Wastewater reuse is a proven strategy to mitigate water stress in drought-prone regions. However, this practice is still limited due to high implementation costs, regulatory hurdles, and limited public acceptance. In regions with low reclaim rates, a thorough evaluation of the potential for reuse is needed to support decision-making, focusing on opportunities that address both low-hanging fruit and high-leverage projects. This paper introduces a streamlined, data-centric methodology for assessing wastewater reuse potential, adaptable to various regional contexts. The methodology involves comprehensive data collection and processing to evaluate wastewater treatment plant (WWTP) capabilities and identify potential users, allowing the prioritisation of case studies based on demand alignment. Different treatment and distribution systems are analysed to match WWTP capabilities with user needs, considering volume, quality, and infrastructure requirements. Cost analysis incorporates capital expenditure (CAPEX), operational expenditure (OPEX) and unit costs using novel cost functions for treatment and distribution. Risk analysis adheres to WHO methodology to ensure safety and sustainability. A case study in the Lisbon and Oeste areas in Portugal validates this approach, revealing key insights into the potential and economic viability of water reuse. This article examines the multifaceted landscape of Rare Earth Elements (REEs), crucial components in traditional industries like glass and metallurgy, as well as pivotal in the growing green and digital economies. The transition toward a greener economy heavily relies on REEs for batteries, wind turbines, and solar panels. The global REE supply chain, dominated by China with nearly 95% of the world's reserves, introduces complexities and geopolitical considerations, and prompting nations like the United States and countries in the European Union to seek alternatives and reduce dependence on China's dominant control. The United States, once a key player in rare earth mining, now heavily relies on Chinese imports, posing national security concerns. This article examines the challenges and opportunities in the global rare earth elements market, addressing environmental controversies, geopolitical tensions, and the race for a secure supply chain. Aspergilli comprise a diversity of species that have been extensively studied due to their catabolic diversity, biotechnological and ecological value, and pathogenicity. An impressive level of structural and functional conservation has been shown for aspergilli, regardless of many (yet) cryptic genomic elements. We have hypothesized the existence of conserved genes responsive to stress in aspergilli. To test the hypothesis of such conserved stress regulators in aspergilli, a straightforward computational strategy integrating well-established bioinformatic tools was used as the starting point. Specifically, five transcriptome-based datasets on exposure to organic compounds were used, covering three distinct Aspergillus species. Among the identified up-regulated genes, only one gene showed the same response in all conditions, AN9181. This gene encodes a protein containing a phenylcoumaran benzylic ether reductase-like domain and a Nitrogen metabolite repressor regulator domain (NmrA). Deletion of this gene caused significant phenotypic alterations compared to that of the parental strain across diverse conditions. Specifically, the deletion of AN9181 raised the mutant's metabolic activity in different nitrogen sources. The acquired data supports that AN9181 acts by repressing (slowing down) A. nidulans growth when exposed to aromatic compounds in a concentration dependent manner. The same phenotype was observed for amphotericin B. Finally, AN9181 underwent differential upregulation under oxidative stress conditions. Collectively, the data suggest that AN9181, herein assigned as NmrB (Nitrogen Metabolite Repression Regulator B), builds up the genetic machinery of perception of oxidative stress by negatively regulating growth under such conditions.
