Abstract

The flux of cosmic ray hadrons at the atmospheric depth of 820 g/cm^2 has been measured by means of the EAS-TOP hadron calorimeter (Campo Imperatore, National Gran Sasso Laboratories, 2005 m a.s.l.). The hadron spectrum is well described by a single power law : S(E_h) = (2.25 +- 0.21 +- 0.34(sys)) 10^(-7)(E_h/1000)^(-2.79 +- 0.05) m^(-2) s^(-1) sr^(-1) GeV^(-1) over the energy range 30 GeV-30 TeV. The procedure and the accuracy of the measurement are discussed. The primary proton spectrum is derived from the data by using the CORSIKA/QGSJET code to compute the local hadron flux as a function of the primary proton spectrum and to calculate and subtract the heavy nuclei contribution (basing on direct measurements). Over a wide energy range E_0 = 0.5-50 TeV its best fit is given by a single power law : S(E_0) = (9.8 +- 1.1 +- 1.6(sys)) 10^(-5) (E_0/1000)^(-2.80 +- 0.06) m^(-2) s^(-1) sr^(-1) GeV^(-1). The validity of the CORSIKA/QGSJET code for such application has been checked using the EAS-TOP and KASCADE experimental data by reproducing the ratio of the measured hadron fluxes at the two experimental depths (820 and 1030 g/cm^2 respectively) at better than 10% in the considered energy range., Comment: 16 pages, 9 figures, accepted for publication in Astroparticle Physics

Abstract

Background The purpose of the study was to evaluate the benefit of adjuvant chemotherapy (AC) versus surgery alone in patients with muscle-invasive bladder cancer (MIBC). Patients and methods One hundred and ninety-four patients with pT2G3, pT3–4, N0–2 transitional cell bladder carcinoma were randomly allocated to control (92 patients) or to four courses of AC (102 patients). These latter patients were further randomly assigned to receive gemcitabine 1000 mg/m2 days 1, 8 and 15 and cisplatin 70 mg/m2 day 2 or gemcitabine as above plus cisplatin 70 mg/m2 day 15, every 28 days. Results At a median follow-up of 35 months, the 5-year overall survival (OS) was 48.5%, with no difference between the two arms [P = 0.24, hazard ratio (HR) 1.29, 95% confidence interval (CI) 0.84–1.99]. Mortality hazard was significantly correlated with Nodes (N) and Tumor (T) stage. The control and AC arms had comparable disease-free survival (42.3% and 37.2%, respectively; P = 0.70, HR 1.08, 95% CI 0.73–1.59). Only 62% of patients received the planned cycles. A significant higher incidence of thrombocytopenia was observed in patients receiving cisplatin on day 2 (P = 0.006). A similar global quality of life was observed in the two arms. Conclusion The study was underpowered to demonstrate that AC with cisplatin and gemcitabine improves OS and disease-free survival in patients with MIBC.

Abstract

A search for transient point sources of ultra-high-energy (UHE) gamma-rays has been performed, based on the correlation of two extensive air shower arrays, BAKSAN (North Caucasus, 1700 m a.s.l., BAKSAN Neutrino Observatory, Russia) and EAS-TOP (Campo Imperatore, 2005 a.s.l., Laboratori Nazionali del Gran Sasso, Italy), which are located at very similar latitudes (phi approximate to 43 degrees N), and separated in longitude by Delta lambda approximate to 33.7 degrees. The search has been conducted at primary energy E (gamma) greater than or equal to 5 x 10(13) eV on the time scale of a daily source transit over the sky, observable in the northern hemisphere (19 degrees < delta < 69 degrees), and for three sources, namely the Crab Nebula, Markarian 421 and Markarian 501, observed at TeV energies by atmospheric Cherenkov detectors. The stability of the single detectors has been studied and verified up to high significance level. No coincident excess, not compatible with the expectations from statistical fluctuations, has been observed. The obtained upper limits to the rate of transient events, n gamma, at 90% c.l., are concerning the sky survey, in the declination band corresponding to the zenith, n gamma/(Omega t)< 12/(yr sr) with Phi(gamma),(E-gamma > 5 x 10(13) eV)> 2.0 x 10(-11) cm(-2)s(-1) and duration Delta t < 8 h; concerning the candidate sources, e.g., for Markarian 421: n gamma/t < 7.7/yr with Phi(gamma),(E-gamma, > 5 x 10(13) eV) > 1.3 x 10(11) cm(-2) s(-1) and Delta t < 7.5 h. cm A coincident episode from Markarian 421. observed on 15 January, 1994, with expected chance imitation rate n(ch) = 0.01 is discussed. Abstract

