Abstract

Abstract Currently, the assessment of sperm function in a raw or processed semen sample is not able to reliably predict sperm ability to withstand freezing and thawing procedures and in vivo fertility and/or assisted reproductive biotechnologies (ART) outcome. The aim of the present study was to investigate which parameters among a battery of analyses could predict subsequent spermatozoa in vitro fertilization ability and hence blastocyst output in a goat model. Ejaculates were obtained by artificial vagina from 3 adult goats (Capra hircus) aged 2 years (A, B and C). In order to assess the predictive value of viability, computer assisted sperm analyzer (CASA) motility parameters and ATP intracellular concentration before and after thawing and of DNA integrity after thawing on subsequent embryo output after an in vitro fertility test, a logistic regression analysis was used. Individual differences in semen parameters were evident for semen viability after thawing and DNA integrity. Results of IVF test showed that spermatozoa collected from A and B lead to higher cleavage rates (0 < 0.01) and blastocysts output (p < 0.05) compared with C. Logistic regression analysis model explained a deviance of 72% (p < 0.0001), directly related with the mean percentage of rapid spermatozoa in fresh semen (p < 0.01), semen viability after thawing (p < 0.01), and with two of the three comet parameters considered, i.e tail DNA percentage and comet length (p < 0.0001). DNA integrity alone had a high predictive value on IVF outcome with frozen/thawed semen (deviance explained: 57%). The model proposed here represents one of the many possible ways to explain differences found in embryo output following IVF with different semen donors and may represent a useful tool to select the most suitable donors for semen cryopreservation.

The Drift Tube Linac (DTL) of the European Spallation Source (ESS) is designed to operate at 352.2 MHz with a duty cycle of 4 % (3 ms pulse length, 14 Hz repetition period) and will accelerate a proton beam of 62.5 mA pulse peak current from 3.62 to 90 MeV. This paper presents the approach taken in order to preserve field flatness of DTL Tanks. This strategy required a set of simulations and consequent choices about RF design of DTL cells, RF coupler tuning and compensation, cooling of the DTL cells. Abstract

The IFMIF-EVEDA RFQ is composed of 18 modules for a total length of 9.8 m and is designed to accelerate the 125 mA D⁺ beam up to 5 MeV at the frequency of 175 MHz. The RFQ is subdivided into three Super-Modules of six modules each. The Super-Modules were shipped to Rokkasho (Japan) at the beginning of 2016, pre-assembled 3 m far from the final location and tuned to reach target field flatness requirements. Just after conclusion of injector commissioning, the tuned RFQ was disassembled, moved and reassembled in the final location. After confirmation that field flatness was not affected by this movement, high power couplers were installed and tuned and all the structure was baked. Abstract

All IFMIF - EVEDA RFQ modules were completed in summer 2015. In the previous year the last three modules were RF tested at LNL at nominal power up to cw operation. At the beginning of this year all the modules were assembled in three 3.3 m long super-modules structures that were shipped to Japan. RFQ was then installed and tuned with provisional aluminum tuners and end plates to nominal frequency and field distribution. Abstract

Abstract

Mouflon (Ovis aries musimon) became extinct from mainland Europe after the Neolithic, but remnant populations from the Mediterranean islands of Corsica and Sardinia have been used for reintroductions across Europe since the 19th-century. Mouflon x sheep hybrids are larger-bodied than mouflon, potentially showing increased male reproductive success, but little is known about genomic levels of admixture, or about the adaptive significance of introgression between resident mouflon and local sheep breeds. Here we analysed Ovine medium-density SNP array genotypes of 92 mouflon from six geographic regions, along with data from 330 individuals of 16 domestic sheep breeds. We found lower levels of genetic diversity in mouflon than in domestic sheep, consistent with past bottlenecks in mouflon. Introgression signals were bidirectional and affected most mouflon and sheep populations, being strongest in one Sardinian mouflon population. Developing and using a novel approach to identify chromosomal regions with consistent introgression signals, we infer adaptive introgression from mouflon to domestic sheep related to immunity mechanisms, but not in the opposite direction. Further, we infer that Soay and Sarda sheep carry introgressed mouflon alleles involved in bitter taste perception and/or innate immunity. Our results illustrate the potential for adaptive introgression even among recently diverged populations.

