isolated from the aerial parts of Hottonia palustris. Their chemical structures were determined through the use of spectral, spectroscopic and crystallographic methods. The quantitative analysis of the compounds (1–7) and the zapotin (ZAP) in methanol (HP1), petroleum (HP6), and two chloroform extracts (HP7 and HP8) were also determined using HPLC-PDA. The biological activity of these compounds and extracts on the oral squamous carcinoma cell (SCC-25) line was investigated by considering their cytotoxic effects using the MTT assay. Subsequently, the most active compounds and extracts were assessed for their effect on DNA biosynthesis. It was found that all tested samples during 48 h treatment of SCC-25 cells induced the DNA biosynthesis-inhibitory activity: compound 1 (IC50, 29.10 ± 1.45 µM), compound 7 (IC50, 40.60 ± 1.65 µM) and extracts ZAP (IC50, 20.33 ± 1.01 µM), HP6 (IC50, 14.90 ± 0.74 µg), HP7 (IC50, 16.70 ± 0.83 µg), and HP1 (IC50, 30.30 ± 1.15 µg). The data suggest that the novel polymethoxyflavones isolated from Hottonia palustris evoke potent DNA biosynthesis inhibitory activity that may be considered in further studies on experimental pharmacotherapy of oral squamous cell carcinoma. Lysimachia sedoides, a rare new species of Primulaceae from a karst cave in northern Guangxi, China, is described and illustrated. It is similar to L. rupestris F.H.Chen & C.M.Hu in having rosette leaves and copiously glandular dots on young parts of shoot apex, but differs markedly in the smaller plant size, fleshy leaf blade, with a size of 10-30 × 3-8 mm, and the length of offsets never exceeding 5 cm. Chemical investigation of the Vietnamese plant Aegiceras floridum Roem. & Schult. (Primulaceae) led to the isolation of the new compound 3-methoxy-5-nonylphenol (1) along with five known ones 2,8,10-trihydroxy-6H-benzo[c]chromen-6-one (2), 2-hydroxy-5-methoxy-3-nonylbenzo-1,4-quinone (3), 5-(3-hydroxypropyl)-7-methoxy-3-(methylbenzofuran-2-yl)-3-methoxyphenol (4), 2,8-dihydroxy-7-methoxy-3,9-diundecyldibenzofuran-1,4-dione (5) and 10-hydroxy-4-methoxy-2,11-diundecylgomphilactone (6). The structures were elucidated by analysis of their HRESIMS and NMR data as well as the comparison of their NMR data with those reported in the literature. The cytotoxic activity of selected isolated compounds against some cancer cell lines such as human epithelial carcinoma (HeLa), human lung cancer (NCI-H460), liver hepatocellular carcinoma (HepG2), human breast cancer (MCF-7), and acute T cell leukemia (Jurkat) was evaluated. Among them, 3 showed moderate activities against MCF-7 with an IC50 of 17.77 µM and NCI-H460 with an IC50 of 25.02 µM. Background and aimsSoil fertility and topographic microclimate are common determinants of plant species distributions. However, biotic conditions also vary along these abiotic gradients, and may mediate their effects on plants. In this study, we investigated whether soils and topographic microclimate acted directly on the performance of a focal understorey plant, or indirectly via changing biotic conditions.MethodsWe examined direct and indirect relationships between abiotic variables (soil fertility and topographic microclimate) and biotic factors (overstorey and understorey cover, litter depth and mycorrhizal colonization) and the occurrence, density and flowering of a common understorey herb, Trientalis latifolia, in the Klamath-Siskiyou Mountains, Oregon, USA.ResultsWe found that the positive effects of soil fertility on Trientalis occurrence were mediated by greater overstorey shading and deeper litter. However, we did not find any effects of topographic microclimate on Trientalis distribution that were mediated by the biotic variables we measured. The predictive success of Trientalis species distribution models with soils and topographic microclimate increased by 12 % with the addition of the biotic variables.ConclusionsOur results reinforce the idea that species distributions are the outcome of interrelated abiotic gradients and biotic interactions, and suggest that biotic conditions, such as overstorey density, should be included in species distribution models if data are available.