A panel of international breast cancer experts formulated a declaration of consensus regarding many key issues in the use of primary systemic therapy (PST) either in clinical routine or research practice. The attainment of pathological complete response (pCR), defined as no residual invasive tumor in the surgical specimens both in breast and in axillary nodes, is one of the main goals of PST, and pCR can be used as the primary objective in prospective clinical trials. However, pCR is not a reliable endpoint with all treatment approaches, and alternatives such as Ki67 index of the residual invasive disease or after 2 weeks of PST are also potential endpoints. PST has several advantages: breast conservation and the unique opportunity to obtain information on the interaction between treatment and tumor biology. Changes in tumor biology after PST are an early phenomenon; so, an additional core biopsy performed after 14 days from treatment start should be considered in clinical trials.

Abstract

The cross section for large p(t) jet production for transverse momentum 10 less than or equal to p(t) less than or equal to 20 GeV/c and rapidity 1.6 less than or equal to eta less than or equal to 2.6 in p-N (p-'air') interactions is studied from the analysis of multicore Extensive Air Showers recorded in the EAS-TOP calorimeter. The projectiles are the leading particles interacting at atmospheric depths between 250 and 480 cm(-2), the CMS energy of interaction being root s = 500 GeV. The slope of the p(t) distribution agrees with the expected one from the p-(p) over bar collider data at the same CMS energy. The measured jet production cross section in p-N interactions with respect to p-(p) over bar interactions is (d sigma/dp(t))(pN)(jet) = (d sigma/dp(t))(p (p) over bar)(jet) . A(alpha) with alpha = 1.56 +/- 0.07 for A = 14.7 (average mass number of 'air' nuclei). Such value is compatible within the experimental uncertainties with the one obtained in p-nucleus accelerator measurements at root s similar to 30 GeV in the same range of transverse momentum and rapidity. Abstract

We describe and discuss the operation, calibration and stability of the EAS-TOP calorimeter (Campo Imperatore, National Gran Sasso Laboratories), a large area hadron and muon detector devoted to cosmic-ray physics. It consists of iron slabs (for a total thickness of 818 g cm(-2)) and Iarocci tubes as sensitive layers, operating in the streamer mode and the "quasi proportional" regime. Using a model describing the operation of the "quasi proportional" chambers, we derive a calibration curve in the energy range 50-5000 GeV, whose reliability has been indirectly checked through on-site measurements, by means of an accelerator beam run (up to similar or equal to 600-700 GeV) and by comparing the model predictions on hadron shower transition curves with the data. Abstract

The attenuation length of proton primaries and the proton-air inelastic cross section are studied at energies E-0 approximate to 10(15) eV by using the EAS-TOP array data. Proton initiated Extensive Air Showers, in the energy range (2 divided by 4) 10(15) eV and near maximum development, are selected from their N-mu(E-mu > 1 GeV) and N-e sizes. The experimental attenuation length Lambda(obs) is compared with those obtained from simulations, including the full detector response, based on different interaction models (HDPM, VENUS, DPMJET, QGSJET and SIBYLL) in the frame of CORSIKA code. The proton-air inelastic cross section is also inferred by using the factor k = Lambda(obs)(sim)/lambda(p-air)(sim) obtained from each interaction model.

Abstract

BACKGROUND: A phase 2 trial was carried out to assess the antineoplastic activity of 2 courses of cyclophosphamide-etoposide in relapsed osteosarcoma patients. METHODS: Twenty-six relapsed osteosarcoma patients with a median age of 18.5 years (8.3-47.1) were enrolled. Seven patients were in first relapse (27%), 11 in second relapse (42%), 7 in third relapse (27%), and 1 in fourth relapse (4%). Eighteen patients had bone metastasis at study entry (69%). Cyclophosphamide was given at 4 g/m 2 on Day 1 followed by etoposide at 200 mg/m 2 on Days 2, 3, and 4. Second cyclophosphamide and etoposide was planned at 21 days to 28 days from the previous one. The primary endpoint of the study was the clinical benefit at 4 months measured as progression-free survival. RESULTS: Progression-free survival at 4 months was 42%. Five patients achieved responses (19%), 9 patients had stable disease (35%), and 12 had tumor progression (46%). Overall survival (OS) at 1 year was 50%. The only grade 4 extrahematological toxicities were fever (5%), acute bronchospasm (4%) and stomatitis (18%). Six patients (23%) underwent radical surgery after cyclophosphamide and etoposide � 2. CONCLUSIONS: Cyclophosphamide and etoposide � 2 may arrest osteosarcoma progression in a significant number of patients (54%). Osteosarcoma progression arrest after cyclophosphamide and etoposide � 2 translates in a better OS. Cyclophosphamide and etoposide � 2 had good tolerability and the toxicity was time-limited and resolved in all cases. Cancer 2009;115:2980–7. V C

