Abstract

We evaluate the Community ocean Vertical Mixing (CVMix) project version of the K-profile parameterization (KPP). For this purpose, one-dimensional KPP simulations are compared across a suite of oceanographically relevant regimes against large eddy simulations (LES). The LES is forced with horizontally uniform boundary fluxes and has horizontally uniform initial conditions, allowing its horizontal average to be compared to one-dimensional KPP tests. We find the standard configuration of KPP consistent with LES across many forcing regimes, supporting the physical basis of KPP. Our evaluation motivates recommendations for "best practices" for using KPP within ocean circulation models, and identifies areas where further research is warranted. Further, our test suite can be used as a baseline for evaluation of a broad suite of boundary layer models. The original treatment of KPP recommends the matching of interior diffusivities and their gradients to the KPP predicted values computed in the ocean surface boundary layer (OSBL). However, we find that difficulties in representing derivatives of rapidly changing diffusivities near the base of the OSBL can lead to loss of simulation fidelity. We propose two alternative approaches. We find the KPP entrainment buoyancy flux to be sensitive to vertical grid resolution and details of how to diagnose the KPP boundary layer depth. We modify the KPP turbulent shear velocity parameterization to reduce resolution dependence. Additionally, our results show that the KPP parameterized non-local tracer flux is incomplete due to the assumption that it solely redistributes the surface tracer flux. However, examination of the LES vertical turbulent scalar flux budgets show that non-local fluxes can exist in the absence of surface tracer fluxes. This result motivates further studies of the non-local flux parameterization.

Abstract

A critical component of preventing the spread of vector borne diseases such as Chagas disease are door-to-door campaigns by public health officials that implement insecticide application in order to eradicate the vector infestation of households. The success of such campaigns depends on adequate household participation during the active phase as well as on sufficient follow-up during the surveillance phase when newly infested houses or infested houses that had not participated in the active phase will receive treatment. Queueing models which are widely used in operations management give us a mathematical representation of the operational efforts needed to contain the spread of infestation. By modeling the queue as consisting of all infested houses in a given locality, we capture the dynamics of the insect population due to prevalence of infestation and to the additional growth of infestation by redispersion, i.e. by the spread of infestation to previously uninfested houses during the wait time for treatment. In contrast to traditional queueing models, houses waiting for treatment are not known but must be identified through a search process by public health workers. Thus, both the arrival rate of houses to the queue as well as the removal rate from the queue depend on the current level of infestation. We incorporate these dependencies through a load dependent queueing model which allows us to estimate the long run average rate of removing houses from the queue and therefore the cost associated with a given surveillance program. The model is motivated by and applied to an ongoing Chagas disease control campaign in Arequipa, Peru.

Abstract

Some argue that political stability is best served through a two-party system. This study refutes this. The author mathematically defines the stability and rigidity of electoral systems comprised of any quantity of electors and parties. In fact, stability is a function of the quantity of electors - i.e., the number of occupied seats at the table. As the number of electors increases, the properties of an electorate are increasingly well resolved, and well described by those of an electorate that is least excessive -- that is to say an electorate that is closest to equilibrium. Further, electoral rigidity is a function of the quantity of parties and their probabilities of representation. An absolutely rigid system admits no fluctuations -- whatever happens to one elector will happen to all electors. As the quantity of parties increases so does the number of party lines, and with it the quantity of alternatives with which to respond to an external stimulus. Rigidity is significant in a social system that places high value on party loyalty. In conclusion, (i) electoral stability is best served by increasing the quantity of electors; (ii) electoral rigidity is best served by decreasing the quantity of parties, and by increasing the representation of some parties at the expense of others; and (iii) the less stable a branch of government, the more concern is placed on those who would hold those offices for the people., Comment: 34 pages (body = 28, appendix = 6), 3 Figures, 3 tables, includes 2 apendices

Abstract

Transient spine enlargement (3-5 min timescale) is an important event associated with the structural plasticity of dendritic spines. Many of the molecular mechanisms associated with transient spine enlargement have been identified experimentally. Here, we use a systems biology approach to construct a mathematical model of biochemical signaling and actin-mediated transient spine expansion in response to calcium-influx due to NMDA receptor activation. We have identified that a key feature of this signaling network is the paradoxical signaling loop. Paradoxical components act bifunctionally in signaling networks and their role is to control both the activation and inhibition of a desired response function (protein activity or spine volume). Using ordinary differential equation (ODE)-based modeling, we show that the dynamics of different regulators of transient spine expansion including CaMKII, RhoA, and Cdc42 and the spine volume can be described using paradoxical signaling loops. Our model is able to capture the experimentally observed dynamics of transient spine volume. Furthermore, we show that actin remodeling events provide a robustness to spine volume dynamics. We also generate experimentally testable predictions about the role of different components and parameters of the network on spine dynamics.
