Abstract

In this work, a method of design and support of the productive process developed for the manufacture of multilayer coated glass is presented. This method has been used in the design and manufacture of a solar control coating in laminated glass, with colorimetric and solar properties similar to a low emissivity insulating glass.En este trabajo, se presenta un método de diseño y apoyo al proceso de producción que se ha desarrollado para la realización de vidrios con estructuras de multicapas depositadas. Este método se ha utilizado en el diseño y realización práctica de un recubrimiento de control solar en estructura de vidrio laminado, cuyas propiedades colorimétricas y de protección solar son similares a un vidrio bajo-emisivo en doble acristalamiento.

Abstract

Background Global health foregrounds trust as a key requirement for the achievement of international health initiatives, but it remains an elusive concept that is often mobilised without consideration of its dimensions, drivers and downstream behavioural consequences. This paper aims to contribute to the conceptual development and measurement of ‘patient trust in primary healthcare’ from the lower middle-income country perspective of rural Lao PDR.Methods A two-phase mixed-method research design was implemented between January 2021 and April 2023. Phase 1 involved exploratory qualitative research to understand the local expressions and dimensions of patient trust in primary healthcare, with 25 semistructured interviews and 17 focus group discussions (120 participants) in eight villages in Bokeo Province. Phase 2 involved explanatory research to assess patterns of trust systematically at scale in 14 villages across four provinces, wherein 26 cognitive interviews, 17 expert interviews and non-participant community observations informed a community census survey with 1838 participants. We analysed qualitative data through content-oriented thematic analysis and developed an 8-item trust scale on that basis. Quantitative data analysis used descriptive statistical and regression analysis.Results We found that trust in primary healthcare is readily understood and intrinsically valuable in rural Lao PDR. Key dimensions included communication, respectful care, relationship, fairness, integrity, reputation, assurance of treatment and competence. The survey highlighted that reputation, competence, integrity and respectful care had the lowest trust scores. Health centre operations predicted the local expressions of trust. The behavioural consequences of trust were limited to a positive statistical association with antenatal care uptake among pregnant women but outweighed by alternative measures that also captured the availability of healthcare facilities.Conclusions Overall, the development of our quantitative trust scale offers a process model for future researchers. We conclude that interpersonal, institutional and service-related trust require more explicit recognition in health system development and integration into health policy.

Abstract

The 235U(n,f) cross section was measured in a wide energy range (25 meV - 170 keV) at the n_TOF facility at CERN, relative to 6Li(n,t) and 10B(n,alpha) standard reactions, with high resolution and accuracy, with a setup based on a stack of six samples and six silicon detectors placed in the neutron beam. In this paper we report on the results in the region between thermal and 10 keV neutron energy. A resonance analysis has been performed up to 200 eV, with the code SAMMY. The resulting fission kernels are compared with the ones extracted on the basis of the resonance parameters of the most recent major evaluated data libraries. A comparison of the n_TOF data with the evaluated cross sections is also performed from thermal to 10 keV neutron energy for the energy-averaged cross section in energy groups of suitably chosen width. A good agreement is found in average between the new results and the latest evaluated data files ENDF-B/VIII and JEFF-3.3, as well as with respect to the IAEA reference files. However, some discrepancies are still present in some specific energy regions. The new dataset here presented, characterized by unprecedented resolution and accuracy, can help improving the evaluations in the Resolved Resonance Region and up to 10 keV, and reduce the uncertainties that affect this region.

Abstract

CoFe2O4 thin films (5 nm and 20 nm thick) were grown by oxygen assisted molecular beam epitaxy on Pt(111) at 523~K and subsequently annealed at 773 K in vacuum or oxygen. They were characterized in-situ using Auger Electron Spectroscopy, Low-Energy Electron Diffraction, Scanning Tunneling Microscopy and Conversion Electron M\"ossbauer Spectroscopy. The as-grown films were composed of small, nanometric grains. Annealing of the films produced an increase in the grain size and gave rise to magnetic order at room temperature, although with a fraction of the films remaining in the paramagnetic state. Annealing also induced cobalt segregation to the surface of the thicker films. The measured M\"ossbauer spectra at low temperature were indicative of cobalt ferrite, the both films showing very similar hyperfine patterns. Annealing in oxygen or vacuum affected the cationic distribution, which was closer to that expected for an inverse spinel in the case of annealing in an oxygen atmosphere., Comment: 9 pages (two columns), 8 figures, 2 tables. Abstract

