The effects induced by Apis mellifera venom (AMV), melittin-free AMV, fraction with molecular mass < 10 kDa (F<₁₀) or melittin in nociceptive and inflammatory pain models in mice were investigated. Subcutaneous administration of AMV (2, 4 or 6 mg/kg) or melittin-free AMV (1, 2 or 4 mg/kg) into the dorsum of mice inhibited both phases of formaldehyde-induced nociception. However, F<₁₀ (2, 4 or 6 mg/kg) or melittin (2 or 3 mg/kg) inhibited only the second phase. AMV (4 or 6 mg/kg), but not F<₁₀, melittin-free AMV or melittin, induced antinociception in the hot-plate model. Paw injection of AMV (0.05 or 0.10 mg), F<₁₀ (0.05 or 0.1 mg) or melittin (0.025 or 0.050 mg) induced a nociceptive response. In spite of inducing nociception after paw injection, scorpion (Tityus serrulatus) or snake (Bothrops jararaca) venom injected into the dorsum of mice did not inhibit formaldehyde-induced nociception. In addition, AMV (6 mg/kg), but not F<₁₀ (6 mg/kg) or melittin (3 mg/kg), inhibited formaldehyde paw oedema. Concluding, AMV, F<₁₀ and melittin induce two contrasting effects: nociception and antinociception. AMV antinociception involves the action of different components and does not result from non-specific activation of endogenous antinociceptive mechanisms activated by exposure to noxious stimuli., (Copyright © 2011 Elsevier Ltd. The aim of this study was to investigate the effect of daily coffee ingestion on hepatocarcinogenesis in rats submitted to the resistant hepatocyte (RH) model. During lactation, the dams were fed a control or a coffee-supplemented diet. After weaning, male pups followed the same dietary protocol and were submitted to the RH model. The animals were sacrificed at 110 days of life. Removal of the medial and left lateral lobes was used as mitogenic stimulus, and the liver regeneration was estimated. Morphometric analyses of preneoplastic lesions were carried out on liver histological sections submitted to the histochemical procedure of the glucose-6-phosphatase activity. The gamma-glutamyltranspeptidase (GGT) activity was analyzed in the homogenate of regenerated livers. Body weight, mass liver regeneration, and hepatic cell architecture were not affected by coffee ingestion. In the group of animals fed the coffee-supplemented diet, the number of persistent and remodeling nodules was reduced (85.5% and 70.5%, respectively). The hepatic area occupied by the persistent nodules was also reduced (92%). There was a reduction of 7.7% in the GGT activity in the group fed the coffee-supplemented diet, although not statistically significant. The results indicate that coffee modulates chemical hepatocarcinogenesis in rats.

Scorpion envenomation is a public health problem in Brazil, with most severe cases occuring in children under the age of 5 years (0.6% lethality). In fact, the toxic fractions of the Tityus serrulatus scorpion venom (TSSV) have greater permeability across the BBB of weanling rats when compared to adults. Although EEG alterations have been reported in up to 75% of pediatric severe cases, the role of the CNS in envenomation morbidity is still in debate. Our working hypothesis is that the neural substrates that play a major role in morbidity generate activity undetectable from EEG scalp leads. Twenty one-day-old rats (n=18) were injected s.c. with the deadliest toxic fraction of the TSSV, tityustoxin (TsTX; 2xDL50=6 mg/kg). EEG leads were stereotaxicaly implanted in the nucleus of the solitary tract (NTS) and left parietal cortex. EEG and ECG were continuously monitored by a video EEG system until death or for a maximum period of 240 min. An experimental group pre-treated with carbamazepine (CBZ) was added in order to better access the cause-effect relationship between neural discharges and the systemic ECG alterations. High amplitude discharges in the NTS, which correlated to cardiac alterations, were recorded soon after administration of TsTX. Abnormal electrographic activity spread throughout the cortex only later in the recording. As expected, the CBZ treatment increased the latency for the first epileptiform discharge, decreased EEG/ECG alterations and increased the general survival time. In summary: peripheral scorpion toxin inoculation recruits brainstem involved in cardiovascular control and initial electrographic activity was undetectable from the cortical electrode.

