Abstract

Hyphocapnodia sichuanensis Z. H. Lu, Wanas., Madrid & Maharachch., sp. nov. FIG 2. MycoBank number: MB 845350 Etymology: Named after Sichuan Province where it was collected Saprobic on living and decaying branches of an unidentified bamboo. Sexual morph: not observed. Asexual morph: hyphomycetous. Colonies on the natural substrate effuse, hairy, black, developing on the upper side of branches of the host. Mycelium immersed and superficial, forming dark brown stromata. Conidiomata synnematous, erect, subcylindrical, dark brown to black, 980–1500 µm tall, 15.5–24 µm wide at the apex, 30–50 µm wide at the base, with conidiogenous cells distributed mostly towards the upper half of the synnemata. Conidiophores macronematous, simple, dark brown, smooth to verruculose 900–1300 × 2–5 µm. Conidiogenous cells mostly lateral, monoblastic, (sub)cylindrical, integrated, determinate, smooth, 2–5 × 2–4 µm. Conidia subcylindrical to fusiform, straight to slightly curved or sigmoid, dark brown, verrucose, with an obtuse apex and a truncate base, dark brown at maturity, 38.5–51 × 10–15 µm (x̅ = 44 × 11.9 µm), with up to 8 transverse septa, conidial secession schizolytic. Germination occurs from the apical cell of the conidia. Culture characteristics: Colonies on PDA slow growing reaching 2 cm diam. in 3 weeks, irregular, superficial to erumpent, black, raised centrally and wrinkled, with an undulate margin, reverse black. Material examined: CHINA, Sichuan Province, Chengdu city, Bai Yun Gou, on dead branches of Bambusaceae, 27 September 2021, 30°47.52′ N, 103°24.19′ E, elev.: 990 m. Lu Z-H, HUEST 22.0031 (HKAS 124021, holotype), ex-type living culture CGMCC 3.23573 = UESTCC 22.0031. Notes: In BLASTn searches with the ITS sequence of this fungus, the closest hits were Polychaeton citri (strain CBS 116435, GenBank OM238161; Identities = 568 / 596 (95 %), 9 gaps (1 %)), Conidiocarpus caucasicus (voucher UBCF23755, GenBank KF263962; Identities = 535 / 564 (95 %), 9 gaps (1 %)), and Capnodium sp. (strain TMS - 2011, GenBank HQ631045; Identities = 532 / 563 (94 %), 8 gaps (1 %)). Based on morphology and sequence data, Hyphocapnodia is clearly distinct from the other members of the Capnodiaceae. In vitro, this fungus showed a remarkably different morphology, characterized by the absence of synnemata and micronematous conidiophores giving rise to conidia 55–132 × 9–16 µm (x̅ = 89.9 × 12.5 µm, n =16), which often appeared sinuous and showed up to 14 septa (mostly 5–10). Hyphocapnodia morphologically resembles Phaeoisaria (Pleurotheciaceae), Dokmaia (Pleosporales genera incertae sedis) and Neosporidesmium (Dothideomycetes genera incertae sedis) with its unbranched conidiophores forming distinct synnematal conidiomata (De Hoog & Papendorf 1976, Promputtha et al. 2003, Verma et al. 2021). However, these genera are phylogenetically distant from our new genus.The new genus has relatively close phylogenetic proximity to the Capnodium species. However, the asexual of these genera is distinct. Hyphocapnodia has synnematal conidiomata, whereas Capnodium is characterized by pycnidial conidiomata (Abdollahzadeh et al. 2020). Neodigitodesmium cheirosporum W. H. Tian & Maharachch. sp. nov. (FIG. 2) MycoBank: MB 844433 Etymology: The name refers to the cheiroid conidia Saprobic on submerged decayed wood in aquatic habitats. Sexual morph: Undetermined. Asexual morph: Hyphomycetous. Conidiomata sporodochial on natural substrate, superficial, compact, dark brown to black, with base attached on the surface of substrate. Conidiophores micronematous, reduced to conidiogenous cell. Conidiogenous cells 7–12 × 5–11 μm (x̅ = 9 × 7 μm, n = 15), holoblastic, at the base of the spore, hyaline. Conidia 35–70 × 9–24 μm (x̅ = 57 ×19 μm, n = 30), solitary, dictyosporous, conical shaped, cheiroid, not complanate, consisting of 1–4 rows with each row composed of 6–10 cells, with a hyaline basal connecting cell, not easy to separate, brown, subhyaline at the tip of peripheral rows. Culture characteristics: Conidia germinating on PDA within 12 h at 25 °C in the dark. Colonies on PDA circular, woolly at the margin, raised at the center, reverse yellow to brown center with a white margin. Material examined: CHINA, Sichuan Province, Chengdu, Baiyungou, on submerged decayed wood, 103°24′19″ E, 30°47′52″ N, 27 September 2021, W. H. Tian BY112_3 (HKAS 124014, holotype), ex-type culture CGMCC 3.23623 = UESTCC 22.0020. Notes: Combined sequences of LSU, ITS, SSU and tef1 confirmed the isolate of Neodigitodesmium formed a distinct clade in Dictyosporiaceae. However, it could not assign to any known genera (FIG. 1). In the combined gene tree, Neodigitodesmium is phylogenetically related to the Vikalpa australiensis (HKUCC 8797), Aquadictyospora lignicola (MFLUCC 17-1318) and Aquadictyospora clematidis (MFLUCC 17-2080) (FIG. 1), but they are not clustered on one branch and have obvious morphological differences. Neodigitodesmium is morphologically similar to Digitodesmium, both having sporodochial conidiomata and acrogenous, cheiroid, digitate conidia (Kirk 1981; Nóbrega et al. 2021). However, Neodigitodesmium differs from Digitodesmium and other Dictyosporiaceae members in conicalshaped conidia, composed of 1–4 compactly arranged rows of dark brown cells, with a hyaline basal connecting conidiogenous cells (FIG. 2). MycoBank: MB 844526 Basionym: Paradictyocheirospora tectonae Rajeshkumar, R. K. Verma, Boonmee, K. D. Hyde, Chandrasiri & Wijayaw, Phytotaxa 509 (3): 259–271 (2021). Notes: The genus Paradictyocheirospora was introduced by Rajeshkumar et al. (2021), with P. tectonae as the type species. The phylogenetic analysis based on a combined dataset of LSU, ITS, SSU and tef1 sequences show that P. tectonae is clustered within Digitodesmium (FIG. 1). In addition, Paradictyocheirospora possesses key characteristics of Digitodesmium such as punctiform, sporodochial conidiomata, acrogenous, cheiroid, digitate conidia, with an apical gelatinous cap (Kirk 1981; Rajeshkumar et al. 2021; Nóbrega et al. 2021). Thus, Paradictyocheirospora and Digitodesmium are congeneric. 73. Co-infection of Fusarium aglaonematis sp. nov. and Fusarium elaeidis Causing Stem Rot in Aglaonema modestum in China

