51. Audio-based AI classifiers show no evidence of improved COVID-19 screening over simple symptoms checkers

Abstract: Recent work has reported that AI classifiers trained on audio recordings can accurately predict severe acute respiratory syndrome coronavirus 2 (SARSCoV2) infection status. Here, we undertake a large scale study of audio-based deep learning classifiers, as part of the UK governments pandemic response. We collect and analyse a dataset of audio recordings from 67,842 individuals with linked metadata, including reverse transcription polymerase chain reaction (PCR) test outcomes, of whom 23,514 tested positive for SARS CoV 2. Subjects were recruited via the UK governments National Health Service Test-and-Trace programme and the REal-time Assessment of Community Transmission (REACT) randomised surveillance survey. In an unadjusted analysis of our dataset AI classifiers predict SARS-CoV-2 infection status with high accuracy (Receiver Operating Characteristic Area Under the Curve (ROCAUC) 0.846 [0.838, 0.854]) consistent with the findings of previous studies. However, after matching on measured confounders, such as age, gender, and self reported symptoms, our classifiers performance is much weaker (ROC-AUC 0.619 [0.594, 0.644]). Upon quantifying the utility of audio based classifiers in practical settings, we find them to be outperformed by simple predictive scores based on user reported symptoms.

Abstract: Since early in the coronavirus disease 2019 (COVID-19) pandemic, there has been interest in using artificial intelligence methods to predict COVID-19 infection status based on vocal audio signals, for example cough recordings. However, existing studies have limitations in terms of data collection and of the assessment of the performances of the proposed predictive models. This paper rigorously assesses state-of-the-art machine learning techniques used to predict COVID-19 infection status based on vocal audio signals, using a dataset collected by the UK Health Security Agency. This dataset includes acoustic recordings and extensive study participant meta-data. We provide guidelines on testing the performance of methods to classify COVID-19 infection status based on acoustic features and we discuss how these can be extended more generally to the development and assessment of predictive methods based on public health datasets.

Abstract: Non-invasive imaging of the human spinal cord is a vital tool for understanding the mechanisms underlying its functions in both healthy and pathological conditions. However, non-invasive imaging presents a significant methodological challenge because the spinal cord is difficult to access with conventional neurophysiological approaches, due to its proximity to other organs and muscles, as well as the physiological movements caused by respiration, heartbeats, and cerebrospinal fluid (CSF) flow. Here, we discuss the present state and future directions of spinal cord imaging, with a focus on the estimation of current flow through magnetic field measurements. We discuss existing cryogenic (superconducting) and non-cryogenic (optically-pumped magnetometer-based, OPM) systems, and highlight their strengths and limitations for studying human spinal cord function. While significant challenges remain, particularly in source imaging and interference rejection, magnetic field-based neuroimaging offers a novel avenue for advancing research in various areas. These include sensorimotor processing, cortico-spinal interplay, brain and spinal cord plasticity during learning and recovery from injury, and pain perception. Additionally, this technology holds promise for diagnosing and optimizing the treatment of spinal cord disorders.

Abstract: The phenylpropanoid pathway is one of the main carbon sinks in plants, channeling phenylalanine towards thousands of products including monolignols, stilbenes, flavonoids and volatile compounds. The enzymatic steps involved in many of these pathways are well characterized, however the physical organization of these enzymes within the plant cell remains poorly understood. Proximity-dependent labeling allows untargeted determination of both direct and indirect protein interactions in vivo, and therefore stands as an attractive alternative to targeted binary assays for determining global protein-protein interaction networks. We used TurboID-based proximity labeling to study protein interaction networks of the core phenylpropanoid and anthocyanin pathways in petunia. To do so, we coupled the endoplasmic reticulum (ER) membrane anchored cytochrome P450 cinnamic acid 4-hydroxylase (C4H, CYP73A412) from Petunia inflata to TurboID and expressed it in protoplasts derived from anthocyanin-rich petunia petals. We identified multiple soluble enzymes from the late anthocyanin pathway among enriched proteins, along with other C4H isoforms, and other ER membrane anchored CYPs. Several of these interactions were subsequently confirmed by bimolecular fluorescence complementation (BiFC). Our results suggest that C4H co-localizes with enzymes from the phenylpropanoid- and downstream anthocyanin pathways, supporting the idea that C4H may serve as ER anchoring points for downstream metabolic pathways. Moreover, this study demonstrates the feasibility of using protoplasts to perform global mapping of protein network for enzymes in their native cellular environment.

