51. Effect of a redox-mediating ligand shell on photocatalysis by CdS quantum dots

Abstract: Semiconductor quantum dots (QDs) are efficient organic photoredox catalysts due to their high extinction coefficients and easily tunable band edge potentials. Despite the majority of the surface being covered by ligands, our understanding of the effect of the ligand shell on organic photocatalysis is limited to steric effects. We hypothesize that we can increase the activity of QD photocatalysts by designing a ligand shell with targeted electronic properties, namely, redox-mediating ligands. Herein, we functionalize our QDs with hole-mediating ferrocene (Fc) derivative ligands and perform a reaction where the slow step is hole transfer from QD to substrate. Surprisingly, we find that a hole-shuttling Fc inhibits catalysis, but confers much greater stability to the catalyst by preventing a build-up of destructive holes. We also find that dynamically bound Fc ligands can promote catalysis by surface exchange and creation of a more permeable ligand shell. Finally, we find that trapping the electron on a ligand dramatically increases the rate of reaction. These results have major implications for understanding the rate-limiting processes for charge transfer from QDs and the role of the ligand shell in modulating it.

53. Sudden Unexpected Infant Deaths: 2015–2020

Abstract: OBJECTIVE Although the US infant mortality rate reached a record low in 2020, the sudden infant death syndrome (SIDS) rate increased from 2019. To understand if the increase was related to changing death certification practices or the coronavirus disease 2019 (COVID-19) pandemic, we examined sudden unexpected infant death (SUID) rates as a group, by cause, and by race and ethnicity. METHODS We estimated SUID rates during 2015 to 2020 using US period-linked birth and death data. SUID included SIDS, unknown cause, and accidental suffocation and strangulation in bed. We examined changes in rates from 2019 to 2020 and assessed linear trends during prepandemic (2015–2019) using weighted least squares regression. We also assessed race and ethnicity trends and quantified COVID-19–related SUID. RESULTS Although the SIDS rate increased significantly from 2019 to 2020 (P < .001), the overall SUID rate did not (P = .24). The increased SIDS rate followed a declining linear trend in SIDS during 2015 to 2019 (P < .001). Other SUID causes did not change significantly. Our race and ethnicity analysis showed SUID rates increased significantly for non-Hispanic Black infants from 2019 to 2020, widening the disparities between these two groups during 2017 to 2019. In 2020, CONCLUSIONS Diagnositic shifting likely explained the increased SIDS rate in 2020. Why the SUID rate increased for non-Hispanic Black infants is unknown, but warrants continued monitoring. Interventions are needed to address persistent racial and ethnic disparities in SUID.

