The establishment of reliable prognostic biomarkers for canine insulinoma are warranted to facilitate optimal patient management. Using univariate and multivariate analyses, the present study evaluated the prognostic power of several clinical and histopathological criteria, as well as the Ki67 index, in 26 dogs with insulinoma. On univariate analysis, stromal fibrosis within these tumours was found to be significantly predictive for survival, while tumour size, the Ki67 index, and TNM (Tumour, Node, Metastasis) stage were significant in the prognosis of both disease-free interval and survival time. On multivariate analysis, tumour size retained predictive power for disease-free intervals, while the Ki67 index proved prognostically significant for both the disease-free interval and overall survival time. This study demonstrates that, in addition to known factors such as tumour size and stage, Ki67 can act as a biomarker of insulinoma that can be used to predict clinical outcome.

An 11-yr-old female hamadryas baboon (Papio hamadryas) that died with a history of diarrhea and anorexia was submitted for necropsy. Major pathologic changes were restricted to the gastrointestinal tract. The small intestinal contents were watery and sanguinous, with a deepening of the red color in the large intestines. The intestinal mucosa was hyperemic. Microscopically, lesions consisted of surface epithelial cell necrosis in association with numerous rod-shaped bacteria and high numbers of Trichuris cynocephalus nematodes. Culturing of the small intestine yielded Clostridium perfringens. No other pathogenic bacteria were cultured using routine bacteriologic techniques. Polymerase chain reaction (PCR) analysis classified the Clostridium perfringens as type A cpb2-positive. Immunohistochemical examination with anti-β2-toxin antibodies revealed β2-toxin in close approximation with the intestinal lesions.

To characterize biologic behavior, clinical outcome, and effect of histologic grade on prognosis for dogs with appendicular chondrosarcoma treated by amputation alone.Case series.Dogs (n=25) with appendicular chondrosarcoma.Medical records were searched to identify dogs with appendicular chondrosarcoma treated by limb amputation alone. Information recorded included signalment, anatomic location, radiographic appearance, and development of metastasis. Histopathologic diagnosis was confirmed and graded (1, 2, or 3). Survival curves were generated by the Kaplan-Meier method and the association between covariates (gender, age, weight, and tumor grade) and survival were evaluated using the univariate proportional hazards model.Histopathology slides were available for 25 dogs. Rates of pulmonary metastasis were as follows: grade 1-0%, grade 2-31%, and grade 3-50%. Overall median survival time (MST) was 979 days. Age, weight, and sex were not significantly associated with survival (P=.16; .33; and .31, respectively). Survival was significantly associated with tumor grade (P=.008), with dogs with tumor grade of 1, 2, and 3 having MSTs of 6, 2.7, and 0.9 years, respectively.Canine appendicular chondrosarcoma can be treated effectively with amputation alone. Low to intermediate grade chondrosarcoma has a good prognosis, whereas high-grade tumors appear to behave aggressively.The overall prognosis for appendicular chondrosarcoma is better than that of appendicular osteosarcoma treated by amputation alone or in combination with chemotherapy.

A 6-year-old, castrated male ferret (Mustela putorius furo) was presented with progressive neurological signs consisting of a right-sided head tilt and ataxia. Neurological examination revealed hemiparesis and absence of proprioception on the right side, consistent with central vestibular syndrome. Measurement of blood glucose excluded hypoglycemia due to insulinoma. Contrast-enhanced computed tomography revealed the presence of an intracranial mass, consistent with either granuloma or neoplasia. Palliative treatment with prednisolone yielded no improvement. At postmortem examination, a final diagnosis of a choroid plexus papilloma originating from the fourth ventricle was made. This is the first report of such a tumor in a ferret.