Abstract

A simple prognostic classification of Ph+ chronic myeloid leukemia (CML) was proposed in 1984 by an international study Group [23] and is now widely used for clinical purposes. That study was retrospective, and was based on disease features at diagnosis. To test prospectively and to check the value of the classification, and to investigate if additional information on the early course of leukemia can help in refining the prognosis, 505 Ph+, nonblastic and nontransplanted patients first registered between 1984 and 1986 were followed up to June 1990. It was found that the prognostic formulation predicted survival in that series exactly the same way as in the original series (median survival: > 60 months for low-risk patients, 46 months for intermediate-risk patients, and 32 months for high-risk ones). It was also found that several objective or subjective assessments of disease course during the first 8 months after diagnosis were significantly related to survival length within any risk group. This study provides full confirmation of Sokal's international prognostic classification, and shows that the definition of prognosis can be improved some months after diagnosis by taking into account the course of the disease and the response to therapy. These conclusions apply to patients receiving conventional treatment. The course of Ph+ chronic myeloid leukemia (CML) is rigidly programmed to progress to an acute phase, which is called blastic metamorphosis (BM) and which can develop either suddenly (blastic crisis) or by a slow progression (accelerated phase) [7,24,29]. In all recent series, 2-year survival ranged between 65% and 80%, median survival was slightly shorter than 4 years, and the proportion of patients who were still alive after 10 years was less than 10% [24,29]. However, a number of prognostic variables were identified [2,8,9,12,15,16,19,23, 28] and a prognostic formulation that was claborated by an international study [23] was shown to provide a reliable estimate of survival length and is currently used for that purpose. The value of that formulation, and the formulation itself, require periodical controls and revisions. For that purpose, the Italian Cooperative Study Group on CML registered and followed up all CML patients who were first seen between 1984 and 1986. A very preliminary analysis of that cohort of patients has already been performed, but it was limited to the first 2 years after diagnosis [10]. This report extends prior findings, confirming the validity of the formula up to the 5th year after diagnosis, and shows that the definition of prognosis can be significantly improved by taking into account dynamic clements which reflect the course of the disease and the response to conventional treatment during the first 8 months after diagnosis.

Abstract

Horizontal Air Showers, i.e. events observed at “quasi” horizontal incidence, are studied from EAS-TOP at Campo Imperatore (National Gran Sasso Laboratories). The “reality” of such events as due to deeply penetrating particles is discussed from the experimental point of view. We deduce: • -the measurement of the HE muon flux at E μ = 20 TeV : F μ (> 20 TeV ) = (1.3 ± 0.4) × 10 −11 cm −2 s −1 sr −1 , • -the upper limit to the extraterrestrial HE neutrino flux: I ν (> 10 5 GeV ) −9 cm −2 s −1 sr −1 . • The possibility to develop the technique for UHE ν detection is discussed.

Abstract

We present the results of a search for Gamma Ray Bursts at energies E(gamma) greater than or equal to 10 GeV and E(gamma) greater than or equal to 100 TeV made by the EAS-TOP Air Shower Array in correlation with similar to 50 events detected by BATSE in the last 3 years. This analysis gives an indication of the sensitivity of air shower arrays in the detection of GRBs at energies beyond the range seen by satellite experiments.

Abstract

A study of the primary cosmic ray composition in the energy range 5ṡ1014−5ṡ1015 eV is performed through the analysis of the deep underground muons and of the e.m. component of Extensive Air Showers detected in coincidence by the MACRO and EAS-TOP experiments at the Gran Sasso Laboratory. We conclude that: a) pure proton and Iron primary beams are excluded by the experimental data; b) a fraction of nuclei heavier than Helium is necessary to explain the experimental data both below and above the knee; c) a mixed composition, obtained a priori from extrapolations of the spectra directly measured below 100 TeV, yields a reasonable account of the observations up to the knee of the primary spectrum.

Il programma AirWatch riguarda l'osservazione dallo spazio degli sciami estesi di particelle prodotti nell'atmosfera da particelle cosmiche di altissima energia. Nell'ambito di tale programma si colloca il progetto EUSO, recentemente sottoposto all'ESA da una collaborazione internazionale. In questa nota si descrivono la problematica scientifica connessa con i raggi cosmici di altissima energia, le problematiche tecniche legate all'osservazione dallo spazio degli sciami atmosferici estesi e si descrivono le linee di una possibile partecipazione dell'INFN al programma AirWatch ed al progetto EUSO in particolare.