Abstract

Sheep are thought to have been one of the first livestock to be domesticated in the Near East, thus playing an important role in human history. The current whole mitochondrial genome phylogeny for the genus Ovis is based on: the five main domestic haplogroups occurring among sheep (O. aries), along with molecular data from two wild European mouflons, three urials, and one argali. With the aim to shed some further light on the phylogenetic relationship within this genus, the first complete mitochondrial genome sequence of a Cypriot mouflon (O. gmelini ophion) is here reported. Phylogenetic analyses were performed using a dataset of whole Ovis mitogenomes as well as D-loop sequences. The concatenated sequence of 28 mitochondrial genes of one Cypriot mouflon, and the D-loop sequence of three Cypriot mouflons were compared to sequences obtained from samples representatives of the five domestic sheep haplogroups along with samples of the extant wild and feral sheep. The sample included also individuals from the Mediterranean islands of Sardinia and Corsica hosting remnants of the first wave of domestication that likely went then back to feral life. The divergence time between branches in the phylogenetic tree has been calculated using seven different calibration points by means of Bayesian and Maximum Likelihood inferences. Results suggest that urial (O. vignei) and argali (O. ammon) diverged from domestic sheep about 0.89 and 1.11 million years ago (MYA), respectively; and dates the earliest radiation of domestic sheep common ancestor at around 0.3 MYA. Additionally, our data suggest that the rise of the modern sheep haplogroups happened in the span of time between six and 32 thousand years ago (KYA). A close phylogenetic relationship between the Cypriot and the Anatolian mouflon carrying the X haplotype was detected. Abstract

The International Large Detector (ILD) is a concept for a detector at the International Linear Collider, ILC. The ILC will collide electrons and positrons at energies of initially 500 GeV, upgradeable to 1 TeV. The ILC has an ambitious physics program, which will extend and complement that of the Large Hadron Collider (LHC). A hallmark of physics at the ILC is precision. The clean initial state and the comparatively benign environment of a lepton collider are ideally suited to high precision measurements. To take full advantage of the physics potential of ILC places great demands on the detector performance. The design of ILD is driven by these requirements. Excellent calorimetry and tracking are combined to obtain the best possible overall event reconstruction, including the capability to reconstruct individual particles within jets for particle ow calorimetry. This requires excellent spatial resolution for all detector systems. A highly granular calorimeter system is combined with a central tracker which stresses redundancy and efficiency. In addition, efficient reconstruction of secondary vertices and excellent momentum resolution for charged particles are essential for an ILC detector. The interaction region of the ILC is designed to host two detectors, which can be moved into the beam position with a push-pull scheme. The mechanical design of ILD and the overall integration of subdetectors takes these operational condition s into account. The International Large Detector (ILD) is a concept for a detector at the International Linear Collider, ILC. The ILC will collide electrons and positrons at energies of initially 500 GeV, upgradeable to 1 TeV. The ILC has an ambitious physics program, which will extend and complement that of the Large Hadron Collider (LHC). A hallmark of physics at the ILC is precision. The clean initial state and the comparatively benign environment of a lepton collider are ideally suited to high precision measurements. To take full advantage of the physics potential of ILC places great demands on the detector performance. The design of ILD is driven by these requirements. Excellent calorimetry and tracking are combined to obtain the best possible overall event reconstruction, including the capability to reconstruct individual particles within jets for particle ow calorimetry. This requires excellent spatial resolution for all detector systems. A highly granular calorimeter system is combined with a central tracker which stresses redundancy and efficiency. In addition, efficient reconstruction of secondary vertices and excellent momentum resolution for charged particles are essential for an ILC detector. The interaction region of the ILC is designed to host two detectors, which can be moved into the beam position with a push-pull scheme. The mechanical design of ILD and the overall integration of subdetectors takes these operational conditions into account.