The scorpion-envenoming syndrome has an incidence of approximately 8000 accidents/year in Brazil; with most severe cases occurring during childhood and elderly. Previous results from our laboratory suggest that the effects of scorpion toxins on the central nervous system play a major role on the lethality induced by scorpion envenoming. Our group has shown that the pre-treatment with carbamazepine (CBZ) is able to increase the latency-to-death in developing animals inoculated with tityustoxin, a toxic fraction of the Tityus serrulatus crude venom. Nevertheless, in order to perceive CBZ as potentially useful in clinical practice, the efficiency of CBZ against crude venom inoculation and the pharmacological treatment introduced after envenomation must be addressed. Thus, the objective of this work was to evaluate CBZ therapeutic efficiency against scorpion envenomation in developing rats. Animals were treated with i.p. injections of either vehicle or CBZ (50 mg/kg or 100 mg/kg) 10 min after injected with a s.c. fixed volume of either saline or crude T. serrulatus venom extract (48 mg/kg). The dose chosen for venom inoculation was 16 times its DL50 for 21-day-old Wistar rats, invariably inducing death within 2 h. Although CBZ did not significantly reduce the pulmonary edema, it was effective in increasing survival rate by approximately 75% in treated rats. In conclusion, CBZ was effective in the treatment of T. serrulatus envenomation even though not blocking the pulmonary edema.

The scorpion envenoming syndrome is a serious public health matter in Brazil. The most severe cases occur during childhood and elderly. Previous results from our laboratory suggest that the effects of scorpion toxins on the central nervous system play a major role on the lethality induced by scorpion envenoming. The aim of this work is to evaluate the therapeutic potential of carbamazepine (CBZ) injected i.p. 90 min before s.c. tityustoxin (TsTX) injection in weanling rats. Rats were divided into six experimental groups according to s.c. injection (saline or TsTX) and i.p. treatment (vehicle or CBZ 12, 50 and 100 mg/kg): Sal/Veh group (n=4); Sal/CBZ100 (n=4); TsTX/CBZ12 (n=6); TsTX/CBZ50 (n=8); TsTX/CBZ100 (n=8) and, at last, TsTX/Veh (n=8). The dose of TsTX was the same for all groups: 6.0mg/kg, twice the DL50 for weanling rats. Video images were recorded until death or for a maximum period of 240 min. Lungs were excised and weighed to evaluate edema. The results showed that CBZ (12, 50 and 100mg/kg) was able to increase the survival rate and latency-to-death of the rats. Only the group treated with 100mg/kg of CBZ had a decrease in the pulmonary edema. The known effect of CBZ reducing neuronal excitability most likely protected the neural substrates targeted by TsTX. Although treatment was performed before TsTX inoculation, the results are promising regarding CBZ as a therapeutic coadjuvant in the treatment of scorpion poisoning. The pharmacokinetics of CBZ can be very much improved by either changing the form of administration or encapsulating the drug in order to enhance solubility.

The scorpion envenoming syndrome is a serious public health matter in Brazil. The objective of this work is to evaluate carbamazepine (CBZ) as a potential therapeutic agent against tityustoxin (TsTX) envenomation. The choice of i.c.v. toxin injection assures that TsTX is readily available in the parenchyma, configuring a worst case scenario for protecting the CNS afflicted by envenomation. Adult Wistar rats were submitted to surgery for guide cannulae (lateral ventricle) and electrodes (EEG-parietal cortices and ECG-thoracic leads) implantation. Animals (n=25) were treated with i.p. injections of either vehicle or CBZ 90 min before injected i.c.v. with a fixed volume of either saline or TsTX: vehicle treated/TsTX injected (1.74 microg, i.c.v.; n=4), CBZ treated (50mg/kg, i.p.)/TsTX injected (n=12); CBZ treated/saline injected (n=5); and vehicle treated/saline injected (n=4). Video EEG/ECG was recorded until death or for a maximum period of 90 min. Lungs were excised and weighed to evaluate edema. The results showed that 10 out of 12 CBZ treated rats survived to TsTX i.c.v. microinjection. CBZ significantly decreased cardiac arrhythmias and pulmonary edema in rats injected with TsTX. Furthermore, CBZ also significantly increased the latency for the first cortical epileptiform discharge. The known effect of CBZ reducing neuronal excitability most likely protected the neural substrates targeted by TsTX. CBZ was efficient in attenuating envenoming symptoms after the i.c.v. inoculation of the TsTX in rats. Thus, CBZ can be proposed as a therapeutic coadjuvant in the treatment of scorpion poisoning.