Abstract

Granulocyte-macrophage-colony stimulating factor (GM-CSF) administration stimulates the proliferation of hemopoietic progenitors. Shortly (48-96 hours) after its discontinuation, feedback phenomena occur and the progenitor proliferation rate drops below baseline levels. As the quiescence of hyperplastic bone marrow suggests that hemopoietic cells may be refractory to the toxic effects of cytostatic drugs, the decision was made to test the hypothesis that GM-CSF given before chemotherapy may be myeloprotective.Fifty-six patients with newly diagnosed Stage II-IV Hodgkin disease, ages 18-77 years, were randomized to receive GM-CSF (5 microg/kg subcutaneously) or placebo from Day 7 to Day 4 before each chemotherapy administration (6 cycles of a hybrid of mechlorethamine, vincristine, procarbazine, and prednisone with doxorubicin, bleomycin, vinblastine, and dacarbazine). The treatment was considered a success if the delivery rate of chemotherapy was90% after 3 cycles and80% after 6 cycles.Thirty patients received GM-CSF and 26 placebo. The dose intensity (85.2% vs. 79.6%) and the overall success in terms of delivery rate (56.7% vs. 50%) were higher in the GM-CSF group, although these differences were not statistically significant. The neutrophil nadirs were higher in the GM-CSF group during the first three cycles and subsequently similar in both groups.No significant differences in terms of myelotoxicity or drug delivery were observed between the two treatment arms. Although the myeloprotective effect of the prechemotherapy administration of GM-CSF seems to be minimal, the data indicate a safe timing between GM-CSF discontinuation and further chemotherapy. Because cumulative myelotoxicity has been observed with other growth factors, given in the interval between the chemotherapy cycles, this may be relevant to the planning of rapid cycling.

Abstract

Epoetin (Epo) is physiologically present in the human body, stimulating erythropoiesis from bone marrow. Anemia is observed in cancer patients submitted to high-dose chemotherapy (HDCT), mainly caused by myelosuppression. Epoetin alfa has been widely used to treat the anemia that develops in the HDCT setting. Controlled studies in patients with hematologic malignancies or solid tumors who received Epo following HDCT have shown a decreased red blood cell transfusion requirement at least in patients receiving allogeneic bone marrow transplantation (BMT), while results in patients receiving autologous BMT have been disappointing. The administration of Epo before HDCT, in a period when bone marrow is still responsive to growth factors, may represent a new strategy aimed at decreasing the degree of anemia in these patients. A combination of granulocyte-colony stimulating factor and Epo has proved to be effective in mobilizing stem cell and committed myeloid/erythroid precursors.

Abstract

Many chemotherapy regimens are associated with variable periods of myelosuppression. In cancer patients, neutropenia (less than 500 neutrophils/microL) is the most important risk factor for infections. The incidence and severity of infectious complications are related to depth and duration of neutropenia, with the highest risk if neutrophils are less than 100/microL for more than a week. The period required for neutrophil recovery is usually short with standard regimens, but prolonged after high dose chemotherapy followed by autologous bone marrow transplant (-ABMT) or peripheral blood stem cell (PBSC) infusion. Under these conditions, the administration of granulocyte colony-stimulating factor (G-CSF) or granulocyte-macrophage colony-stimulating factor (GM-CSF) accelerates neutrophil recovery and shortens the duration of hospitalization. In standard chemotherapy settings, prophylactic use of CSF's is a matter of debate. Several studies have reached contrasting conclusion, but, combining effectiveness and costs, it results that this use of CSF'S is not to be recommended unless the risk of infections (elderly patients, reduced marrow reserve) is high. The administration of G-CSF or GM-CSF to a febrile neutropenic patient (cfr CSF's therapy) shortens the duration of neutropenia, although no great clinical benefits are evident. Nevertheless the identification of subsets of patients with additional risk factors (i.e. absolute neutrophil count100/microL at the onset of fever or delayed neutrophil recovery) should be helpful in establishing the role of CSF's therapy. When prolonged periods of severe neutropenia (less than 500 neutrophils/microL) are expected, antibiotics should be prophylactically administered. Fluoroquinolones seem to be the optimal choice in heavily myelosuppressed patients (ie. bone marrow transplant recipients). Fluoroquinolones are effective in reducing the frequency of gram-negative bacteremia, but, because of incomplete coverage, gram-positive infections are becoming increasingly problematic. The association with an agent that can be absorbed orally, active against gram-positive cocci, seems to be an effective strategy. Fungal infections are an important cause of morbility and mortality in severely neutropenic patients. Safety and efficacy of antifungal triazoles and the lipid formulations of amphotericin B used prophylactically still require investigation. In patients at high risk for fungal infections, monitoring cultures are predictive for systemic mycoses and should guide prophylactic and therapeutic choices. The standard treatment of oncologic patients with potential infectious neutropenia complications is admission to the hospital and treatment with broad-spectrum intravenous antibiotics. Until third generation cephalosporin and carbapenems became available, most neutropenic febrile patients were treated with associations of an aminoglycoside plus a beta-lactam. Monotherapy with the new antibiotics has proven to be effective as an association therapy and offers advantages in terms of cost and tolerability. Whether or not vancomycin is included in the initial antibiotic regimen should be decided on the basis of epidemiological consideration (i.e. prevalence of meticillin-resistant Staphylococcus aureus or Staphylococcus mitis in certain centers). Antifungal therapy is indicated in neutropenic patients who remain febrile after one week of broad-spectrum antibiotics or have recurrent fever. Amphotericin B should be promptly administered in patients suspected of invasive mycoses. Selected patients with fever and neutropenia, that can be identified on the basis of reduced risk of severe complications, do not need hospitalization. In the first reports, outpatient treatment has proven to be effective, cost saving and well received by patients, but further studies are needed to accurately define low risk status and the optimal home antibiotic regimens.

