Rationale: We developed an innovative, minimally invasive, highly efficient extracorporeal CO2 removal (ECCO2R) technique called respiratory electrodialysis (R-ED). Objectives: To evaluate the efficacy of R-ED in controlling ventilation compared with conventional ECCO2R technology. Methods: Five mechanically ventilated swine were connected to a custom-made circuit optimized for R-ED, consisting of a hemofilter, a membrane lung, and an electrodialysis cell. Electrodialysis regionally modulates blood electrolyte concentration to convert bicarbonate to CO2 before entering the membrane lung, enhancing membrane lung CO2 extraction. All animals underwent three repeated experimental sequences, consisting of four steps: baseline (1 h), conventionalECCO2R(2 h), R-ED (2 h), and finalNOECCO2R (1 h). Blood and gas flow were 250 ml/min and 10 L/min, respectively. Tidal volume was set at 8 ml/kg, and respiratory rate was adjusted to maintain arterial PCO2 at 50 mm Hg. Measurements and Main Results: During R-ED, chloride andH1 concentration increased in blood entering the membrane lung, almost doubling CO2 extraction compared with ECCO2R (11266 vs. 6465 ml/min, P,0.001). Compared with baseline, R-ED and ECCO2R reduced minute ventilation by 50% and 27%, respectively. Systemic arterial gas analyses remained stable during the experimental phases. No major complication occurred, but there was an increase in creatinine level. Conclusions: In this first in vivo application, we proved electrodialysis feasible and effective in increasingmembrane lung CO2 extraction. R-ED was more effective than conventional ECCO2R technology in controlling ventilation. Further studies are warranted to assess the safety profile of R-ED, especially regarding kidney function.

Cytosine methylation is a widespread epigenetic regulation mechanism. In healthy mature cells, methylation occurs at CpG dinucleotides within promoters, where it primarily silences gene expression by modifying the binding affinity of transcription factors to the promoters. Conversely, a recent study showed that in stem cells and cancer cell precursors, methylation also occurs at non-CpG pairs and involves introns and even gene bodies. The epigenetic role of such methylations and the molecular mechanisms by which they induce gene regulation remain elusive. The topology of both physiological and aberrant non-CpG methylation patterns still has to be detailed and could be revealed by using the differential stability of the duplexes formed between site-specific oligonucleotide probes and the corresponding methylated regions of genomic DNA. Here, we present a systematic study of the thermal stability of a DNA oligonucleotide sequence as a function of the number and position of non-CpG methylation sites. The melting temperatures were determined by monitoring the fluorescence of donor-acceptor duallabelled oligonucleotides at various temperatures. An empirical model that estimates the methylationinduced variations in the standard values of hybridization entropy and enthalpy was developed.

In the search of new drug delivery carriers for the brain, self-assembled nanoparticles (NP) were prepared from poly(N,N-dimethylacrylamide)-block-polystyrene polymer. NP displayed biocompatibility on cultured endothelial cells, macrophages and differentiated SH-SY5Y neuronal-like cells. The surface-functionalization of NP with a modified fragment of human Apolipoprotein E (mApoE) enhanced the uptake of NP by cultured human brain capillary endothelial cells, as assessed by confocal microscopy, and their permeability through a Transwell Blood Brain Barrier model made with the same cells, as assessed by fluorescence. Finally, mApoE-NP embedding doxorubicin displayed an enhanced release of drug at low pH, suggesting the potential use of these NP for the treatment of brain tumors. In the search of novel drug carriers for brain drug delivery, poly(N,N-dimethylacrylamide)-b-polystyrene nanoparticles (NP) have been functionalized with a modified fragment of Apolipoprotein E (mApoE). These NP show to be uptaken and to overcome a Blood-Brain Barrier (BBB) in vitro cellular model. The cytotoxicity of NP is also investigated on different cell lines, showing high biocompatibility.

A Monte Carlo simulation of a nematic Lebwohl-Lasher liquid crystal model with a certain percentage of randomly oriented frozen spins is presented. The site-site distance dependence of the nematic correlation functions obtained are generally well fitted by stretched exponentials, which evolve into simple exponential at low temperature. The dependence of the correlation length on the disorder density is presented and compared to corresponding experimental results.

