Abstract

IShTAR is a linear magnetized plasma test facility dedicated to the investigation of RF wave/plasma interaction. The IShTAR ICRF system consists of a single strap RF antenna. When using the antenna for plasma production without an external plasma source, it is shown that the plasma is either produced in front of the antenna strap or inside the antenna box depending on the antenna parameters. Here, we present experimental and numerical investigation of the plasma initiation parametric dependencies. Detailed pressure and RF power scans were performed in helium at f = 5.22 MHz and f = 42.06 MHz. The experiment shows the parameter ranges for which the plasma is produced in front of the strap, or inside the antenna box. These ranges are validated by simulations with the RFdinity model, and by theoretical predictions.

Abstract

Cell motility on ECM critically depends on the cellular response to force from the matrix. We find that force-dependent reinforcement of [[alpha].sub.v]/[[beta].sub.3]-integrin-mediated cell-matrix connections requires the receptorlike tyrosine phosphatase [alpha] (RPTP[alpha]). RPTP[alpha] colocalizes with [[alpha].sub.v]-integrins at the leading edge during early spreading, and coimmunoprecipitates with [[alpha].sub.v]-integrins during spreading on fibronectin and vitronectin. RPTP[alpha]-dependent activation of Src family kinases, in particular activation of Fyn, is required for the force-dependent formation of focal complexes and strengthening of [[alpha].sub.v]/[[beta].sub.3]-integrin-cytoskeleton connections during the initial phase of ECM contact. These observations indicate that Src family kinases have distinct functions during adhesion site assembly, and that RPTP[alpha] is an early component in force-dependent signal transduction pathways leading to the assembly of focal complexes on both fibronectin and vitronectin.

Abstract

Nuclear fusion is a promising candidate to supply energy for future generations. At the high temperatures needed for the nuclei to fuse, ions and electrons are no longer bound into atoms. Magnetic fields confine the resulting plasma. One of the heating methods is the ion cyclotron resonant absorption of waves emitted by an external Ion Cyclotron Radio Frequency (ICRF) antenna. The efficiency of ICRF heating is strongly affected by rectified RF electric fields at antenna and other in-vessel components (so-called ‘sheath effects’). The chapter presents an overview of ICRF principles. Attention is given to characterising the detrimental sheath effects through experiments on a dedicated test facility (IShTAR: Ion cyclotron Sheath Test ARrangement). IShTAR has a linear magnetic configuration and is equipped with an independent helicon plasma source. The configuration and capabilities of the test-bed and its diagnostics are described, as well as an analysis of the plasmas.

Abstract

Introduction: CAR T-cell therapy is currently being evaluated in non-oncologic autoimmune diseases (Mougiakakos D et al. N Engl J Med2021; Müller F et al. Lancet2023). In a cohort of patients (pts) with SLE, CD19 CAR T-cell therapy resulted in B-cell depletion and treatment-free, durable remission per Definitions of Remission In SLE (DORIS), suggesting that CD19 CAR T therapy may be feasible, well-tolerated, and effective for patients with SLE (Mackensen A et al. Nat Med2022). Baseline traits associated with greater manufacturing success and lower toxicity profiles for CAR T have been identified in hematologic oncology (Mashadi-Hossein A et al. J Clin Oncol2023; Rytlewski J et al. J Clin Oncol2022) including age, bone marrow function, inflammatory state, and tumor burden. Here, we compared baseline traits in pts with active SLE versus R/R LBCL to predict the potential manufacturing success and toxicity profile for the ongoing phase 1 study to evaluate the investigational NEX-T TMCAR T-cell product BMS-986353 (CC-97540) in pts with severe, refractory SLE (NCT05869955).

Abstract

Udgivelsesdato: 2007-Nov-1, Receptor-like protein tyrosine phosphatase alpha (RPTPalpha)-knockout mice have severe hippocampal abnormalities similar to knockouts of the Src family kinase Fyn. These enzymes are linked to the matrix-rigidity response in fibroblasts, but their function in neurons is unknown. The matrix-rigidity response of fibroblasts appears to differ from that of neuronal growth cones but it is unknown whether the rigidity detection mechanism or response pathway is altered. Here, we report that RPTPalpha is required for rigidity-dependent reinforcement of fibronectin (FN)-cytoskeleton bonds and the rigidity response in hippocampal neuron growth cones, like in fibroblasts. In control neurons, rigid FN surfaces inhibit neurite extension and neuron differentiation relative to soft surfaces. In RPTPalpha(-/-) neurons, no inhibition of extension and differentiation is found on both rigid and soft surfaces. The RPTPalpha-dependent rigidity response in neurons is FN-specific, and requires clustering of alpha(v)beta(6) integrin at the leading edge of the growth cones. Further, RPTPalpha is necessary for the rigidity-dependent concentration of Fyn and p130Cas phosphorylation at the leading edge of the growth cone, like it is in fibroblasts. Although neurons respond to rigid FN surfaces in the opposite way to fibroblasts, we suggest that the mechanism of detecting FN rigidity is similar and involves rigidity-dependent RPTPalpha recruitment of Fyn.