Abstract Background The dynamics of Staphylococcus aureus in patients and the hospital environment are relatively unknown. We studied these dynamics in a tertiary care hospital in the Netherlands. Methods Nasal samples were taken from adult patients at admission and discharge. Isolates cultured from clinical samples taken before and during hospitalization from these patients were included. Environmental samples of patient rooms were taken over a three-year period. Finally, isolates from clinical samples from patients with an epidemiological link to S. aureus positive rooms were included. Staphylococcal protein A (spa) typing was performed. Results Nasal samples were taken from 673 patients. One hundred eighteen (17.5%) were positive at admission and discharge, 15 (2.2%) patients acquired S. aureus during hospitalization. Nineteen patients had a positive clinical sample during hospitalization, 15.9% of the S. aureus were considered as from an exogenous source. One hundred and forty (2.8%) environmental samples were S. aureus positive. No persistent contamination of surfaces was observed. Isolates were highly diverse: spa typing was performed for 893 isolates, identifying 278 different spa types, 161 of these spa types were observed only once. Conclusion Limited transmission could be identified between patients and the hospital environment, and from patient-to-patient. Exogenous acquisition was assumed to occur in 15% of clinical samples. Environmental contamination was infrequent, temporarily, and coincided with the strain from the patient admitted to the room at that time. MRSA was rare and not found in the environment.

67. Exceptional thermal stability and enhanced hardness in a nanostructured Mg-Gd-Y-Zn-Zr alloy processed by high pressure torsion

Abstract

A Mg-8.2Gd-3.8Y-1.0Zn-0.4Zr (wt.%) alloy is processed by solution treatment and high pressure torsion (HPT) at room temperature to produce a nanostructured light material with high hardness. The stability of this alloy is subsequently tested through isochronal annealing for 0.5 h at 373 K to 673 K. The results reveal a thermal stability that is vastly superior to that of conventional Mg-based alloys processed by severe plastic deformation: the grain size remains at around 50 nm on heating to 573 K, and as the temperature is increased to 673 K, grain growth is restricted to within 500 nm. The stability of grain refinement of the present alloy/processing combination allowing grain size to be limited to 55 nm after exposure at 573 K, appears to be nearly one order of magnitude better than for the other SPD processed Mg-RE type alloys, and 2 orders of magnitude better than those of SPD processed RE-free Mg alloys. This superior thermal stability is attributed to formation of co-clusters near and segregation at grain boundaries, which cause a thermodynamic stabilization of grain size, as well as formation of β-Mg5RE equilibrium phase at grain boundaries, which impede grain growth by the Zener pinning effect. The hardness of the nanostructured Mg-Gd-Y-Zn-Zr alloy increases with increasing annealing temperature up to 573 K, which is quite different from the other SPD-processed Mg-based alloys. The high hardness of 136 HV after annealing at 573 K is mainly due to solute segregation and solute clustering at or near grain boundaries.

Abstract

Abstract Arthritogenic alphaviruses are positive-strand RNA viruses that cause debilitating musculoskeletal diseases affecting millions worldwide. A recent discovery identified the four-and-a-half-LIM domain protein 1 splice variant A (FHL1A) as a crucial host factor interacting with the hypervariable domain (HVD) of chikungunya virus (CHIKV) nonstructural protein 3 (nsP3). Here, we show that acute and chronic chikungunya disease in humans correlates with elevated levels of FHL1. We generated FHL1−/− mice, which when infected with CHIKV or o’nyong-nyong virus (ONNV) displayed reduced arthritis and myositis, fewer immune infiltrates, and reduced proinflammatory cytokine/chemokine outputs, compared to infected wild-type (WT) mice. Interestingly, disease signs were comparable in FHL1−/− and WT mice infected with arthritogenic alphaviruses Ross River virus (RRV) or Mayaro virus (MAYV). This aligns with pull-down assay data, which showed the ability of CHIKV and ONNV nsP3 to interact with FHL1, while RRV and MAYV nsP3s did not. We engineered a CHIKV mutant unable to bind FHL1 (CHIKV-ΔFHL1), which was avirulent in vivo. Following inoculation with CHIKV-ΔFHL1, mice were protected from disease upon challenge with CHIKV and ONNV, and viraemia was significantly reduced in RRV- and MAYV-challenged mice. Targeting FHL1-binding as an approach to vaccine design could lead to breakthroughs in mitigating alphaviral disease.

