The present investigation was carried out to identify the potential donors of resistant gene(s)/the source of white rust disease in B. juncea using 30 genotypes, including locally adapted accessions and advanced breeding lines. Out of 30 genotypes, ten lines viz. Bio-YSR, CAULC-1, CAULC-2, CAULC-3, CAULC-4, CAURM-2, CAULR-7, CAURM-4, CAURM 4-1, and CAURM 4-2 exhibited a lower PDI value (lesser than mean 10.83) with a superior agronomic performance related with the disease. The evaluation of these ten genotypes for the presence of the BjuWRR1 gene using a gene-based marker depicted the presence of the functional allele of the BjuWRR1 gene in the five genotypes viz., Bio-YSR, CAULC-1, CAULC-3, CAURM 4-1 and CAURM 4-2. When compared with the sequenced amplicon of these genotypes, it is found to be identical with that of an east European Brassica juncea line, Donskaja-IV, the completely resistant genotype against various isolates of Albugo candida. The findings from the present study suggested that besides Bio-YSR, the local lines of Manipur CAULC-1 (Local Yella of Potshangbam) and CAULC-3 (Local Yella of Kakching Lamjao) can be used as the potential white rust resistance sources/donors in disease resistance breeding programmes for the development of elite B. juncea cultivars in the future. In addition to the local lines, two improved advanced lines, viz. CAURM 4-1 and CAURM 4-2, obtained from a hybridization programme, may be further evaluated for releasing resistant varieties against white rust. [ABSTRACT FROM AUTHOR]

The vegetables used by common man are rich sources of nutrients and have been the important constituent of the human diet. Recently researches have been conducted which elucidate another important aspect of the vegetables that is their capacity as source of important medicinal benefits. The present review article is an effort to understand the importance of vegetables for common man and its role as nutraceuticals for preventing many common and complicated human diseases. Identifying elite lines is a major research priority for developing new hybrids/varieties. A total of 153 tomato lines were characterized based on horticultural traits, total soluble solids (TSS) and tomato yellow leaf curl virus (ToLCV) resistance under natural field conditions. Molecular markers were applied for ToLCV (Ty-2 and Ty-3), late blight (Ph-3) and root-knot nematode (Mi-1.2) resistance genes. Based on horticultural traits such as average fruit weight, fruit yield, TSS and ToLCV resistance, promising lines were identified belonging to improved lines, jointless tomato, cherry tomato, beta-carotene-rich lines, germplasm and varieties. An improved line VRT-02 possessing dwarf plant type was found suitable for pot culture, whereas H-88-78-2 had a delayed and partial fruit ripening. Formyl peptide receptors (FPR), part of the G-protein coupled receptor superfamily, are pivotal in directing phagocyte migration towards chemotactic signals from bacteria and host tissues. Although their roles in acute bacterial infections are well-documented, their involvement in immunity against tuberculosis (TB) remains unexplored. This study investigates the functions of Fpr1 and Fpr2 in defense against Mycobacterium tuberculosis (Mtb), the causative agent of TB. Elevated levels of Fpr1 and Fpr2 were found in the lungs of mice, rabbits and peripheral blood of humans infected with Mtb, suggesting a crucial role in the immune response. The effects of Fpr1 and Fpr2 deletion on bacterial load, lung damage, and cellular inflammation were assessed using a TB model of hypervirulent strain of Mtb from the W-Beijing lineage. While Fpr2 deletion showed no impact on disease outcome, Fpr1 -deficient mice demonstrated improved bacterial control, especially by macrophages. Bone marrow-derived macrophages from these Fpr1 -/- mice exhibited an enhanced ability to contain bacterial growth over time. Contrarily, treating genetically susceptible mice with Fpr1-specific inhibitors caused impaired early bacterial control, corresponding with increased bacterial persistence in necrotic neutrophils. Furthermore, ex vivo assays revealed that Fpr1 -/- neutrophils were unable to restrain Mtb growth, indicating a differential function of Fpr1 among myeloid cells. These findings highlight the distinct and complex roles of Fpr1 in myeloid cell-mediated immunity against Mtb infection, underscoring the need for further research into these mechanisms for a better understanding of TB immunity., Competing Interests: Competing interests The authors declare no competing interests.

The prokaryotic adaptive immune system has clustered regularly interspaced short palindromic repeat. CRISPR-associated protein (CRISPR-Cas) genome editing systems have been harnessed. A robust programmed technique for efficient and accurate genome editing and gene targeting has been developed. Engineered cell therapy, in vivo gene therapy, animal modeling, and cancer diagnosis and treatment are all possible applications of this ground-breaking approach. Multiple genetic and epigenetic changes in cancer cells induce malignant cell growth and provide chemoresistance. The capacity to repair or ablate such mutations has enormous potential in the fight against cancer. The CRISPR-Cas9 genome editing method has recently become popular in cancer treatment research due to its excellent efficiency and accuracy. The preceding study has shown therapeutic potential in expanding our anticancer treatments by using CRISPR-Cas9 to directly target cancer cell genomic DNA in cellular and animal cancer models. In addition, CRISPR-Cas9 can combat oncogenic infections and test anticancer medicines. It may design immune cells and oncolytic viruses for cancer immunotherapeutic applications. In this review, these preclinical CRISPRCas9- based cancer therapeutic techniques are summarised, along with the hurdles and advancements in converting therapeutic CRISPR-Cas9 into clinical use.