Abstract

Background Study results demonstrated that IFN augments BEV activity and improves median PFS in pts with mRCC. Thus, combination BEV + IFN is a standard first-line treatment option for mRCC. Combining BEV with the mTOR inhibitor EVE may be an efficacious and well-tolerated treatment option. The open-label, phase II RECORD-2 trial compared first-line EVE + BEV and IFN + BEV in mRCC. Patients and methods: Therapy-naive pts with clear cell mRCC and prior nephrectomy were randomized 1:1 to BEV 10 mg/kg IV every 2 weeks with either EVE 10 mg oral daily or IFN (9 MIU SC 3 times/week, if tolerated). Tumour assessments were every 12 weeks. Primary objective was treatment effect on progression-free survival (PFS) per central review based on an estimate of the chance of a subsequent phase III trial success (50% threshold for phase II success). Results In EVE + BEV (n = 182) and IFN + BEV (n = 183) arms, median age was 60/60 years, 76/72% of pts were men, MSKCC risk was favourable/intermediate/poor in 36/57/7% and 36/57/7% of pts, and 43/46% of pts had >2 organs involved, respectively. For EVE + BEV and IFN + BEV, median treatment duration was 8.5/8.3 months, respectively; 23/26% of pts discontinued due to AEs. In EVE + BEV and IFN + BEV arms, median PFS by central review was 9.3/10.0 months (HRIFN/EVE, 0.91; 95% CI, 0.69-1.19; P =0.485), respectively; probability of subsequent phase III success was 5.1%. Results of central and local PFS analysis were consistent. Objective response rate was 27/28% in EVE + BEV and IFN + BEV arms, respectively. Median overall survival (OS) was not reached in the EVE + BEV arm and was 25.9 months (95% CI: 21.1, 30.2) in the IFN + BEV arm. Most frequent AEs (%) were stomatitis (63), proteinuria (49), diarrhoea (39), hypertension (38), and epistaxis (35) in EVE + BEV arm and decreased appetite (45), fatigue (41), proteinuria (37), and pyrexia (35) in IFN + BEV arm. Conclusions In RECORD-2, PFS and tolerability were similar for first-line EVE + BEV and IFN + BEV. Final OS analysis will occur after 2-year follow-up. Disclosure A. Abstract