Scorpion venom (TX) promotes gastric acid and pepsin secretion leading to acute gastric mucosal lesions (AGML), when injected in animals. The goal of the present study was to observe the effects of acid gastric secretion blockers over the incidence of TX-induced AGML in vivo. To verify this model, we used male albino rats, fasted 18-20 h (n=122) and anaesthetized with urethane (1.4 g/kg, i.p.). Their trachea and left femoral vein were both cannulated; the first to avoid airway obstructions during scorpion intoxication and the second for administration of saline, TX and acid blockers. Following the surgical procedure, the animals were divided in 10 groups of at least 10 animals each. Control groups were injected with NaCl 0.9% 1 ml/kg (n=10) or TX 375 microg/kg (n=32). Test groups (n=10, each) received atropine 5 mg/kg, cimetidine 10mg/kg, ranitidine 2.5mg/kg, ranitidine 5mg/kg, omeprazol 1 mg/kg, omeprazol 4 mg/kg, octreotide 80 and octreotide 100 microg/kg 10 min before the TX was injected. After 1h of intoxication, the stomach was resected for macroscopic study and the gastric secretion was collected for volume, pH and acid output assessment. We observed that all blockers were able to completely or partially prevent the TX-induced acid secretion as well as the AGML (p<0.05). Our data suggest the TX-induced AGML can be prevented by different class of acid blockers injected before the intoxication.

In this study we characterized the nociceptive response and edema induced by the venom of the scorpion Tityus serrulatus in rats and mice and carried out a preliminary pharmacological investigation of the mechanisms involved in these responses. Intraplantar injection of the venom (1 or 10mug) induced edema and a marked ipsilateral nociceptive response, characterized by thermal and mechanical allodynia and paw licking behaviour. The nociceptive response was inhibited by previous intraperitoneal administration of indomethacin (4mg/kg), dipyrone (200mg/kg), cyproheptadine (10mg/kg) or morphine (5 or 10mg/kg), but not by dexamethasone (1 or 4mg/kg) or promethazine (1 or 5mg/kg). The edema was inhibited by previous treatment with promethazine (5 or 10mg/kg) or cyproheptadine (5 or 10mg/kg), but not by indomethacin (2 or 4mg/kg), dexamethasone (1 or 4mg/kg) or cromolyn (40 or 80mg/kg). Some bioactive amines, including histamine and 5-hydroxytryptamine, were found in the venom in low concentrations. In conclusion, the nociceptive response and edema induced by the venom of T. serrulatus may result from the action of multiple mediators including eicosanoids, histamine and 5-hydroxytryptamine. These results may lead to a better understanding of the host response to potent animal toxins and also give insights into a more rational pharmacological approach to alleviate the intense pain associated with the scorpion envenomation.

The effect of tityustoxin (TsTX) on the release of [3H] dopamine in rat brain prefrontal cortical slices was investigated. The stimulatory effect of TsTX was dependent on incubation time and TsTX concentration with an EC50 of 0.05 microM. The release of [3H] dopamine stimulated by TsTX is dependent of Na+ channels and thus, was completely, inhibited by tetrodotoxin. Tityustoxin-induced release of [3H] dopamine was not blocked by ethylene glycol-bis(beta-aminoethyl) ether (EGTA) and thus was independent of extracellular calcium. However, [3H] dopamine release induced by TsTX was inhibited by 52% by BAPTA, a calcium chelator. Moreover, dantrolene (100 microM) and tetracaine (500 microM) partially inhibited by 38 and 29%, respectively, the tityustoxin-induced release of [3H] dopamine from prefrontal cortical slices suggesting a role from intracellular calcium increase. In conclusion, part of the TsTX-induced release [3H] dopamine may be due to an effect of the toxin on the reversal of the dopamine transporter (DAT), but the majority of the toxin stimulated release of [3H] dopamine involves the mobilization of intracellular calcium stores.