Abstract: PURPOSE: This study examined the epidemiology of self-harm emergency department (ED) visits among Asian American and Pacific Islander (AAPI) youth, and associated factors. METHODS: We used California ED visit records in 2010 and 2011 to calculate incidence rates of self-harm ED visits for AAPI versus non-Hispanic White (NHW) patients aged 10-29 years. Demographic and clinical characteristics were compared for AAPI versus NHW patients presenting with self-harm. We used modified Poisson regression models to estimate the relative risk of recurrent ED self-harm visits for AAPI versus NHW patients and examined the association of insurance type and gender with recurrent self-harm among AAPIs. RESULTS: Rates of self-harm ED visits for young AAPI patients were 38 and 26 per 100,000 among females and males, respectively. Although AAPI patients presenting with self-harm were equally or less likely than NHW patients to have comorbid psychological and substance use diagnoses at their index visit, they were 25% more likely to be admitted to hospital. However, they were 40% less likely to have a recurrent ED self-harm visit. Among AAPI patients, those who used Medicaid were significantly more likely than those with other insurance to be admitted as inpatients. DISCUSSION: Young AAPI patients presenting to EDs with deliberate self-harm have different sociodemographic and clinical profiles compared to NHW patients. Our study also demonstrates significant heterogeneity in risk of recurrent self-harm by gender and insurance type among AAPI patients. This information may be useful for future intervention programs among self-harming AAPI youth.

Abstract: ContextSegregation has been linked to unequal life chances. Individuals from marginalized communities experience more crime, higher levels of poverty, poorer health, and less civic engagement. In addition, segregated metropolitan regions have been found to display inequality in access to basic services. This article builds on these findings by linking segregation to infection and deaths from COVID-19.MethodsUsing census data matched to COVID infection and death statistics at the county level, this article offers a theoretical basis for the researchers' choice of segregation measures and predictions for different racial groups. It analyzes the relationship between two dimensions of segregation-racial isolation and racial unevenness-and COVID outcomes for different racial and ethnic groups.FindingsIn counties where Black and Latino residents lived in more racially isolated neighborhoods, they were much more likely to contract COVID-19. This pattern was exacerbated in counties with a high proportion of frontline workers. In addition, racial segregation increased COVID-19 death rates for Black, Latino, and white residents.ConclusionsThese findings suggest that devastating outcomes of the coronavirus pandemic were linked to a long history of racial marginalization and entrenched discrimination produced by structural inequalities embedded in our geographies. This knowledge should be used to inform public health planning.

Abstract: We present the award-winning submission to the WikiKG90Mv2 track of OGB-LSC@NeurIPS 2022. The task is link-prediction on the large-scale knowledge graph WikiKG90Mv2, consisting of 90M+ nodes and 600M+ edges. Our solution uses a diverse ensemble of $85$ Knowledge Graph Embedding models combining five different scoring functions (TransE, TransH, RotatE, DistMult, ComplEx) and two different loss functions (log-sigmoid, sampled softmax cross-entropy). Each individual model is trained in parallel on a Graphcore Bow Pod$_{16}$ using BESS (Balanced Entity Sampling and Sharing), a new distribution framework for KGE training and inference based on balanced collective communications between workers. Our final model achieves a validation MRR of 0.2922 and a test-challenge MRR of 0.2562, winning the first place in the competition. Abstract: The effects of a finite temperature on the equilibrium structures of hydrocarbon molecules are computationally explored as a function of size and relative chemical composition in hydrogen and carbon. Using parallel tempering Monte Carlo simulations employing a reactive force field, we find that in addition to the phases already known for pure carbon, namely cages, flakes, rings and branched structures, strong effects due to temperature and the addition of little amounts of hydrogen are reported. Both entropy and the addition of moderate amounts of hydrogen favor planar structures such as nanoribbons over fullerenes. Accurate phase diagrams are proposed, highlighting the possible presence of multiple phase changes at finite size and composition. Astrophysical implications are also discussed.

Abstract: We present Sparrow, an information-seeking dialogue agent trained to be more helpful, correct, and harmless compared to prompted language model baselines. We use reinforcement learning from human feedback to train our models with two new additions to help human raters judge agent behaviour. First, to make our agent more helpful and harmless, we break down the requirements for good dialogue into natural language rules the agent should follow, and ask raters about each rule separately. We demonstrate that this breakdown enables us to collect more targeted human judgements of agent behaviour and allows for more efficient rule-conditional reward models. Second, our agent provides evidence from sources supporting factual claims when collecting preference judgements over model statements. For factual questions, evidence provided by Sparrow supports the sampled response 78% of the time. Sparrow is preferred more often than baselines while being more resilient to adversarial probing by humans, violating our rules only 8% of the time when probed. Finally, we conduct extensive analyses showing that though our model learns to follow our rules it can exhibit distributional biases.