Copyright © 2023 The Author(s). Background: Sleep disturbance is common following hospital admission both for COVID-19 and other causes. The clinical associations of this for recovery after hospital admission are poorly understood despite sleep disturbance contributing to morbidity in other scenarios. We aimed to investigate the prevalence and nature of sleep disturbance after discharge following hospital admission for COVID-19 and to assess whether this was associated with dyspnoea. Methods: CircCOVID was a prospective multicentre cohort substudy designed to investigate the effects of circadian disruption and sleep disturbance on recovery after COVID-19 in a cohort of participants aged 18 years or older, admitted to hospital for COVID-19 in the UK, and discharged between March, 2020, and October, 2021. Participants were recruited from the Post-hospitalisation COVID-19 study (PHOSP-COVID). Follow-up data were collected at two timepoints: an early time point 2–7 months after hospital discharge and a later time point 10–14 months after hospital discharge. Sleep quality was assessed subjectively using the Pittsburgh Sleep Quality Index questionnaire and a numerical rating scale. Sleep quality was also assessed with an accelerometer worn on the wrist (actigraphy) for 14 days. Participants were also clinically phenotyped, including assessment of symptoms (ie, anxiety [Generalised Anxiety Disorder 7-item scale questionnaire], muscle function [SARC-F questionnaire], dyspnoea [Dyspnoea-12 questionnaire] and measurement of lung function), at the early timepoint after discharge. Actigraphy results were also compared to a matched UK Biobank cohort (non-hospitalised individuals and recently hospitalised individuals). Multivariable linear regression was used to define associations of sleep disturbance with the primary outcome of breathlessness and the other clinical symptoms. PHOSP-COVID is registered on the ISRCTN Registry (ISRCTN10980107). Findings: 2320 of 2468 participants in the PHOSP-COVID study attended an early timepoint research visit a median of 5 months (IQR 4–6) following discharge from 83 hospitals in the UK. Data for sleep quality were assessed by subjective measures (the Pittsburgh Sleep Quality Index questionnaire and the numerical rating scale) for 638 participants at the early time point. Sleep quality was also assessed using device-based measures (actigraphy) a median of 7 months (IQR 5–8 months) after discharge from hospital for 729 participants. After discharge from hospital, the majority (396 [62%] of 638) of participants who had been admitted to hospital for COVID-19 reported poor sleep quality in response to the Pittsburgh Sleep Quality Index questionnaire. A comparable proportion (338 [53%] of 638) of participants felt their sleep quality had deteriorated following discharge after COVID-19 admission, as assessed by the numerical rating scale. Device-based measurements were compared to an age-matched, sex-matched, BMI-matched, and time from discharge-matched UK Biobank cohort who had recently been admitted to hospital. Compared to the recently hospitalised matched UK Biobank cohort, participants in our study slept on average 65 min (95% CI 59 to 71) longer, had a lower sleep regularity index (–19%; 95% CI –20 to –16), and a lower sleep efficiency (3·83 percentage points; 95% CI 3·40 to 4·26). Similar results were obtained when comparisons were made with the non-hospitalised UK Biobank cohort. Overall sleep quality (unadjusted effect estimate 3·94; 95% CI 2·78 to 5·10), deterioration in sleep quality following hospital admission (3·00; 1·82 to 4·28), and sleep regularity (4·38; 2·10 to 6·65) were associated with higher dyspnoea scores. Poor sleep quality, deterioration in sleep quality, and sleep regularity were also associated with impaired lung function, as assessed by forced vital capacity. Depending on the sleep metric, anxiety mediated 18–39% of the effect of sleep disturbance on dyspnoea, while muscle weakness mediated 27–41% of this effect. Interpretation: Sleep disturbance following hospital admission for COVID-19 is associated with dyspnoea, anxiety, and muscle weakness. Due to the association with multiple symptoms, targeting sleep disturbance might be beneficial in treating the post-COVID-19 condition. Abstract: We report the development of a specialty optical fiber preform fabrication system based on carbon monoxide (CO) laser heating. The laser heating is accomplished via a single-beam optical arrangement integrated into a rotating glass lathe. The CO laser output power and its beam quality are affected by absorption of the laser radiation by water vapor present in the surrounding air. This is addressed by construction of an enclosed and fully motorized system to enable preform processing in a dry air environment. The performance of the system is evaluated, and the ability to maintain a desired preform processing temperature is demonstrated. Relevant aspects of preform manufacturing, such as glass cutting, splicing, tapering, and overcladding, are described in detail. The process of using these aspects to fabricate optical fiber preforms made of highly dissimilar materials and of various core-to-cladding ratios is discussed. Specialty fibers drawn from these preforms exhibit low-loss and show good optical performance.