Previous research from our Laboratory has shown a greater susceptibility of young animals, when compared to adults, to envenomation by tityustoxin (TsTX), one of the main toxins from Tityus serrulatus scorpion venom. Our hypothesis is that a differential body distribution of TsTX among adult and young animals could account for the worse prognosis of scorpion envenomation in infants. Thus, TsTX labeled with technetium-99m was injected (6 microg, subcutaneous) in adult (150-160 day-old) and young (21-22 day-old) male rats. Groups of animals were sacrificed at different times after TsTX injection (0.08, 1.0, 3.0, 6.0, 12.0 and 24.0 hours) under Urethane anesthesia (140 mg/100 g, i.p.). The brain, heart, lungs, liver, kidneys, spleen and thyroid were excised and blood collected. Young rats presented a shorter latency toxin concentration peak in all studied organs except for the liver and the kidney, when compared to adults. The ratio between the area under the curve of the toxin concentration in each organ and that in blood (Kp) indicates higher accumulation in the organs of young animals mainly for brain, liver and heart. These observations suggest a faster toxin distribution in the organs of young rats. The higher uptake of TsTX in the brain is suggestive of a greater permeability for the toxin along the blood-brain barrier of young rats. In conclusion, the higher uptake in heart, together with data from the brain, may help to elucidate the clinical manifestations frequently observed in children under scorpion envenomation.

In this work we submitted adult male Wistar rats to intracerebroventricular (icv) and iv microinjections of the fraction tityostoxin (TsTX) from the Tityus serrulatus scorpion venom, to address whether the CNS could account for the systemic alterations previously reported: cardiac arrhythmias, lung edema, and seizures. Animals were injected icv, total volume of 1.0 microl, with either sterile saline (n = 4) or differing doses of TsTX (1.74, n = 5; 0.174, n = 4; 0.087, n = 5; and 0.058 microg, n = 4). The peripheral effect of the highest dose of TsTX used (1.74 microg) was tested through iv injections in the femoral vein (n = 4). All animals were recorded by a Video EEG/ECG system for a maximum period of 90 mins or until death. After recording, the lungs were harvested and weighed to evaluate edema (lung/body wt x100). Our results show that icv injections of TsTX, but not iv injections, were able to provoke heart arrhythmias, lung edema, and seizures. Furthermore, the toxin was capable of producing epileptiform discharges in all animals injected with 1.74 microg of the toxin. In conclusion, the action of TsTX in the CNS may solely account for the peripheral alterations observed in severe cases of Tityus serrulatus scorpion poisoning.

1. Voltage-gated Na+ channels are responsible for initiation and conduction of action potentials. The arrival of an action potential at nerve terminal increases intracellular Na+ and Ca2+ concentrations. Calcium entry into neurons through voltage-dependent calcium channels is associated with a variety of intracellular processes. Scorpion neurotoxins have been used as tools to investigate mechanisms involved in neurotransmitter release. Tityustoxin (TsTX) is an alpha-type toxin that delays Na+-channel inactivation. Toxin-gamma (TiTX-gamma) is a beta-type toxin that induces Na+-channel activation at resting potentials. 2. In the present work, we describe the effects of both toxins on [3H]acetylcholine ([3H]ACh) release from rat cerebrocortical synaptosomes, in the presence or absence of the calcium channels blockers: omega-conotoxin-GVIA (omega-CgTx), 1 microM; omega-agatoxin-IVA (omega-Aga), 30 nM; omega-conotoxin-MVIIC (omega-MVIIC), 1 microM; or verapamil, 1 microM. 3. TsTX evokes [3H]ACh release in a concentration-dependent manner with a gradual increase up to saturation at concentrations of 500 nM. However, release of ACh evoked by TiTX-gamma was not linear regarding the toxin concentration. The [3H]-ACh release evoked by TsTX or TiTX-gamma was partially inhibited by omega-CgTx or omega-Aga, and blocked with omega-MVIIC. Verapamil (1 microM) had no effect. Tetrodotoxin blocked [3H]ACh release evoked by both toxins. 4. These results show that different actions on Na+-channels produce different effects on [3H]ACh release with involvement of distinct presynaptic Ca2+-channels, which supports the idea that sodium channels may modulate neurotransmitter release.

