Mediator is a large and evolutionarily conserved coactivator complex essential for RNA polymerase II (Pol II)-mediated gene regulation at multiple steps of the transcription process, including preinitiation complex (PIC) assembly and function. Here, we used the MultiBac baculovirus expression system to generate recombinant human core Mediator subcomplexes and subsequent biochemical approaches to dissect the mechanism by which Mediator facilitates direct recruitment of Pol II to core promoters. Our results highlight a pivotal role in this process for the N-terminal half (NTD) of the MED14 subunit. We show that a reconstituted 15-subunit human core Mediator complex that contains only the MED14-NTD is fully functional in facilitating both basal and activated (p53) transcription. This complex directly interacts with the C-terminal domain (CTD) of the RPB1 subunit of Pol II (RPB1 CTD) and is required for recruiting Pol II to core promoters. Moreover, recombinant RPB1 can completely reverse the human core Mediator-Pol II interaction. Notably, the human MED14-NTD region has secondary structure conservation with Schizosaccharomyces pombe. In addition, reanalysis of published cryo-EM structures of yeast Mediator-Pol II complexes strongly supports our conclusion. Thus, our analyses provide critical new insights into how Mediator binds to Pol II and recruits it to the promoters to facilitate transcription.

Bu doğrultuda araştırmanın amacı HolidayCheck.de web sitesinde Alanya’da faaliyet gösteren dört ve beş yıldızlı konaklama işletmeleri için 2009-2017 yılları arasında farklı kriterler için verilen puanların, işletmelerin tavsiye edilmesi üzerindeki etki düzeyinin ve en etkili değişkenin belirlenmesidir. HolidayCheck.de web sitesinde genel durum, lokasyon, çalışanlar, yiyecek-içecek hizmetleri, spor-havuz-eğlence hizmetleri ve odalar olarak belirlenen altı kritere verilen puanlamalara İkili Lojistik Regresyon Analizi uygulanmıştır. Elde edilen bulgulara göre konaklama işletmelerinin tavsiye edilmesi üzerinden en yüksek etkiye sahip kriter yiyecek-içecek hizmetleri ve en düşük etkiye sahip kriter odalar olarak belirlenmiştir. İkinci derecede etkili kriter ise aile dostu, engelli dostu gibi özelliklerin puanlandığı genel durum olarak tespit edilmiştir.

Introduction Incorrect positioning is one of the main factors for glenoid component loosening in reverse shoulder arthroplasty and component placement can be challenging. This study aimed to assess whether Patient-Specific Instrumentation (PSI) provides better guide pin positioning accuracy and is superior to standard guided and freehand instrumentation methods in cases of glenoid bone deformity. Materials and Methods Based on the Walch classification, five different scapula types were acquired by computed tomography (CT). For each type, two different surgeons placed a guide pin into the scapula using three different methods: freehand method, conventional non-patient-specific guide, and PSI guide. Each method was repeated five times by both surgeons. In these experiments, a total of 150 samples of scapula models were used (5 × 2 × 3 × 5 = 150). Post-operative CT scans of the samples with the guide pin were digitally assessed and the accuracy of the pin placement was determined by comparison to the preoperative planning on a three-dimensional (3D) model. Results The PSI method showed accuracies to the preoperative plan of 2.68 (SD 2.10) degrees for version angle ( p < .05), 2.59 (SD 2.68) degrees for inclination angle ( p < .05), and 1.55 (SD 1.26) mm for entry point offset ( p < .05). The mean and standard deviation errors compared to planned values of version angle, inclination angle, and entry point offset were statistically significant for the PSI method for the type C defected glenoid and non-arthritic glenoid. Conclusion Using the PSI guide created by an image processing software tool for guide pin positioning showed advantages in glenoid component positioning over other methods, for defected and intact glenoid types, but correlation with clinical outcomes should be examined.