Abstract: We report on the influence a preferential magnetic vortices motion has on the magnitude of the inverse ac Josephson effect (the appearance of dc current Shapiro steps) and the coherent operation of asymmetrical parallel arrays of YBaCuO Josephson junctions (JJ) irradiated with microwave (MW) radiation in the presence of an applied magnetic field B. The preferential direction of motion of the Josephson vortices is due to the asymmetry-induced ratchet effect and has a dramatic impact: for a particular positive dc bias current I when the flux-flow is robust multiple pronounced Shapiro-steps are observed consistent with a coherent operation of the array. This suggests an efficient emission/detection of MW in related applications. In contrast, when we reverse the direction of I, the flux-flow is reduced and the Shapiro-steps are strongly suppressed due to a highly incoherent operation that suggests an inefficient emission/detection of MW. Remarkably, by changing B slightly, the situation is reversed: Shapiro steps are now suppressed for a positive I, while well pronounced for a reverse current -I. Our results suggest that a preferential vortex-flow has a very significant impact on the coherent MW operation of superconducting devices consisting of either multiple JJs or a single long JJ asymmetrically biased. This is particular relevant in the case of flux-flow oscillators for sub-terahertz integrated-receivers, flux-driven Josephson (travelling-wave) parametric amplifiers, or on-chip superconducting MW generators which usually operate at bias currents in the Shapiro step region., Comment: 15 pages, 5 figures that includes a figure in the Supplementary material

70. Movement correction and clinical implementation of wearable magnetoencephalography (MEG)

Abstract: Magnetoencephalography (MEG) is the non-invasive measurement of magnetic fields due to neuronal current flow. The magnitude of the magnetic fields (10 fT to 1000 fT) is millions of times smaller than the Earth's static field. Consequently, highly sensitive magnetic sensors are required for MEG. Until recently, MEG systems have been based on sensors requiring cryogenic cooling. Hardware limitations from this cooling have made MEG systems large, immobile and expensive. In recent years, Optically Pumped Magnetometers (OPMs) have become viable sensors with which to measure neuromagnetic fields. These can be placed directly on the scalp. This wearability means that the participant is no longer required to remain still and the cost of the system, both financial and in terms of space, is generally lower. The freedom of movement opens up new neuroscientific and clinical applications. However, this new system is not without limitations. Movement in particular leads to artefacts unlike those previously seen in MEG; the OPM properties (gain, sensitive axis orientation, phase) are dependent on the ambient magnetic field at the sensor, which changes with position. In this thesis, we look at the impact of movement on OPM based MEG (OP-MEG) and how it can be reduced. In Chapter 2, we look into the cause of movement artefacts in OP-MEG, by mapping the spatial variation in the background magnetic field in our OP-MEG system. We show that the field varies both spatially and temporally, and that by modelling it we can reduce the interference in an OP-MEG recording. In Chapters 3 and 4, we correct for this changing field in real-time, first in simulation and then empirically. Based on the simulation results, we updated our empirical method to remove reliance on recording the position of the participant and to minimise time delays in providing the correction. Finally, in Chapters 5 and 6, we record interictal (between seizure) and ictal (seizure) OP-MEG in patients with epilepsy, while considering the impact movement has on the recordings and interictal event detection.

Abstract: Abstract Background To explore the demographic and clinical features of current depressive episode that discriminate patients diagnosed with major depressive disorder (MDD) from those with bipolar I (BP-I) and bipolar II (BP-II) disorder who were misdiagnosed as having MDD . Methods The Mini-International Neuropsychiatric Interview (MINI) assessment was performed to establish DSM-IV diagnoses of MDD, and BP-I and BP-II, previously being misdiagnosed as MDD. Demographics, depressive symptoms and psychiatric comorbidities were compared between 1463 patients with BP-I, BP-II and MDD from 8 psychiatric settings in mainland China. A multinomial logistic regression model was performed to assess clinical correlates of diagnoses. Results A total of 14.5% of the enrolled patients initially diagnosed with MDD were eventually diagnosed with BP. Broad illness characteristics including younger age, higher prevalence of recurrence, concurrent dysthymia, suicidal attempts, agitation, psychotic features and psychiatric comorbidities, as well as lower prevalence of insomnia, weight loss and somatic symptoms were featured by patients with BP-I and/or BP-I, compared to those with MDD. Comparisons between BP-I and BP-II versus MDD indicated distinct symptom profiles and comorbidity patterns with more differences being observed between BP-II and MDD, than between BP-I and MDD . Conclusion The results provide evidence of clinically distinguishing characteristics between misdiagnosed BP-I and BP- II versus MDD. The findings have implications for guiding more accurate diagnoses of bipolar disorders.

