Rice is characterized by a broad range of metabolic and morphological adaptations to flooding, such as germination and mobilization of stored nutrients under submergence until seedlings reach the water surface to carry out photosynthesis, and sustainable growth of mature plants for long durations under partial submergence. The underlying mechanisms of the molecular basis of adaptation to anaerobic germination and seedling growth in rice are being uncovered. Induction of an ensemble of hydrolases to mobilize endosperm nutrient reserves is one of the key factors for successful germination and coleoptile elongation in rice under submergence. To compensate for reduced efficiency of Tricarboxylic Acid cycle and oxidative respiration in mitochondria under O2 deficient conditions, α-amylases play a central role in the hydrolysis of starch to provide sugar substrates for glycolysis and alcohol fermentation for generating ATP. We review the progress on the molecular mechanism regulating α-amylase expression that involves the integration of signals generated by the hormone gibberellin (GA), sugar starvation and O2 deprivation that results in germination and sustainable seedling growth in rice under anaerobic conditions. Comparisons are also made between dicots and monocots for the molecular mechanism of induction of genes involved in alcohol fermentation and sugar/O2 deficiency sensing system.

In plants, source-sink communication plays a pivotal role in crop productivity, yet the underlying regulatory mechanisms are largely unknown. The SnRK1A protein kinase and transcription factor MYBS1 regulate the sugar starvation signaling pathway during seedling growth in cereals. Here, we identified plant-specific SnRK1A-interacting negative regulators (SKINs). SKINs antagonize the function of SnRK1A, and the highly conserved GKSKSF domain is essential for SKINs to function as repressors. Overexpression of SKINs inhibits the expression of MYBS1 and hydrolases essential for mobilization of nutrient reserves in the endosperm, leading to inhibition of seedling growth. The expression of SKINs is highly inducible by drought and moderately by various stresses, which is likely related to the abscisic acid (ABA)–mediated repression of SnRK1A under stress. Overexpression of SKINs enhances ABA sensitivity for inhibition of seedling growth. ABA promotes the interaction between SnRK1A and SKINs and shifts the localization of SKINs from the nucleus to the cytoplasm, where it binds SnRK1A and prevents SnRK1A and MYBS1 from entering the nucleus. Our findings demonstrate that SnRK1A plays a key role regulating source-sink communication during seedling growth. Under abiotic stress, SKINs antagonize the function of SnRK1A, which is likely a key factor restricting seedling vigor.

Cytosolic CuZnSOD removes deleterious superoxides from plant cells. In order to understand its function better, we sought to express a monocot CuZnSODgene in transgenic Arabidopsis. We constructed a transgene usingthe CaMV 35S promoter to express a rice cytosolic CuZnSOD gene in Arabidopsis and generated over 200 transformants. A 16 kD polypeptide, the same size as the native rice CuZnSOD polypeptide, was detected inthe transgenic Arabidopsis. Interestingly, two forms of riceCuZnSOD, rSODI and rSODII, having the same dimeric size, were detectedin the transgenic plants. rSODII protein was relatively abundant but hadlow specific activity. In contrast, rSODI protein was relatively rareand had high specific activity. Inter-conversion of rSODI and rSODIIcould be achieved by the addition and removal of copper ions into the purifiedrecombinant SOD and to the leaf extract of transgenic plants. Ouranalysis indicates that rSODI most likely corresponds to native riceCuZnSOD that has incorporated the Cu and Zn ions required for fullactivity, whereas the less active rSODII form may not have properlyincorporated the necessary copper ions.

Standard Bonner spheres and proposed high-sensitivity Bonner cylinders were calibrated in a neutron calibration room, using a (252)Cf source. The Bonner sphere system consists of 11 polyethylene (PE) spheres of various diameters and 4 extended spheres that comprise embedded metal shells. Similar to the design of Bonner spheres, a set of Bonner cylinders was assembled using a large cylindrical (3)He tube as the central probe, which was wrapped using various thicknesses of PE. A layer of lead was employed inside one of the PE cylinders to increase the detection efficiency of high-energy neutrons. The central neutron probe used in the Bonner cylinders exhibited an efficiency of ∼17.9 times higher than that of the Bonner spheres. However, compared with the Bonner spheres, the Bonner cylinders are not fully symmetric in their geometry, exhibiting angular dependence in their responses to incoming neutrons. Using a series of calculations and measurements, this study presents a systematic comparison between Bonner spheres and cylinders in terms of their response functions, detection efficiencies, angular dependences and spectrum unfolding.