It is said in Bilge Kaghan inscription, one of the Orkhon inscriptions, that “Turkish people give up this habit and regret it! People of the holy Otuken Mountain, you left your fatherland”. This expression used by Bilge Kaghan to remark the importance of staying in Otuken Forest and not leaving there, in a sense, can be as a principal motivator and slogan of the travels for the Turks worldwide in order to visit their homelands to learn about the roots and heritage. Nowadays, the urge for searching the ancestor and the roots, finding and reinforcing the identity and experiencing the old times is an important tourism field. The primary aim of this study is to conceptualize the new notions used in the international literature that arise from new tourism activities such as nostalgia and root tourism in the Turkish language. Besides, contributing to the planning process of the supply and demand conditions that might be developed between Turkey and Central Asia based on the common cultural heritage is another important aim of the study. The research, based on the literature review and secondary data, reveal that root tourism is a significant opportunity for the Turks and Turkey and Central Asia., Orhon yazıtlarından biri olan Bilge Kağan yazıtında “Türk halkı (bu huyundan) vazgeç ve nadim ol! Kutsal Ötüken Dağları Halkı, (sen kendin yerini yurdunu bırakıp) gittin” ifadesi yer almaktadır. Bilge Kağan tarafından Ötüken ormanında kal, oradan ayrılma anlamında kullanılan bu ifade bir anlamda bütün dünyadaki ve özellikle Türkiye’deki Türklerin ata yurtlarını ziyaret etme ve kökleri ile ilgili mirası görme amacıyla yapacakları seyahatlerin temel motivasyonu ve sloganı olabilir. Günümüzde bireylerin, atalarını ve köklerini araştırma, kimliğini bulma, güçlendirme ve geçmiş zamanı yaşama isteği önemli bir turizm alanı haline gelmiştir. Bu çalışmanın öncelikli amacını miras ve nostalji odaklı ortaya çıkan bu yeni turizm faaliyetleriyle ilgili uluslararası alanyazında kullanılan kavramların Türkçe karşılıklarının geliştirilmesidir. Ayrıca, bu kapsamda Türkiye ile Orta Asya arasında ortak kültürel mirasa dayalı olarak geliştirilebilecek turizm arz ve talep koşullarının planlanmasına katkı sunulması çalışmanın bir diğer amacını oluşturmaktadır. Alanyazına ve ikincil verilere dayalı olarak yapılan bu çalışma, miras odaklı gelişen turizm türlerinin Türkler ve Türkiye ve Orta Asya Ülkeleri için önemli bir fırsat olduğunu ortaya koymaktadır.

Proliferating trichilemmal tumor (PTT) is a rare, mostly benign neoplasm which stems from the follicular outer root sheath epithelium. PTT occurs as a subcutaneous cystic nodule slowly enlarging to a larger noduler mass, and is usually localized on the scalp of elderly women. To the best of our knowledge, PTT localized on a psoriatic plaque has not been reported previously. Herein, we report an unusual case of benign PTT arising from a psoriatic plaque on the knee of a 63-year-old male patient. Keywords: proliferating trichilemmal tumor, psoriasis, trichilemmal cyst

Bu çalışmada, Kars yöresinde halk elinde yetiştirilen kaz işletmelerinden kaz kesimi esnasında alınan kan serum örneklerinde Salmonella Enteritidis antikorlarının Enzyme-Linked Immunosorbent Assay (ELISA) yönetimi ile araştırılması amaçlanmıştır. Bu amaçla, Kars il merkezinde 8 ve Arpalı köyünde 2 kaz işletmesinden olmak üzere toplam 10 farklı işletmeden ve önceden S. Enteritidis ve diğer Salmonella enfeksiyonlarına karşı herhangi bir aşı geçmişi olmayan 1 yaşındaki 315 adet erkek ve dişi yerli kazdan örnek alındı. İncelenen 315 kaz kan serum örneğinin 16 (%5.07)’sı S. Enteritidis antikorları yönünden pozitif, 48 (%15.23)’i şüpheli ve 251 (%79.68)’i negatif saptandı. En yüksek S. Enteritidis antikor pozitifliği Kars il merkezindeki işletme 7 ile Arpalı köyündeki işletme 10’da (sırasıyla %16.66 ve %12.5) saptandı ve işletmeler arası bu pozitiflik istatistiksel olarak anlamlı (P0.05). Kars yöresinde kazlarda S. Enteritidis’in varlığı ve yaygınlığı üzerine yapılan bu çalışmada, kaz kan serumu örneklerinden %5.07 oranında antikor pozitiflik saptanmasının önemli olduğu; bununla birlikte vertikal bulaşma özelliğine sahip olan ve karkas kontaminasyonunun yaygın olduğu Salmonella enfeksiyonlarında benzer tarama testleri yapılmasının hastalığın prevalansının azaltılmasına fayda sağlayacağı düşünülmektedir., In this study, it was aimed to investigate Salmonella Enteritidis antibodies by Enzyme-Linked Immunosorbent Assay (ELISA) in blood serum samples obtained from the goose enterprises grown in Kars Region during the goose slaughtering period. For this purpose, a total of 10 different flocks, 8 from Kars Province center and 2 from Arpalı Village, were included in the study. Totally, one year old 315 female and male domestic geese which had no vaccination history against S. Enteritidis and other Salmonella infections were sampled. Of the 315 goose blood serum samples examined, 16 (5.07%) were positive, 48 (15.23%) were suspicious and 251 (79.68%) were negative for S. Enteritidis antibodies. The highest S. Enteritidis antibody positivity was detected in the Flock 7 in Kars city center and in the Flock 10 in Arpalı Village with a rate as 16.66% and 12.5%, respectively. This positivity between the flocks was statistically significant (P0.05). In this study conducted on the presence and prevalence of S. enteritidis in geese in Kars province, it was found that 5.07% antibody positivity was considered to be important in geese blood serum samples; in addition to this, similar screening tests for Salmonella infections, which have vertical transmission characteristic and carcass contamination are common, are thought to be beneficial in decreasing the prevalence of the disease.

