Sonodynamic therapy (SDT), which involves a combination of low-intensity ultrasound (US) and a chemical sonosensitizer, has emerged as a promising minimally invasive and selective approach for the treatment of deep tumors. US not only increases cell membrane permeability, thereby enhancing the cellular uptake efficiency of chemical sonosensitizers, but also excites chemical sonosensitizers in deep tissues to generate reactive oxygen species (ROS), which in turn kill cancer cells. As compared with inorganic materials, organic sonosensitizers exhibit a defined chemical structure, controlled synthesis process, excellent biodegradability, and high ROS generation. However, they frequently exhibit poor water solubility, strong phototoxicity, and low tumor tissue targeting capacity. In this review, we address the currently accepted mechanisms in SDT and the recent progresses in organic sonosensitizers, discuss the strategies to improve the water solubility, tumor targeting capacity, biocompatibility, and therapeutic effects of organic sonosensitizers. The advantages of combining SDT with other cancer treatments, including phototherapy, chemodynamic therapy, chemotherapy, starvation therapy, gas therapy, and immune therapy are also demonstrated. Finally, the existing challenges and future perspectives on clinical SDT of tumor are discussed.

C dots (CDs) have shown great potential in bioimaging and phototherapy. However, it is challenging to manipulate their fluorescent properties and therapeutic efficacy to satisfy the requirements for clinic applications. In this study, we prepared S, Se-codoped CDs via a hydrothermal method and demonstrated that the doping resulted in excitation wavelength-independent near-infrared (NIR) emissions of the CDs, with peaks at 731 and 820 nm. Significantly, the CDs exhibited a photothermal conversion efficiency of ~58.2%, which is the highest reported value for C nanostructures and is comparable to that of Au nanostructures. Moreover, the CDs had a large two-photon absorption cross section (~30,045 GM), which allowed NIR emissions and the photothermal conversion of the CDs through the two-photon excitation (TPE) mechanism. In vitro and in vivo tests suggested that CDs can function as new multifunctional phototheranostic agents for the TPE fluorescence imaging and photothermal therapy of cancer cells.

A family of anthracene carboxyimides with multi-color fluorescence have been successfully prepared, and their spectroscopic properties were investigated by UV–vis absorption spectroscopy, steady-state and time-resolved fluorescence spectroscopy. The photostability and solvatochromic fluorescence in organic solvents were also investigated. The parent anthracene carboxyimide 4a displays very strong green fluorescence (Φ F = 0.91 in EtOH), good pH stability and photostability, as well as a large Stokes shift. By attachment of different substituents at the 10-position of the anthracene carboxyimide, the fluorescence maxima could be effectively modulated from 500 nm to 615 nm. The 10-(picolylamine) substituted anthracene carboxyimides 5a and 5b exhibit large Stokes shifts (>80 nm) and significant positive solvatochromic fluorescence in organic solvents due to their intramolecular charge-transfer (ICT) character. Furthermore, cell staining experiments indicate that anthracene carboxyimides have excellent cell membrane permeability and good photo-bleaching resistance in living cells. Particularly, compound 5a is a specific mitochondria-targeting dye with red fluorescence. All these features enable anthracene carboxyimides to serve as promising candidates for bio-labeling/imaging applications.

Surface ligand-removed and Cd-rich CdSe quantum dots (QDs) exhibited exceptional activity as photocatalyst for the conversion of CO

Photodynamic therapy (PDT) has been a preferred clinical technology for treating superficial tumors due to its advantages of high selectivity, non-invasiveness and negligible drug resistance. However, the hypoxic tumor microenvironment weakens the efficiency of O2-dependent PDT. Moreover, the PDT process consumes a large amount of O2 and destroys the tumor blood vessels and further blocks the O2 supply to tumor sites. Therefore, developing more advanced materials and methods for PDT of the hypoxic tumor is an essential scientific significance. This tutorial review summarizes the strategies for improving the efficacy of PDT in hypoxic tumor therapy, which is categorized into three sections: (I) enhancing O2 concentration in the tumor; (II) disregarding hypoxia; and (III) exploiting hypoxia. The advantages of combining PDT with other therapeutics, such as chemotherapy, chemo-dynamic therapy, gas therapy, immunotherapy and gene therapy, are also demonstrated. Finally, the existing challenges and future perspectives on clinical PDT for hypoxic tumors are discussed.

