Developmental brain health is paramount to the future of all society. As such, many western countries have heavily invested in several large-scale studies of brain development including the Adolescent Brain Cognitive Development (ABCD) study, and the Healthy Brain and Child Development (HBCD) study. However, since brain development and brain determinants of health are multidimensional and variable, true advancement requires a broader sampling of our populations – a global effort beyond western countries. To this end, advancing global brain health science requires a large effort in developing measures and testing protocols, methodological approaches, theoretical perspectives and systems that have cross-cultural relevance, and can be employed to generate globally representative data to inform inclusive public health interventions. In this White Paper, we discuss the daunting challenges of population neuroscience with specific focus on children and adolescents. After an executive summary and a concise overview of the major issues yet to be resolved, we present the global longitudinal study on child and adolescent development (GLAD) initiative and explain how it could potentially be deployed to generate globally relevant data to optimize brain health in young people across diverse contexts. Throughout the document, we emphasize that to optimize children’s brain health, we need to foster strong collaboration among multiple stakeholders including researchers, clinicians, educators, policy experts, and funding organizations.

International audience; Model-based data analysis of whole-brain dynamics links the observed data to model parameters in a network of neural masses. Recently, studies focused on the role of regional variance of model parameters. Such analyses however necessarily depend on the properties of preselected neural mass model. We introduce a method to infer from the functional data both the neural mass model representing the regional dynamics and the region-and subject-specific parameters while respecting the known network structure. We apply the method to human resting-state fMRI. We find that the underlying dynamics can be described as noisy fluctuations around a single fixed point. The method reliably discovers three regional parameters with clear and distinct role in the dynamics, one of which is strongly correlated with the first principal component of the gene expression spatial map. The present approach opens a novel way to the analysis of resting-state fMRI with possible applications for understanding the brain dynamics during aging or neurodegeneration.

Quantitative maps of neurotransmitter receptor densities are important tools for characterising the molecular organisation of the brain and key for understanding normal and pathologic brain function and behaviour. We describe a novel method for reconstructing 3-dimensional cortical maps for data sets consisting of multiple different types of 2-dimensional post-mortem histological sections, including autoradiographs acquired with different ligands, cell body and myelin stained sections, and which can be applied to data originating from different species. The accuracy of the reconstruction was quantified by calculating the Dice score between the reconstructed volumes versus their reference anatomic volume. The average Dice score was 0.91. We were therefore able to create atlases with multiple accurately reconstructed receptor maps for human and macaque brains as a proof-of-principle. Future application of our pipeline will allow for the creation of the first ever set of ultra-high resolution 3D atlases composed of 20 different maps of neurotransmitter binding sites in 3 complete human brains and in 4 hemispheres of 3 different macaque brains.

Population neuroimaging allows for extracting general principles of brain-phenotype relationships. Capturing individual brain-behavior profiles in groups with pronounced inter-individual variability, like the older adult population, however, remains challenging. Therefore, deep characterization is required to link multilevel brain, cognitive and lifestyle data. We here proposed a use case of five older males scoring low on a dementia screening test. We showed quite heterogeneous individual cognitive, lifestyle and grey matter atrophy profiles. Integrating additional regional genetic, molecular and connectional data using a multilevel atlas framework revealed (dis-) similarities between the atrophied brain areas, thereby helping to explain the individual phenomena and emphasizing the need for integrating multifactorial and multilevel information on the way toward individualized predictions.

The temporal cortex encompasses a large number of different areas ranging from the six-layered isocortex to the allocortex. The areas support auditory, visual, and language processing, as well as emotions and memory. The primary auditory cortex is found at the Heschl gyri, which develop early in ontogeny with the Sylvian fissure, a deep and characteristic fissure that separates the temporal lobe from the parietal and frontal lobes. Gyri and sulci as well as brain areas vary between brains and between hemispheres, partly linked to the functional organization of language and lateralization. Interindividual variability in anatomy makes a direct comparison between different brains in structure-functional analysis often challenging, but can be addressed by applying cytoarchitectonic probability maps of the Julich-Brain atlas. We review the macroanatomy of the temporal lobe, its variability and asymmetry at the macro- and the microlevel, discuss the relationship to brain areas and their microstructure, and emphasize the advantage of a multimodal approach to address temporal lobe organization. We review recent data on combined cytoarchitectonic and molecular architectonic studies of temporal areas, and provide links to their function.