Abstract

We report a novel β+-thalassemia mutation found in a Vietnamese family. The molecular defect T→A lies at -72 of the β-globin gene promoter, within the conserved CCAAT box. The index case was a 5-year-old child having red blood cells indices close to normal and slightly increased level of HbA2 (3.96%). The expression of the mutated β allele was inferred by luciferase reporter assay in K562 cells. The β -72 determinant is the eighth β-thalassemic mutation identified in Vietnam and it was not previously reported in any population. The absence of homozygous or compound heterozygous states did not allow us to precisely predict either its clinical impact or its relevance in management programs. Abstract

During a screening for hemoglobinopathies, we found a carrier of the Sardinian ββ-thalassemia condition. The proband's hematology and hemoglobin (Hb) profile agreed with those of the other carriers previously identified during our diagnostic program except for the fetal Hb (HbF) composition, which consisted of both α2 Aγ2 and α2 Gγ2 instead of nearly 100% α2 Aγ2 . In order to explain the unusual γ-chain ratio, sequencing of the Gγ promoter was carried out and revealed two nucleotide substitutions in cis : C → T at position -474 and A → G at position -309 from the Cap site. The latter had previously been observed in subjects with raised HbF levels, although it has not yet been evaluated at functional level. We used the luciferase assay to determine whether the two mutations modify the transcriptional activity of the Gγ promoter. Results indicated that the observed in vivo Gγ- globin production cannot be translated into increased in vitro promoter function, suggesting that the assessed muta- tions cannot be considered as functional single nucleotide polymorphisms per se; instead, a more complex regulatory mechanism might be involved. Abstract

This study compares the developmental capacity and cryotolerance of embryos produced from oocytes of stimulated prepubertal and adult Sarda goats. Twelve prepubertal and 13 adult goats were each given 110 and 175 IU FSH, respectively, and cumulus–oocyte complexes (COCs) were collected by laparoscopic oocyte-pick-up (LOPU). After in vitro maturation, fertilisation and culture (IVMFC), blastocysts were vitrified, warmed and blastocoel re-expansion and gene expression were evaluated. Prepubertal goats produced a higher COCs number than adults (mean ± s.e.m., 89.67 ± 5.74 and 26.69 ± 3.66, respectively; P < 0.01). Lower developmental competence was demonstrated in the prepubertal oocytes as shown by a higher number of COCs discarded before IVM (21.1% and 14.7% for prepubertals and adults, respectively; P < 0.01) and IVF (23.4% v. 9.1%; P < 0.01) and by the lower cleavage (55.6% and 70.3%, respectively; P < 0.01) and blastocyst rates (24.2% and 33.9%, respectively; P < 0.05). Compared with the adult, prepubertal vitrified/warmed blastocysts showed significantly (P < 0.05) lower in vitro viability, as determined by the re-expansion rate (62.5% and 40.3%). No differences were observed in the time required for blastocoel re-expansion or in cyclin B1, E-cadherin, Na/K ATPase, HSP90β and aquaporin 3 messenger RNA quantity. These results show that in vitro-produced embryos produced from prepubertal goat oocytes have a lower developmental rate and cryotolerance compared with their adult counterparts. However, we can assume that the quality of re-expanded embryos does not differ between the two groups.

Abstract

A new variant of the fetal hemoglobin (Hb) was observed in a newborn baby subjected to phototherapy due to jaundice, by means of electrophoretic and chromatographic techniques. The variant Hb resulted unstable by the isopropanol stability test. After HBG2 gene sequencing, the G to A transversion at codon 64, position eight of the E helix, was found, which corresponds to the Asp for Gly amino acid substitution. The new variant was called Hb F-Turritana [Gγ64(E8)Gly→Asp, HBG2:c.194G>A]. Incoming aspartic acid residue, bulky and negatively charged, may be responsible for alteration of the heme pocket steric configuration and for instability. Two healthy newborns, heterozygous for two different γ-globin chain mutations, were observed during an electrophoretic screening for hemoglobinopathies in Sassari, North Sardinia (Italy). The variants were characterized by reversed phase high performance liquid chromatography (HPLC) and sequencing of amplified γ-globin genes. One of the two abnormalities was a novel Aγchain variant and the tetramer was named Hb F-Osilo [Aγ119(GH2)Gly→Ser]. Abstract