Abstract: Objective: The study objective was to determine whether adequately delivered bilateral remote ischemic preconditioning is cardioprotective in young children undergoing surgery for 2 common congenital heart defects with or without cyanosis. Methods: We performed a prospective, double-blind, randomized controlled trial at 2 centers in the United Kingdom. Children aged 3 to 36 months undergoing tetralogy of Fallot repair or ventricular septal defect closure were randomized 1:1 to receive bilateral preconditioning or sham intervention. Participants were followed up until hospital discharge or 30 days. The primary outcome was area under the curve for high-sensitivity troponin-T in the first 24 hours after surgery, analyzed by intention-to-treat. Right atrial biopsies were obtained in selected participants. Results: Between October 2016 and December 2020, 120 eligible children were randomized to receive bilateral preconditioning (n = 60) or sham intervention (n = 60). The primary outcome, area under the curve for high-sensitivity troponin-T, was higher in the preconditioning group (mean: 70.0 ± 50.9 μg/L/h, n = 56) than in controls (mean: 55.6 ± 30.1 μg/L/h, n = 58) (mean difference, 13.2 μg/L/h; 95% CI, 0.5-25.8; P = .04). Subgroup analyses did not show a differential treatment effect by oxygen saturations (pinteraction = .25), but there was evidence of a differential effect by underlying defect (pinteraction = .04). Secondary outcomes and myocardial metabolism, quantified in atrial biopsies, were not different between randomized groups. Conclusions: Bilateral remote ischemic preconditioning does not attenuate myocardial injury in children undergoing surgical repair for congenital heart defects, and there was evidence of potential harm in unstented tetralogy of Fallot. The routine use of remote ischemic preconditioning cannot be recommended for myocardial protection during pediatric cardiac surgery.

Abstract: Abstract Huntington’s disease (HD) is a dominant neurological disorder caused by an expanded HTT exon 1 CAG repeat that lengthens huntingtin’s polyglutamine tract. Lowering mutant huntingtin has been proposed for treating HD, but genetic modifiers implicate somatic CAG repeat expansion as the driver of onset. We find that branaplam and risdiplam, small molecule splice modulators that lower huntingtin by promoting HTT pseudoexon inclusion, also decrease expansion of an unstable HTT exon 1 CAG repeat in an engineered cell model. Targeted CRISPR-Cas9 editing shows this effect is not due to huntingtin lowering, pointing instead to pseudoexon inclusion in PMS1. Homozygous but not heterozygous inactivation of PMS1 also reduces CAG repeat expansion, supporting PMS1 as a genetic modifier of HD and a potential target for therapeutic intervention. Although splice modulation provides one strategy, genome-wide transcriptomics also emphasize consideration of cell-type specific effects and polymorphic variation at both target and off-target sites.

74. STELA: a community-centred approach to norm elicitation for AI alignment

Abstract: Abstract Value alignment, the process of ensuring that artificial intelligence (AI) systems are aligned with human values and goals, is a critical issue in AI research. Existing scholarship has mainly studied how to encode moral values into agents to guide their behaviour. Less attention has been given to the normative questions of whose values and norms AI systems should be aligned with, and how these choices should be made. To tackle these questions, this paper presents the STELA process (SocioTEchnical Language agent Alignment), a methodology resting on sociotechnical traditions of participatory, inclusive, and community-centred processes. For STELA, we conduct a series of deliberative discussions with four historically underrepresented groups in the United States in order to understand their diverse priorities and concerns when interacting with AI systems. The results of our research suggest that community-centred deliberation on the outputs of large language models is a valuable tool for eliciting latent normative perspectives directly from differently situated groups. In addition to having the potential to engender an inclusive process that is robust to the needs of communities, this methodology can provide rich contextual insights for AI alignment.

76. Examining High School Career and Technical Education Programs and the Postsecondary Outcomes of Career and Technical Education Students in the Round Rock Independent School District. Policy Brief