Primula xinningensis Wei Zhang bis & J.W.Shao, a new species from Hunan Province, China, is described. Its leaf morphology is similar to the P. merrilliana complex and flower morphology similar to P. cicutariifolia, but it can be distinguished from the former by the black pollen sac, corolla lobes apex obviously emarginate and can be differed from the latter by cotyledon triangular obovate, plants densely covered with glandular hairs and special habitat (karst caves). The whole plastid genome of this new species is 151, 601–151, 630 bp in length. Based on the whole plastid genome sequences, phylogenetic trees revealed that the new species did not genetically relate to the above two mentioned morphologically similar species, but it was closely related to P. hubeiensis. Currently, only three populations were discovered within a small distribution area, thus, it is preliminarily considered as Vulnerable (VU) according to criteria of the IUCN Red List. Flower colour has played a decisive role as an evolutionary force in many groups of plants by driving speciation. A well-known example of colour polymorphism is found across the Mediterranean populations of Lysimachia arvensis and L. monelli , in which blue- and red-flowered plants can be found. Previous studies recognized two lineages within L. arvensis differing in flower colour, but this variation has not yet been considered in a phylogenetic context. We have reconstructed the ancestral states of flower colour across Mediterranean Lysimachia spp. aiming at understanding its phylogenetic signal using the nuclear internal transcribed spacer (ITS) sequences and three plastid markers. All blue- and red-flowered specimens were nested in different clades in the ITS tree, thus supporting that L. arvensis and L. monelli are polyphyletic, whereas low phylogenetic resolution was found in plastid markers. Monophyly was reconstructed for blue-flowered L. arvensis and L. monelli samples, and similarly for red-flowered individuals of each species: (1) blue-flowered L. arvensis was reconstructed as sister to the strictly blue-flowered L. talaverae in a monophyletic clade sister to remaining Lysimachia ; (2) red-flowered L. arvensis was resolved as sister to red-flowered L. monelli in a monophyletic clade; and (3) clade 2 was sister to blue-flowered L. monelli and the strictly blue-flowered L. foemina. Our results suggest that colour lineages in L. arvensis and L. monelli constitute different species, but flower colour did not promote the separation of these lineages. We propose a new name for blue-flowered L. arvensis (L. loeflingii) and a new combination for red-flowered L. monelli (L. collina), maintaining L. arvensis for red-flowered plants and L. monelli for blue-flowered plants. Ardisia kteniophylla (Primulaceae) is highly valued in traditional medicine due to its production of the pharmacologically active secondary metabolites, especially triterpenoid saponins in its roots. Although A. kteniophylla is very important in traditional medicine, the genetic basis for its production of triterpenoid saponins remains largely unknown. Therefore, we sequenced transcriptomes of A. kteniophylla to identify putative genes involved in production of triterpenoid saponins in both leaves and roots, and we used the transcriptomes to compare expression levels of these genes between the two organ systems. The production of triterpenoid saponins in plants is usually induced through hormonal signaling on account of the presence of pests. Thus, we treated plants with the hormones salicylic acid (SA) and methyl jasmonate (MeJA) and used quantitative real‐time PCR (qRT‐PCR) to investigate expression levels of genes involved in triterpenoid saponin biosynthesis. In total, we obtained transcriptomes for leaf and root tissues representing 52,454 unigenes. Compared with the leaf transcriptome, we found that 6092 unigenes were upregulated in the root, especially enzymes involved in the direct synthesis of triterpenoid saponins, while 6001 genes appeared downregulated, including those involved in precursory steps in the triterpenoid saponin biosynthesis pathway. Our results from qRT‐PCR indicate that genes within the upstream parts of the triterpenoid saponin biosynthesis pathway may be upregulated under exposure to the applied hormones, but downstream