Due to the advantages of in situ, real-time, and non-destructive detection of target molecules, fluorescent probes have become an indispensable tool in the fields of disease diagnosis and environmental and food safety monitoring. However, with the development of modern science and technology, sensing techniques have new challenges and requirements, such as increased sensitivity, better selectivity, and high-throughput visual detection in complicated practical samples. Developing effective fluorescent probes is still ongoing research. Fluorescent carbon dots (CDs) are a novel carbon-based nanomaterial that have attracted tremendous attention because of their excellent biocompatibility, tunable fluorescence properties, good photostability, and easy synthesis and surface modification, and they have shown great potential in fluorescent sensing and imaging applications. In this review, we summarize the recent advances in CD-based fluorescent probes, including the design strategies, sensing mechanisms, and their applications in sensitive and selective detection of public safety substances, organic amines, phenols, bioactive molecules, biological enzymes, and inorganic salts. A future perspective is introduced to highlight the existing challenges and possible strategies to overcome the problems in practical application of currently developed CD-based fluorescent probes, which may be instructive to the design of effective fluorescent probes in the future.

Carbon dots (CDs) are novel zero-dimensional carbonaceous nanomaterial, and they are featured with an attractive combination of physical, chemical, and biomedical properties including wide absorption spectrum, tunable fluorescence, excellent water solubility, outstanding photostability, and biocompatibility, which leads to their diverse applications as optically functional materials in various fields, e.g., fluorescence sensing, bioimaging, and drug delivery. Recently, it was revealed that CDs could be used as an effective photosensitizer to generate reactive oxygen species under light excitation, and/or a photothermal agent to convert light energy into heat, which enable them to be applied in photodynamic therapy or photothermal therapy for cancer treatment. In this review, we summarize the latest progress of research on CDs in phototherapy, including their use as nanocarriers for organic phototherapy agents, and highlight the strategies which are adopted to improve the therapeutic efficacy of CDs-based phototheranostics. Furthermore, we also discuss the challenges and prospects of the CDs-based phototheranostics towards clinical applications. We hope this review will provide critical insights to inspire new exciting discoveries on CDs from both fundamental and practical views so as to prompt their advances in the phototherapy.

In using nanomaterials for advanced photodynamic therapy, it is important to improve surface modification to increase stability, minimize non-specific loss of drugs in blood circulation, enhance the photostability of photosensitizers, and achieve highly efficient intracellular uptake. In this report, we for the first time covalently conjugate photosensitizer Chlorin e6 (Ce6) to polyethylene glycol modified layered double hydroxides and produce hybrid nanoflakes. These nanoflakes display many superior characteristics and lead to excellent in vivo photodynamic therapeutic efficiency and safety profiles.

A carbon dot (CD)-based fluorometric probe for oxytetracycline (OTC) detection utilizing a Forster resonance energy transfer (FRET) mechanism was firstly developed. Upon addition of OTC, the blue fluorescence of the CDs peaked at 405 nm was quickly quenched, with a new fluorescence peaked at 505 nm attributed to OTC was observed within 30 s. A visual fluorescent color change of the CDs solution from blue to green was discerned under a 365 nm UV light irradiation. The CDs displayed a high sensitivity and selectivity toward OTC with a low detection limit of 0.41 μM. Furthermore, the probe was applied to detect OTC in water, milk, and pork samples with a satisfied recovery.

