Abstract

The use of next-generation probiotics (NGP) in pigs for combating diseases has been subject to limited research. Here we explored the potential of a well-known NGP candidate Akkermansia muciniphilatargeting pig gut health. In the first screening experiment, we found that the abundance of A. muciniphilapeaked at 14 d old but decreased at weaning (21 d old; P < 0.05), suggesting the weaning period may be an effective window for A. muciniphilaintervention. Following that, 48 crossbred weaned pigs at 28 d old were randomly assigned to five groups: control (CON), high/low live A. muciniphila(HA/LA), and high/low heat-killed A. muciniphila(HIA/LIA). From 1 to 28 d old, the CON group received gastric infusion of anaerobic sterile saline every other day; the HA and LA groups were gavaged every other day with 1 × 1010 CFU/5 mL and 5 × 108 CFU/5 mL live A. muciniphila, respectively; and the HIA and LIA groups were gavaged every other day with 1 × 1010 CFU/5 mL and 5 × 108 CFU/5 mL heat-killed A. muciniphila, respectively. At d 29, pigs in the CON group were randomly and equally divided into two groups, one of which was named the enterotoxigenic Escherichia coli(ETEC) group, and all groups except CON received a 5-d ETEC challenge. The supplementation of A. muciniphilanumerically reduced the diarrhea rate of weaned pigs compared to the pigs that only received the ETEC challenge (P = 0.57), but the LIA group had a higher diarrhea rate than the CON group (P < 0.05). Consistent with this, the supplementation of A. muciniphilaimproved the small intestinal morphology and structure, proportion of CD4+T lymphocytes in the blood, as well as the expression of genes related to intestinal barrier and antioxidant indices of pigs with ETEC challenge, especially for the LA group (P < 0.05). Meanwhile, A. muciniphilasupplementation reduced the expression of ETEC virulence factor genes in the ileum and colon of pigs challenged by ETEC (P < 0.05). Therefore, A. muciniphilamay protect the intestinal health of weaned piglets from damage caused by ETEC infection, but the effect may vary depending on the concentration and activity of A. muciniphila.

Abstract

Simple Summary: Protease plays a crucial role in enhancing amino acid utilization in animals. This study was conducted to investigate the effects of exogenous protease in weaned pig diets with low protein. The findings demonstrated that protease added in the diets with a 1.5% reduction in soybean meal contributed to improving intestinal morphology and enhancing nutrient absorption, resulting in better growth performance. This experiment was conducted in weaned piglets to determine the effects of exogenous protease to low soybean meal (SBM) diets on growth performance, diarrhea rate, nutrient digestibility, and intestinal morphology. Seventy-two Duroc × Landrace × Yorkshire weaned barrows (21-day-old, 5.88 ± 0.95 kg) were randomly divided into four treatments with six replicates in each following a 2 × 2 factorial arrangement of SBM levels (0 to 14 d, 9%, 7.5%; 15 to 42 d, 20%, 18.5%) and protease (0 or 150 mg/kg) for a 42-day trial. Fecal samples were collected on days 11 to 14 and 38 to 42 of the experiment, and serum, intestinal tissue, and chyme samples were taken at the end of the experiments. Adding protease in low SBM diets had a significant increase in ADG (p < 0.05) and a decrease in F/G (p < 0.05). Protease significantly reduced the diarrhea rate (p < 0.05). Low SBM level decreased the apparent total tract digestibility (ATTD) of crude protein (CP) and ash (p < 0.05) but increased the ATTD of dry matter (DM), ash, organic matter (OM), and CP after the addition of protease (p < 0.05). The apparent ileal digestibility (AID) of aspartic acid (Asp), threonine (Thr), serine (Ser), alanine (Ala), lysine (Lys), and total amino acids (AAs) were significantly increased by protease supplementation (p < 0.05). Both the SBM-reduced and protease-added diets lead to lower albumin (ALB), albumin/globulin (A/G), and urea nitrogen (UREA) (p < 0.05), but greater globulin (GLOB) with low SBM diets (p < 0.05). The SBM-reduced and protease-added diets decreased the duodenum pH, respectively (p < 0.05). The protease increased the villus:crypt (V:C) in the duodenum and ileum, and ileal villus length (p < 0.05). In conclusion, the dietary supplementation of 150 mg/kg protease improved the intestinal health and performance of the weaned piglets and reversed the negative effect of a 1.5% SBM reduction in nutrient utilization, intestinal pH, and intestinal morphological parameters of weaned piglets. Abstract

Co-seismic groundwater-level and temperature changes caused by the Mw9. 0 Japan earthquake in Chinese mainland are described. The recorded water-level changes were mostly oscillations, with some step decreases mostly in the coastal area of Southeast China and step increases mostly in Northeast China and the North–south Seismic Belt. The water-temperature changes were mainly decrease with slow recovery in Sichuan-Yunnan, South China, and lower reaches of Jiangzhong, and sharp increase followed by sharp decrease, or vice versa, in North China and Northeast China. For wells that showed step changes in both water level and temperature, more showed them in the same direction than in the opposite direction.

Abstract

Tilt and strain meters of the deformation-observation network in Hubei Province all responded to the Mw9. 0 Japan earthquake on March 11, 2011. By analyzing the co-seismic responses, we found that firstly there was essentially a linear correlation between response time and epicentral distance. Secondly, there was some correlation between maximum response amplitude and earthquake magnitude as well as between the duration and earthquake magnitude. Thirdly, the response amplitudes and decay rates were different for different types of instruments. Due to less data-sampling frequency, the deformation instruments, could not display the first motion of P and S waves, but responded mainly to far-field surface waves. Before the earthquake, the NS earth tide component recorded by the cave stainmeter at Yichang was distorted for nearly eight hours. While digital deformation observation did not show complete information about the earthquake source, it still reflected some key features of seismic-wave propagation.