The rotational correlation function of anisotropic colloidal particles can be measured by observing the fluctuations of the depolarized intensity scattered in the forward direction. In this work we present a theoretical treatment of forward depolarized light scattering. Our calculations show the importance of the insertion of a quarter-wave plate between the scattering cell and the analyzer. The role of the quarter-wave plate is to generate a heterodyne signal between the transmitted beam and the depolarized forward scattered field. In order to illustrate the theoretical calculations, we present experimental results concerning the measurement of the rotational diffusion coefficient in dispersions of fluorinated polymer colloids

An enormous dissipation of the order of 2 kJ/L takes place during the natural mixing process of fresh river water entering the salty sea. "Capacitive mixing" is a promising technique to efficiently harvest this energy in an environmentally clean and sustainable fashion. This method has its roots in the ability to store a very large amount of electric charge inside supercapacitor or battery electrodes dipped in a saline solution. Three different schemes have been studied so far, namely, Capacitive Double Layer Expansion (CDLE), Capacitive Donnan Potential (CDP) and Mixing Entropy Battery (MEB), respectively based on the variation upon salinity change of the electric double layer capacity, on the Donnan membrane potential, and on the electrochemical energy of intercalated ions.

One dimensional theory of the electro-diffusion of ions in activated carbon porous electrodes is applied to describe the dynamic cycle of the capacitive mixing (CAPMIX) based on the double layer expansion (CDLE) technique to harvest renewable energy from salinity gradients. The model combines the electro-diffusion of ions with adsorption and desorption of charge and neutral salt into the double layers at the solid liquid interface, providing a comprehensive and accurate description of the full CAPMIX cycle experimentally measured in prototype cells. A careful analysis of the simulated cycles identifies key parameters for the optimization of the extracted power, like the appropriate thickness and micro-structure of the electrodes, best materials and operation conditions of the electrochemical cell. These directions will be fundamental in the development of this technique as an economically competitive renewable energy source.

The "capacitive mixing" (CAPMIX) technique is an emerging technology aimed at the extraction of energy from salinity differences, e.g. between sea and river waters. CAPMIX benefits from the voltage rise that takes place between two electrodes dipped in a saline solution when its salt concentration is changed. Several kinds of electrodes have been proposed so far: activated carbon materials (Brogioli, 2009), membrane-based ion-selective electrodes (Sales et al., 2010), and battery electrodes (Biesheuvel and van der Wal, 2010). The power production mainly depends on two properties of each single electrode: the amplitude of the potential rise upon salinity change, and the potential in the high-salinity solution. The various electrode materials that have been used returned different values of the two parameters, and hence to different power productions. In this paper, we apply electrokinetic and electrochemical models to qualitatively explain the experimentally observed behaviors of various materials under different experimental conditions. The analysis allows to devise techniques for tailoring new materials, particularly suited for the CAPMIX technique. © 2013 Elsevier Inc.

Recently, we demonstrated that TLQP-21 triggers lipolysis and induces resistance to obesity by reducing fat accumulation [1]. TLQP-21 is a 21 amino acid peptide cleavage product of the neuroprotein VGF and was first identified in rat brain. Although TLQP-21 biological activity and its molecular signaling is under active investigation, a receptor for TLQP-21 has not yet been characterized. We now demonstrate that TLQP-21 stimulates intracellular calcium mobilization in CHO cells. Furthermore, using Atomic Force Microscopy (AFM), we also provide evidence of TLQP-21 binding-site characteristics in CHO cells. AFM was used in force mapping mode equipped with a cantilever suitably functionalized with TLQP-21. Attraction of this functionalized probe to the cell surface was specific and consistent with the biological activity of TLQP-21; by contrast, there was no attraction of a probe functionalized with biologically inactive analogues. We detected interaction of the peptide with the binding-site by scanning the cell surface with the cantilever tip. The attractive force between TLQP-21 and its binding site was measured, statistically analyzed and quantified at approximately 40 pN on average, indicating a single class of binding sites. Furthermore we observed that the distribution of these binding sites on the surface was relatively uniform. © 2012 Elsevier B.V. All rights reserved.