Abstract

Aglaonema modestum (A. modestum) (Araceae) is an evergreen herbage, which is intensively grown as an ornamental plant in South China. A new disease was observed in A. modestum from 2020 to 2021 in Guangdong province, China. The disease symptoms associated with plants were initial leaf wilt, stem rot, and resulting plant death, leading to severe economic losses. In total, six Fusarium isolates were obtained from diseased plants. The putative pathogen was identified using both morphological characteristics and molecular phylogenetic analysis of calmodulin A (cmdA), RNA polymerase largest subunit 1 (rpb1), RNA polymerase II (rpb2), translation elongation factor-1α (tef1-α), and beta-tubulin (β-tubulin) sequences. Two Fusarium species were identified, namely, one new species, Fusarium aglaonematis (F. aglaonematis) belonging to Fusarium fujikuroi species complex. In addition, Fusarium elaeidis (F. elaeidis) belonging to the Fusarium oxysporum (F. oxysporum) species complex was also identified. Pathogenicity assays were conducted by inoculating each species into potted A. modestum plants and co-inoculating two species. The results showed that two Fusarium species could infect plants independently and can infect them together. Co-infection of these two species enhanced the disease severity of A. modestum. Compared to single inoculation of F. elaeidis, severity was higher and disease development was quicker when plants were only inoculated with F. aglaonematis. In addition, these two Fusarium species could infect Aglaonema plants without wounds, while inoculation with a physical injury increased disease severity. This is the first report of co-infection by F. aglaonematis and F. elaeidis causing stem rot on A. modestum worldwide. Abstract

Periconia is a polyphyletic and asexual morphic genus within the family Periconiaceae (Pleosporales). The genus is characterized by a pale to dark brown stipe with an apical conidial head and ellipsoidal to oblong conidia. Species of Periconia are widely distributed throughout the world in various hosts, while most species are isolated from graminaceous plants. During our investigations of microfungal in Sichuan Province, China, 26 Periconia isolates were collected from a wide variety of graminaceous plants. These isolates corresponded to 11 species based on the examination of morphology and multi-locus phylogenetic analysis (SSU, ITS, LSU, TEF1, RPB2). This includes six new species (P. chengduensis, P. cynodontis, P. festucae, P. imperatae, P. penniseti, and P. spodiopogonis) and five new records (P. byssoides, P. chimonanthi, P. cookie, P. pseudobyssoides, and P. verrucosa). A comprehensive description and illustrations of the new species are provided and discussed with comparable taxa. These discoveries expand our knowledge of the species diversity of Periconia taxa in graminaceous plants in China. [ABSTRACT FROM AUTHOR]