69. Magnitude and variability of individual elbow load in repetitive baseball pitching

Abstract

Abstract In baseball pitchers the elbow is exposed to high and repetitive loads (i.e. external valgus torque), caused by pitching a high number of balls in a practice session or game. This can result in overuse injuries like the ulnar collateral ligament (UCL) injury. To understand injury mechanisms, the effect of repetitive pitching on the elbow load magnitude and variability was investigated. In addition, we explored whether repetitive pitching affects elbow muscle activation during pitching. Fifteen pitchers threw each 60 to 110 balls. The external valgus torque and electromyography of three elbow muscles were quantified during each pitch. Linear mixed model analyses were performed to investigate the effect of repetitive pitching. On a group level, the linear mixed models showed no significant associations of repetitive pitching with valgus torque magnitude and variability and elbow muscle activity. Significant differences exist between pitchers in their individual trajectories in elbow valgus torque and muscle activity with repetitive pitching. This shows the importance of individuality in relation to repetitive pitching. In order to achieve effective elbow injury prevention in baseball pitching, individual characteristics of changes in elbow load and muscle activity in relation to the development of UCL injuries should be investigated.

Abstract

Electroconvulsive Therapy (ECT) is arguably the most effective intervention for treatment-resistant depression. While large interindividual variability exists, a theory capable of predicting individual response to ECT remains elusive. To address this, we posit a quantitative, mechanistic framework of ECT response based on Network Control Theory (NCT). Then, we empirically test our approach and employ it to predict ECT treatment response. To this end, we derive a formal association between Postictal Suppression Index (PSI) - an ECT seizure quality index - and whole-brain modal and average controllability, NCT metrics based on white matter brain network architecture, respectively. Exploiting the known association of ECT response and PSI, we then hypothesized an association between our controllability metrics and ECT response mediated by PSI. We formally tested this conjecture in N=50 depressive patients undergoing ECT. We show that whole-brain controllability metrics based on pre-ECT structural connectome data predict ECT response in accordance with our hypotheses. In addition, we show the expected mediation effects via PSI. Importantly, our theoretically motivated metrics are at least on par with extensive machine learning models based on pre-ECT connectome data. In summary, we derived and tested a control-theoretic framework capable of predicting ECT response based on individual brain network architecture. It makes testable, quantitative predictions regarding individual therapeutic response, which are corroborated by strong empirical evidence. Our work might constitute a starting point for a comprehensive, quantitative theory of personalized ECT interventions rooted in control theory.

Abstract

Candidemia is an opportunistic mycosis with high morbidity and mortality rates. Even though Candida albicans is the main causative agent, other Candida species, such as Candida tropicalis, are relevant etiological agents of candidiasis and candidemia. Compared with C. albicans, there is currently limited information about C. tropicalis’ biological aspects, including those related to the cell wall and the interaction with the host. Currently, it is known that its cell wall contains O-linked mannans, and the contribution of these structures to cell fitness has previously been addressed using cells subjected to chemical treatments or in mutants where O-linked mannans and other wall components are affected. Here, we generated a C. tropicalis pmt2∆ null mutant, which was affected in the first step of the O-linked mannosylation pathway. The null mutant was viable, contrasting with C. albicans where this gene is essential. The phenotypical characterization showed that O-linked mannans were required for filamentation; proper cell wall integrity and organization; biofilm formation; protein secretion; and adhesion to extracellular matrix components, in particular to fibronectin; and type I and type II collagen. When interacting with human innate immune cells, it was found that this cell wall structure is dispensable for cytokine production, but mutant cells were more phagocytosed by monocyte-derived macrophages. Furthermore, the null mutant cells showed virulence attenuation in Galleria mellonella larvae. Thus, O-linked mannans are minor components of the cell wall that are involved in different aspects of C. tropicalis’ biology.

Abstract

Background: A therapeutic intervention in psychiatry can be viewed as an attempt to influence the brain's large-scale, dynamic network state transitions underlying cognition and behavior. Building on connectome-based graph analysis and control theory, Network Control Theory is emerging as a powerful tool to quantify network controllability - i.e., the influence of one brain region over others regarding dynamic network state transitions. If and how network controllability is related to mental health remains elusive. Methods: From Diffusion Tensor Imaging data, we inferred structural connectivity and inferred calculated network controllability parameters to investigate their association with genetic and familial risk in patients diagnosed with major depressive disorder (MDD, n=692) and healthy controls (n=820). Results: First, we establish that controllability measures differ between healthy controls and MDD patients while not varying with current symptom severity or remission status. Second, we show that controllability in MDD patients is associated with polygenic scores for MDD and psychiatric cross-disorder risk. Finally, we provide evidence that controllability varies with familial risk of MDD and bipolar disorder as well as with body mass index. Conclusions: We show that network controllability is related to genetic, individual, and familial risk in MDD patients. We discuss how these insights into individual variation of network controllability may inform mechanistic models of treatment response prediction and personalized intervention-design in mental health., Comment: 24 pages, 1 figure