The opening of DNA double strands is extremely relevant to several biological functions, such as replication and transcription or binding of specific proteins. Such opening phenomenon is particularly sensitive to the aqueous solvent conditions in which the DNA molecule is dispersed, as it can be observed by considering the classical dependence of DNA melting temperature on pH and salt concentration. In the present work, we report a single-molecule study of the stability of DNA against denaturation when subjected to changes in solvent. We investigated the appearance of DNA instability under specific external applied force and imposed twist values, which was revealed by an increase in the temporal fluctuations in the DNA extension. These fluctuations occur in the presence of a continuous interval of equilibrium states, ranging from a plectonemic state to a state characterized by denaturation bubbles. In particular, we observe the fluctuations only around a characteristic force value. Moreover, this characteristic force is demonstrated to be notably sensitive to variations in the pH and ionic strength. Finally, an extension of a theoretical model of plectoneme formation is used to estimate the average denaturation energy, which is found to be linearly correlated to the melting temperature of the DNA double strands.

A model of the electro-diffusion of ions in porous electrodes is applied to analyze the dynamics of capacitive-mixing extraction of energy from salinity gradients with carbon porous electrodes. The complex time-evolution of the cell voltage observed in experiments is satisfactorily described. The asymmetry on the duration of the solution-change steps performed in open circuit is found to be due to the nonlinear voltage-concentration relationship of the electric double layers and to a current that redistributes the counterions along the depth of the electrode leading to nonuniform charge and salt adsorption. The validated model is an essential tool for the design and optimization of renewable energy extraction by this technique. © 2012 American Chemical Society.

We use the "magnetic tweezers" technique to show the structural transitions that the DNA undergoes in the force-torsion space. In particular, we focus on the regions corresponding to negative supercoiling. These regions are characterized by the formation of the so-called denaturation bubbles, which play an essential role in the replication and transcription of DNA. We experimentally map the region of the force-torsion space where the denaturation takes place. We observe that large fluctuations in DNA extension occur at one of the boundaries of this region, i.e., when the formation of denaturation bubbles and of plectonemes compete. To describe the experiments, we introduce a suitable extension of the classical model. The model correctly describes the position of the denaturation regions, the transition boundaries, and the measured values of the DNA extension fluctuations. © 2012 American Physical Society.

Electrochemical cells containing two electrodes dipped in an ionic solution are widely used as charge accumulators, either with polarizable (supercapacitor) or nonpolarizable (battery) electrodes. Recent applications include desalination ("capacitive deionization") and energy extraction from salinity differences ("capacitive mixing"). In this Letter, we analyze a general relation between the variation of the electric potential as a function of the concentration and the salt adsorption. This relation comes from the evaluation of the electrical and mechanical energy exchange along a reversible cycle, which involves salt adsorption and release by the electrodes. The obtained relation thus describes a connection between capacitive deionization and capacitive mixing. We check this relation with experimental data already reported in the literature, and moreover by some classical physical models for electrodes, including polarizable and nonpolarizable electrodes. The generality of the relation makes it very useful in the study of the properties of the electric double layer. © 2012 American Physical Society.

The process of mixing sea and river water can be utilised as a power source. At present, three groups of technology are established for doing so; i) mechanical; Pressure Retarded Osmosis PRO, ii) electrochemical reactions; Reverse ElectroDialysis (RED) and Nano Battery Electrodes (NBE) and iii) ultra capacitors; Capacitive Double Layer Expansion (CDLE) and Capacitors charge by the Donnan Potentials (CDP). The chemical potential for salt gradient power systems is only limited by the feed solution concentrations and is the same for all types of salt power branches, but the electric work to the grid, however, relies on the route of conversion and means chosen therein. The CAPMIX project is a joint project to develop and explore ultra capacitors for doing so. Ultra-capacitor materials can interact with sea and river water in order to be deployed as an electricity source. The author consortium is currently exploring two routes to extract the potential free energy from mixing sea and river water by such means. These two routes are the Capacitive Double Layer Expansion (CDLE) and Capacitors charge by the Donnan Potentials (CDP), which are both recently reported, since 2009. The denominator of the two processes is the porous carbon capacitors constituting the capacitors where the chemical energy is converted into electric energy (current). The CDP differs from the CDLE mainly because it includes the use of membranes in addition to the capacitor materials. © 2012 Published by Elsevier Ltd