Background: Most studies of immunity to SARS-CoV-2 focus on circulating antibody, giving limited insights into mucosal defences that prevent viral replication and onward transmission. We studied nasal and plasma antibody responses one year after hospitalisation for COVID-19, including a period when SARS-CoV-2 vaccination was introduced. Methods: In this follow up study, plasma and nasosorption samples were prospectively collected from 446 adults hospitalised for COVID-19 between February 2020 and March 2021 via the ISARIC4C and PHOSP-COVID consortia. IgA and IgG responses to NP and S of ancestral SARS-CoV-2, Delta and Omicron (BA.1) variants were measured by electrochemiluminescence and compared with plasma neutralisation data. Findings: Strong and consistent nasal anti-NP and anti-S IgA responses were demonstrated, which remained elevated for nine months (p < 0.0001). Nasal and plasma anti-S IgG remained elevated for at least 12 months (p < 0.0001) with plasma neutralising titres that were raised against all variants compared to controls (p < 0.0001). Of 323 with complete data, 307 were vaccinated between 6 and 12 months; coinciding with rises in nasal and plasma IgA and IgG anti-S titres for all SARS-CoV-2 variants, although the change in nasal IgA was minimal (1.46-fold change after 10 months, p = 0.011) and the median remained below the positive threshold determined by pre-pandemic controls. Samples 12 months after admission showed no association between nasal IgA and plasma IgG anti-S responses (R = 0.05, p = 0.18), indicating that nasal IgA responses are distinct from those in plasma and minimally boosted by vaccination. Interpretation: The decline in nasal IgA responses 9 months after infection and minimal impact of subsequent vaccination may explain the lack of long-lasting nasal defence against reinfection and the limited effects of vaccination on transmission. Abstract: The roosterfish (Nematistius pectoralis) is a piscivorous predator targeted extensively in recreational fisheries throughout the eastern tropical Pacific; however, its biology is poorly understood. To address these shortcomings, we investigated vertical habitat use and behaviour of roosterfish in coastal Panama using pop-up satellite archival tags. Nearly 5000 h of vertical movement data across 274 days from five fish showed that roosterfish largely used the upper 10 m and 20 m of the water column during the night-time and daytime respectively, and rarely left the mixed layer. Roosterfish diving behaviour showed a clear diurnal pattern, with oscillatory dives beginning during dawn and continuing through dusk. Abstract: We present a measurement of the cosmic-ray spectrum above 100 PeV using the part of the surface detector of the Pierre Auger Observatory that has a spacing of 750 m. An inflection of the spectrum is observed, confirming the presence of the so-called second-knee feature. The spectrum is then combined with that of the 1500 m array to produce a single measurement of the flux, linking this spectral feature with the three additional breaks at the highest energies. The combined spectrum, with an energy scale set calorimetrically via fluorescence telescopes and using a single detector type, results in the most statistically and systematically precise measurement of spectral breaks yet obtained. These measurements are critical for furthering our understanding of the highest energy cosmic rays.

Abstract: There is a need to understand the feeding behavior ontogeny of newly received high-risk steers. Understanding how calves learn to eat out of a bunk will facilitate the implementation of management practices that expedite this process and allow animal managers to exploit this information to monitor individual animal health. To address this knowledge gap, the feeding behavior of high-risk steers that were received for backgrounding was quantified and characterized using electronic feedbunks in this context. To create this paradigm, crossbred steers (n = 20) were purchased in two groups (Group: n = 64 steers; Group 2: n = 56 steers) from a local auction market, transported 800 km to the Prairie Research Unit in Prairie, MS, and immediately weighed (shrunk body weight, BW). Steers were purchased according to research unit space availability. Therefore, purchases occurred 133 days apart. The subsequent day, steers were ranked by shrunk BW (227 ±.3 kg) and allocated to 1 of 8 drylot pens (n = 8 steers/pen) with two GrowSafe automated feeding systems per pen. Steers were fed a total mixed ration ad libitum for a 28-day period and steer BW was recorded on d 0, 2, 6, 10, 13, 21 and 28. Daily feeding behavior [e.g., meal duration (sec), intake (g/d), meal frequency (count/d), bunk visit frequency (count/d), head down duration (sec), maximum non-feeding interval (NFI,s), ratio of head down duration to bunk visit duration)] were calculated using Meal Criterion Calculation Software. The impact of time on feeding behavior and performance was analyzed using a mixed linear model (PROC MIXED). To determine when steers had established consistent dry matter intake (DMI), consistency was defined as when the DMI of a steer remained within one standard deviation of the 7-day average DMI of the steer. Latency to first bunk attendance (d), first feed consumption, attainment of NEm requirement, and latency (d) to establish consistent feed intake levels were analyzed using a generalized linear mixed model (PROC GLIMMIX) for binomial data. Steer BW increased across days (P < 0.01), whereas average daily gain was.48 ± 0.4 kg/d. All feeding behaviors changed across days (P < 0.01). Meal and bunk visit (duration and frequency), meal intake (mean and sum), head down duration, and maximum NFI increased over time. All steers visited the bunk and consumed their first meal by day 4 (P < 0.01). All steers achieved NEm requirements by day11 (P < 0.01). These results quantify the acclimation period of steers to a new environment and characterizes the learning and development of novel modes of eating. These results inform the development of behavioral expectations for newly received high-risk calves that can be used to monitor cattle health, understand cattle perception, and promote cattle welfare.