S,N-doped carbon dots (S,N-CDs) were facilely prepared via a hydrothermal method by using thiourea and citric acid (CA) as precursors. The as-prepared S,N-CDs exhibited high stability in aqueous solution and were applied for fluorometric detection of tetracyclines (TCs). The approach was developed on the basic of the fluorescence of S,N-CDs at 440 nm, which was selectively quenched by TCs through the inner filter effect (IFE) mechanism. Moreover, the fluorescence of TCs with maximum at 600 nm could be used as the internal standard signal, resulting in a fluorometric spectral response. The limit of detection (LOD) was 0.25, 0.39 and 0.26 μM for tetracycline (TC), doxycycline (DC) and oxytetracycline (OTC), respectively. Furthermore, S,N-CDs were successfully utilized for the detection of residual TCs in fish and pork samples with acceptable recovery and good RSD.

Phototherapy, including photodynamic therapy (PDT) and photothermal therapy (PTT) holds great potential for efficient cancer therapy by inducing reactive oxygen species (ROS) or heat into tumor. Nevertheless, PDT or PTT suffers from some limitations, e.g., PTT requires long-time and high-power laser irradiation to generate enough heat, while the hypoxia microenvironment of tumor and the limit diffuse distance of ROS hamper the efficacy of oxygen-dependent PDT. Here we reported the carbon dots (CDs) which could simultaneously generate singlet oxygen (1O2), hydroxyl radical (OH ), and heat under a 635 nm laser irradiation, with a 1O2 generation quantum yield of 5.7% and photothermal conversion efficiency of 73.5% (the highest thus far for CDs). Significantly, the CDs can selectively accumulate in lysosome, which is an ideal organelle for phototherapy because of its key role in sustaining cellular activity and stability. Moreover, the CDs present one-photon excited (OPE) and two-photon excited (TPE) fluorescence, and excellent photoacoustic (PA) imaging capability. Combining the good biocompatibility, the as-prepared CDs was served as multi-functional phototheranostic agent for synergistic PA/fluorescence imaging, and PDT/PTT.

This paper reports on new applications of water- dispersible graphene quantum dots (GQDs) that we recently developed. The prepared GQDs not only show broad absorption in the visible spectrum from 400 to 700 nm, but can also serve as smart photosensitizers with high singlet oxygen (1O2) production under visible-light irradiation (≥420 nm). We showed that the prepared G QDs can potentially be used as a metal-free, visible-light-active, sensitized photocatalyst via e nergy transfer mechanism, in which the light energy was converted by GQDs to produce 1O2, which can kill nearby microorganisms and degrade organic dyes.

We report an amphiphilic macromolecule containing a cyclometalated gold(III) complex that self-assembles into nano-sized micelles and also displays a long-lived emissive triplet excited state with a lifetime of 84 μs in degassed water. This amphiphilic AuIII complex exhibits good biocompatibility and activity towards in vitro photo-toxicity, as well as enhanced permeability and retention effects in vivo.

Cancer nanotheranostics combining therapeutic and imaging functions within a single nanoplatform are extremely important for nanomedicine. In this study, carbon dots (C-dots) with intrinsic theranostic properties are prepared by using polythiophene benzoic acid as carbon source. The obtained C-dots absorb light in the range of 400-700 nm and emit bright fluorescence in the red region (peaking from 640 to 680 nm at different excitations). More importantly, the obtained C-dots exhibit dual photodynamic and photothermal effects under 635 nm laser irradiation with a singlet oxygen ((1)O2) generating efficiency of 27% and high photothermal conversion efficiency of 36.2%. These unique properties enable C-dots to act as a red-light-triggered theranostic agent for imaging-guided photodynamic-photothermal simultaneous therapy in vitro and in vivo within the therapeutic window (600-1000 nm).

Nitrogen and sulfur codoped carbon dots (CDs) were prepared from garlic by a hydrothermal method. The as-prepared CDs possess good water dispersibility, strong blue fluorescence emission with a fluorescent quantum yield of 17.5%, and excellent photo and pH stabilities. It is also demonstrated that the fluorescence of CDs are resistant to the interference of metal ions, biomolecules, and high ionic strength environments. Combining with low cytotoxicity properties, CDs could be used as an excellent fluorescent probe for cellular multicolor imaging. Moreover, the CDs were also demonstrated to exhibit favorable radical scavenging activity.