Abstract: The policy brief describes a study done by the Regional Educational Laboratory Southwest. Round Rock Independent School District (ISD) wanted to enhance its high school career and technical education (CTE) programming to prepare CTE graduates to either enter high-wage, in-demand careers in Central Texas or continue their education after high school. However, district leaders lacked key information about the district's CTE programming and their CTE graduates' postsecondary performance. In response to a request from the Round Rock ISD, this study investigated the percentage of Round Rock ISD graduates from 2012/13 through 2017/18 who completed one or more career and technical education (CTE) programs of study. The study also examined the percentage of CTE programs of study in Round Rock ISD that were aligned with high wage, in demand career pathways in Central Texas, plus the postsecondary outcomes of these graduates who completed a program of study. Key study findings include: (1) The percentage of students who graduated with one or more CTE programs of study increased more than 10 fold across the six graduating cohorts, from 4 percent for the 2012/13 cohort to 47 percent for the 2017/18 cohort; (2) Seventy-six percent of the CTE graduates completed course requirements in the 13 programs of study aligned with regional high wage, in demand career pathways in the Central Texas labor market; and (3) More than 80 percent of the CTE graduates from each cohort enrolled in two or four year colleges or were employed within one year of high school graduation. 77. Alternative Career Readiness Measures for Small and Rural Districts in Texas. Policy Brief

Abstract: Texas House Bill 3 established a college, career, and military readiness (CCMR) outcomes bonus, which provides extra funding to districts for each annual graduate demonstrating CCMR under the state accountability system. Some small district and rural district leaders expressed concern about the ability of their graduates to meet the career readiness component of the CCMR accountability standards due to a lack of career and technical education (CTE) program or course resources. Texas Education Agency (TEA) partnered with Regional Educational Laboratory (REL) Southwest to conduct a study focused on attainment of alternative career readiness options among high school graduates, and specifically graduates in small districts and rural districts. The study provides information on the implications of the CCMR accountability standards for high school graduates by examining the extent to which the cohort of 2017-18 Texas public school graduates did not meet CCMR accountability standards, particularly for graduates in small districts and rural districts. The study also provides information on the extent to which graduates demonstrated career readiness via alternative career readiness options identified by TEA. Finally, the study explores whether graduates who met the alternative career readiness options attained similar postsecondary college and career outcomes to graduates who met CCMR accountability standards to inform the Commissioner of Education as he considers alternative career readiness options. Abstract: The fundamental quandle is a powerful invariant of knots, links and spatial graphs, but it is often difficult to determine whether two quandles are isomorphic. One approach is to look at quotients of the quandle, such as the $n$-quandle defined by Joyce \cite{JO}; in particular, Hoste and Shanahan \cite{HS2} classified the knots and links with finite $n$-quandles. Mellor and Smith \cite{MS} introduced the $N$-quandle of a link as a generalization of Joyce's $n$-quandle, and proposed a classification of the links with finite $N$-quandles. We generalize the $N$-quandle to spatial graphs, and investigate which spatial graphs have finite $N$-quandles. We prove basic results about $N$-quandles for spatial graphs, and conjecture a classification of spatial graphs with finite $N$-quandles, extending the conjecture for links in \cite{MS}. Abstract: The error-backpropagation (backprop) algorithm remains the most common solution to the credit assignment problem in artificial neural networks. In neuroscience, it is unclear whether the brain could adopt a similar strategy to correctly modify its synapses. Recent models have attempted to bridge this gap while being consistent with a range of experimental observations. However, these models are either unable to effectively backpropagate error signals across multiple layers or require a multi-phase learning process, neither of which are reminiscent of learning in the brain. Here, we introduce a new model, Bursting Cortico-Cortical Networks (BurstCCN), which solves these issues by integrating known properties of cortical networks namely bursting activity, short-term plasticity (STP) and dendrite-targeting interneurons. BurstCCN relies on burst multiplexing via connection-type-specific STP to propagate backprop-like error signals within deep cortical networks. These error signals are encoded at distal dendrites and induce burst-dependent plasticity as a result of excitatory-inhibitory top-down inputs. First, we demonstrate that our model can effectively backpropagate errors through multiple layers using a single-phase learning process. Next, we show both empirically and analytically that learning in our model approximates backprop-derived gradients. Finally, we demonstrate that our model is capable of learning complex image classification tasks (MNIST and CIFAR-10). Abstract: Large language models produce human-like text that drive a growing number of applications. However, recent literature and, increasingly, real world observations, have demonstrated that these models can generate language that is toxic, biased, untruthful or otherwise harmful. Though work to evaluate language model harms is under way, translating foresight about which harms may arise into rigorous benchmarks is not straightforward. To facilitate this translation, we outline six ways of characterizing harmful text which merit explicit consideration when designing new benchmarks. We then use these characteristics as a lens to identify trends and gaps in existing benchmarks. Finally, we apply them in a case study of the Perspective API, a toxicity classifier that is widely used in harm benchmarks. Abstract: With age, metabolic perturbations accumulate to elevate our obesity burden. While age-onset obesity is mostly driven by a sedentary lifestyle and high calorie intake, genetic and epigenetic factors also play a role. Among these, members of the large histone deacetylase (HDAC) family are of particular importance as key metabolic determinants for healthy ageing, or metabolic dysfunction. Here, we aimed to interrogate the role of class 2 family member HDAC5 in controlling systemic metabolism and age-related obesity under non-obesogenic conditions. Starting at 6 months of age, we observed adult-onset obesity in chow-fed male global HDAC5-KO mice, that was accompanied by marked reductions in adrenergic-stimulated ATP-consuming futile cycles, including BAT activity and UCP1 levels, WAT-lipolysis, skeletal muscle, WAT and liver futile creatine and calcium cycles, and ultimately energy expenditure. Female mice did not differ between genotypes. The lower peripheral sympathetic nervous system (SNS) activity in mature male KO mice was linked to higher dopaminergic neuronal activity within the dorsomedial arcuate nucleus (dmARC) and elevated hypothalamic dopamine levels. Mechanistically, we reveal that hypothalamic HDAC5 acts as co-repressor of STAT5b over the control of Tyrosine hydroxylase (TH) gene transactivation, which ultimately orchestrates the activity of dmARH dopaminergic neurons and energy metabolism in male mice under non-obesogenic conditions.