Based on quantitative cyto- and receptor architectonic analyses, we identified 35 prefrontal areas and introduced a novel subdivision of Walker’s areas 10, 9, 8B and 46. Statistical analysis of receptor densities revealed regional differences in lateral and ventrolateral prefrontal cortex. Since structural and functional organization of subdivisions encompassing areas 46 and 12 demonstrated significant differences in the interareal levels of α2 receptors. Furthermore, multivariate analysis included receptor fingerprints of previously identified 16 motor areas in the same macaque brains, and revealed five clusters encompassing frontal lobe areas. Based on the functional connectivity analysis, clustered areas showed similar connectivity distribution pattern. In particular, rostrally located areas (in clusters 1-2) were characterized by bigger fingerprints, i.e., higher receptor densities, and stronger regional interconnections. Whereas, more caudal areas (in clusters 3-5) had smaller fingerprints, but showed a widespread connectivity pattern with distant cortical regions. Taken together, present study provides a comprehensive insight into the molecular structure underlying the functional organization of the cortex and, thus, reconcile discrepancies between the structural and functional hierarchical organization of the primate frontal lobe. Finally, our data are publicly available via the EBRAINS and BALSA repositories for the entire scientific community.

In rodents, gene-expression, neuronal tuning, connectivity and neurogenesis studies have postulated that the dorsal, the intermediate and the ventral hippocampal formation (HF) are distinct entities. These findings are underpinned by behavioral studies showing a dissociable role of dorsal and ventral HF in learning, memory, stress and emotional processing. However, up to now, the molecular basis of such differences in relation to discrete boundaries is largely unknown. Therefore, we analyzed binding site densities for glutamatergic AMPA, NMDA, kainate and mGluR2/3 , GABAergic GABAA (including benzodiazepine binding sites), GABAB , dopaminergic D1/5 and noradrenergic α1 and α2 receptors as key modulators for signal transmission in hippocampal functions, using quantitative in vitro receptor autoradiography along the dorsal-ventral axis of the mouse HF. Beside general different receptor profiles of the dentate gyrus (DG) and Cornu Ammonis fields (CA1, CA2, CA3, CA4/hilus), we detected substantial differences between dorsal, intermediate and ventral subdivisions and individual layers for all investigated receptor types, except GABAB . For example, striking higher densities of α2 receptors were detected in the ventral DG, while the dorsal DG possesses higher numbers of kainate, NMDA, GABAA and D1/5 receptors. CA1 dorsal and intermediate subdivisions showed higher AMPA, NMDA, mGluR2/3 , GABAA , D1/5 receptors, while kainate receptors are higher expressed in ventral CA1, and noradrenergic α1 and α2 receptors in the intermediate region of CA1. CA2 dorsal was distinguished by higher kainate, α1 and α2 receptors in the intermediate region, while CA3 showed a more complex dissociation. Our findings resulted not only in a clear segmentation of the mouse hippocampus along the dorsal-ventral axis, but also provides insights into the neurochemical basis and likely associated physiological processes in hippocampal functions. Therein, the presented data has a high impact for future studies modeling and investigating dorsal, intermediate and ventral hippocampal dysfunction in relation to neurodegenerative diseases or psychiatric disorders.

Cytoarchitecture is a basic principle of microstructural brain parcellation. We introduce Julich-Brain, a three-dimensional atlas containing cytoarchitectonic maps of cortical areas and subcortical nuclei. The atlas is probabilistic, which enables it to account for variations between individual brains. Building such an atlas was highly data- and labor-intensive and required the development of nested, interdependent workflows for detecting borders between brain areas, data processing, provenance tracking, and flexible execution of processing chains to handle large amounts of data at different spatial scales. Full cortical coverage was achieved by the inclusion of gap maps to complement cortical maps. The atlas is dynamic and will be adapted as mapping progresses; it is openly available to support neuroimaging studies as well as modeling and simulation; and it is interoperable, enabling connection to other atlases and resources.

