Abstract

Response of aquatic organisms to pollutants has been studied following different biomarkers in feral fish at different locations of the Moselle River (France). Both ethoxyresorufin O-deethylase (EROD) and acetylcholinesterase (AChE) activities were measured respectively in the liver and muscle of chub (Leuciscus cephalus) caught by electrofishing. Parallel to this, level of DNA damage (in terms of DNA single strand breaks) was quantified in chub erythrocytes applying the comet assay. Results show a strong difference between males and females in consideration of EROD activity exhibiting 10 fold-values in males compared to females, but not for DNA damage and AChE activity. Moreover response of the 3 biomarkers does not depend on size of fish (highly correlated to their age). EROD activity appears to be induced in males and females according to a growing gradient between upstream and downstream stations but level of DNA damage follows an inverse tendency (i.e. fish from upstream stations exhibit a higher DNA damage than those downstream). Chemical analyses of organic and metallic compounds both in fish muscle and sediment do not clearly explain the responses of studied biomarkers. This study points out the difficulty to assess effects of pollutants in the case of complex contamination and highlights the necessity to use a large array of biomarkers at different levels of biological organisation., Cette étude menée par des laboratoires du Cemagref et de l'ENTPE s'inscrit dans le cadre du programme "PNETOX", destiné à évaluer et comparer des biomarqueurs et des indicateurs biologiques sur des poissons et des invertébrés de la Moselle sur 4 stations réparties à l'amont de Nancy en juin 1998. Il ressort de ces mesures une induction de l'activité EROD chez les cyprinidés de plus en plus marquée de l'amont vers l'aval. L'activité AChE ne présente quant à elle pas d'effet station. Enfin, l'impact génotoxique semble plus marqué pour les trois stations les plus amont. Les résultats de ces biomarqueurs sont à confronter aux mesures de bioaccumulation toxique dans les poissons et aux mesures de contamination dans les sédiments.

The radiation consists in large part of geosynchrotron radiation. The intensity of this radiation depends, among other factors, on the energy of the primary particle and the angle of the shower axis with respect to the geomagnetic field. Aims: Since the radiation mechanism is based on particle acceleration, the atmospheric electric field can play an important role. Especially inside thunderclouds large electric fields can be present. In this paper we examine the contribution of an electric field to the emission mechanism theoretically and experimentally. Methods: Two mechanisms of amplification of radio emission are considered: the acceleration radiation of the shower particles and the radiation from the current that is produced by ionization electrons moving in the electric field. For both mechanisms analytical estimates are made of their effects on the radio pulse height. We selected lopes data recorded during thunderstorms, periods of heavy cloudiness and periods of cloudless weather. We tested whether the correlations with geomagnetic angle and primary energy vary with atmospheric conditions. Results: We find that during thunderstorms the radio emission can be strongly enhanced. The present data suggests that the observed amplification is caused by acceleration of the shower electrons and positrons. In the near future, extensions of lopes and the construction of lofar will help to identify the mechanism in more detail. No amplified pulses were found during periods of cloudless sky or heavy cloudiness, suggesting that the electric field effect for radio air shower measurements can be safely ignored during non-thunderstorm conditions.

This is important, since a possible neutrino detection has to fulfill two requirements. First: antennas should be able to detect very inclined cosmic ray air showers, and second: it should be possible to estimate the distance to the shower maximum, since neutrinos are most likely to travel far through the Earth's atmosphere without interaction and induce air showers close to the ground. Methods: LOPES (LOFAR PrototypE Station; LOFAR - LOw Frequency ARray), an array of dipole antennas, is used for the detection of inclined cosmic ray air showers. LOPES is co-located and triggered by the KASCADE (KArlsruhe Shower Core and Array DEtector) experiment, which also provides information on air shower properties such as electron and muon numbers on the ground, as well as the arrival direction. Radio emission of nearly vertical cosmic ray air showers has been detected by LOPES. Results: LOPES-10 (the first phase of LOPES, consisting of 10 antennas) detected a significant number of cosmic ray air showers with a zenith angle larger than 50°, and many of these have very high radio field strengths. The most inclined event that has been detected with LOPES-10 has a zenith angle of almost 80°. This is proof that the new technique is also applicable for cosmic ray air showers with high inclinations, which in the case that they are initiated close to the ground, can be a signature of neutrino events. Conclusions: .Our results indicate that arrays of simple radio antennas can be used for the detection of highly inclined air showers, which might be triggered by neutrinos. In addition, we found that the radio pulse height (normalized with the muon number) for highly inclined events increases with the geomagnetic angle, which confirms the geomagnetic origin of radio emission in cosmic ray air showers.