The aim of this study was to evaluate the ability of phenobarbital to block the lung edema observed after intracerebroventricular (icv) injections of tityustoxin (TsTX), a toxic fraction of the Tityus serrulatus venom. We injected 1.74 microg icv (1.0 microl) of TsTX in Wistar rats pre-treated with 0.1 ml intramuscular injections of sterile saline or phenobarbital (60 or 170 mg/kg body weight). After the experiments the lungs were harvested and the pulmonary index (PI = lung/body weight x 100) calculated. The animals pre-treated with saline developed severe lung edema (PI = 1.8 +/- 0.2) after TsTX icv injection whereas those that received 170 mg/kg of phenobarbital presented no lung edema (PI = 0.71 +/- 0.02). Our results suggest that the lung edema induced by TsTX is of neurogenic nature and that 170 mg/kg of phenobarbital blocks TsTX induced lung edema., (Copyright 2002 Elsevier Science Ireland Ltd.)

In this paper, the effect of the alpha-scorpion toxin tityustoxin (TsTX) in the release of gamma-[(3)H]aminobutyric acid ([(3)H]GABA) from rat brain cortical slices is described. The TsTX-stimulatory effect on the release of [(3)H]GABA was dependent on incubation time and TsTX concentration, having an EC(50) of 0.33 microM. Tetrodotoxin (TTX) completely inhibited the TsTX action on [(3)H]GABA release. The scorpion toxin effect was calcium-dependent and involves P/Q calcium channels. beta-Alanine also induces the release of [(3)H]GABA that was not inhibited by TTX but was additive in the presence of TsTX. The data suggest a neuronal origin for the release of [(3)H]GABA by TsTX.

One particularly important class of ion channels in excitable cells are the voltage-dependent sodium channels (VDSC). Knowledge of the distribution of VDSC in living cells is important for studies of neuronal excitability, development, and plasticity. Here, we demonstrate a new method for visualizing the spatial distribution of VDSC in living cells. To illustrate the capabilities of the technique, the distribution of VDSC in GH3 cells was revealed with fluorescent derivatives of the alpha-type and beta-type scorpion toxins in conjunction with laser scanning confocal microscopy. Cells exhibited fluorescent hot spots on the surface of the membrane. This characteristic staining pattern was prevented by pre-incubation with unlabeled native toxins and blocked by membrane depolarization for alpha-type toxins. Labeling was not observed in cells lacking sodium channels (HEK 293) after incubation with fluorescent-labeled toxins. Image processing techniques were applied to identify the location of each cluster of labeled VDSC in these cells. The proposed method eliminates artefacts commonly introduced during sample preparation for immunostaining and should prove to be a valuable research tool for investigating VDSC distribution in living specimens.

The effect of the beta-scorpion toxin, TiTX gamma on the release of [3H]GABA from rat brain cortical slices is described. The stimulatory effect of TiTX gamma on the release of [3H]GABA was dependent on incubation time and TiTX gamma concentration with an EC50 of 0.19 microM. The scorpion toxin effect was calcium dependent and was completely inhibited by tetrodotoxin. beta-Alanine also induced the release of [3H]GABA and this effect was not inhibited by tetrodotoxin but was additive in the presence of TiTX gamma. The data suggest a neuronal origin for the release of [3H]GABA by TiTX gamma.