Metal-free, organic-dye-based fluorescent nanorods were fabricated through a simple solvent-exchange procedure. The as-prepared nanorods exhibit low toxicity to living cells and excellent photostability. Furthermore, they are stable in solutions of various pHs and high ionic strength and in solutions with interfering metal ions. Compared with the free DPP-Br molecules in THF, these nanorods exhibit larger Stokes shift, broader absorption spectra, and greatly improved photostability. We successfully demonstrated the application of the nanorods, including their aforementioned beneficial characteristics, as a good fluorescence probe for bio-imaging.

A colorimetric and fluorescent dual-responses chemosensor ( PT3 , a water-soluble polythiophene) for streptomycin was designed and synthesized. The structure of PT3 was characterized by using infrared spectroscopy, 1 H NMR and gel-permeation chromatography analyses. The conformational change of PT3 induced by streptomycin resulted in the red shift of absorption spectra and fluorescent quenching. Moreover, PT3 showed excellent selectivity for streptomycin over other antibiotics and biomolecules. PT3 could quantificationally detect streptomycin in the range of 2–70 μM with a detection limit of 0.2 μM (116 ppb), which is lower than the maximum residue limit defined by World Health Organization (200 ppb).

Carbon nano-dots were synthesized by a hydrothermal method and integrated into the poly(vinyl alcohol) matrix as charge carrier trapping centers for nonvolatile memory devices. The devices with the configuration of ITO/carbon nano-dots-PVA/Ag exhibited bistable electrical switching at low voltage, long retention time and excellent reading stability.

Metallic lithium (Li) is a promising anode material for next-generation rechargeable batteries. However, the dendrite growth of Li and repeated formation of solid electrolyte interface during Li plating and stripping result in low Coulombic efficiency, internal short circuits, and capacity decay, hampering its practical application. In the development of stable Li metal anode, the current collector is recognized as a critical component to regulate Li plating. In this work, a lithiophilic Cu-CuO-Ni hybrid structure is synthesized as a current collector for Li metal anodes. The low overpotential of CuO for Li nucleation and the uniform Li+ ion flux induced by the formation of Cu nanowire arrays enable effective suppression of the growth of Li dendrites. Moreover, the surface Cu layer can act as a protective layer to enhance structural durability of the hybrid structure in long-term running. As a result, the Cu-CuO-Ni hybrid structure achieves a Coulombic efficiency above 95% for more than 250 cycles at a current density of 1 mA cm-2 and 580 h (290 cycles) stable repeated Li plating and stripping in a symmetric cell.

Positively charged water-soluble polythiophene (PT0) that could self-assemble into nanoparticles in pure water solution was designed and synthesized. PT0 exhibited high photostabilities and pH stabilities, excellent biocompatibility, strong 1O2 generation capability, and large two-photon absorption cross sections. Moreover, we showed that the fluorescence of PT0 was unaffected by the interference of biomolecules and metal ions. As an example application, PT0 was demonstrated to be capable of simultaneous cell imaging and photodynamic therapy under either one-photon or two-photon excitation modes.

Carbon dots (CDs) with blue fluorescence were synthesized through hydrothermal treatment of p-dihydroxybenzene and hydrazine hydrate. The fluorescence of the investigated CDs could be selectively quenched by iron ions (Fe3+) because of the strong metal-ligand coordination and electrostatic interactions between the large amount of hydroxyl, amino, and carboxyl groups on CDs’ surface and Fe3+. Interestingly, this non-fluorescent CDs + Fe3+ complex displayed a sensitive and selective turn-on fluorescence toward ampicillin with a limit of detection of 0.70 μM. The sensing process could be observed by naked eyes under the 365 nm UV light irradiation. Importantly, CDs + Fe3+ complex had been successfully applied in the detection of ampicillin in river water.

