A key drawback of hydrogel materials for tissue engineering applications is their characteristic swelling response, which leads to a diminished mechanical performance. However, if a solution can be found to overcome such limitations, there is a wider application for these materials. Herein, we describe a simple and effective way to control the swelling and degradation rate of nucleophilic thiol-yne poly(ethylene glycol) (PEG) hydrogel networks using two straightforward routes: (1) using multiarm alkyne and thiol terminated PEG precursors or (2) introducing a thermoresponsive unit into the PEG network while maintaining their robust mechanical properties. In situ hydrogel materials were formed in under 10 min in PBS solution at pH 7.4 without the need for an external catalyst by using easily accessible precursors. Both pathways resulted in strong tunable hydrogel materials (compressive strength values up to 2.4 MPa) which could effectively encapsulate cells, thus highlighting their potential as soft tissue scaffolds. Biodegradable polyester fibers have been loaded with two engineered analogues of gramicidin soviet. In these cyclic peptide derivatives, which were designed in a previous work to stabilize the bioactive conformation while enhancing the antimicrobial activity, the D-Phe was replaced by D-Pro, and the L-Pro was changed by 1-aminocyclopropanecarboxylic acid (Ac3c) or by an Ac3c derivative with two vicinal phenyl substituents in a trans relative disposition (S,S-c3diPhe). The diameter, topography, thermal stability and wettability of the polyester fibers, which have been obtained by electrospinning, strongly depend on the molecular constraints and stability of the loaded peptides. More specifically, unloaded and linear gramicidin-loaded fibers (used as control) are hydrophobic, rough and micrometric, while fibers loaded with the cyclic peptides are hydrophilic, ultra-smooth, nanometric and less thermally stable. The activity of the two cyclic peptides increases when loaded into polyester fibers, suggesting that the polymeric matrix stabilizes the bioactive b-sheet structure. The peptide with S,S-c3diPhe displays higher antibiotic potency and biocompatibility than that with Ac3c, which indicates not only that the bioactive conformation is better preserved by the former but also the significant role played by the phenyl rings in the recognition by living cells. Peptides homing tumor vasculature are considered promising molecular imaging agents for cancer detection at an early stage. In addition to their high binding affinity, improved tissue penetrating ability, and low immunogenicity, they can deliver targeted anticancer drugs, thus expanding therapeutic treatments. Among those, CREKA, a linear peptide that specifically binds to clotted-plasma proteins in tumor vessels, has been recently employed to design bioactive systems able to target different cancer types. Within this context, this paper explores the biorecognition event between CR( NMe)EKA, an engineered CREKA-analog bearing a noncoded amino acid ( N-methyl-Glu) that is responsible for its enhanced activity, and clotted-plasma proteins (fibrin and fibrinogen) by nanomechanical detection. Specifically, the tumor-homing peptide was covalently attached via epoxysilane chemistry onto silicon microcantilever chips that acted as sensors during dynamic mode experiments. Before that, each step of the functionalization process was followed by contact angle measurements, interferometry, X-ray photoelectron spectroscopy, and atomic force microscopy, thus revealing the applied protocol as a suitable strategy. The fibrin(ogen)-binding induced by CR( NMe)EKA was detected by the resonance frequency shift of the cantilevers, and a detection limit of 100 ng/mL was achieved for both proteins. Even though further development is required, this work reflects the promising application of emerging technologies capable of assisting in the comprehension of biological interactions and their implications in the biotechnological field. Conducting polymer nanocomposites (CPnC) have been prepared by loading different concentrations of poly(3-thiophene methyl acetate) (P3TMA) nanoparticles to an epoxy network. Therefore, the typical limitations of CPs applicability, i.e., reduced solubility and difficult processability, have been overcome. The influence of different factors (i.e., curing temperature, solvent used to disperse nanoparticles, and concentration of P3TMA) on the CPnC properties has been evaluated. The major requirement to obtain homogeneously distributed materials has been found to be the P3TMA-solvent