Abstract: Glycogen-autophagy (‘glycophagy’) is a selective autophagy process involved in delivering glycogen to the lysosome for bulk degradation. Glycophagy protein intermediaries include STBD1 as a glycogen tagging receptor, delivering the glycogen cargo into the forming phagosome by partnering with the Atg8 homolog, GABARAPL1. Glycophagy is emerging as a key process of energy metabolism and development of reliable tools for assessment of glycophagy activity is an important priority. Here we show that antibodies raised against the N-terminus of the GABARAPL1 protein (but not the full-length protein) detected a specific endogenous GABARAPL1 immunoblot band at 18kDa. A stable GFP-GABARAPL1 cardiac cell line was used to quantify GABARAPL1 lysosomal flux via measurement of GFP puncta in response to lysosomal inhibition with bafilomycin. Endogenous glycophagy flux was quantified in primary rat ventricular myocytes by the extent of glycogen accumulation with bafilomycin combined with chloroquine treatment (no effect observed with bafilomycin or chloroquine alone). In wild-type isolated mouse hearts, bafilomycin alone and bafilomycin combined with chloroquine (but not chloroquine alone) elicited a significant increase in glycogen content signifying basal glycophagy flux. Collectively, these methodologies provide a comprehensive toolbox for tracking cardiac glycophagy activity to advance research into the role of glycophagy in health and disease.

Abstract: Posttraumatic stress disorder (PTSD) is a heritable (h2 = 24-71%) psychiatric illness. Copy number variation (CNV) is a form of rare genetic variation that has been implicated in the etiology of psychiatric disorders, but no large-scale investigation of CNV in PTSD has been performed. We present an association study of CNV burden and PTSD symptoms in a sample of 114,383 participants (13,036 cases and 101,347 controls) of European ancestry. CNVs were called using two calling algorithms and intersected to a consensus set. Quality control was performed to remove strong outlier samples. CNVs were examined for association with PTSD within each cohort using linear or logistic regression analysis adjusted for population structure and CNV quality metrics, then inverse variance weighted meta-analyzed across cohorts. We examined the genome-wide total span of CNVs, enrichment of CNVs within specified gene-sets, and CNVs overlapping individual genes and implicated neurodevelopmental regions. The total distance covered by deletions crossing over known neurodevelopmental CNV regions was significant (beta = 0.029, SE = 0.005, P = 6.3 × 10-8). The genome-wide neurodevelopmental CNV burden identified explains 0.034% of the variation in PTSD symptoms. The 15q11.2 BP1-BP2 microdeletion region was significantly associated with PTSD (beta = 0.0206, SE = 0.0056, P = 0.0002). 89. Examining the Relation between Exercise and Word Learning in Preschool-Age Children

Abstract: Purpose: There is evidence suggesting that aerobic exercise immediately after vocabulary training can improve word recall in school-age children. This work examined whether the previously identified word-learning benefits associated with exercise can be extended to preschoolers. Additionally, we evaluated whether the effects of physical activity on vocabulary learning may be influenced by existing language skills that the child possesses. Method: Children ages 3-6 years completed the study (N = 42). Data were collected via a virtual testing session in which participants completed a word-learning task that included two experimental conditions ("exercise" and "resting"). In the resting measure, children were taught names of novel objects and then sat down and colored for 3 min before being tested on their ability to identify the trained words. The exercise condition was identical, except that participants engaged in 3 min of guided aerobic exercise before testing. Additionally, at the end of the visit, participants completed the Quick Interactive Language Screener (QUILS), which measured general language skills. Results: Accuracy of word recognition was significantly higher after the exercise condition compared to the resting condition. Furthermore, this pattern of results was not related to children's existing language skills, as measured by the QUILS. Conclusions: This study is one of the first to closely examine the relation between physical activity and word-learning abilities in children as young as 3-6 years of age. Results align with previous findings stating that aerobic exercise can boost vocabulary learning and suggest that this is the case regardless of existing language skills.

