Population rate or activity equations are the foundation of a common approach to modeling for neural networks. These equations provide mean field dynamics for the firing rate or activity of neurons within a network given some connectivity. The shortcoming of these equations is that they take into account only the average firing rate while leaving out higher order statistics like correlations between firing. A stochastic theory of neural networks which includes statistics at all orders was recently formulated. We describe how this theory yields a systematic extension to population rate equations by introducing equations for correlations and appropriate coupling terms. Each level of the approximation yields closed equations, i.e. they depend only upon the mean and specific correlations of interest, without an {\it ad hoc} criterion for doing so. The digital reconstruction of a slice of rat somatosensory cortex from the Blue Brain Project provides the most complete simulation of a piece of excitable brain matter to date. To place these efforts in context and highlight their strengths and limitations, we introduce a Biological Imitation Game, based on Alan Turing’s Imitation Game, that operationalizes the difference between real and simulated brains.

Mean field theories have been a stalwart for studying the dynamics of networks of coupled neurons. They are convenient because they are relatively simple and possible to analyze. However, classical mean field theory neglects the effects of fluctuations and correlations due to single neuron effects. Here, we consider various possible approaches for going beyond mean field theory and incorporating correlation effects. Statistical field theory methods, in particular the Doi–Peliti–Janssen formalism, are particularly useful in this regard.

Much progress has been made in uncovering the computational capabilities of spiking neural networks. However, spiking neurons will always be more expensive to simulate compared to rate neurons because of the inherent disparity in time scales - the spike duration time is much shorter than the inter-spike time, which is much shorter than any learning time scale. In numerical analysis, this is a classic stiff problem. Spiking neurons are also much more difficult to study analytically. One possible approach to making spiking networks more tractable is to augment mean field activity models with some information about spiking correlations. For example, such a generalized activity model could carry information about spiking rates and correlations between spikes self-consistently. Here, we will show how this can be accomplished by constructing a complete formal probabilistic description of the network and then expanding around a small parameter such as the inverse of the number of neurons in the network. The mean field theory of the system gives a rate-like description. The first order terms in the perturbation expansion keep track of covariances.

The manner in which different distributions of synaptic weights onto cortical neurons shape their spiking activity remains open. To characterize a homogeneous neuronal population, we use the master equation for generalized leaky integrate-and-fire neurons with shot-noise synapses. We develop fast semi-analytic numerical methods to solve this equation for either current or conductance synapses, with and without synaptic depression. We show that its solutions match simulations of equivalent neuronal networks better than those of the Fokker-Planck equation and we compute bounds on the network response to non-instantaneous synapses. We apply these methods to study different synaptic weight distributions in feed-forward networks. We characterize the synaptic amplitude distributions using a set of measures, called tail weight numbers, designed to quantify the preponderance of very strong synapses. Even if synaptic amplitude distributions are equated for both the total current and average synaptic weight, distributions with sparse but strong synapses produce higher responses for small inputs, leading to a larger operating range. Furthermore, despite their small number, such synapses enable the network to respond faster and with more stability in the face of external fluctuations., Author Summary Neurons communicate via action potentials. Typically, depolarizations caused by presynaptic firing are small, such that many synaptic inputs are necessary to exceed the firing threshold. This is the assumption made by standard mathematical approaches such as the Fokker-Planck formalism. However, in some cases the synaptic weight can be large. On occasion, a single input is capable of exceeding threshold. Although this phenomenon can be studied with computational simulations, these can be impractical for large scale brain simulations or suffer from the problem of insufficient knowledge of the relevant parameters. Improving upon the standard Fokker-Planck approach, we develop a hybrid approach combining semi-analytical with computational methods into an efficient technique for analyzing the effect that rare and large synaptic weights can have on neural network activity. Our method has both neurobiological as well as methodological implications. Sparse but powerful synapses provide networks with response celerity, enhanced bandwidth and stability, even when the networks are matched for average input. We introduce a measure characterizing this response. Furthermore, our method can characterize the sub-threshold membrane potential distribution and spiking statistics of very large networks of distinct but homogeneous populations of 10s to 100s of distinct neuronal cell types throughout the brain.

We examine simultaneously recorded spikes from multiple grid cells, to elucidate mechanisms underlying their activity. We demonstrate that grid cell population activity, among cells with similar spatial periods, is confined to lie close to a 2-dimensional manifold: grid cells differ only along two dimensions of their responses and are otherwise nearly identical. The relationships between cell pairs are conserved despite extensive deformations of single-neuron responses. Results from novel environments suggest such structure is not inherited from hippocampal or external sensory inputs. Across conditions, cell-cell relationships are better conserved than the responses of single cells. Finally, the system is continually subject to perturbations that were the 2-d manifold not attractive, would drive the system to inhabit a different region of state-space than observed. Together, these findings have strong implications for theories of grid cell activity, and provide compelling support for the general hypothesis that the brain computes using low-dimensional continuous attractors.