genes are downregulated. This suggests possible conflicting effects of SA and MeJA in promoting the production of secondary metabolites on the one hand, and, on the other, limiting plant growth processes to devote energy to combating pests. We also performed an analysis of transcription factors (TFs) and found 997 unique transcripts belonging to 16 TF families. Primula longipilosa from SW Yunnan, China, is described as a species new to science and illustrated. The systematic placement of this new species is also discussed based on an nrITS molecular tree. It is morphologically most similar to P. mollis, but differs from the latter in its racemose inflorescence, green calyx tube, pink to pink rose corolla, stamens at 1/3 length above the base of the corolla tube and applanate globose capsule. The genus Primula (Primulaceae) comprises more than 500 species, with 300 species distributed in China. The contradictory results between systematic analyses and morphology-based taxonomy make taxonomy studies difficult. Furthermore, frequent introgression between closely related species of Primula can result in non-monophyletic species. In this study, the complete chloroplast genome of sixteen Primula obconica subsp. obconica individuals were assembled and compared with 84 accessions of 74 species from 21 sections of the 24 sections of the genus in China. The plastome sizes of P. obconica subsp. obconica range from 153,584 bp to 154,028 bp. Genome-wide variations were detected, and 1915 high-quality SNPs and 346 InDels were found. Most SNPs were detected in downstream and upstream gene regions (45.549% and 41.91%). Two cultivated accessions, ZP1 and ZP2, were abundant with SSRs. Moreover, 12 SSRs shared by 9 accessions showed variations that may be used as molecular markers for population genetic studies. The phylogenetic tree showed that P. obconica subsp. obconica cluster into two independent clades. Two subspecies have highly recognizable morphological characteristics, isolated geographical distribution areas, and distinct phylogenetic relationships compared with P. obconica subsp. obconica. We elevate the two subspecies of P. obconica to separate species. Our phylogenetic tree is largely inconsistent with morphology-based taxonomy. Twenty-one sections of Primula were mainly divided into three clades. The monophyly of Sect. Auganthus, Sect. Minutissimae, Sect. Sikkimensis, Sect. Petiolares, and Sect. Ranunculoides are well supported in the phylogenetic tree. The Sect. Obconicolisteri, Sect. Monocarpicae, Sect. Carolinella, Sect. Cortusoides, Sect. Aleuritia, Sect. Denticulata, Sect. Proliferae Pax, and Sect. Crystallophlomis are not a monophyletic group. The possible explanations for non-monophyly may be hybridization, polyploidization, recent introgression, incorrect taxonomy, or chloroplast capture. Multiple genomic data and population genetic studies are therefore needed to reveal the evolutionary history of Primula. Several species of the genus Lysimachia L. (Primulaceae) are well known in folk medicine, but very few studies have been conducted on them. The aim of our study was to investigate the microscopical characteristics and chemical composition of Lysimachia insignis Hemsl., L. decurrens G. Forst., and L. congestiflora Hemsl. as well as to evaluate some biological activities of their methanol extracts. The transverse sections of stem and powder characteristics of the three Lysimachia species shared some similarities. Also, some differences were indicated. The chemical investigation of three aforesaid species including the preliminary phytochemical screening, the quantification of the total polyphenol content and total flavonoid content, as well as the determination of metabolites. The phytochemical screening showed the presence of flavonoids, saponins, tannins and sterols in all the three species. On the other hand, coumarins were detected in L. insignis while anthranoids were present in L. decurrens and L. congestiflora. The total polyphenol content and the total flavonoid content of the aforesaid species were determined by the spectrometric method. The metabolite profiling approach of three species was performed by LC-Q-ToF-HRMS and HPTLC. The methanol extract of L. insignis showed a weak inhibitory effect on NO production (1% = 57.4 at the concentration