74. Effects of temperament on reproductive performance of Bos taurus heifers enrolled in the 7-day CO-Synch + controlled internal drug release protocol

Abstract: Cattle temperament significantly impacts production traits such as reproduction. The objective of this study was to assess the effects of temperament on pregnancy rates to fixed-timed artificial insemination (TAI) in Bos taurus beef heifers. A total of 297 Angus influenced heifers from 3 different locations were evaluated for temperament based on chute score and exit velocity on the first day of the estrus synchronization protocol (d-9) and classified by temperament type based on temperament score (calm ≤ 3 < excitable). Pregnancy status was determined by transrectal ultrasonography approximately 40 d after TAI. Hair from the tail switch was collected at d-9 and at d0 (TAI) for cumulative cortisol evaluation. A subset of 43 heifers from location 3 had blood samples collected at all handling events and evaluated for cortisol concentrations. Overall, 71% of heifers were classified as calm whereas 29% as excitable. Pregnancy rates to TAI were reduced (P = 0.042) in excitable heifers compared to calm heifers (36% vs. 55%, respectively). Mean concentration of cortisol in the hair was reduced over time (P < 0.001) from d-9 (3.5 ± 0.3 pg/mg) to d0 (1.74 ± 0.3 pg/mg) in all heifers, regardless of temperament. In addition, excitable heifers had increased circulating concentrations of cortisol when compared to calm heifers in all handling events (P = 0.015). More interestingly, despite of temperament, concentrations of circulating cortisol were reduced as the protocol events progressed, with greater cortisol concentrations on d-9, intermediate on d-3, and lesser on d0 (P = 0.031). Therefore, the present study demonstrates that heifer temperament has negative effects in pregnancy rates to TAI programs. However, according to blood cortisol concentrations, no chronic stress response was detected due to cattle handling for the TAI protocol. In fact, blood cortisol concentration was reduced between the initiation and completion of the protocol, indicating that heifers were acclimated to handing. These results support the adoption of acclimation protocols and proper cattle handling as a strategy to increase fertility of heifers that are exposed to TAI.

Abstract: Perennial legumes have potential to increase pasture productivity in the high rainfall zone (600–850 mm) of south-eastern Australia through their ability to use summer rainfall and fix nitrogen (N2). Various perennial legumes are being evaluated for this environment; however, little information exists on legume–rhizobia cross-host compatibility and its consequences for biological N2 fixation. This is especially important when legumes are sown into fields with a background of competitive rhizobia such as WSM1325 or sown as a pasture mix with different host–symbiont pairs. We studied the effectiveness and cross-host compatibility of five commercial rhizobial strains for a range of pasture legumes (nine species, 18 cultivars) under controlled environment conditions, and further evaluated nodule occupancy and competitiveness of a newly established pasture (13 species, 20 cultivars) in the field, by determining nodulation and production (biomass and N2 fixation). Three of the commercial inoculant strains formed root nodules with multiple legume species; commonly however, less N2 was fixed in cases where the inoculant was not the recommended strain for the legume species. Within a legume species, cultivars could differ in their ability to form effective root nodules with multiple rhizobial strains. White clover cvv. Trophy, Haifa and Storm, strawberry clover cv. Palestine, and Talish clover cv. Permatas formed effective nodules with both TA1 and WSM1325 rhizobial strains. White clover cultivars that could not form an effective symbiosis with the common background strain WSM1325 fixed less N2. The white clover × Caucasian clover hybrid formed effective symbiosis with strain TA1 but not with other commercial strains. Some species such as birdsfoot trefoil, Talish clover, sulfur clover and tetraploid Caucasian clover formed ineffective symbiosis in the field. Until resolved, this will likely inhibit their further development as pasture plants for similar permanent pasture environments.