In the present work the pH and arterial blood gases were measured in fasted and fed male albino rats, weighing 297 +/- 13 g, anaesthetized with urethane (1.4 g/kg, i.p.) before and after injection of T1 fraction from Titys serrulatus scorpion venom, during 60 min. Arterial blood samples were collected at 0, 5, 15, 30 and 60 min for pH, pCO2, pO2, bicarbonate and base-excess analysis. The data showed that the scorpion toxin induced a continuous drop in the blood pH along the time. Hypercapnia and hypoxemia peaking at 30 min and followed by a recovery towards normal values at 60 min were also observed. A pronounced decrease in the blood bicarbonate levels at 60 min and negative base-excess values along with time were evident at 60 min. The comparisons between fasted and fed animals have shown that in the last group the effects of scorpion toxin on the arterial blood gases were less pronounced. We conclude that T1 fraction of Tityus serrulatus scorpion venom induces in anaesthetized rats an acute respiratory acidosis followed by metabolic acidosis.

The effect of reducing reagents on omega-conotoxin GVIA (omega-CgTX) inhibition of the release of [3H]-acetylcholine ([3H]-ACh) induced by tityustoxin, K+ 50 mM and electrical stimulation was investigated in rat brain cortical slices. 2. In cortical slices the inhibition of tityustoxin or electrically-stimulated [3H]-ACh release by omega-CgTX was dramatically increased by reducing reagents ascorbate or beta-mercaptoethanol. Dehydroascorbic acid did not substitute for ascorbate. 3. Depolarization induced by K+ 50 mM caused [3H]-ACh release from cortical slices which was not inhibited by omega-CgTX, even in the presence of ascorbate. 4. In the guinea-pig myenteric plexus, omega-CgTX inhibition of the tityustoxin induced release of [3H]-ACh was independent of ascorbate. 5. It is suggested that N-type-like calcium channels in guinea-pigs myenteric plexus may have pharmacological/biochemical diversity from similar channels of rat cerebral cortex.

The effects of toxin Ts-gamma and tityustoxin purified from Tityus serrulatus scorpion venom were investigated on isolated rat atria. Rat atria were placed in an organ bath containing Krebs-Ringer solution, 30 degrees C, pH 7.4, and bubbled with a gas mixture of 95% O2 and 5% CO2. The atrial rate and contractile force were simultaneously recorded. Addition of toxin Ts-gamma to the bath (0.14 microM) evoked an initial reduction of both atrial rate and contractile force, followed by a small increase in force and a decrease in rate, and finally a long reduction of rate and force. Addition of an identical dose of Ts-gamma 30 or 60 min later did not evoke any effect. Addition of tityustoxin to the bath (0.14 microM) induced an increase of atrial rate and force. Addition of an identical dose of tityustoxin 30 min later evoked similar effects. The negative chronotropic and inotropic effects induced by Ts-gamma were abolished by tetrodotoxin (TTX, 1 microM) or atropine (1.5 microM), whereas the positive effects observed in the presence of atropine were prevented by TTX (1 microM) or alprenolol (10 microM). The negative chronotropic effect of 0.14 microM tityustoxin was only observed in the presence of physostigmine (0.3 microM). This negative effect was abolished by TTX (1 microM) or atropine (1.5 microM). The positive inotropic effect of tityustoxin was decreased by TTX (1 microM and 10 microM), but was totally prevented by guanethidine (10 microM) or alprenolol (10 microM).(ABSTRACT TRUNCATED AT 250 WORDS)

A semi-solid corn-meal diet sterilized in glass flasks in a hospital-type autoclave is described. The values of weight gain, feed efficiency, apparent digestibility, and cecal diameters were the same for germ-free mice fed either on this diet or on a soybean meal and alfalfa diet (L-485), widely used in germ-free research. The corn-meal diet has been successfully used for eight years.