of 100 [zg/ml) while the extract of L. decurrens exhibited moderate anti-oxidant activity in vitro (SC% = 62.5 at the concentration of 100 pg/mL). Phytochemical investigation of the methanol extract of the aerial parts of Lysimachia laxa led to the isolation of four new oleanane-type saponins, lysimosides A-D (1-4) and one known compound, lysimachigenoside B (5). Their structures were elucidated using a combination of HR-ESI-MS, 1D and 2D-NMR spectral data, chemical methods, and comparison with previous literature. The cytotoxic activity of these compounds was evaluated against human lung cancer (A-549) and human breast cancer (MCF-7) cell lines. All compounds exhibited cytotoxic activity against A-549 and MCF-7 cell lines with IC 50 values ranging from 6.1-16.0 μM, comparable to the positive control, mitoxantrone. The escalating climate imbalance, coupled with rising water demands in rapidly expanding urban areas, is forcing scientists and policymakers to seek alternative strategies for efficient water resource management. Nature Based Solutions (NBS) are gaining prominence due to their ability to provide multiple ecosystem services. However, the quantification of benefits and drawbacks mediated by different vegetation species remains inadequate. In this study, we investigated the performance of a pot-based vertical greening system (VGS) designed to integrate the functions of green facades with those of treatment wetlands. The VGS was vegetated with Mentha aquatica L. (hereafter Mentha), Oenanthe javanica (Blume) DC. (hereafter Oenanthe) and Lysimachia nummularia L. (hereafter Lysimachia), and their respective effects on water balance and mass removal of common greywater pollutants were compared. Results indicated that VGS lines vegetated with Oenanthe and Mentha exhibited comparable pollutant removals. Oenanthe showed a preference for greywater that had already undergone partial treatment, while Mentha was not affected by any pollutant load in water removal -48.1 % of total inflow- and in nutrients uptake in aboveground biomass -14.3 % N and 7 % P- due to sustained and robust growth, outperforming Oenanthe and Lysimachia. This study examined effects of mangrove plants Kandelia obovata and Aegiceras corniculatum on harmful algal species. While A. corniculatum leaf extract had no inhibitory effect, K. obovata leaf extract significantly inhibited the growth of two harmful algal species Alexandrium tamarense and Karenia mikimotoi. The inhibitory effect was concentration-dependent, with over 90 % inhibition at the highest concentration. Morphological changes and cell size reduction were observed in both microalgae. Excessive production of reactive oxygen species and damage to algal photosynthetic system were found. The allelopathic effect of K. obovata on K. mikimotoi with low-concentration repeated exposure was more effective than high-concentration single exposure. The EC 50 of K. obovata (0.33 g L -1 ) was lower than reported values on other coastal plants. Higher inhibitory effects of K. obovata were found on naked algal species than the armoured ones. Primula calliantha subsp. bryophila (Balf. f. et Farrer) W.W. Smith and Forrest (1928) is a perennial alpine species with ornamental value. It is distributed in northwestern Yunnan and adjacent eastern Tibet of China, and northern Myanmar. Here, we sequenced and assembled complete plastid genome of P. calliantha subsp. bryophila, which is a circular molecule of 152,045 bp in length, including a large single-copy region (83,966 bp), a small single-copy region (17,663 bp), and a pair of inverted repeats (25,208 bp). The chloroplast genome contained 113 genes, including 79 protein-coding genes, four rRNA genes, and 30 tRNA genes. The phylogenetic tree based on chloroplast genomes showed the relative relationship of P. calliantha subsp. bryophila and P. calliantha, which further supports P. calliantha subsp. bryophila as a subspecies of P. calliantha in taxonomy. The complete chloroplast (cp) genome of P. calliantha subsp. bryophila provides valuable data for further phylogenetic studies of Primulaceae. [ABSTRACT FROM AUTHOR]

