Abstract

Superparamagnetic iron oxide nanoparticles (IONPs) are promising bioactive additives to fabricate magnetic scaffolds for bone tissue engineering. To date, there has been no report on osteoinductivity of IONP-incorporated calcium phosphate cement (IONP-CPC) scaffold on stem cells using an exterior static magnetic field (SMF). The objectives of this study were to: (1) develop a novel magnetic IONP-CPC construct for bone tissue engineering, and (2) investigate the effects of IONP-incorporation and SMF application on the proliferation, osteogenic differentiation and bone mineral synthesis of human dental pulp stem cells (hDPSCs) seeded on IONP-CPC scaffold for the first time. The novel magnetic IONP-CPC under SMF enhanced the cellular performance of hDPSCs, yielding greater alkaline phosphatase activities (about 3-fold), increased expressions of osteogenic marker genes, and more cell-synthesized bone minerals (about 2.5-fold), compared to CPC control and nonmagnetic IONP-CPC. In addition, IONP-CPC induced more active osteogenesis than CPC control in rat mandible defects. These results were consistent with the enhanced cellular performance by magnetic IONP in media under SMF. Moreover, nano-aggregates were detected inside the cells by transmission electron microscopy (TEM). Therefore, the enhanced cell performance was attributed to the physical forces generated by the magnetic field together with cell internalization of the released magnetic nanoparticles from IONP-CPC constructs.

Abstract

Rationale: Bone homeostasis is maintained by a balanced interplay of osteoblasts and osteoclasts. Osteoclasts are derived from monocyte/macrophage lineage. Major vault protein (MVP) is known to promote apoptosis and prevent metabolic diseases in macrophage. However, whether MVP is involved in osteoclastogenesis is unknown. Here, we identified an important function of MVP as a negative regulator of osteoclastogenesis and its therapeutic potential in preventing bone loss. Methods: Expression of MVP in osteoclasts was investigated in human tumor tissues with immunohistochemical staining. Next, we generated total body (Mvp-/- ) and monocyte-specific (Mvpf/fLyz2-Cre) MVP gene knockout mice to observe bone phenotype and osteoclastogenesis using micro-CT and bone histomorphometry. Moreover, we examined the effects of MVP on osteoclast differentiation, bone resorption, NFATc1 activation and calcium oscillations in vitro. Finally, we explored the clinical potential of targeting MVP in two osteoporosis mouse models and used an adeno-associated virus (AAV) gene to overexpress MVP locally in mice. Results: We found that Mvp-/- and Mvpf/fLyz2-Cre mice both exhibited osteoporosis-like phenotypes. MVP-deficiency also enhanced calcineurin-NFATc1 signaling and promoted NFATc1 activity, which led to enhanced osteoclastogenesis and bone resorption. Calcineurin inhibition using the small molecule inhibitor FK506 corrected the enhanced osteoclastogenesis in Mvpf/fLyz2-Cre group. Additionally, MVP reexpression in Mvpf/fLyz2-Cre group rescued calcineurin expression. MVP overexpression in wild-type mice prevented pathologic bone loss in mouse models of ovariectomized (OVX) and calvaria-adjacent lipopolysaccharide (LPS)-injected. Abstract

Titanium (Ti) alloys, particularly Ti6Al4V, are the most commonly used biomedical implant material. Ti alloys are biologically inert, so there have been continuous efforts to improve their osteogenic properties and clinical performance. Since TiO2 nanotubes (NT) appear to be excellent drug platforms, and strontium reportedly enhances osteogenesis, we constructed a TiO2 nanotube coating on the surface of Ti6Al4V and immersed it in Sr (OH)2 solution in order to incorporate Sr into TiO2 nanotubes (NT‐Sr). The results of field emission scanning electron microscope and X‐ray diffraction analysis verified the fabrication of NT‐Sr. We next added polydopamine (PDA) and cyclo‐ (arginine–glycine–aspartic acid–phenylalanine–cysteine) [c(RGDfC)] peptides to further promote biocompatibility of the implant. Fourier transform infrared spectroscopy and X‐ray photoelectron spectroscopy confirmed the existence of PDA and c(RGDfC). Mesenchymal stem cells (MSCs) were planted on Ti, NT, NT‐Sr, NT‐Sr/PDA, and NT‐Sr/PDA‐RGD surfaces. The adhesion and differentiation of MSCs on different surfaces were evaluated. The mRNA expression of alkaline phosphatase, runt‐related transcription factor 2 (Runx2) and type I collagen (Col I) of different groups were also tested. Finally, we observed that the NT‐Sr/PDA‐RGD group showed significantly better performance than other groups in terms of the differentiation and osteogenesis‐related gene expression of MSCs. Abstract