Purpose BMS-936558 is a monoclonal antibody that blocks the PD-1 co-inhibitory receptor expressed by activated T cells. This study describes its activity and safety in pts with previously treated advanced MEL. Methods BMS-936558 was administered IV q2wk to pts with various tumors at 0.1 - 10 mg/kg during dose-escalation and/or cohort expansion. Pts received up to 12 cycles (4 doses/cycle) of treatment or until unacceptable toxicity, confirmed progressive disease, or complete response. Clinical activity was assessed by RECIST v1.0. Results As of Feb 24, 2012, 104 MEL pts had received BMS-936558 at 0.1 (n = 17), 0.3 (n = 19), 1 (n = 31), 3 (n = 17), or 10 mg/kg (n = 20). ECOG performance status was 0/1/2 in 63/38/3 pts, respectively. Most pts (67/104) had received prior immunotherapy (IT); prior anti-CTLA-4, -PD-1, or -PD-L1 was not permitted. The number of prior therapies was 1 (39%), 2 (35%), or ≥3 (26%). Median therapy duration was 20 wks (range 2.0 - 121.7 wks). The incidence of grade 3 - 4 related AEs was 20% and included gastrointestinal (4%), endocrine (2%), and hepatobiliary disorders (1%). There were no drug-related deaths in MEL pts. Clinical activity (responses or prolonged stable disease) was observed at all doses (Table). Of the 26/94 (28%) evaluable responders, 19 (73%) are ongoing ranging from 1.9+ to 24.9+ months. For the 23 responders followed ≥6 months from first dose on study, 16 (70%) are progression free. ORs occurred in pts with visceral or bone metastases. Six pts (6%; 95% CI 2 - 13%) had prolonged SD (≥24 wk); 3 pts had a persistent decrease in target lesion tumor burden in the presence of new lesions and were not categorized as responders. Conclusions BMS-936558 had durable clinical benefit in pts with advanced MEL, including those who had received prior IT. Additional long-term follow-up data will be reported. Abstract

The Large Volume Detector (LVD) in the Gran Sasso underground Laboratory is a multipurpose detector consisting of a large volume of liquid scintillator interleaved with limited streamer tubes. In this paper we discuss its power to study low energy cosmic neutrinos. The results show that the first LVD tower (368 tons of liquid scintillator) is well suited to detect neutrinos from gravitational stellar collapses within all of our Galaxy over a wide range of burst duration (up to a few hundred seconds). No burst candidates have been observed in the first two months of data taking.

Abstract

Phase I and II clinical studies have shown that the hematopoietic growth factors such as granulocyte-macrophage colony-stimulating factor (GM-CSF) and granulocyte colony-stimulating factor (G-CSF) are able to reduce the period of neutropenia following standard-dose chemotherapy [1–5] and can accelerate autologous marrow engraftment after high-dose chemotherapy [6, 7]. Although the hematopoietic growth factors have been shown to be effective at increasing the absolute numbers of circulating and marrow neutrophils, the exact clinical advantage that therapy with these agents may confer remains to be demonstrated. To be of clinical advantage the circulating neutrophils need to be available in tissues where their function is necessary for host defenses. In vitro studies have shown that neutrophils exposed to GM-CSF show a decreased migration in agar [8]. By using a skin chamber assay, Peters et al. suggested a defective neutrophils migration during GM-CSF continuous infusion in patients after bone marrow transplantation (BMT) [9].

Abstract

Upper limits have been obtained to the diffuse gamma-ray emission from the Galactic disk. The limits I(gamma)/I(p) less than 0.2 percent, i.e., I(gamma)(greater than 130 TeV) less than 3.2 x 10 exp -13/q cm/s/rad provide the first measurement obtained in the angular window corresponding to the distribution of the ISM. It is still, however, a factor of about 30 higher than the flux calculated from the cosmic ray interactions by Berezinsky and Kudryavtsev (1990). A lower limit of gamma greater than 2.3 is obtained to the power-law index of the differential energy spectrum of Galactic gamma rays from 100 MeV to 100 TeV, setting a limit to the possibility of a hard extrapolation of the Galactic gamma-ray spectrum to the 100 TeV region. 21 refs.

Abstract

Transforming Growth Factor beta (TGF beta) exerts different effects on the hemopoietic system which range from the growth stimulation of more mature myeloid progenitors to the growth inhibition of more immature and multilineage hemopoietic precursors. TGF beta is also an inhibitor of the proliferation and functional activities of normal T and B lymphocytes. The aim of this study was to evaluate its effect on the growth of a human leukemic cells of lymphoid and myeloid origin.We tested its activity on the doubling time, the DNA synthesis rate and the clonal growth of a panel of lymphoid and myeloid leukemic cell lines.Among the myeloid cell lines, the proliferation in liquid cultures as well as the clonal growth of KG1, HL60 and U937 were suppressed by TGF beta 1 at doses ranging from 0.025 to 2.5 ng/ml; the degree of inhibition was, however, variable. BV 137, a Ph1-positive cell line derived from a very undifferentiated stem cell, was also highly responsive to TGF beta 1 inhibition. Among the six lymphoid neoplastic cell lines, only Nalm 6, a pre-B leukemic cell line, was consistently and reproducibly inhibited by the same doses of TGF beta 1. Conversely, two Burkitt lymphoma cell lines, Raji and Daudi, and three T-cell leukemias (Molt 4, Jurkat and PF 382) were insensitive to TGF beta inhibition.