Objective— To investigate the histologic characteristics of feline osteosarcoma (OS) and compare the histologic data with phenotypically comparable canine OS. The effects of histologic and clinical variables on survival statistics were evaluated. Study Design— Retrospective study. Animals— Cats (n=62) and dogs (22). Methods— Medical records of 62 cats with OS were reviewed for clinically relevant data. Clinical outcome was obtained by telephone interview. Histologic characteristics of OS were classified using a standardized grading system. Histologic characteristics in 22 feline skeletal OS were compared with 22 canine skeletal OS of identical location and subtype. Prognostic variables for clinical outcome were determined using multivariate analysis. Results— Feline OS was characterized by moderate to abundant cellular pleomorphism, low mitotic index, small to moderate amounts of matrix, high cellularity, and a moderate amount of necrosis. There was no significant difference between histologic variables in feline and canine OS. Histologic grade, surgery, and mitotic index significantly influenced clinical outcome as determined by multivariate analysis. Tumor invasion into vessels was not identified as a significant prognosticator. Conclusion— Feline and canine skeletal OS have similar histologic but different prognostic characteristics. Clinical Relevance— Prognosis for cats with OS is related to histologic grade and mitotic index of the tumor.

Post-mortem investigation of a harbor porpoise (Phocoena phocoena) found dead on the beach of the island of Vlieland, The Netherlands, revealed severe granulomatous changes in the right lung lobe. Ziehl Neelsen staining demonstrated relatively large acid-fast rods. Mycobacterial culture yielded a fast-growing mycobacterium, which was identified by molecular biological methods as Mycobacterium mageritense. Autolysis prevented histopathology. It was tentatively concluded that the granulomatous changes were the cause of porpoise's death and that M. mageritense was the causative agent. This is the first report of the isolation and molecular identification of this mycobacterium in a nonhuman animal species and the first association with the marine environment.

The jejunal mucosa of pigs fed diets containing Phaseolus vulgaris beans was characterized grossly as mucosal atrophy and microscopically as atrophy and blunting of the villus in association with elongation of crypts with cells with increased mitotic activity. These morphologic findings were most severe in the proximal and middle parts of the jejunum. Compared to controls, goblet cells were significantly decreased in the villus but markedly increased in the crypt region. The activity of aminopeptidase and sucrase-isomaltase in the test animals was also significantly lower than in the controls. The findings in this study suggested that feeding Phaseolus vulgaris beans reduced the digestive and absorptive capacity of the mucosa, resulting in weight loss and diarrhea of affected pigs.

Zusammenfassung Der Einflus von Rationen mit Pisum sativum auf Wachstum und Organgewichte bei Ferkeln, Ratten und Kuken An Ferkel, Ratten und Kuken wurden Rationen, die 30% Pisum sativum enthielten und durch Aminosauren und Energie bedarfsgerecht erganzt waren, bei zwei Futterungsintensitaten gefuttert. Gemessen wurde bei allen drei Tierspezies die Futteraufnahme, die Gewichtszunahme, die Futterverwertung und das Gewicht verschiedener Organe. Dabei sollte gepruft werden, ob die bei Ratten und Kuken ermittelten Ergebnisse auf das Ferkel ubertragbar sind. Wahrend bei Kuken und Ratten keine Reduzierung der Gewichtszunahme beobachtet wurde, zeigten Ferkel niedrigere Gewichtszunahmen. Bei Ferkeln konnte ein Einflus auf das Thymusgewicht, bei Ratten dagegen auf Pankreas- und Nierengewichte und bei Kuken auf Pankreas-, Nieren- und Dunndarmgewichte beobachtet werden. Daraus last sich schliesen, das Ergebnisse, die an Ratten und Kuken erhoben wurden, nicht fur Rationen mit Pisum auf Ferkel ubertragbar sind.

In The Netherlands between January 2002 and December 2004, numerous psittaciformes died showing severe splenomegaly and hepatomegaly with multifocal acute necrosis. At the start of the outbreaks mostly parakeets were affected, but later larger parrots were also involved. Seventy-eight birds showed the same features and six were examined completely, including a virological examination. Tests for polyomavirus, Pacheco's disease (herpesvirus) and circovirus psittacine beak and feather disease (PBFD) viruses and Chlamydophila psittaci were carried out. All results were negative, except for two cases of circovirus infection. Many concurrent bacterial and parasitic infections were seen. Immunohistochemistry revealed reovirus antigen in intralesional mononuclear cells, and reovirus-like particles could be observed by negative contrast electron microscopy. A reovirus was grown and the isolates reacted with polyclonal reovirus antiserum but did not react with monoclonal antibodies against chicken reovirus. The virus was therefore considered a psittacine reovirus. Because reoviruses were seen consistently, they seemed to be the most probable cause of the outbreaks. Climate, the introduction of new birds and the transportation of birds might be other factors involved in the disease seen in The Netherlands. No regional influence could be seen; therefore, we suggested that the virus might be widespread and carriers could be a source of re-introduction.