A novel nanohybrid ratiometric fluorescence sensor is developed for selective detection of mercuric ions (Hg(2+)), and the application has been successfully demonstrated in HEPES buffer solution, lake water, and living cells. The sensor comprises water-soluble fluorescent carbon nanoparticles (CNPs) and Rhodamine B (RhB) and exhibits their corresponding dual emissions peaked at 437 and 575 nm, respectively, under a single excitation wavelength (350 nm). The photoluminescence of the CNPs in the nanohybrid system can be completely quenched by Hg(2+) through effective electron or energy transfer process due to synergetic strong electrostatic interaction and metal-ligand coordination between the surface functional group of CNPs and Hg(2+), while that of the RhB remains constant. This results in an obviously distinguishable fluorescence color variation (from violet to orange) of the nanohybrid solution. This novel sensor can effectively identify Hg(2+) from other metal ions with relatively low background interference even in a complex system such as lake water. The detection limit of this method is as low as 42 nM. Furthermore, the sensing technique is applicable to detect Hg(2+) in living cells.

Cyan-emitting carbon dots (CDs) with a high fluorescence quantum yield of 49% were synthesized through hydrothermal treatment of 1,3,6-trinitropyrene and Na2SO3 in alkaline solution. The as-prepared CDs could be used as a label-free fluorescent probe for sensitive and selective detection of ferric ions (Fe3+). Spectral measurements revealed that the fluorescence of the CDs was quenched significantly and the emission wavelength was red-shifted from 490 nm to 515 nm upon the addition of Fe3+, which was demonstrated to be due to the formation of Fe3+-phenol complex induced inner filter effect. Furthermore, with the addition of dopamine (DA), the strong binding interaction between the catechol groups of DA and Fe3+ can extract Fe3+ from the surface of CDs, and turn on the quenched fluorescence. The formation of polydopamine (PDA) by DA and Fe3+ generated an emission peak at ∼470 nm, making the CDs + Fe3+ system a ratiometric fluorescence sensor for DA as well and the sensing processes could be directly observed by naked eye under a 365 nm UV light. Low limits of detection of 0.86 μM and 0.18 μM were achieved for Fe3+ and DA, respectively.

As an emerging clinical modality for cancer treatment, photodynamic therapy (PDT) takes advantage of the cytotoxic activity of reactive oxygen species (ROS) that are generated by light irradiating photosensitizers (PSs) in the presence of oxygen (O2 ). However, further advancements including tumor selectivity and ROS generation efficiency are still required. Substantial efforts are devoted to design and synthesize smart PSs with optimized properties for achieving a desirable therapeutic efficacy. This review summarizes the recent progress in developing intelligent PSs for efficient PDT, ranging from single molecules to delicate nanomaterials. The strategies to improve ROS generation through optimizing photoinduced electron transfer and energy transfer processes of PSs are highlighted. Moreover, the approaches that combine PDT with other therapeutics (e.g., chemotherapy, photothermal therapy, and radiotherapy) and the targeted delivery in cancer cells or tumor tissue are introduced. The main challenges for the clinical application of PSs are also discussed.

A water-soluble, polythiophene-based colorimetric sensor was designed for selective and sensitive detection of carbenicillin in HEPES buffer solution. Quaternized quinine was linked to thiophene through bis-functionality benzyl group, which can interact with carbenicillin via electrostatic interaction and geometric match effect. The sensor exhibited a colorimetric signal change upon the addition of carbenicillin because of the formation of more nonplanar structures. However, the addition of other beta-lactam antibiotics or dicarboxylic acids into the sensor solution caused no obvious changes in absorbance intensity ratio. This result may be attributed to the cavity formed by the semi-rigid framework of PTQ2, which is suitable for the special binding with carbenicillin. This novel sensor can effectively distinguish carbenicillin from other beta-lactam antibiotics and has a wide linear range response in HEPES buffer solution. Linear calibration curves are obtained with 0 to 18 μmol/L of HEPES buffer solution, with a limit of detection of 0.54 μmol/L.

3-Phenylthiophene-based water-soluble copolythiophenes (CPT9) were designed for colorimetric and fluorometric detection of lysophosphatidic acid (LPA) based on electrostatic interaction, hydrophobic interaction, and hydrogen bonding. Other negatively charged species gave nearly no interference, and the detection limit reached to 0.6 μM, which is below the requisite detection limits for LPA in human plasma samples. The appealing performance of CPT9 was demonstrated to originate from the multipoint interaction-induced conformational change of conjugated backbone and weakened electron transfer effect. To our best knowledge, this is the first polythiophene based optical sensor which displays emission peak red-shift followed with fluorescence enhancement.

