Abstract

Traditional associative learning theories predict that training with feature negative (A+/AB-) contingencies leads to the feature B acquiring negative associative strength and becoming a conditioned inhibitor (i.e., prevention learning). However, feature negative training can sometimes result in negative occasion setting, where B modulates the effect of A. Other studies suggest that participants learn about configurations of cues rather than their individual elements. In this study, we administered simultaneous feature negative training to participants in an allergist causal learning task and tested whether evidence for these three types of learning (prevention, modulation, configural) could be captured via self-report in the absence of any procedural manipulation. Across two experiments, we show that only a small subset of participants endorse the prevention option, suggesting that traditional associative models that predict conditioned inhibition do not completely capture how humans learn about negative contingencies. We also show that the degree of transfer in a summation test corresponds to the implied causal structure underlying conditioned inhibition, occasion-setting, and configural learning, and that participants are only partially sensitive to explicit hints about causal structure. We conclude that feature negative training is an ambiguous causal scenario that reveals individual differences in the representation of inhibitory associations, potentially explaining the modest group-level inhibitory effects often found in humans. Abstract

Studying generalisation of associative learning requires analysis of response gradients measured over a continuous stimulus dimension. In human studies, there is often a high degree of individual variation in the gradients, making it difficult to draw conclusions about group-level trends with traditional statistical methods. Here, we demonstrate a novel method of analysing generalisation gradients based on hierarchical Bayesian curve-fitting. This method involves fitting an augmented (asymmetrical) Gaussian function to individual gradients and estimating its parameters in a hierarchical Bayesian framework. We show how the posteriors can be used to characterise group differences in generalisation and how classic generalisation phenomena such as peak shift and area shift can be measured and inferred. Estimation of descriptive parameters can provide a detailed and informative way of analysing human generalisation gradients. [ABSTRACT FROM AUTHOR]

Abstract

VGX-1027, a novel oral immune modulator, is under development for the treatment of rheumatoid arthritis. The safety, tolerability, and pharmacokinetics of single (1-800 mg) and multiple (40-400 mg) oral doses were evaluated in 2 clinical studies. The doses were well tolerated up to 800 mg in a single dose and 200 mg twice daily in multiple doses. Adverse events were mild to moderate in severity with no identifiable dose-related pattern. There were no clinically significant physical or laboratory findings. The pharmacokinetic data indicated that increases in Cmax and AUC0-inf were dose-proportional, and AUC0- τ was approximately dose-proportional. For the single-dose study, median Tmax ranged from 0.5 to 2 hours and mean t1/2 ranged from 4.9 to 8.7 hours. For the multiple-dose study, median Tmax ranged from 0.5 to 2.0 hours and mean t1/2 ranged from 7.05 to 10.05 hours. No accumulation of the drug was observed after day 1, indicating that steady-state concentrations were attained with single and multiple dosing for 5 days. Approximately 90% of the administered dose was excreted in urine as unchanged drug.

60. Negative evidence and inductive reasoning in generalization of associative learning.

Abstract

When generalizing properties from known to novel instances, both positive evidence (instances known to possess a property) and negative evidence (instances known not to possess a property) must be integrated. The current study compared generalization based on positive evidence alone against a mixture of positive evidence and perceptually dissimilar negative evidence in an interdimensional discrimination procedure. In 2 experiments, we compared generalization following training with a single positive stimulus (that predicted shock) against groups where an additional negative stimulus (that did not predict shock) was presented in a causal judgment (Experiment 1) and a fear conditioning (Experiment 2) procedure. In contrast to animal conditioning studies, we found that adding a "distant" negative stimulus resulted in an overall increase in generalization to stimuli varying on the dimension of the positive stimulus, consistent with the inductive reasoning literature. We show that this key qualitative result can be simulated by a Bayesian model that incorporates helpful sampling assumptions. Our results suggest that similar processes underlie generalization in inductive reasoning and associative learning tasks. [ABSTRACT FROM AUTHOR]