These structural variants are present in 0.5% to 1.0% of the population, making 15q11.2 BP1-BP2 the site of the most prevalent known pathogenic copy number variation (CNV). It is unknown to what extent this CNV influences brain structure and affects cognitive abilities., [Objective] To determine the association of the 15q11.2 BP1-BP2 deletion and duplication CNVs with cortical and subcortical brain morphology and cognitive task performance., [Design, Setting, and Participants] In this genetic association study, T1-weighted brain magnetic resonance imaging were combined with genetic data from the ENIGMA-CNV consortium and the UK Biobank, with a replication cohort from Iceland. In total, 203 deletion carriers, 45 247 noncarriers, and 306 duplication carriers were included. Data were collected from August 2015 to April 2019, and data were analyzed from September 2018 to September 2019., [Main Outcomes and Measures] The associations of the CNV with global and regional measures of surface area and cortical thickness as well as subcortical volumes were investigated, correcting for age, age2, sex, scanner, and intracranial volume. Additionally, measures of cognitive ability were analyzed in the full UK Biobank cohort., [Results] Of 45 756 included individuals, the mean (SD) age was 55.8 (18.3) years, and 23 754 (51.9%) were female. Compared with noncarriers, deletion carriers had a lower surface area (Cohen d = −0.41; SE, 0.08; P = 4.9 × 10−8), thicker cortex (Cohen d = 0.36; SE, 0.07; P = 1.3 × 10−7), and a smaller nucleus accumbens (Cohen d = −0.27; SE, 0.07; P = 7.3 × 10−5). There was also a significant negative dose response on cortical thickness (β = −0.24; SE, 0.05; P = 6.8 × 10−7). Regional cortical analyses showed a localization of the effects to the frontal, cingulate, and parietal lobes. Further, cognitive ability was lower for deletion carriers compared with noncarriers on 5 of 7 tasks., [Conclusions and Relevance] These findings, from the largest CNV neuroimaging study to date, provide evidence that 15q11.2 BP1-BP2 structural variation is associated with brain morphology and cognition, with deletion carriers being particularly affected. The pattern of results fits with known molecular functions of genes in the 15q11.2 BP1-BP2 region and suggests involvement of these genes in neuronal plasticity. Publisher's version (útgefin grein), Carriers of large recurrent copy number variants (CNVs) have a higher risk of developing neurodevelopmental disorders. The 16p11.2 distal CNV predisposes carriers to e.g., autism spectrum disorder and schizophrenia. We compared subcortical brain volumes of 12 16p11.2 distal deletion and 12 duplication carriers to 6882 non-carriers from the large-scale brain Magnetic Resonance Imaging collaboration, ENIGMA-CNV. After stringent CNV calling procedures, and standardized FreeSurfer image analysis, we found negative dose-response associations with copy number on intracranial volume and on regional caudate, pallidum and putamen volumes (β = −0.71 to −1.37; P < 0.0005). In an independent sample, consistent results were obtained, with significant effects in the pallidum (β = −0.95, P = 0.0042). The two data sets combined showed significant negative dose-response for the accumbens, caudate, pallidum, putamen and ICV (P = 0.0032, 8.9 × 10−6, 1.7 × 10− 9, 3.5 × 10−12 and 1.0 × 10−4, respectively). Full scale IQ was lower in both deletion and duplication carriers compared to non-carriers. This is the first brain MRI study of the impact of the 16p11.2 distal CNV, and we demonstrate a specific effect on subcortical brain structures, suggesting a neuropathological pattern underlying the neurodevelopmental syndromes., 1000BRAINS: 1000BRAINS is a population-based cohort based on the Heinz-Nixdorf Recall Study and is supported in part by the German National Cohort. The sagittal stratum is a prominent and macroscopically clearly visible white-matter structure within occipital and parietal lobes with a highly organized structure of parallel fibers running in rostro-caudal direction. Apart from the major tract running through, i.e., the optic radiation, the source and arrangement of other fibers within the sagittal stratum is only partially understood. Recent diffusion imaging studies in-vivo suggest additional minor fiber directions, perpendicular to the major rostro-caudal ones, but the spatial resolution does not allow to resolve them, and to unambiguously distinguish it from noise. Taking this previous evidence as motivation, the present study used 3D polarized light imaging (3D-PLI) for micrometer resolution analysis of nerve fibers in postmortem specimens of a vervet monkey brain. The analysis of coronal occipital and parietal sections revealed that the sagittal stratum consisted of an external and an internal layer, which are joined and crossed by fibers from the surrounding white matter and the tapetum. Fibers from different parietal and occipital regions entered the sagittal stratum in the dorsal, ventral or middle sector, as solid large bundles or as several small fiber aggregations. These patterns were remarkably similar to published results of tracer experiments in macaques. Taking this correspondence as external validation of 3D-PLI enabled translation to the human brain, where a similarly complex fiber architecture within the sagittal stratum could be exemplified in a human hemisphere in our study. We thus argue in favor of a dedicated fiber microstructure within the sagittal stratum as a correlate of the additional fiber directions typically seen in in-vivo diffusion imaging studies.