The "capacitive mixing" (CAPMIX) technique is aimed at the extraction of energy from the salinity difference between the sea and rivers. It is based on the voltage rise that takes place at the electrodes when changing the salt concentration of the solution in which the two electrodes are dipped. In this paper, we focus on activated carbon electrodes, produced with various methods and treatments, and we measure their behaviour in CAPMIX experiments. We find that they behave as polarizable electrodes only on time scales of the order of minutes, while on longer time scales they tend to move to a specific "spontaneous" potential, likely due to chemical redox reactions. This analysis sheds light on the charge leakage, i.e. the loss of the stored charge due to undesired chemical reactions, which is one of the main hurdles of the CAPMIX technique when performed with activated carbon electrodes. We show that the leakage finds its origin in the tendency of the electrode to move to its spontaneous potential. Our investigation allows us to completely get rid of the leakage and demonstrates the striking result that CAPMIX cycles can be performed without an external power supply. © The Royal Society of Chemistry 2013.

When a coherent laser beam impinges on a random sample (e.g. a colloidal suspension), the scattered light exhibits characteristic speckles. If the temporal coherence of the light source is too short, then the speckles disappear, along with the possibility of performing homodyne or heterodyne scattering detection or photon correlation spectroscopy. Here we investigate the scattering of a so-called 'skewed coherence beam', i.e. a short-coherence beam modified such that the field is coherent within slabs that are skewed with respect to the wave fronts. We show that such a beam generates speckles and can be used for heterodyne scattering detection, despite its short temporal coherence. Moreover, we show that the heterodyne signal is not affected by multiple scattering. We suggest that the phenomenon presented here can be used as a means of carrying out heterodyne scattering measurement with any short-coherence radiation, including x-rays.

Binding of ligands to DNA gives rise to several relevant biological and biomedical effects. Here, through the use of atomic force microscopy (AFM), we studied the consequences of drug binding on the morphology of single DNA molecules. In particular, we quantitatively analyzed the effects of three different DNA-binding molecules (doxorubicin, ethidium bromide, and netropsin) that exert various pharmacologic and therapeutic effects. The results of this study show the consequences of intercalation and groove molecular binding on DNA conformation. These single-molecule measurements demonstrate morphological features that reflect the specific modes of drug-DNA interaction. This experimental approach may have implications in the design of therapeutically effective agents.

Photon correlation spectroscopy (PCS) is based on measuring the temporal correlation of the light intensity scattered by the investigated sample. A typical setup requires a temporally coherent light source. Here, we show that a short-coherence light source can be used as well, provided that its coherence properties are suitably modified. This results in a "skewed-coherence" light beam allowing that restores the coherence requirements. This approach overcomes the usual need for beam filtering, which would reduce the total brightness of the beam. © 2011 Optical Society of America.

Herein, we study the nanomechanical characteristics of single DNA molecules in the presence of DNA binders, including intercalating agents (ethidium bromide and doxorubicin), a minor groove binder (netropsin) and a typical alkylating damaging agent (cisplatin). We have used magnetic tweezers manipulation techniques, which allow us to measure the contour and persistence lengths together with the bending and torsional properties of DNA. For each drug, the specific variations of the nanomechanical properties induced in the DNA have been compared. We observed that the presence of drugs causes a specific variation in the DNA extension, a shift in the natural twist and a modification of bending dependence on the imposed twist. By introducing a naive model, we have justified an anomalous correlation of torsion data observed in the presence of intercalators. Finally, a data analysis criterion for discriminating between different molecular interactions among DNA and drugs has been suggested.