1. The effect of dietary sucrose on carcass composition was determined in conventional (CV) and germ-free (GF) mice fed on a control diet and on diets containing 20 and 40% sucrose. 2. Germ-free mice fed the control diet were heavier than their conventional counterparts. CV mice fed on sucrose diets were, however, heavier than their GF counterparts. 3. GF mice fed on high-sucrose diets showed a lower lipid content on carcass than the GF controls and their CV counterparts. 4. CV mice fed the high-sucrose diets were heavier than the conventional animals fed on the control diet. 5. The water content in the carcass of GF mice fed on high sucrose diets was higher than that of controls, as well as their CV counterparts. 6. The present results suggest different lipidogenic effect of dietary sucrose in germ-free and conventional mice.

1. The effects of Tityus serrulatus scorpion venom and its most important toxin (toxin gamma) were investigated on isolated guinea-pig hearts, perfused with Locke solution, by the Langendorff's method. 2. The cardiac contraction, the coronary flow and the electrocardiogram (ECG) were simultaneously recorded. 3. Bolus injections of 25, 50 or 100 micrograms of scorpion venom and 2.5, 5 or 10 micrograms of toxin gamma in the heart evoked complex effects which were divided into 3 phases: an initial phase (tachycardia or bradycardia associated with an increase in contractile force), an intermediate phase (oscillations of cardiac rate, contractile force and coronary flow, due to wandering pacemakers) and a third phase (sinus tachycardia). 4. The bradycardia and the oscillations of rhythm were prevented by atropine, whereas the tachycardia and the increase in contractile force were prevented either by reserpine or propranolol. 5. Scorpion venom or toxin gamma induced a ST segment displacement in the ECG, explained by a transitory myocardial hypoxia, due to an increase in the contractile force and a simultaneous decrease of the coronary flow. 6. Perfusion of the heart with Locke solution containing 2% scorpion antivenom prevented almost totally the effects elicited by the venom. 7. It is concluded that the complex effects induced by scorpion venom and toxin gamma are due to the simultaneous release of acetylcholine and catecholamines from postganglionic nerve fibers in the heart.

Rats showing signs of pellagra-like disease associated with gelatin ingestion were injected with [14C]-nicotinic acid into the portal vein and the incorporation of the label into hepatic NAD+ and NADP+ was determined. The quantity of NADP+ synthesized within 1 h after [14C]-nicotinic acid injection by the experimental group, supplemented or not with dietary niacin, was 75% lower than that synthesized by the control group fed a casein control diet (141 nmol h-1 g-1). The NAD+ and NADP+ contents of the liver of the experimental animals were 245 and 93 micrograms/g liver, respectively, i.e., significantly lower than those for the controls (597 and 210 micrograms/g, respectively). Dietary supplementation with niacin increased the NAD+ content to 317 micrograms/g liver but had no statistical effect on NADP+ content (75 micrograms/g liver) or NADP+ synthesis (46.5 in the free niacin group vs 37.0 nmol h-1 g-1 in the niacin-supplemented group). Therefore, NAD+ content did not seem to limit NADP+ synthesis. These results suggest that the lower availability of NADP+ is responsible, at least in part, for some metabolic derangements in pellagra-like disease, such as a decrease in the activity of NADP(+)-dependent enzymes observed in quail muscle.

A study was carried out to determine the chemical composition and dietary effects of the oil from "mandi" (Pimelodus clarias). Findings revealed that it had a low proportion of essential C18:2 fatty acids and a high percentage of oleic acid (20.93%), as well as polyunsaturated fatty acids with more than 18 carbon atoms. Of the long-chain fatty acids, C22:6 was present in the highest percentage (1.94%). When rats were fed a diet the lipid source of which was the oil of mandi, they showed a reduced growth rate as compared to animals receiving the control diet (corn oil). The fatty acid composition of the liver and heart of rats from the experimental group was modified. The greatest variation occurred in the percentage of C22:6 in the heart muscle, wherein a five-fold increase was observed. Reduction of growth and alteration in the polyunsaturated fatty acids levels may be due to a deficiency in C18:2-w6, or to a possible imbalance between C18:2-w6 and C18:3-w3.