In order to overcome the obstacles in traditional experimenting and practical training courses, as well as in enhancing the functions of the present e-learning system, the study took sensor network technology as the foundation in developing a web services system. The system will be able to make presentations of the students ‘operations and results on an immediate basis, allowing the students to be guided adequately as they face problems during experiment and practical training.

Mobile technological products have now become part of people's lives. As such, the tablet computer is really a new-generation PC that is very popular among business people as well as students, due to its comfort and mobility, and because it functions well. The purpose of this explanatory and exploratory research survey was to test a hypothesized model regarding customers' perceptions of brand image, the sacrifices involved in the purchase, value, satisfaction, brand loyalty, and repurchase intention. Data was collected from 818 responses via questionnaire survey, and all scales were examined for reliability and construct validity. The findings indicated that (a) customers' positive perceptions of brand image have a significant positive effect on brand loyalty; and (b) customers' perceived level of sacrifice has a significant negative effect on customer satisfaction. The limitations and recommendations for future research are also included in this study.

The purpose of this research is introduce the design of an improved graphical user interface (GUI) and verifies the operational efficiency of the proposed interface. Until now, clicking the toolbar with the mouse is the usual way to operate software functions. In our research, we designed an improved graphical user interface – a tool ball that is operated by a mouse wheel to perform software functions. Several experiments are conducted to measure the time needed to operate certain software functions with the traditional combination of “mouse click + tool button” and the proposed integration of “mouse wheel + tool ball”. The results indicate that the tool ball design can accelerate the speed of operating software functions, decrease the number of icons on the screen, and enlarge the applications of the mouse wheel.

Sugars repress α-amylase expression in germinating embryos and cell cultures of rice (Oryza sativa) through a sugar response complex (SRC) in α-amylase gene promoters and its interacting transcription factor MYBS1. The Snf1 protein kinase is required for the derepression of glucose-repressible genes in yeast. In this study, we explored the role of the yeast Snf1 ortholog in rice, SnRK1, in sugar signaling and plant growth. Rice embryo transient expression assays indicated that SnRK1A and SnRK1B act upstream and relieve glucose repression of MYBS1 and αAmy3 SRC promoters. Both SnRK1s contain N-terminal kinase domains serving as activators and C-terminal regulatory domains as dominant negative regulators of SRC. The accumulation and activity of SnRK1A was regulated by sugars posttranscriptionally, and SnRK1A relieved glucose repression specifically through the TA box in SRC. A transgenic RNA interference approach indicated that SnRK1A is also necessary for the activation of MYBS1 and αAmy3 expression under glucose starvation. Two mutants of SnRK1s, snrk1a and snrk1b, were obtained, and the functions of both SnRK1s were further studied. Our studies demonstrated that SnRK1A is an important intermediate in the sugar signaling cascade, functioning upstream from the interaction between MYBS1 and αAmy3 SRC and playing a key role in regulating seed germination and seedling growth in rice.

Using transfer DNA (T-DNA) with functions of gene trap and gene knockout and activation tagging, a mutant population containing 55,000 lines was generated. Approximately 81% of this population carries 1-2 T-DNA copies per line, and the retrotransposon Tos17 was mostly inactive in this population during tissue culture. A total of 11,992 flanking sequence tags (FSTs) have been obtained and assigned to the rice genome. T-DNA was preferentially ( approximately 80%) integrated into genic regions. A total of 19,000 FSTs pooled from this and another T-DNA tagged population were analyzed and compared with 18,000 FSTs from a Tos17 tagged population. There was difference in preference for integrations into genic, coding, and flanking regions, as well as repetitive sequences and centromeric regions, between T-DNA and Tos17; however, T-DNA integration was more evenly distributed in the rice genome than Tos17. Our T-DNA contains an enhancer octamer next to the left border, expression of genes within genetics distances of 12.5 kb was enhanced. For example, the normal height of a severe dwarf mutant, with its gibberellin 2-oxidase (GA2ox) gene being activated by T-DNA, was restored upon GA treatment, indicating GA2ox was one of the key enzymes regulating the endogenous level of GA. Our T-DNA also contains a promoterless GUS gene next to the right border. GUS activity screening facilitated identification of genes responsive to various stresses and those regulated temporally and spatially in large scale with high frequency. Our mutant population offers a highly valuable resource for high throughput rice functional analyses using both forward and reverse genetic approaches.