A 6-mo-old female scarlet macaw (Ara macao) was presented after a 2-mo period of anorexia and weakness. The bird was reluctant to fly 1 wk before referral due to a painful left wing. Physical examination revealed a firm swelling around the left shoulder. On radiographs, the diaphysis and proximal metaphysis of the left scapula were radiolucent. Computer tomography revealed an osteolytic process, suggestive of a bone tumor, affecting the left scapula. Cytology of a fine needle aspiration biopsy of the mass showed erythrocytes, a proliferation of spindle-shaped mesenchymal cells, and multinucleated giant cells (osteoclasts) suggestive of a giant cell tumor. The left wing, including the scapula, was amputated. The bird showed a fast recovery but died 1 hr later. Findings during the pathological examination were compatible with shock due to blood loss. The shoulder process was characterized as a giant cell tumor. To our knowledge, this is the first complete report of a giant cell tumor of the bone in a bird.

Generalized cowpox infection in the Patagonian cavy may represent a threat to the health of immunocompromised persons. We report the first case of cowpoxvirus infection in the Patagonian cavy in an educational animal park. The mara developed extensive pox lesions, shedding large amounts of viral particles. The ending of vaccination programmes against smallpox in the late 1970's may lead to an increase in susceptibility of humans to zoonotic poxviruses.

Objectives The purpose of this study was to investigate scaling of the duration of late diastolic left ventricular (LV) filling in relation to AV conduction time (delay) (PR interval on the ECG) in mammals. Background From mouse to whale, AV delay increases 10-fold, whereas body mass increases one million-fold. The apparent "mismatch" results from scaling of AV delay versus body and heart mass. Methods We measured (1) mitral orifice diameter in 138 postmortem hearts of 48 mammalian species weighing between 17 g and 250 kg and (2) transmitral diastolic flow using magnetic resonance imaging (MRI) recordings of 10 healthy human individuals. (3) We visually inspected early and late diastolic LV filling. (4) We developed two physical models to explain scaling of late diastolic LV filling time. Results (1) Diameter of the mitral orifice proportionally relates to heart length (third root of heart mass). (2) Atrial contraction starts at a fixed instant (± 80%) of the (normalized) cardiac cycle and contributes 31% ± 5% to LV filling. (3) MRI shows that during diastole, the left atrium (LA) and LV form a single space. (4) The physical models relate the duration of late diastolic LV filling directly to heart length, the third root of heart mass. Conclusions (1) Late diastolic (LV) filling time scales with heart length (third root of heart mass). (2) No "mismatch" exists between AV delay and heart size. (3) Knowledge of the actual starting time of atrial contraction may contribute to better treatment of patients with heart failure. (4) The findings suggest that in evolution of mammalian species, hemodynamics commands electrical behavior of the heart.

A 10-year-old German shepherd dog was presented with unilateral uveitis and hyphaema. Treatment was unsuccessful and the eye was enucleated. Intraocular osteosarcoma was diagnosed by histological examination.

Hyperadrenocorticism in ferrets is usually associated with unaltered plasma concentrations of cortisol and adrenocorticotropic hormone (ACTH), although the urinary corticoid/creatinine ratio (UCCR) is commonly elevated. In this study the urinary glucocorticoid excretion was investigated in healthy ferrets and in ferrets with hyperadrenocorticism under different circumstances. In healthy ferrets and in one ferret with hyperadrenocorticism, approximately 10% of plasma cortisol and its metabolites was excreted in the urine. High-performance liquid chromatography (HPLC) revealed one third of the urinary corticoids to be unconjugated cortisol; the other peaks mainly represented cortisol conjugates and metabolites. In 21 healthy sexually intact ferrets, the UCCR started to increase by the end of March and declined to initial values halfway the breeding season (June). In healthy neutered ferrets there was no significant seasonal influence on the UCCR. In two neutered ferrets with hyperadrenocorticism the UCCR was increased, primarily during the breeding season. In 27 of 31 privately owned ferrets with hyperadrenocorticism, the UCCR was higher than the upper limit of the reference range (2.1×10 −6 ). In 12 of 14 healthy neutered ferrets dexamethasone administration decreased the UCCR by more than 50%, whereas in only 1 of the 28 hyperadrenocorticoid ferrets did the UCCR decrease by more than 50%. We conclude that the UCCR in ferrets primarily reflects cortisol excretion. In healthy sexually intact ferrets and in ferrets with hyperadrenocorticism the UCCR increases during the breeding season. The increased UCCR in hyperadrenocorticoid ferrets is resistant to suppression by dexamethasone, indicating ACTH-independent cortisol production.