The nutritive value of calf stomach waste from rennet production was assessed. The by-product was either drum-dried at the industry (DDSW) or mildly dried (60 degrees C) in a ventilated oven at the laboratory (MDSW). Part of the sample treated at the laboratory was autoclaved before drying (ASW). The protein and ether extract concentrations ranged from 34.25% to 44.31% and from 31.14% to 38.95%, respectively, and were lower in DDSW than in the other two samples. The ash content which ranged from 16.04% to 27.38%, was higher in the DDSW sample. Methionine, cystine, and available lysine were lower in the DDSW by-product than in the other two samples. Lysine availability was affected by the drum-drying and autoclaving procedures. Tryptophan was shown to be the limiting amino acid in all samples. Nutritive value, assayed both in DDSW and ASW, was lower in DDSW when compared with ASW or with casein diets as far as PER is concerned. The digestibility was shown to be ranked in increasing order for DDSW, ASW and casein. In conclusion, the results indicate that the waste from rennet production may be recommended as in inexpensive source of protein for animal feeding.

Drum-dried calf stomach waste (DDSW) from rennet production, after washing with water for salt elimination, as well as drum-dried porcine pancreas waste (DDPW) from protease extraction were used. Their chemical composition and nutritional values were studied. The protein content of DDPW was two-thirds that of the DDSW sample. The amount of ashes (acid soluble and insoluble) in DDPW was twice that of DDSW. Four diets, the protein sources of which were casein, DDSW, DDPW and a mixture of equal parts of both by-products (MSPW) were then prepared. Their nutritive value was assessed in weanling rats through the following parameters: food ingestion, body weight gain, protein efficiency ratio (PER) and and apparent protein digestibility. The DDSW sample showed the lowest nutritional quality for all parameters measured. The weight gain of rats fed diets containing casein, DDPW, MSPW, and DDSW were, 59.60 +/- 8.12, 84.63 +/- 9.89, 56.22 +/- 6.74, and 20.08 +/- 3.72, respectively. The PER for the same diets were: 2.82 +/- 0.33, 2.67 +/- 0.19, 2.17 +/- 0.25, and 1.30 +/- 0.20, in the same order. The protein digestibility of all experimental diets was about 70%, and that of the control diets, about 90%. As a conclusion, the by-product of protease production from porcine pancreas seems adequate as a protein source for animal feeding, despite its high level of ashes. Also, this by-product appears to be useful for feeding purposes, when mixed with the other animal by-products studies.

1. The objective of the present study was to examine the reversibility of the effect of neonatal malnutrition on in vivo neuronal protein synthesis. 2. Neonatal malnutrition was achieved by letting the pups suckle mothers fed a low casein diet (6.4%) which decreased the amount of milk produced. The control group was composed of pups from mothers fed a 17% casein diet and kept on the same diet after weaning. A group of pups was undernourished during the suckling period, as above, but fed a 17% casein diet after weaning (recovered group). All animals were killed at 60 days of age. 3. Neuronal cell bodies were separated from forebrain by mechanical disruption of the tissue and centrifugation in a sucrose gradient. 4. Protein biosynthesis was measured by the incorporation of [14C]-leucine into proteins of neuronal cell bodies, after correcting for the specific activity of the precursor. 5. Forebrain cell number and cell size were evaluated by DNA content and the protein/DNA ratio, respectively. 6. The forebrains of the recovered group weighed less than those of the controls, perhaps due to a lower number of cells, which exhibited an average size similar to that of the control group. 7. The rate of protein biosynthesis was similar in malnourished and recovered groups, but both were only 57% of the rate of the control group. 8. We conclude that neonatal malnutrition promotes an irreversible adverse effect on in vivo neuronal protein synthesis by rats.

Rats inoculated with Trypanosoma cruzi were divided into two groups maintained, respectively, on a low and a rich protein diet. Ninety-eight days after inoculation the cardiac levels of noradrenalin in the well-nourished infected rats were not different from those of uninfected controls. In the malnourished group, however, these levels were around 50% of the control values. In addition, the adrenergic nerve terminals were clearly reduced in the auricular appendages of 87.5% of the malnourished infected rats. These results indicate that protein deficiency at least delays the recovery of noradrenalin levels that normally occurs during the chronic phase of experimental Chagas' disease.