Abstract

In many eukaryotes, translation initiation is regulated by proteins that bind to the mRNA cap-binding protein eukaryotic translation initiation factor 4E (eIF4E). These proteins commonly prevent association of eIF4E with eIF4G or form repressive messenger ribonucleoproteins that exclude the translation machinery. Such gene-regulatory mechanisms in plants, and even the presence of eIF4E-interacting proteins other than eIF4G (and the plant-specific isoform eIFiso4G, which binds eIFiso4E), are unknown. Here, we report the discovery of a plant-specific protein, conserved binding of eIF4E 1 (CBE1).We found that CBE1 has an evolutionarily conserved eIF4E-binding motif in its N-terminal domain and binds eIF4E or eIFiso4E in vitro. CBE1 thereby forms cap-binding complexes and is an eIF4E-dependent constituent of these complexes in vivo. Of note, plant mutants lacking CBE1 exhibited dysregulation of cell cycle-related transcripts and accumulated higher levels of mRNAs encoding proteins involved in mitosis than did WT plants. Our findings indicate that CBE1 is a plant protein that can form mRNA cap-binding complexes having the potential for regulating gene expression. Because mammalian translation factors are known regulators of cell cycle progression, we propose that CBE1 may represent such first translation factor- associated plant-specific cell cycle regulator. Jaded magazine is a form of alternative media to encourage political, cultural, and personal discourse among UCI students. We celebrate and support the Asian Pacific Islander community through the retelling of the past, engaging of the present, and sharing a vision for the future. We hope to build connections and bridge gaps between people of different ethnicities and ways of thinking. The goal of the publication is not only to provide a space where Asian Pacific Islander students can voice different opinions and artistic expressions, but also as a form of community activism through education and awareness. Despite the fact that we are misrepresented, our images misconstrued, and our cultures misunderstood we are not JADED in spirit. This is what we are doing about it., Action Figures. We have all become stoic bodies that watch bad reality TV, eat bad fast food, and listen to bad one dimensional news reporting. The sense of urgency is completely gone. The only people moving are the action figures in our community. In the issue of Jaded, we found some cool-hippy folks to inspire us, to get off our collective ass and do something awesome. Let it be art, making a zine, starting a strike, standing up for your body, inventing sunks, or saying “No” to bad Asian porn, we should at least waste our time on something interesting. This is what we are doing about it., Americans are afraid of the wrong things. Our administration employs the politics of fear to take advantage of the legitimate concerns of citizens. We are told to be afraid for our security when the Technicolor alert is elevated from tangerine orange to salmon pink. However, our real security problem lies in the inability of our national emergency response system to help Americans in catastrophic events. The fear of terrorism is reframed to target illegal immigrants from Latin America, while the government sends more secret agents to track Fidel Castro than Osama Bin Ladin. Our fears are exploited to justify unchecked powers and invasion of nations. Issue seven of Jaded discusses the scary tactics of military recruitment in our schools, the profit motives of petition collectors, the antagonism of civilian border patrollers, the fear of the “other” in suburbia, the lack of accountability in police brutality cases, and our future with the oil dilemma. This is what we are doing about it., The second amalgamation of our brains ... I am exhausted in trying to make my identity known, but jaded I am not. It's hard enough dealing with post teen angst and , for some of us, quarter-life crisis, to really sit down and discuss the issues with identity. Our identities are always questioned, whether by the media, family, friends, politicians, cultural studies experts, and even ourselves. This second issue of Jaded will focus on our individual identities-and on our collective identity. Often in the media, we are portrayed as simplistic, one dimensional caricatures of kung fu masters, lotus blossoms, sex fetishes, or geekish, slightly tone deaf, American idol enthusiasts. 75. Clinical validation of the watershed sign as a marker for neuropsychiatric systemic lupus erythematosus

Abstract

To study the relationship between single‐photon–emission computed tomography (SPECT) brain imaging and neuropsychiatric signs/symptoms in a cohort of patients with systemic lupus erythematosus (SLE), analyzed using a stereotactic surface projection (SSP) technique.Thirty‐seven SLE patients were referred for 99mTc–ethyl cysteinate dimer SPECT brain imaging because of neuropsychiatric signs/symptoms. Nineteen normal controls were studied with the identical protocol. Reconstructed images were computed and Z scores were calculated using the SSP technique with the 2‐sample t‐tests comparing normal controls with SLE patients, and patients with mild cognitive dysfunction with those with severe cognitive dysfunction. The clinical characteristics of SLE patients were collected by retrospective chart review and categorized according to American College of Rheumatology case definitions for neuropsychiatric SLE. Cognitive dysfunction was rated by the treating physician on a scale of 0–3.Thirty of 37 SLE patients had abnormal SPECT results. SLE patients had reduced perfusion in the watershed areas of the frontal lobes bilaterally compared with controls. Additionally, SLE patients with severe cognitive dysfunction had more severe perfusion deficits than those with mild cognitive dysfunction. In some patients with severe cognitive dysfunction, the watershed areas had Z scores ≥4 SDs below controls.A convenience sample of patients with SLE and neuropsychiatric signs/symptoms demonstrated reduced perfusion in the watershed areas of the frontal lobes on SPECT scanning analyzed by the SSP technique. The severity of findings correlated with severity of cognitive dysfunction. The area of the brain affected is one that is susceptible to ischemia.

76. A Mixture of Experts in Associative Generalization

Abstract

After learning that one stimulus predicts an outcome (e.g., an aqua-colored rectangle leads to shock) and a very similar stimulus predicts no outcome (e.g., a slightly greener rectangle leads to no shock), some participants generalize the predictive relationship on the basis of physical similarity to the predictive stimulus, while others generalize on the basis of the relational difference between the two stimuli (e.g., “higher likelihood of shock for bluer stimuli”). To date, these individual differences in generalization rules have remained unexplored in associative learning. Here, we present evidence that a given individual simultaneously entertains belief in both “similarity” and “relational” rules, and generalizes using a mixture of these strategies. Using a “mixture of experts” modelling framework constrained by participants self-reported rule beliefs, we show that considering multiple rules predicts generalization gradients better than a single rule, and that generalization behavior is better described as switching between, rather than averaging over, different rules.

The article reports on the study conducted by the periodical regarding the rise of recruitment in the county's largest law firms in Orange County, California. It states that the boost in mergers and acquisitions, securities work, patents and deals fueled hiring at these law firms in 2007. Research shows that the 50 largest law firms based in the county saw a 6% gain in lawyers for a total of 2,117 attorneys. Law firms foretold that such growth will continue in 2008.