Abstract: Double-cantilever beams (DCBs) are widely used to study mode-I fracture behavior and to measure mode-I fracture toughness under quasi-static loads. Recently, the authors have developed analytical solutions for DCBs under dynamic loads with consideration of structural vibration and wave propagation. There are two methods of beam-theory-based data reduction to determine the energy release rate: (i) using an effective built-in boundary condition at the crack tip, and (ii) employing an elastic foundation to model the uncracked interface of the DCB. In this letter, analytical corrections for a crack-tip rotation of DCBs under quasi-static and dynamic loads are presented, afforded by combining both these data-reduction methods and the authors’ recent analytical solutions for each. Convenient and easy-to-use analytical corrections for DCB tests are obtained, which avoid the complexity and difficulty of the elastic foundation approach, and the need for multiple experimental measurements of DCB compliance and crack length. The corrections are, to the best of the authors’ knowledge, completely new. Verification cases based on numerical simulation are presented to demonstrate the utility of the corrections.

Abstract: Summary A 7-year-old male neutered Labrador Retriever dog presented with acute-onset fever, shifting limb lameness and anorexia, with development of acute respiratory distress. At necropsy, there was vegetative endocarditis, which effaced the aortic valve. Gram staining of impression smears from the aortic valve and kidney revealed numerous gram-positive rods with some coryneform bacteria. Similar coryneform bacteria were isolated on aerobic culture of the aortic valve. Identification was attempted by matrix-assisted laser desorption ionization time-of-flight mass spectrometry and 16S sequencing, the latter of which indicated Corynebacterium mustelae. This is the second reported case of endocarditis in a dog involving C. mustelae and the first with a description of post-mortem pathology. This case is an example of the utility of various modalities to identify facultative anaerobic bacterial pathogens that may be difficult to culture and may be more widespread than previously diagnosed.

The aim of this study was to: a) understand medical students’ attitude towards the humanities in medical education and b) assess their understanding of the necessary qualities of doctors and how interaction with the humanities affects the development of such attributes. Methods A mixed methods survey was designed to elicit demographics, engagement, interest and perspective on curricular positioning, and to explore how students ranked the qualities of a doctor. It was distributed to medical students of all year groups in the 6-year bachelor of medicine, bachelor of surgery (MBBS) course at Imperial College London. Results One hundred nine fully completed questionnaires were received. No significant difference was found in engagement or interest in the humanities between genders. Students felt strongly that humanities subjects shouldn’t be assessed (71:18) though some felt it was necessary for engagement, while no consensus was reached on whether these subjects should be elective or not (38:31). The majority of students wanted more medical humanities to be incorporated into the traditional medical course with a preference of incorporation into the first 3 years. Junior medical students were more likely to rank empathy as a highly desirable attribute than senior students. Students provided qualitative insights into curricular positioning, assessment and value. Conclusions This study provides the perspective of medical students on how and whether the humanities should be positioned in medical education. It may be helpful to medical schools that are committed to student involvement in curriculum design.

Abstract: The objective of this study was to evaluate the use of corpus luteum (CL) color Doppler (CD) ultrasonography and pregnancy-associated glycoproteins (PAG) for early pregnancy diagnosis and examine their ability to predict late embryonic/early fetal mortality (LEM) in Bos taurus beef replacement heifers. Beef heifers (n = 178) were exposed to a 7-d CO-Synch + CIDR protocol followed by fixed-time artificial insemination (day 0). On days 20 and 22, B-mode and CD ultrasonography were performed to evaluate CL morphometries and blood perfusion, respectively. Heifers were considered nonpregnant when CL area was2 cm2 or estimated luteal blood perfusion was ≤30% of the total luteal area. Blood samples were collected on days 25 and 29 to estimate circulating concentrations of PAG. Conventional ultrasonography on days 29 and 94 was utilized to determine pregnancy status and considered the gold standard method for pregnancy diagnosis. Pregnant heifers had greater (P0.01) CL diameter, area, volume, and blood perfusion when compared with nonpregnant heifers on days 20 and 22. Accuracy of CD on days 20 and 22, and PAG on days 25 and 29 were 91%, 94%, 96%, and 98%, respectively. No false-negative results were observed for CD on both days 20 and 22 (negative predicted value = 100%) and false-positive results represented 8% and 6% of the diagnoses. Heifers that experienced LEM between days 29 and 94 of gestation had decreased luteal (P = 0.02) volume on day 20 and tended (P = 0.07) to have decreased concentrations of PAG on day 29 compared with heifers that maintained pregnancy. However, both CD and PAG failed to predict embryonic mortality. In conclusion, CD successfully detected most nonpregnant replacement heifers as early as day 20 of gestation, while resulting in no false negative diagnoses. Both CD and PAG failed to predict LEM in the present study.Identifying nonpregnant females early after breeding allows cattle producers to rapidly rebreed females that failed to conceive after their first artificial insemination and consequently increases reproductive efficiency. Additionally, embryonic and fetal mortality during early gestation are the main drivers of pregnancy failure and represents a significant economic burden to both the beef and dairy industries. This study evaluated the use of color Doppler (CD) ultrasonography of specific ovarian structures and blood concentrations of pregnancy-associated glycoproteins (PAG) as potential methods to diagnose pregnancy earlier than industry-standard techniques. Moreover, the present study investigated the use of CD and blood PAG as predictor of pregnancy loss. Data generated here indicates that CD and blood PAG can accurately detect pregnancy as early as 20 and 25 d after insemination, respectively. In the present study, heifers that experienced pregnancy loss between days 29 and 94 of gestation tended to have decreased plasma concentrations of PAG during early pregnancy. Heifers experiencing pregnancy loss between days 29 and 94 of gestation were also more likely to have a luteal cavity on day 20 of gestation and had decreased luteal volume on the same day. However, results reported herein indicate that both CD and blood concentrations of PAG failed to predict pregnancy loss in replacement heifers.