Given their easy structural modification and good biocompatibility advantages, near-infrared (NIR) organic dyes with a large molar extinction coefficient, while a superlow fluorescence quantum yield shows considerable potential application in photothermal therapy (PTT). Herein, a new NIR-absorbing asymmetric cyanine dye, namely, RC, is designed and synthesized via the hybrid of rhodamine and hemicyanine derivatives. RC-BSA nanoparticles (NPs) are fabricated by using the bovine serum albumin (BSA) matrix. The NPs exhibit a strong NIR absorption peak at ∼868 nm and 28.7% photothermal conversion efficiency. Based on these features, RC-BSA NPs exhibit excellent performance in ablating tumor under a 915 nm laser radiation through a PTT mechanism. These NPs show no obvious toxicity to the treated mice. Thus, RC-BSA NPs can used as a new NIR laser-triggered PTT agent in cancer treatment.

We have developed a conceptually new design strategy for fluorescent probes based on a combinational use of photo-induced electron transfer (PET) and excited-state intramolecular proton transfer (ESIPT) mechanisms. This turn-on fluorescence thiol-probe shows fast responses to mercapto-biomolecules with high sensitivity and selectivity. Furthermore, it was used for imaging in living cells with excellent performance.

A carbon dot-based fluorescence turn-on sensor for hydrogen peroxide (H2O2) with a photo-induced electron transfer mechanism was developed. The sensor exhibits good selectivity, sensitivity, and rapid response to H2O2 with a detection limit of 84 nM. The sensor maintains excellent sensing performance over a wide pH range.

Clinical applications of current photodynamic therapy (PDT) agents are often limited by their low singlet oxygen (1O2) quantum yields, as well as by photobleaching and poor biocompatibility. Here we present a new PDT agent based on graphene quantum dots (GQDs) that can produce 1O2 via a multistate sensitization process, resulting in a quantum yield of ~1.3, the highest reported for PDT agents. The GQDs also exhibit a broad absorption band spanning the UV region and the entire visible region and a strong deep-red emission. Through in vitro and in vivo studies, we demonstrate that GQDs can be used as PDT agents, simultaneously allowing imaging and providing a highly efficient cancer therapy. The present work may lead to a new generation of carbon-based nanomaterial PDT agents with overall performance superior to conventional agents in terms of 1O2 quantum yield, water dispersibility, photo- and pH-stability, and biocompatibility., Photosensitisers are used in cancer therapy to promote the formation of reactive oxygen species on irradiation with light. Here, the authors present a graphene quantum dot photosensitiser with a singlet oxygen quantum yield of approximately 1.3, and investigate its in vitro and in vivo applications

Volvox-like Cdx Zn1-x S solid solutions with a cubic zinc blend structure were synthesized through a template-free ethylene glycol process. Cd(Ac)2 ⋅2 H2 O, Zn(Ac)2 ⋅2 H2 O, and thiourea are used as the starting materials and dissolved in ethylene glycol. These reaction precursors and solvent not only contributed to control over the formation of the volvox-like spherical geometry, but also exerted vigorous domination for existence of cubic-phase Cdx Zn1-x S nanostructures. As-prepared volvox-like Cdx Zn1-x S nanospheres have a diameter of around 100 nm with extensional shells. These samples show excellent photocatalytic H2 evolution activity from water splitting under visible-light irradiation without any cocatalyst or scaffolding, owing to their tunable band gap, cubic zinc blend structure, and unique hierarchical porous structure with a high surface area (as high as 95.2 m(2) g(-1) ).

A new ratiometric fluorescent sensor based on a quaternized quinine substituted 3-phenylpolythiophene derivative (PTQ1) was designed and developed for facile and reliable detection of carbenicillin with high sensitivity and selectivity.