Abstract

We performed a 3-m3 Gd experiment by doping (up to 0.1% in weight) two counters of the LVD experiment, at LNGS, with a Gd organic salt developed and produced as the result of a joint INFN/INR research activity. Feasibility of the experiment and performances of the Gd doped liquid scintillator (Gd-LS) have been presented. The chemical and physical properties of the Gd-LS and its performance as a neutron detector, namely neutron capture efficiency and average capture time are being monitored since the doping time in 2005. From laboratory survey we can state the stability of the trasmittance (T) at the reference wavelength (425 nm) with a C.L. of 81% and 96% respectively for the first and the second doped counter. This is the largest stable Gd-doped organic liquid scintillator target ever produced and continuously operated for a long period.

Abstract

The effect of recombinant human tumor necrosis factor alpha (TNF-alpha) on normal and chronic myeloid leukemia granulocyte-macrophage progenitors (CFU-GM) growing in semisolid agar cultures in the presence of recombinant granulocyte colony-stimulating factor and granulocyte-macrophage colony-stimulating factor was studied. Granulocyte-macrophage colony-stimulating factor-dependent growth of normal and chronic myeloid leukemia bone marrow CFU-GM was greatly enhanced by TNF-alpha at doses of 0.1 to 100 units/ml. Growth enhancement included neutrophil, eosinophil, and monocyte-macrophage colonies and clusters at 7 and 14 days of culture. Since similar results were achieved with highly enriched progenitor cell populations, devoid of accessory cells, an indirect effect on CFU-GM growth through the release by accessory cells of other cytokines upon TNF-alpha stimulation was thus ruled out. By contrast, the same doses of TNF-alpha inhibited the growth of normal CFU-GM in granulocyte colony-stimulating factor-dependent cultures. Taken together, our findings indicate that the final effect of TNF-alpha on normal bone marrow granulocyte-macrophage progenitor growth is dependent on the specific growth factor interacting with it, and that both normal and chronic myeloid leukemia CFU-GM are equally responsive to the combined effects of TNF-alpha and a given colony-stimulating factor.

Abstract

The EAS-TOP extensive air shower array has been operating since 1992 in the search for gamma-ray bursts at primary energies E(1) greater than or equal to 10 GeV and E(2) greater than or equal to 80 TeV. The study is performed by searching for short transients in the cosmic-ray intensity in the single particle (E(1)) and extensive air shower (E(2)) counting rates at mountain altitude (2005 m above sea level). We discuss the method and the results obtained both in sky survey and in correlation with BATSE events. In both energy ranges, the observed fluctuations in the event rate obtained in the sky survey during similar to 800 days of live time are compatible with the statistical fluctuations of the cosmic-ray background. A single candidate of time duration Delta t similar to 2 s and energy fluence F(10 < E < 100 GeV) = 1.7 x 10(-4)/(cos theta)(10.5) ergs cm(-2) (where theta is the unknown zenith angle) has been observed on 1992 July 15 at 13:22:26 UT in the energy range E(1) greater than or equal to 10 GeV with significance 10.6 and 20.1 sigma in two measurement channels. In the analysis made in correlation with similar to 50 events detected by BATSE, no burst candidate was found in time coincidence or in the 2 hr interval, around the BATSE detection time. The following ranges of upper limits F-max to the energy fluence in the time interval Delta t(90) in which BATSE detected 90% of the counts are obtained: F-max = 2.3 x 10(-5)-7.4 x 10(-3) ergs cm(-2) (10 < E < 100 GeV) F-max = 1.6 x 10(-6)-3.3 x 10(-5) ergs cm(-2) (100 < E < 1000 TeV).

The fully contained events detected in the NUSEX nucleon stability experiment have been analysed to search for possible anomalies in the fluxes of atmospheric neutrinos. The measured flux of muon neutrinos is in very good agreement with predictions and no anomaly has been found in the ratio between the rate of electron and muon neutrino events.

Abstract

We present measurements of the large-scale cosmic-ray (CR) anisotropies in R.A., using data collected by the surface detector array of the Pierre Auger Observatory over more than 14 yr. We determine the equatorial dipole component, , through a Fourier analysis in R.A. that includes weights for each event so as to account for the main detector-induced systematic effects. For the energies at which the trigger efficiency of the array is small, the “east–west” method is employed. Besides using the data from the array with detectors separated by 1500 m, we also include data from the smaller but denser subarray of detectors with 750 m separation, which allows us to extend the analysis down to ∼0.03 EeV. The most significant equatorial dipole amplitude obtained is that in the cumulative bin above 8 EeV, %, which is inconsistent with isotropy at the 6σ level. In the bins below 8 EeV, we obtain 99% CL upper bounds on d⊥ at the level of 1%–3%. At energies below 1 EeV, even though the amplitudes are not significant, the phases determined in most of the bins are not far from the R.A. of the Galactic center, at αGC = −94°, suggesting a predominantly Galactic origin for anisotropies at these energies. The reconstructed dipole phases in the energy bins above 4 EeV point instead to R.A. that are almost opposite to the Galactic center one, indicative of an extragalactic CR origin. Abstract