Complex systems are generally analytically intractable and difficult to simulate. We introduce a method for deriving an effective stochastic equation for a high-dimensional deterministic dynamical system for which some portion of the configuration is not precisely specified. We use a response function path integral to construct an equivalent distribution for the stochastic dynamics from the distribution of the incomplete information. We apply this method to the Kuramoto model of coupled oscillators to derive an effective stochastic equation for a single oscillator interacting with a bath of oscillators and also outline the procedure for other systems.

We analyze neocortical dynamics using field theoretic methods for non-equilibrium statistical processes. Assuming the dynamics is Markovian, we introduce a model that describes both neural fluctuations and responses to stimuli. We show that at low spiking rates, neocortical activity exhibits a dynamical phase transition which is in the universality class of directed percolation (DP). Because of the high density and large spatial extent of neural interactions, there is a "mean field" region in which the effects of fluctuations are negligible. However as the generation and decay of spiking activity becomes balanced, there is a crossover into the critical fluctuation driven DP region, consistent with measurements in neocortical slice preparations. From the perspective of theoretical neuroscience, the principal contribution of this work is the formulation of a theory of neural activity that goes beyond the mean-field approximation and incorporates the effects of fluctuations and correlations in the critical region. This theory shows that the scaling laws found in many measurements of neocortical activity, in anesthetized, normal and epileptic neocortex, are consistent with the existence of DP and related phase transitions at a critical point. It also shows how such properties lead to a model of the origins of both random and rhythmic brain activity.

A well-defined stochastic theory for neural activity, which permits the calculation of arbitrary statistical moments and equations governing them, is a potentially valuable tool for theoretical neuroscience. We produce such a theory by analyzing the dynamics of neural activity using field theoretic methods for nonequilibrium statistical processes. Assuming that neural network activity is Markovian, we construct the effective spike model, which describes both neural fluctuations and response. This analysis leads to a systematic expansion of corrections to mean field theory, which for the effective spike model is a simple version of the Wilson-Cowan equation. We argue that neural activity governed by this model exhibits a dynamical phase transition which is in the universality class of directed percolation. More general models (which may incorporate refractoriness) can exhibit other universality classes, such as dynamic isotropic percolation. Because of the extremely high connectivity in typical networks, it is expected that higher-order terms in the systematic expansion are small for experimentally accessible measurements, and thus, consistent with measurements in neocortical slice preparations, we expect mean field exponents for the transition. We provide a quantitative criterion for the relative magnitude of each term in the systematic expansion, analogous to the Ginsburg criterion. Experimental identification of dynamic universality classes in vivo is an outstanding and important question for neuroscience.

The incoherent state of the Kuramoto model of coupled oscillators exhibits marginal modes in mean field theory. We demonstrate that corrections due to finite size effects render these modes stable in the subcritical case, i.e. when the population is not synchronous. This demonstration is facilitated by the construction of a non-equilibrium statistical field theoretic formulation of a generic model of coupled oscillators. This theory is consistent with previous results. In the all-to-all case, the fluctuations in this theory are due completely to finite size corrections, which can be calculated in an expansion in 1/N, where N is the number of oscillators. We present an approach for the description of fluctuations that are due to finite system size induced correlations in the Kuramoto model of coupled oscillators. We construct a hierarchy for the moments of the density of oscillators that is analogous to the Bogoliubov-Born-Green-Kirkwood-Yvon hierarchy in the kinetic theory of plasmas and gases. To calculate the lowest order system size effect, we truncate this hierarchy at second order and solve the resulting closed equations for the two-oscillator correlation function around the incoherent state. We use this correlation function to compute the fluctuations of the order parameter, including the effect of transients, and compare this computation with numerical simulations.

Understanding the influence of network structure on neural dynamics is a fundamental step toward deciphering brain function, yet presents many challenges. We show how networks may be described in terms of the occurrences of certain patterns of edges, and how the frequency of these motifs (see Figure ​Figure1)1) impacts global dynamics. Through analysis and simulation of neuronal networks, we have found that two edge directed paths (two-chains) have the most dramatic effect on dynamics. Our analytic results are based on equations for mean population activity and correlations that we derive using path integrals and moment hierarchy expansions. These equations reveal the primary ways in which the network motifs influence dynamics. For example, the equations indicate that the propensity of a network to globally synchronize increases with the prevalence of two-chains, and we verify this result with network simulations. Finally, we present ongoing work investigating when these second-order equations break down, and how they might be corrected by higher order approximations to the network structure, such as the prevalence of three edge chains beyond that predicted by the two-chains.