This so-called knee is believed to be an imprint of corresponding steepenings or even cut-offs in the energy spectra of single cosmic ray elements, thus implying a change of composition in the range between 1 PeV and 100 PeV. One of the sophisticated experiments aiming at detailed measurements in the knee region is the KASCADE experiment and its successor, KASCADE-Grande. In the following, existing data on the knee and their limitations are briefly discussed. Concluding, an update on the KASCADE composition analysis is presented.

This radio emission has advantages for the detection of high energy cosmic rays compared to secondary particle or fluorescence measurement methods. Radio antennas like the LOPES30 antenna system are suited to investigate this emission process by detecting the radio pulses. The characteristic observable parameters like electric field strength and pulse length require a calibration which was done with a reference radio source resulting in an amplification factor representing the system behavior in the environment of the KASCADE-Grande experiment. Knowing the amplification factor and the gain of the LOPES antennas LOPES30 is calibrated absolutely for systematic analyses of the radio emission.

Abstract

The present study examines the potential of the comet assay using the rainbow trout gonad cell line-2 (RTG-2) as an in vitro indicator test for genotoxicity assessment of aquatic contaminants and native surface waters. Initially, the comet assay protocol was adapted to the RTG-2 cell line. An exposure period of 2 h was found to be optimal, because DNA damage decreased when exposure was prolonged. Then, the sensitivity of the comet assay with RTG-2 cells toward six genotoxic reference substances was evaluated. The lowest-observed-effect concentration values for the directly acting genotoxins, 4-nitroquinoline-N-oxide and N-methyl-N-nitro-N-nitrosoguanidine, were in the low nanomolar range. The RTG-2 test system clearly was less sensitive for the indirectly acting genotoxins benzo[a]pyrene, nitrofurantoin, 2-acetylaminofluorene, and dimethylnitrosamine, despite the presence of xenobiotic metabolic capacities in RTG-2 cells. The two effect endpoints used, tail length (TL) and tail moment (TM), did not differ with respect to sensitivity, but the linearity of the concentration-response curve was better with TM than with TL. The overall reproducibility of the assay results was good. Finally, the applicability of the comet assay with RTG-2 cells for genotoxicity screening of native surface water samples was studied. The assay tolerated the use of nonsterile water samples and was able to detect genotoxic potentials in native water samples; that is, extraction and concentration of the samples were not needed. The results of the present study indicate the suitability of the comet assay with the fish cell line, RTG-2, as in vitro screen for detecting genotoxic potencies of xenobiotics and environmental samples.

Abstract

Screening methods to indicate the genotoxic potential of individual chemicals or environmental mixtures rely mainly on short-term bacterial tests. Differences in the genotoxic response of prokaryotic and eukaryotic cells necessitate the development of nonbacterial screening assays. A promising approach for this purpose could be the comet (single-cell gel electrophoresis) assay performed with fish cells in vitro. In the present study, we evaluated the comet assay with two different fish cell lines from rainbow trout (Oncorhyhnchus mykiss), the fibroblast-like RTG-2 cell line established from gonad tissue, and the epitheloid RTL-W1 cell line established from liver tissue. The cells were exposed in vitro during 2 hr to the genotoxins, 4-nitroquinoline-1-oxide (NQO), and benzo(a)pyrene (BaP), as well as to environmental samples. The LOEC values for NQO were similar in both cell lines, whereas for BaP, the RTL-W1 cells were found to be more sensitive than the RTG-2 cells. The slopes of the concentration–response curves of the two test compounds differed between the two cell lines, with RTG-2 cells showing a steeper slope for NQO, and RTL-W1 cells showing a steeper slope for BaP. When exposed to environmental samples from a remediation site, the RTL-W1 cell line, but not the RTG-2 cell line, indicated a genotoxic potential of the samples. The differences in the genotoxic response pattern of the two cell lines could be only partly explained in relation to metabolic enzymes, cytochrome P4501A, glutathione-S-transferase, and xenobiotic reductase. The findings of this study demonstrate that the comet assay with fish cell lines is suitable as in vitro screening assay in environmental genotoxicity testing, but the choice of test cell line may be critical
