Abstract

Traumatic head injury is the second leading cause of catastrophic injuries for sports and physical activities. Previous studies on American football players reveal that the incidence of sport-related concussion (SRC) is associated with the occurrence of subdural hematoma. It follows that SRC prevention is essential in order to improve the safety of organized sports. A study that investigated the SRC knowledge among schoolteachers and coaches in Japan reported that nearly half had insufficient knowledge of SRC. In addition, the SRC knowledge of students in teacher training courses at Japanese universities remains unclear. In light of this, we conducted a survey to discern the SRC knowledge of students in a teacher training course. Knowledge was assessed by the number of correct answers that tested knowledge of ten general SRC facts and eight questions regarding SRC symptoms. The average score for general SRC knowledge was 8.65 (median: 9) and for SRC symptoms, 5.62 (median: 6). More significantly, only 35% of respondents correctly answered a question about long-term complications from repeated SRC trauma; only 25% correctly answered the symptoms relating to drowsiness and anxiety. The participants’ characteristic showed little ability in explaining, general knowledge of SRC (R-squared = 0.023). This study suggests the immediacy of ensuring students in teacher training courses have sufficient knowledge of SRC, its symptoms and risks as essential knowledge for the next generation of teachers and coaches., 本研究は，科学研究費補助金（課題番号18J15017）の助成を受けて実施した研究成果の一部です。

66. Visualization of human induced pluripotent stem cells-derived three-dimensional cartilage tissue by gelatin nanospheres

Abstract

Recently, many studies on the three-dimensional (3D) fabrication of cells have been performed. Under these circumstances, it is indispensable to develop the imaging technologies and methodologies for noninvasive visualization of 3D cells fabricated. The objective of this study is to develop the labeling method of human induced pluripotent stem (iPS) cells-derived 3D cartilage tissue with gelatin nanospheres coincorporating three kinds of quantum dots (QD) and iron oxide nanoparticles (IONP) (GNSQD+IONP). In this study, two labeling methods were performed. One is that a cartilage tissue was labeled directly by incubating with octaarginine (R8)-treated GNSQD+IONP (direct labeling method). The other one is a "dissociation and labeling method." First, the cartilage tissue was dissociated to cells in a single dispersed state. Then, the cells were incubated with R8-GNSQD+IONP in a monolayer culture. Finally, the cells labeled were fabricated to 3D pellets or cell sheets. By the direct labeling method, only cells residing in the surrounding site of cartilage tissue were labeled. On the contrary, the 3D cartilage pellets and the cell sheets were homogenously labeled and maintained fluorescently visualized over 4 weeks. In addition, the cartilage properties were histologically detected even after the process of dissociation and labeling. Homogenous labeling and visualization of human iPS cells-derived 3D cartilage tissue was achieved by the dissociation and labeling method with GNSQD+IONP. Impact statement The homogenous labeling and visualization of human iPS cells-derived three-dimensional (3D) cartilage tissue was achieved over 4 weeks by the dissociation and labeling method with gelatin nanospheres coincorporating quantum dots (QD) and iron oxide nanoparticles (IONP) (GNSQD+IONP). The cartilage properties of cells treated were maintained. It is concluded that the dissociation and labeling method with GNSQD+IONP is a promising to visualize the human iPS cells-derived 3D cartilage tissue.

Abstract

Acute myocardial infarction is a leading cause of death among single organ diseases. Despite successful reperfusion therapy, ischaemia reperfusion injury (IRI) can induce oxidative stress (OS), cardiomyocyte apoptosis, autophagy and release of inflammatory cytokines, resulting in increased infarct size. In IRI, mitochondrial dysfunction is a key factor, which involves the production of reactive oxygen species, activation of inflammatory signalling cascades or innate immune responses, and apoptosis. Therefore, intercellular mitochondrial transfer could be considered as a promising treatment strategy for ischaemic heart disease. However, low transfer efficiency is a challenge in clinical settings. We previously reported uptake of isolated exogenous mitochondria into cultured cells through co-incubation, mediated by macropinocytosis. Here, we report the use of transactivator of transcription dextran complexes (TAT-dextran) to enhance cellular uptake of exogenous mitochondria and improve the protective effect of mitochondrial replenishment in neonatal rat cardiomyocytes (NRCMs) against OS. TAT-dextran-modified mitochondria (TAT-Mito) showed a significantly higher level of cellular uptake. Mitochondrial transfer into NRCMs resulted in anti-apoptotic capability and prevented the suppression of oxidative phosphorylation in mitochondria after OS. Furthermore, TAT-Mito significantly reduced the apoptotic rates of cardiomyocytes after OS, compared to simple mitochondrial transfer. These results indicate the potential of mitochondrial replenishment therapy in OS-induced myocardial IRI.

Abstract

Transanal total mesorectal excision (taTME) has been rapidly accepted as a promising surgical approach to the distal rectum. The benefits include ease of access to the bottom of the deep pelvis linearly over a short distance in order to easily visualize the important anatomy. Furthermore, the distal resection margins can be secured under direct vision. Additionally, a two‐team approach combining taTME with a transabdominal approach could decrease the operative time and conversion rate. Although taTME was expected to become more rapidly popularized worldwide, enthusiasm for it has stalled due to unfamiliar intraoperative complications, a lack of oncologic evidence from randomized trials, and the widespread use of robotic surgery. While international registries have reported favorable short‐ and medium‐term outcomes from taTME, a Norwegian national study reported a high local recurrence rate of 9.5%. The characteristics of the recurrences included rapid, multifocal growth in the pelvis, which was quite different from recurrences following traditional transabdominal TME; thus, the Norwegian Colorectal Cancer Group reached a consensus for a temporary moratorium on the performance of taTME. To ensure acceptable baseline quality and patient safety, taTME should be performed by well‐trained colorectal surgeons. Although the appropriate indications for taTME remain controversial, the transanal approach is extremely important as a means of goal setting in difficult TME cases and as an aid to the transabdominal approach in various types of extended pelvic surgeries. This article discusses the usefulness of fluorescent ureteral navigation in total pelvic exenteration for locally recurrent pelvic malignancies. The authors highlight the challenges of redo surgery for these malignancies due to fibrosis caused by previous surgeries or radiotherapy. They introduce a novel technique using fluorescent ureteral navigation with a two-team approach in laparoscopic pelvic exenteration for locally recurrent endometrial cancer. The authors believe that this technique is beneficial for detecting the ureter intraoperatively and can improve outcomes for patients with locally recurrent pelvic malignancies.
