Introduction and Aims. Accurate knowledge of individual hepatitis C virus (HCV) status is an important component ofcomprehensive health services for people who inject drugs (PWID). In this paper we compare the perceived HCV status ofPWID in a longitudinal cohort study with their actual status, as verified by HCV-RNA testing. Methods. Participants whoconsented to blood testing at first follow up (352/688) were included. Self-reported HCV status (positive/negative/don’t know),was compared with serology test results for participants. Results. In comparing self-report with HCV-RNA results, 274 of 352participants had valid serology and self-report results; of these, 220 (80%) accurately reported their HCV status. Discussionand Conclusions. The findings of this study suggest that large proportions of PWID know their true HCV infection status,but the discordant participants represent potential HCV infection risks. Despite the majority of participants displayingconcordance, this study reinforces the need for regular blood testing and the giving of accurate, practical and comprehensiveHCV result information. We provide the results from a spectral analysis of nuclear decay data displaying annually varying periodic fluctuations. The analyzed data were obtained from three distinct data sets: Si-32 and (CI)-C-36 decays reported by an experiment performed at the Brookhaven National Laboratory (BNL), Mn-56 decay reported by the Children's Nutrition Research Center (CNRC), but also performed at BNL, and Ra-226 decay reported by an experiment performed at the Physikalisch-Technische Bundesanstalt (PTB) in Germany. All three data sets exhibit the same primary frequency mode consisting of an annual period. Additional spectral comparisons of the data to local ambient temperature, atmospheric pressure, relative humidity, Earth-Sun distance, and their reciprocals were performed. No common phases were found between the factors investigated and those exhibited by the nuclear decay data. This suggests that either a combination of factors was responsible, or that, if it was a single factor, its effects on the decay rate experiments are not a direct synchronous modulation. We conclude that the annual periodicity in these data sets is a real effect, but that further study involving additional carefully controlled experiments will be needed to establish its origin. Two reliable small-plant bioassays were developed using tissue-cultured banana, resulting in consistent symptom expression and infection by Fusarium oxysporum f. sp. cubense (Foc). One bioassay was based on providing a constant watertable within a closed pot and the second used free-draining pots. Culture medium for spore generation influenced infectivity of Foc. Inoculation of potted banana by drenching potting mix with a conidial suspension, consisting mostly of microconidia, few macroconidia and no chlamydospores, generated from one-quarter-strength potato dextrose agar + streptomycin sulfate, resulted in inconsistent infection. When a conidial suspension that consisted of all three spore types, microconidia, macroconidia and chlamydospores, prepared from spores generated on carnation leaf agar was used, all plants became infected, indicating that the spore type present in conidial suspensions may contribute to inconsistency of infection. Inconsistency of infection was not due to loss of virulence of the pathogen in culture. Millet grain precolonised by Foc as a source of inoculum resulted in consistent infection between replicate plants. Sorghum was not a suitable grain for preparation of inoculum as it was observed to discolour roots and has the potential to stunt root growth, possibly due to the release of phytotoxins. For the modified closed-pot system, a pasteurised potting mix consisting of equal parts of bedding sand, perlite and vermiculite plus 1 g/L Triabon slow release fertiliser was suitable for plant growth and promoted capillary movement of water through the potting mix profile. A suitable potting mix for the free-draining pot system was also developed.

To investigate how diphtheria toxin (DT) undergoes pH-dependent membrane translocation in mammalian cells, we have isolated and characterized mutants of the toxin that are defective in acidic-pH-dependent killing of Escherichia coli. Cloned DT secreted to the periplasm of E. coli kills the bacteria under acidic conditions (near pH 5.0) by inserting into and permeabilizing the inner membrane (a mechanism independent of the toxin's ADP-ribosylation activity). Mutant forms of DT with reduced lethality for E. coli were selected by plating the bacteria under acidic conditions. CRM503, one of the full-length mutants selected by this protocol, also showed diminished cytotoxicity for mammalian cells. We traced the altered cytotoxicity of CRM503 to a Glu-349----Lys mutation (E349K), one of three point mutations, within the B fragment. The E349K mutation alone inhibited cytotoxicity and membrane translocation in mammalian cells and lethality for E. coli but did not affect enzymic activity or receptor binding. The recently determined crystallographic model of DT shows that Glu-349 resides within a short loop connecting two long hydrophobic alpha-helices of the translocation domain. Protonation of Glu-349 and two other nearby acidic residues, Asp-352 and Glu-362, may enable these helices to undergo membrane insertion and the intervening loop to be transferred to the opposite face of the bilayer. The E349K mutation introduces a positive charge at this site, which would be expected to inhibit membrane insertion and the insertion-dependent activities of DT.