Binary mixtures of larger and smaller colloids having charges of equal sign display unusual response to low-frequency electric fields. We show here that the previously reported negative torque acting on rodlike particles when in presence of a sea of smaller particles is accompanied by a field-induced clearing of the suspension. Measurements of transmitted intensity performed on mixtures of large (dilute) and small (semidilute) spherical particles indicate that such a clearing effect is universal and due to an anisotropic E2 -dependent redistribution of the small spheres around the large ones. We interpret this behavior as resulting from O (E2) electro-osmotic flows whose magnitude is greatly enhanced by the presence of the small particles. © 2009 The American Physical Society.

The diffusive process of dispersed particles in a semi-infinite cell of an anisotropic fluid limited by an adsorbing surface is theoretically investigated. The densities of bulk and surface particles are analytically determined, taking into account the conservation of the number of particles immersed in the sample. It is shown that an anomalous diffusion process can take place according to the time scale considered. For short time interval there is a sub-diffusion process, followed by a super-diffusive regime. For large time interval the diffusive process tends to a normal behavior. The analysis is suitable for the description of the adsorption phenomena of neutral as well as charged particles in anisotropic media. Copyright © 2009 EPLA.

Light scattering techniques are widely used in many fields of condensed and soft matter physics. Usually these methods are based on the study of the scattered light in the far field. Recently, a new family of near field detection schemes has been developed, mainly for the study of small angle light scattering. These techniques are based on the detection of the light intensity near to the sample, where light scattered at different directions overlaps but can be distinguished by Fourier transform analysis. Here we report for the first time data obtained with a dynamic near field scattering instrument, measuring both polarized and depolarized scattered light. Advantages of this procedure over the traditional far field detection include the immunity to stray light problems and the possibility to obtain a large number of statistical samples for many different wave vectors in a single instantaneous measurement. By using the proposed technique we have measured the translational and rotational diffusion coefficients of rod-like colloidal particles. The obtained data are in very good agreement with the data acquired with a traditional light scattering apparatus. © 2009 Optical Society of America.

By using scattering in near field techniques, a microscope can be easily turned into a device measuring static and dynamic light scattering, very useful for the characterization of nanoparticle dispersions. Up to now, microscopy based techniques have been limited to forward scattering, up to a maximum of 30°. In this paper we present a novel optical scheme that overcomes this limitation, extending the detection range to angles larger than 90° (back-scattering). Our optical scheme is based on a microscope, a wide numerical aperture objective, and a laser illumination, with the collimated beam positioned at a large angle with respect to the optical axis of the objective (Tilted Laser Microscopy, TLM). We present here an extension of the theory of near field scattering, which usually applies only to paraxial scattering, to our strongly out-of-axis situation. We tested our instrument and our calculations with calibrated spherical nanoparticles of several different diameters, performing static and dynamic scattering measurements up to 110°. The measured static spectra and decay times are compatible with the Mie theory and the diffusion coefficients provided by the Stokes-Einstein equation. The ability of performing backscattering measurements with this modified microscope opens the way to new applications of scattering in near field techniques to the measurement of systems with strongly angle dependent scattering. © 2009 Optical Society of America.

Light scattering detection in the near field, a rapidly expanding family of scattering techniques, has recently proved to be an appropriate procedure for performing dynamic measurements. Here we report an algorithm, first suggested by Oh et al. (Phys. Rev. E 69, 21106 (2004)), based on the evaluation of the Exposure Time Dependent Spectrum (ETDS), which makes it possible to measure the fast dynamics of a colloidal suspension with the aid of a simple near field scattering apparatus and a CCD camera. The algorithm consists in acquiring static spectra in the near field at different exposure times, so that the measured decay times are limited only by the exposure time of the camera and not by its frame rate. The experimental set-up is based on a modified microscope, where the light scattered in the near field is collected by a commercial objective, but (unlike in standard microscopes) the light source is a He-Ne laser which increases the instrument sensitivity. The apparatus and the algorithm have been validated by considering model systems of standard spherical nano-particle.