Abstract: Topic: To review clinical evidence on systemic factors that might be relevant to update diabetic retinal disease (DRD) staging systems, including prediction of DRD onset, progression, and response to treatment. Clinical relevance: Systemic factors may improve new staging systems for DRD to better assess risk of disease worsening and predict response to therapy. Methods: The Systemic Health Working Group of the Mary Tyler Moore Vision Initiative reviewed systemic factors individually and in multivariate models for prediction of DRD onset or progression (i.e., prognosis) or response to treatments (prediction). Results: There was consistent evidence for associations of longer diabetes duration, higher glycosylated hemoglobin (HbA1c), and male sex with DRD onset and progression. There is strong trial evidence for the effect of reducing HbA1c and reducing DRD progression. There is strong evidence that higher blood pressure (BP) is a risk factor for DRD incidence and for progression. Pregnancy has been consistently reported to be associated with worsening of DRD but recent studies reflecting modern care standards are lacking. In studies examining multivariate prognostic models of DRD onset, HbA1c and diabetes duration were consistently retained as significant predictors of DRD onset. There was evidence of associations of BP and sex with DRD onset. In multivariate prognostic models examining DRD progression, retinal measures were consistently found to be a significant predictor of DRD with little evidence of any useful marginal increment in prognostic information with the inclusion of systemic risk factor data apart from retinal image data in multivariate models. For predicting the impact of treatment, although there are small studies that quantify prognostic information based on imaging data alone or systemic factors alone, there are currently no large studies that quantify marginal prognostic information within a multivariate model, including both imaging and systemic factors. Conclusion: With standard imaging techniques and ways of processing images rapidly evolving, an international network of centers is needed to routinely capture systemic health factors simultaneously to retinal images so that gains in prediction increment may be precisely quantified to determine the usefulness of various health factors in the prognosis of DRD and prediction of response to treatment. Abstract: Background Early life stress (ELS) is an important risk factor in the aetiology of depression. Developmental glucocorticoid exposure impacts multiple brain regions with the hippocampus being particularly vulnerable. Hippocampal mediated behaviours are dependent upon the ability of neurones to undergo long-term potentiation (LTP), an N-methyl-D-aspartate receptor (NMDAR) mediated process. In this study we investigated the effect of ELS upon hippocampal NMDAR function. Methods Hooded Long-Evans rat pups (n=82) were either undisturbed or maternally separated for 180 minutes per day (MS180) between post-natal day (PND) 1 and PND14. Model validation consisted of sucrose preference (n=18) and novelty supressed feeding (NSFT, n=34) tests alongside assessment of corticosterone (CORT) and paraventricular nucleus (PVN) cFos reactivity to stress and hippocampal neurogenesis (all n=18). AMPA/NMDA ratios (n=19), miniEPSC currents (n=19) and LTP (n=15) were assessed in whole-cell patch clamp experiments in CA1 pyramidal neurones. Results MS180 animals showed increased feeding latency in the NSFT alongside increased overall CORT in the restraint stress experiment and increased PVN cFos expression in males but no changes in neurogenesis or sucrose preference. MS180 was associated with a lower AMPA/NMDA ratio with no change in miniEPSC amplitude or area. There was no difference in short- or long-term potentiation between MS180 and control animals nor were there any changes during the induction protocol. Conclusions The MS180 model showed a behavioural phenotype consistent with previous work. MS180 animals showed increased NMDAR function with preliminary evidence suggesting that this was not concurrent with an increase in LTP.