RNA polymerase II (Pol II) is a 12 subunit protein complex from yeast to human that is required for gene expression. Gdownl containing Pol II [Pol II (G)] is a special form of Pol II that is catalytically inactive and heavily depends on the 30-subunit Mediator complex for its activator and basal dependent function in vitro. Here we report for the first time, the identification and the generation of a 15-subunit human Mediator complex via the novel multibac baculovirus expression system that is fully responsive to Pol II (G). Our results show complete recovery of Pol II (G) dependent transcription both with full 30-subunit Mediator and also with 15-subunit recombinant Mediator that we synthesized. Moreover, we also show that the recombinant Mediator interacts with Pol II (G) as well. These results enlighten us towards understanding how a certain population of Pol II that is involved in selected gene regulation is activated by Mediator complex.

Brucellosis is one of the infectious diseases that may increase osteoporosis risk. Bone formation and destruction markers analyzed in the examination of osteoporosis risk. There been many studies on the effect of a variety of diseases on the bone, but no studies have conducted for brucellosis. This study is one of the rare studies showing the effect of brucellosis on a bone. The purpose of this study is that whether age, agglutination level, having brucellosis at the time or before, and gender knowledge of patients have effects on osteoporosis by using bone formation and destruction markers. Between 01/04/2015 and 31/12/2017, blood samples were taken from 40 patients with brucellosis and seven patients whose treatment completed at least six months before. Then biochemical markers were studied on these blood samples. ELISA washer and reader (Biotek, Novatek, Istanbul, Turkey) was used to obtain the values of bone formation and destruction markers. There was a significant difference, according to H. Osteocalcin (Human Osteocalcin/Bone Gla Protein), in terms of bone formation markers and was higher in women. Having brucellosis at the time was significant according to Human deoxypyridinoline (DPD) and Human C-telopeptide of type I collagen (CTX-I) in terms of bone destruction markers and was higher in brucellosis. The bone formation and destruction markers strongly correlated with each other in the same direction. It is thought that brucellosis can increase bone destruction markers, especially DPD (Human deoxypyridinoline) and CTX-I; therefore, osteoporosis risk in brucella patients can reduce by implementing a treatment plan that closely monitors bone destruction markers.

Pasini ve Pierini’nin İdiyopatik Atrofoderması (PPİA) nadir görülen etyolojisi bilinmeyen bir dermal atrofi tipidir. Lezyonlar tek ya da birden fazla oval ya da yuvarlak, simetrik , deriden hafif çökük, keskin sınırlı plaklar şeklindedir. Çoğunlukla sırtta lokalize olan lezyonlar sonrasında göğüs ve abdominal bölgede de ortaya çıkabilmektedir. Lezyonların boyutu 1-20 cm arasında değişebilmektedir ve lezyonlar zaman içerisinde yavaşça genişler. Hastaların çoğu kadın olup, genelde ikinci-üçüncü dekatlarda görülmektedir. Nadiren PPİA ve morfea bir arada görülebilir. Bu makalede sırtında asemptomatik çökük kahverengi leke ile polikliniğimize başvuran, klinik ve histopatolojik bulgularla PPİA ve morfea tanısı konulan 27 yaşında bayan hasta sunulmaktadır.

Aim: Prostatic adenocarcinoma is the most common cancer among men in the world and prostatectomy specimens are one of the most commonly encountered materials in pathology laboratories. Histopathological evaluation of radical prostatectomy specimens provides very important prognostic parameters to predict patient’s prognosis and to choose an appropriate treatment. There is no globally accepted standard grossing method for radical prostatectomy materials. Different grossing protocols are preferred in different centers considering financial condition, storage spaces, number of technicians and pathological workload. Methods: In this study, we evaluated 50 radical prostatectomy specimens using total and partial sampling methods and compared the results. Results: As a result of the partial sampling method the number of blocks per case was reduced prominently, and depending on this workload and financial burden also reduced. The correlation between total and partial sampling methods was statistically significant. Conclusion: Partial sampling method can be a choice of grossing of radical prostatectomy specimens with the help of macroscopic, clinical and radiological findings.

Introduction: Lichen planus (LP) is a chronic inflammatory disease, where the psychogenic factors seem to play an important role in the pathogenesis. Aim: To determine the expression of corticotropin-releasing hormone (CRH) receptor type 1 (CRH-R1) in LP. Material and methods: Thirty-two LP patients and 17 age/gender-matched controls were included in the study. Detailed information about the disease and body surface area (BSA) covered by the lesions was recorded. Immunohistochemically, the expression of CRH-R1 was stained in the lesional skin of patients with LP and in the control group. Results: The comparison of CRH-R1 expression showed a statistically significant difference between LP patients and the controls (p < 0.05). Additionally, we did not observe any correlation between BSA and staining intensity in LP patients. Conclusions: Our study showed an increase in CRH-R1 expression in LP lesions. These results support the participation of the cutaneous CRH/CRH-R1 system in the pathogenesis of LP skin lesions.