Multifunctional composite nanostructure prepared via electrospinning has attracted wide attention. In this study, Fe2O3-carbon composite nanofiber with particle–nanorod structure was successfully prepared via electrospinning and followed calcination. Then, the electromagnetic properties of this material have been fully characterized, and the influence of different preparation conditions on these properties has been studied. In addition, compared to pure γ -Fe2O3 nanoparticles and hollow Fe2O3 nanofibers, the composite nanofibers with a thickness of 2.64 mm exhibited an additional absorption peak at a frequency of 13.92 GHz and an enhancement in absorption at a frequency of 15.45 GHz, which may be attributed to the increase in electrical loss introduced by amorphous carbon and the enhanced magnetic loss resulting from the multi-stage reflection introduced by the particle–nanorod structure. This study shows that the composite of Fe2O3 and carbon, and the introduction of the particle–nanorod structure can improve the microwave absorption efficiency of materials, and more nanocomposites can be designed like this to further improve their electromagnetic properties and absorption efficiency in the future. Fe2O3-carbon composite nanofibers with particle–nanorod structure are obtained via electrospinning. The size of nanoparticle will increase with the increase of calcination temperature and time, and further electromagnetic and microwave test indicates that the microwave absorption efficiency of materials could be improved by the introduction of the particle–nanorod structure. Abstract

One of the key factors in tissue engineering and regenerative medicine is to optimize the interaction between seed cells and scaffolds such that the cells can grow in naturally biomimetic conditions. Their similarity to macromolecules and many unique properties mean that functional nanoparticles have promising potential for the modification and improvement of traditional scaffolds to obtain excellent biocompatibility, tunable stiffness, physical sensing, and stimulus-response capabilities. In the present study, we report magnetic poly(lactic- co-glycolic acid)/polycaprolactone (PLGA/PCL) scaffolds that were fabricated using a combination of the electrospinning technique and layer-by-layer assembly of superparamagnetic iron oxide nanoparticles (IONPs). PLGA/PCL scaffolds assembled with gold nanoparticles were prepared using the same method for comparison. The results showed that the assembled film of nanoparticles on the surface greatly enhanced the hydrophilicity and increased the elastic modulus of the scaffold, which subsequently improved the osteogenesis of the stem cells. Furthermore, the magnetic property of the IONPs proved to be the key factor in enhancing osteogenic differentiation, which explained the superior osteogenic capacity of the magnetic scaffolds compared with that of the gold nanoparticle-assembled scaffold. These results demonstrated the importance of magnetic nanomaterials as a bioactive interface between cells and scaffolds and will promote the design of biomaterials to improve tissue engineering and regenerative medicine efficacy.

Abstract

Piezoelectric catalysis is an emerging research field in recent years. Since the catalytic reaction usually occurs on the surface, the specific surface area is very important for piezoelectric catalysis. Therefore, nanoparticles and nanowires have been developed for piezoelectric catalysis and have attracted widespread attention. However, in actual water applications, the use of nanoparticles will cause secondary pollution. Therefore, an overall deformable nanoporous system is urgently needed to adapt to the shapes of various ultrasonic transducer heads. We have obtained macroscopically inorganic flexible barium titanate nanofiber membranes through electrospinning technology. Without traditional organic mixtures, the surface of the piezoelectric material can fully come into contact with water. The fiber membrane is macroscopically integrated, deformable, and can fully adapt to multiple ultrasonic transducers of various shapes. Calcium doping can effectively improve the catalytic effect, and the catalytic rate (k × 103) of the material with the highest doping concentration can reach 24.47 min−1. In addition, studies have found that lowering the calcination temperature can effectively improve the flexibility of the fiber membrane, which can be attributed to smaller barium titanate grains formed at low temperature. This work provides a generic method for preparing flexible inorganic piezoelectric materials that can be adapted to a variety of surfaces, and successfully applies it in the field of piezoelectric catalysis, which provides an important way for the practical research of piezoelectric catalysis in the future. Abstract

Abstract Background Maxillary expansion (ME) is a common practice in orthodontics that aims to increase the constricted maxillary arch width. Relapse often occurs, however, and better treatment strategies are needed. In order to develop a more effective method, this study was designed to further examine the process of tissue remodeling during ME, to identify the changes in expression of several proteins of interest, and to clarify the molecular mechanism responsible for tissue remodeling. Methods Male Wistar rats were randomly divided into control and ME groups. The rats were euthanized at various intervals over 11 days, and the dissected palates were prepared for histological examination. The structure of the midpalatal sutures changed little during the first three days. Proteins from samples in the ground midpalatal tissues obtained on the third day were subjected to two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF MS) analysis. Validation of protein expression was performed by Western blot analyses. Results From day 5, chondrocytes in the inner layer of suture cartilage and osteoblasts at the end of the suture cartilage began to proliferate, and the skeletal matrix increased later adjacent to the cartilage in the ME group. Comparative proteomic analysis showed increases in 22 protein spots present in the ME group. The changes in three proteins closely related to osteogenesis (parathyroid hormone, osteoprotegerin and vimentin) were confirmed by Western blotting. Conclusion Many proteins are over-expressed during ME, and they may play an important role in the remodeling process.