The study of cosmic rays at primary energies E-0 > 10(14) eV requires the detection of the different components of Extensive Air Showers through ground based stations. EAS-TOP is such an array (located at Campo Imperatore, 2000 m a.s.l., National Gran Sasso Laboratories), including detectors of the electromagnetic, muon (E-mu approximate to GeV at the surface, and E-mu approximate to TeV in coincidence with the detectors operating in the underground Gran Sasso Laboratories), hadron and atmospheric Cerenkov light components. The combined operation of the detectors is discussed from the point of view of the capability of measuring the cosmic ray primary spectrum and composition. The main lines for checking the significant features of the high energy hadron interaction model used for the interpretation of the data are outlined.

Abstract

Experimental results on the shapes of atmospheric Cherenkov light images from Extensive Air Showers at E(0) = 10(13)-10(14) eV are presented, The results are based on data obtained by the first element of the Cherenkov light array of the EAS-TOP experiment at Campo Imperatore (2005 m a.s.l., National Gran Sasso Laboratories, Italy), Images are obtained with pixel field of view omega = 1.5 x 10(-5) sr and full field of view Omega approximate to 10(-3) sr. The shapes of the images agree with the expectations and their fluctuations are large and significant, Experimental data on the dependence of the image shapes on the detection geometry are reported, The images are well reproduced by two independent side by side detectors, within measured accuracies, Such accuracies (approximate to 15% for large photoelectron content) define the level at which each pixel content is related to the shower structure, The presence of events characterized by patterns with multiple structures is proved, It is shown that such features are related to physical fluctuations of the shower structure.

Abstract

Using data collected at the Pierre Auger Observatory during the past 3.7 years, we demonstrated a correlation between the arrival directions of cosmic rays with energy above ~ 6x10^{19} electron volts and the positions of active galactic nuclei (AGN) lying within ~ 75 megaparsecs. We rejected the hypothesis of an isotropic distribution of these cosmic rays with at least a 99% confidence level from a prescribed a priori test. The correlation we observed is compatible with the hypothesis that the highest energy particles originate from nearby extragalactic sources whose flux has not been substantially reduced by interaction with the cosmic background radiation. Abstract

A study of the in vitro growth model of human granulo-monopoietic precursors (CFU-GM) before and after cryopreservation using both leukocyte feeder layers and GCT conditioned medium as the source of colony stimulating activity (CSA) is reported. The number of colonies produced with fresh cells was linearly related to the amount of marrow seeded with both CSA sources, whereas after cryopreservation this was true with feeder layers, and with GCT only at relatively high cell concentrations. This might indicate the production of granulopoietic stimulators on the part of a second population that is at least partly resistant to freezing. It seems more likely, however, that these results depend mainly on a sublethal damage to CFU-GM induced by freezing, thus making the cells hyporesponsive to some forms of CSA, such as those contained in GCT conditioned medium.

Abstract

A 90 tons Liquid Scintillation Detector (LSD) is fully running since October 1984 in the Mont Blanc Laboratory, at a depth of 5200 hg/cm2 under ground. The detection of cosmic neutrinos of different origin is the main goal of this experiment. The detector is very well shielded against the local radioactivity and is extremely sensitive to detect low energy particles with a threshold of 5 MeV.

Abstract

The influence of prostaglandin E1 (PGE1) on colonies produced by different subtypes of committed granulomacrophagic precursors (CFU-GM) was studied. Even though they represent a more immature class of precursors than day 7 CFU-GM, day 14 CFU-GM have the same sensitivity to PGE1: their growth is significantly inhibited by low PGE1 concentrations (10(-10) M or higher). Macrophagic colony formation is most sensitive to PGE1. Pure granulocytic colonies are relatively resistant and significantly inhibited by high concentrations only. Mixed granulomacrophagic colonies display an intermediate degree of sensitivity. Both day 7 and day 14 CFU-GM from chronic myeloid leukemia and idiopathic myelofibrosis patients are significantly less sensitive than normal CFU-GM. This insensitivity is not dependent on a lack of production of macrophagic colonies, since these may even be present in a higher than normal frequency.
