In experiments with rooted cuttings of aspen (Populus tremula L). with a small leaf area, it was found that the roots grew well as long as there was no shoot growth. The onset of shoot growth was followed by a period of decreased root growth. When the leaf area had increased sufficiently, root growth recovered. Decreasing the shoot growth by removal of growth points in the shoot or by short day treatment increased the fraction of photosynthesis products used for root growth, leading to increased root/shoot ratios. Competition between growing shoots and roots for carbohydrates formed in photosynthesis is considered to cause the effects noted and to be of importance for maintaining the balance between the root and shoot systems.

The major ether-soluble, growth-stimulating substance detected by the Avena coleoptile straight-growth test in extract from sprouting buds of Scots pine (Pinus silvestris L.) was identified as indole-3-acetic acid by Rf values in 5 solvent systems and by its elution volume in ethanol on a Sephadex LH-20 column. When the substance was applied to the growth solution of wheat roots in a special test the growth in length of the roots was at first inhibited, but growth was recovered after about 6 hours in the same manner as when small quantities of IAA were applied. The extracts also contained large amounts of growth inhibitors which interfered with the auxin response if they were not removed.

Application of 10 to 100 μg indol-3-ylacetic acid to the leaves of rooted cuttings of aspen caused inhibition of root growth after three hours. Root growth recovered within 24 hours after IAA treatment. Swelling of the root tips occurred during the period of inhibition. The roots responded in the same way if IAA was applied in solution to the cut stem surface above the mature leaves. IAA-1-14C applied through a cut stem surface or to mature leaves was translocated downwards in the plants and labelled IAA could be isolated from the roots 3 to 24 hours after application. The ethanol-soluble activity decreased rapidly indicating a rapid metabolism or binding of IAA. IAA-1-14C applied to growing leaves was not translocated. From the rapid response of root growth it was concluded that IAA was translocated into the roots at a rate of about 7 cm per hour. This rate of translocation indicates that the sieve tubes are involved in the translocation. Implications of the results for the translocation of endogenous auxin into the roots are discussed.

Stem cuttings from 3-year-old seedlings of Picea abies L. (Karst.) (Norway spruce) were rooted in tap water in a growth chamber. The effect of two levels of irradiance given to either the upper two thirds of the cuttings or to the whole cutting was investigated. When only the upper two thirds of the cuttings were irradiated, the number of cuttings which rooted was lower at the higher irradiance level (40 W m−2) as compared with the number rooting at the lower irradiance level (8 W m−2). Rooting was inhibited almost completely at either of the irradiances if the base of the cutting was also irradiated.

Indoleacetic acid substantially increased the inhibitory influence of abscisic acid on growth measured on fresh weight basis of Lemna gibba L. A similar synergistic action was obtained with indolebutyric acid while neither naphthylacetic acid nor 2,4-dichlorophenoxyacetic acid showed any synergism. The antiauxin para-chlorophenoxy-isobutyric acid did not counteract the synergistic action of IAA and ABA. The results indicate that the enhancing effect of IAA on the ABA action is not a typical auxin effect.

The inhibitory effects of indole-3-acetic acid (IAA) and 1-aminocyclopropane-1-carboxylic acid (ACC) on elongation growth of pea (Pisum sativum L.) seedling roots were investigated in relation to the effects of these compounds on ethylene production by the root tips. When added to the growth solution both compounds caused a progressively increasing inhibition of growth within the concentration range of 0.01 to 1 micromolar. However, only ACC increased ethylene production in root tips excised from the treated seedlings after 24 hours. High auxin concentrations caused a transitory increase of ethylene production during a few hours in the beginning of the treatment period, but even in 1 micromolar IAA this increase was too low to have any appreciable effect on growth. ACC, but not IAA, caused growth curvatures, typical of ethylene treatment, in the root tips. IAA caused conspicuous swelling of the root tips while ACC did not. Cobalt and silver ions reversed the growth inhibitory effects induced by ACC but did not counteract the inhibition of elongation or swelling caused by IAA. The growth effects caused by the ACC treatments were obviously due to ethylene production. We found no evidence to indicate that the growth inhibition or swelling caused by IAA is mediated by ethylene. It is concluded that the inhibitory action of IAA on root growth is caused by this auxin per se.