compatibility. Xylene, which is typically used in epoxy resins, has been practically eliminated to avoid the formation of aggregates and has been replaced by tetrahydrofuran. Although properties like chemical structure and doping level as well as morphology and P3TMA nanoparticle distribution into the epoxy matrix have been examined for CPnC samples, particular attention has been paid to evaluate their mechanical and electrical properties. The Young's modulus and tensile strength values increase upon the addition of a small concentration of P3TMA, independently of the curing temperature, with the exception of 25 wt% P3TMA composition that exceeds the percolation limit for this material. The electrical DC conductivity determined for samples with 12 and 25 wt% P3TMA ranged from 6.0 ×·10−7 to 8.2·× 10−7 S/cm. Therefore, CPnC films exhibited a semiconductor behavior with conductivity values at least three to four orders of magnitude higher compared to that of the pure insulating epoxy (10−11 to 10−13 S/cm). In this manuscript we report a critical evaluation of the ability of natural DNA to mediate the nitroaldol (Henry) reaction at physiological temperature in pure water. Under these conditions, no background reaction took place (i.e., control experiment without DNA). Both heteroaromatic aldehydes (e.g., 2-pyridinecarboxaldehyde) and aromatic aldehydes bearing strong or moderate electron-withdrawing groups reacted satisfactorily with nitromethane obeying first order kinetics and affording the corresponding β-nitroalcohols in good yields within 24 h. In contrast, aliphatic aldehydes and aromatic aldehydes having electron-donating groups either did not react or were poorly converted. Moreover, we discovered that a number of metal-free organic buffers efficiently promote the Henry reaction when they were used as reaction media without adding external catalysts. Lithium-ion batteries are today among the most efficient devices for electrochemical energy storage. However, an improvement of their performance is required to address the challenges of modern grid management, portable technology, and electric mobility. One of the most important limitations to solve is the slow kinetics of redox reactions associated to inorganic cathodic materials, directly impacting on the charging time and the power characteristics of the cells. In sharp contrast, redox polymers such as poly(2,2,6,6-tetramethyl-1-piperidinyloxy methacrylate) (PTMA) exhibit fast redox reaction kinetics and pseudocapacitors characteristics. In this contribution, we have hybridized high energy Li(NixMnyCoz)O2 mixed oxides (NMC) with PTMA. In this hybrid cathode configuration, the higher voltage NMC (ca. 3.7 V vs. Li/Li+) is able to transfer its energy to the lower voltage PTMA (3.6 V vs. Li/Li+) improving the discharge power performances and allowing high power cathodes to be obtained. However, the NMC-PTMA hybrid cathodes show an important capacity fading. Our investigations indicate the presence of an interface degradation reaction between NMC and PTMA transforming NMC into an electrochemically dead material. Moreover, the aqueous process used here to prepare the cathode is also shown to enable the degradation of NMC. Indeed, once NMC is immersed in water, alkaline surface species dissolve, increasing the pH of the slurry, and corroding the aluminum current collector. Additionally, the NMC surface is altered due to delithiation which enables the interface degradation reaction to take place. This reaction by surface passivation of NMC particles did not succeed in preventing the interfacial degradation. Degradation was, however, notably decreased when Li(Ni0.8Mn0.1Co0.1)O2 NMC was used and even further when alumina-coated Li(Ni0.8Mn0.1Co0.1)O2 NMC was considered. For the latter at a 20C discharge rate, the hybrids presented higher power performances compared to the single constituents, clearly emphasizing the benefits of the hybrid cathode concept. The stereochemistry of polymers has a profound impact on their mechanical properties. While this has been observed in thermoplastics, studies on how stereochemistry affects the bulk properties of swollen networks, such as hydrogels, are limited. Typically, changing the stiffness of a hydrogel is achieved at the cost of changing another parameter, that in turn affects the physical properties of the material and ultimately influences the cellular response. Herein, we report that by manipulating the stereochemistry of a double bond, formed in situ during gelation, materials with diverse mechanical properties but comparable physical properties can be