A replacement name Ardisia lallaniana R.Kr. Singh is here proposed for the illegitimate A. montana King & Gamble, being a later homonym of A. montana C.F. Gaertn. Also, the name A. zeylanica C.B. Clarke is a later homonym of A. zeylanica (Gaertn.) Lam. ex Forsyth f. and for the former, Tinus clarkeana Kuntze is the only available legitimate name. Therefore, Ardisia clarkeana (Kuntze) R.Kr. Ardisia phankelociana (Primulaceae) is restricted to the limestone areas in Ha Giang and Bac Kan provinces, northeastern Vietnam. Its vegetative characters and fruits were first described in 2017, but not the flowers, which are most important to distinguish the species from morphologically similar ones. Primula nghialoensis D.W.H. Rankin (Primulaceae) is reduced to the synonym of P. wenshanensis Chen & C.M. Hu. A new species, P. tsaiana G. Hao & Y. The genus Ardisia (Myrsinoideae, Primulaceae) has 16 subgenera and over 700 accepted names, mainly distributed in tropical Asia and America. The circumscription of Ardisia is not well-defined and sometimes confounded with the separation of some small genera. A taxonomic revision focusing on Ardisia and allies is necessary. In the Ardisia subgenus Crispardisia, symbiotic association with leaf-nodule bacteria is a unique character within the genus. The endosymbionts are vertically transmitted, highly specific and highly dependent on the hosts, suggesting strict cospeciation may have occurred in the evolutionary history. In the present study, we aimed to establish a phylogenetic framework for further taxonomic revision. We also aimed to test the cospeciation hypothesis of the leaf-nodulate Ardisia and their endosymbiotic bacteria. Nuclear ITS and two chloroplast intergenic spaces were used to reconstruct the phylogeny of Asian Ardisia and relatives in Myrsinoideae, Primulaceae. The 16S-23S rRNA were used to reconstruct the bacterial symbionts' phylogeny. To understand the evolutionary association of the Ardisia and symbionts, topology tests and cophylogenetic analyses were conducted. The molecular phylogeny suggested Ardisia is not monophyletic, unless Sardiria, Hymenandra, Badula and Oncostemum are included. The results suggest the generic limit within Myrsinoideae (Primulaceae) needs to be further revised. The subgenera Crispardisia, Pimelandra, and Stylardisia were supported as monophyly, while the subgenus Bladhia was separated into two distant clades. We proposed to divide the subgenus Bladhia into subgenus Bladhia s.str. and subgenus Odontophylla. Both of the cophylogenetic analyses and topology tests rejected strict cospeciation hypothesis between Ardisia hosts and symbiotic Burkholderia. Cophylogenetic analyses showed general phylogenetic concordance of Ardisia and Burkholderia, and cospeciation events, host-switching events and loss events were all inferred. [ABSTRACT FROM AUTHOR]

In boreal species, populations at a southern range edge are at risk of being extirpated due to rising temperatures from climate change and potentially from demographic factors that affect small populations. The ability to track climate through phenological responsiveness can affect survival and reproductive success, with implications for long-term population persistence. This study examines phenological responsiveness and fecundity in populations of an understory plant species, Trientalis borealis, in the Midwestern USA using herbarium specimens and field observations. Date of flowering was well predicted by the mean May temperature. We detected a shift in the mean flowering date for populations north of latitude 45° N of an average of 1.4 days earlier per decade. We did not find shifts in mean flowering date for populations south of 45° N. Trientalis borealis exhibits phenological responsiveness differently in different parts of its range. In populations north of latitude 45° N, T. borealis advances its date of flowering, on average, by approximately 3 days for every 1 °C increase in mean May temperature. In populations south of 45° N, T. borealis advances its flowering date less than 1 day for every 1 °C increase in mean May temperature. Flowering and seed set increased along a latitudinal gradient away from the range edge. If population growth is limited by seed production in this species, this suggests that populations near the southern range limit are not performing well, perhaps due to direct effects of warming, a lack of phenological responsiveness that exacerbates exposure to warming, or a lack of genetic diversity.
