Abstract

Milicia excelsa (Welw.) C.C. Berg is an important species for timber production and refor- estation purposes in west Africa. Polyphenols, as secondary compounds, are found in both conifers and broadleaves and the concentrations of these substances vary among populations, families and clones. Phenolics are involved in the defence chemistry of the plants and these compounds can affect the sucepti- bility of the tree to the attack of insects and fungi. The purpose of this preliminary phytochemical investigation on fresh leaves, was to determine the geographic variability of different populations of M. excelsa. Leaves were collected from seedlings that represent three characteristic natural areas in Ghana, Sierra Leone and Cote d'Ivoire. A preliminary screening was carried out by High Per- formance Thin-Layer Chromatography (HP-TLC) analysis. This methodology allowed to easily compare the polyphenolic content of total raw extract of the leaves. The chemical structure of the main compounds was detected by means of High Performance Liquid Chromatography/Diode Array Detector (HPLC/DAD) and High Performance Liquid Chromatography/Mass Spectrometry (HPLC/MS). Our preliminary results show that the phenolic composition varied between the provenances and that phenolic markers could be an useful tool for selection of ecophysiologically adapted indigenous genotypes for sustainable timber production.

79. An in vitro evaluation of the antibacterial activity of the non-volatile phenolic fraction from rosemary leaves.

Abstract

In this study, the aim was to evaluate the antimicrobial action of the non-volatile phenols of rosemary leaves against two Gram-negative bacteria (Escherichia coli and Pseudomonas aeruginosa) and two Gram-positive bacteria (Staphylococcus aureus and Staphylococcus epidermidis). Three extracts with different phenolic compositions were tested. By the agar disc diffusion method, Gram-positive bacteria were more sensitive to the extracts, and S. epidermidis showed the highest inhibition zones. Overall, all the extracts tested by the broth dilution method showed higher activity than results from the agar disc diffusion method. The minimal bactericidal concentration values indicated that E. coli was the most susceptible strain. This study demonstrated that the flavonoidic fraction of rosemary leaves does not play a crucial role as antimicrobial agents against these microorganisms. Abstract

Pharmacological interest in drug discovery has increased in recent years. Accordingly, an increasing part of the scientific body has thus far studied several thousands of compounds of natural and synthetic origin to ascertain their biological activity. The occurrence in plant tissues of secondary metabolites characterized by a O-heterocyclic structure and exhibiting antimicrobial properties is a well-known phenomenon. Among them, catechins and proanthocyanidins are two classes of compounds that exhibit antiviral or antimicrobial properties towards prokaryotic and eukaryotic microorganisms. Yet, despite the profusion of studies published so far, the real potentialities and limitations presented by the use of the above-mentioned class of molecules as antiviral or antimicrobial agents have not been critically evaluated. The present chapter represents an overview of the recent literature regarding the antiviral and antimicrobial properties exhibited by this class of compounds. Their mode of action as well as their synergy with currently used antibiotic molecules are also reviewed. 87. Antitumoural activity of viniferin-enriched extracts from Vitis vinifera L. cell cultures.

Abstract

The aim of this work was to evaluate the effect of stilbenes from different cultivars ofVitis viniferaon tumour proliferation. Extracts were obtained from elicitedV. viniferacell cultures and characterised by HPLC/DAD/MS. Cell growth was evaluated in four human cancer cell lines and in normal human fibroblasts. The cells were exposed to the extracts or totrans-resveratrol, used as reference molecule, for 48 h, at 1–10 μM concentrations of total stilbenoids. All the extracts exhibited antiproliferative activity, mediated by modulation of the cell cycle and induction of cytotoxicity in cancer but not in normal cell lines, and positively correlated with the content in dimeric stilbenoids. The Alphonse Lavallée extract was the most active, and the obtained stilbenoid fraction resulted 8–10 times more active thantrans-resveratrol. Extracts fromV. viniferacell cultures could represent new sources of active stilbenoid compounds to be further assayed inin vivostudies for their antitumoural properties. [ABSTRACT FROM PUBLISHER]