86. Impact of preweaning vaccination on host gene expression and antibody titers in healthy beef calves

Abstract: The impact of preweaning vaccination for bovine respiratory viruses on cattle health and subsequent bovine respiratory disease morbidity has been widely studied yet questions remain regarding the impact of these vaccines on host response and gene expression. Six randomly selected calves were vaccinated twice preweaning (T1 and T3) with a modified live vaccine for respiratory pathogens and 6 randomly selected calves were left unvaccinated. Whole blood samples were taken at first vaccination (T1), seven days later (T2), at revaccination and castration (T3), and at weaning (T4), and utilized for RNA isolation and sequencing. Serum from T3 and T4 was analyzed for antibodies to BRSV, BVDV1a, and BHV1. Sequenced RNA for all 48 samples was bioinformatically processed with a HISAT2/StringTie pipeline, utilizing reference guided assembly with the ARS-UCD1.2 bovine genome. Differentially expressed genes were identified through analyzing the impact of time across all calves, influence of vaccination across treatment groups at each timepoint, and the interaction of time and vaccination. Calves, regardless of vaccine administration, demonstrated an increase in gene expression over time related to specialized proresolving mediator production, lipid metabolism, and stimulation of immunoregulatory T-cells. Vaccination was associated with gene expression related to natural killer cell activity and helper T-cell differentiation, enriching for an upregulation in Th17-related gene expression, and downregulated genes involved in complement system activity and coagulation mechanisms. Type-1 interferon production was unaffected by the influence of vaccination nor time. To our knowledge, this is the first study to evaluate mechanisms of vaccination and development in healthy calves through RNA sequencing analysis.

Abstract: Quantifying the timing (duration and frequency) of brief visual events is vital to human perception, multisensory integration and action planning. Tuned neural responses to visual event timing have been found in association cortices, in areas implicated in these processes. Here we ask how these timing-tuned responses are related to the responses of early visual cortex, which monotonically increase with event duration and frequency. Using 7-Tesla functional magnetic resonance imaging and neural model-based analyses, we find a gradual transition from monotonically increasing to timing-tuned neural responses beginning in the medial temporal area (MT/V5). Therefore, across successive stages of visual processing, timing-tuned response components gradually become dominant over inherent sensory response modulation by event timing. This additional timing-tuned response component is independent of retinotopic location. We propose that this hierarchical emergence of timing-tuned responses from sensory processing areas quantifies sensory event timing while abstracting temporal representations from spatial properties of their inputs.