We discuss the translocation of inhaled asbestos fibers based on pulmonary and pleuro-pulmonary interstitial fluid dynamics. Fibers can pass the alveolar barrier and reach the lung interstitium via the paracellular route down a mass water flow due to combined osmotic (active Na+ absorption) and hydraulic (interstitial pressure is subatmospheric) pressure gradient. Fibers can be dragged from the lung interstitium by pulmonary lymph flow (primary translocation) wherefrom they can reach the blood stream and subsequently distribute to the whole body (secondary translocation). Primary translocation across the visceral pleura and towards pulmonary capillaries may also occur if the asbestos-induced lung inflammation increases pulmonary interstitial pressure so as to reverse the trans-mesothelial and trans-endothelial pressure gradients. Secondary translocation to the pleural space may occur via the physiological route of pleural fluid formation across the parietal pleura; fibers accumulation in parietal pleura stomata (black spots) reflects the role of parietal lymphatics in draining pleural fluid. Asbestos fibers are found in all organs of subjects either occupationally exposed or not exposed to asbestos. Fibers concentration correlates with specific conditions of interstitial fluid dynamics, in line with the notion that in all organs microvascular filtration occurs from capillaries to the extravascular spaces. Concentration is high in the kidney (reflecting high perfusion pressure and flow) and in the liver (reflecting high microvascular permeability) while it is relatively low in the brain (due to low permeability of blood-brain barrier). Ultrafine fibers (length < 5 mu m, diameter < 0.25 mu m) can travel larger distances due to low steric hindrance (in mesothelioma about 90% of fibers are ultrafine). By combined light scattering and circular dichroism measurements (CD), we have investigated the coil-to-globule transition of the thermosensitive polymer poly(N-isopropylacrylamide) (pNIPAAm) copolymerized with a 1/10 fraction of valine- or leucine-derived groups randomly positioned along the chains. The comonomers provide the pNIPAAm chains with chirality, electric charge, and increased hydrophobicity. For valine-derived copolymers, the coil-globule transition is basically unmodified with respect to pNIPAAm, whereas doping with leucine-derived groups significantly lowers the transition temperature and makes the transition discontinuous. We find the CD signal of the chiral comonomers to cleanly depend on the local chain density. We interpret this behavior as an effect of the whole chain conformation on the conformations accessible to the chiral groups.

Prompted by the results obtained by Mantegazza et al. [Nature Physics 1 (2005) 103], where the electric birefringence of suspensions of elongated particles was strikingly affected by the presence of a sea of very small (size ratio lower than 10:1) colloidal spheres, we have undertaken an investigation of other electrokinetic phenomena in suspensions containing various relative concentrations of large (Teflon or polystyrene latex) and small (nanometer-sized silica spheres) colloids. We have determined the quantities that might be greatly affected by the size distribution of the particles, mainly in the presence of ac electric fields, since the response of the suspensions will show very characteristic relaxations, dominated in principle by the size of the particles. In this work, we report on measurements of the dielectric dispersion of mixed particles as a function of the concentration, ionic strength, and field frequency. The results indicate that the response is not just a simple combination of those obtained with suspensions of the individual particles, and in fact the presence of even small amounts of the small particles affects considerably the frequency response of the suspensions. We present a combined experimental and Monte Carlo study of a nematic phase in the presence of quenched disorder. The turbidity of a nematic liquid crystal embedded in a porous polymer membrane is measured under different applied field conditions for field-cooled and zero-field-cooled samples. We find that a significant permanent alignment of the nematic can be induced by fields as low as 0.1 V/ ¿ m applied during the isotropic to nematic transition. An analogous effect and dependence on sample history is found by studying the order parameter of a sprinkled disorder Lebwohl-Lasher spin model, indicating that dilute quenched randomness is sufficient to produce memory effects in nematics. The large memory induced by field cooling appears to be written in the system during the transition as a result of the field action on freely oriented nematic nuclei. At lower temperature the nuclei consolidate into permanent nematic textures developed from the interaction with quenched disorder