Abstract: Background Gluteal tendinopathy (GT) is a degenerative tendon condition characterised by pain over the greater trochanter of the hip. A randomised controlled trial (RCT) in Australia found that 14 sessions of EDucation on load management plus eXercise (EDX) delivered over 8 weeks resulted in greater improvements in global rating of change and pain outcomes at 8 and 52 weeks, compared with corticosteroid injection or ‘wait and see’. Typically, 5-6 physiotherapy sessions are provided in public and private physiotherapy settings in Ireland, therefore, the aim of this study is to examine the feasibility of conducting a future definitive RCT to investigate effectiveness of 6 sessions of the EDX programme compared to usual care. Methods We will randomly allocate 64 participants with GT to physiotherapist-administered EDX or usual care. The EDX intervention (EDX-Ireland) will be delivered in 6 sessions over 8 weeks. To determine feasibility of an RCT, we will assess recruitment and retention and outcome measure completion. The health status outcomes to be assessed at baseline, 8 weeks and 3 months include: Global Rating of Change, pain severity, the Victorian Institute of Sport Assessment-Gluteal Questionnaire (VISA-G), the Patient-Specific Functional Scale, the Pain Catastrophizing Scale, Patient Health Questionnaire (PHQ), Pain Self-Efficacy Questionnaire, the EQ-5D-5L, the Central Sensitisation Inventory and hip abductor muscle strength. We will explore acceptability of the EDX-Ireland intervention from the perspective of patients and treatment providers, and the perspective of referrers to the trial. A Study Within A Trial will be also applied to compare recording of exercise adherence using app-based technology to paper diaries. Discussion There is a need to establish effective treatments for GT that potentially can be implemented into existing health systems. The findings of this feasibility trial will inform development of a future definitive RCT. Registration The trial is registered prospectively on ClinicalTrials.gov (NCT05516563, 27/10/2022).

Abstract: The aim of this study is to utilize Semiotic analysis to analyze the representation of Gulf men in advertising a Kuwaiti brand. Based on two YouTube commercials as a case study, this article asks, How is Arab masculinity represented in advertising? The findings indicate the existence of masculinity in flux, suggesting a subtle form of socialization and identity negotiation between foreign and local ideas, especially regarding gender relations and the representation of Arab masculinity. The analysis highlights the contrast between traditional and modern forms of masculinity, as represented by local and foreign football teams in the advertisement, suggesting that modern (or Western) forms of masculinity can be used to counteract oppressive local practices. The study argues that masculinity is not a fixed or objective concept and is instead shaped by the social and cultural context in which it exists. Here, the media's representation of masculinity is particularly influential in shaping how young people understand and internalize this idea.

Abstract: Introduction: United States emergency departments (ED) visit rates for nonfatal self-harm increased by 42% from 2001 to 2016. Previous suicide mortality research has provided conflicting evidence on the use of suicide-related Internet searches as a surveillance tool for self-harm and suicidal ideation. However, few have used rigorous approaches to account for autocorrelation at the aggregate level, and none have focused on Internet searches related to suicide prevention. Methods and results: Over a 9-year study period (2007–2015), suicidality-related search data were extracted using the Google Health Application Programming Interface (API) for Arizona and California – states, chosen for their differing age distributions and rigorous ED injury coding policies. We examined several combined suicide prevention-related search queries. Using autoregressive integration moving average (ARIMA) models and a Box-Jenkins approach, we assessed whether increased prevention-related Internet searches related to suicidality are predictive of lower subsequent ED visits related to suicidal ideation with or without self-harm injury. In both states, greater prevention-related queries were associated with lower ED visits approximately four to six weeks later. Conclusions: Our results indicate that Internet-based search volumes related to suicide prevention may have the potential to monitor suicidality and online suicide prevention resources offer meaningful opportunities for mental health support.

Abstract: Introduction: Patients with sarcoidosis have a lower survival rate than the general population, in part due to cardiovascular disease, infections and neoplasms. Our objective was to evaluate the impact of haematological neoplasms (HN) and lymphomas on sarcoidosis patient mortality in a nation-wide analysis conducted in Spain, a country with a population of 47 million. Methods: Retrospective and observational comparison of the HN related deaths in sarcoidosis patients and the general Spanish population reported in the Spanish Hospital Discharge Database. To determine the impact of sarcoidosis on the risk of dying from each HN lineage, a binary logistic regression considering age, female sex, tobacco and alcohol consumption, was performed. 97. Theoretical study of laser intensity noise effect on CW-STED microscopy

Abstract: Spatial resolution of stimulated emission depletion (STED) microscopy varies with sample labeling techniques and microscope components, e.g., lasers, lenses, and photo-detectors. Fluctuations in the intensity of the depletion laser decrease achievable resolution in STED microscopy; the stronger the fluctuations, the higher the average intensity needed to achieve a given resolution. This phenomenon is encountered in every STED measurement. However, a theoretical framework that evaluates the effect of intensity fluctuations on spatial resolution is lacking. This article presents an analytical formulation based on a stochastic model that characterizes the impact of the laser fluctuations and correlation time on the depletion efficiency in the continuous wave (CW) STED microscopy. We compared analytical results with simulations using a wide range of intensity noise conditions and found a high degree of agreement. The stochastic model used considers a colored noise distribution for the laser intensity fluctuations. Simple analytical expressions were obtained in the limit of small and large fluctuations correlation time. These expressions fitted very well the available experimental data.