Abstract: Sepsis is a leading cause of death in hospitals requiring prompt recognition and treatment. The sepsis bundle is the cornerstone of sepsis treatment. Studies have evaluated the impact of a sepsis huddle on sepsis bundle compliance but not in sepsis identification.Measure the effect of a multidisciplinary sepsis bedside huddle in the Emergency Department (ED) on sepsis identification and sepsis bundle compliance.Retrospective, single-center, cohort study. Pre-huddle patients were identified via Best Practice Advisory (BPA) alert on the electronic medical record from 11/01/2019-3/31/2020. The post-huddle group were patients for whom a sepsis huddle was activated from 11/01/2020-3/31/2021.116 patients met inclusion criteria and 15 were determined to not have sepsis for a total of 21 pre-huddle and 80 post-huddle patients. Comparing pre-post results, sepsis huddle increased code sepsis activation (10% vs 91%, p0.001); sepsis bundle compliance (24% vs 80%, p0.001); antibiotics within one hour (33% vs 90%, p0.001); culture within one hour (67% vs 95%, p0.001), order entry30 min. (29% vs 86%, p0.001); and median order entry time (48 vs. 3 min, p0.001). Post-huddle, 80% of order entries were ≤ 20 min. Logistic regression predicting sepsis code found huddle to be the first predictor, (p0.0000005). Hour-1 bundle compliance was predicted by physician/physician assistant order ≤30 min (RSepsis bedside huddle in the ED improves identification and sepsis bundle compliance. Results suggest increased order entry speed caused bundle improvement.

Abstract: Accumulation of tau is observed in dementia, with human tau displaying 6 isoforms grouped by whether they display either 3 or 4 C-terminal repeat domains (3R or 4R) and exhibit no (0N), one (1N) or two (2N) N terminal repeats. Overexpression of 4R0N-tau in rat hippocampal slices enhanced the L-type calcium (Ca2+) current-dependent components of the medium and slow afterhyperpolarizations (AHPs). Overexpression of both 4R0N-tau and 4R2N-tau augmented CaV1.2-mediated L-type currents when expressed in tsA-201 cells, an effect not observed with the third 4R isoform, 4R1N-tau. Current enhancement was only observed when the pore-forming subunit was co-expressed with CaVβ3 and not CaVβ2a subunits. Non-stationary noise analysis indicated that enhanced Ca2+ channel current arose from a larger number of functional channels. 4R0N-tau and CaVβ3 were found to be physically associated by co-immunoprecipitation. In contrast, the 4R1N-tau isoform that did not augment expressed macroscopic L-type Ca2+ current exhibited greatly reduced binding to CaVβ3. These data suggest that physical association between tau and the CaVβ3 subunit stabilises functional L-type channels in the membrane, increasing channel number and Ca2+ influx. Enhancing the Ca2+-dependent component of AHPs would produce cognitive impairment that underlie those seen in the early phases of tauopathies.

Abstract: Low birthweight and reduced height gain during infancy (stunting) may arise at least in part from adverse early life environments that trigger epigenetic reprogramming that may favor survival. We examined differential DNA methylation patterns using targeted methyl sequencing of regions regulating gene activity in groups of rural Gambian infants: (a) low and high birthweight (DNA from cord blood (n = 16 and n = 20, respectively), from placental trophoblast tissue (n = 21 and n = 20, respectively), and DNA from peripheral blood collected from infants at 12 months of age (n = 23 and n = 17, respectively)), and, (b) the top 10% showing rapid postnatal length gain (high, n = 20) and the bottom 10% showing slow postnatal length gain (low, n = 20) based on z score change between birth and 12 months of age (LAZ) (DNA from peripheral blood collected from infants at 12 months of age). Using BiSeq analysis to identify significant methylation marks, for birthweight, four differentially methylated regions (DMRs) were identified in trophoblast DNA, compared to 68 DMRs in cord blood DNA, and 54 DMRs in 12‐month peripheral blood DNA. Twenty‐five DMRs were observed to be associated with high and low length for age (LAZ) at 12 months. With the exception of five loci (associated with two different genes), there was no overlap between these groups of methylation marks. Of the 194 CpG methylation marks contained within DMRs, 106 were located to defined gene regulatory elements (promoters, CTCF‐binding sites, transcription factor‐binding sites, and enhancers), 58 to gene bodies (introns or exons), and 30 to intergenic DNA. Distinct methylation patterns associated with birthweight between comparison groups were observed in DNA collected at birth (at the end of intrauterine growth window) compared to those established by 12 months (near the infancy/childhood growth transition). The longitudinal differences in methylation patterns may arise from methylation adjustments, changes in cellular composition of blood or both that continue during the critical postnatal growth period, and in response to early nutritional and infectious environmental exposures with impacts on growth and longer‐term health outcomes.