obtained. Click-hydrogels that possess a high % trans content are stiffer than their high % cis analogues by almost a factor of 3. A wide range of biorelevant applications, particularly in pharmaceutical formulations and the food and cosmetic industries, require the stabilization of two water-soluble blended components which would otherwise form incompatible biphasic mixtures. Such water-in-water emulsions can be achieved using Pickering stabilization, where two-dimensional (2D) nanomaterials are particularly effective due to their high surface area. However, control over the shape and size of the 2D nanomaterials is challenging, where it has not yet been possible to examine chemically identical nanostructures with the same thickness but different surface areas to probe the size-effect on emulsion stabilization ability. Hence, the rationale design and realization of the full potential of Pickering water-in-water emulsion stabilization have not yet been achieved. Herein, we report for the first time 2D poly(lactide) platelets with tunable sizes (with varying coronal chemistry) and of uniform shape using a crystallization-driven self-assembly methodology. We have used this series of nanostructures to explore the effect of 2D platelet size and chemistry on the stabilization of a water-in-water emulsion of a poly(ethylene glycol) (PEG)/dextran mixture. We have demonstrated that cationic, zwitterionic, and neutral large platelets (ca. 3.7 × 10 6 nm 2 ) all attain smaller droplet sizes and more stable emulsions than their respective smaller platelets (ca. 1.2 × 10 5 nm 2 ). This series of 2D platelets of controlled dimensions provides an excellent exemplar system for the investigation of the effect of just the surface area on the potential effectiveness in a particular application., Competing Interests: The authors declare no competing financial interest.

This manuscript describes a new route to prepare rapidly Ca 2+ -free hydrogels from unmodified sodium alginate by simply mixing with small organic molecules such as poly(carboxylic acid) compounds as cross-linker agents instead of classical divalent metal salts such as CaCl 2 . Dimethyl sulfoxide (DMSO) was also found to induce the rapid gelation of aqueous alginate solutions. The gelation process takes place at room temperature, and depending on the composition, gels with good thermal (90-100 °C) and mechanical properties compared to classical metal-containing analogs are obtained. DMSO-based gels showed remarkable self-supporting and thixotropic properties, which can be tuned by the biopolymer concentration. Furthermore, oxalic acid-based gels show superior elasticity than HCl, CaCl 2 and DMSO-based gels. The possibility to prepare monoliths, beads, and films of these gels provide them with significant versatility. In particular, films made of alginate and oxalic acid show good potential as synergistic anticancer drug delivery carrier. Computational studies using both quantum mechanical and classical force-field methodologies reveal that hydrogen bonding networks between water and DMSO molecules located close to the alginate chains are responsible for the stability of DMSO-based gels. In contrast, the cohesion of oxalic acid-based gels is a consequence of the coexistence of multiple ionic associations involving oxalate, alginate, and Na + counterions, which stabilize the system and keep all the interacting species grouped.

Three different tetraphenylalanine (FFFF) based peptides that differ at the N- and C-termini have been synthesized by using standard procedures to study their ability to form different nanoassemblies under a variety of conditions. The FFFF peptide assembles into nanotubes that show more structural imperfections at the surface than those formed by the diphenylalanine (FF) peptide under the same conditions. Periodic DFT calculations (M06L functional) were used to propose a model that consists of three FFFF molecules defining a ring through head-to-tail NH3(+)⋅⋅⋅(-)OOC interactions, which in turn stack to produce deformed channels with internal diameters between 12 and 16 Å. Depending on the experimental conditions used for the peptide incubation, N-fluorenylmethoxycarbonyl (Fmoc) protected FFFF self-assembles into a variety of polymorphs: ultra-thin nanoplates, fibrils, and star-like submicrometric aggregates. DFT calculations indicate that Fmoc-FFFF prefers a parallel rather than an antiparallel β-sheet assembly. Finally, coexisting multiple assemblies (up to three) were observed for Fmoc-FFFF-OBzl (OBzl = benzyl ester), which incorporates aromatic protecting groups at the two peptide terminals.