Een mannelijke Aziatische olifant leed sinds enkele jaren aan gewrichtsproblemen. Tevens had het 50 jaar oude dier regelmatig problemen met de nagels, zoals het loslaten daarvan in combinatie met een necrotiserende nagelbedontsteking. Na verloop van tijd verslechterde het dier zienderogen met als verschijnselen: sloomheid, toenemende gewrichtsontsteking, zucht onderaan de buik (ventraal oedeem) en overmatig drinken en plassen.

This paper describes the signs, clinical pathology, and postmortem findings in 14 young African grey parrots (Psittacus erithacus erithacus) that were naturally infected with psittacine beak and feather disease (PBFD) virus (psittacine circovirus). All but two of the parrots had severe leukopenia at clinical presentation. Two other parrots also had severe anemia. All birds died within 3 wk after presentation. Postmortem examination documented liver necrosis in 11 of 14 birds and secondary bacterial or fungal infections in 9 of 14 birds. Tests for Chlamydia psittaci, polyomavirus, and Salmonella sp. were negative. PBFD viral infection could be demonstrated in all birds by polymerase chain reaction. Supporting evidence of PBFD viral infection was gathered by histologic examination of the bursa of Fabricius, electron microscopy, and DNA in situ hybridization. Electron microscopic examination of both the bursa of Fabricius and liver revealed virus particles resembling circovirus. DNA in situ hybridization of six liver tissue samples confirmed the presence of PBFD virus and excluded the presence of avian polyomavirus. Our findings suggest that a specific presentation of peracute PBFD viral infection, characterized by severe leukopenia, anemia, or pancytopenia and liver necrosis in the absence of feather and beak abnormalities, may occur in young African grey parrots.

We have investigated the effects of soybean agglutinin on the cytoskeletal element actin in differentiated Caco-2 cells. The actin cytoskeleton of the cells was visualized by fluorescence microscopy using 7-nitrobenz-2-oxa-1, 3-diazole phallacidin as a specific marker for F-actin. Compared with control Caco-2 cells no changes in the fluorescence pattern were observed after incubation with soybean agglutinin. However, using the deoxyribonuclease-I inhibition assay a dose-related response was noted in the increase of intracellular G-actin after a 2-hour incubation period with soybean agglutinin. Already after exposure for 15 min to soybean agglutinin a decrease in intracellular F-actin was demonstrable. This apparent depolymerization could be prevented by incubating the Caco-2 cells with soybean agglutinin and the appropriate monosaccharide simultaneously. The increase in the amount of G-actin appeared to be correlated with a shortening of microvilli on the Caco-2 cells.

Zusammenfassung Eine Biopsietechnik zur Gewinnung von Dunndarm-Schleimhaut bei fistulierten Schweinen Um in vivo fur Versuchszwecke bei Schweinen Dunndarm-Schleimhaut zu gewinnen, wurde eine Biopsie-Technik bei fistulierten Schweinen mittels Entnahme von Dunndarm-Schleimhaut uber eine Jejunalfistel entwickelt und vorgestellt.

The literature concerning the effects of plant lectins on the small intestinal epithelium is reviewed. It appears that after oral intake, intact plant lectins can reach the small intestinal lumen. Their binding to the mucosal surface evokes an increased synthesis of glycoproteins and a degeneration of the intestinal epithelium. The epithelial alterations may result in hyperregenerative villus atrophy and endogenous nitrogen loss. These changes ultimately can lead to less efficient feed conversion, diminished growth, scouring, wasting and death. The possible significance of plant lectins in digestive disturbances in farm animals is suggested.