On the other hand, its usefulness for photochemical reactions is more questionable, as different energy surfaces are involved. So far, no systematic studies on the validity of the Hammond postulate for photochemical reactions are available. The present work aims at filling this gap by providing a simple, unbiased series of test reactions based on the stereospecificity of isotopically labelled substrates., In this work, a series of photochemical reactions were performed on isotopically labelled substrates to explore the validity of Hammond's postulate for the excited state.

Abstract: Objective The A6702 multisite trial confirmed that apparent diffusion coefficient (ADC) measures can improve breast MRI accuracy and reduce unnecessary biopsies, but also found that technical issues rendered many lesions non-evaluable on diffusion-weighted imaging (DWI). This secondary analysis investigated factors affecting lesion evaluability and impact on diagnostic performance. Methods The A6702 protocol was IRB-approved at 10 institutions; participants provided informed consent. In total, 103 women with 142 MRI-detected breast lesions (BI-RADS assessment category 3, 4, or 5) completed the study. DWI was acquired at 1.5T and 3T using a four b-value, echo-planar imaging sequence. Scans were reviewed for multiple quality factors (artifacts, signal-to-noise, misregistration, and fat suppression); lesions were considered non-evaluable if there was low confidence in ADC measurement. Associations of lesion evaluability with imaging and lesion characteristics were determined. Areas under the receiver operating characteristic curves (AUCs) were compared using bootstrapping. Results Thirty percent (42/142) of lesions were non-evaluable on DWI; 23% (32/142) with image quality issues, 7% (10/142) with conspicuity and/or localization issues. Misregistration was the only factor associated with non-evaluability (P = 0.001). Smaller (≤10 mm) lesions were more commonly non-evaluable than larger lesions (p Conclusion Image quality remains a technical challenge in breast DWI, particularly for smaller lesions. Protocol optimization and advanced acquisition and post-processing techniques would help to improve clinical utility.

Abstract: Purpose Toll-like receptor 4 (TLR4) is an inflammatory receptor expressed ubiquitously in immune cells as well as skeletal muscle and other metabolic tissues. Skeletal muscle develops favorable inflammation-mediated metabolic adaptations from exercise training. Multiple inflammatory myokines, downstream from TLR4, are proposed links to the metabolic benefits of exercise. Additionally, activation of TLR4 alters skeletal muscle substrate preference. The role of skeletal muscle TLR4 (mTLR4) in exercise metabolism has not previously been investigated. Herein, we aimed to specifically test the significance of mTLR4 to exercise-induced metabolic adaptations. Methods We developed a novel muscle-specific TLR4 knockout (mTLR4) mouse model on C57BL/6J background. Male mTLR4 mice and wild type (WT) littermates were compared under sedentary (SED) and voluntary wheel running (WR) conditions for 4 weeks. Results mTLR4 deletion revealed marked reductions in downstream interleukin-1 receptor-associated kinase-4 (IRAK4) phosphorylation. In addition, the disruption of mTLR4 signaling prominently blunted the metabolic adaptations in WR-mTLR4 mice as oppose to substantial improvements exhibited by the WT counterparts. Voluntary WR in WT mice, relative to SED, resulted in significant increases in skeletal muscle fatty acid oxidation (FAO), glucose oxidation (GO), and associated mitochondrial enzyme activities, all of which were not significantly changed in mTLR4 mice. Abstract: Distributed temperature sensing (DTS) uses Raman scatter from laser light pulsed through an optical fiber to observe temperature along a cable. Temperature resolution across broad scales (seconds to many months, and centimeters to kilometers) make DTS an attractive oceanographic tool. Although DTS is an established technology, oceanographic DTS observations are rare since significant deployment, calibration, and operational challenges exist in dynamic oceanographic environments. Here, results from an experiment designed to address likely oceanographic DTS configuration, calibration, and data processing challenges provide guidance for oceanographic DTS applications. Temperature error due to suboptimal calibration under difficult deployment conditions is quantified for several common scenarios. Alternative calibration, analysis, and deployment techniques that help mitigate this error and facilitate successful DTS application in dynamic ocean conditions are discussed.