We report a new hemoglobin (Hb) variant on theHBA2gene, Hb Zara [α91(FG3)Leu→Ile (α2);HBA2: c.274C > A], which was found in a Caucasian man from Croatia. It was observed by routine cation exchange chromatography as an abnormal 21.8% fraction overlapping Hb A2, and associated with normal hematology. It was slightly unstable by the standard isopropanol precipitation test. DNA analysis revealed theCTT > ATT mutation at codon 91 on an α2 gene of a normal α-globin gene arrangement. This new variant represents the sixth described mutation at codon α91 and fourth on the α2 locus. As a result of the slight instability due to the significant role of the α91 residue in the α1β2 contact, the level of the Hb Zara variant was lower than levels observed for several stable variants codified by the α2 locus. Abstract

In this report we describe the complete sequence of the mtDNA genome of Ammotragus lervia (Barbary sheep or aoudad) as obtained by PCR and sequencing with primer walking using flanking sequences. The molecule is 16,530 base pairs in length, resulting similar to those of goat and sheep. The genome organization matches to those of other mammalian mitochondrial genomes. The phylogenetic position of the Ammotragus relative to twelve other mammalian species was assessed based on protein-coding sequences. A goat-aoudad split of 9.9-8.9 MYBP has been estimated, whereas the divergence ancestor/caprines was dated at 12.1-10.8 MYBP.

In the framework of the IFMIF/EVEDA project, the RFQ is a 9.8 m long cavity, with very challenging mechanicals specification. In the base line design, the accelerator tank is composed of 18 modules that are flanged together. The construction procedure of each module foresees the horizontal brazing of the four electrodes and then the vertical brazing of the flanges. Abstract

The Linear IFMIF Prototype Accelerator, the high intensity deuteron linac compact demonstrator of the IFMIF machine, is in an advanced installation phase at BA site (Rokkasho, Japan), within a European-Japan collaboration coordinated respectively by F4E and JAEA. The RFQ (5 Mev, 130 mA) is an Italian in-kind contribution under the responsibility of INFN. Is it a 9,8 m-long structure made of 18 modules, pre-assembled in three parts. Abstract

Mouflons are flagship species of the Mediterranean islands werethey persist. Once thought to be the remnants of a Europeanpopulation, archaeology suggests they were imported by humansto the islands of Cyprus in the Early Neolithic and laterto Corsica and Sardinia, and their status as wild animals hassince been disputed. To study the relationship between thisisland populations and other domestic and wild sheep fromthe Mediterranean we sequenced mitogenomes of 44 mouflonsfrom the islands, plus modern and ancient Sardinian domesticsheep and Anatolian mouflons. We used those in addition withpublicly available mitogenomes to reconstruct the phylogeny ofsheep and its closest wild relative, the Asiatic mouflon (Ovisgmelini) to describe how the free-ranging populations on theMediterranean islands fit. Our analysis highlights the structureof haplogroup B, where Sardinian mouflons form two geographicallyseparate clusters with gene flow between them and withdomestic sheep, and the isolation of the Corsican population,which appears as a basal lineage to all other sheep from thishaplogroup. While Corsican and Sardinian mouflon belong tobasal lineages of the domestic haplogroups associated with theearly European expansion, Cyprus mouflons are more relatedto Anatolian and Iranian mouflons belonging to the wild haplogroupX, which seems to be basal to the domestic C-E complex.These results highlight the unique genetic structure of thisisland populations while placing them in the wider context ofthe evolution of the Ovis genus.

Abstract

As the first domestic livestock species, sheep has had a fundamentalrole in human populations since the Neolithic. Theirdemographic history is, however, poorly understood. We successfullysequenced 13 ancient sheep from 2 sites in NorthernIberia to up to 8.74× and 1 modern Corsican mouflon to describethis demographic history at the Western extreme of theMediterranean. Our results support an initial maritime expansioninto Iberia and that European mouflons descended fromferalized Neolithic sheep. We also describe a secondary expansionof Eastern ancestry in sheep at the same time when humanSteppe ancestry arrived to Iberia, and when we expect woollysheep to expand through Europe. Lastly, we found a third expansionin the Mediterranean area during the Roman period,when some historical sources mentioned fine-wool sheep beingtraded. We see evidence that this expansion had a significanteffect in shaping the modern European sheep gene pool leadingto modern breeds like the popular Merino. These resultsillustrate the dynamic history of Iberian sheep populations, andhow human cultural and demographic changes left their footprintsin the sheep gene pool, marking them as a useful proxyfor describing complex human demographic events.
