Abstract

People with newly-discovered mutations in the gene for cardiac troponin T may have few symptoms of hypertrophic cardiomyopathy but nevertheless have a high risk of sudden death. Hypertrophic cardiomyopathy is a type of heart disease. Researchers used DNA analysis to detect mutations in the genes for the heart muscle proteins alpha tropomyosin and cardiac troponin T in 27 families and 126 people with a family history of hypertrophic cardiomyopathy. Five families and six people had a mutation in the gene for cardiac troponin T. Only one person had a mutation in the gene for alpha tropomyosin. Eight mutations in the cardiac troponin T gene could be linked to hypertrophic cardiomyopathy and five have never been described before. Death rates were higher in young patients with a mutation in cardiac troponin T, and more died suddenly, compared to patients with the more common mutation in the heavy chain of beta cardiac myosin. However, many of the patients with the cardiac troponin T mutation had no clinical signs of heart disease.

77. Cetirizine a histamine H1 receptor antagonist improves viral myocarditis

Abstract

Abstract Background We showed that mast cells played a critical role in the progression of heart failure induced by pressure overload and viral myocarditis in mice. In this study, we investigated the effect of cetirizine, a selective H1 receptor antagonist, on experimental viral myocarditis induced by encephalomyocarditis (EMC) virus. Methods Four-week-old inbred male DBA/2 mice were inoculated intraperitoneally with 10 plaque-forming units (pfu) of the EMC virus. Cetirizine was administered orally at a dose of 1 or 10 mg/kg per day for the survival study, and 1 mg/kg for the histologic and gene expression studies, beginning on the day of viral inoculation. Results Cetirizine improved survival dose dependently. Heart weight to body weight ratio was significantly decreased in mice treated with cetirizine. The area of myocardial necrosis was significantly smaller in the hearts of mice treated with cetirizine compared with controls. Gene expressions of tumor necrosis factor, interleukin 6, and metalloproteinase 2 were significantly suppressed in the hearts of mice treated with cetirizine. Conclusion These results suggest that cetirizine exerts its beneficial effects on viral myocarditis by suppressing expression of pro-inflammatory cytokines, genes related to cardiac remodeling in the hearts of mice.

Abstract

原子力機構は、新型転換炉ふげん発電所の新型転換炉原型炉施設の廃止措置計画を平成18年11月7日に認可申請し、平成20年2月12日に認可を受けた。これに伴い、新型転換炉ふげん発電所を原子炉廃止措置研究開発センター(以下「ふげん」という。)に改組し、施設の解体撤去作業に着手するとともに、自らの廃止措置に関する技術の開発及びこれに必要な研究(以下「廃止措置技術開発」という。)を実施している。この廃止措置技術開発を計画・実施するにあたり、「ふげん」を国内外に開かれた技術開発の場及び福井県が目指すエネルギー研究開発拠点化計画における研究開発拠点として十分に活用するとともに、当該技術開発で得られる成果を有効に活用することを目的として、原子力機構内外の有識者で構成される「ふげん廃止措置技術専門委員会」を設置している。本稿は、平成22年9月14日に開催した第22回ふげん廃止措置技術専門委員会において発表された資料「廃止措置の状況」,「重水系機器を用いた放射性腐食性生物(CP)除染試験」,「管理データ評価システムの「ふげん」への適用」,「クリアランス制度運用に向けた準備状況」についてまとめたものである。, In planning and carrying out our decommissioning technical development, "Technical special committee on Fugen decommissioning", which consists of the members well-informed, is established, aiming to make good use of Fugen as a place for technological development which is opened inside and outside the country, as the central point in the energy research and development base making project of Fukui prefecture, and to utilize the outcome in our decommissioning to the technical development effectively. Abstract

Purpose: NT-proBNP has emerged as a powerful diagnostic and prognostic biomarker in heart disease. Studies showed that NT-proBNP is a sensitive biomarker for identifying patients with heart failure caused by hepatitis C virus (HCV) related myocarditis. The purpose of this study was to evaluate the correlation between the serum concentration of NT-proBNP and hepatitis virus infection/liver disease. Methods: 223 serum samples from blood donors (aged 19~50 years old) were collected as a control group, and 644 samples were obtained from patients infected by hepatitis viruses including 493 HBV: 364 chronic hepatitis (CH), 86 hepatocellular carcinoma (HCC) and 43 liver cirrhosis (LC) and 151 HCV (85 CH, 14 HCC, 52 LC). All samples were assayed with an Elecsys immunoassay analyzer for NT-proBNP concentration. Results: The mean concentration of NT-proBNP in the control group was 21.77 pg/ml and showed no significant variation with either age or gender. Both the mean value and the rate of abnormality of NT-proBNP were significantly higher for the HBV- and HCV-infected groups in comparison with the control group. The mean NT-proBNP value (380.24 pg/ml) and abnormality rate (38.41%) in the HCV group were higher than that of the HBV group. For samples from patients with HBV/HCV-related hepatic disease/pathology, the mean NT-proBNP value (517.19 pg/ml/597.18 pg/ml) were the highest in the liver cirrhosis group. Conclusions: Hepatic pathologic lesions, particularly cirrhosis, may contribute to the elevation of NT-proBNP in subjects with HBV/HCV infection.
